A novel indole derivative (1) and three known compounds (24) were isolated from the fruiting bodies of Tricholoma flavovirens. Their structures were determined or identified by the interpretation of spectroscopic data. Compounds 1 and 2 promoted root growth of lettuce and inhibited hypocotyl growth at 1 μmol/paper. Compound 3 inhibited hypocotyl and root growth at 100 nmol/paper.  相似文献

Phenylalanine ammonia-lyase, which catalyzes the conversion of l-phenylalanine to trans-cinnamic acid and ammonia, has been partially purified from the cells of Rhodotorula. Some of the properties of this phenylalanine ammoyia-lyase were investigated. The enzyme was stable in phosphate buffer of pH over the range of 6.0 to 7.0 On heating, the enzyme was stable up to 50°C, but above 60°C, it was destroyed. The enzyme activity was strongly inhibited by p-chloromercuribenzoate at 10?5 m and almost recovered by the addition of glutathione or mercaptoethanol at 10?3 m. The present enzyme preparation of Rhodotorula also catalyzed the deamination of l-tyrosine to trans-p-coumaric acid. trans-p-Coumaric acid was isolated from the reaction mixture and identified by its absorption spectra. The rates of deamination showed optima at pH 9.0 and 9.5 for l-phenylalanine and l-tyrosine, respectively.  相似文献

3-Methylthiopropylamine (MTPA) formation from l-methionine in Streptomyces sp. K37 was studied in detail. The reaction was confirmed to be catalyzed by the decarboxylase of l-methionine. The properties of the enzyme were studied in detail using acetone dried cells or cell-free extract. The enzyme was specific for l-methionine. Pyridoxal phosphate stimulated the reaction and protected the enzyme against heat inactivation. The optimum pH for the reaction was 6.0~8.0 and the optimum temperature was about 40°C. Carbonyl reagents (10?2~10?3 m) inhibited the reaction completely, and silver nitrate and mercuric chloride (10?3~10?4 m) markedly inhibited the reaction. Km value for the reaction was 1.21 × 10?5 m. l-Methionine assay using the decarboxylase was attempted and was found to be applicable to practical use.  相似文献

Certain Streptomyces strains were found to accumulate an unknown substance in culture broth when the microorganisms were grown in the medium containing dl-methionine. The substance was isolated from the culture broth as hydrochloride and was identified as 3-methylthiopropylamine (MTPA), decarboxylated product of methionine, from its melting point, chemical composition, infrared spectrum, and other properties. Cultural conditions for MTPA formation in Streptomyces sp. K 37 were investigated. The yield of MTPA from l-methionine reached about 90% with a culture medium containing corn steep liquor. Namely, 6.47 mg of MTPA per millilitre of culture broth was produced from 10 mg of l-methionine per millilitre of the growth medium. The transforming activity was found in the cells of the early culture period. MTPA-producing activity was induced by l- methionine in the medium. d-Methionine was not utilized as a substrate of the reaction with intact cells. Optimum pH for the reaction appeared to be 6.0~8.0.  相似文献

Certain strains of Streptomyces were found to convert l-methionine into 3-methylthio-propylamine (MTPA), but not d-methionine. Now, optical resolution of DL-methionine was attempted using this phenomenon. Streptomyces sp. K37 was cultured in a medium containing DL-methionine (10 mg/ml). The culture filtrate was applied to a column of Diaion SA-21A (OH form). MTPA was recovered from the effluent by ether exraction. The Diaion SA-21A was eluted with 1N HCl and the eluate was applied to a column of Diaion SK-1 (H form). d-Methionine was eluted from the column with 1N NH4OH and recovered after concentration, decolorization with active carbon, and precipitation with ethanol. The yields of MTPA and d-methionine from the broth were 69.5% and 89.5%, respectively.  相似文献

不同产地松茸氨基酸与矿质元素的比较研究   总被引:2,自引:0,他引:2  
以西南地区的三种松茸子实体为材料 ,用柱前衍生高效液相色谱法和原子吸收分光光度法分别对松茸子实体的氨基酸和矿质元素进行了测定 ,氨基酸的测定结果表明 :采自四川通江的松茸子实体的氨基酸总含量最高 ,为 10 .5 82 % ;其次为采自重庆的松茸子实体为 9.6 0 7% ;最低的为采自云南昌隆的松茸子实体 ,为 3.381%。矿质元素的测定结果表明 :所测定的七种元素中 ,含量较高的有三种 ,分别为K ,Ca和Mg ,平均含量分别为 3.1× 10 4mg/Kg ,1.6 2× 10 3mg/Kg和 5 .5 8× 10 2mg/Kg。在这三种松茸子实体中云南昌隆的Fe ,Mn含量最高 ,分别为 6 2 .2 82mg/Kg和 6 .0 6 4mg/Kg ;重庆的Cu含量最高 ,为 5 2 .84 5mg/Kg;Zn的含量在这三种子实体中差异不大。  相似文献

Aromatic amino acids (phenylalanine, tyrosine and tryptophan) were heated at 300°C under nitrogen and volatile compounds generated were examined. Twelve compounds in which many of them have aromatic rings were identified in the volatiles from thermal degradation of phenylalanine. Tyrosine and tryptophan produced some phenols and indoles, respectively, besides several compounds. Formation mechanisms of some compounds were also discussed.  相似文献

建立了红豆杉悬浮培养细胞中游离芳香族氨基酸的高效毛细管电泳的分离直接紫外吸收测定方法。在优化的实验条件下 ,以间苯二酚为内标 ,未涂层融硅毛细管 (75 μmi.d .,总长 37cm ,有效分离长度 30cm)为分离通道 ,40mmol/L磷酸缓冲液 (pH10 .46 )为电泳介质 ,3sec(0 .5 psi)压力进样 ,15kV恒压电泳 ,2 0 0nm检测 ,在 9min内三种氨基酸得到分离测定。色氨酸、苯丙氨酸和酪氨酸的线性范围分别为 2 .5 0~ 2 5 0 μmol/L(r =0 .996 2 ,RSD =2 .5 2 % )、1.2 5~ 2 5 0 μmol/L(r =0 .996 6 ,RSD =2 .12 % )和 2 .5 0~ 2 5 0 μmol/L(r=0 .9940 ,RSD =2 .93% ) ,苯丙氨酸回收率为 96 .8%± 1.37% ,酪氨酸回收率为 99.2 %± 3.0 4%。  相似文献

Kinetic studies of the transport of aromatic amino acids by Pseudomonas aeruginosa revealed the existence of two high-affinity transport systems which recognized the three aromatic amino acids. From competition data and studies on the exchange of preformed aromatic amino acid pools, the first transport system was found to be functional with phenylalanine, tyrosine, and tryptophan (in order of decreasing activity), whereas the second system was active with tryptophan, phenylalanine, and tyrosine. The two systems also transported a number of aromatic amino acid analogues but not other amino acids. Mutants defective in each of the two and in both transport systems were isolated and described. When the amino acids were added at low external concentrations to cells growing logarithmically in glucose minimal medium, the tryptophan pool very quickly became saturated. Under identical conditions, phenylalanine and tyrosine each accumulated in the intracellular pool of P. aeruginosa at a concentration which was 10 times greater than that of tryptophan.  相似文献

观察了支链氨基酸(BCAA)对大鼠运动能力和血清游离氨基酸代谢的影响。实验用21只雄性wistar大鼠,随机分为3组:正常组、游泳对照组和游泳补充BCAA组。2个运动组每天游泳训练1h,10天后游泳6h,观察补充BCAA对大鼠游泳运动能力和血清游离氨基酸水平的影响。实验结果表明,补充BCAA可明显提高大鼠游泳存活率,抑制血清中必需氨基酸、非必需氨基酸和总氨基酸水平升高,游泳运动后大鼠的血清中乳酸和LDH的升高幅度有所降低,抑制骨骼肌LDH活力的下降。说明补充BCAA可明显提高大鼠的运动能力,减少运动造成的蛋白质分解  相似文献

应用外标法测定当归静脉注射液中游离氨基酸和总氨基酸的含量。当归静脉注射液中含有15种氨基酸,其中游离氨基酸最高的是精氨酸,游离氨基酸最低的是甘氨酸。总氨基酸最高的是谷氨酸,总氨基酸最低的是赖氨酸。当归静脉注射液中氨基酸种类丰富,特定氨基酸如精氨酸、谷氨酸含量较高,是其临床特定一些疗效的基础之一。  相似文献

Shikimate, anthranilate, indole, l -tryptophan, phenylpyruvate, l -p henylalanine, p-hydroxyphenylpyruvate or l -tyrosine were added to suspension-cultured Nicotiana tabacum (tabacco) and Daucus carota (carrot) tissues and incubated for 24 hours. Uptake of each compound was substantial as measured by its decrease in the medium. The levels of free tryptophan, phenylalanine and tyrosine were determined in the tissues after the 24 hours incubation. Shikimate did not change the aromatic animo acid levels in carrot tissue, but did increase all three in tobacco (3-fold or more), indicating a less stringent feedback control in tobacco. Anthranilate and indole increased the tissue tryptophan levels in both species by at least 17-fold, showing that the flow from anthranilate and indole to tryptophan was apparently unhindered by enzymatic control mechanisms. When tryptophan levels were elevated in both carrot and tobaccotissues by anthranilate, indole or tryptophan addition, there was also an increase in free phyenylalanine and tyrosine. This might be due to the reversal of phenylalanine and tyrosine feedback inhibition of chorismate mutase by the high tryptophan in the tissue. Chorismate mutase activity in tobacco crude extracts could be inhibited by 66–90% by 1 mM phenylalanine and /or tyrosine. Tryptophan at 1 mM stimulated the enzyme activity by 1/3 and completely reversed the phenylalanine and/or tyrosine inhibition of enzyme activity. Chorsimate mutase activity amino acids under a variety of conditions. Phenylpyruvate increased the phenylalanine levels and p-hydroxyphenylpyruvate increased the tyrosine levels in carrot and tobacco tissues indicating that there was no feedback control of the last step in phenylalanine and tyrosine biosynthesis.  相似文献

The transport of neutral amino acids through the brain capillary endothelial wall, which makes up the blood-brain barrier (BBB) in vivo, is an important control point for the overall regulation of cerebral metabolism, including protein synthesis and neurotransmitter production. The Michaelis-Menten kinetics of BBB amino acid transport have been investigated in vivo with the brain uptake index (BUI) technique, and in vitro with the isolated human brain capillary preparation. The only amino acid that is albumin-bound is tryptophan, and the majority of albumin-bound tryptophan in the plasma is available for transport through the BBB via an enhanced dissociation mechanism that operates at the surface of the brain capillary endothelium. The availability in brain of amino acids is predicted from the BBB Km values to be sharply influenced by supra-physiological concentrations of phenyalanine in the 200–500 M range. Moreover, the measurement of cerebral protein synthesis with an internal carotid artery perfusion technique and HPLC-based measurements of aminoacyl-transfer RNA specific activities shows an inverse relationship between cerebral protein synthesis and plasma phenyalanine concentrations in the 200–500 M range. These findings indicate the neurotoxicity of hyperphenylalninemia is not restricted to the phenylketonuria range of approximately 2000 M, but is exerted in the supra-physiological range of 200–500 M.  相似文献
