Dextran is a polysaccharide produced commercially by Leuconostoc mesenteroides B512(F) growing on excess sucrose. The high viscosity of the resulting broth makes process analysis and control extremely difficult. In an attempt to overcome this the broth rheology was monitored with time and correlated with other process variables. It was determined that the viscosity and pseudo-plasticity increase through most of the fermentation, but then go through a sharp maximum immediately prior to completion of dextran synthesis and sucrose consumption. This was attributed to release of dextran molecules from the cell wall, reducing the size of polymer aggregates in solution. As such, the change in rheology provides a marker for the completion of the fermentation. The analysis requires no pretreatment of the broth, and it is envisaged that an on-line viscometer, determining apparent broth viscosity at a single shear rate, could be used for process monitoring and control. This would enable early detection of operating upsets, as well as avoid errors introduced by manual sampling and analysis, ans permit the optimum point at which to harvest the broth to be selected more precisely. Summary Aclacinomycin A, an antitumour antibiotic, was produced by fermentation with Streptomyces lavvendofoliae DKRS. Aclacinomycin A analogue compounds, such as aclacinomycins B and Y which are less effective as antitumour agents, were also produced in significant amounts. These analogue compounds could be converted to aclacinomycin A by incubation with the washed cell preparation as previously reported with S. galilaeus(Hoshino, T., Sekine, Y., Fujiwara, A. (1983) J.Antibiot. 36, 1458–1462). It was also found that the analogue compounds could be metabolically converted to aclacinomycin A by changing the pH of the culture broth at a later stage of fermentation. This metabolic conversion is expected to lead to additional benefits in the purification of aclacinomycin A as well as its increased production.  相似文献

A method for the accumulation of the streptomycin precursor (L) in the culture broth of Streptomyces griseus was developed and the precursor was successfully isolated from the broth.

When the microorganism was cultured under shaking in the glucose-meat extract-peptone medium (0.5% glucose, 0.2% yeast extract, 0.2% meat extract, 0.4% peptone, 0.5% sodium chloride, 0.025% magnesium sulfate, pH 7.0), the accumulation of the precursor in the broth was induced by the addition of supplementary glucose (e.g., 2 g glucose per 100 ml broth) 24 hr after inoculation followed by further cultivation for 48 hr. Increased accumulation of L component was obtained merely by increasing glucose content in the culture medium (e.g., 5% glucose-containing medium in the above-indicated one) instead of glucose supplement on the way of fermentation. For the accumulation of a large amount of L component in a culture broth, it looked to be necessary for pH value of the broth to be maintained between 6 and 7 during fermentation.

L component was isolated from the culture broth by adsorption on Amberlite IRC-50 and elution with 2% NaCl solution. The L component was separated on this column from contaminated streptomycin which requires 5% NaCl solution to be eluted. The L component in the 2% NaCl eluate was adsorbed on active carbon at neutral or slightly alkaline pH and eluted with 95% methanol at acidic pH, Partially purified L component precipitated as hydrochloride by addition of acetone to the methanol extract which had been concentrated in vacuo.  相似文献

The capability of the yeast Lodderomyces elongisporus to utilize solved paraffins in fermentation brothes could be demonstrated. The growth rate of this microorganism in the case of utilization of solved paraffins is higher as the most known dates. The saturated concentrations of solved hydrocarbons in the fermentation brothes are higher as in real solvent systems. The part of the solved hydrocarbon is a function of the power input, the diameter of oil drops, the fermentation conditions and the length of the paraffin chain. The organism growth rate depends on the solved paraffin concentration in the fermentation broth. This fact is one of the reasons for the variability of the consumption coefficients by utilization of paraffins with different chain lenghts. The results confirm the assumption that the transport of the paraffins from the oil drops to the cells takes place over water soluble phase.  相似文献

A bacterium isolated from Burdwan (India) soil was found to accumulatel-lysine in the growth medium and was identified asArthrobacter globiformis. The strain grew and accumulated lysine in a purely synthetic medium. Supplementation of the synthetic medium stimulated growth but did not improve the yield. The entire fermentation period could be divided into a growth phase and a production phase, which could be prolonged by adjustment of pH to the neutral range. Among the different hydrocarbon and nitrogen sources tested SR gas oil at 4 % and ammonium sulphate at 0.4 %, respectively, were found most to be suitable. Different vitamins and antibiotics stimulated growth and lysine yield; inoculum of 7 % (V/V) of the medium was found to be optimal. The yield of lysine under optimal conditions was 3.4 g per litre medium. Lysine was isolated in crystalline form from the fermented broth by IEC and found to be a purel-isomer.  相似文献

采用研磨法分离纯化猪笼草各组织器官中的内生菌,并采用水解圈法和摇瓶发酵法分别进行初筛与复筛,对筛选出的菌株进行16SrDNA分析鉴定。结果表明:在猪笼草叶片、捕虫囊、根和茎等4种器官中共分离出25株内生菌;进一步从中筛选出2株可产胞外蛋白酶的细菌A3、L5,其中菌株A3的产酶能力较高,水解圈/菌落直径比(D/d)值为6.5,发酵液的蛋白酶活力为17.58U/mL;菌株L5的D/d值为3.0,发酵液的蛋白酶活力为15.77U/mL;16SrDNA鉴定结果表明,菌株A3、L5与芽孢杆菌属成员具有99%的同源性,其中A3是枯草芽孢杆菌(Ba-cillus subtilis),L5可能是其的新变种或新亚种。  相似文献

【背景】由禾谷镰刀菌(Fusarium graminearum)引起的小麦赤霉病严重威胁我国的小麦生产。【目的】筛选对禾谷镰刀菌具有拮抗能力的链霉菌菌株,为生防菌剂开发提供理论基础。【方法】利用平板对峙法筛选对禾谷镰刀菌具有拮抗能力的链霉菌;通过形态特征、生理生化特征和16S rRNA基因序列分析对其进行鉴定;通过病原菌菌丝生长、孢子产生及萌发抑制试验分析其发酵液的抑菌活性;利用人工接种试验测定该菌株发酵液的防病效果。【结果】筛选到一株对禾谷镰刀菌具有较强拮抗活性的链霉菌21-1,抑菌率为59.5%。依据形态特征、生理生化特性和16S rRNA基因序列分析,将该菌株鉴定为黄三素链霉菌(Streptomycesflavotricini)。菌株21-1发酵液能够抑制禾谷镰刀菌的菌丝生长、孢子产生及萌发过程,而且可以降低禾谷镰刀菌菌丝中可溶性蛋白质的含量,并增加丙二醛的含量。菌株21-1可以产生蛋白酶及纤维素酶。菌株21-1菌液10倍稀释液对小麦赤霉病的防效最佳,为70.1%。此外,菌株21-1发酵液对其他8种植物病原菌均有较好的抑制作用。【结论】菌株21-1对禾谷镰刀菌有较好的抑菌活性,具...  相似文献

Summary Candida lipolytica is shown to produce an extracellular polymer with emulsifying properties when grown on n-tetradecane or a mixture of linear hydrocarbons. A device for biosurfactant isolation is presented. The polymers recovered from the fermentation broth were found to be complex molecules with a protein, a lipid and a carbohydrate moiety. Their surface active properties suggest a possible role in hydrocarbon uptake by cells. Effectively, the addition of crude polymer resulted in an enhancement of respiration rate which was dependent on n-alkane concentration with glucose grown cells. Likewise in batch culture, maximum growth rate, cell productivity and yield were increased by the presence of the biosurfactant.  相似文献

Intermittent broth replacement was carried out to enhance the productivity and purity of sodium gluconate usingAspergillus niger by reducing the concentration of unmetabolized glucose. As inoculum size increased, length of lag phase was shortened and high initial production rate of sodium gluconate was achieved. However, too high inoculum concentration lowered productivity during the later stage of fermentation and increased residual glucose at the end of cultivation. When culture broth was replaced intermittently with distilled water, fresh medium, or recycled medium for comparison with traditional fermentation method, production of sodium gluconate was enhanced more than 1.5 fold and active production period could be prolonged without residual glucose. In addition, productivity was maintained at a level higher than 6 g/L/nr. Therefore, it was found that the reduction of biomass and viscosity by intermittent broth replacement could enhance the productivity.  相似文献

A stable and fast l-tryptophan producer, AGX1757, was isolated from Escherichia coli W3110 trpAE1 trpR tnaA, which carried pSC101-trpI15·14. Cells of AGX1757 did not lose the composite plasmid during fermentation. Whenever a fed-batch culture of AGX1757 attained an l-tryptophan concentration of about 30 g/l, indole began to appear in the broth. The emergence of indole was caused by inhibition of tryptophan synthase due to accumulated l-tryptophan. Hence, the production rate of l-tryptophan sharply decreased. A higher solubility of l-tryptophan in the supernatant of culture broth (about 32 g/l) than that in the initial medium (about 22 g/l) was attributed to some unknown interaction between l-tryptophan and certain macromolecular material(s) coming from the bacterial cells. An addition of non-ionic detergents into the supernatant was effective for decreasing the solubility of l-tryptophan, hence causing crystallization of l-tryptophan. Pluronic L-61 was supplied from outside to an extent of 0.5% in terms of wt% concentration at around 45 h of fermentation when the l-tryptophan accumulated reached about 25 g/l. This addition actually caused crystallization of l-tryptophan and, as a result, the inhibitory effect of tryptophan synthase by l-tryptophan accumulated in the broth could be alleviated. A physico-chemical, two phase simulated pseudoplastic fermentation (SPF) broth was investigated in which Solka Floc cellulose fibre was used to simulate the filamentous biomass, and a mixture of 0.1% (w/v) carboxymethyl cellulose (CMC) and 0.15 M aqueous sodium chloride was used to simulate the liquid fraction of the fermentation broth. An investigation of the rheological behaviour and hydrodynamic properties of the SPF broth was carried out, and compared to both a fungal Tolypocladium inflatum fermentation broth and a CMC solution in a 50 L stirred tank bioreactor equipped with conventional Rushton turbines. The experimental data confirmed the ability of the two phase SPF broth to mimic both the T. inflatum broth bulk rheology as well as the mixing and mass transfer behaviour. In contrast, using a homogeneous CMC solution with a similar bulk rheology to simulate the fermentation resulted in a significant underestimation of the mass transfer and mixing times. The presence of the solid phase and its microstructure in the SPF broth appear to play a significant role in gas holdup and bubble size, thus leading to the different behaviours. The SPF broth seems to be a more accurate simulation fluid that can be used to predict the bioreactor mixing and mass transfer performance in filamentous fermentations, in comparison with CMC solutions used in some previous studies.  相似文献

The enzymatic digestibility of alkali/peracetic acid (PAA)-pretreated bagasse was systematically investigated. The effects of initial solid consistency, cellulase loading and addition of supplemental β-glucosidase on the enzymatic conversion of glycan were studied. It was found the alkali-PAA pulp showed excellent enzymatic digestibility. The enzymatic glycan conversion could reach about 80% after 24 h incubation when enzyme loading was 10 FPU/g solid. Simultaneous saccharification and fermentation (SSF) results indicated that the pulp could be well converted to ethanol. Compared with dilute acid pretreated bagasse (DAPB), alkali-PAA pulp could obtain much higher ethanol and xylose concentrations. The fermentation broth still showed some cellulase activity so that the fed pulp could be further converted to sugars and ethanol. After the second batch SSF, the fermentation broth of alkali-PAA pulp still kept about 50% of initial cellulase activity. However, only 21% of initial cellulase activity was kept in the fermentation broth of DAPB. The xylose syrup obtained in SSF of alkali-PAA pulp could be well converted to 2,3-butanediol by Klebsiella pneumoniae CGMCC 1.9131.  相似文献

Two strains of Bacillus, one from a culture collection (B. subtilis ATCC 6633) and a wild type (Bacillus sp. UFLA 817CF) isolated during coffee fermentation in the south of Minas Gerais, Brazil, were evaluated in relation to secretion of alkaline proteases. The strains were grown on nutrient broth, nutrient broth with sodium caseinate and nutrient broth with three different concentrations of cheese whey powder for 72 h. Samples were collected at 24-h intervals to evaluate the proteolytic activity, protein content and cell population. Maximum protease activity was observed after 24-h growth for both the microorganisms, a period that coincided with the end of the exponential phase. The specific activity values were, respectively, 839.8 U/mg for B. subtilis ATCC 6633 and 975.9 U/mg for Bacillus sp. UFLA 817CF. The 60% saturation presented the best results for specific protease activity in all the growth culture media tested with B. sp. UFLA 817CF. Bacillus sp. UFLA 817CF showed highest enzymatic activity at pH 9.0 and 40°C in the three culture media tested. The protease obtained from culture of the wild Bacillus strain presented stability at pH 7.0 and considerable heat stability at 40°C and 50°C, and could be an alternative for the industry to utilize cheese whey to produce proteolytic enzymes.  相似文献

【目的】通过中药美白成分的复配,进而研究复方中药发酵液活性成分及功效性。【方法】以酪氨酸酶抑制率为指标研究最佳复配含量,采用分光光度法测定复方中药发酵液中总酚、多糖、黄酮、蛋白质含量,通过HPLC技术进行氨基酸分析,抗氧化能力通过总还原力和自由基清除试验进行评价,通过酪氨酸酶活性抑制实验进行美白功效分析。【结果】正交实验得出4种中药美白成分中覆盆子对酪氨酸酶抑制效果最好;复方中药发酵液中多酚、多糖、黄酮和蛋白质含量分别为147.80、4.36、1.17、2.22 g/kg;复方中药发酵液具有较高的总还原力、羟自由基清除能力和DPPH自由基清除能力;5%(W/V)复方中药发酵液的酪氨酸酶活性抑制率为82.41%。【结论】复方中药发酵液中含有多种活性成分,具有一定的抗氧化和美白功效。  相似文献

Beauveria bassiana endophytically colonises corn (Zea mays) reducing tunneling from European corn borer (Ostrinia nubilalis). Endophytic colonisation of other plants by B. bassiana has been reported, and potentially, may reduce insect feeding on these plants. We evaluated the effects on larval growth and development, and mortality of different rates of dried, ground mycelia and water-soluble metabolites from fermentation broth culture of different isolates of B. bassiana incorporated into a synthetic diet and fed to neonate bollworm, Helicoverpa zea larvae. Development was delayed, weights of larvae were lower, and mortality was high for larvae fed the highest rates (1.0 and 5.0%, w/v) of mycelia incorporated diet compared to control. Insects fed diets containing mycelia of B. bassiana isolate 11-98 had the greatest mortality. Mortality was 100% for larvae fed 5% (w/v) mycelia incorporated diet of isolate 11-98, and 61% for isolate 3-00. For insects fed low rates (0.1 to 0.5%, w/v) of mycelia incorporated diet, mortality was lower, approximately 5% for isolate 11-98, and 5 to 14% for isolate 3-00. At the 0.1% (w/v) rate of mycelia incorporated diet, development occurred at an accelerated rate, compared to fungus-free controls, indicating increased nutrition in the lowest rate fungal diet. Mortality was low for all larvae fed diets containing spent fermentation broth of B. bassiana; however, development was delayed. Insects fed the highest rate (0.5%, v/v) of spent fermentation broth-amended diet had lower pupal weights, and a greater number of days to pupation than insects fed the lowest (0.1%, v/v) rate. Insects fed the 5% (v/v) rate of spent fermentation broth of isolates 11-98 and 3-00 had the longest days to pupation.  相似文献

水稻内生细菌群落是反映植株内环境是否健康稳定的重要生物学指标,芽胞杆菌是防治水稻病害的重要生防微生物。为揭示芽胞杆菌浸种处理对水稻内生细菌群落结构的影响,采用Illumina MiSeq测序的方法对水稻内生细菌的16S rRNA基因进行测序,剖析了芽胞杆菌浸种处理对不同水稻组织内生细菌的微生态调控作用。结果表明,3种芽胞杆菌浸种处理可以提高水稻根和茎部内生细菌群落的丰富度和均匀度,降低叶部内生细菌群落的丰富度和均匀度,显著增加根部内生细菌群落多样性。变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)和拟杆菌门(Bacteroidetes)是水稻根部和茎部共有优势菌门,厚壁菌门和芽胞杆菌属(Bacillus)是叶部共有优势菌门和属。芽胞杆菌浸种处理显著提高了叶部内生厚壁菌门和芽胞杆菌属的相对丰度,增加了根系和茎部组织内生细菌的分类单元OTU(Operational Taxonomic Units)数量,对叶部组织影响不明显;降低了茎部和叶部中参与各种代谢通路的内生细菌丰度,显著增加了根部参与代谢通路的内生细菌丰度。因此,3种芽胞杆菌浸种处理可以显著改变水稻根部、茎部和叶部内生细菌群落结构,改善水稻生长的微生态环境。  相似文献

Screening was carried out for erythorbic acid (EA)-producing strains from about 5,000 newly isolated fungi and bacteria. Penicillium notatum FY 115 was screened out as most powerful EA producer. Only Penicillium, but no other genera, was obtained as EA producers from our screening program. Monospore selections and mutagenic treatments succeeded to elevate the yield of EA over 40% to glucose supplied. Various cultural conditions were studied, and pH change during fermentation process was proved to be most important for favorable EA production. Over 80% yield could be obtained when washed mycelium was used in dilute glucose solution.

Abundant accumulation of EA by the strain FY 115, Penicillium sp., in fermentation broth was studied, and EA, both free and Na-salt, was obtained as crystal in the yield of about 45% to glucose supplied, in the media of 8% glucose by jar fermentor, in considering the inhibitory effect of some metal ion.

Extraction processes were improved to elevate the yield and was developed the continuous multi-bed extraction system of anion-exchange resin, which resulted in the yield of 90.9% of EA from fermentation broth in sum total.  相似文献

微生物发酵过程是细胞新陈代谢进行物质转化的过程,为了提高目标产物的转化率,需要对微生物发酵动态特性进行实时分析,以便实时优化发酵过程。拉曼光谱(Raman spectroscopy)量化测试作为一种有应用前景的在线过程分析技术,可以在避免微生物污染的条件下,实现精准监测,进而用于优化控制微生物发酵过程。【目的】以运动发酵单胞菌(Zymomonas mobilis)为例,建立微生物发酵过程中葡萄糖、木糖、乙醇和乳酸浓度拉曼光谱预测模型,并进行准确性验证。【方法】采用浸入式在线拉曼探头,收集运动发酵单胞菌发酵过程中多个组分的拉曼光谱,采用偏最小二乘法对光谱信号进行预处理和多元数据分析,结合离线色谱分析数据,对拉曼光谱进行建模分析和浓度预测。【结果】针对运动发酵单胞菌,首先实现拉曼分析仪对单一产品乙醇发酵过程的精准检测,其次基于多元变量分析,建立葡萄糖、乙醇和乳酸浓度变化的预测模型,实现对发酵过程中各成分浓度变化的准确有效分析。【结论】成功建立了一种评价资源微生物尤其是工业菌株发酵液多种组分的拉曼光谱分析方法。该方法为运动发酵单胞菌等工业菌株利用多组分底物工业化生产不同产物的实时检测,以及其他微生物尤其工业菌株的选育和过程优化提供了新方法。  相似文献

Yarrowia lipolytica is able to secrete large amounts of citric acid (CA), which is greatly affected by the dissolved oxygen concentration (DOC) in the fermentation medium. In this study, oleic acid was selected as oxygen‐vector to improve DOC during CA fermentation. When 2% (v/v) of oleic acid was added to the culture broth, higher DOC (>42.1%) was determined throughout the CA synthesis phase. The yield of CA reached a maximum of 32.1 g/L (25.4% higher than the control) and the biomass was 8.8 g/L. The substrate uptake rate, products formation rate and key enzyme activities were also determined, and the results indicated that CA synthesis was strengthened with oleic acid addition. Furthermore, it was detected that oleic acid could be assimilated by the cells, which means that oleic acid could be served both as oxygen‐vector and co‐substrate for CA synthesis by Y. lipolytica. In a bioreactor with working volume of 3 L, the highest concentration of CA reached to 36. 4 g/L in the presence of 2% (v/v) oleic acid after 192 h of fermentation. These results confirmed that oleic acid could be applied in the large‐scale production of CA by Y. lipolytica.  相似文献
