为探究岩溶植物的光合生理适应机制,采用Li-6400XT便携式光合作用测量系统,对广西平果市岩溶区8种适生植物的叶片净光合速率(Pn)、气孔导度(Gs)、胞间CO2浓度(Ci)、蒸腾速率(Tr)、水分利用效率(WUE)和气孔限制值(Ls)等光合特征参数进行了测定分析。结果表明:(1)6个光合特征参数在种内和种间均存在不同程度的变异,并且种内变异均大于种间变异。(2)Gs和Tr的变化主要来源于种间变异(46.72%~49.76%),而Pn、Ci、WUE和Ls变化主要来源于种内变异(48.66%~64.50%)。在生活型水平上,Pn、Gs和Tr的种内变异表现为常绿植物小于落叶植物,而Ci、WUE和Ls则相反。(3)各参数的种间变异均表现为落叶植...  相似文献

Ellenberg’s indicator values have been suggested as useful method of estimating site conditions using plants. We examined whether Ellenberg’s R values are suitable for indicating soil reaction and if calibration to physical pH measurements can improve bioindication in oligotrophic and mesotrophic submontane broad-leaved forests in Slovakia. Vegetation relevés and pH-H2O and pH-CaCl2 soil reaction were recorded for this purpose. Ellenberg’s R values (R e) were compared to Jurko’s indicator values (R j) and a set of species R values and tolerances (T), which were calibrated with physical pH data using the weighted averaging (R w, T w) and Huisman-Olff-Fresco modelling (R h, T h). Original R e values were then recalibrated with measured pH data to establish new, adjusted set of scores (R c, T c) at Ellenberg’s scale. The Re values are significantly correlated with the other R values, and they demonstrate similar frequency distribution to R j and R w values for the studied species pool. The frequency distribution becomes similar across all the R values when indifferent species were excluded. The performance of all the indicator values in terms of bioindication was tested. Relevé means of the R values were regressed on the field pH measurements. The performance of bioindication varied from 36% to 49% of the explained variance for pH-CaCl2, with the R e and R c values yielding 46% and 49% respectively. The bioindication slightly improved for all calibrated methods (R w, R h and R c) when species were weighted inversely with their tolerances — the performance varied from 42% to 51%, and the R c values performed most effectively. We concluded that Ellenberg’s R values represent a powerful system for bioindicating soil acidity when compared to the other alternatives, with pH-CaCl2 showing better results than pH-H2O. Recalibration of Ellenberg’s values to the measured data improved the indicator system.  相似文献

以沙质草地优势物种猪毛蒿、胡枝子和糙隐子草为研究对象,利用开顶式生长室(OTC)模拟增温,研究降水减少20%、40%和60%与增温的交互作用对3种典型植物叶片光合气体交换特征及叶绿素荧光特征的影响,以揭示沙质草地3种优势植物对气候变化的响应规律。结果显示:(1)与自然温度相比,OTC模拟增温增加了猪毛蒿C_(i),显著降低了胡枝子G_(s)、P_(n)和T_(r)、糙隐子草G_(s)和P_(n)、猪毛蒿WUE和L_(s),也显著降低了猪毛蒿和胡枝子F_(v)/F_(m)和F_(v)/F_(o)。(2)无论增温与否,随着降水减少幅度的增加,猪毛蒿G_(s)和P_(n)呈下降趋势,且中度以上的干旱胁迫下(降水减少>40%)胡枝子和糙隐子草P_(n)显著低于对照。(3)在自然温度条件下,轻度干旱胁迫时(降水减少20%)猪毛蒿T_(r)显著低于对照,重度干旱胁迫时(降水减少60%)其WUE、F_(v)/F_(m)和F_(v)/F_(o)显著低于对照;重度干旱胁迫时,胡枝子C_(i)显著高于对照,差异幅度达10.7%,L_(s)显著低于对照,轻度干旱胁迫时(降水减少20%)其F_(v)/F_(m)和F_(v)/F_(o)显著高于中度以上的干旱胁迫;中度以上的干旱胁迫下糙隐子草T_(r)和G_(s)显著低于对照,重度干旱胁迫时,其C_(i)、F_(v)/F_(m)和F_(v)/F_(o)显著低于对照,WUE和L_(s)显著高于对照。(4)增温与降水减少交互作用下,所有处理猪毛蒿C_(i)均高于对照,差异幅度分别达4.5%,6.0%和8.4%;胡枝子T_(r)均显著低于对照,差异幅度达57.8%;重度干旱胁迫时猪毛蒿L_(s)和WUE显著低于对照,糙隐子草F_(v)/F_(m)和F_(v)/F_(o)随降水减少而降低,中度以上的干旱胁迫时其值显著低于对照。(5)相关性分析表明,3个优势物种的P_(n)与F_(v)/F_(m)和F_(v)/F_(o)均呈显著正相关关系,其中猪毛蒿和糙隐子草的P_(n)—F_(v)/F_(m)和P_(n)—F_(v)/F_(o)斜率明显高于胡枝子。研究表明,气候变暖会在一定程度上加剧降水减少对沙质草地3种群落优势物种光合作用的抑制。  相似文献

Hybrids were produced with relative ease from controlled crosses of Elymus canadensis L. with European Agropyron caninum (L.) Beauv., North American A. trachycaulum (Link) Malte ex H. F. Lewis, and Asian A. striatum Nees ex Steud. All hybrids appeared to be completely sterile and were, for the most part, morphologically intermediate between their parents. The E. canadensis × A. caninum hybrids were exceptionally vigorous and leafy and may have some potential as forage grasses if fertility can be achieved. All parent plants were tetraploid, 2n = 28, and they behaved cytologically as alloploids. Chromosome pairing in the hybrids indicated that both E. canadensis genomes were closely homologous with those of A. trachycaulum and somewhat less homologous with those of A. caninum. Interchanged and inverted chromosome segments apparently constitute the major differences between E. canadensis, A. trachycaulum, and A. caninum genomes; however, cryptic structural differences must also exist. Partial homologies were detected between one A. striatum and E. canadensis genome, but their other genomes were distinctly different. The genome relations between the parent species were expressed in terms of the following genome formulas: E. canadensis, S1S1X1X1; A. trachycaulum, S2S2X2X2; A. caninum, S3S3X3X3 : and A. striatum S4S4YY or X4X4YY, where “S” refers to a genome derived from A. spicatum and “X” and “Y” are genomes of unknown origin.  相似文献

We quantified the effect of stand age and tree species composition on canopy transpiration (EC) by analysing transpiration per unit leaf area (EL) and canopy stomatal conductance (GS) for boreal trees comprising a five stand wildfire chronosequence. A total of 196 sap flux sensors were used on 90 trees consisting of Betula papyrifera Marsh (paper birch; present in the youngest stand), Populus tremuloides Michx (quaking aspen), Pinus banksiana Lamb. (jack pine), and Picea mariana (Mill.) (black spruce). While fine roots were positively correlated with stand EC; leaf area index, basal area, and sapwood area were not. Stands less than 70 years old were dominated by Populus tremuloides and Pinus banksiana and stands greater than 70 years old were composed almost entirely of Picea mariana. As Populus tremuloides and Pinus banksiana increased in size and age, they displayed an increasing sapwood to leaf area ratio (AS : AL), a constant minimum leaf water potential (ΨL), and a constant proportionality between GS at low vapour pressure deficit (Dj GSref) and the sensitivity of GS to D (–δ). In contrast, AS : AL, minimum ΨL, and the proportionally between –δ and GSref decreased with height and age in Picea mariana. A GS model that included the effects of D, AS : AL, tree height, and for Picea mariana an increasing soil to leaf water potential gradient with stand age, was able to capture the effects of contrasting hydraulic properties of Picea mariana, Populus tremuloides and Pinus banksiana during stand development after wildfire.  相似文献

Three chlorophyll c-type pigments were separated by reversed-phase high Performance liquid chromatography and thin-layer chromatography from pigment extracts of the prymnesiophyte, Prymnesium parvum Carter. Based on spectral characteristics, retention times, and comparison with reference pigments isolated from the diatom Phaeodactylum tricornutum Bohlin, two of these pigments were identijied as chlorophyll c1 and c2. The other pigment was identified by its absorption spectrum and thin-layer chromatography retention times as the newly described chlorophyll c3. However, in other prymnesiophytes so far examined, chlorophyll c1 and chlorophyll c3 were present with no chlorophyll cl. The discovery of chlorophyll c3 with chlorophyll c1 and chlorophyll c3 in Prymnesium parvum therefore represents the first report of this combination of pigments in prymnesiophytes.  相似文献

Partitioning of Respiration in an Intensively Managed Grassland   总被引:1,自引:0,他引:1  
Total (RTOT) and heterotrophic (RH) respiration were measured in an intensively managed perennial ryegrass (Lolium perenne L.) grassland. The overall aim of the study was to partition RTOT into RH and autotrophic respiration (RA). This was achieved as follows: (1) analyse the effect of air temperature, soil moisture content and leaf area index on RTOT and the influence of soil temperature and soil moisture content on RH; (2) combine these effects into separate empirical models for RTOT and RH and; (3) use these models to determine temporal trends in RTOT and RH and to assess the relative contribution of RH and RA to RTOT. CO2 fluxes were measured using a vented and thermostatically controlled perspex chamber in conjunction with a portable infrared gas analyser. RTOT was measured in plots with grass and RH in plots with bare soil. RTOT was related to air temperature and RH to soil temperature using exponential relationships. Both RTOT and RH were related to soil moisture content using lognormal relationships. RTOT was related to leaf area index using a linear relationship. These relationships were combined to produce statistical response functions that explained 87% and 84% of the variation in RTOT and RH, respectively. These relationships were combined with meteorological and leaf area index data to reconstruct daily and seasonal fluxes. RTOT values in wintertime were ~4 g C m−2 day−1 increasing to ~10 g C m−2 day−1 in summertime when temperatures and leaf area index were higher and soils were drier. RH has a similar seasonal trend to RTOT but was consistently lower. Wintertime values were ~2 g C m−2 day−1 and increased to ~5 g C m−2 day−1 in summertime. Before day of year 143, and after day of year 259 RH and RA represented 62% and 38% of RTOT, respectively. In the period between these days RH and RA both accounted for 50% of RTOT. In total during 2004 RTOT, RH and RA were 2.34, 1.31 and 1.03 kg C m−2, respectively.  相似文献

Summary Antibodies of two specificities in alloimmune sheep anti-L sera, anti-L P and anti-L l , were separated by a new technique and characterized. Absorption of anti-L serum with trypsinized LK (LL) sheep red cells left anti-L P antibodies; the absorbed anti-L l antibodies were then eluted. Anti-L P was only weakly lytic in the presence of complement; it had no effect on passive K influx, but stimulated active K influx. The stimulation could be reversed by eluting the antibody in glycine buffer at low pH. Stimulatory activity in the eluted cells could be restored by resensitization with anti-L P . Anti-L l was more strongly lytic than anti-L P in the presence of complement; it had no effect on active K influx, but inhibited passive K influx. Pig anti-ruminant IgG conjugated to hemocyanin was used to visualize by electron microscopy the number of L P and L l antigen sites onLL sheep red cells sensitized with anti-L P and anti-L l . The values obtained were 590 L P sites/cell and 847 L l sites/cell.  相似文献

基于全球647套通量数据,定量分析了全球尺度下生态系统光合作用和呼吸作用的温度敏感性(Q10)随纬度、气候和植被的分布规律。结果表明:在全球尺度下,光合作用和呼吸过程的温度敏感性(Q10,G和Q10,R)都随纬度的升高而增加,其中Q10,G和Q10,R的均值分别为3.99±0.21和2.28±0.074。除热带多树草原、常绿落叶林外,Q10,G均大于Q10,R值。不同植被类型的温度敏感性存在显著性差异,表现为:针叶林阔叶林;落叶林常绿林,其中生态系统的季节性变异是造成差异的主要原因。当植被类型和纬度区域共同影响Q10值时,植被类型对Q10值的总变异贡献更大。气候类型对Q10,G和Q10,R都有显著影响。在气候带上,干旱带的Q10,G最小,而冷温带的Q10,G最高。不同气候类型下(除温带草原气候外)的Q10,G都大于Q10,R。在极端条件下,温度可能不在是主导因素,而水分对温度敏感性的影响不可忽略,今后的研究需要更多的关注生态系统温度敏感性对水分变化的响应。  相似文献

Genetics of tolerance to iron chlorosis was investigated in eight crosses involving parents distinctly different in their level of tolerance. The segregating populations with parents and F1s were screened under actual stress conditions in the field. Also, selected crosses were studied for Fe3+ uptake capacity. Tolerance/moderate tolerance to Fe chlorosis was dominant over susceptibility and it was controlled by two sets of nonallelic genes with complementary interaction. Gene Ic 1 has been found to be basic and in complementation with Ic 3 it confers tolerance. Likewise, Ic 2 with Ic 4 confers tolerance. The basic genes Ic 1 and Ic 2 are nonallelic and, in the absence of their respective complementary genes Ic 3 and 4 , ineffective, which results in susceptibility. Of tolerant cultivars, ARC 10372 and Cauvery have been tentatively assigned the genotype of Ic 1 , Ic 2 , Ic 3 , Ic 4 , and moderately tolerant IET 7613, Prasanna and Akashi Ic 1 , 2 Ic 3 Ic 4 . The susceptible ARC 5723 has been assigned Ic 1 , 2 , Ic 3 , Ic 4 , and IET 9829, Ic 1 , 2 Ic 3 Ic 4 . IET 7614 is susceptible, due to the presence of inhibitory genes I-Ic 1 , I-Ic 2 together with ic 1 pt>, ic 2 , Ic 3 , Ic 4 . Further, the gene Pc for purple coleoptile shows linkage with one of the complementary genes with a crossover value of 15.26%, while the gene(s) for seedling height Ts with Ic 1 with a crossover value of 1.7%. It is possible that the gene(s) for iron chlorosis tolerance might belong to the second linkage group, where genes for purple leaf were located.  相似文献

We investigate the utility of an improved isotopic method to partition the net ecosystem exchange of CO2 (F) into net photosynthesis (FA) and nonfoliar respiration (FR). Measurements of F and the carbon isotopic content in air at a high‐elevation coniferous forest (the Niwot Ridge AmeriFlux site) were used to partition F into FA and FR. Isotopically partitioned fluxes were then compared with an independent flux partitioning method that estimated gross photosynthesis (GEE) and total ecosystem respiration (TER) based on statistical regressions of night‐time F and air temperature. We compared the estimates of FA and FR with expected canopy physiological relationships with light (photosynthetically active radiation) and air temperature. Estimates of FA and GEE were dependent on light as expected, and TER, but not FR, exhibited the expected dependence on temperature. Estimates of the isotopic disequilibrium D , or the difference between the isotopic signatures of net photosynthesis (δA, mean value ?24.6‰) and ecosystem respiration (δR, mean value ?25.1‰) were generally positive (δAR). The sign of D observed here is inconsistent with many other studies. The key parameters of the improved isotopic flux partitioning method presented here are ecosystem scale mesophyll conductance (gm) and maximal vegetative stomatal conductance (gcmax). The sensitivity analyses of FA, FR, and D to gcmax indicated a critical value of gcmax (0.15 mol m?2 s?1) above which estimates of FA and FR became larger in magnitude relative to GEE and TER. The value of D decreased with increasing values of gm and gcmax, but was still positive across all values of gm and gcmax. We conclude that the characterization of canopy‐scale mesophyll and stomatal conductances are important for further progress with the isotope partitioning method, and to confirm our anomalous isotopic disequilibrium findings.  相似文献

Summary Touch smears of the cerebellum and cerebrum of ageing rats were fixed with methanol, hydrolyzed with 2N HCl at various temperatures and for various periods, and stained with pararosaniline-Schiff reagent. The hydrolysis curves were determined by fluorescence cytophotometry and were computer fitted to the Bateman function to determine the kinetic parameters, the initial yield of apurinic acid or single-stranded DNA (y 0), and the rate constants for depurination or denaturation (k 1) and depolymerization (k 2). The values for k 1 (1/k 1 is correlated with the degree of chromatin condensation) and k 2 (which reflects the degree of DNA instability) steadily increased with age. The values for y 0, which may indicate the degree of DNA denaturation or damage present before acid hydrolysis, also increased with age in both the cerebellum and cerebrum; however, this value was lower in the cerebellum untill 15 weeks, with the situation being reversed after 35 weeks, the cross-over time being at about 25 weeks. The values of lnk 1 and lnk 2 were plotted as the function of the reciprocal of the absolute temperature (T) (Arrhenius plot) for both the cerebellum and cerebrum of 15- and 74-week-old rats, and the activation energies (E) for depurination and depolymerization were calculated from the slopes. In particular, the values of E for k 2 decreased much more quickly with age and were smaller in cerebellum. In conclusion, the degree of DNA damage and DNA instability steadily increases in both the cerebellum and cerebrum of ageing rats, and this process is much faster in the cerebellum.In honour of Prof. P. van Duijn  相似文献

以浙江天童国家森林公园常绿阔叶林为研究对象,采用空间代替时间的方法,研究了5个不同演替阶段常见的4种乔木以及4种灌木叶片的光饱和速率(Pmax)、光合氮素利用效率(PNUE)及其与叶片氮含量(NL)、叶片氮素在细胞壁的分配比例(细胞壁N/叶片总N,NCW/NL)、氮素在光合酶中的分配比例(NR/NL)、单位面积叶干重(LMA)的相互关系。结果表明:(1)演替系列4种乔木和4种灌木各种间指标除NL外均表现出显著差异,前期种较后期种具有更高的NR/NL、PNUE、Pmax,而后期种LMA、NCW/NL、MCW/ML(细胞壁干重/叶片总干重)更大,NL在乔木各种间差异不明显,在灌木种间则差异显著;乔木种较灌木种具有更大的LMA、NCW/NL、MCW/ML,而NR/NL则较灌木小;8种植物的Pmax与NL以杨梅为最高,连蕊茶最低;苦槠具有最高的PNUE,而栲树最低。(2)随着演替的进行,前期种的NR/NL、PNUE、Pmax有减小趋势,而LMA、NCW/NL、MCW/ML逐渐增大,后期种则表现出相反的趋势。(3)NR/NL与Pmax、PNUE之间呈显著正相关关系,而LMA、NCW/NL、MCW/ML则与Pmax、PNUE、NR/NL显著负相关。研究认为,NR/NL与NCW/NL之间的负相关性及其对PNUE的影响可以在一定程度上解释树木在光合与维持两方面的权衡关系以及演替的生理机制。  相似文献

Cross-compatibility relationships in almond are controlled by a gametophytically expressed incompatibility system partly mediated by stylar RNases, of which 29 have been reported. To resolve possible synonyms and to provide data for phylogenetic analysis, 21 almond S-RNase alleles were cloned and sequenced from SP (signal peptide region) or C1 (first conserved region) to C5, except for the S 29 allele, which could be cloned only from SP to C1. Nineteen sequences (S 4 , S 6 , S 11 S 22 , S 25 S 29 ) were potentially new whereas S 10 and S 24 had previously been published but with different labels. The sequences for S 16 and S 17 were identical to that for S 1 , published previously; likewise, S 15 was identical to S 5 . In addition, S 4 and S 20 were identical, as were S 13 and S 19 . A revised version of the standard table of almond incompatibility genotypes is presented. Several alleles had AT or GA tandem repeats in their introns. Sequences of the 23 distinct newly cloned or already published alleles were aligned. Sliding windows analysis of Ka/Ks identified regions where positive selection may operate; in contrast to the Maloideae, most of the region from the beginning of C3 to the beginning of RC4 appeared not to be under positive selection. Phylogenetic analysis indicated four pairs of alleles had ‘bootstrap’ support > 80%: S 5 /S 10 , S 4 /S 8, S 11 /S 24 , and S 3 /S 6 . Various motifs up to 19 residues long occurred in at least two alleles, and their distributions were consistent with intragenic recombination, as were separate phylogenetic analyses of the 5′ and 3′ sections. The localization, distribution and orientation of O2 chemoreceptors associated with the control of cardio-respiratory responses were investigated in the neotropical, Hoplias lacerdae. Selective denervation of the cranial nerves (IX and X) was combined with chemical stimulation (NaCN) to characterize the gill O2 chemoreceptors, and the fish were then exposed to gradual hypoxia to examine the extent of each cardio-respiratory response. Changes in heart rate (fH) and ventilation amplitude (Vamp) were allied with chemoreceptors distributed on both internal and external surfaces of all gill arches, while ventilation rate (fR) was allied to the O2 chemoreceptors located only in the internal surface of the first gill arch. H. lacerdae exposed to gradual hypoxia produced a marked bradycardia (45%) and 50% increase in Vamp, but only a relatively small change in fR (32%). Thus, the low fR response yet high Vamp were in accord with the characterization of the O2 chemoreceptors. Comparing these results from H. lacerdae with hypoxia-tolerant species revealed a relationship existent between general oxygenation of the individual species environment, its cardio-respiratory response to hypoxia and the characterization of O2 chemoreceptors.  相似文献

Nuclear magnetic resonance T 1 and T 2 relaxivities (r 1 and r 2) exhibit efficiency of a material to alter the relaxation rates (1/T 1 and 1/T 2), and they are being used for diagnostic purposes. The determination of total relaxivities (r 1t and r 2t) of cystic fluid content and ameloblastoma may therefore be useful for discriminative purposes. In order to determine what makes total relaxivities of hemorrhagic cysts, four sets of tubes containing pooled cyst were doped with increasing concentrations of iron, copper, albumin, and γ-globulins. These sets were replaced in a phantom together with six individual cysts and one ameloblastoma. The relaxation times were measured by magnetic resonance imaging operating at 1.5 T. The relaxivities of individual ions and proteins were determined from the slope of the relation between relaxation rates and concentration, while total relaxivities were determined by using the increases in relaxation rates and material content of cystic fluid (MC). Iron, copper, albumin, and γ-globulins were found to be the sources of r 1t and r 2t. Each of r 1t, r 2t, r 1tMC, r 2tMC, and r 2t/r 1t are distinctive parameters for each cystic category and ameloblastoma. Except for MC, the parameters measured for ameloblastoma are significantly smaller than those of cysts. The similarity of the present results to those used in clinical applications suggests that each of r 1t, r 2t, r 1tMC, r 2tMC, and r 2t/r 1t has an ability to discriminate various fluids and masses. The present work also suggests that r 1tMC, r 2tMC, and r 2t/r 1t can be determined in vivo.  相似文献

The present investigation concerns with the development and optimization of an in situ forming formulation using 33 full factorial design experimentation. Metformin, an antidiabetic drug with upper part of gastrointestinal tract as absorption window was used as a model drug. The formulations were designed with an objective to retain in stomach for an extended time period. The effect of three independent factors—concentrations of sodium alginate (X 1), gellan gum (X 2), and metformin (X 3) on in vitro drug release were used to characterize and optimize the formulation. Five dependent variables—release exponent (Y 1), dissolution efficiency (Y 2), drug release at 30 min (Y 3), 210 min (Y 4), and 480 min (Y 5) were considered as optimization factors. The data were statistically analyzed using ANOVA, and a p < 0.05 was considered statistically significant. Three dimensional surface response plots were drawn to evaluate the interaction of independent variables on the chosen dependent variables. Of the prepared 27 formulations, the responses exhibited by batch F17 containing medium level sodium alginate (X 1), low level gellan (X 2), and medium level metformin (X 3) were similar to the predicted responses.  相似文献

The cytological possibility of gene transfer from Sinapis pubescens to Brassica napus was investigated. Intergeneric hybrids between Brassica napus (2n = 38) and Sinapis pubescens (2n = 18) were produced through ovary culture. The F1 hybrids were dihaploid and the chromosome configurations were (0–1) III + (2–11) II + (5–24) I . One F2 plant with 38 chromosomes was obtained from open pollination of the F1 hybrid. Thirty-one seeds were obtained from the backcross of the F2 plant with B. napus. Five out of seven plants had 38 chromosomes, and the pollen stainability ranged from 0% to 81.4%. In the B2 plants obtained from the backcross of B1 plants with B. napus, 66.7% of the plants examined had 38 chromosomes. S. pubescens may become a gene source for the improvement of B. napus.  相似文献

Abstract Acetylcholinesterase (AChE) in the susceptible (S) and the resistant (R) strains of housefly (Musca domestica) was investigated using kinetic analysis. The Vmax values of AChE for hydrolyzing acetylthiocholine (ATCh) and butyrylthiocholine (BTCh) were 4578.50 and 1716.08nmol/min/mg* protein in the R strain, and were 1884.75 and 864.72 nmol/min/mg. protein in the Sstrain, respectively. The Vmax ratios of R to S enzyme were 2.43 for ATCh and 1.98 for BTCh. The Km values of AChE for ATCh and BTCh were 0.069 and 0.034 mmol/L in the S strain, and 0.156, 0.059 mmol/L in the R strain, respectively. The Km ratios of R to S enzyme were 2.26 for ATCh and 1.74 for BTCh. The ki ratios of S to R enzyme for three insecticides propoxur, methomyl and paraoxon were 46.04, 4.17 and 2. 86, respectively. In addition, kcat and kcat/Km for measuring turnover and catalytic efficiency of AChE were determined using eserine as titrant. The kcat values of AChE from the R strain for both ATCh and BTCh were higher than those values from the S strain. But the values of kcat/Km were in contrary to the kcat values with R enzyme compared to S enzyme. The AChE catalytic properties and sensitivity to the inhibition by three insecticides in the R and S strains of housefly were discussed based on contribution of Vmax, Km, ki, kcat and kcat/Km. All these data implied that AChE from the R strain might be qualitatively altered. We also observed an intriguing phenomenon that inhibitors could enhance the activity of AChE from the resistant strain. This “flight reaction” of the powerful enzyme might be correlated with the developing resistance of housefly to organophosphate or carbamate insecticides.  相似文献
