Nine hours after rats fed ad libitum for 14 days a 10% caein diet (10C), a 10% casein diet containing 7% glycine (10C7G) and a 10% casein diet containing 7% glycine with 1.4% l-arginine HCI and 0.9% l-methionine (10C7GArgMet) were force-fed 10 ml of each diet suspension containing 5 μCi of glycine-U-14C per 100 g of body weight, the radioactivity recoveries of 14C in expired CO2, tissue components and urine were determined.

The radioactivity recovery of 14C in the expired C02 of the 10C7G group was generally higher than that of the 10C7GArgMet group, and those of both groups would have been much higher than that of the 10C group unless the isotope had been diluted. The amount of expiratory 14C of rats fed a 25 % casein diet containing 7% glycine was not different from that of the 10C7G group. The recovery of 14C in the trichloroacetic acid (TCA) soluble fraction of muscle of the 10C7G and the 10C7GArgMet groups were greater than that of the 10C group, but there was no difference between the 10C7G and the 10C7GArgMet groups. The recoveries of 14C in the TCA soluble fraction and protein of plasma and liver, and the muscle protein were negligible in all the groups. The amount of glycine-14C incorporated into the carcass lipids of the 10C7GArgMet group was larger than that of other groups. Those in the carcass lipids of the 10C7G and the 10C7GArgMet groups would have been much higher than that of the 10C group unless the dilution of the isotope had taken place. The recoveries of 14C in the liver and muscle glycogen, and liver lipids were remarkably small in all the groups. From the above results, it was suggested that the degradation of glycine-14C to expiratory CO2 was not accelerated, but the rate of incorporation of the isotope into carcass lipids was increased by the supplementation of l-arginine and l-methionine to the 10C7G diet as compared with that of rats fed the 10C7G diet.  相似文献

Cytological effects of glycine on Bacillus subtilis var. amyloliquefaciens were compared between the cells of the glycine-sensitive parent and resistant mutant. Glycine induced disruption of the protoplasts which had been prepared by treating the glycine-sensitive cells with lysozyme. This effect of glycine was almost completely prevented by preincubating the protoplasts with spermine. The protoplasts prepared from the resistant cells were markedly stable in the presence of glycine. In this mutant, neither cell lysis nor cessation of the enzyme production by glycine occurred, contrary to the results obtained with the glycine-sensitive parent. Between both type of cells little difference could be observed in the metabolic activity for glycine, but free amino acid content was higher in the glycine-resistant cells than in the parent ones.  相似文献

Summary Membrane units from lysed spheroplasts induced by lysozyme or glycine from Mycobacterium spec. smegmatis were isolated in a biological active state by differential centrifugation and by density gradient technique. They were compared morphologically with membraneous fractions obtained from mycobacterial cells disintegrated under a high hydrostatic pressure.Higher homogeneity of membraneous structures isolated from spheroplasts was confirmed. Three types of membraneous structures could be distinguished. They include empty ghosts of spheroplasts, tubular structures containing cytoplasmic material and fragments of typical membraneous structures relatively free of contaminants. By studying protoplasts in the process of lysis it was determined that these structures correspond with cytoplasmic membranes and mesosomes.Differences between lysozome and glycine induced spheroplasts as a starting material for isolation of membraneous structures include the proportion of contamination by other cellular components, reasons of which are discussed.  相似文献

Fourteen new and three known o-arylethenylbenzoic acids (10 ~ 18), required for testing as potential plant growth regulators, have been synthesized by the Wittig reaction. The cis- and trans-isomers were separated by fractional crystallization and identified by NMR spectroscopy. The preliminary assay results on cress seedlings are reported and show that both geometric isomers appear to have nearly identical anit-geotropic activity in this test system.  相似文献

Some strict anaerobic bacteria catalyze with glycine as substrate an internal Stickland reaction by which glycine serves as electron donor being oxidized by glycine-cleavage system or as electron acceptor being reduced by glycine reductase. In both cases, energy is conserved by substrate level phosphorylation. Except for the different substrate-activating proteins P B , reduction of sarcosine or betaine to acetyl phosphate involves inEubacterium acidaminophilum the same set of proteins as observed for glycine, e.g. a unique thioredoxin system as electron donor and an acetyl phosphate-forming protein P c interacting with the intermediarily formed Secarboxymethylselenoether bound to protein P A .  相似文献

Clostridium histolyticum grew on glycine, arginine, or threonine as sole substrate. Arginine degradation preceded that of glycine and partially inhibited that of threonine when two amino acids were present. Each amino acid seemed to be individually catabolized, not by a Stickland type of reaction. Glycine fermentation required the presence of complex ingredients. Therefore, an effect of selenite on glycine catabolism could only be demonstrated after scavenging selenium contamination by preculturing Peptostreptococcus glycinophilus in that medium. C. acidiurici was not suited as selenium accumulating organism as C. histolyticum was inhibited by the residual uric acid. Arginine catabolism was unaffected by seleniuum depriviation. The labelling pattern obtained in acetate after incubation of C. histolyticum with [1-14C]- or [2-14C]glycine strongly indicated the metabolism of glycine via the glycine reductase pathway.  相似文献

The aim of the study was to determine the relative bioavailability of zinc gluconate stabilized with glycine in a Petit Suisse cheese from an infant dessert. Weight gain and bone zinc content were the nutritional responses evaluated for the diets of different zinc content: 2 ppm (basal) and 5, 10, and 30 ppm from zinc gluconate stabilized with glycine and zinc sulfate. Nonlinear regression analysis of the fitted curves for weight gain determined a relative zinc bioavailability of 100% for the Y max ratio and 96% for Y max/t 1/2 ratio for zinc gluconate stabilized with glycine (R 2=0.7996 for zinc sulfate and 0.8665 for zinc gluconate stabilized with glycine). The slope ratio analysis from linear regression of femur zinc determined a relative zinc bioavailability of 93% for zinc gluconate stabilized with glycine (R 2=0.8693 for zinc sulfate and 0.8307 for zinc gluconate stabilized with glycine). Zinc gluconate stabilized with glycine has similar bioavailability as zinc sulfate in a Petit Suisse cheese nutritional matrix, with the advantage that the stabilized compound does not modify the sensorial characteristics of the fortified cheese.  相似文献

A simple electronic device was constructed which, in combination with a conventional titrator, records close approximation of the buffer capacity curve (β-pH curve) for the solution of unknown composition. Since the recorded curves provide the overall picture of the distribution of weak Brönsted acids in the solution on the pKa axis, this apparatus may be useful in various fields such as chemistry of food and agricultural products and clinical medicine, where the characterization of the complex mixtures of weak electrolytes — carboxylic acids, amono acids, proteins, amines, phenols and etc. — are important.  相似文献

Anaerobic, Gram-positive cocci were obtained from chicken feces by direct isolation, which grew on the purines uric acid, xanthine, 6,8-dihydroxypurine, guanine, and hypoxanthine. Adenine and glycine were fermented, but not as readily. Acetate, formate, ammonia, and CO2 were products. The isolated strains were nutritionally non-fastidious, however, they required selenite, molybdate, and tungstate as micronutrients. The cells were spherical and 0.5–0.9 m in diameter. The addition of bile salts enhanced the growth rate in most cases. The organisms proved to be quite resistant to lysis. The guanosine-plus-cytosine (G+C) content of their deoxyribonucleic acid was 33.6 to 34.8 mol%. The peptidoglycan was of the same structure (Gly-Lys-d-Asp) as reported for the anaerobic cocci of Hare group IX. However, the latter strains could only utilize glycine, not purines. Summary A glycine cleavage enzyme system, inducible by glycine, has been demonstrated in Salmonella typhimurium. The induced enzyme levels, however, are only about 20% of the induced levels found in Escherichia coli. Starting with a serine auxotroph, mutants were isolated that grow with a serine supplement, but not with a glycine supplement. Three independently isolated mutants have reduced or nondetectable glycine cleavage enzyme levels. Biosynthesis of glycine betaine from simple carbon sources as compatible solute is rare among aerobic heterotrophic eubacteria, and appears to be almost exclusive to the non-halophilic and slightly halophilic phototrophic cyanobacteria. Although Synechococcus sp. WH8102 (CCMP2370), a unicellular marine cyanobacterium, could grow up to additional 2.5% (w/v) NaCl in SN medium, natural abundance 13C nuclear magnetic resonance spectroscopy identified glycine betaine as its major compatible solute. Intracellular glycine betaine concentrations were dependent on the osmolarity of the growth medium over the range up to additional 2% NaCl in SN medium, increasing from 6.8 ± 1.5 to 62.3 ± 5.5 mg/g dw. The ORFs SYNW1914 and SYNW1913 from Synechococcus sp. WH8102 were found as the homologous genes coding for glycine sarcosine N-methyltransferase and sarcosine dimethylglycine N-methyltransferase, heterologously over-expressed respectively as soluble fraction in Escherichia coli BL21(DE3)pLysS and purified by Ni-NTA His•bind resins. Their substrate specificities and the values of the kinetic parameters were determined by TLC and 1H NMR spectroscopy. RT-PCR analysis revealed that the two ORFs were both transcribed in cells of Synechococcus sp. WH8102 growing in SN medium without additional NaCl, which confirmed the pathway of de novo synthesizing betaine from glycine existing in these marine cyanobacteria.  相似文献

Two strains of Rhizobia isolated from nodules of Vicia faba var. major and one strain isolated from nodules of Cicer arietinum L. were characterized for salt resistance. The presence of 1 mM glycine betaine or choline in a minimal medium with added NaCl had a beneficial role on the growth of the three strains. Both molecules were found to be taken up by cells obtained at low osmolarity, and whereas glycine betaine uptake activity was stimulated significantly in cells grown in the presence of 0.15 M NaCl, choline uptake activity was strongly inhibited by salt in all tested strains. However, in cells grown with exogenous choline, the uptake inhibition exerted by salt was relieved, mainly in the strain isolated from nodules of C. arietinum L. On the basis of kinetics determinations, in control cells as well as in salt-stressed cells, only high-affinity activities were observed for glycine betaine and choline (apparent K m s between 3 and 18 μM). Periplasmic proteins that bound glycine betaine or choline were identified. In nondenaturing conditions, these proteins extracted from the various strains showed different electrophoretic mobility with always a less negative entire charge than the analogous proteins from Rhizobium meliloti. Humans are exposed much more often to exogenous Saccharomyces cerevisiae (a baker’s yeast) than exogenous Candida albicans (a highly infectious yeast) but suffer no apparent complications from S. cerevisiae. We hypothesize that variations in characteristics between these two species may be due, in part, to differences in glycine metabolism. In this study, we examined differences in glycine oxidation between C. albicans and S. cerevisiae. Both C. albicans and S. cerevisiae were cultured in glycine enriched media, followed by determination of glycine oxidation and amino acid concentrations in cells. Glycine was degraded to a much greater extent in C. albicans than in S. cerevisiae. Threonine concentrations and glycine oxidation were also elevated in C. albicans. Almost all of the disappearance of glycine from incubation media was accounted for by the formation of serine, threonine, and CO2 in S. cerevisiae, whereas these products represented only 50% of the metabolized glycine in C. albicans. The unidentified metabolites of glycine in C. albicans, presumably purines, could contribute to its infectious capacity and this warrants further study.  相似文献

A growing culture of Clostridium saccharoperbutylacetonicum partially lost its turbidity in the presence of 0.3 to 0.6 m sucrose without any extraneous supplements for cell wall degradation. The maximum effect was shown at 0.35 m of sucrose and the culture lost 40 to 50% of initial turbidity. The rate of lysis depended on the age of culture. The most rapid lysis occurred in the organisms of early exponential growing cultures, but no lysis was observed on those of late exponential and stationary phase cultures. The optimal pH was 5.5 to 6.0, and the optimal temperature 30 to 35°C. The sucrose-induced lysis was inhibited by bivalent cations (such as Ca2+, Mg2+), heavy metal cations (such as Cu2+, Pb2+), enzymic inhibitors (such as PCMB) and fixative agents (such as formalin, glutaraldehyde), while organisms whose growth had been inhibited by antibiotics (such as chloramphenicol, tetracycline) were also resistant to sucrose-induced lysis. The sucrose-induced lysis was accompanied by striking morphological conversion from original rod cells (3.0~6.0}0.4~0.6 μ) to spherical cells (1.0~ 1.2 μ diameter). The sucrose-induced lysis was also observed on the relative strains of C. saccharoperbutylacetonicum and C. sporogenes, but not observed on many other species of Clostridium and aerobic bacteria tested. It was suggested that sucrose-induced lysis was a kind of bacterial autolysis which was induced by sucrose treatment. The bacterial spheres developed during the lysis may be the protoplasts.  相似文献

裂解作为合成生物学中的常见功能模块单元,在基因线路设计方面具有广泛的运用。裂解主要通过诱导表达噬菌体的裂解基因盒子实现,不过,尚未有关于裂解基因的详细表征。本研究首先利用阿拉伯糖和鼠李糖诱导系统在大肠杆菌(Escherichia coli)Top10表达了5种裂解基因盒子(S105, A52G, C51S S76C, LKD, LUZ),通过OD600值的测量,表征了在不同生长阶段、诱导剂浓度、质粒拷贝数等条件下,含有不同裂解基因盒子的大肠杆菌菌株Top10的裂解行为。结果表明,虽然5种裂解基因盒子在大肠杆菌内都可以有效引起宿主的裂解,但是裂解行为在不同条件下有较大差异。进一步地,由于诱导系统在不同宿主之间背景表达水平的差异,导致在铜绿假单胞菌(Pseudomonas aeruginosa) PAO1中难以构建可诱导的裂解系统。经过筛选,通过基因组插入的方式,利用鼠李糖诱导系统对裂解基因盒子在铜绿假单胞菌中引起的裂解行为进行表征。结果显示,LUZ和LKD具有较强的使铜绿假单胞菌裂解的能力,而S105、A52G、C51S S76C对铜绿假单胞菌的裂解能力较差。最后,利用LUZ和光遗传学工...  相似文献

From an acarid mite, Lardoglyphus konoi (Sasa et Asanuma) (Acarina: Acaridae), with its culture medium, an aggregation pheromone was isolated. The structure of the pheromone was determined as 1,3,5,7-tetramethyldecyl formate and identified by 11 steps of organic synthesis. The absolute configuration of C1 was assigned as R. The compound has been assigned the trivial name lardolure. The synthesized diastereomeric mixture of racemates showed one-tenth of the activity of the natural pheromone against the test mite, Carpoglyphus lactis.  相似文献

The extraction of nucleic acids from a given environment marks a crucial and essential starting point in any molecular investigation. Members of Halococcus spp. are known for their rigid cell walls, and are thus difficult to lyse and could potentially be overlooked in an environment. Furthermore, the lack of a suitable lysis method hinders subsequent molecular analysis. The effects of six different DNA extraction methods were tested on Halococcus hamelinensis, Halococcus saccharolyticus and Halobacterium salinarum NRC-1 as well as on an organic rich, highly carbonated sediment from stromatolites spiked with Halococcus hamelinensis. The methods tested were based on physical disruption (boiling and freeze/thawing), chemical lysis (Triton X-100, potassium ethyl xanthogenate (XS) buffer and CTAB) and on enzymatic lysis (lysozyme). Results showed that boiling and freeze/thawing had little effect on the lysis of both Halococcus strains. Methods based on chemical lysis (Triton X-100, XS-buffer, and CTAB) showed the best results, however, Triton X-100 treatment failed to produce visible DNA fragments. Using a combination of bead beating, chemical lysis with lysozyme, and thermal shock, lysis of cells was achieved however DNA was badly sheared. Lysis of cells and DNA extraction of samples from spiked sediment proved to be difficult, with the XS-buffer method indicating the best results. This study provides an evaluation of six commonly used methods of cell lysis and DNA extraction of Halococcus spp., and the suitability of the resulting DNA for molecular analysis.  相似文献
