Xylose-to-xylitol conversion was investigated in a bench-scale bioreactor using Candida guilliermondii cells entrapped within polyvinyl alcohol-hydrogel beads in a system operated in repeated-batch mode with cell recycling. Yeast-viable cells were immobilized in the support using the freezing–thawing method. Bioconversion assays were performed in a stirred tank reactor operated at 400-rpm agitation speed, 30°C temperature, and 1.04-vvm air flow rate. The system was explored during six successive cycles, and a small decrease in the conversion performance in the fifth cycle was observed, but the biocatalytic activity of the microorganism was recovered in the sixth cycle after washing the particles. During the process, the hydrogel beads maintained their shape and size without appreciable deterioration. Xylitol production, yield factor, and volumetric productivity increased with progressive recycling of cells and achieved their maximum values (P F = 39.7 g l−1; Y P/S = 0.77 g g−1; Q P = 0.53 g l−1 h−1, respectively) after the third cell recycling, probably because of cells’ adaptation to the medium.  相似文献

In hot water (50–60°C), polyvinyl alcohol (PVA) molecules have coordination reaction with Au3+ cations, forming an Au3+-PVA polymer complex. In the proposed model reaction in small templates, the complex converts to Au0 capping in PVA molecules. Adding sucrose (5–10 times the PVA in mass) in a typical batch promotes Au3+→Au0 reaction, showing absorption coefficient α in Au0 surface plasmon band to be enhanced as much as 28 times the value in reaction with PVA. The band shifts at 547 nm from 566 nm (α  =  21.4 cm−1 mol−1) in the PVA sample. Drying Au0-PVA/sucrose (2–5 wt% Au0) colloid at 60–70°C and then heating at 450°C in air burns off the organic part, leaving behind a light ash colored powder with Au0 nanoprisms or nanofibrils (∼30 nm average width). X-ray diffractogram has six reflections, (111), (200), (220), (311), (222), and (400), of Fm3m fcc Au0 of lattice parameter a  =  0.4080 nm. The powder has photoluminescence in transversal and longitudinal Au0 plasmon bands of 535 and 585 nm, respectively.  相似文献

Strains producing higher levels of cellulolytic enzymes were selected from among 520 strains of plant pathogenic fungi, Fusarium species, and F. oxysporum strain SUF850 was found to be the best producer. When strain SUF850 was cultured using one of three polysaccharides, Avicel, carboxy- methyl cellulose (CMC) or xylan, as a carbon source, the culture filtrate contained degrading activi- ties toward all three substrates, i.e., irrespective of the carbon source used. From the culture filtrate of Avicel-grown cells, four distinct enzymes were purified to homogeneity, as judged on SDS-PAGE. They were designated as CMCase I, CMCase II, /Miitrophenyl-β-d-cellobiosidase and xylanase, and the characteristics of the individual enzymes were examined and compared.  相似文献

This study aimed to investigate the effects of dry and humid heat curing on the physical and drug release properties of polyvinyl acetate–polyvinyl pyrrolidone matrices. Both conditions resulted in increased tablet hardness; tablets stored under humid conditions showed high plasticity and deformed during hardness testing. Release from the matrices was dependent on the filler's type and level. Release profiles showed significant changes, as a result of exposure to thermal stress, none of the fillers used stabilized matrices against these changes. Density of neat polymeric compacts increased upon exposure to heat; the effect of humid heat was more evident than dry heat. Thermograms of samples cured under dry heat did not show changes, while those of samples stored under high humidity showed significant enlargement of the dehydration endotherm masking the glass transition of polyvinyl acetate. The change of the physical and release properties of matrices could be explained by the hygroscopic nature of polyvinyl pyrrolidone causing water uptake; absorbed water then acts as a plasticizer of polyvinyl acetate promoting plastic flow, deformation, and coalescence of particles, and altering the matrices internal structure. Results suggest that humid heat is more effective as a curing environment than dry heat for polyvinyl acetate–polyvinyl pyrrolidone matrices.  相似文献

Novel acremolactones B and C were isolated from the acremolactone A-producing Acremonium roseum I4267. The structure of acremolactone B having a phenylpyridyl γ-lactone was elucidated by spectroscopic methods. It showed plant growth inhibitory activity toward Chinese cabbage seedlings. The congener of acremolactone C having a phenylcyclopentenone γ-lactone showed weak activity.  相似文献

We describe a procedure for the selection of alcohol dehydrogenase negative mutants in Drosophila. The method consists of exposing eggs and larvae to low concentrations of 1-pentyne-3-ol dissolved in the culture medium. Only those flies with greatly reduced levels of alcohol dehydrogenase activity survive. In addition, genotypically negative flies die if their mothers are alcohol dehydrogenase positive. Using this procedure and formaldehyde to generate mutants, we were able to detect seven alcohol dehydrogenase negative mutants out of 350,000 individuals subjected to selection. At least five of the mutants contain small deletions that include the alcohol dehydrogenase locus.  相似文献

The authors report an experimental study of a perceptual hypothesis of subjects (ages 17 to 25 years) on a drawing task (drawing a still life). It is assumed that perception can be studied as a process of solving a direct-sensory task where the solution to the task requires removing uncertainty from the situation. From subjects’ solution of the drawing task, three classes of hypothesis were determined, and each divided into three types depending on the number of components (a one, two, or multicomponent hypothesis). Most biofilm studies employ single species, yet in nature biofilms exist as mixed cultures, with inevitable effects on growth and development of each species present. To investigate how related species of bacteria interact in biofilms, two Pseudomonas spp., Pseudomonas fluorescens and Pseudomonas putida, were cultured in capillary bioreactors and their growth measured by confocal microscopy and cell counting. When inoculated in pure culture, both bacteria formed healthy biofilms within 72?h with uniform coverage of the surface. However, when the bioreactors were inoculated with both bacteria simultaneously, P. putida was completely dominant after 48?h. Even when the inoculation by P. putida was delayed for 24?h, P. fluorescens was eliminated from the capillary within 48?h. It is proposed that production of the lipopeptide putisolvin by P. putida is the likely reason for the reduction of P. fluorescens. Putisolvin biosynthesis in the dual-species biofilm was confirmed by mass spectrometry.  相似文献

In the present study, factors influencing the synthesis and activity of β-galactosidase of two strains of Propionibacterium acidipropionici with some probiotic properties are described for the first time. The enzyme 6-phospho-β-D-galactosidase of the PEP-PTS system was not detected, suggesting that P. acidipropionici metabolize lactose only by using β-galactosidase. The highest enzymatic activities were obtained from cultures developed in a basal broth medium containing 1.0% sodium lactate or 0.25% lactose. Maximum β-galactosidase activity from cell-free extracts of the strains was obtained at pH 7.0 and 50°C, but a high activity was even detected at 37°C. The enzyme was competitively inhibited by lactose and activated by glucose and sodium lactate. The remaining activities after heating cell-free extracts up to 20 min at 60°C were 70% and 25% of untreated control activities for P. acidipropionici Q4 and CRL 1198, respectively. Cations like Mg2+, Mn2+, Li+, Na+, and K+ acted as stimulators of the β-galactosidase activity whereas Ca2+, Co2+, Ni2+, Hg2+ and Cu2+ showed inhibitory effect in different extent. These results suggest that the environmental conditions commonly present in the human's intestine may be adequate for the synthesis and activity of β-galactosidase from these strains of Propionibacterium. The enzyme resist the cooking temperature of Swiss-type cheeses in different extent depending on the strain tested and most of the cations present in milk stimulate the enzymatic activity. Our results suggest that a cheese would be an appropriate vehicle for delivery of β-galactosidase from propionibacteria to the host and efforts to develop a Swiss-type probiotic cheese for lactose intolerant persons should be done.  相似文献

The endo α-1,4 polygalactosaminidase from Pseudomonas sp. 881 was purified from the culture nitrate by ethanol precipitation and sequential column chromatographies on CM-Sephadex C-25, Sephadex G-50 and Phenyl-Sepharose CL-4B. The purified enzyme was electrophoretically homogeneous and its molecular weight and isoelectric point were 31,000 and 6.7, respectively. The optimum pH and temperature for hydrolysis of polygalactosamine were 5.0 and 55°C, respectively. The enzyme was stable up to 45°C for 15min and from pH 4.0 to 7.6 at 37°C for 1 hr.

The Km value was 0.05% α-1,4 polygalactosamine and the V was 0.154μmol reducing sugar (galactosamine)/min/μg protein. This polygalactosaminidase was inhibited by Sn2+ , Fe2+ , Fe3+ , Hg2+, Cu2+ ions and SDS. The enzyme did not hydrolyze oligo galactosamines (n < tetramer) or N-acetyl-polygalactosamines. It acted only on oligo galactosamine (n > trimer) and polygalactosamine endogeneously so far tested.  相似文献

1. Bacilysin, a peptide which yields l-alanine and l-tyrosine on acid hydrolysis, was produced by a strain of Bacillus subtilis (A 14) in a chemically defined medium containing glucose, ammonium acetate or ammonium chloride, potassium phosphate and other inorganic salts, and ferric citrate. 2. Under the conditions used growth was diphasic. Bacilysin was formed during the second phase of slower growth, and there was little production during the stationary phase. Nevertheless, bacilysin production occurred when protein synthesis was inhibited by chloramphenicol. It thus appears that there is no obligatory coupling of protein synthesis and bacilysin synthesis. 3. When dl-[1-(14)C]alanine was added to a growing culture of B. subtilis, (14)C was incorporated into bacilysin, which contains an N-terminal alanine residue. 4. Under similar conditions virtually no (14)C was incorporated into bacilysin from dl-[2-(14)C]tyrosine, l-[U-(14)C]tyrosine or [1-(14)C]acetate, although these compounds were used by the cell for the biosynthesis of other substances. These results indicate that neither tyrosine nor acetate is a precursor of the fragment of bacilysin which yields tyrosine on hydrolysis with hot 6n-hydrochloric acid. 5. The tyrosine-yielding fragment of bacilysin was labelled with (14)C from [1,6-ring-(14)C(2)]shikimic acid. The biosynthesis of bacilysin thus appears to involve a diversion from the pathway leading to aromatic amino acids at the shikimic acid stage, or a subsequent one.  相似文献

A crystalline 3α-hydroxysteroid: NAD+-oxidoreductase (EC 1 1.1.50) which had been obtained from the cell-free extracts of Pseudomonas putida NRRL B-11064 in the presence of added polyethylene glycol, was found to be a native monomer form with a specific activity of 63.0 and a molecular weight of 45,000. Isoelectric focusing exhibited the enzyme to be composed of two isoenzymes: one major part focusing at pH 4.75 and a minor part focusing at pH 5.10. Whereas the enzyme was changed from the monomeric form to a dimeric one with a considerable decrease in the specific activity during the course of crystallization in the absence of the added polyethylene glycol.

The enzyme showed an absolute specificity with regard to 3α-hydroxyl group besides a high requirement for cis A: B fusion of steroids. Typical substrates are cholic acid (Km = 1.33 × 10?5 m), deoxycholic acid, chenodeoxycholic acid, 3α-hydroxy-12-keto-9,11-cholanoic acid, and etiocholan-3α-ol-17-one. Conjugated bile acids such as taurocholic acid and glycocholic acid are also rapidly oxidized. The pH optima for oxidation of cholic acid and reduction of etiocholan-3,17-dione were 11.5 and 7.0, respectively. The enzyme could be employed for the sensitive and specific assay of bile acids.  相似文献

Agar obtained from the red alga Hydropuntia cornea was blended with polyvinyl alcohol (PVOH) in order to produce biodegradable films. In this study, we compare the properties of biopolymeric films formulated with agars extracted from H. cornea collected at different seasons (rainy and dry) in the Gulf of Mexico coast and PVOH as synthetic matrix. The films were prepared at different agar contents (0%, 25%, 50%, 75%, and 100%) and their optical, mechanical, thermal, and morphological properties analyzed. The tensile strength of PVOH–agar films increased when agar content was augmented. The formulation with 50% agar from rainy season (RS) had a significant higher tensile strength when compared to those from dry season (DS; p < 0.05). Tensile modulus also displayed an increasing trend and likewise, for 50% and 75% agar blends from RS showed higher values than those from DS (p < 0.05). In contrast, elongation at break decreased as the agar content increased, independently of the season. Environmental scanning electron microscopy images of PVOH–agar 75% biofilms from RS showed a homogeneous structure with good interfacial adhesion between the two components. The changes evidenced in the FTIR spectrum of this blend suggest that hydrogen bonding is taking place between the agar ether linkages (C-O-C) and the hydroxyl groups (OH) of the PVOH. Based on the above mentioned results, blends of PVOH and 75% agar from H. cornea collected in rainy season showed good properties for applications in the biodegradable packaging industry.  相似文献

Objective

In the pathogenesis of coronary atherosclerosis, local macrophage-driven inflammation and secretion of proinflammatory cytokines, interleukin-1β (IL-1β) in particular, are recognized as key factors. Moderate alcohol consumption is associated with a reduced risk of coronary artery disease mortality. Here we examined in cultured human macrophages whether ethanol modulates the intracellular processes involved in the secretion of IL-1β.

Results

Ethanol decreased dose-dependently the production of mature IL-1β induced by activators of the NLRP3 inflammasome, i.e. ATP, cholesterol crystals, serum amyloid A and nigericin. Ethanol had no significant effect on the expression of NLRP3 or IL1B mRNA in LPS-primed macrophages. Moreover, secretion of IL-1β was decreased in parallel with reduction of caspase-1 activation, demonstrating that ethanol inhibits inflammasome activation instead of synthesis of pro-IL-1β. Acetaldehyde, a highly reactive metabolite of ethanol, had no effect on the ATP-induced IL-1β secretion. Ethanol also attenuated the secretion of IL-1β triggered by synthetic double-stranded DNA, an activator of the AIM2 inflammasome. Ethanol conferred the inhibitory functions by attenuating the disruption of lysosomal integrity and ensuing leakage of the lysosomal protease cathepsin B and by reducing oligomerization of ASC.

Conclusion

Ethanol-induced inhibition of the NLRP3 inflammasome activation in macrophages may represent a biological pathway underlying the protective effect of moderate alcohol consumption on coronary heart disease.  相似文献

The characteristics of electron drift in a mixture of argon with mercury vapor at reduced electric fields of E/N = 1–100 Td are calculated and analyzed with allowance for inelastic collisions. It is shown that even a minor additive of mercury to argon at a level of a fraction of percent substantially affects the discharge parameters, in particular, the characteristics of inelastic processes. The influence of the concentration of mercury vapor in argon on the kinetic characteristics, such as the diffusion and mobility coefficients and ionization frequency, is investigated.  相似文献
