Lactoferrin is an iron-binding glycoprotein found in the milk of most mammals for which various biological functions have been reported, such as antimicrobial activity and bifidogenic activity. In this study, we compared the bifidogenic activity of bovine lactoferrin (bLF) and pepsin hydrolysate of bLF (bLFH), isolated bifidogenic peptide from bLFH, and investigated the bifidogenic spectra of bLF, bLFH, and its active peptide against 42 bifidobacterial strains comprising nine species. Against Bifidobacterium breve ATCC 15700T, minimal effective concentrations of bLF and bLFH were 300 and 10 μg/ml. Against Bifidobacterium longum subsp. infantis ATCC 15697T, the minimal effective concentration of bLFH was 30 μg/ml, and bLF did not show bifidogenic activity within 300 μg/ml. As an active peptide, a heterodimer of A1-W16 and L43-A48 linked by a disulfide bond was isolated. Previously, this peptide was identified as having antibacterial activity. An amino acid mixture with the same composition as this peptide showed no bifidogenic activity. The strains of each species whose growth was highly promoted (>150%) by this peptide at 3.75 μM were as follows: B. breve (7 out of 7 strains [7/7]), B. longum subsp. infantis (5/5), Bifidobacterium bifidum (2/5), B. longum subsp. longum (1/3), Bifidobacterium adolescentis (3/6), Bifidobacterium catenulatum (1/4), Bifidobacterium pseudocatenulatum (0/4), Bifidobacterium dentium (0/5), and Bifidobacterium angulatum (0/3). Growth of none of the strains was highly promoted by bLF at 3.75 μM. We demonstrated that bLFH showed stronger bifidogenic activity than natural bLF, especially against infant-representative species, B. breve and B. longum subsp. infantis; furthermore, we isolated its active peptide. By use of pepsin a plastein was synthesized from a peptic hydrolysate of soybean protein and characterized as a protein-like substance, based on its behavior against trichloroacetic acid and its affinity to dyes. The contribution of the S–S bridge to plastein formation was little if any. The fractionation of the protein-like substance by solubility showed that the whole plastein-reaction product was constituted of 81.5 parts of the 50% ethanol-insoluble fraction; 63.8% of this fraction was also insoluble in 0.035 M phosphate buffer (pH 7.6). The phosphate buffer-insoluble fraction was mostly solubilized in 0.3 M sodium dodecyl sulfate (SDS). Although it was a minor component, there was a 50% ethanol-soluble and water-insoluble (prolamine-like) fraction which showed a reversible aggregation-dispersion change by a repeated heating-cooling treatment. A characteristic point of the SDS-soluble and prolamine-like fractions was found in their amino acid compositions. As compared with the substrate (peptic hydrolysate of soybean protein), they contained larger amounts of hydrophobic amino acids such as leucine than hydrophilic ones typified by glutamic acid. 相似文献

香菇粉经10℃pH 10的水提取制备香菇蛋白,得率13.1%,其蛋白含量47.5%,多糖含量24.2%.香菇蛋白经DEAE Sepharose CL-6B柱层析分级得5个级分,收集级分F1、F2、F3、F4,它们都是由蛋白和多糖构成的复合物.Sepharose CL-6B凝胶色谱显示,F2和F4的分子量分布较为均匀,且以蛋白为主,多糖含量很低;F3主要由两个分子量不同的蛋白级分构成,含有一定的多糖;F1中多糖含量较高,蛋白含量较少,且多糖分子量分布均匀.香菇蛋白的分子量主要集中在20 kDa~40 kDa之间.F1、F3、F4都属于酸性蛋白质,含有除色氨酸之外的7种必需氨基酸,除蛋氨酸含量较低外,其余必需氮基酸含量接近,且赖氨酸含量较高.红外光谱分析表明,香菇蛋白的二级结构主要为α-螺旋和无规卷曲. 相似文献

鱼类抗菌肽是鱼体天然免疫的重要组成部分,其结构和组成复杂多样.根据生化和结构特点,可将它们分为4种基本类型:具有疏水或双亲性α螺旋结构的抗菌肽、含多对二硫键并可形成β折叠结构的抗菌肽、组蛋白样抗菌肽和具有糖基化修饰的抗菌肽.鱼类抗菌肽多以前肽原的形式合成,通过酶解切除信号肽和羧基端酸性片段后形成有活性的成熟肽.成熟肽具有很强的抑菌活性,其最小抑制浓度多在毫摩尔水平.目前,已克隆了多个鱼类抗菌肽基因,揭示了pleurocidin等基因家族的结构及其转录调控特点.鱼类抗菌肽的抗菌机制已建立了“桶-桶板”和“地毯”样两种模型,基本阐明了抗菌肽分子结构与抗菌功能间的关系. 相似文献

Commercial Philippine fish sauce (Patis) was steam-distilled and the distillate was fractionated into four fractions, neutral, basic, acidic and phenolic and each fraction was analyzed using gas chromatography and gas chromatography-mass spectrometry.As a result of this study, a total of 66 compounds were identified in Patis, 14 of which were only a tentative identification. Out of these 66 identified compounds, 40 identified compounds have not been reported in previous studies on fish sauces. These identified compounds include 19 acids, 14 alcohols, 12 nitrogen containing compounds, 5 esters, 3 sulfur containing compounds, 1 phenol, 3 carbonyls, 7 hydrocarbons and 2 others. In the acidic fraction, 5 acids were considered major constituents which accounted for about 98% of the total acids. n-Butanoic acid was found to be the most abundant accounting for about 50% of the total acids. 相似文献

Sesame peptide powder (SPP) exhibited angiotensin I-converting enzyme (ACE) inhibitory activity, and significantly and temporarily decreased the systolic blood pressure (SBP) in spontaneously hypertensive rats (SHRs) by a single administration (1 and 10 mg/kg). Six peptide ACE inhibitors were isolated and identified from SPP. The representative peptides, Leu-Val-Tyr, Leu-Gln-Pro and Leu-Lys-Tyr, could competitively inhibit ACE activity at respective Ki values of 0.92 μM, 0.50 μM, and 0.48 μM. A reconstituted sesame peptide mixture of Leu-Ser-Ala, Leu-Gln-Pro, Leu-Lys-Tyr, Ile-Val-Tyr, Val-Ile-Tyr, Leu-Val-Tyr, and Met-Leu-Pro-Ala-Tyr according to their content ratio in SPP showed a strong antihypertensive effect on SHR at doses of 3.63 and 36.3 μg/kg, which accounted for more than 70% of the corresponding dosage for the SPP-induced hypotensive effect. Repeated oral administration of SPP also lowered both SBP and the aortic ACE activity in SHR. These results demonstrate that SPP would be a beneficial ingredient for preventing and providing therapy against hypertension and its related diseases. 相似文献

ABSTRACT:?Fish metabolism needs special enzymes that have maximum activity at very different conditions than their mammalian counterparts. Due to the differences in activity, these enzymes, especially cold-adapted proteases, could be used advantageously for the production of some foods. In addition to the enzymes, this review describes some other unique fish polypeptides such as antifreeze proteins, fluorescent proteins, antitumor peptides, antibiotics, and hormones, that have already been cloned and used in food processing, genetic engineering, medicine, and aquaculture. Recombinant DNA technology, which allows these biological molecules to be cloned and overexpressed in microorganisms is also described, highlighting innovative applications. Cell-penetrating peptides (CPPs) are short peptides which can carry various types of molecules into cells; however, although most CPPs rapidly penetrate cells in vitro, their in vivo tissue-targeting specificities are low. Herein, we describe cell-binding, internalization, and targeting characteristics of a newly identified 10-residue CPP, denoted ECP32–41, derived from the core heparin-binding motif of human eosinophil cationic protein (ECP). Besides traditional emphasis on positively charged residues, the presence of cysteine and tryptophan residues was demonstrated to be essential for internalization. ECP32–41 entered Beas-2B and wild-type CHO-K1 cells, but not CHO cells lacking of cell-surface glycosaminoglycans (GAGs), indicating that binding of ECP32–41 to cell-surface GAGs was required for internalization. When cells were cultured with GAGs or pre-treated with GAG-digesting enzymes, significant decreases in ECP32–41 internalization were observed, suggesting that cell-surface GAGs, especially heparan sulfate proteoglycans were necessary for ECP32–41 attachment and penetration. Furthermore, treatment with pharmacological agents identified two forms of energy-dependent endocytosis, lipid-raft endocytosis and macropinocytosis, as the major ECP32–41 internalization routes. ECP32–41 was demonstrated to transport various cargoes including fluorescent chemical, fluorescent protein, and peptidomimetic drug into cultured Beas-2B cells in vitro, and targeted broncho-epithelial and intestinal villi tissues in vivo. Hence this CPP has the potential to serve as a novel vehicle for intracellular delivery of biomolecules or medicines, especially for the treatment of pulmonary or gastrointestinal diseases. 相似文献

Here, we describe peptide aggregation, which is also known as enzymatic protein resynthesis. Whey protein hydrolysate (WPH) is the starting material for assembling peptides. Analyses of the involved amino acids, intrinsic fluorescence, fluorescence phase diagram, secondary structure, turbidity, and surface hydrophobicity were performed to investigate the reaction process. The aggregation mechanism consists of two parts: 1) formation and 2) aggregation of the building blocks that form the ordered secondary β-sheet structure. Constructing the building blocks requires at least one intermediate state, which is formed after 0.5 hours. Non-synergistic changes in the secondary and tertiary structures then allow the intermediate state to emerge. 相似文献

With the aid of papain, a plastein was synthesized from a 1 : 10 mixture of l-methionine ethyl ester and a peptic hydrolysate of soybean protein. Dialysis of the whole reaction-product yielded a methionine-incorporated plastein (Met-plastein) as the nondialyzable fraction, its yield being 78.2% on a dry-matter basis, of the whole reaction-product. Methionine content in this Met-plastein was 7.22% on a weight basis, while its content in the material hydrolysate was only 1.25%. Carboxypeptidase A treatment of Met-plastein liberated methionine at an outstandingly rapid rate. A similar, but not so outstanding, rate was observed for the methionine liberation from Met-plastein by treatment with leucine aminopeptidase. Methyl isothiocyanate treatment and subsequent cyclization yielded a mixture of methylthiohydantoins from Met-plastein. Gas chromatographic analysis of this mixture after trimethylsilylation showed a result that methionine occupied 33.2%, on a molar basis, of the total N-terminal amino acids. Lithium borohydride reduction and 6 n hydrochloric, acid hydrolysis of Met-plastein produced monomeric aminois, and their 2,4-dinitrophenylation followed by thin-layer chromatography gave a result that methionine occupied 84.9%, on a molar basis, of the total C-terminal amino acids; the residues amounting to 14.4% of the C-terminal methionine residues remained as an ethyl eater form. The selective degradation probe employing cyanogen bromide to generate free homoserine disclosed that the occurrence of the polymeric methionine-methionine sequence was little if any in Met-plastein. Based on the above experiments as well as an evaluation of the esterase activity against l-methionine ethyl ester, a possible mechanism was discussed of the papain-catalyzed synthesis of plastein in a system containing such an ester. 相似文献

Dry hydrolysate was prepared from protein-containing waste of Icelandic scallop processing by means of a proteinase complex from king crab hepatopancreas. The resulting product contains no less than 80% free amino acids and oligopeptides. Predominant are aspartic acid, leucine, isoleucine, arginine, and lysine, which account for more than 50% of the free amino acids. The potential of using the protein hydrolysate as a nutrient for microorganism cultivation is estimated. It is demonstrated that the hydrolysate can be used for growing test cultures. 相似文献
