Endophytic Bacillus sp. CY22 was previously isolated from the root interior of the balloon flower (Platycodon grandiflorum) (Cho et al., Biosci. Biotechnol. Biochem., 66, 1270-1275 (2002)). Three-month-old balloon flower seedlings were inoculated with 107 cfu/ml of strain CY22R3, a rifampicin-resistant strain of CY22, and external and internal root colonization was assessed 2 and 4 weeks later. After inoculation, large numbers of bacteria were observed on the root surface by scanning electron microscopy. More detailed studies using optical and transmission electron microscopy confirmed that Bacillus sp. CY22 was endophytically established within intercellular spaces, cortical cells, and aerenchymas of root. Also, Bacillus sp. CY22 showed antibiotic activities against several phytopathogens by producing the antibiotic iturin A. In the pot test, root rot of balloon flower seedlings caused by Rhizoctonia solani was suppressed when the Bacillus sp. CY22R3 was inoculated into the soil.  相似文献

Natural gypsum (NG) and phosphogypsum (PG) were tested for resistance to fungal growth based on standard test ASTM D 3273-86, with the recommended mixture of three fungal species, and using the same test modified by the use of a Cladosporium sp. A, isolated from a gypsum plaster ceiling. In the standard test little growth occurred on any of the test specimens. However, abundant fungal growth was produced by the Cladosporium sp. A on phosphogypsum, which was much more susceptible than natural gypsum. Phosphogypsum heated to 600°C to destroy organic residues was resistant to growth of Cladosporium sp. A, as well as other fungi isolated from phosphogypsum panels stored in the environment for 2 years: Cladosporium sp. B, Aspergillus niger and Trichoderma sp. Phosphogypsum moulded in Petri dishes was susceptible to growth of a wide range of fungi, although Fusarium sp. and Rhizopus sp. caused practically no discoloration of the substrate. Six biocides were separately incorporated into the phosphogypsum at concentrations recommended by the suppliers and test specimens incubated on Sabouraud agar inoculated with various fungal isolates. The biocide 2-N-octyl-4-isothiazolin-3-one was the most efficient compound. It prevented the growth of the fungi most likely to cause health problems in buildings, but not that of Helminthosporium sp., isolated from powdered phosphogypsum in the factory. This was the most resistant fungus showing growth on all biocide-containing specimens.  相似文献

Different Gram-positive and Gram-negative bacteria (Staphylococcus xylosus, S. aureus, S. cohnii, Bacillus sp., Corynebacterium sp., Pseudomonas vesicularis) were isolated from homogenized shoot tips of Drosera rotundifolia, Spatiphyllum sp., Syngonium cv. White butterfly, Nephrolepis exaltata cv. Teddy Junior. Growth inhibition of selected bacterial strains was examined using 28 different single antibiotics and 7 antibiotic mixtures. It was found that with the two mixtures Imipenem/Ampicillin and Imipenem/Penicillin G at concentrations of 5 mg l–1 each, bacterial growth inhibition was most effective. Because of the lack of toxic effects on in vitro plants of 7 species it was proposed that these antibiotic mixtures can be applied advantageously to inhibit bacterial growth in tissue culture.  相似文献

A new antifungal antibiotic named acmycin was isolated from a soil streptomycete. Detailed comparative taxonomic studies showed that the organism differed from three related species of streptomycetes. The organism was referred to asStreptomyces sp. AC2. Microbial transformation of the nucleoside analogue antibiotic showdomycin was performed using some Streptomyces species. Both the growing culture and the resting cells of Streptomyces sp. No. 383 arrested the antibacterial activity of showdomycin. The inactivated showdomycin was isolated from the reaction mixture by carbon chromatography and was identified with an isoshowdomycin sample which has been chemically derived from showdomycin. It is conjectured that the conversion of showdomycin to isoshowdomycin results from isomerization by an enzyme of Streptomyces sp. No. 383.  相似文献

An actinomycete strain KN-0647 was isolated from a forest soil sample collected from Dali Cangshan mountain, Yunnan Province, China. The strain was identified as Streptomyces sp. according to the morphological, physiological characteristics and whole nucleotide sequence analysis of 16S rRNA gene, and could not be identified up to species level, just suggesting a potential new taxon. The ethyl acetate extract from this strain displayed growth inhibition on the test pathogenetic insects, such as Spodoptera exigua, Dendrolimus punctatus, Plutella xylostella, Aphis glycines and Culex pipiens. The active compound was isolated and identified through a combination of spectral and chemical methods (UR, MS, and 1HNMR) as quinomycin A. This is the first report on the insecticidal activity of antibiotic quinomycin A.  相似文献

A screening for antibacterial antibiotics was carried out with psychrophilic microorganisms. The most active microorganism, a soil actinomycete, was selected and characterized to be a facultative psychrophile, Streptomyces sp. No. 81. This strain was found to produce antibiotic(s) in the culture fluid only at low temperature cultivation below 20°C but not at moderate temperature. Mycelial growth at low temperature seemed to be indispensable for the antibiotic production. The antibiotic produced by Streptomyces sp. No. 81 was isolated and characterized. It appeared that the antibiotic had the selective toxicity against several Gram-positive bacteria. From the comparative studies with several known antibiotics, the antibiotic appears to be a new compound derived from the new metabolic routes involving temperature-sensitive mechanisms.  相似文献

金荞麦和苦荞麦抗菌活性内生真菌的筛选及鉴定   总被引:1,自引:0,他引:1  
从药用植物金荞麦和苦荞麦的根、茎、叶、花中分离到62株内生真菌,并以金黄色葡萄球菌(Staphylococcus aureus)、大肠杆菌(Escherichia coli)、枯草芽孢杆菌(Bacillus subtilis)[CMCC(B)63501]、小麦赤霉病菌(Fusarium graminearum)、黄瓜枯萎病菌(Fusarium oxysporum f.sp.cu-cumerinum)和绵腐病菌(Pythium aphanidermatum)6种微生物为指示菌对分离到的内生真菌进行抗菌活性检测。结果发现,分离的内生真菌菌株KQH-01、KQH-02和JQY-1的发酵醇提取物具有较好的抑菌活性。形态学特征和分子鉴定确定菌株KQH-01为炭角菌属(Xylaria sp.)真菌,菌株KQH-02为球毛壳菌(Chaetomium globosum),菌株JQY-1为葡萄座腔菌(Botryosphaeria dothidea)。  相似文献

Morphological, cultural and physiological-biochemical properties ofStreptomyces sp. strain 1000 and its antibiotic production were investigated. Antibiotics 1011 (identical with the streptovaricin complex) and 1012 (with antibacterial action) were isolated from the cultural broth of this strain. The overproducing natural variant 1011 was isolated from the population of a strain producing antibiotic 1011 at a concentration of 1000 mg/L (activity of the parent strain represents 41 mg/L only). Comparative taxonomical characteristic ofStreptomyces sp. strain 1000 with strains fromS. spectabilis showed that the strain 1000 differed in some properties and antibiotic production being considered as a new variant ofS. spectabilis. The strain shows an expressed antibiotic activity against G+ as well as G− bacterial and yeasts.  相似文献

A streptomycete isolated in our laboratories was found to produce a new antibiotic effective for the control of sheath blight of the rice plant. As a result of taxonomical study it was proved to belong to a new species and therefore it was named Streptomyces multispiralis nov. sp. From its fermented broth, the antibiotic was isolated and crystallized from benzene. After the comparison of its physicochemical and biological properties with those of known antibiotics, it was considered to be a new antibiotic and named neohumidin. It inhibited the growth of certain phytopathogenic fungi, saprophytic fungi, yeasts and gram-positive bacteria.  相似文献

From the results of taxonomic studies, Streptomyces sp. strain No. KT–151 isolated from a soil sample collected in Kumamoto City, was identified as a strain belonging to Streptomyces luteogriseus Schmitz, Deak, Crook and Hooper 1964. A new antibiotic, produced by this strain, was isolated as a leaflet crystal by ion-exchange chromatography and found to be an amino acid with the molecular formula, C5H12N2O2, and named antibiotic KT–151 (refered to as KT–151 hereinafter). The antibiotic showed antimicrobial activity against various Gram-positive and Gram-negative bacteria in a chemically defined medium but it was antagonized by several amino acids such as valine, leucine, isoleucine and threonine.  相似文献

The inside of Pinus thunbergii could be a reliable screening source for a useful agent in controlling plant disease. Isolation of endophytic actinomycetes from P. thunbergii and their potential as biocontrol agents against the plant pathogen Cylindrocladium sp. were investigated. Two endophytic actinomycetes, Streptomyces sp. and Microbispora sp., were isolated from surface-sterilised root tissues of P. thunbergii seedlings. The recovery test of these two endophytic actinomycetes from pine seedling showed that Streptomyces sp. was isolated from only roots, but Microbispora sp. was isolated from both roots and leaves. Thus, Microbispora sp. is able to move to leaves from roots. Moreover, we evaluated the potential of both strains as biocontrol agents against Cylindrocladium sp. Two weeks after inoculation of Cylindrocladium sp. alone, pine seedlings showed a 50% mortality rate. Co-inoculation of Cylindrocladium sp. and Microbispora sp. did not affect seedling mortality rate. However, inoculation with both Cylindrocladium sp. and Streptomyces sp. reduced seedling mortality to 12%. Streptomyces sp. could be a useful agent in controlling pine disease caused by Cylindrocladium sp. Thus, it seems that Streptomyces sp. may induce a local host defence reaction and Microbispora sp. systemically spreads to aerial parts through the transpiration stream.  相似文献

Twenty-one strains belonging to 18 species of basidiomycetes from different ecological groups of fungi were isolated from natural sources. Light and electron microscopy was used to determine the morphological properties of the cultures, which confirmed their classification as basidiomycetes and facilitated their identification in monocultures. The capacity of the fungal strains for biosynthesis of antibiotics was determined by one- or two-stage cultivation on seven nutrient media. It was established that, under submerged cultivation, antimicrobial substances were formed by 13 strains (81.25%) of 12 fungal species (Armillaria sp., Coprinus comatus, Flammulina velutipes, Hypsizygus ulmarius, Lentinus tigrinus, Lycoperdon pyriforme, Macrolepiota procera, Panellus serotinus, Pholiota aurivella, Pholiota lenta, Rhodocollybia maculate, and Sparassis crispa). The antibiotics formed were efficacious against bacterial test strains, including the methicillin-resistant strain Staphylococcus aureus (MRSA) and the strain Leuconostoc mesenteroides VKPM B-4177 that is resistant to the glycopeptide antibiotics. No antibiotic activity was revealed against fungal test cultures (Aspergillus niger INA 00760 and Saccharomyces cerevisiae RIA 259).  相似文献

During early Spring (April–May) when the seawater salinity drops suddenly and the seawater temperature increases drastically, severe lesions and thallus bleaching were observed in the Laminaria religiosa population at Oshoro Bay, Otaru, Hokkaido, Japan. The healthy and diseased kelp blades were collected and subjected to enumeration of total number of culturable bacteria and bacterial species. Bacterial enumerations were done using 3 different media formulations; high-nutrient media (Media 1), low-nutrient media (Media 2) and modified low-nutrient media with 5% kelp extract (Media 3). Seven bacterial species were isolated from the healthy kelp. These were Alcaligenes aquamarinas, Alteromonas sp., Azomonas agilis, Azotobacter beijerinckii, Escherichia coli, Halobacterium sp. and Halococcus sp. All 7 bacterial species were isolated on Media 2 and Media 3, but only 5 species were isolated using Media 1 with the absence of Halobacterium sp. and Halococcus sp. Highest total number of culturable bacteria was 2050 CFU/cm2 on Media 3. Eight species of bacteria were isolated from the diseased kelp thallus with the addition of Erwinia amylovora. All 8 bacteria grew on Media 2 and Media 3, but only 6 species were isolated using Media 1 with the absence of Halobacterium sp. and Halococcus sp. Highest total number of culturable bacteria was 5830 CFU/cm2 on Media 3. However, only 3 species were isolated from the lesioned area. The most abundant species was Alteromonas sp. followed by Halococcus sp. and Alcaligenes aquamarinas. The surface bacteria showed best growth on Media 3. Scanning Electron Microscopic images of the healthy and diseased thallus gave distinctive evidence of the severity of the lesions as well as the relative abundance in the bacterial population. In an effort to identify the symptoms causative organism, the isolated bacterial species were cultured and used to test Koch's postulates. Out of the 8 species, only Alteromonas sp. induced lesions on the axenic kelp blades. The inoculated bacteria were also re-isolated without any significant contamination. Hence, Alteromonas sp. is suggested as the possible disease causing organism.  相似文献

Active substances which increased RNA content and RNA productivity in yeast culture without affecting the growth rate of yeast were investigated.

The remarkable effect of zinc ion on RNA accumulation was found in flask cultures of Candida utilis.

The active substance of culture of Saccharomyces cerevisiae was isolated from the culture filtrate of Streptomyces sp. S–22 and it was identified as anisomycin, an antiprotozoal and antifungal antibiotic. The effect of anisomycin on the enhancement of yeast RNA formation was shown only with the Saccharomyces genus, which was more sensitive to the antibiotic than other genus. This phenomenon was exhibited only in the case of anisomycin and cycloheximide, whose modes of action were similar among various antibiotics. The ratio of four nucleotides in RNA fraction was almost equal to that of ribosomal RNA.  相似文献

APseudomonas sp. and aFusarium sp. were isolated from pasture soil. The organisms grew syntrophically on a mineral medium with benzylpenicillin as carbon and nitrogen source. During synergistic degradation of the antibiotic, benzylpenicilloic and benzylpenilloic acid were intermediates. Activities of both organisms were necessary for further decomposition, during which the phenylacetate side chain was degraded. 6-Aminopenicillanic acid did not occur. During syntrophic growth, a red pigment appeared in fungal hyphae growing through bacterial colonies.  相似文献

Deschampsia antarctica Desv. is the only gramineae capable of colonizing the Antarctic due to the region’s extreme climate and soil environment. In the present research, bacteria colonizing the rhizospheric soil of D. antarctica were isolated and characterized. The soil studies showed that D. antarctica possesses a wide spectrum of psychrotolerant bacteria with extensive and varied antibiotic resistance, as well as heavy metal tolerance. The bacterial strains isolated from the rhizosphere of D. antarctica also produced a diverse pattern of enzymes. Based on the strain identification with partial characterization of the 16S rRNA gene, the majority of the isolates correspond to different Pseudomonas species, and species of the genus Flavobacterium sp. and Arthrobacter sp. The isolated strains collected from this research constitute a unique collection for future, more detailed taxonomic analysis and physiological characterization, contributing to the search for potential biotechnological uses. These findings and others have great potential for developing new biotechnological products from Antarctic microorganisms.  相似文献

An unusual Phomopsis sp. was isolated as endophyte of Odontoglossum sp. (Orchidaceae), associated with a cloud forest in Northern Ecuador. This fungus produces a unique mixture of volatile organic compounds (VOCs) including sabinene (a monoterpene with a peppery odor) only previously known from higher plants. In addition, some of the other more abundant VOCs recorded by GC/MS in this organism were 1-butanol, 3-methyl; benzeneethanol; 1-propanol, 2-methyl and 2-propanone. The gases of Phomopsis sp. possess antifungal properties and an artificial mixture of the VOCs mimicked the antibiotic effects of this organism with the greatest bioactivity against a wide range of plant pathogenic test fungi including: Pythium, Phytophthora, Sclerotinia, Rhizoctonia, Fusarium, Botrytis, Verticillium, and Colletotrichum. The IC50 values for the artificial gas mixture of Phomopsis sp. varied between 8 and 25.65 μl/mL. Proton transfer reaction-mass spectrometry monitored the concentration of VOCs emitted by Phomopsis sp. and yielded a total VOC concentration of ca. 18 ppmv in the head space at the seventh day of incubation at 23°C on PDA. As with many VOC-producing endophytes, this Phomopsis sp. did survive and grow in the presence of the inhibitory gases of Muscodor albus. A discussion is presented on the possible involvement of VOC production by the fungus and its role in the biology/ecology of the fungus/plant/environmental relationship.  相似文献

A total of 228 bacteria with an ability to resist toxic heavy metals were isolated from 8 selected sites of the Lagos Lagoon. The bacteria isolated wereStaphylocaccus sp.,Bacillus sp.,Pseudomonas sp.,Streptococcus sp.,Moraxella sp.,Escherichia coli, Proteus sp.,Klebsiella sp. andSalmonella sp. The heavy metals to which resistance was recorded were mercury, lead, zinc, cobalt, copper and chromium. The lagoon sites from which the highest number of resistant bacteria were isolated were Marina and Ebute-Ero. The heavy metal to which most bacteria were resistant was cobalt, while the least was chromium. The significance of the result is discussed in relation to the Nigerian environment and human health.  相似文献

Rhizobium huakuii bv. renge B3, a native symbiont of Astragalus sinicus, outcompeted Rhizobium sp. strain ACMP18, which was isolated from Astragalus cicer nodules, in the formation of root nodules on A.␣sinicus when plants were co-inoculated with these strains. The strains occupying the nodules were identified by antibiotic resistance and phage sensitivity markers and also by polymerase chain reaction (PCR) genomic fingerprintings, which were performed by using enterobacterial repetitive intergenic consensus sequences. In PCR genomic fingerprintings, the total genomic DNA isolated from pure bacterial culture and from squashed root nodules showed identical profiles, indicating that this technique can be a useful tool for identification of rhizobia in ecological studies. When Rhizobium sp. strain ACMP18 outnumbered R. huakuii bv. renge strain B3 by a factor of ten, and even when strain ACMP18 was added to plants 1 week before bacterization with strain B3, the strain B3 occupied most nodules. Dually infected nodules were not observed, although Rhizobium sp. ACMP18 formed active nodules on A. sinicus when the bacterial strain was inoculated alone.
