(R,S)-trans-8-Hydroxy-2-[N-n-propyl-N-(3′-iodo-2′-propenyl)amino]tetralin 7 , a new radioiodinated ligand based on 8-OH-DPAT, was reported as a potential ligand for 5-HT1A receptors. The optically active (+)-(R)- and (?)-(S)- 7 were prepared to investigate the stereoselectivity of (R,S)- 7 . Racemic intermediate 8-methoxy-2-N-n-propyltetralin was reacted with the acyl chloride of (?)-(R)-O-methylmandelic acid to form a mixture of (S,R)- and (R,R)-diastereoisomers, which were separated by flash column chromatography. After removing the N-acyl group from the diastereoisomers, the desired (+)-(R)-or (?)-(S)- 7 was obtained by adding an N-iodopropenyl group. In vitro homogenate binding studies showed the stereoselectivity of this new compound for 5-HT1A receptors. (+)-(R)- 7 isomer displayed 100-fold higher affinity than the (?)-(S)- 7 isomer. Biochemical study indicated that (+)-(R)- 7 potently inhibited forskolin-stimulated adenylyl cyclase activity in hippocampal membranes (Emax and EC50 were 24.5% and 5.4 nM, respectively), while (?)-(S)- 7 showed no effect at 1 μM. The radioiodinated (+)-(R)- and (?)-(S)-[125I] 7 were confirmed by coelution with the resolved unlabeled compound on HPLC (reverse phase column PRP-1, acetonitrile/pH 7.0 buffer, 80/20). The active isomer, (+)-(R)-[125I] 7 , displayed high binding affinity to 5-HT1A receptors (Kd = 0.09 ± 0.02 nM). In contrast, the (?)-(S)- 7 isomer displayed a significantly lower affinity to the 5-HT1A receptor (Kd > 10 nM). Thus, (+)-(R)-[125I]trans-8-OH-PIPAT, (+)-(R)- 7 , an iodinated stereoselective 5-HT1A receptor agonist, is potentially useful for study of in vivo and in vitro function and pharmacology of 5-HT1A receptors in the central nervous system. © 1995 Wiley-Liss, Inc.  相似文献

Abstract: Previous work from this laboratory has shown that retinal adenosine A2 binding sites are localized over outer and inner segments of photoreceptors in rabbit and mouse retinal sections. In the present study, adenosine receptor binding has been characterized and localized in membranes from bovine rod outer segments (ROS). Saturation studies with varying concentrations (10–150 nM) of 5′-(N-[2,8-3H]ethylcarboxamido)adenosine ([3H]NECA) and 100 μg of ROS membrane protein show a single site with a KD of 103 nM and a Bmax of 1.3 pM/mg of protein. Cold Scatchards, which used nonradiolabeled NECA (concentrations ranging from 10 nM to 250 nM) in competition with a fixed amount of [3H]NECA (30 nM), demonstrated the presence of a low-affinity site (KD, 50 μM) in addition to the high-affinity site. To confirm the presence of A2abinding sites, saturation analyses with 2-p-(2-[3H]-carboxyethyl)phenylamino-5′-N-ethylcarboxamido adenosine (0–80 nM) also revealed a single population of high-affinity A2a receptors (KD, 9.4 nM). The binding sites labeled by [3H]NECA appear to be A2 receptor sites because binding was displaced by increasing concentrations of 5′-(N-methylcarboxamido)adenosine and 2-chloroadenosine. ROS were fractionated into plasma and disk membranes for localization studies. Receptor binding assays, used to determine specific binding, showed that the greatest concentration of A2 receptors was on the plasma membranes. Therefore, adenosine A2 receptors are in a position to respond to changes in the concentration of extracellular adenosine, which may exhibit a circadian rhythm.  相似文献

Oxidation of methvl 2-trans-β-ionylideneacetate with X-bromosuccinimide afforded methyl 2-cis and trans-3′-hydroxy-β-ionylideneacetates. NaBH4 reduction of methyl 2-cis-3′-keto-β-ionylideneacetate and ethyl 4′-keto-α-ionylideneacetate gave methyl 2-cis-3′-hydroxy-β-ionylideneacetate and ethyl 4′-hydroxy-α-ionyiideneacetate respectively. Further, methyl 4′-methoxy-epoxy-α-ionylideneacetate was prepared by epoxidation of methyl 4′-methoxy-α-ionylideneacetate. And then methyl 4′-hydroxy-l′, 2′-dihydro-β-ionylideneacetate was synthesized from ethyl 4-keto-α-cyclogeranate. Growth inhibitory activities of the above compounds on rich seedlings were examined.  相似文献

Cell-free systems were prepared from germinating seed and seedlings of Phaseolus coccineus. Gibberellin A4 (GA4)-metabolising activity was detected in vitro using preparations from roots, shoots and cotyledons of germinating seed, but only up to 24 h after imbibition. Cell-free preparations from cotyledons converted [3H]GA4 to GA1, GA34, GA4-glucosyl ester and a putative O-glucoside of GA34, and, in addition converted [3H]GA1 to GA8. Preparations from embryo tissues contained 2-hydroxylase activity, converting [3H]GA4 to GA34 and [3H]GA1 to GA8.The presence of GA-metabolising enzymes was also indicated by in-vivo feeds of [3H]GA4 to epicotyls of intact 4-d-old seedlings, which resulted in the accumulation of GA1, GA8, GA3-3-O-glucoside, GA4-glucosyl ester, GA8-2-O-glucoside and a putative O-glucoside of GA34. Gibberellin A1 was the first metabolite detected, 15 min after application of [3H]GA4, but after 24 h most of the label was associated with GA8-2-O-glucoside. Over 90% of the recovered radioactivity was found in the shoot. The main component of Japanese Ho-leaf oil has been shown to be (?)-linalool (80~90%), and the following twenty minor constituents newly have been identified; methyl vinyl ketone, methyl isobutyl ketone, mesityl oxide, β-pinene, myrcene, (+)-limonene, cis- and trans-ocimene, n-hexanol, cis-3-hexenol, cis- and trans-linalool oxide, (?)-1-terpinen-4-ol, (+)-cis and (+)-trans-2,6,6-trimethyl-2-vinyl-5-hydroxytetrahydropyran, citronellol, nerol, (+)-β-selinene, (+)-tagetonol and (?)-trans-hotrienol. (+)-Tagetonol and (?)-trans-hotrienol have been demonstrated to be (+)-3,7-dimethyl-3-hydroxy-1-octen-5-one (III) and (3R)-(?)-trans-3,7-dimethyl-3-hydroxy-1,5,7-octatriene (IX), respectively.  相似文献

Prothallia of Lygodium japonicum (Thunb.) Sw. were aseptically cultured under white light in a mineral solution. Solvent fractionation of the resultant culture medium and subsequent preparative thinlayer chromatography yielded a fraction that induced antheridium formation and inhibited archegonium formation. Combined gas chromatography-selected ion monitoring analysis of this fraction confirmed the presence of gibberellin A9 methyl ester (GA9-me) as an antheridiogen and an inhibitor of archegonium formation. Exogenously applied [3H]GA9 was rapidly converted to [3H]GA9-me in the prothallial tissue. Abstract: The specific binding of [3H]WAY-100635 {N-[2-[4-(2-[O-methyl-3H]methoxyphenyl)-1-piperazinyl]ethyl]-N-(2-pyridinyl)cyclohexane carboxamide trihydrochloride} to rat hippocampal membrane preparations was time, temperature, and tissue concentration dependent. The rates of [3H]WAY-100635 association (k+1 = 0.069 ± 0.015 nM?1 min?1) and dissociation (k?1 = 0.023 ± 0.001 min?1) followed monoexponential kinetics. Saturation binding isotherms of [3H]WAY-100635 exhibited a single class of recognition site with an affinity of 0.37 ± 0.051 nM and a maximal binding capacity (Bmax) of 312 ± 12 fmol/mg of protein. The maximal number of binding sites labelled by [3H]WAY-100635 was ~36% higher compared with that of 8-hydroxy-2-(di-n-[3H]-propylamino)tetralin ([3H]8-OH-DPAT). The binding affinity of [3H]WAY-100635 was significantly lowered by the divalent cations CaCl2 (2.5-fold; p < 0.02) and MnCl2 (3.6-fold; p < 0.05), with no effect on Bmax. Guanyl nucleotides failed to influence the KD and Bmax parameters of [3H]WAY-100635 binding to 5-HT1A receptors. The pharmacological binding profile of [3H]WAY-100635 was closely correlated with that of [3H]8-OH-DPAT, which is consistent with the labelling of 5-hydroxytryptamine1A (5-HT1A) sites in rat hippocampus. [3H]WAY-100635 competition curves with 5-HT1A agonists and partial agonists were best resolved into high- and low-affinity binding components, whereas antagonists were best described by a one-site binding model. In the presence of 50 µM guanosine 5′-O-(3-thiotriphosphate) (GTPγS), competition curves for the antagonists remained unaltered, whereas the agonist and partial agonist curves were shifted to the right, reflecting an influence of G protein coupling on agonist versus antagonist binding to the 5-HT1A receptor. However, a residual (16 ± 2%) high-affinity agonist binding component was still apparent in the presence of GTPγS, indicating the existence of GTP-insensitive sites.  相似文献

Summary The binding properties of A1 adenosine receptors in brain membranes were compared in two congeneric marine teleost fishes which differ in their depths of distribution. Adenosine receptors were labeled using the A1 selective radioligand [3H]cyclohexyladenosine ([3H]CHA). The A1 receptor agonist [3H]CHA bound saturably, reversibly and with high affinity to brain membranes prepared fromSebastolobus altivelis andS. alascanus; however, the meanK d values differed significantly (Figs. 1–3, Table 1). Saturation data fit to a one site model indicated that the A1 receptor inS. alascanus exhibited a higher affinity (K d=1.49 nM) for [3H]CHA whereas A1 receptors inS. altivelis exhibited a significantly lower affinity (K d=3.1 nM). Moreover,S. altivelis, but notS. alascanus, parameter estimates for [3H]CHA binding to two sites of receptor were obtained (Fig. 3, Table 1). The mean dissociation constant values for the high and low affinity sites for [3H]CHA inS. altivelis were 0.43 nM and 16.3 nM, respectively. In equilibrium competition experiments the adenosine analogs R-phenylisopropyladenosine (R-PIA), N-ethylcarboxamidoadenosine (NECA) and S-phenylisopropyladenosine (S-PIA) all displayed higher affinities for A1 receptors inS. alascanus as compared toS. altivelis brain membranes (Table 2, Fig. 6). The specific binding of [3H]CHA was significantly increased by 0.1 and 1.0 mM MgCl2 in both fishes; however, the sensitivity (95–131% increase) ofS. altivelis to this effect was significantly greater than that ofS. alascanus (48–91% increase) (Fig. 5). A silver ion-loaded microparticulate cation-exchange resin column has been used for high-performance liquid chromatographic (HPLC) separation of the p-nitrophenacyl esters of several series of closely related prostaglandins: 8-iso-PGE2, 11-epi-PGE2, 5-trans-PGE2, PGE2, PGF, PGE1, and PGF, PGA2 and PGB2; 15 (R)-methyl-PGE2 and 15 (S)-methyl-PGE2; 5-trans-PGA2 and PGA2; and 5-trans-PGF and PGF. ABSTRACT

In Chinese Hamster Ovary (CHO) cells expressing cloned human 5-hydroxy-tryptamine1A (5-HT1A) receptors, (R)-3-N,N-dicyclobutylamino-8-fluoro-[6-3H]-3,4-dihydro-2H-1-benzopyran-5-carboxamide ([3H]NAD-299) exhibited high affinity (Kd = 0.16?nM) and labeled 34% more receptors than 8-hydroxy-2-([2,3-3H]di-n-propylamino)tetralin ([3H]8-OH-DPAT). NAD-299 behaved as a silent antagonist in [35S]GTPγS binding similar to N-tert-butyl-3-(4-(2-methoxyphenyl)-piperazin-1-yl)-2-phenylpropanamide (WAY-100635) and (S)-5-fluoro-8-hydroxy-2-(di-n-propylamino)tetralin ((S)UH-301). 5-HT and 5-carboxamidotryptamine (5-CT) stimulated [35S]GTPγS binding 2.5-fold while spiperone and methiothepin inhibited [35S]GTPγS binding 1.4-fold. Furthermore, NAD-299 antagonised both the 5-HT stimulated and the spiperone inhibited [35S]GTPγS binding to basal levels. The KiL/KiH ratios for spiperone (0.66), methiothepin (0.39), WAY-100635 (0.32), (S)UH-301 (0.94), NAD-299 (1.29), NAN-190 (1.23), (S)pindolol (5.85), ipsapirone (13.1), buspirone (24.6), (±)8-OH-DPAT (47.3), flesinoxan (55.8), 5-HT (200) and 5-CT (389) correlated highly significantly with the intrinsic activity obtained with [35S] GTPγS (r = 0.97).  相似文献

In aqueous solutions, 13C- and 1H-nmr studies show that the percentage of trans conformation of proline oligomers +H2H Pro-(Pro)n-CO increases substantially from n = 1 (65% trans) to n = 2 (90% trans). The relatively low percentage of trans structure for the dimer (n = 1) very likely is caused by the extra stability acquired by the end-to-end intramolecular H-bonding of the cis dimer. As n increase from 2 to 3 (or 5) in +H2N-Pro-(Pro)n-CO, the percentage of trans conformation stays more or less constant (~0.9). A high salt concentration (4M CaCl2) causes a conformation randomization, so that the short-chain oligomer (n = 1, 3, 5) and the long-chain poly (L -proline) all show about the same frantion of trans conformation (0.7-0.8).  相似文献

A cell-free preparation from Streptomyces UC5319 has been developed which catalyzes the conversion of trans, trans-farnesyl pyrophosphate (1) to pentalenene (2), the parent hydrocarbon of the pentalenolactone family of sesquiterpene antibiotics. Incubation of [9-3H2, 12,13-14C]farnesyl pyrophosphate with the pentalenene synthetase gave [1,8-3H2, 14,15-14C]pentalenene, as established by a combination of chemical and microbial degradation methods. The retention of both equivalents of tritium in the enzymatically derived pentalenene establishes that the cyclization of trans, trans-farnesyl pyrophosphate to 2 is catalyzed by a single enzyme.  相似文献

We synthesized several novel 2-O- or 11-O-substituted N-alkylnoraporphines and assessed their affinities at dopamine D1 and D2, and serotonin 5-HT1A receptors in rat forebrain tissue. Tested compounds displayed moderate to high affinities to D2 receptors but low affinities to D1 and 5HT1A receptors. The findings accord with previous evidence of a lipophilic cavity on the surface of the D2 receptor to accommodate N-alkyl moieties of aporphines. The most D2-potent (Ki = 97 nM) and selective novel agent (>100-fold vs. D1 and 5-HT1A sites) was R(−)-2-(2-hydroxyethoxy)-11-hydroxy-N-n-propylnoraporphine (compound 11).  相似文献

The enantioselectivity potential of five strains of Saccharomyces cerevisiae was studied for the reduction of ethyl N-{2-{4-[(2-oxocyclohexyl)methyl]phenoxy}ethyl} carbamate (1), an insect juvenile hormone bioanalog. The products of the reaction, the cis and trans isomers of ethyl N-{2-{4-[(2-hydroxycyclohexyl)methyl]phenoxy}ethyl} carbamate (2 and 3), were obtained in 45–49% (w/w) chemical yields and with 79 to > 99% enantiomeric purity values. The absolute configurations of the major products were assigned as ethyl (1S,2S)-N-{2-{4-[(2-hydroxycyclohexyl)methyl]phenoxy}ethyl} carbamate (2) and ethyl (1S,2R)-N-{2-{4-[(2-hydroxycyclohexyl)methyl]phenoxy}ethyl} carbamate (3). The products 2 and 3 belong to the series of the chiral insect juvenile hormone analogs.  相似文献

Synthesis of (±)-trans-chrysanthemic acid from (±)-1′-hydroxydihydro-trans-chrysanthemic acid by the dehydration with p-toluene-sulfonic acid was attempted. However, the attempt was found to be unsuccessful giving a compound believed to be methyl methyl 2,6 dimethylhepta-3.6-diene-5-carboxylate upon dehydration.

A cleavage upon cyclopropane ring was confirmed by deriving the acid obtained by the hydrolysis of the above ester to already known 2,6-dimethyl-heptane-5-carboxylic acid.

Analogous mode of dehydration and cleavage upon the ester of (±)-2,2-dimethyl-3-trans-hydroxylbenzyl-cyclopropane-l-carboxylic acid was also observed to give 1-phenyl-4-methyl-penta-1,3-diene-3-carboxylic acid. On the other hand, (±)-trans-caronic acid being derived to (±)-1′-oxo-2′-hydroxy-dihydro-trans-chrysanthemic acid, the synthesis of (±)-trans-chrysanthemic acid from (±)-trans-caronic acid became possible using (±)-1′-oxo-2′-hydroxy-dihydro-trans-chrysanthemic acid as a relay substance.  相似文献

Polarized Raman spectra have been obtained from single microcrystals of the duplex of the decamer d(A5T5)2 using a Raman microscope. This is the first report of Raman spectra from a crystal of a deoxyoligomer that contains only long, nonalternating sequences of adenine and thymine. Sequences containing d(A)n and d(T)n are of interest in view of recent suggestions that they induce bends in DNA and that they might exist in a nonstandard B-conformation. Polarized Raman spectra of a crystal of d(pTpT) have also been obtained. Both crystals display Raman bands whose intensities are very sensitive to the orientation of the crystal with respect to the direction of polarization of the incident laser beam. These spectra indicate that the helical axes of the oligonucleotides are parallel to the long axes of the crystals and that the d(A5T5)2 is not appreciably bent in the crystal. The Raman spectrum from the d(pTpT) crystal indicates that all of the furanose ring puckers are in a C2′-endo configuration since only the C2′-endo marker band at 835 ± 5 cm?1 is present. Crystals of d(A5T5)2 show measurable Raman intensities in both the 838- and 816-cm?1 bands. This indicates the presence of both the C2′-endo and C3′-endo, or possibly other non-C2′-endo, furanose conformations. The 816-cm?1 band is weak so that only a small fraction of the residues are estimated to be in the non-C2′-endo conformation. In both the d(pTpT) and d(A5T5)2 crystals the intensity of the bands due to vibrations of the backbone show only a small dependence on orientation of the crystals. This result is explained by the low symmetry of the puckered sugar rings. It is concluded that Raman spectra obtained from oligonucleotide crystals in which the orientation of the crystal axes to the laser polarization is not carefully controlled may contain intensity artifacts that are due to polarization effects.  相似文献

Eight fructo-oligosaccharides were isolated from purified oligosaccharide fractions of the roots of Asparagus officinalis L. (Liliaceae). By examination of constituent sugars, gas-liquid-chromatographic analysis of methyl derivatives, and investigation of partial acid hydrolyzates and products of β-fructofuranosidase action, they were confirmed to be 1F(1-β -fructofuranosyl)n sucrose [n = 1 (1-kestose), 2 (nystose), and 3], 6G (1-β-fructofuranosyl)n sucrose [n = 1 (neokestose), 2, and 3], 1F,6G-di-β-fructofuranosyl sucrose, and a new pentasaccharide 1F (1-β-fructofuranosyl)2-6G-β-fructofuranosyl sucrose.  相似文献

Cyclo(L -Pro-Sar)n (n = 2–4) with moderate flexibility and hydrophobicity of molecular structure was synthesized, and the characteristics of these cyclic peptides and their metal complexes in acetonitrile were investigated in connection with the residual properties using 13C-nmr measurements. The cyclic tetrapeptide cyclo(L -Pro-Sar)2 showed a sterically hindered phenomenon in acetonitrile in which the amide backbone adopted a cis-trans-cis-trans sequence. The cyclic hexapeptide cyclo(L -Pro-Sar)3 existed as a mixture of several conformers whose interconversion is slow on the nmr time scale, including cis-cis-trans and/or cis-trans-trans arrangement of the Sar-Pro bond. Finally, it was demonstrated that the cyclic octapeptide cyclo(L -Pro-Sar)4 behaved as a mixture of multiple conformers which allowed for cis-trans isomerism about the Pro-Sar peptide bond, of which 20–30% had the all-cis Sar-Pro bond isomer and the remaining 70–80% had one (or more) cis Sar-Pro bond isomer. 13C-nmr spectra also demonstrated that cyclo(L -Pro-Sar)n (n = 3,4) formed a 1:1 ion complex whose conformation was characterized by an all-trans peptide bond in the presence of excess metal salt. Cation binding studies, using CD measurements, established that the ion selectivity of cyclo(L -Pro-Sar)4 in acetonitrile decreased in the order, Ba2+ > Ca2+ > Na+ > Mg2+ > Li+.  相似文献

In connection with our investigation on the structure-activity relationships of the American cockroach sex pheromone mimic, (+)-trans-verbenyl acetate, a number of alcohols having 6,6-dimethylbicyclo[3, 1. l]heptane skeleton were synthesized, and measured their PMR spectra in the presence of the shift reagent, Eu(dpm)3. The induced shifts of the 6, 6-gem-dimethyl protons and α-proton at the C–7 guided us to distinction between cis- and trans-alcohol. The configuration of secondary methyl groups was discussed from the induced shift values of the methyl protons.  相似文献
