许多不皂化物成分作为药效成分被应用于恶性肿瘤的临床治疗中,五谷虫不皂化物的提取分离可以充分发挥这种传统中药材的功效。通过单因素实验得到不皂化物酸值较低而产率较高的最佳工艺条件是石油醚添加量为10mL/g,碱添加过量5%,在60℃的条件下反应15min,0.5mol/L的KCl溶液添加量为50mL/g。通过响应面优化后,得到最佳的不皂化物酸值和提取率分别为70.21mg KOH/g和58.05%。  相似文献

Six different solvents namely: methanol, ethanol, water, acetone, pet-ether or n-hexane was used to extract the essential oils from cashew kernel. The extracted oils were evaluated on Sitophilus zeamais (Motschulsky 1855) for mortality, oviposition and adult emergence effects. The long-term storage and water absorption capacity of the treated maize grains was also investigated. Results showed that the steam distillate was most lethal on S. zeamais. The least potent extracts on S. zeamais were those extracted with methanol and ethanol. The extracts drastically reduced oviposition in S. zeamais. Maize grains treated with steam distillate extract had the lowest water absorption capacity, while the highest water absorption capacity was obtained in grains treated with ethanol and methanol. Extracts of steam distillate, n-hexane, petroleum ether and acetone completely prevented infestation and subsequence damage of the treated maize grains for a period of three months. In the grains treated with the methanol and ethanol extracts, damage of 28.61% and 6.24%, respectively were obtained. The results obtained reveal that extracts of cashew kernel are effective in controlling S. zeamais and could serve as an alternative to synthetic insecticides.  相似文献

Cheese wood, Alstonia boonei De Wild stem bark was extracted with five different solvents namely: methanol, ethanol, acetone, petroleum ether and n-hexane. The extracts were tested for insecticidal activity on cowpea bruchid, Callosobruchus maculatus (Fab.). Adult mortality and adult emergence of the insects were investigated. Oviposition deterrences using dual-choice and multiple-choice tests were also investigated as well as percentage damage and weight loss at temperature of 28?±?2?°C and 75?±?5% relative humidity. Results showed that at the rate of 2% extract per 20?g of cowpea seeds, methanol, petroleum ether and n-hexane extracts caused 100% mortality of adult cowpea bruchid after four?days of post treatment. When the bruchid had choice of oviposition substrate, they laid significantly (p?<?0.05) fewer eggs on cowpea seeds treated with extracts compared to untreated seeds. There was no adult emergence in seeds treated with methanol, ethanol, petroleum ether and n-hexane extracts and 100% reduction in F1 progeny was recorded compared with untreated that had 81.86% adults emergence and 0% reduction in F1 progeny. The results obtained from this study revealed that methanol, ethanol, acetone, petroleum ether and n-hexane extracts of A. boonei stem bark were effective in controlling C. maculatus and could serve as an alternative to synthetic insecticides for the protection of stored cowpeas against bruchids.  相似文献

Propolis has been used in traditional folk medicine for ages owing to a number of biological effects. Four propolis samples of Czech and one of Slovak origin were extracted using Soxhlet apparatus and analysed by thin-layer chromatography. Raw propolis samples and their extracts were tested by microdilution broth method to determine minimal inhibitory concentration (MIC) in eight strains of human pathogenic fungi. Raw propolis samples showed a lower in vitro antifungal activity than their extracts. In general, the petroleum ether extracts exhibited the highest in vitro antifungal activity (MIC range of 16–64 μg/ml). The content of flavonoids in the samples varied according to region. The highest amount of flavonoids was found in sample A that originated from Broumov (4%). The most susceptible to the propolis extracts were Trichophyton mentagrophytes and Candida albicans. The propolis samples of Czech and Slovak origin and their extracts showed a considerable in vitro antifungal effect which was associated especially with nonpolar petroleum ether and toluene extracts. There was only a partial correlation between flavonoids content and in vitro antifungal activity.  相似文献

Three new compounds, 1 – 3 , and 20 known compounds were isolated from the AcOEt and BuOH extract of edible Opuntia Milpa Alta. The petroleum ether extract was examined by GC and MS. A total of 26 compounds were identified, representing 95.6% of the total extract, phytosterol (36.03%) being the most abundant component, and polyunsaturated fatty acids (18.57%) represented the second largest group, followed by phytol (12.28%), palmitic acid, palmitate (13.54%), vitamin E (4.51%), and other compounds (7.47%). The effects of various extracts from edible Opuntia Milpa Alta (petroleum ether extract, AcOEt extract, BuOH extract, aqueous extract, H2O parts) and the positive control (received dimethylbiguanide) were tested on streptozotocin (STZ)‐induced diabetic mice. The results indicated that all the treatment groups could significantly decrease blood glucose levels in STZ‐induced diabetic mice compared to the model control group (P<0.01), except the aqueous extract group (P<0.05). Especially, the petroleum ether extract group and the positive control group showed remarkable decrease of blood glucose levels. Taken together, the results indicate that the petroleum ether extract is the major hypoglycemic part in edible Opuntia Milpa Alta, which may be developed to a potential natural hypoglycemic functional ingredient.  相似文献

Equimolar aqueous solutions of d-glucose and n-butylamine were heated at 95°C for various times at pH 4, 6.5 and 11.48. The resulting brown solutions were extracted with ether. The volatile components in the ether extracts were analyzed by gas chromatography and gas chromatography-mass spectrometry, with a fused silica capillary column. The major components formed were identified as three alcohols, four N-butylpyrroles, N-butylacetamide, N-butylformamide, N-butylsuccinimide, one pyranone and 5-(hydroxymethyl)-2-furfural. In addition, eleven minor components were also identified. The relative amount of each component changed markedly with pH. At pH 4.0, higher-boiling heterocyclic compounds without C-C fission of glucose were largely formed, and at pH 11.48, lower-boiling fission compounds were mainly formed. Cyanobacterial cultures tolerating 200 mmol l−1 sodium chloride isolated from terrestrial and freshwater habitats of North Maharashtra region of India were evaluated for antifungal activity. Aqueous, methanol, n-propanol, and petroleum ether extracts of 40 cyanobacterial isolates belonging to nine genera were examined for inhibitory activity against five fungal pathogens. Eighteen isolates belonging to genus Oscillatoria dominated the population of halotolerant cyanobacterial cultures. Four antifungal bioassays viz. double layer agar method, disc diffusion assay, silica gel method, and minimum inhibitory concentration (MIC) were used to screen the cultures for antifungal activity. Among the solvents used, methanol extracts showed 34.9% inhibition followed by n-propanol, petroleum ether, and water exhibiting 30.2%, 18.6% and 16.2% inhibition, respectively. The double agar layer method was found to be a suitable method in preliminary screening for handling large number of cultures without extraction of compounds. However, in later screening experiments, silica gel method was seen to be advantageous over MIC and agar disc diffusion methods.  相似文献

The composition of the unsaponifiable matter of the lipids of six Adansonia species (A. grandidieri, A. za, A. fony, A. madagascariensis, A. digitata and A. suarezensis) was investigated. The total unsaponifiable content, its general composition and the identity of the components of the hydrocarbon, sterol and tocopherol fractions are presented. The unsaponifiable content in oil ranges from 0.4 to 1.1% (hexane method) and from 0.6 to 2.2% (diethyl ether method). In two species (A. grandidieri and A. suarezensis) the major components are 4-demethylsterols (23–42%) tocopherols (37-10%) and hydrocarbons (15–17%). In both species examined, eight 4-demethylsterols occur in the sterol fraction with sitosterol (81–88%) being predominant. Among the four tocopherols present, γ-tocopherol (68–98%) is the major compound. Each Adansonia species shows a characteristic gas liquid chromatography pattern for the hydrocarbon fraction. Squalene is the major component for five species (40–75%). Iso-, anteiso- and other branched hydrocarbons were not identified but were present in small amounts in comparison with n-alkanes. The dominance of odd- over even-carbon number chain length of n-alkanes was not observed in any species. The results show that C22, C25, C26, C27, C28 and C29 are the most frequent major constituents.  相似文献

Pithecellobium dulce has been used in traditional medicine to treat various ailments owing to its restorative properties. The biological activities and chemical profiles of the lipophilic fraction of P. dulce bark and leaves were assessed herein. Fatty acid methyl esters (FAME) and unsaponifiable matter (USM) were prepared and analyzed by GC/MS. A total of 40 compounds were identified in the bark saponifiable fraction, whereas 9 compounds were annotated in the leaves. Palmitic acid methyl ester was the major compound identified accounting for 41.48 % of the bark and 19.03 % of the leaves composition. Besides, linolenic acid methyl ester (22.40 %) and linoleic acid (12.69 %) were annotated in the leaves saponifiable fraction. A total of 63 compounds were detected in the bark USM and 4 compounds were identified in the leaves. Phytol represented the major component in the leaves (52.57 %) followed by lupeol (20.68 %) and lupenone (8.60 %). Meanwhile, n‐dodecane dominated in the bark USM accounting for 24.69 % of the total composition. The leaves and bark lipophilic fractions revealed moderate antioxidant and antibacterial activities. Both extracts showed no antifungal activity. No cytotoxicity was observed for both lipophilic fractions. P. dulce offers a good source of antioxidant compounds that can be introduced to food and pharmaceutical industry.  相似文献

Hyoscyamus species is one of the four plants used in Ayurveda for the treatment of Parkinson’s disease (PD). Since Hyoscyamus niger was found to contain negligible levels of L-DOPA, we evaluated neuroprotective potential, if any, of characterized petroleum ether and aqueous methanol extracts of its seeds in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) model of PD in mice. Air dried authenticated H. niger seeds were sequentially extracted using petroleum ether and aqueous methanol and were characterized employing HPLC-electrochemistry and LCMS. Parkinsonian mice were treated daily twice with the extracts (125–500 mg/kg, p.o.) for two days and motor functions and striatal dopamine levels were assayed. Administration of the aqueous methanol extract (containing 0.03% w/w of L-DOPA), but not petroleum ether extract, significantly attenuated motor disabilities (akinesia, catalepsy and reduced swim score) and striatal dopamine loss in MPTP treated mice. Since the extract caused significant inhibition of monoamine oxidase activity and attenuated 1-methyl-4-phenyl pyridinium (MPP+)-induced hydroxyl radical (·OH) generation in isolated mitochondria, it is possible that the methanolic extract of Hyoscyamus niger seeds protects against parkinsonism in mice by means of its ability to inhibit increased ·OH generated in the mitochondria.  相似文献

Acaricidal and ovicidal activities of Clerodendrum viscosum Ventenat (Verbenaceae), a common weed of India, were investigated on tea red spider mite, Oligonychus coffeae Nietner (Acarina: Tetranychidae). Different solvent extracts (water, methanol, acetone and petroleum ether) of C. viscosum at different concentrations (1, 2, 4 and 8%) were used. These solvent extracts exhibited mortality of O. coffeae in the range of 40–90% in water, 67–97% in petroleum ether, 50–80% in acetone and 43–87% in methanol extract. Depending on LC50 values, the relative toxicity was found to be significantly highest against petroleum ether extracts and lowest in water extracts. Acetone extract of C.viscosum recorded maximum ovicidal activity followed by petroleum ether, methanol and the less by water extracts. In the field trials, mite population were significantly lower (P < 0.05) on plots sprayed with different of C. viscosum extracts than the control and at par with chemical acaricide and neem biopesticide. No phytotoxic effect (score, 0–5% and grade 1) was observed in the field from tea bushes sprayed with different doses of extracts of C. viscosum. Made tea samples were taint free. Organoleptic test revealed leaf infusions and liquor strength as good, scoring 6.5–7.0 on a 10-point scale. Availability and distribution of this weed (C. viscosum) in and around tea growing areas of sub-Himalayan region, along with its processing for the feasibility of including C.visosum extracts in the current IPM programme are discussed.  相似文献

The growing demand of medicinal plants is posing a major threat to biodiversity conservation. Sclerocarya birrea and Harpephyllum caffrum (Anacardiaceae) are traditionally used to treat numerous ailments increasing their susceptibility to overexploitation. The aim of the study was to evaluate the medicinal efficacy of renewable aerial parts of the plants using in vitro assays, thus providing a conservation measure by plant part substitution. Plant samples were sequentially extracted with petroleum ether, dichloromethane and 80% ethanol, sonicated on ice and concentrated under vacuum. Antimicrobial activities were determined by the minimum inhibition concentration (MIC) using the micro-dilution assay. The ability of extracts to inhibit COX-1 and COX-2 enzymes was used to evaluate anti-inflammatory activity. Ethanolic extracts of S. birrea and H. caffrum exhibited high antibacterial activity (MIC < 1.0 mg/ml). S. birrea twig extract was the most active with a total activity of 1609.1 ml/g against Bacillus subtilis and an MIC of 0.098 mg/ml. Petroleum ether and dichloromethane extracts exhibited high COX-1 (90.7–99.8%) and COX-2 (69–92.6%) enzyme inhibition at a concentration of 250 μg/ml. The extracts of S. birrea and H. caffrum exhibited high antimicrobial and anti-inflammatory properties. Based on these results, plant part substitution can be a practical conservation strategy for the two species.  相似文献

Six plant crude extracts were chosen to evaluate their acaricidal activity against Tetranychus cinnabarinus adults. The crude extract from the stems and leaves of Arachis hypogaea L. presented the highest activity in leaf‐dip bioassays, with an LC50 value of 3,545.98 mg/L. Further extraction using four different solvents (petroleum ether, ethyl acetate, n‐butyl alcohol and distilled water) demonstrated that the active components mainly existed in the petroleum ether phase. Then, the active compound, palmitic acid, was isolated from the petroleum ether phase via two‐step column chromatography using columns filled with silica gel and C‐18 and identified via mass spectrometry and nuclear magnetic resonance analyses. The LC50 value of active palmitic acid against T. cinnabarinus was 534.58 mg/L. The present study demonstrated that the active compound extracted from A. hypogaea is a potential novel botanical acaricide for controlling T. cinnabarinus.  相似文献

Introduction – The sclerotia of Inonotus obliquus (Chaga) are effective therapeutic agents to treat several human malignant tumours and other diseases without unacceptable toxic side‐effects. Objective – To investigate solvent effects on metabolic profiles and antioxidant activities of extracts of Chaga. Methodology – Chaga was extracted by petroleum ether, chloroform, ethyl acetate, acetone, ethanol and water. Solvent effects on metabolites in the extracts were assayed by NMR‐based metabolomic analysis. Antioxidant activities were indicated as capacities for scavenging superoxide anion, DPPH and hydroxyl radicals. Results – Petroleum ether and chloroform extracts contained primarily lanostane‐type triterpenoids (LT), whereas the extracts of ethyl acetate, acetone and ethanol were characterised by the predominant presence of hispidin analogues and LT, and water extracts by polysaccharides and phenolic compounds. The ethyl acetate, acetone, ethanol and water extracts revealed remarkable potential for scavenging the tested radicals, while those of petroleum ether and chloroform did not. Polyphenols are the major contributors for quenching the tested free radicals, while in LT only compounds 16 , 17 and 22 participated in scavenging hydroxyl radicals. Conclusion – Polyphenols in Chaga are the principles for quenching free radicals while polysaccharides and a few LT compounds contribute partially in scavenging DPPH and hydroxyl radicals, respectively. NMR‐based metabolomic analysis is a useful method by which to correlate 1H‐NMR spectra of Chaga extracts with their antioxidant activities, and this allows the prediction of potentials for scavenging free radicals by 1H‐NMR spectroscopy. Egg masses laid by Spodoptera littoralis mated female moths were extracted by petroleum ether (PE), ethanol (E) and Ringer's solution (RS). Egg-wash extracts were evaporated and the weights of crude materials were obtained. Different aqueous concentrations were made. The amount of extracted material increased as the weight of eggs used increased and vice versa. Coating Nerium oleander leaves with aqueous egg-wash extracts prepared from S. littoralis egg-masses deterred the mated conspecific female moths from ovipositing their eggs on treated leaves, as well as causing a decrease in the total number of deposited eggs per female during the moth's life span. The highest deterrent effect on conspecific female moths to oviposit their eggs was obtained after treatment of N. oleander leaves with PE or E egg-wash extract. The deterrent effects of the tested egg-wash extracts was concentration dependant; an increase in the concentration of any extract caused an obvious decrease in the number of deposited egg-masses and the total number of laid eggs on the treated N. oleander leaves.  相似文献

Abstract

The biological effect of plant extracts on the two predators C. carnea and C. undecimpunctata was studied in the laboratory. Treated B. brassicae was offered to the newly hatched larvae of the two predators. The rate of prey consumption and duration of the larval instars of the two predators were slightly affected as a result of the treatments of the plant extracts. Adults of the aphid parasite D. Rapae and the whitefly parasite E. Mundus were exposed to the plant extracts via feeding on treated honey droplets or parasitized treated hosts. Mortality percentages of D. Rapae fed on honey contaminated with plant extracts were 50 and 80% for extracts of Melia fruits in ethanol and in petroleum ether, respectively. Mortalities in parasites developed in hosts treated with plant extracts of Neem fruits in ethanol and Melia flowers in petroleum ether in were 33.3 and 81.5%, respectively. Similar results were obtained when E. Mundus received the same treatments of the plant extracts.  相似文献

采用液体摇瓶法培养草菇菌丝体,并用不同有机溶剂对培养液及菌丝体中的代谢成分分别进行分离提取,获得了不同来源的次生代谢提取物。对各提取物的成分分析表明,石油醚相、乙酸乙酯相和乙醇相提取物中均含有粗三萜和黄酮类物质,但菌丝体提取物中粗三萜和黄酮含量明显高于培养液提取物中的含量。石油醚抽提菌丝体获得的提取物中粗三萜含量最高,达17%,而乙酸乙酯抽提菌丝体获得的提取物中黄酮含量最高,达9.31%。抗氧化活性检测结果显示,石油醚相、乙酸乙酯相、乙醇相中的代谢提取物均有较高的抗氧化活性,但乙酸乙酯相提取物的抗氧化活性明显高于石油醚相提取物,具最高抗氧化活性的提取物分别来自乙酸乙酯、乙醇对菌丝体的抽提物。MTT法检测各提取物对胃癌细胞增殖的抑制作用,结果显示各组分均有较高的抗肿瘤活性,且抗肿瘤活性与提取物浓度存在明显的量效关系。  相似文献

Abstract

The potency of Piper nigrum seed and leaf, Aframomum meleguata seed and Ageratum conyzoides leaf extracts in the control of cassava tuber rot caused by Rhizopus oryzae was investigated. Water, ethanol and petroleum ether were used as extracting solvents. These extracts were fungitoxic both in vitro and in vivo against the test pathogen. P. nigrum seed extracts were the best followed by those of A. meleguata seed, A. conyzoides leaf and then P. nigrum leaf. The extracts were more effective in controlling rot development in unwounded than wounded tubers especially when they were applied before inoculation with the test pathogen. Ethanol extracts gave the highest growth inhibition in vivo followed by water and the petroleum ether extract whereas water extracts showed marked superiority over the extracting solvents in checking rot development in cassava tuber. Water and ethanol extracts of P. nigrum seed and leaf, A. meleguata seed and A. conyzoides leaf could be used as pesticide of plant origin in the control of R. oryzae causing cassava tuber rot in storage.  相似文献

Model-experiments with kinetin, 6-furfuryl-amino-purine, to determine the quantitative yield in purification of cytokinin extracts have been performed. If kinetin in acidic water solution is partitioned three times with equal volumes of ethyl ether, about 50 per cent of the kinetin passes into the ether phases. In similar experiments with ethyl acetate more than 90 per cent of the kinetin goes over to the ethyl acetate phases. Accordingly, use of these two solvents in purification of cytokinin extracts leads to very large losses. Use of petroleum ether or n-hexane on the other hand leads to none or very small losses. Partitioning of alkaline kinetin solutions three times with equal volumes of 1-butanol results in an almost quantitatitve extraction of the kinetin from the water solution. About 7 per cent of the original amount of kinetin follows the acid auxin, when a saturated ether solution of kinetin is extracted according to a common method for acid auxins. Kinetin may thus interfere in later analyses for auxins. The dangers involved when cytokinins are “purified” according to normal practice are pointed out.  相似文献

Petroleum ether, acetone, water and aqueous extract of Pedalium murex L. (Pedalaceae) leaves, root and fruit were tested against fourth instar larvae of the tobacco cutworm, Spodoptera litura (Fabr.) under laboratory conditions using leaf dip method. Larval mortality; larval, pupal and adult periods; pupal weight, pupation and adult emergence; larval and pupal deformities of S. litura were recorded. All the tested solvent extracts of P. murex were effective against S. litura life stages by causing mortality in a dose dependent manner. However, the efficacy was more significant with respect to acetone (leaf and root) and petroleum ether (root) at higher concentrations (0.8%) which leads to 100% larval mortality. The water and aqueous extracts of root caused 86.6 and 88.0% larval mortality respectively at 4% concentration. Among the plant parts tested, root had more importance followed by leaf and fruit. Plant extracts extended the larval, pupal and adult periods; reduced the pupal weight, pupal and adult emergence and caused larval–pupal deformities. Total life time was highly prolonged in acetone extracts of P. murex root. Though all the extracts were found to have insecticide activity, acetone and petroleum ether extracts of P. murex root can be used as an effective alternative to modern synthetic insecticides. Bioactive principles from these extracts can be isolated, identified and integrated in S. litura management.  相似文献
