Feedlot waste contains essentially all the necessary nutrients for batch fermentation with the fungus Trichoderma viride. The organism utilizes two-thirds of the carbohydrate in feedlot waste while elaborating cellulase in quantities comparable to commercial preparations. Essentially odor-free, the fermented waste contains all of the original nitrogen but has 24% less organic matter.  相似文献

In order to have increased extracellular production of cellulase by Trichoderma viride ITCC 1433, the organism was grown on various growth factors. Cellulose Powder ?123 was found to be the best C-source while amongst raw materials, alkali-treated rice straw gave the best yield. A combination of peptone, urea and ammonium sulphate gave better production of cellulase than when a single nitrogen source was used. Sugars when added into the cellulose medium, generally suppressed the yield. When the organism was grown on sugars as the sole source of carbon, only lactose and maltose induced any cellulase production. Acetate and ascorbate were conspicuous in increasing cellulase production and when given together they had a cummulative effect and the yeild was doubled.  相似文献

Cellulase was produced by Trichoderma viride in semisolid cultures of rice bran, rice straw and rice hulls. T. viride QM 9414 generally produced higher cellulolytic activity on CM-cellulose (Cx activity) using rice bran-rice hull mixture (2:1 w/w) as substrate compared to strains ITCC 1433 and D 4014. It showed higher Cx activity on rice bran-rice straw mixtures than on rice bran-rice hull mixtures. Maximal extraction of the enzyme from mold bran was obtained with 0.05 m sodium citrate buffer, pH 3.5.  相似文献

利用筛选的绿色木霉ZY-1(Trichoderma viride ZY-1)固态发酵产纤维素酶,采用稻草和麸皮为底物,考察稻草与麸皮比例随发酵时间对产酶的影响。结果表明:底物中,在m(稻草):m(麸皮)为0:5和1:4时,发酵48h,pH保持4.5左右,还原糖量急剧上升,胞外蛋白产量最低;仅以稻草作底物时,整个发酵过程中pH约为7,还原糖量最低,胞外蛋白产量较高而滤纸酶活、羧甲基纤维素酶(CMCase)和β-葡萄糖苷酶(β-Gase)酶活均较低;在m(稻草):m(麸皮)为3:2时,发酵96h,滤纸酶活达最大值5.01U/g干曲;m(稻草):m(麸皮)为1:4时,发酵96h,β-Gase酶活达最大值4.6U/g干曲;m(稻草):m(麸皮)为4:1时,发酵72h,CMCase酶活达最大值6.01U/g干曲。因此,底物中存在适量的稻草和麸皮有利于Trichoderma viride ZY—1产纤维素酶。  相似文献

A plate clearing assay was devised to screen for high-producing cellulase mutants of Trichoderma viride. The method employs (i) the use of either rose bengal or oxgall to limit colony size and (ii) Phosfon D (tributyl-2, 4-dichloroben-zylphosphonium chloride) to enhance cellulase detection, in combination with acid-swollen cellulose on agar plates. The method was used to isolate constitutive cellulase mutants of T. viride and should prove useful for isolating high-producing mutants from a range of organisms. This technique has been also used to determine the concentration at which glucose and glycerol inhibit cellulase synthesis by catabolite repression in the wild-type strains.  相似文献

A series of 4-alkylthio-1,2-dithiolanes were synthesized from S,S′-(2-alkylthiotrimethylene) di(benzenethiosulfonates), and their biological activities were tested on Culex pipience molestus, Laodelphax striatellus and Tetranychus urticae. These compounds showed potent activity against both insect species, the strongest being displayed by 4-ethylthio-1,2-dithiolane and 4-isopropylthio-1,2-dithiolane.  相似文献

绿色木霉(Trichodermaviride)在pH控制发酵条件下,采用流加葡萄糖发酵策略,可显著提高综合滤纸酶活力(FPA)和内切酶(endo—β—1,4-glucanase,EG)、外切酶exo—β-1,4-glucanase,CBH)、纤维二糖酶(cellobiase,CB)酶活。在5L发酵罐中采用pH控制和流加葡萄糖工艺,可提高CB酶含量,改变酶组分之间的比例,使得FPA、EG、CB和CBH酶活分别达到50.0U/mL,210.0U/mL,4.0U/mL和2.5U/mL,比摇瓶发酵分别提高了6.7.4.2、19、2.5倍。  相似文献

利用纤维素酶高产菌绿色木霉Trichoderma viride降解木质纤维素是实现废料资源化的重要手段。本研究选取来自不同生境的两株T. viride,分别以玉米秸秆和甘草药渣为基质,测定两者滤纸纤维素酶(filter paper cellulase,FPase)活性和还原糖产量。从时间、温度、水分、pH 4个方面比较两株T. viride的环境适应性和不同基质的差异性。结果表明,以玉米秸秆为基质,T. viride XJ最适初始料液比为1:4-1:5.5,T. viride AG最适初始料液比为1:5-1:5.5。初始料液比1:5.5时,T. viride AG产FPase活性显著高于T. viride XJ。两株T. viride最适发酵温度均为28℃,各温度处理下不同菌株间无显著差异。两株T. viride均表现为还原糖消耗。以甘草药渣为基质,T. viride XJ最适初始料液比为1:2-1:2.5,T. viride AG最适初始料液比为1:3-1:3.5。料液比高于1:3,T. viride AG产FPase活性显著高于T. viride XJ。T. viride AG最适发酵温度为28℃,T. viride XJ最适发酵温度为23-28℃。温度低于28℃,T. viride XJ产FPase活性显著高于T. viride AG。两株T. viride均表现为还原糖积累。两株T. viride最适初始pH均为6-7,最适发酵时间均为3d。最优发酵条件下FPase活性:T. viride AG>T. viride XJ。对T. viride产FPase诱导能力:甘草药渣>玉米秸秆。变差分解表明两株T. viride产FPase活性差异主要源于菌株对生境的生态适应。比较分析菌种来源、基质类型、环境条件对T. viride发酵效果的影响,将有助于该菌大规模应用性研究。  相似文献

利用绿色木霉,以稻草为唯一碳源,采用液态发酵的方法,分别加入生物表面活性剂鼠李糖脂和化学表面活性剂Tween 80,重点研究了生物表面活性剂对绿色木霉产纤维素酶的影响。实验分析了加入不同浓度的表面活性剂时滤纸酶活、羧甲基纤维素酶活、微晶纤维素酶活及酶液的表面张力随时间的变化情况。结果表明,添加鼠李糖脂能够促进绿色木霉产酶,分别使滤纸酶活、羧甲基纤维素酶活、微晶纤维素酶活最大提高了1.08倍,1.6倍和1.03倍。与Tween 80相比,鼠李糖脂促进产酶的效果明显优于Tween 80。  相似文献

Chemical modification of histidine residues in ricin E was studied with regard to saccharide binding. The analytical data indicate that 6 out of 7 histidine residues in ricin E are eventually modified with diethylpyrocarbonate (DEP) at pH 6.0 and 25°C in the absence of specific saccharides. Modification of histidine residues greatly decreased the cytoagglutinating activity of ricin E, and only 10% of the residual activity was found after modification of 6 histidine residues/mol. The data of affinity chromatography using lactamyl- and galactosamine-cellulofine columns suggest that modification of histidine residues does not have much effect on the binding ability at the low affinity saccharide-binding site of ricin E but abolishes the binding ability at the high affinity saccharide-binding site. In the presence of lactose, one histidine residue/mol was protected from the DEP modification with retention of a fairly high cytoagglutinating activity. Such a protective effect was also observed for specific saccharides such as galactose and A^-acetylgalactosamine, but not for glucose, a non-specific saccharide. On treatment with hydroxylamine, the modified ricin E restored 67 % of the cytoagglutinating activity. Based on these findings, it is suggested that in the high affinity saccharide- binding site of ricin E there exists one histidine residue responsible for saccharide binding.  相似文献

The contribution of three exoglucanases from a commercial Trichoderma viride cellulase to transcellobiosylation, and the tolerance of these enzymes to acetonitrile co-solvent were studied. The enzymatic reactions were performed with p-nitrophenyl-β-d-cellobioside as the starting substrate. Among these enzymes, the least anionic exoglucanase (Exo I) showed the highest transcellobiosylation activity and acetonitrile tolerance. Exo I retained considerable activity even in 30% MeCN/water and produced p-nitrophenyl-β-d-cellotetraoside at about 1.5% conversion from the initial substrate in 30% MeCN/water. The residual activity of Exo I after incubation in MeCN/water mixture was almost identical to that of the crude cellulase and a considerable amount of the transcellobiosylation properties of the crude cellulase seemed to be attributable to this Exo I component.  相似文献

Fermentations with mixed cultures of the cellulolytic fungus Trichoderma viride and the yeast Saccharomyces cerevisiae or Candida utiliswere examined. The fermentations were carried out in an aerated 5 liter fermentor with NaOH treated barley straw as the cellulose source (2–4%). Yeast was inoculated 24–32 hr after the fungus and the growth of the two organisms was followed through the production of CO2 and cell protein. In comparison with fermentations with T. viridealone, the production time for maximum yields of cellulases and cell protein was reduced by several days, depending on the straw concentrations. The protein content of the growth product was 21–22% and the amino acid composition of the product resembled that of T. viride alone.  相似文献

Bioaccumulation of copper by Trichoderma viride   总被引:3,自引:0,他引:3  
Studies were carried out on interaction of Trichoderma viride with copper and reports bioaccumulation as a mechanism of copper tolerance during growth. There was a marked increase in the lag phase of the growth, which was concentration dependent. At a concentration of 100 mg/L of CuCl2.2H2O, 81% of Cu(II) were removed by 3.4 g/L of the biomass in 72 h. The process was temperature and pH dependent. The maximum copper bioaccumulation occurred at 30 degrees C, pH 5.0. Metabolic inhibitors such as sodium azide (NaN3) and 2,4-dinitrophenol (2,4-DNP) drastically reduced the extent of Cu(II) bioaccumulation. Electron microscopy and cell fractionation studies revealed that 70-80% of copper was present as a layer on the cell wall surface.  相似文献
