(3H)anisomycin binding to eukaryotic ribosomes   总被引:2,自引:0,他引:2  
Anisomycin, a well-known inhibitor of eukaryotic ribosomes' peptidyl-transferase activity, specifically binds to the 60 S ribosome subunit. Quantitative studies on [3H]anisomycin binding to yeast and human tonsil ribosomes have shown that a maximum of one molecule of the antibiotic is bound per ribosome in both cases. There is a single type of binding to 60 S subunits but ribosomes themselves are not homogeneous with respect to [3H]anisomycin binding, since the interaction between antibiotic and ribosome occurs with two different affinities. Only ribosomes having the higher type of affinity for [3H]anisomycin are active in catalysing peptide bond formation, as tested in both the puromycin and the fragment reaction assays. Affinity of [3H]anisomycin for ribosomes is higher at 0 °C than at 30 °C. Affinity is decreased in the presence of ethanol.The acetate group in the 3′ position of the pyrrolidine ring of anisomycin is important for the anisomycin—ribosome interaction since deacetylanisomycin appears to have a mode of action similar to anisomycin but has an affinity for the ribosome that is 350 times smaller.The effect of certain peptidyl-transferase inhibitors has been tested on [3H]anisomycin binding to ribosomes. Using either yeast or human tonsil ribosomes a number of sesquiterpene antibiotics of the trichodermin group (trichodermin, trichodennol, fusarenon X and trichothecin) totally block [3H]anisomycin binding whereas puromycin and verrucarin A only partially inhibit the [3H]anisomycin interaction with ribosomes. Gougerotin, blasticidin S and actinobolin have no effect. Tenuazonic acid and sparsomycin inhibit [3H]anisomycin binding to ribosomes but the degree of inhibition differs between yeast and human tonsil ribosomes.  相似文献

Many strains of the hydrocabon-utilizing yeasts were isolated from various kinds of natural sources by accumulation culture.

Among those yeasts, two strains, S315YI and S131YI, which were identified with Candida tropilcais, assimilated hydrocarbons abundantly. As for type cultures, it was found that many strains of them could utilize hydrocarbons too, especially the strains which belonged to Genus Candida. However, as regards to the ability in utilizing hydrocarbons, no yeast from type culture collections utilized hydrocarbons better than the yeasts newly isolated from nature. Addition of the natural nutrients such as corn steep liquor to the cultural broth of Strain S315Y1 showed no effect on the production of yeast cells.

The yeast Strain S315Y1 assimilated the higher boiling points fraction of n-paraffins in comparison with the hydrocarbon-utilizing bacterium, Pseudomonas aeruginosa S7B1, which had been reported by the authors. Ribonucleic acid contents of dried cells of the yeast Strain S315Y1 and S131Y1 were 5.3 and 4.4% respectively by Schmidt-Thannhauser-Schneider method.  相似文献

Various conditions were studied which gave influences on polyalcohol production by Pichia miso. Pichia miso gave an excellent yield of polyalcohol showing good growth in the vitamin-free medium. Unlike Zygosaccharomyces high concentrations of phosphate such as 2% as KH2PO4 in the medium showed no detrimental effect on polyalcohol production. Remarkable reduction in polyalcohol yield was observed in the medium of high concentration of nitrogen sources, among which yeast extract showed the most striking effect. Polyalcohol fermentation was provoked in the medium of 0.1% yeast extract while the metabolic activity distinctly converted to ethanol fermentation when the organisms were incubated in the medium of 4.0% yeast extract. The fact that a large amount of ethanol more than 6% in the medium was produced aerobically by genus Pichia of oxidative dissimilation type seemed to be very interesting and noticeable. For the purpose of industrial production, it was shown that polyalcohol production in jar-fermenter scale was achieved with as good yield as that in shaking flask culture.  相似文献

The production of pigment by the molds belonging to the genus Monascus in a submerged culture was examined. The extracellular pigment was mainly studied. Monascus sp. No. 2 was found to be the most potent pigment producer. The optimum cultural conditions were: pH of the medium, 6.5; the temperature, 25°C; carbon sources, glucose or ethyl alcohol; nitrogen sources, polypeptone, yeast extract, monosodium glutamate or casamino acids. Glycine, l-threonine, l-arginine, l-alanine and l-tyrosine were found to be the most effective substances promoting pigment production.

Mycelial forms of this strain were correlated with pigment formation in submerged culture. As it grew into pellet type, the yield of pigment was at high level.

The Monascus-pigment in the fermentation liquid seemed to be firmly bound to the protein-like substances which made the pigment apparently soluble.  相似文献

The antibiotic anisomycin, an inhibitor of protein synthesis in eucaryotic cells, which blocks long-term memory in mice, is shown to interact with the cholinergic system by inhibiting reversibly the acetylcholinesterase. The inhibition is a competitive one, the inhibition constant Ki being 5.0 × 10?3 for human brain acetylcholinesterase and 1.7 × 10?3 for acetylcholinesterase of bovine erythrocytes. The anisomycin effect on acetylcholinesterase is compared with the puromycin and cycloheximide-inhibition of the enzyme. The significance of the cholinergic effect of anisomycin in addition to its inhibitory effect on protein synthesis for the interpretation of memory experiments is discussed.  相似文献

A culture was obtained from a spore print of a basidiocarp of Mycogloea nipponica collected in Taiwan. A yeast stage and basidia identical to those of M. nipponica developed in laboratory media. The Taiwanese specimen of M. nipponica and its yeast anamorph were characterised in the present study. Comparative morphological, molecular, and ultrastructural studies indicated that the yeast stage can be assigned to the genus Kurtzmanomyces. The revealed connection between the sexual species Mycogloea nipponica and the asexual genus Kurtzmanomyces demonstrates the importance of anamorphic characteristics in the modern systematics of heterobasidiomycetous fungi. Summary Dose and phase response curves for phase shifting the circadian oscillator in the dinoflagellateGonyaulax polyedra were measured with pulses of the antibiotic anisomycin (an inhibitor of protein synthesis on 80 S ribosomes), using the bioluminescent glow rhythm as the assay. The three dimensional surface of final phase, initial phase, and concentration was found to be a right handed helix, with the axis at a critical initial phase near circadian time 12 h, and critical concentration near 0.2 micromolar anisomycin (for 1 h pulses). The normally rhythmic glow of populations ofGonyaulax was significantly disrupted by pulses with these critical parameters, and in many instances appeared nearly arrhythmic.With increasing drug concentration, phase response curves appear to move bodily to earlier phases, and no saturation is evident in the phase shifting effect. These results are interpreted as indicating that anisomycin at sufficiently high doses causes an immediate strong (type 0) phase shift, then holds the clock stationary for a time interval that increases with concentration.the possibility that the 80 S ribosomal complex may be centrally involved in the fundamental circadian oscillation is put forward.Abbreviations DRC dose response curve - PRC phase response curve  相似文献

A bacterium R–4 which produces a novel type of lytic enzyme which lyses fungal and yeast cell walls was isolated from the air and was identified to belong to the genus Bacillus.

Production of the enzyme appeared to require a high concentration of nitrogen source in medium. No inducing substance was needed for the enzyme production.

A crude preparation of the enzyme was used to characterize the lytic activity. From the lytic spectrum, the enzyme seemed to have the highest activity toward the cell walls of species in the genus Rhizopus among various fungi and yeasts tested, A proteolytic activity was shown to be parallel with the lytic activity. The lytic activity was also accompanied with the liberation of reducing sugars from Rhizopus cell wall, but no activity on some known carbohydrates tested was detected in the preparation.  相似文献

The fission yeast genus Schizosaccharomyces contains important model organisms for biological research. In particular, S. pombe is a widely used model eukaryote. So far little is known about the natural and artificial habitats of species in this genus. Finding out where S. pombe and other fission yeast species occur and how they live in their habitats can promote better understanding of their biology. Here we investigate in which substrates S. pombe, S. octosporus, S. osmophilus and S. japonicus are present. To this end about 2100 samples consisting of soil, tree sap fluxes, fresh fruit, dried fruit, honey, cacao beans, molasses and other substrates were analyzed. Effective isolation methods that allow efficient isolation of the above mentioned species were developed. Based on the frequency of isolating different fission yeast species in various substrates and on extensive literature survey, conclusions are drawn on their ecology. The results suggest that the primary habitat of S. pombe and S. octosporus is honeybee honey. Both species were also frequently detected on certain dried fruit like raisins, mango or pineapple to which they could be brought by the honey bees during ripening or during drying. While S. pombe was regularly isolated from grape mash and from fermented raw cacao beans S. octosporus was never isolated from fresh fruit. The main habitat of S. osmophilus seems to be solitary bee beebread. It was rarely isolated from raisins. S. japonicus was mainly found in forest substrates although it occurs on fruit and in fruit fermentations, too.

The effects of the aminoglycoside antibiotic paromomycin on the fidelity of translation of the synthetic template poly(U), and two natural mRNAs (rabbit globin mRNA and Brome Mosaic virus RNA), were examined in an mRNA-dependent cell-free system from the yeast Saccharomyces cerevisiae. At antibiotic concentrations that did not inhibit translation (100 μM) optimal mistranslation of all three templates was observed, with the effects declining at higher antibiotic concentrations. Synthesis of the opal termination read-through protein of rabbit β-globin mRNA was induced by paromomycin, but only in lysates prepared from a [psi+] strain of yeast. The antibiotic did not induce detectable levels of either ochre or amber read-through, but did induce general misreading of Brome Mosaic virus RNA to the same degree in both [psi+] and [psi?] lysates. This misreading was enhanced by addition of the polyamine spermidine.  相似文献

【目的】开展拉鲁湿地水体酵母菌多样性研究,探究理化因子与酵母菌群落结构的相互关系。【方法】采用原位培养法从拉鲁湿地11个水样中分离酵母菌,应用26S r RNA D1/D2区域序列分析,并结合经典分类法对获得的菌株进行分类鉴定,运用SPSS和CANOCO软件分析酵母菌多样性及其与环境因子相关性。【结果】从拉鲁湿地水体中分离得到169株酵母菌,鉴定分属为15个属31个种。优势种为Ustilentyloma graminis和Filobasidium magnum,优势属为Naganishia、Ustilentyloma、Filobasidium和Cystofibasidium。统计分析结果表明,化学需氧量(COD)是影响拉鲁湿地水体酵母菌数量的显著因素,另外,此理化因子是影响Ustilentyloma分布的重要环境条件。【结论】西藏拉鲁湿地酵母菌资源比较丰富,且存在明显的空间异质性。  相似文献

Nutritional requirement of Candida lipolytica AJ 5004 and its productivity of α-ketoglutarate were further studied.

It became clear that this yeast required only thiamine as grown factor, and even if the yeast was cultured in chemically defined medium containing adequate amount of thiamine, it was able to produce as high yield of α-ketoglutarate as in the medium containing 0.02% of corn steep liquor.

It was also shown that the rate of convertion of n-paraffin to α-ketoglutarate gradually increased as the concentration of n-paraffins was decreased or as the incubation time was prolonged. A very high rate of conversion, 71%, was obtained after prolonged culture, for 5 days, with a culture medium containing 8% of n-paraffins.

The productivity of α-ketoglutarate from C9- to C20-alkanes by the yeast was maximum in the range from C15 to C19, especially from C17 to C19.  相似文献

Abstract

Tyrosine phenol lyase (TPL) is a robust biocatalyst for the production of L-dihydroxyphenylalanine (L-DOPA). The improvement of TPL production is conducive to the industrial potential. In this study, the optimization of culture medium of recombinant Escherichia coli harboring TPL from Fusobacterium nucleatum (Fn-TPL) was carried out. Sucrose and combination of yeast extract and peptone were selected as carbon and nitrogen source, respectively. Their optimal concentrations were determined by Box-Behnken design and the synergistic effect between yeast extract and peptone was found to be significant, with p-value < 0.05. The DO-STAT fed-batch fermentation under optimized culture condition was established and the oxygen level was fixed at 20%. Both the biomass and Fn-TPL activity were significantly increased, which were 35.6 g dcw/L and 12292 U/L, respectively. The results obtained significantly promote the industrial production of L-DOPA production.  相似文献

Background

In recent years, acetic acid bacteria have been shown to be frequently associated with insects, but knowledge on their biological role in the arthropod host is limited. The discovery that acetic acid bacteria of the genus Asaia are a main component of the microbiota of Anopheles stephensi makes this mosquito a useful model for studies on this novel group of symbionts. Here we present experimental results that provide a first evidence for a beneficial role of Asaia in An. stephensi.

Results

Larvae of An. stephensi at different stages were treated with rifampicin, an antibiotic effective on wild-type Asaia spp., and the effects on the larval development were evaluated. Larvae treated with the antibiotic showed a delay in the development and an asynchrony in the appearance of later instars. In larvae treated with rifampicin, but supplemented with a rifampicin-resistant mutant strain of Asaia, larval development was comparable to that of control larvae not exposed to the antibiotic. Analysis of the bacterial diversity of the three mosquito populations confirmed that the level of Asaia was strongly decreased in the antibiotic-treated larvae, since the symbiont was not detectable by PCR-DGGE (denaturing gradient gel electrophoresis), while Asaia was consistently found in insects supplemented with rifampicin plus the antibiotic-resistant mutant in the diet, and in those not exposed to the antibiotic.

Conclusions

The results here reported indicate that Asaia symbionts play a beneficial role in the normal development of An. stephensi larvae.

The utilization of hydrocarbons by microorganisms was studied in many fields, but the production of biotin vitamers by hydrocarbon-utilizing bacteria has never been reported.

We have screened many hydrocarbon-utilizing bacteria which produce biotin vitamers in the culture broth. The effects of cultural conditions on biotin vitamers production by strain 5–2, tentatively assigned to the genus Pseudomonas, were studied.

More than 98% of biotin vitamers produced from hydrocarbons by strain 5–2 was chromatographically determined as desthiobiotin. As nitrogen source, natural nutrients were more effective than inorganic nitrogen sources. The production of biotin vitamers was increased under the condition of good aeration. Exogenous pimelic or azelaic acid enhanced biotin vitamers production by strain 5–2.

The production of biotin vitamers from n-alkanes, n-alkenes or glucose by an isolated bacterium, strain 5-2, tentatively assigned to the genus Pseudomonas, was investigated. Among these carbon sources, n-undecane was the most excellent for biotin vitamers production.

The biosynthetic pathway of biotin vitamers, especially desthiobiotin, from n-undecane was also studied. It was found by thin-layer and gas-liquid chromatographical methods that pimelic and azelaic acids were the main acid components in n-undecane culture.

This result, together with previously reported enhancement of biotin vitamers production by these acids, suggests that pimelic and azelaic acids may be the intermediates of biotin vitamers biosynthesis from n-undecane.  相似文献

Summary Mutants of Methanobacterium formicicum resistant to the anti-80S ribosome-targeted inhibitor anisomycin were isolated and characterized. The resistance phenotype is correlated with a mutationally altered 50S ribosomal subunit. Anisomycin resistance in the mutants is accompanied by cross-resistance to other inhibitors of the 80S peptidyl-transferase centre like narciclasine, bruceantin, trichodermin and verrucarin A and by hypersensitivity to sparsomycin. This phenotype is identical to that reported for anisomycin-resistant mutants of yeast; it appears therefore, that the anisomycin interaction sites on the 70S ribosomes from M. formicicum bear the structural features typical of eukaryotic 80S organelles.  相似文献

Yeasts associate with numerous insects, and they can assist the metabolic processes within their hosts. Two distinct yeasts were identified by PCR within the planthopper Perkinsiella saccharicida, the vector of Fiji disease virus to sugarcane. The utility of both microbes for potential paratransgenic approaches to control Fiji leaf gall (FLG) was assessed. Phylogenetic analysis showed one of the microbes is related to yeast-like symbionts from the planthoppers: Laodelphax striatellus, Nilaparvata lugens, and Sogetella furcifera. The second yeast was a member of the Candida genus, a group that has been identified in beetles and recently described in planthoppers. Microscopy revealed the presence of yeast in the fat body of P. saccharicida. The Candida yeast was cultured, and transformation was accomplished by electroporation of Candida albicans codon optimized plasmids, designed to integrate into the genome via homologous recombination. Transgenic lines conferred resistance to the antibiotic nourseothricin and expression of green fluorescent protein was observed in a proportion of the yeast cells. Stably transformed yeast lines could not be isolated as the integrative plasmids presumably replicated within the yeast without integration into the genome. If stable transformation can be achieved, then this yeast may be useful as an agent for a paratransgenic control of FLG.  相似文献

为了从土壤环境中筛选产生抗菌活性物质的放线菌,采用琼脂扩散法对分离自湖南省高望界自然保护区森林土壤的169株放线菌进行抗菌活性检测,采用16S RNA基因序列分析法进行系统发育分析,采用常规方法进行生物学特性研究。结果表明,有47株受试菌株的发酵产物显示抗菌活性阳性(27.8%),其中6株具有较强抗菌活性。形态观察结果表明,这6株菌株均为典型的产丝产孢放线菌,在多数培养基上生长良好,气生菌丝和孢子丝丰富。系统发育分析表明,6株菌株均属于链霉菌属(Streptomyces),归为5个物种。菌株JSM 147612、JSM 147786、JSM 147831和JSM 147846分别与链霉菌属的已知物种S.violascens(序列相似性为99.93%)、S.malachitospinus(99.65%)、S.anulatus(99.72%)和S.aureus(99.38%)系统发育关系最为密切;菌株JSM 147823和JSM 147842之间的序列相似性为99.86%,应该属于同一物种,与它们系统发育关系最为密切的是S.albidoflavus(相似性分别为99.72%和99.58%)。  相似文献
