The heating of 7S globulin caused changes in the intensities, but hardly affected the positions of the peaks and troughs of the second derivative absorption spectra at wavelengths below 270 nm. On the other hand, above 271 nm, changes were reflected both in the intensities and in the positions of peaks and troughs. The difference-second derivative absorption spectra indicated that 60 and 70 percent, respectively, of phenylalanine and tyrosine residues buried in the native 7S globulin remained as the buried form even after heating.

A spectrofluorimetry and fluorescence-quenching study suggested that one residue of tryptophan in the 7S globulin tended to be transferred to the more hydrophobic interior on heating.  相似文献

A new indole-3-acetic acid metabolite was isolated from broad bean (Vicia faba L. cv Chukyo) seedlings. It was a conjugate of dioxindole-3-acetic acid, aspartic acid, and glucose and was identified as 3-(O-β-glucosyl)-2-indolone-3-acetylaspartic acid (molecular weight 484) from ultraviolet, infrared, nuclear magnetic resonance, and mass spectra. Its natural content in 4-day-old Vicia seedlings was estimated to be 8.6 nanomoles per gram fresh weight. It was suggested that oxidation of indole-3-acetic acid not accompanied by decarboxylation might regulate endogenous level of the hormone.  相似文献

An important role of protein ADP-ribosylation in bacterial morphogenesis has been proposed (J. Bacteriol. 178, 3785-3790; 178, 4935-4941). To clarify the detail of ADP-ribosylation, we identified a new kind of target protein for ADP-ribosylation in Streptomyces coelicolor A3(2) grown to the late growth phase. All four proteins (MalE, BldKB, a periplasmic protein for binding branched-chain amino-acids, and a periplasmic solute binding protein) were functionally similar and participated in the regulation of transport of metabolites or nutrients through the membrane. ADP-ribosylation was likely to occur on a cysteine residue, because the modification group was removed by mercuric chloride treatment. The modification site may be the site of lipoprotein modification necessary for protein export. This report is the first suggesting that certain proteins involved in membrane transport can be ADP-ribosylated.  相似文献

Peanut seeds (Arachis hypogea L. Yue-you 551) contain 50 to 100 nanomoles per gram conjugated 1-aminocyclopropanecarboxylic acid (ACC). Based on paper chromatography, paper electrophoresis, and gas chromatography-mass spectrometry, it was verified that the major ACC conjugate was N-malonyl-ACC (MACC). Germinating peanut seeds converted [2-14C]ACC to ethylene 70 times more efficiently than N-malonyl-[2-14C]ACC; when ACC was administered, most of it was metabolized to MACC. Germinating peanut seeds produced ethylene and converted l-[3,4-14C]methionine to ethylene; this ethylene biosynthesis was inhibited by aminoethoxyvinylglycine. These data indicate that MACC occurs in peanut seeds but does not serve as the source of ethylene during germination; ethylene is, however, synthesized from methionine via ACC.  相似文献

目的:对引进法国罗地亚EZAL-MY96直投式酸奶发酵剂(DVS)进行菌株分离鉴定。方法:利用半选择性培养基进行发酵剂菌株分离,并采用糖发酵实验和RAPD指纹图谱进行鉴定。结果:EZAL-MY96中球菌的平板菌落与光镜显微观察、糖发酵实验和RAPD指纹图谱均只有1种情况,而杆菌均有2种情况。结论:从EZAL-MY96菌种中共分离到3株菌,其中1株鉴定为嗜热链球菌,1株为德氏乳杆菌保加利亚亚种,另1株为未知的乳杆菌。  相似文献

In the experiments involving incubation of the liver, brain cortex, muscle and adipose tissues homogenates with [3-14C] tryptophan for an hour 43.2-89.3% of the label was found in proteins, 7.2-47.2%--in lipids, 2.6-9.4%--in CO2. Following incubation of the above-mentioned tissue homogenates with [2-14C] alanine, proteins, lipids and CO2 contain 28.8-49.3%; 22.6-31.9% and 21.6-49.3% of radioactive label, respectively. Radioactivity of lipids synthesized by the homogenates of the investigated tissues from [3-14C] tryptophan and [2-14C] alanine is 23.5-63.5 and 21.1-56.0%, respectively, the radioactivity of CO2 being 1.4-5.1 and 9.3-11.8% of the above-mentioned compounds synthesized from [1-14C] acetate. The results obtained testify to the considerable contribution of [3-14C] tryptophan and [2-14C] alanine to protein synthesis as well as to their involvement in the substrate supply of lipogenesis and energetic processes in various organs and tissues of cattle.  相似文献

N-Carboxymethyl-β-alanine and four γ-glutamyl peptides—γ-l-glutamyl-l-leucine, γ-l-glutamyl-l-methionine, γ-glutamylphenylalanine and γ-glutamyltyrosine—were isolated from green gram seeds. N-Carboxymethyl-β-alanine is a compound which is isolated from natural products for the first time. An amount of γ-glutamylmethionine was far more abundance than all others.  相似文献

An improved method for regenerating Bacillus subtilis protoplasts at the frequency of 92~100% on a semi-synthetic medium was found. Protoplasts were preincubated in HCP-3 medium, an isotonic semi-synthetic medium supplemented with polyvinylpyrrolidone, and then plated on HCP-1.5 agar medium by overlaying. By this method, even on the regeneration medium supplemented with minimal nutritional requirements protoplasts regenerated at a frequency of as high as 20%. The modified method was applicable to the direct-selection of prototrophic recombinants after fusion (the highest recombination yield from the input protoplasts was 1.3%) and to protoplast transformation with plasmid DNA.  相似文献

In addition to the four cytokinins, 6-(3-methyl-2-butenylamino)purine, 6-methylaminopurine and the cis and trans isomers of 6-(4-hydroxy-3-methyl-2-butenylamino)purine, reported earlier from our laboratories, three cytokinin-active fractions have been obtained from the aqueous medium of 6-day-old Corynebacterium fascians cultures. One of these has been identified as 6-(4-hydroxy-3-methyl-cis-2-butenylamino)-2-methylthiopurine (2-methylthio-cis-zeatin, c-ms2io6 Ade).  相似文献

Apple seeds soaked in a solution of [±]-[2-14C]abscisicacid for up to 70 d formed phaseate and the epimeric dihydrophaseatesand all four acids were converted into alkali-hydrolysable conjugates.This metabolism occurred in the husks and in the embryos. Bothparts of untreated control seeds contained the four acids andtheir conjugates. In addition to these characterized metabolites,a number of other, labelled, acidic products were isolated fromthe seeds. 14CO2 was evolved from unsterilized seeds but notfrom surface-sterilized ones. Bacterial cultures isolated fromsoil and rotting fruit metabolized [14C]ABA to a range of compounds,but not to phaseic or the dihydro-phaseic acids. After oral administration of AF-2 (3-(5-nitro-2-furyl)-2-(2-furyl)acrylamide) to rabbits, the two unique metabolites, M-I and M-II, were isolated from the urine. M-I, yellow needles of mp 117°, was identified as a new type metabolite of nitrofuran derivative, 2-(β-carboxypropionyl)-3-(5-methylthio-2-furyl) acrylamide by its mass, ir and nmr spectrometries. M-II, yellow solid, appears to be cis-trans isomer of M-I considering from its uv and mass spectral data, and the behavior on tlc.  相似文献

Three hydroxy-1,8-cineole glucopyranosides, (1R,2R,4S)- and (1S,2S,4R)-trans-2-hydroxy-1,8-cineole β-D-glucopyranosides, and (1R,3S,4S)-trans-3-hydroxy-1,8-cineole β-D-glucopyranoside, which are possible precursors of acetoxy-1,8-cineoles as unique aroma components, were isolated from the rhizomes of greater galangal (Alpinia galanga W.). Their structures were analyzed by FAB-MS and NMR spectrometry, and the absolute configulation of each aglycone was determined by using a GC-MS analysis with a capillary column coated with a chiral stationary phase. The composition of the diastereomers of (1R,2R,4S)- and (1S,2S,4R)- trans-2-hydroxy-1,8-cineole β-D-glucopyranosides in the rhizomes was determined as 3:7 by a GC-MS analysis after preparing the trifluoroacetate derivatives of the glucosides.  相似文献

Abstract

A new modified nucleoside, 3-(3-amino-3-carboxypropyl)-uridine was isolated from a 24 hour collection of a normal human urine. The structure was assigned on the basis of UV, NMR and mass spectrometry data and confirmed by comparison of the spectral data and HPLC mobilities with those of an authentic sample. Origin and significance of this nucleoside in relation to tRNA is discussed. The new nucleoside is present also in the urine of cancer patients but in smaller amounts.  相似文献

The seed oil content of Microula sikkimensis (Clarke) Hemsl. is up to 45% There is 8.1% of γ-linolenic acid which has the pharmacological action in the fatty acids composition, It has showed that this oil has a stronger effect on reducing triglyceride in serum. Fifteen different kinds of fatty acids were analysed. The unsaturated C20, C22, C24 acid, C18 triene-acid and tetraene-acid of the seed oil were separated on AgNO3-silica gel column and HPLC. and were identified by Periodata-Permanganate Oxidation, GLC, IR, UV, and MS. They are cis-11-eicosenoic, cis-13-docosenoic, cis-15-tetracosenoic, cis-6,9,12-octadecatrienoic and cis-6,9,12,15- octadecatetraenoic acids.  相似文献

Electrophilic additions of DL- and L-Cys to propenoic acid afforded (RS)- and (R)-2-amino-3-(2-carboxyethylthio)propanoic acids [(RS)- and (R)-ACE], respectively. (RS)-ACE was found to exist as a conglomerate based on its melting point, solubility, and infrared spectrum. (RS)-ACE was optically resolved by preferential crystallization to yield (R)- and (S)-ACE. The obtained (R)- and (S)-ACE were efficiently recrystallized from water, taking account of the solubility of (RS)-ACE, to give them in optically pure form.  相似文献

14C-labelled shikimic acid and double labelled shikimic acid tritiated stereospecifically at C-6 are incorporated into 3-(3-carboxyphenyl)alanine, 3-(3-carboxyl-4-hydroxyphenyl)alanine, phenylalanine, and tyrosine in Resda lutea L., Reseda odoratta L., Iris x Hollandica cv. Prof. Blauw, and Iris x hollandica cv. Wedgwood. The experiments with 14C-labelled shikimic acid confirm that the aromatic carboxyl groups and rings in 3-(3-carboxyphenyl)-alanine and 3-(3-carboxy-4-hydroxyphenyl)alanine derive from the carboxyl group and ring in shikimic acid whereas the experiments with double labelled shikimic acid demonstrate that the pro-6S-hydrogen atom is retained and the pro-6R-hydrogen atom lost in the biosynthesis of 3-(3-carboxyphenyl)alanine, phenylalanine, and tyrosine in the plants used. 3H was located in the ortho-position in the aromatic rings of phenylalanine and tyrosine but in a position para to the alanine side chain of 3-(3-cabroxyphenyl)alanine. No 3H was found in 3-(3-carboxy-4-hydroxyphenyl)alanine. This supports a derivation of the last two compounds from chorismic acidvia isochorismic acid, isoprephenic acid, and 3′-carboxyphenylpyruvic acid and 3′-carboxy-4′-hydroxyphenylpyruvic acid. The 3H/14 C ratio in 3-(3-carboxyphenyl)alanine was found higher than in the precursor used. This isotope effect must operate by competition between the pathways from isoprephenic acid to 3′-carboxyphenylpyruvic acid and to 3′-carboxy-4′-hydroxyphenylpyruvic acid. The proposed biosynthetic pathways for the two carboxy-substituted amino acids are in agreement with their distribution patterns in the plant kingdom and suggest that they may derive from minor changes of enzymes involved in the general pathways of aromatic biosynthesis.  相似文献

To elucidate the formation mechanism of N,N′-dialkylpyrazine cation radical during browning reaction of sugars with amino compounds, main products in an early stage of the reaction were determined quantitatively by TLC and GLC. It was shown that the Schiff base, two-carbon fragmental product of sugar, the free radical and deoxyosone were successively produced prior to the browning. Polarographical measurements indicated that the radical formation was induced by the production of some reducing substances in the reaction mixture. These results suggest that the free radical was formed by the reduction of N,N′-dialkylpyrazinium; a compound, which demonstrated to have a strong activity to browning, might be formed by condensation of two-carbon enaminol followed by oxidation.  相似文献
