The influence of the methodology used in the evaluation of feed ingredients for rabbits on the precision of determination is reviewed. The procedures for calculation of the energy values and their standard errors are shown and discussed for each method. The direct method used for evaluating some relatively balanced feedstuffs such as alfalfa hay or wheat bran gives consistent results, but over- or underestimation and low repeatability is obtained with most imbalanced ingredients. For the substitution method, two calculation procedures of the energy values are compared, showing greater accuracy with the equation in which the gross energy of the ingredient is determined directly instead of estimated by difference. The basal diet and the rate/s of substitution must be designed to avoid a great nutrient imbalance in all the experimental diets. The substitution rate is the main factor in the precision of estimation of feedstuff evaluation, being directly related to the accuracy. Thus, the standard error of the ingredient energy value estimated by difference is 13.4, 6.4 and 2.9 times higher than the standard error of diets for 10%, 20% and 40% substitution rates, respectively. The number of replicates is of limited importance in comparison with the substitution rate; thus, the numbers of rabbits required to estimate the mean energy value within ± 5% with a 95% confidence are 37 and 10 for 20% and 40% substitution rates, respectively. Therefore, the energy value of ingredients that are usually included in diets in high proportions (20–30%) can be obtained with good precision by difference using only one rate of substitution. Several levels of inclusion (four or more) must be used when interactions between ingredients are expected or very low rates of inclusion are used. In this case, the energy value is estimated by extrapolation of the regression between the digestible energy of diets and the substitution rates and a greater precision than by difference is obtained. The multiregression method is rarely used because of the great number of ingredients included in the diets and because of the existence of interactions among them, preventing the correct estimation of their energy values.  相似文献

Calculating the estimated resting energy expenditure (REE) in severely obese patients is useful, but there is controversy concerning the effectiveness of available prediction equations (PE) using body weight (BW). We evaluated the efficacy of REE equations against indirect calorimetry (IC) in severely obese subjects and aimed to develop a new equation based on body composition compartments. One hundred and twenty severely obese patients had their REE measured (MREE) by IC and compared to the most commonly used PE (Harris-Benedict (HB), Ireton-Jones, Owen, and Mifflin St. Jeor). In a random sample (n = 60), a new REE equation based on fat-free mass (FFM) was developed and validated. All PE studied failed to estimate REE in severe obesity (low concordance correlation coefficient (CCC) and limits of agreement of nearly 50% of the sample ±10% of MREE). The HB equation using actual BW exhibited good results for all samples when compared to IC (2,117 ± 518 kcal/day by HB vs. 2,139 ± 423 kcal/day by MREE, P > 0.01); these results were blunted when patients were separated by gender (2,771 vs. 2,586 kcal/day, P < 0.001 in males and 1,825 vs. 1,939 kcal/day, P < 0.001 in females). A new resting energy expenditure equation prediction was developed using FFM, Horie-Waitzberg, & Gonzalez, expressed as 560.43 + (5.39 × BW) + (14.14 × FFM). The new resting energy expenditure equation prediction, which uses FFM and BW, demonstrates higher accuracy, precision, CCC, and limits of agreement than the standard PE in patients when compared to MREE (2,129 ± 45 kcal/day vs. 2,139 ± 423 kcal/day, respectively, P = 0.1).The new equation developed to estimate REE, which takes into account both FFM and BW, provides better results than currently available equations.  相似文献

The aim of this work was to test the robustness of the 0.68 estimate of the efficiency of conversion of metabolisable protein into true milk protein (Agriculture and Food Research Council (AFRC), 1993) for protein-limiting diets and to determine whether a different value is appropriate for practical rationing. Seventy-two multiparous cows were blocked on the basis of milk energy output per unit of dry matter intake (DMI), and allocated at random to one of four treatments. Treatments supplied metabolisable energy (ME) at a fixed level to individuals within a block, but varied metabolisable protein (MP) supply from 25% below the estimated requirements, through -12.5% and +12.5% up to 25% above requirements for the average performance of animals within blocks at the start of the study. Cows were offered diets to meet their predicted ME requirements for each 3-week period with measurements performed in the last week of each period. Milk protein output was regressed against the estimated MP available for production for each cow and the efficiency of conversion of MP into milk true protein was calculated, assuming a maintenance requirement according to the MP system. The efficiency of conversion of MP into milk true protein decreased with the increasing supply of MP from 0.77 to 0.50. Using an iterative approach to determine the best fit of the data when supply matched requirement resulted in a range of efficiency values between 0.62 and 0.64 g of true milk protein per g of MP.  相似文献

Chicks were fed on diets containing 23% of protein with various levels of energy from 60 to 75%. Total digestible nutrient is used to show the dietary energy level. The relationship between body weight gain (y) of chicks of 4 weeks fed with standard diets and dietary energy level (x) can be shown as:

By substitution of y of chicks fed on unknown diet, x of the diet can be obtained.

With this principle, it was found that the total digestible nutrients of raw and cooked sweet potato were 47% and 65%, respectively. The former is about 3/4 of the value previously estimated by Morimoto. Results of the determination of metabolizable energy of the test diets endorse the reliability of this conclusion.  相似文献

The objective of this study was to estimate the effect of increasing the dietary content of bacterial protein meal (BPM) on energy and protein metabolism in growing mink kits. Sixteen male mink kits of the standard brown genotype were randomly fed one of four diets: A control (Diet I) based on high-quality fish meal, and three experimental diets in which 20% (Diet II), 40% (Diet III) and 60% (Diet IV) of the digested nitrogen (DN) was replaced with BPM. Nitrogen balance and respiration experiments (indirect calorimetry) were carried out when the animals were approximately 9.5, 14.5, 17.5, 23.5 and 28.5 weeks of age. The apparent digestibility of crude protein and energy decreased significantly with increasing dietary BPM. The retained nitrogen was 0.45, 0.54, 0.52 and 0.40 g/kg0.75 on Diets I, II, III and IV, respectively, the observed differences between diets being non-significant (p = 0.06). Heat production (HE) was between 645 and 665 kJ/kg0.75 on all diets (p = 0.78). Retained energy (RE) was approximately 150-160 kJ/kg0.75 on Diets I to III, whereas it was -11 kJ/kg0.75 on Diet IV, the differences being significant (p< 0.001). A lower feed intake and apparent digestibility of energy caused the negative RE on Diet IV. The amount of HE from oxidation of protein decreased from 32.7% on Diet I to 26.6% on Diet IV, and oxidation of fat increased from 53.8% on Diet I to 63.5% Diet IV. In conclusion, protein and energy metabolism remained unaffected when up to 40% of DN was derived from BPM.  相似文献

Biological availability of 33 esters, 17 ethers and 2 acetals of ethanediol, 1,2-propanediol, 1,3-butanediol and 1,4-butanediol was compared by mini-test with chicks. Chicks can utilize esters of ethanediol, 1,2-propanediol and 1,3-butanediol with acetic acid and fatty acids of carbon chain length from 5 to 12 with more improved palatability than that of free acids, while availability of esters of these glycols with propionic and butyric acids was low. Esters of 1,4-butanediol and ether derivatives of these glycols was not available, except ethyl ether of di-ethanediol which was partially available. Acetacetal of ethanediol was partially available but n-butyracetal was not.  相似文献

True metabolizable energy (TME) is a measure of avian dietary quality that accounts for metabolic fecal and endogenous urinary energy losses (EL) of non-dietary origin. The TME is calculated using a bird fed the test diet and an estimate of EL derived from another bird (Paired Bird Correction), the same bird (Self Correction), or several other birds (Group Mean Correction). We evaluated precision of these estimators by using each to calculate TME of three seed diets in blue-winged teal (Anas discors). The TME varied by <2% among estimators for all three diets, and Self Correction produced the least variable TMEs for each. The TME did not differ between estimators in nine paired comparisons within diets, but variation between estimators within individual birds was sufficient to be of practical consequence. Although differences in precision among methods were slight, Self Correction required the lowest sample size to achieve a given precision. Feeding trial methods that minimize variation among individuals have several desirable properties, including higher precision of TME estimates and more rigorous experimental control. Consequently, we believe that Self Correction is most likely to accurately represent nutritional value of food items and should be considered the standard method for TME feeding trials.  相似文献

Currently, most analytical methods assume all observed genotypes are correct; however, it is clear that errors may reduce statistical power or bias inference in genetic studies. We propose procedures for estimating error rate in genetic analysis and apply them to study the GeneChip Mapping 10K array, which is a technology that has recently become available and allows researchers to survey over 10,000 SNPs in a single assay. We employed a strategy to estimate the genotype error rate in pedigree data. First, the "dose-response" reference curve between error rate and the observable error number were derived by simulation, conditional on given pedigree structures and genotypes. Second, the error rate was estimated by calibrating the number of observed errors in real data to the reference curve. We evaluated the performance of this method by simulation study and applied it to a data set of 30 pedigrees genotyped using the GeneChip Mapping 10K array. This method performed favorably in all scenarios we surveyed. The dose-response reference curve was monotone and almost linear with a large slope. The method was able to estimate accurately the error rate under various pedigree structures and error models and under heterogeneous error rates. Using this method, we found that the average genotyping error rate of the GeneChip Mapping 10K array was about 0.1%. Our method provides a quick and unbiased solution to address the genotype error rate in pedigree data. It behaves well in a wide range of settings and can be easily applied in other genetic projects. The robust estimation of genotyping error rate allows us to estimate power and sample size and conduct unbiased genetic tests. The GeneChip Mapping 10K array has a low overall error rate, which is consistent with the results obtained from alternative genotyping assays.  相似文献

In spring 2006, we conducted a collaborative U.S.–Russia survey to estimate abundance of the Pacific walrus (Odobenus rosmarus divergens). The Bering Sea was partitioned into survey blocks, and a systematic random sample of transects within a subset of the blocks was surveyed with airborne thermal scanners using standard strip‐transect methodology. Counts of walruses in photographed groups were used to model the relation between thermal signatures and the number of walruses in groups, which was used to estimate the number of walruses in groups that were detected by the scanner but not photographed. We also modeled the probability of thermally detecting various‐sized walrus groups to estimate the number of walruses in groups undetected by the scanner. We used data from radio‐tagged walruses to adjust on‐ice estimates to account for walruses in the water during the survey. The estimated area of available habitat averaged 668,000 km2 and the area of surveyed blocks was 318,204 km2. The number of Pacific walruses within the surveyed area was estimated at 129,000 with 95% confidence limits of 55,000–507,000 individuals. Poor weather conditions precluded surveying in other areas; therefore, this value represents the number of Pacific walruses within about half of potential walrus habitat.  相似文献

Hematein-free hematoxylin (HFH) was prepared by a modification of the procedure of Palmer and Lillie (Histochemie, 5: 44-54, 1965). Fifty mg of HFH were dissolved in 5 mg of ethylene glycol and then 45 nil of an aqueous solution of 2.25 gm KAl(SO4)2. 12H2O and 5.445 mg KIO3 were added. Since this amount of KIO3 would be sufficient to oxidize 25 mg of HFH to hematein we have termed this half-oxidized hematoxylin (HOH). The peak absorbance (560 nm) of this purple solution remained constant for at least a week. With omission of the KIO3 the solution was colorless. A curve was constructed by plotting absorbance against concentration of hematein in HOH at various dilutions. For analyses of hematein content of commercial hematoxylins 50 mg of sample and 100 mg of hydroquinone were dissolved in 5 ml of ethylene glycol and then 45 ml of a 5% solution of KAl(SO4)2. 12H2O were added. The addition of the hydroquinone stabilized the absorbance for about 5 min. The hematein content could then be calculated by comparing the observed absorbance with the standard curve. Eleven samples of hematoxylin certified by the Biological Stain Commission had hematein concentrations varying from 0.01 to 0.43%. For analyses of the available hematein content of commercial hemateins, 50 mg of sample were dissolved in 10 ml of ethylene glycol, then 45 ml of water and 45 ml of 5% KAI(SO4)2. 12H2O added. The hematein content could then be calculated by comparing the observed absorbance with the standard curve. In 9 samples of hematein from 4 different sources the active hematein content varied from 19 to 97%.  相似文献

The cranial capacity of Olduvai Hominid 7 is estimated to be 690 cc, with a standard uncertainty range of 538 to 868 cc. The estimate is derived from a systematic consideration of the relationships between Bregma-Asterion chords and cranial capacities obtained from a large sample of Homo sapiens and Pan troglodytes and from available fossil hominids. Several researchers have developed prediction equations to estimate the metabolisable energy (ME) of energetic and protein concentrate feedstuffs used in diets for broilers. The ME is estimated by considering CP, ether extract, ash and fibre contents. However, the results obtained using traditional regression analysis methods have been inconsistent and new techniques can be used to obtain better estimate of the feedstuffs’ energy value. The objective of this paper was to implement a multilayer perceptron network to estimate the nitrogen-corrected metabolisable energy (AMEn) values of the energetic and protein concentrate feeds, generally used by the poultry feed industry. The concentrate feeds were from plant origin. The dataset contains 568 experimental results, all from Brazil. This dataset was separated into two parts: one part with 454 data, which was used to train, and the other one with 114 data, which was used to evaluate the accuracy of each implemented network. The accuracy of the models was evaluated on the basis of their values of mean squared error, R2, mean absolute deviation, mean absolute percentage error and bias. The 7-5-3-1 model presented the highest accuracy of prediction. It was developed an Excel® AMEn calculator by using the best model, which provides a rapid and efficient way to predict the AMEn values of concentrate feedstuffs for broilers.  相似文献

A direct injection high-performance liquid chromatography method is described for the determination of mitomycin C (MMC) in human plasma. The stationary phase consisted of hydrophilic and hydrophobic functional groups covalently bound to silicone-coated silica beads (CAPCELL PAK MF Ph-1, 150×4.6 mm I.D., 5 μm). A mobile phase using 100% water gave a better separation of MMC from endogenous interferences as compared to a mobile phase with 12.5% acetonitrile and 2.5 mM phosphate buffer (pH 6.9). Using water as the eluent (1 ml/min) and UV detection at 365 nm, MMC was found to elute at 5.0 min with a peak width of 0.3 min, whereas endogenous interferences eluted before 3 min. Total assay time per sample was 6 min. Internal standard was not required because the recovery of MMC was nearly complete, about 90% from 20 to 5000 ng/ml. The standard curve was linear from 20 to 5000 ng/ml in plasma, and the intra- and inter-day variation was between 3 to 6%. The lower detection limit was 5 ng/ml with a 25 μl sample, which represent a two- to four-fold improvement over the 10 ng/ml detection limit by previous methods using liquid-liquid extraction and comparable sample size. The simplicity of this method, i.e., no sample extraction, no internal standard, 100% aqueous mobile phase, isocratic elution and short analysis time (6 min/sample), makes it suitable for large scale routine sample analysis, whereas its small sample volume requirement and high sensitivity are useful for pharmacokinetic studies in small animals where limited sample is available.  相似文献

Over a 14-day period, male SPF Wistar rats with an initial weight of 60, 200 and 250 g (ages 30, 75 and 90 days) were given diets with a constant protein content (10% casein) and a mounting fat content (10, 15, 20, 25, 30, 40 and 50% margarine, at the expense of the saccharides in the standard diet). The utilization parameter of protein biological value (NPU) for the various diets was determined from protein intake and the body nitrogen values. The reciprocal relation of the intake of the two nutrients was determined on the basis of linearity between th growth parameter NPR and protein and fat intake and, after substituting the optimum protein intake, was used to compute the optimum fat content of the diet. From the aspect of the maximum NPU value the optimum fat contents were 30% for 30-day-old, 15% for 75-day-old and 10% for 90-day-old animals. In all three age groups (according to the growth curve for the standard Larsen diet animals with an equal growth rate of 3 g/day), protein utilization under optimum nutritional conditions was the same. The optimum fat content of the diet computed from the reciprocal relationship of fat and protein intake is in agreement with the value obtained by the biological method - 29.4% (49.5 energy %) for 30-day-old animals, 15,8% (30.4% of the total energy value of the diet) for 75-day-old animals and 11.83% (23.7% of total energy) for 90-day-old rats. By using a biological and a mathematical method, this study contributes to the determination of optimum physiological doses of nutrients.  相似文献

Abstract. Two sets of 40 relevés, made independently by two observers on the same 5m x 5m sample plots, were compared to estimate the sampling error and to assess the effect of this sampling error on (1) estimates of species richness and diversity (2) results of multivariate analyses, and (3) estimation of species turnover in repeated sampling. The relevés were made according to the standard Braun-Blanquet method. The sampling error was estimated for (1) recording of species in sample plots and (2) visual estimation of the degree of cover (or of the general population size). Despite the fact that the sample plots were searched thoroughly for 30 - 40 min, the number of overlooked species was high with a discrepancy of 13% between corresponding relevés. Regarding multivariate analysis, the error caused by missing species was at least as important as the error in visual estimation of species cover. The estimates of degree of cover using the Braun-Blanquet scale are sufficiently reliable for use in multivariate analysis when they are subjected to ordinal transformation. When average cover values are used, the patterns detected are based solely on dominants. Species richness and species diversity could be reliably estimated from the relevés, but the estimates of equitability are very unreliable. The classical relevé method remains one of the most efficient survey methods for recognition of vegetation types on the macro-community and landscape scales.  相似文献

Two experiments were conducted to estimate the metabolisable energy (ME) and net energy (NE) of rice straw and wheat straw for beef cattle. In each experiment, 16 Wandong bulls (Chinese indigenous yellow cattle) were assigned to 4 dietary treatments in a completely randomised design. Four dietary treatments included one corn silage-concentrate basal diet and three test diets in which the basal diet was partly substituted by rice straw (Exp. 1) or wheat straw (Exp. 2) at 100, 300 and 600 g/kg. Total collection of faeces and urine was conducted for 5 consecutive days after a 2-week adaption period, followed by a 4-d period where gas exchange measurements were measured by an open-circuit respiratory cage. Linear regression equations of rice straw- or wheat straw-associated ME and NE contribution in test diets against rice straw or wheat straw substitution amount were developed to predict the ME and NE values of rice straw and wheat straw. These regression equations resulted in ME and NE values (dry matter basis) of 6.76 and 3.42 MJ/kg for rice straw and 6.43 and 3.28 MJ/kg for wheat straw, respectively. The NE and ME requirement for maintenance of Wandong cattle fed a straw-based diet were 357 and 562 kJ·kg?0.75·d?1, respectively. The regression-derived ME and NE have lower standard errors and coefficients of variation than those estimated by any single substitution ratio. Our study found that the regression method based on multiple point substitution is more reliable than the substitution method for energy evaluation of feedstuffs for beef cattle.  相似文献

Protein is essential for living organisms, but digestibility of crude protein is poorly understood and difficult to predict. Nitrogen is used to estimate protein content because nitrogen is a component of the amino acids that comprise protein, but a substantial portion of the nitrogen in plants may be bound to fiber in an indigestible form. To estimate the amount of crude protein that is unavailable in the diets of mountain gorillas (Gorilla beringei) in Bwindi Impenetrable National Park, Uganda, foods routinely eaten were analyzed to determine the amount of nitrogen bound to the acid-detergent fiber residue. The amount of fiber-bound nitrogen varied among plant parts: herbaceous leaves 14.5+/-8.9% (reported as a percentage of crude protein on a dry matter (DM) basis), tree leaves (16.1+/-6.7% DM), pith/herbaceous peel (26.2+/-8.9% DM), fruit (34.7+/-17.8% DM), bark (43.8+/-15.6% DM), and decaying wood (85.2+/-14.6% DM). When crude protein and available protein intake of adult gorillas was estimated over a year, 15.1% of the dietary crude protein was indigestible. These results indicate that the proportion of fiber-bound protein in primate diets should be considered when estimating protein intake, food selection, and food/habitat quality.  相似文献

In the captive Indian rhinoceros (Rhinoceros unicornis), two disease complexes with a high incidence—chronic foot problems and uterine leiomyomas—may be linked to excess body weight (BW). In this study, intake and digestion trials were conducted (by means of 7‐day weigh‐backs, and 5‐day total fecal collections, respectively) with 11 Indian rhinoceroses at four zoological institutions in Europe and the United States to quantify energy and mineral nutrition on conventional or roughage‐only diets. Diets comprising a variety of forages (grass hay only, a combination of grass hay and grass silage, straw, or a mixture of grass and legume hay) were offered as the roughage source, along with various concentrates, produce, and supplements. Water intake was quantified, and urine samples were obtained opportunistically. The animals consumed 0.5–1.1% of their BW in dry matter (DM) daily, with calculated digestible energy (DE, in megajoules MJ) values ranging from 0.27 to 0.99 MJ DE/kg BW0.75/day compared to an estimated requirement of 0.49–0.66 MJ DE/kg BW0.75/day. Seven of 11 rhinos (64%) fed restricted levels of concentrate plus forage consumed DE in excess of this estimate. Even on roughage‐only diets, some individuals consumed energy well above the presumed metabolic requirements. Hence, restriction of both concentrates and roughage may be important for weight management in this species. Water intake ranged from 30 to 49 mL/kg BW daily (3.4–5.2 L/kg ingested DM), similar to values that have been reported for domestic equids. Excretion amounts and patterns also resembled those found in horses. Endogenous fecal losses measured for Ca, P, Cu, Fe, and Zn indicate that the maintenance requirements of these minerals should be met in Indian rhinoceroses by diets that meet recommendations for domestic horses. It is particularly important to evaluate dietary adequacy in mineral nutrition in this species in concert with the need for restricted energy intake, especially with regard to the hypothetical involvement of a low Zn supply in chronic foot problems. Summary The wombats Vombatus ursinus and Lasiorhinus latifrons have a capacious proximal colon with only a vestigial caecum. The pattern of microbial fermentation in the hindgut of both species was studied in captive animals fed a pelleted straw diet and in wild wombats feeding on their natural winter diets. Digesta pH was low in the stomach but near neutrality along the hindgut, indicating effective absorption and/or buffering of the colonic contents. Initial proportions and production rates of short chain fatty acids in vitro reflected the fermentation of plant cell walls. Proportions of isobutyrate, isovalerate and n-valerate increased towards the distal colon indicating proteolysis and subsequent fermentation of amino acids. The low ammonia content of digesta fluid suggested that ammonia released from these amino acids was absorbed and utilized by the wombats and their gut microbes. Wild wombats had higher concentrations and production rates of short chain fatty acids than captive animals, which was consistent with the higher apparent digestibility of their natural diet. The energy from short chain fatty acids in captive animals was 30–33% of digestible intake. Energy intakes were low and similar to resting metabolic rates estimated for marsupials. Actual resting metabolic rates of the wombats are probably lower than these estimates, and the proportion of energy derived from fermentation substantially higher than the 53–61% estimated in wild wombats. Many fruits contain high levels of available energy but very low levels of protein and other nutrients. The discrepancy between available energy and protein creates a physiological paradox for many animals consuming high-fruit diets, as they will be protein deficient if they eat to meet their minimum energy requirement. We fed young grizzly bears both high-energy pelleted and fruit diets containing from 1.6% to 15.4% protein to examine the role of diet-induced thermogenesis and fat synthesis in dealing with high-energy-low-protein diets. Digestible energy intake at mass maintenance increased 2.1 times, and composition of the gain changed from primarily lean mass to entirely fat when the protein content of the diet decreased from 15.4% to 1.6%. Daily fat gain was up to three times higher in bears fed low-protein diets ad lib., compared with bears consuming the higher-protein diet and gaining mass at the same rate. Thus, bears eating fruit can either consume other foods to increase dietary protein content and reduce energy expenditure, intake, and potentially foraging time or overeat high-fruit diets and use diet-induced thermogenesis and fat synthesis to deal with their skewed energy-to-protein ratio. These are not discrete options but a continuum that creates numerous solutions for balancing energy expenditure, intake, foraging time, fat accumulation, and ultimately fitness, depending on food availability, foraging efficiency, bear size, and body condition.  相似文献
