Biological availability of 23 alcohols, 16 aldehydes and 2 ketones was compared by the mini-test with chicks. Chicks can utilize methanol and ethanol, but not the alcohols of carbon chain from 3 to 9. Lauryl and myristyl alcohols were well utilized but those of higher carbon chain than 14 were not, mainly due to low digestibility. Glycerol and D-sorbitol were well utilized but not 4 other poly alcohols tested. Aldehydes and ketones with free carbonyl group showed low availability or even toxicity, but their derivatives with masked group showed no detrimental effect. Acetaldehyde, acetal and aldehydes of carbon chain longer than 9 were partially utilized. 相似文献

Biological availability of 33 esters, 17 ethers and 2 acetals of ethanediol, 1,2-propanediol, 1,3-butanediol and 1,4-butanediol was compared by mini-test with chicks. Chicks can utilize esters of ethanediol, 1,2-propanediol and 1,3-butanediol with acetic acid and fatty acids of carbon chain length from 5 to 12 with more improved palatability than that of free acids, while availability of esters of these glycols with propionic and butyric acids was low. Esters of 1,4-butanediol and ether derivatives of these glycols was not available, except ethyl ether of di-ethanediol which was partially available. Biological availability of 106 esters of alcohols and aliphatic mono-, di- and tri-carboxylic acids and diethylene glycol succinate was compared by the mini-test with chicks. Chicks can utilize methyl esters of saturated fatty acids of carbon chain from 10 to 14, ethyl esters of those from 9 to 12, propyl caprate, n-butyl esters of those from 8 to 12, n-amyl esters of those from 6 to 12, n-hexyl n-butyrate and i-vaterate, and n-octyl and n-decyl acetates. Only 3 dicarboxylates, i.e. di-octyl and di-lauryl succinates and di-methyl cis-cyclopropane-l,2-dicarboxylate, were available among the dicarboxylates tested. Nutritive value of 4 glycol esters, i.e. ethanediol diacetate, 1,2-propanediol diacetate, 1,3-butanediol diacetate and 1,3-butanediol dioctylate, was estimated biologically by feeding the esters to growing chicks and rats. Energy in the esters taken by both chicks and rats was well utilized, though feed intake of the diets containing the esters at high level tended to decrease. Bitter taste of the esters was suspected to be related to low appetite. The acetates were somewhat volatile and released free acetic acid in the diet during storage. These properties of the acetates makes their use for dietary energy source difficult in practical condition. 相似文献

Growth rate, efficiencies of utilization of dietary energy and protein, and carcass fat level of the chicks decreased linearly with the increase of the intensity of restriction of either feed or energy alone, while the levels of carcass components other than fat were almost kept constant regardless of the restriction. The data indicated that chicks have ability to tolerate severer restriction than feeding moderately lipogenic diet at the level of 40% of full-feeding. It was confirmed that responses of chicks can be predicted based on the dietary composition and feeding conditions, using equations presented in this and previous papers. 相似文献

A number of amino acid derivatives of DL-Zra/w-epoxysuccinic acid, with a general formula of R1O-ES-AA-OR2 (ES, DL-trans-epoxysuccinyl group; AA, amino acid residue) were newly synthesized and used for the study of structure-activity relationships of papain inhibition. Branched-alkyl amino acids, such as Leu, He and Val, as AA and hydrogen or an alkyl group substituted with a phenyl or cycloalkyl group as R1 were desirable for activity, respectively. A new bacterial method for determining amino acids in protein foods is described. Instead of the 'natural'microbial auxotrophs e.g. Tetrahymena, Streptococcus , and Leuconostoc , currently used for such assays, an 'artificial'mutant is used, viz. an auxotroph of Escherichia coli . Test proteins (Bovine serum albumin, legume and maize meals) were predigested with a mixture of pronase and intestinal peptidases, the efficiency and extent of proteolysis being monitored by pH stat titration. Final digests were examined by Sephadex gel filtration to ensure that all protein cleavage products were small enough to pass through the E. coli cell wall and to reach its cyto-plasmic amino acid and peptide permeases. The lysine content of the meals, as determined from the growth of an E. coli lysine auxotroph upon the digests, was found to be greater than 90° of the lysine determined chemically in acid hydrolysates. Practical and theoretical advantages of using this latter type of bacterium rather than the fastidious species are discussed. In addition, the particular value of using an intestinal bacterium like E. coli to assay nutritional availability of amino acids is considered in relation to its normal utilization of digested protein foods in vivo , and the similarities between its amino acid and peptide permeases and those of the intestine. 相似文献

逆转录酶 (RT)是目前发现的所有逆转录病毒的一个重要标志 ,凡是逆转录病毒均携带逆转录酶 ,逆转录酶活性的测定可以反映出制品中是否有逆转录病毒的污染 ,不受病毒基因序列的影响。采用PCR法进行逆转录酶活性检测方法 ,以整个生活周期中无DNA过程的MS2RNA为模板 ,设计一对引物 ,在逆转录系统中加入待检样品 ,经过RT PCR扩增后 ,放大检测对象物进行观察 ,从而了解制品中是否存在逆转录酶的污染 ,继而间接推测其是否污染逆转录病毒。对不同生物制品厂家生产的减毒活疫苗的逆转录酶活性进行检测 ,发现在一些疫苗里有逆转录酶活性阳性。 相似文献

The metabolic fate of the carbon skeleton of l-serine-U-14C has been investigated, in vivo and in vitro, in growing rats and chicks fed the diets with various protein calories percents (PC%) at 410 kcal of metabolizable energy.The incorporation of 14C into body protein at 12 hr after the injection of serine-14C was about 49% of the injected dose in rats fed the 10 or 15 PC % diet, though the value was reduced in rats fed lower and higher protein diets. The 14CO2 production was smaller in rats fed the 10 and 15 PC% diet, and it showed an inverse pattern to that of the 14C incorporation into body protein. Urinary excretion of 14C was higher in rats fed 10 and higher PC% diets, whose growth rate and net body protein retention were maximum.In contrast to the case of rats, the incorporation of 14C into body protein of chicks at 6 hr after the injection was rather reduced in the 15 PC% group. The proportion of 14C excreted as uric acid was remarkably increased above the 10 PC% group, and about 19% of the injected dose was recovered in the 50 PC% group.The catabolic rate of serine in the liver slices of rats and chicks was increased by high protein diets.These results support the concept that the nutritional significance of metabolism of the carbon skeleton of serine in growing rats and chicks is different from each other, especially at high protein diets. 相似文献

I. The chemical and spectrophotometric methods adopted in the Stain Commission laboratories for the certification of stains are described. Spectrophotometric examination is by the Beckman spectrophotometer in which the color density at the absorption maximum is determined as well as a ratio of color densities designed to detect abnormal symmetry of the absorption curve. A somewhat modified titanous chloride assay is described; eerie sulfate is used to determine the normality of the unstable titanous chloride, and the oxidant solution, which is stable, is standardized against Bureau of Standards arsenious oxide. The general approach used for re-study of the stains by these methods and the criteria necessary for adoption of the spectrophotometric method of assay are described.
II. This report gives the details of the spectrophotometric examination and assay of the individual stains in the thiazin group, namely methylene blue, thionin, methylene violet, toluidine blue O, and azures A, B, and C. Bioassy procedure to evaluate biologically available energy of chemicals applicable to rats was established, and available energy of 36 chemicals was determined and compared with that estimated by chicks previously. Rats can utilize energy of propionic and butyric acids and n-hexyl propionate and butyrate well, while chicks cannot. Succinic acid, lauryl alcohol and dilauryl succinate at 5% dietary level were available by rats, though at 10% level lauryl alcohol was toxic. Ethyl lactate, octyl and decyl acetates and 1,2-propanediol dilaurate were available by both rats and chicks. Availability of other 6 esters including ethyl succinate and citrate was low. Experimental research involving Arabidopsis thaliana often involves the quantification of phenotypic traits during cultivation on compost or other growing media. Many commercially-available growing media contain peat, but peat extraction is not sustainable due to its very slow rate of formation. Moreover, peat extraction reduces peatland biodiversity and releases stored carbon and methane into the atmosphere. Here, we compared the experimental performance of Arabidopsis on peat-based and several types of commercially-available peat-free growing media (variously formed from coir, composted bark, wood-fibre, and domestic compost), to provide guidance for reducing peat use in plant sciences research with Arabidopsis. Arabidopsis biomass accumulation and seed yield were reduced by cultivation on several types of peat-free growing media. Arabidopsis performed extremely poorly on coir alone, presumably because this medium was completely nitrate-free. Some peat-free growing media were more susceptible to fungal contamination. We found that autoclaving of control (peat-based) growing media had no effect upon any physiological parameters that we examined, compared with non-autoclaved control growing media, under our experimental conditions. Overall, we conclude that Arabidopsis performs best when cultivated on peat-based growing media because seed yield was almost always reduced when peat-free media were used. This may be because standard laboratory protocols and growth conditions for Arabidopsis are optimized for peat-based media. However, during the vegetative growth phase several phenotypic traits were comparable between plants cultivated on peat-based and some peat-free media, suggesting that under certain circumstances peat-free media can be suitable for phenotypic analysis of Arabidopsis. 相似文献

Malondialdehyde (MDA) is assayed in femtomole quantities in biological samples by gas chromatography-mass spectrometry (GC-MS). The MDA trapped in protein as a Schiff base is released by H2SO4, the protein precipitated using Na2WO4, and the MDA derivatized with pentafluorophenylhydrazine to form the stable adduct, N-pentafluorophenylpyrazole. Negative chemical ionization (NCI) capability allows the sensitive detection of this MDA adduct in biological samples at a level of 5 nM on-column. A stable-isotope-labeled MDA, [2H2]MDA, was used as an internal standard for quantitation. MDA recovery from plasma was 76%. This assay provides two forms of confirmation of the analyte, retention time and mass ion, thus minimizing error due to interfering compounds. The commonly used thiobarbituric acid assay for MDA overestimates the MDA levels by over 10-fold, possibly resulting from cross-reactivity with other aldehydes and artifactual oxidation due to 100°C temperature conditions. In our assay, all steps were performed at room temperature thereby suppressing artifactual oxidation of the sample. We have successfully applied this assay to biological samples including plasma, tissue homogenates, and sperm. 相似文献

In this continuation paper of the work on the chemical and spectrophotometic characteristics of commercial stains, data on the xanthene dyes are presented. In the xanthene group of dyes, it has been found possible to assay pyronin B, eosins B and Y and ethyl eosin by spectrophotometric means. Phloxine B, rose Bengal, and erythrosin B are assayed by die color acid precipitation method. Typical absorption curves are given for these dyes as well as representative spectral and assay data. 相似文献
