This research investigates the effect of bacterial chemotaxis on biodegradation rate in an experimental model for in situ bioremediation. The novel experimental methodology of this work has provided for the systematic evaluation of the effect of the chemotaxis phenomenon in a saturated porous medium. The methodology has been developed to measure enhancement of degradation rate of serine, a simulated contaminant and chemoattractant. Escherichia coli RP437 was used as a representative, chemotactic, in situ bacterium, whereas E. coli RP5700, a tsr? mutant strain of RP437 that lacks the serine chemoreceptor, was used as the specifically nonchemotactic control strain. These two strains were highly characterized for this work. Swimming speeds, run lengths, and turn angles were compared using a tracking microscope and were statistically similar, as were serine uptake rates, making this pair of strains an excellent choice for chemotaxis studies. For these experiments, a model aquifer introduces bacteria to serine in saturated sand via a sharp gradient. The aquifer successfully achieved biodegradation at an 18% level; however, the degradation rate of serine was similar for both strains over 21 h, indicating that chemotaxis enhancement did not occur. This result is in agreement with certain prior works which did not detect enhanced chemotactic migration.  相似文献

A large number of yeasts, including a variety of species other than Candida albicans, were isolated from clinical specimens. C. tropicalis and Torulopsis glabrata were each found one-third as frequently as C. albicans. A schema is presented which made possible, by simple procedures, the identification of the great majority of the isolated yeasts. Preliminary isolation and differentiation was aided by the use of molybdate-agar medium. The use of the schema by diagnostic bacteriological laboratories is discussed.  相似文献

The need for very durable mounting is especially felt in the teaching of parasitology and mycology; otherwise, the availability of microscope slides may depend on the use of fresh specimens. Resinous mounts and those in aqueous media sealed with fingernail lacquer, paraffin or asphalt do not preserve specimens satisfactorily. Polyvinylpyrrolidone (pvp), a water-soluble mounting medium described by Burstone (1962), cannot be applied directly for mounting of insects and certain other parasites which have water-repelling integuments; moreover, pvp bleaches eosin. Grimley et al. (1965) prepared large epoxy sections of tissues from which areas for electron microscopy could be selected. This procedure however is designed for electron microscopic techniques whereas the present paper describes a direct epoxy mounting method to produce permanent mounts for light microscopy.  相似文献

Rigid endoscopes degrade during clinical use due to sterilization, ionizing radiation and mechanical forces. Despite visual checks on functionality at the department of sterilization, surgeons are still confronted with suboptimal instruments as it is difficult to assess this degradation in an objective manner. To guarantee that endoscopes have sufficient optical quality for minimal invasive surgery, an experimental opto-electronic test bench has been developed in order to be used at the department of sterilization. Transmission of illumination fibres and lens contrast values are stored in a database to enable empirical criteria to reject endoscope for further clinical usage or to accept endoscopes after repair. Results of the test bench are given for an eight month period, where a trained operator performed 1599 measurements on 46 different types. Stability of the system, trends in quality of clinical endoscopes, and effect of repair or replacement were assessed. Although the period was too short to draw firm conclusions, a slow downwards trend in quality of clinically used endoscopes could be observed. Also, endoscopes generally improve in quality after repair or replacement, while endoscope replacement seems to slightly outperform endoscope repair. To optimize the measurement process, a new system is being developed requiring less user interaction and measuring more optical parameters of an endoscope. By commercializing this system, we hope that measurements at different hospitals will give improved insight which acceptance and rejection criteria to use and which factors (usage, cleaning protocol, and brands) determine the economic lifetime of endoscopes.  相似文献

Blood was cultured in two vaccum bottles containing Columbia broth with sodium polyanethol sulfonate and CO2. Filtered air was admitted to one bottle, and the bottles were incubated at 35 C until growth was detected or for a maximum of 7 days. Bottles were examined daily for macroscopic growth. Gram stains were made routinely on the 1st, 4th, and 7th days, and samples were routinely subcultured to sheep blood agar (incubated in GasPak jar) and chocolate agar (incubated in CO2) on the 1st and 4th days of incubation. Of 1,127 positive blood cultures, 65% were first detected by macroscopic examination, 23% were first detected by Gram stain, and 12% were first detected only by subculture.  相似文献
