Emerging pattern of Cercaria tondoni sp., a distome (possessing a ventral sucker lying some distance from the posterior end and approximately at anterior third of the body) belonging to gymnocephalus group (having an oval body and tail devoid of stylets or spines), shows a daily rhythm, being greater during the light phase (0630:1830?h) – similar to that found in a freshwater snail Lymnea stagnalis, Linnaeus, 1758. The peak time of emergence 1230–1430?h has been measured, and there is a high risk for invertebrate or vertebrate hosts and other diurnal animals to acquire infection during this period. However 20% of total shedding in a population of cercariae has also been recorded during the dark phase (0630–1830?h) but there was no significant rhythmicity within this period. An attempt has been made to record cercarial emergence throughout the year, which is of great importance for human welfare. It was observed that the months of April, May, and June are those where frequency of emergence was highest. Implications of this rhythmicity are considered.  相似文献

Abstract

Swertia chirayita, a critically endangered medicinal herb, is being over-harvested in the wild. Understanding seed germination is a pre-requisite to ensure species conservation. The germination of seeds collected from six microhabitats was studied at 20°C, 25°C, and 30°C, both under a 14/10 h light/dark photoperiod and in continuous darkness. Two-way ANOVA indicated that microhabitat and temperature significantly affect seed germination, germination rate, germination recovery (GR), and GR rate. Overall, the seeds collected from under canopy showed a significantly (p < 0.05) higher germination than those from open habitats, at 20°C, 25°C, and 30°C (14/10 h light/dark photoperiod). Germination was negligible in continuous darkness but after transfer to a 14/10 h light/dark photoperiod, the seeds from under canopy significantly recovered at 20°C and at 25°C (p < 0.05), and showed the highest germination percentage compared to seeds collected from tree base, stump base, shrubberies, and grassy slope. Similarly, at 30°C, seeds from under canopy recorded the highest GR percentage. In general, seed germination, mean germination rate, seed GR, and GR rate were significantly greater (p < 0.05) at 25°C. Among the microhabitats tested, variation in GR rate was significant (p < 0.05). Seeds were confirmed to be positively photoblastic.  相似文献

Five-week-old seedlings of Capsicum annuum variety SAMPEP 4, Californian Wonder and Ex Dandamasa drenched with 15,000 infectious units per ml of Phytophthora capsici were incubated at 5°C, 20°C, 30°C and 35°C in alternating light–dark cool cycle Gallenkamp incubators and monitored for root rot development. Each host–pathogen system was replicated five times. Successful disease development was contingent on been incubated at ambient temperature for not less than 3.5 ± 0.5 h. Depending on variety, degenerate tissue rots were aggravated ≤2–3 days after a preconditioning temperature treatment for 24 h possibly linked to cell wall constitution, composition and permeability. Lesion development on stem heightened (27.8%) when incubated at temperatures above 20°C. Ten days after treatment, plant mortality and disease severity were not affected significantly by post-inoculation temperature.  相似文献

In the unicellular algae Pyrocystis lunula Schütt and Gonyaulax polyedra Stein, bioluminescence and its circadian regulation are similar in several respects, but there are also several important differences. As in G. polyedra, P. lunula emits light both as bright flashes and as a low intensity glow. At 20° C, the individual flashes are considerably brighter than in G. polyedra, and their durations are typically less than 500 ms. Both species show a circadian rhythm in the frequency of spontaneous flashes, which peaks in the night-phase under light–dark cycles and continues in both continuous light and dark. However, compared to G. polyedra, the circadian system in P. lunula is more sensitive to light: 10 min exposures (500 μmol · m–2· s–1 white light) can shift the phase of the rhythm by more than 8 h, and rhythmicity is completely suppressed at an irradiance above 20 μmol · m–2· s–1, where the G. polyedra rhythym persists for weeks. Like G. polyedra, period length increases with increasing irradiance of continuous red light but decreases with increasing intensity of continuous blue light. The glow in P. lunula differs markedly from that in G. polyedra in that it occurs at about the same intensity at all times during the circadian cycle; thus, it is not under circadian control but may fluctuate 5–10-fold in intensity within a time frame of seconds. This suggests that the glow may differ in its physiological basis in the two organisms. The results also indicate that the circadian regulation of luciferase activity differs in the two species. In G. polyedra, the organelle responsible for bioluminescence and luciferase is lost and then reformed on a daily basis; in P. lunula, the luciferase is conserved and localized elsewhere during the nonbioluminescent phase of the cycle.  相似文献

We report an investigation into the impact of temperature and illumination on in vitro symbiotic and asymbiotic germination of the threatened taxon Caladenia huegelii, and three other orchid spp. namely—Caladenia latifolia, Microtis media and Pterostylis sanguinea, all species from south-west Western Australia, a recognized biodiversity hotspot. High symbiotic germination on oatmeal agar (OMA + fungal symbionts specific to each species) was recorded in three species in continuous dark incubation i.e. C. huegelii seeds (98 % germination at 25 °C), and M. media and P. sanguinea (93 and 98 % respectively at 20 °C). Highest symbiotic germination for C. latifolia (100 %) was observed at 15 and 20 °C under light treatment (12/12 h light/dark). Low temperature incubation (10 °C) significantly suppressed symbiotic germination/development of seedlings across all species. Asymbiotic media treatments assessed (OMA minus fungal symbionts, Pa5 and ½ MS), failed to stimulate any germination with C. latifolia seeds at 20 °C in either light or dark treatments after an 8 week incubation period. Seeds of M. media sown onto ½ MS medium resulted in higher germination in all developmental stages (3–5) in dark treatment than OMA and Pa5. Seeds of P. sanguinea sown onto ½ MS medium resulted in higher overall germination in all developmental stages (3–5) in light and dark incubation compared to OMA and Pa5. OMA supported the highest asymbiotic germination (100 %) in both light and dark incubation with M. media (only to stage 3) but did not support germination and development with other spp. tested. Caladenia huegelii seeds reached developmental Stage 3 (i.e. germinated), but only on Pa5 medium and only at a relatively low rate in either light (2.6 %) or dark (2.1 %). Germination was higher and development of seedlings faster overall in all test species in symbiotic compared with asymbiotic media treatments. P. sanguinea seeds demonstrated the best response (among species tested) to asymbiotic germination on ½ MS with 40–53 % of germinated seeds spread over developmental stages 3–5 in light or dark incubation (at 20 °C) respectively. Illumination had no effect on fungal symbiont growth across all species, however incubation temperature treatments (10, 15, 20 and 25 °C) affected fungal growth rate. Growth of the fungal symbionts of C. huegelii, M. media and C. latifolia demonstrated significantly lower activity at 10 °C, but the cumulative radial growth rate of the P. sanguinea fungal symbiont reached 64 cm2 after only 2 weeks at all temperatures tested, including 10 °C. The study highlights differences in symbiotic and aysmbiotic germination and early protocorm development in vitro between co-occurring herbaceous terrestrial Australian orchid taxa in response to variations in basal media, temperature and light.  相似文献

Allium stracheyi Baker (Alliaceae, 2600–3000 m asl), an endangered species of Central Himalaya, India, has low seed germination in its natural habitat. This study is an attempt to improve seed germination by determining the seed viability with a low mean germination time (MGT) and germination index (GI) under optimum temperature, light, and pre-soaking treatments. The seeds were pre-soaked in hot water (80°C), cold water (10°C), and gibberellic acid (GA3 at 50 and 100 mg/l) for 24 h and subjected to light (12 h light and 12 h dark) and continuous dark (24 h) conditions with different temperature regimes (10, 15, 20, 25, and 30°C). The viability varied between 66.0% and 69.67% and declined rapidly after 12 months of storage. Our studies suggest that the 100 mg/l GA3 treatment was beneficial for seed germination and seedling growth. Pre-soaking in a 100 mg/l GA3 solution and incubation at 20°C under light conditions enhanced the germination significantly (p < 0.05) and resulted in the highest (97.3%) germination with the lowest MGT = 5.7 days, with GI = 8.11. The recommendations of this study support the conservation of alpine A. stracheyi via simple and cost-effective techniques for optimal seed germination.  相似文献

This research aimed at investigating the light synchronization and endogenous origin of daily expression rhythms of eight key genes involved in epigenetic mechanisms (DNA methylation and demethylation) in zebrafish gonads. To this end, 84 zebrafish were distributed into six tanks, each one containing 14 fish (7 males and 7 females). Animals were subjected to 12 h light:12 h dark cycles (LD, lights on at ZT0 h) and fed randomly three times a day during the light phase. Locomotor activity rhythms were recorded in each tank for 20 days to test their synchronization to light. Then, zebrafish were fasted for one day and gonad samples were collected every 4 h during a 24 h cycle (ZT2, 6, 10, 14, 18, and 22 h). The results revealed that most of the epigenetic genes investigated exhibited a significant daily rhythm. DNA methylation genes (dnmt4, dnmt5, dnmt7) exhibited a daily rhythm of expression with a nocturnal acrophase (ZT14:01~ZT22:17 h), except for dnmt7 in males (ZT2:25 h). Similarly, all DNA demethylation genes (tet2, tdg, mb4, gadd45aa, and apobec2b) revealed the existence of statistically significant daily rhythms, except for gadd45aa in females. In females, tdg, mb4, and apobec2b presented a nocturnal peak (ZT14:20 ~ ZT22:04 h), whereas the tet2 acrophase was diurnal (ZT4:02 h). In males, tet2, tdg, and gadd45aa had nocturnal acrophases (ZT18:26~ZT21:31 h), whereas mb4 and apobec2b displayed diurnal acrophases (ZT5:28 and ZT4:02 h, respectively). To determine the endogenous nature of gene expression rhythms, another experiment was performed: 12 groups of 14 fish (7 males and 7 females) were kept in complete darkness (DD) and sampled every 4 h during a 48 h cycle (CT2, 6, 10, 14, 18, 22, 26, 30, 34, 38, 42, and 46 h). Under DD, most of the genes (7 out of 8) presented circadian rhythmicity with different endogenous periodicities (tau), suggesting that the epigenetic mechanisms of DNA methylation and demethylation in the gonads follow an internal control, functioning as part of the translation network linking the environment into somatic signals in fish reproduction.  相似文献

Biological clocks are innate timing mechanisms that regulate many behavioral and physiological parameters in most organisms. In our modern life, heavy use of mobile phones (MPs) exerts a massive stress on organisms because their electromagnetic radiation usually results in varying degrees of damage to their biological systems including the biological rhythms. In the present study, the possible effects of exposure to radiofrequency–electromagnetic radiation (RF–EMR) from MPs on two characteristic circadian rhythms, locomotor activity and melatonin hormone rhythms, were investigated. Rats were exposed to RF–EMR from MPs at 900 MHz frequency (2-h/day for 2 weeks) during nighttime (20:00–22:00 h) followed by another two weeks without exposure for recovery. Locomotor activity rhythms of the control and treated groups (n = 5/group) were daily recorded using running wheels along the experimental period. For evaluating melatonin hormone rhythm, blood samples of control and treated groups (n = 12/group), were collected at the end of exposure and recovery periods, at 6-h time intervals per day (at 4:00, 10:00, 16:00, and 22:00 h). Rats exposed to RF–EMR exhibited phase shifting as well as a significant increased acrophase level in locomotor activity. Meanwhile, a significant decrease in serum melatonin levels with retaining lower amplitude rhythmicity was observed. Ceasing exposure for two weeks did not restore melatonin levels and circadian locomotor activity rhythms. It could be concluded that, under the current conditions, exposure to RF–EMR revealed disturbances in locomotor activity and melatonin level, although they maintained rhythmicity.  相似文献

Abstract

The purpose of this research was to explore the effect of temperature, light, and post-harvest period, and their interactions, on seed germination ecology of four common pasture species in the Mediterranean environment. Mature seeds of Diplotaxis erucoides, Hirschfeldia incana, Hyoseris scabra (Mediterranean distribution) and Sonchus oleraceus (cosmopolitan distribution) were subjected to seven constant temperatures (10–40°C, at intervals of 5°C) under continuous darkness, or a 12 h/12 h light/dark photoperiod at 30, 150 and 270 days after harvest (DAH). Cumulative germination and germination speed were determined. In all the tested species, except S. oleraceus, light significantly enhanced germination. S. oleraceus seeds maintained germination values over 90%, in a wide range of temperatures (10–35°C), in the dark as well as in light. Seeds of H. incana germinated well soon after seed dispersal. In D. erucoides and H. scabra, germination increased with storage period, while in S. oleraceus there was no effect of seed age. In all the species, moreover, no germination was recorded at 40°C. Temperature, light, and post- harvest requirement may be regarded as an adaptation strategy to ensure optimal conditions for seedling development and survival in Mediterranean species, while the species with a cosmopolitan distribution germinates under almost all tested conditions.  相似文献

The locomotor activity rhythm of the isopod, Porcellio olivieri, was investigated in Gannouch site in the south of Tunisia. The rhythm was monitored under constant temperature in individual animals in winter under two simultaneous regimens: the light–dark (LD) cycle and the continuous darkness (DD). Results revealed that whatever regimens, actograms, and mean activity curves showed that specimens of P. olivieri concentrated their activity during the experimental and subjective night. The species exhibited a locomotor rhythm period significantly shorter under LD (T = 23h13 ± 0h44) than DD (τ = 24h28 ± 0h58). However, the locomotor activity rhythm was less stable and the individuals were significantly more active under entraining conditions than constant darkness. The activity pattern of this species will be discussed as an adaptative strategy to respond to environmental conditions.  相似文献

Few studies have estimated fate and persistence of the hepatotoxic microcystins (MCs) in situ, making ecological and human health risk assessments challenging. We determined fate and persistence of MC congeners during 2 years of Microcystis blooms in a small, shallow, closed-basin lake in Ontario, Canada. In situ half-lives were compared to estimates obtained in vitro under controlled temperature and light. The blooms produced elevated microcystin-LA (MC-LA) (maximum ~4.2 mg L?1) with minor concentrations of MC-LR, -RR, and -YR. Dissolved MC-LA declined more slowly and persisted longer than particulate MC-LA with in situ half-lives (total 1.5–8.5 days) shorter than in vitro (total 6.8–60.0 days). Half-lives in 2010 were two to eight times shorter compared to 2009, likely due to differences in bloom phenology and species/strain composition. In vitro, higher temperature (4°C → 25°C in dark), and irradiance (dark → 45 → 260 μE m?2s?1 at 25°C) accelerated particulate and dissolved MC-LA decline, respectively. MC-RR accumulated in surface sediments while MC-LA was near detection despite elevated surface water concentrations. MC-LA appears to persist longer in surface waters than the equally toxic MC-LR, requiring almost the entire recreational season (9.5 weeks) to reach guideline concentrations (20 μg L?1).  相似文献

The supratidal amphipod Talorchestia longicornis Say has a circadian rhythm in activity, in which it is active on the substrate surface at night and inactive in burrows during the day. The present study determined: (1) the circadian rhythms in individual versus groups of amphipods; (2) the range of temperature cycles that entrain the circadian rhythm; (3) entrainment by high-temperature cycles versus light?:?dark cycles, and (4) seasonal substrate temperature cycles. The circadian rhythm was determined by monitoring temporal changes in surface activity using a video system. Individual and groups of amphipods have similar circadian rhythms. Entrainment occurred only to temperature cycles that included temperatures below 20°C (10–20, 15–20, 17–19, 15–25°C) but not to temperatures above 20°C (20–25, 20–30°C), and required only a 2°C temperature cycle (17–19°C). Diel substrate temperatures were above 20°C in the summer and below 20°C during the winter. Upon simultaneous exposure to a diel high-temperature cycle (20–30°C) and a light?:?dark cycle phased differently, amphipods entrained to the light?:?dark cycle. Past studies found that a temperature cycle below 20°C overrode the light?:?dark cycle for entrainment. The functional significance of this change in entrainment cues may be that while buried during the winter, the activity rhythm remains in phase with the day?:?night cycle by the substrate temperature cycles. During the summer, T. longicornis switches to the light?:?dark cycle for entrainment, perhaps as a mechanism to phase activity precisely to the short summer nights.  相似文献

Marmosets, Saguinus oedipus oedipus and S. fuscicollis, have been shown to have a diurnal heart rate pattern that has a marked difference between high and low values (about 55 beats/minute) and shows a low point during daylight hours around 1300–1400 hours. A similar pattern for body temperature was seen. A species difference existed; the larger S. o. oedipus has a higher heart rate during both light and dark periods. All measurements were made on undisturbed animals at hourly intervals using radiotelemetry. They were kept in a controlled environment with a light cycle of 12L: 12D and a temperature of 27 ± 1°C.  相似文献

Limonium supinum, a perennial herb with interest for the restoration and gardening of arid zones, is widely distributed in saline areas from southeastern Iberian Peninsula. Laboratory experiments were carried out to assess the effects of temperature and salinity on seed germination and on germination recovery from the effects of saline conditions after transfer to distilled water. Seed germination responses were determined over a four temperature regimes (20/10, 25/15, 30/20 and 35/25 °C; 12 h light/12 h dark photoperiod) and six salinities (0, 100, 150, 200, 400 and 600 mM NaCl). The higher germination percentages were obtained in non-saline conditions, under all temperature regimes. An alternating temperature of 20 °C light and 10 °C dark yielded the maximum germination for any saline concentration. Increase in salinity delayed the beginning and end of germination and reduced the final percentage of germination, which becomes completely inhibited at 600 mM NaCl. The adverse effect of salinity is reinforced by high temperatures (30/20 and 35/25 °C). The germination rate was also negatively affected by the increase in salinity and temperature. The final recovery percentages in high salt treatments were near 100%, indicating that exposure to high concentration of NaCl did not inhibit germination permanently.  相似文献

The aim of this study was to investigate the daily rhythms of hematological, biochemical and enzymatic parameters of the blood of a nocturnal model of fish (Lophiosilurus alexandri) bred in the laboratory (F1). Thirty-six juveniles were stocked in six tanks of a recirculation aquaculture system for 20 days. The fish were exposed to a light:dark cycle of 12:12 h and were fed 1% of biomass twice a day with commercial diet. The daily rhythms of hematological, biochemical and enzymatic parameters were then measured at six sampling times “zeitgeber time = ZT” at four-hour intervals under light:dark 12:12 h (lights on = ZT0, at 8.00 a.m). No differences were observed to alkaline phosphatase, glucose, cortisol, aspartate aminotransferase, superoxide dismutase, total protein and hematocrit (p > 0.05). However, white blood cell count, Lymphocytes (LYN), Neutrophils (NEU), Eosinophil and Neutrophils to Lymphocytes ratio were significant different between sample times (p < 0.05). Also, a significant difference in alanine transaminase was observed, with a peak of production at nighttime. In contrast, glutathione peroxidase peaked at 8:00. Uric acid, magnesium and Calcium (Ca++) showed statistically significant differences (p < 0.05). A significant difference was observed (p < 0.05), with a peak of albumin at 08:00 and triglycerides at 12:00, while cholesterol was low (p < 0.05) at 08:00 and higher from 12:00 to 04:00. Cosinor analysis revealed also rhythmicity to SOD, UA, Mg and Ca++, ALB and CHO (p < 0.05). In conclusion, the time of day must be considered a key factor when using blood parameters as biomarkers for disease, health and welfare in the L. alexandri aquaculture.  相似文献

Entomophthora grylli Fres. has been found on larvae of Bradysia (Sciaridae, Diptera) on wood in Kansas State University greenhouses since November 1967. Infected larvae crawled to an exposed site in the night and by morning spores of E. grylli were being discharged. In the greenhouse and under controlled environments spore discharge showed a marked periodicity; spore discharge occurred in the light with peaks in the first 1–4 h; discharge then gradually declined but extended into the dark. On the second and third days peaks occurred in the light but were progressively smaller. In a greenhouse under fluctuating conditions twin peaks occurred at 10·00 and 22·00; in a growth chamber, at constant 21° C. 90% r.h., alternating 12 h light and dark periods, spore discharge was similar, with maximum numbers about 21/2 h after the onset of the photo-period. In continuous dark and continuous light no endogenous pattern was evident. Temperature range for spore discharge was 2–28·, with optimum at 15·. Decreasing humidities resulted in decreased spore production and cessation of discharge below 40% r.h.  相似文献

Abstract Saxifraga cernua, a perennial herb distributed throughout the arctic and subarctic regions, shows high levels of dark respiration. The amount of respiration exhibited by leaves and whole plants at any temperature is influenced by the pretreatment temperature. Plants grown at 10°C typically show higher dark respiration rates than plants grown at 20°C. The levels of alternative-pathway respiration (or cyanide-insensitive respiration) in leaves of S. cernua grown at high and low temperatures were assessed by treating leaf discs with 0.25 mol m?3 salicylhydroxamic acid during measurements of oxygen consumption. Alternative pathway respiration accounted for up to 75% of the total respiration. Tissues from 20°C-grown plants yielded a Q10 of 3.37 for normal respiration, and of 0.97 for alternative-pathway respiration. Tissues from 10°C-grown plants yielded a Q10 of 2.55 for normal respiration, and of 0.79 for alternative-pathway respiration. The alternative pathway does not appear to be as temperature sensitive as the normal cytochrome pathway. A simple energy model was used to predict the temperature gain expected from these high rates of alternative-pathway respiration. The model shows that less than 0.02°C can be gained by leaves experiencing these high respiration rates.  相似文献

The effects of irradiance, temperature, thermal‐ and chilling‐light sensitivities on the photosynthesis of a temperate alga, Sargassum macrocarpum (Fucales) were determined by a pulse amplitude modulation (PAM)‐chlorophyll fluorometer and dissolved oxygen sensors. Oxygenic photosynthesis–irradiance curves at 8, 20, and 28°C revealed that the maximum net photosynthetic rates (NP max) and saturation irradiance were highest at 28°C, and lowest at 8°C. Gross photosynthesis and dark respiration determined over a range of temperatures (8–36°C) at 300 μmol photons m?2 s?1 revealed that the maximum gross photosynthetic rate (GPmax) occurred at 27.8°C, which is consistent with the highest seawater temperature in the southern distributional limit of this species in Japan. Additionally, the maximum quantum yields of photosystem II (F v/F m) during the 72‐h temperature exposures were stable at 8–28°C, but suddenly dropped to zero at higher temperatures, indicative of PSII deactivation. Continuous exposure (12 h) to irradiance of 200 (low) and 1000 (high) μmol photons m?2 s?1 at 8, 20, and 28°C revealed greater declines in their effective quantum yields (Φ PSII) under high irradiance. While Φ PSII under low irradiance were very similar with the initial F v/F m under 20 and 28°C, values rapidly decreased with exposure duration at 8°C. At this temperature, F v/F m did not recover to initial values even after 12 h of dark acclimation. Final F v/F m of alga at 28°C under high irradiance treatment also did not recover, suggesting its sensitivity to photoinhibition at both low and high temperatures. These photosynthetic characteristics reflect both the adaptation of the species to the general environmental conditions, and its ability to acclimate to seasonal changes in seawater temperature within their geographical range of distribution.  相似文献

The role of environmental factors in seasonal succession of six clones of marine diatoms was examined by testing effects of various combinations of temperature (5°, 10°, 15°, 20°C) and photoperiod (10, 12, 14, 16 h) on time to germination of resting stages and subsequent vegetative cell division rates. Resting stages were induced in batch cultures using a low nutrient medium and a cold, dark period. A three-factor ANOVA showed that species, temperature, daylength, and their interactions signficantly (P =0.0001) injuenced germination time and division rate. Resting stages of Chaetoceros similis Cleve and Odontella aurita (Lyngbye) Agardh were most successful in producing populations of cells on warm, long days (20°C/16 h). Chaetoceros didymus Ehrenberg and C. cinctus Gran were most successful under more moderate conditions (15°C/14 h). Resting stages of Ditylum bright-wellii (West) Grunow germinated and cells divided rapidly under all conditions. No consistent pattern was evident with Thalassiosira rotula Meunier. We compared results of the laboratory study with environmental conditions prevailing during the presence of these species in a British Columbia fjord. Favorable growth conditions determined for C. didymus, D. brightwellii, and T. rotula were similar to conditions present when these species were commonly found in the fjord.  相似文献
