The pretreatment of Saccharomyces cerevisiaeand Debaryomyces vanrijiwith sodium azide was found to induce thermotolerance in both yeasts, whereas sodium azide used in combination with heat shock enhanced the thermotolerance of S. cerevisiaeand substantially decreased the thermotolerance of D. vanriji.It is suggested that the different responses of the yeasts to sodium azide during heat shock are due to the different functional organizations of their mitochondrial apparatus.  相似文献

The investigation of the effect of the cytochrome oxidase inhibitors sodium cyanide and sodium azide on the thermotolerance of the yeasts Rhodotorula rubra, Debaryomyces vanriji, and Saccharomyces cerevisiae showed that these inhibitors diminish the thermotolerance of R. rubra and D. vanriji, but do not affect the thermotolerance of S. cerevisiae. Taking into account the fact that, unlike the latter yeast, R. rubra and D. vanriji are nonfermentative yeasts, the difference in the effects of the inhibitors on the yeast thermotolerance can be readily explained by the different types of glucose utilization (either oxidative or fermentative) in these yeasts. The data obtained also provide evidence that there is a correlation between the functional activity of mitochondria and the thermotolerance of yeast cells.  相似文献

The study of the growth of the yeasts Rhodotorula rubra, Saccharomyces cerevisiae, and Debaryomyces vanriji at elevated temperatures and their survival after transient lethal heat shock showed that the ability of these yeasts to grow at supraoptimal temperatures (i.e., their thermoresistance) and their ability to tolerate lethal heat shocks (i.e., their thermotolerance) are determined by different mechanisms. It is suggested that the thermotolerance of the yeasts is mainly determined by the division rate of cells before their exposure to heat shock.  相似文献

The effect of sodium azide in heat shock-induced cell death was studied in Debaryomyces vanrijiae, Candida albicans, and Saccharomyces cerevisiae yeasts. The results presented demonstrate that the azide addition induced a drastic decrease in the thermotolerance of glucose-grown D. vanrijiae. In contrast, glucose-grown S. cerevisiae and C. albicans cells treated with NaN3 became more resistant to heat shock than control cells. Nevertheless, in galactose medium the decrease of thermotolerance of S. cerevisiae and C. albicans cells was observed in the presence of sodium azide. It was suggested that the decreasing effect of sodium azide on thermotolerance takes place only when the yeast cell is incapable of using fermentation for ATP synthesis and obtains energy via oxidative phosphorylation. Yeast were isolated from the intestine of farmed rainbow trout (Salmo gairdneri), turbot (Scophtalmus maximus), and free-living flat-fish (Pleuronectes platessa and P. flesus). The average number of viable yeasts recovered from farmed rainbow trout was 3.0 × 103 and 0.5 × 102 cells per gram homogenized intestine for white and red-pigmented yeasts, respectively. The dominant species were Debaryomyces hansenii, Saccharomyces cerevisiae, Rhodotorula rubra, and R. glutinis. In 5 of 10 free-lving marine fish, > 100 viable yeast cells per gram intestinal mucus were recovered. Red-pigmented yeasts dominated and composed >90% of the isolates. Colonization experiments were performed by inoculating rainbow trout and turbot with fish-specific, isolated yeast strains and by examining the microbial intestinal colonization at intervals. Inoculation of experimental fish with pure cultures of R. glutinis and D. hansenii HF1 yielded colonization at a level several orders of magnitude higher than before the inoculation. Up to 3.8 × 104, 3.1 × 106, and 2.3 × 109 viable yeast cells per gram intestine or feces were recovered in three separate colonization experiments. The high level of colonizing yeasts persisted for several weeks. The concentrations of yeasts in the tank water never exceeded 103 viable cells per milliliter. No traces of fish sickness as a result of high yeast colonization were recorded during any of the colonization experiments. For periods of the experiments, the concentration of aerobic bacteria in the fish intestine was lower than the intestinal yeast concentration. Scanning electron microscopy studies demonstrated a close association of the yeasts with the intestinal mucosa. The mucosal colonization was further demonstrated by separating intestinal content, mucus, and tissue. All compartments were colonized by >103 viable yeast cells per gram. The study of the effect of malonate (an inhibitor of the succinate dehydrogenase complex of the respiratory chain of mitochondria) on the thermotolerance of the fermentative Saccharomyces cerevisiae and nonfermentative Rhodotorula rubra yeasts showed that malonate augmented the damaging effect of heat shock on the yeasts utilizing glucose (or other sugars) by means of oxidative phosphorylation. At the same time, malonate did not influence, and sometimes even improved, the thermotolerance of the yeasts utilizing glucose through fermentation. The suggestion is made that cell tolerance to heat shock depends on the normal functioning of mitochondria. On the other hand, their increased activity at elevated temperatures may accelerate the formation of cytotoxic reactive oxygen species and, hence, is not beneficial to cells.  相似文献

The yeast community associated with deep-sea hydrothermal systems of the Mid-Atlantic Rift was surveyed for the first time. This study relied on a culture-based approach using two different growth media: a conventional culture medium for yeasts supplemented with sea salts (MYPss) and the same medium additionally supplemented with sulfur (MYPssS). For the evaluation of species diversity, a molecular approach involving minisatellite-primed polymerase chain reaction (MSP-PCR) strain typing and sequence analysis of the D1/D2 domains of the 26S rDNA was followed. In the seven water samples that were studied, the number of colony-forming units per liter (cfu/L) ranged from 0 to 5940. The nonpigmented yeasts were much more abundant than the pink-pigmented ones. This disproportion was not observed in studies of other marine systems and may be due to the unique conditions of hydrothermal vents, characterized by a rich animal and microbial diversity and therefore by the availability of organic compounds utilizable by yeasts. Higher counts of nonpigmented yeast were obtained using MYPss, whereas for pink yeasts, higher counts were obtained using MYPssS. Moreover, among pink yeasts, some of the MSP-PCR classes obtained were composed of isolates obtained only on MYPssS, which might be an indication that these isolates are adapted to the ecosystems of the hydrothermal vents. Twelve phylotypes belonged to the Ascomycota and seven phylotypes belonged to the Basidiomycota. The nonpigmented yeasts were identified as Candida atlantica, C. atmosphaerica, C. lodderae, C. parapsilosis, Exophiala dermatitidis, Pichia guilliermondii, and Trichosporon dermatis, whereas the pigmented yeasts were identified as Rhodosporidium diobovatum, R. sphaerocarpum, R. toruloides, and Rhodotorula mucilaginosa. Some of the yeasts that were found belong to phylogenetic groups that include species reported from other marine environments, and eight phylotypes represent undescribed species. The new phylotypes found at Mid-Atlantic Ridge hydrothermal fields represent 33% of the total number of yeast taxa that were found.  相似文献

Four formaldehyde-resistant yeasts were isolated from soil. Three were tentatively identified as Debaryomyces vanriji and one as Trichosporon penicillatum. These yeasts almost completely consumed formaldehyde at 0.15 to 0.55% in growth medium containing glucose as carbon source, but the carbon of formaldehyde was not incorporated into the cell constituents. In formaldehyde-containing medium, yeast growth occurred after formaldehyde consumption. The yeasts showed relatively high activities of formaldehyde dehydrogenase, S-formylglutathione hydrolase and formate dehydrogenase. The resistance to formaldehyde is attributed to detoxification by oxidation.  相似文献

Summary At two locations in the Sonoran Desert, yeasts were sampled from species of Drosophila, the flies' cactus hosts, and other neighboring sources of cactophilic yeasts to determine the relation between the yeasts vectored by the fly and the yeasts found in their breeding sites. D. mojavensis, D. nigrospiracula, and D. mettleri vectored yeast assemblages significantly more similar to the yeast species found on the rot from which the flies were collected than to the yeasts found on other rots from the flies host cactus or other rotting cactus at the same site. Rots with Drosophila had fewer yeast species than those without flies, suggesting that flies were associated with younger rots. Rots with flies and the Drosophila also had more yeast species with the capability to produce ethyl acetate than rots without flies. The results support the contention that cactophilic Drosophila feed on a subset of the yeasts available in an area, and may act to maintain differences among the yeast communities found on different species of cactus.  相似文献

Phosphatase activities of yeasts belonging to the genera Saccharomyces, Kluyveromyces and Rhodotorula were studied. Rhodotorula rubra exhibited activities at acid, neutral and alkaline pH; the other yeasts only had activity at acid pH. Growing yeasts in a constant pH (4.5) medium decreased phosphatase activities in Saccharomyces and Kluyveromyces, while neutral activity was enhanced in Rhodotorula rubra which excreted more enzyme under these conditions. Washing cells with sucrose solutions lowered phosphatase activities in all yeasts, due to enzyme liberation. Acid phosphatase activities in isolated and purified cell walls were very small. Phosphatases thus appear not to be strongly bound to yeast cell walls.  相似文献

The thermophilic bacterium Bacillus sp. strain TB-1 was isolated in association with the yeast Debaryomyces vanriji from hot springs at 46°C. It was shown that TB-1 excreted thiamine into the culture broth, which not only promoted D. vanriji growth in mixed culture but also increased the maximal temperature for yeast growth.  相似文献

The yeast diversity in a paleo-karstic lake of the Lagoa Santa plateau was studied during March 1986–March 1987. Water samples were collected monthly at five stations and the yeasts were isolated at 25 °C on Sabourad dextrose agar supplemented with 0.5% of yeast extract and 10 mg% of chloramphenicol. August and February showed the highest numbers of isolates, coinciding, respectively with the beginning and the ending of stratification. Of 56 isolated species, Aureobasidium pullulans, Candida famata, Cryptococcus albidus, Cryptococcus laurentii, Rhodotorula glutinis, Rhodotorula rubra and Trichosporon cutaneum occurred at the highest frequencies. A. pullulans showed greatest prevalence during the dry period (winter), whereas C. famata, Cr. albidus, Cr. laurentii and Rh. rubra were prevalent during the rainy season (summer). Tr. cutaneum was distributed between July and November. Most isolates yeasts are associated with plants and soils, and are important in decomposition of aquatic macrophytes. Furthermore, a possible role of these species as indicators of pollution is discussed.  相似文献

Summary Soil samples from the Schirmacher Oasis of Antarctica were examined for the presence of yeasts. The number of yeast cells in the samples varied between 600 and 3000 g of wet soil. Eight pure cultures were obtained. All of the eight isolates were nonfermentative and all but one was Diazonium Blue B (DBB)-positive, indicating their basidiomycetous affinity. Based on their morphology, reproductive behavior, growth at different temperatures and salt concentrations, nutritional characteristies and various biochemical tests, all the eight cultures have been identified. Three of them are red yeasts, Rhodotorula rubra, one Bullera alba, one a dimorphic, Candida humicola and one Candida famata. The remaining two cultures have been tentatively identified as Candida ingeniosa and Candida auriculariae.  相似文献

Summary The growth and ethanol production by the d-xylose-fermenting yeasts Pichia stipitis and Pachysolen tannophilus under various conditions of aerobiosis responded similarly to the addition of the respiratory inhibitors potassium cyanide (KCN), antimycin A (AA), sodium azide and rotenone. However, the d-glucose-fermenting yeast Saccharomyces cerevisiae differed markedly from these yeasts in response to the inhibitors. In general the growth of the d-xylose-fermenting yeasts was inhibited by the respiratory inhibitors while ethanol production was either stimulated (especially when oxygen was available) or unaffected or inhibited by rotenone or AA or KCN and sodium azide, respectively. However, by exception KCN and AA stimulated ethanol production under aerobic conditions by Pichia stipitis and Pachysolen tannophilus respectively. Stimulatory or inhibitory effects by respiratory inhibitors were less marked in S. cerevisiae. These data suggest that unimpaired mitochondrial function is necessary for growth on d-xylose and optimal d-xylose fermentation. A requirement for membrane generated energy during d-xylose utilisation is indicated by 2,4-dinitrophenol inhibition of growth and fermentation.  相似文献

A rapid molecular identification technique was applied on microbial microflora isolated from Brazilian cassava roots given a yeast profile presented in the samples analyzed. A total of 24 strain isolated from cassava were initially grouped and identified in five groups using restriction-fragment length polymorphism (RFLPs) of 5.8S-ITS rDNA region. Sequencing analysis of the domains D1 and D2 of the 26S rRNA gene or 5.8S rRNA-ITS region were used to identify different groups of yeasts. Representative colonies of yeasts of each group were isolated and identified as Debaromyces hansenii, Kodamaea ohmeri, Candida glabrata, C. haemulonii, and Pichia gullhermondii. It is hoped that these results will contribute toward selecting yeast from this microflora capable to degrade cassava starch in the near future.  相似文献

浙江雁荡山山脉叶栖酵母菌资源与物种多样性   总被引:1,自引:0,他引:1  
对北雁荡山、中雁荡山、南雁荡山、西雁荡山和东雁荡山等广阔区域内的叶栖酵母菌资源进行考察,旨在弄清雁荡山山脉叶栖酵母菌资源和多样性。采用PDA平板对雁荡山地区的叶栖酵母菌资源进行分离和纯化,根据酵母菌培养物26S rDNA D1/D2序列和系统发育关系明确酵母菌的分类地位,分析该地区叶栖酵母菌群落的多样性、组成和分布情况。结果发现,分离自雁荡山山脉的1118株叶栖酵母菌可以被鉴定为56个已知类群和19个疑似新种,其中38种酵母菌在该地区比较常见。常见的酵母菌类群中,包括Derxomyces属5种,Sporobolomyces属3种,Bannoa、Bulleribasidium、Rhodosporidiobolus、Symmetrospora、Taphrina、Tilletiopsi和Udeniomyces属各2种,Bullera、Coniochaeta、Coniosporium、Cryptococcus、Elsinoe、Erythrobasidium、Fellozyma、Golubevia、Kockovaella、Kondoa、Leucosporidium、Moesziomyces、Oberwinklerozyma、Phyllozyma、Ruinenia和Saitozyma属各1种。根据常见酵母菌类群的相对频率,在该地区分离到的叶栖酵母菌中Derxomyces mrakii具有明显分布优势,并探讨了Derxomyces mrakii株间26S rDNA D1/D2序列的稳定性和变异幅度。根据雁荡山地区常见叶栖酵母菌组成和相似程度,可以推测地理上相近地区的酵母菌物种分布具有更大相似性。基于雁荡山山脉常见叶栖酵母菌种类和数量而绘制出的物种累积曲线,证明雁荡山地区绝大多数的叶栖酵母菌种类已经被分离到,研究结果可以真实反映该地区叶栖酵母菌资源和物种多样性的存在状况。  相似文献

The action mechanism of the mitochondrial inhibitor sodium azide on thermotolerance in Saccharomyces cerevisiae was studied. At ambient growth temperature, pretreatment with sodium azide was shown to improve the thermotolerance of parent cells and the hsp104 mutant. Treating with the inhibitor during a mild heat shock suppressed the development of induced thermotolerance due to the inhibition of heat shock protein (Hsp104) synthesis. Treating with the inhibitor immediately before lethal heat shock produced a variety of effects on thermotolerance depending on whether the yeast metabolism was oxidative or fermentative. The conclusions are: (1) the protective effect of sodium azide on the thermotolerance of S. cerevisiae cells grown on glucose-containing medium is not related to Hsp104 functioning, and (2) the mechanisms of basic and induced thermotolerance differ considerably.  相似文献

The growth of yeast strainsSaccharomyces cerevisiœ, Rhodotorula rubra andBullera alba isolated from natural lake microflora was examined in the presence of prehydrolysis lignin and/or its water-soluble derivative. The stimulation effect of the water-soluble lignin derivative was higher in comparison with that of unmodified lignin. The structural changes of the lignin macromolecule by the yeasts determined by IR spectroscopy indicate oxidative degradation and demeth(ox)ylation, similar to that found with lignin-degrading hyphal fungi. The results indicate a partial utilization of lignin by the yeasts as carbon source.  相似文献

The fungal pathogen Botrytis cinerea causes severe rots on tomato fruit during storage and shelf life. Biological control of postharvest diseases of fruit may be an effective alternative to chemical control. Yeasts are particularly suitable for postharvest use, proving to be highly effective in reducing the incidence of fungal pathogens. Yeast fungi isolated from the surface of solanaceous plants were evaluated for their activity in reducing the postharvest decay of tomato caused by B. cinerea. Of 300 isolates, 14 strains of Rhodotorula rubra and Candida pelliculosa were found to be strongly antagonistic to the pathogen in vitro and were selected for further storage experiment. The antagonists were evaluated for their effect on the biological control of postharvest grey mould. Artificially wounded fruits were treated by means of a novel technique: small sterile discs of filter paper imbibed separately in suspensions of each yeast and the pathogen were superposed onto each wound. After 1‐week, 11 isolates were significantly effective in reducing the diameter of lesions by more than 60% compared to the control treated with B. cinerea alone. Total protection was obtained with the strain 231 of R. rubra on fruits challenged with pathogen spores. To our knowledge, R. rubra and C. pelliculosa have not been described as biocontrol agents against grey mould caused by B. cinerea. Our data demonstrate that the application of antagonistic yeasts represents a promising and environmentally friendly alternative to fungicide treatments to control postharvest grey mould of tomato.  相似文献

Summary The yeast communities from slime fluxes of three deciduous trees (Prosopis juliflora, Populus fremontii and Quercus emoryi) and the necroses of two cacti (Opuntia phaeacantha and Carnegiea gigantea) were surveyed in the region of Tucson, Arizona. In addition, the yeasts carried by dipterans associated with the fluxes or necroses (Drosophila carbonaria, D. brooksae, D. nigrospiracula, D. mettleri, and Aulacigaster leucopeza) were sampled. The results indicate that each host sampled had a distinct community of yeasts associated with it. The dipterans, which can act as vectors of the yeasts, deposited yeasts from other sources in addition to those found on their associated hosts. It is argued that host plant physiology is relatively more important than the activity of the vector in determining yeast community composition. Furthermore, the average number of yeast species per flux or necrosis is not different from the average number of yeast species per fly. It is hypothesized that the vector may affect the number of species per individual flux or not, and that the number is lower than the rot or necrosis could potentially support.  相似文献
