Reovirus is a double-stranded RNA-virus which induces myocarditis in newborn mice. Due to the large diameter of the viral particles (70-75 nm) it can be detected by electron microscopy. Subcutaneous inoculation of 0.05 ml reovirus type 3 (TCID50-titer: 10(8.5)/ml) into newborn NMRI-mice (12-18 h after birth) caused a grey-yellow mottling on the ventricular surface first seen on the 5th day after birth. At the same time muscle fiber necrosis was observed which increased with time. Electron microscopic investigations of the diseased heart muscle disclosed a marked interstitial oedema, swelling of the tubular system and sarcoplasmic reticulum, and degenerative changes in the mitochondria of individual myocardiocytes as early as the 2nd post-inoculation day. Simultaneously, an enlarged Golgi-apparatus and an increasing number of lysosomes, partially exhibiting acid phosphatase activity, was detected in the perinuclear region of ventricular myocardiocytes. On the 5th day after infection, viruses were detected either within single membrane vesicles, dispersed in cytoplasm or as aggregated clusters in the perinuclear region. These in vivo electron microscopic findings correspond with observations of virus propagation in cell-culture systems.  相似文献

Summary Labelling of renal tubule cytosomes with electron dense iron granules can be attained by daily intramuscular injections to mice of an iron sorbitol citric-acid compound in a total of approximately 50 mg Fe+++/100 g of body weight. The labelled cytosomes correspond to secondary lysosomes and represent heterolysosomes or ambilysosomes. The evidence suggests that tubule and lysosome function are undisturbed by the labelling procedure. The use of this method for fine structural studies of the interaction between secondary lysosomes and other cytoplasmic organelles and elements is indicated.Microbodies do not incorporate administered Fe+++. A strain of food-poisoning bacterium has been isolated by Jin Jiexiang (1963) in China from the fermented cornflour that has gone bad. These pathogenic microorganism has been identified and named Pseudomonas by Zhao Naixin in 1988, which is the same species as P. cocovenenans. The characteristics of them were conformed to these of the species P. cepacia of section 2 of the genus Pseudomonas. In view of the fact that the fine structures of the above mentioned three strains of Pseudomonas have not been described yet, we decided to observe them with electron microscope. Results indicate there are many things in common among the three strains, such as: appearing short rods, 0.6-0.8 microns in diameter by 1.5-2.0 microns in length, one polar multiflagella; non-pili, non-capsules, non-endospores; containing intranuclear inclusions (electron-dense bodies or concentric laminae bodies), accumulating intracytoplasmic PHB granules; forming filaments, minicells and bizarrecells; producing extracellular cellulose-like materials by the three strains have not been reported previously.  相似文献

To clarify the pathogenesis of Bordetella in vivo infection, the tracheal epithelia of mice were examined in detail by electron microscopy at various intervals after intranasal inoculation with graded doses of phase I Bordetella bronchiseptica. In mice infected with a lethal dose (6 to 7 x 10(7) CFU), a remarkable rupture of the cell membranes of cilia and microvilli of the middle trachea was found on day I postinfection. The rupture of the membrane was observed over the entire tracheal epithelia, on day 2 after infection. The affected cilia were constricted at the transitional region and were broken off. In the ciliated cells the adherence of organisms to ciliary apexes and colonization in the interciliary spaces were also remarkable. In both the ciliated and nonciliated epithelial cells, the cytoplasmic vacuolation and pyknosis or karyorrehexis were also notable. In mice infected with one-tenth of the lethal dose, similar findings were seen, but appeared more slowly and the bacteria were not seen attaching to ciliary apexes. In mice receiving one-hundredth of the lethal dose, only mild cilial abnormality such as aggregation of cilia, and slight cytoplasmic vacuolation were found 6 days postinfection. Based on these findings, a possible mechanism of the ciliary damages produced by B. bronchiseptica was postulated.  相似文献

Three strains of B. thetaiotaomicron have been isolated from caeca of BALB/c-nu/+ mice of SPF. These Bacteroides are obligately anaerobic, Gram-negative, non-spore-forming and non-motil without flagella rods. A characteristic of staining is deep at ends and stainless at the medium of a rod. The isolation and identification of these strains has been reported in 1987. This paper introduced only the results of EM observations. Under the SEM, the unstained area of rods is always showing a concavity, which is just a nucleoid in sections under the TEM. Many lamellar corpuscles have been found in cell plasma. Some of them have been secreted out of the cells. The chemical properties and physiological functions of them are unknown.  相似文献

Protein synthesizing activity of the rat hypothalamic arcuate nucleus following partial or total deafferentation of the medial basal hypothalamus was studied by light and electron microscopic autoradiography when administering tritiated leucine into the lateral ventricle. There were significantly more grains over the arcuate nucleus 21 days after disconnection of this hypothalamic region than over the intact nucleus. Isolation of a temporal cortical region induced similar changes in the isolated area, although this effect was not so pronounced as in the arcuate region. Data suggest that the protein synthesizing activity of arcuate neurons increases significantly after interruption of neural connections of the medial basal hypothalamus. It is assumed that the effect is primarily due to transneuronal alteration and/or interruption of inhibitory afferents.  相似文献
