To improve the application of essential oils as natural antimicrobial preservatives, the objective of the present study was to determine physical, antimicrobial, and biophysical properties of eugenol after nanoencapsulation by sodium caseinate (NaCas). Emulsions were prepared by mixing eugenol in 20.0 mg/mL NaCas solution at an overall eugenol content of 5.0–137.9 mg/mL using shear homogenization. Stable emulsions were observed up to 38.5 mg/mL eugenol, which had droplet diameters of smaller than 125 nm at pH 5–9 after ambient storage for up to 30 days. The encapsulated eugenol had similar minimal inhibitory and minimal bactericidal concentrations as free eugenol against Escherichia coli O157:H7 ATCC 43895, Listeria monocytogenes Scott A, and Salmonella Enteritidis but showed better inhibition of E. coli O157:H7 than free eugenol during incubation at 37 °C for 48 h. After 20 min interaction at 21 °C, bacteria treated with encapsulated eugenol had a greater reduction of intracellular ATP and a greater increase of extracellular ATP than free eugenol, suggesting the enhanced permeation of eugenol after nanoencapsulation. However, such overall trend was not observed when examining bacterial morphology and uptake of crystal violet, suggesting the possible membrane adaptation. Findings from this study showed the feasibility of preparing nanoemulsions with high loading of eugenol using NaCas.  相似文献

The ultrasound-assisted extraction (UAE) parameters of total water-soluble polysaccharides (TABPs) from Acanthopanaxbrachypus fruit were optimized by Box-Behnken design (BBD) and response surface methodology (RSM). Physicochemical, structural, and functional properties of TABPs were investigated by chemical analysis, inductively coupled plasma optical emission spectrometry (ICP-OES), high performance liquid chromatography (HPLC), scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA), water-holding capacity (WHC), oil-holding capacity (OHC), emulsion capacity (EC), emulsion stability (ES), as well as DPPH. and ABTS.+scavenging assays. The results showed that the maximal UAE-yield of TABPs was 3.81±0.18 % under the optimal conditions (ultrasonic power 325 W, extraction temperature 47 °C, extraction time 22 min). TABPs was rich in some beneficial element (Mg, K, Fe, Zn and Na) but little in harmful elements (Hg, Cd, As and Pb), and displayed rough surface with flake-like features and large dents, contained 93.89±0.08 % of total carbohydrate with more different monosaccharides including glucose, galactose, rhamnose, arabinose, mannose, xylose, and uronic acid in a molar ratio of 8.83 : 7.90 : 4.74 : 4.55 : 2.80 : 2.39 : 1.00, respectively. TABPs exhibited broad weight distribution (11.2–133.5 kDa), excellent thermal stability (>280 °C), WHC (0.61±0.08 g water/g sample) and OHC (4.53±0.12 g oil/g sample), as well as higher EC (43.75±1.23 %) and ES (38.32±1.50 %). Furthermore, TABPs also displayed remarkable scavenging activities on DPPH. and ABTS.+ in vitro. These findings provide a scientific basis for the applications of TABPs in functional additives for food, medicine, and cosmetics.  相似文献

The interactions between sodium caseinate (NaCas) and basil seed gum (BSG) in the presence of calcium chloride (CaCl2) were investigated. The phase behavior of the mixed aqueous dispersions and their gels revealed a homogeneous mixture, obtained at the higher concentrations of both CaCl2 and BSG. The Herschel-Bulkley model sufficiently fitted the flow behavior of the mixture solution data. Apparent viscosity increased significantly (p < 0.05) by increasing the concentration of BSG, where the addition of CaCl2 had no significant effect on the viscosity of the samples (p > 0.05). Furthermore, there was an increase in thixotropy due to the higher concentrations of BSG and CaCl2. Based on the frequency sweep test, at the low frequencies, a more gel-like behavior was observed in the case of the higher concentrations of either BSG or CaCl2. The rheological and SEM data suggested that the stronger structure of NaCas-BSG gel in the presence of the higher concentrations of CaCl2 was related to the induction of complex formation between the two biopolymers.

采用罗氏泡沫仪和流变仪对海藻酸钠(NaAlg)/十二烷基聚氧乙烯醚硫酸钠(AES)复配体系的起泡稳泡性能及流变行为进行研究,以探索NaAlg对AES起泡稳定性的影响及作用机理。泡沫性能试验显示,NaAlg的加入,提高了AES溶液的起泡能力及泡沫稳定性,溶液的pH值由7.0降到5.0,混合体系的起泡稳泡性明显增加。流变性研究展示,随着NaAlg浓度的增加,溶液的粘度呈增大的趋势,溶液总体呈现牛顿流体性质,当NaAlg浓度大于0.5%时,低剪切速率区,溶液仍呈现牛顿流体性质,而在高剪切速率区,有剪切变稀的现象出现。总体来看,各试液的G’相似文献

目的探讨油橄榄叶提取物(OLE)对铅中毒小鼠视网膜组织结构及抗氧化能力的影响。方法选健康小鼠,每日灌胃醋酸铅溶液的同时灌胃不同剂量的OLE进行治疗,连续用药30d,观察小鼠视网膜结构的变化,检测视网膜铅含量、超氧化物歧化酶(SOD)、谷胱苷肽过氧化物酶(GSH-PX)、过氧化氢酶(CAT)活性及丙二醛(MDA)含量。结果与模型对照组相比,小鼠灌胃OLE后视网膜总厚度、外核层厚度及其细胞数量、内核层厚度及其细胞数量增加,视网膜铅含量下降,SOD、GSH-PX和CAT活性升高,MDA含量降低。结论 OLE对铅中毒小鼠有一定的疗效,能影响视网膜结构及抗氧化能力,阻止铅对视网膜的毒性。  相似文献

A full understanding of the molecular mechanism of ion transport and energetics of the Na,K-ATPase will require both structural and functional data. During recent years biophysical methods have provided a number of important pieces of information on ion binding and release and the charge transfer process. This allows the formulation of kinetic models of the transport process. Although a breakthrough has not been obtained due to the lack of detailed knowledge on the three-dimensional structure with a resolution high enough to identify the ion-binding sites and the transport pathway, the functional information has structural implications that create constraints on possible mechanisms of active transport. The influence of actively/passively encapsulated oleuropein on DPPC liposomes thermal and structural properties, and its antioxidant capacity against lipid peroxidation were investigated. Also, an oleuropein-rich olive leaf extract was encapsulated in soy phosphatidylcholine (PL-90 g) and incorporated in model and commercial drinks. Oleuropein induced a concentration-dependent broadening and splitting of the gel-to-liquid phase transition temperature. Fluorescence measurements revealed a fluidizing effect on liposomes below their gel-to-liquid phase transition temperature, and a higher lipid ordering above, especially to active encapsulation. Oleuropein also showed an antioxidant effect against lipid peroxidation in PL-90 g liposomes. PL-90 g Liposomes with olive leaf extract showed a mean diameter of 405 ± 4 nm and oleuropein encapsulation efficiency of 34% and delayed oleuropein degradation at pH 2.0 and 2.8 model drinks. In conclusion, greater effects were observed on the structure and fluidity of DPPC liposomes when oleuropein was actively encapsulated, while its incorporation into acidic foods in encapsulated form could enhance its stability.

本文观察了番茄叶水提物对小鼠的急性毒性及其对脂多糖诱导急性炎症模型大鼠的影响。采用经典的急性毒性试验方法,观察小鼠口服给予番茄叶水提物的死亡率,按寇氏法计算半数致死量(LD50)。50只SD大鼠随机分为正常组、模型组、阳性药泼尼松5 mg/kg组与番茄叶水提物3.19、1.59 g/kg组,连续给药10 d,每天一次。以腹腔注射脂多糖(LPS)建立急性炎症模型,ELISA法测定血清白细胞介素1(IL-1)、白细胞介素6(IL-6)、肿瘤坏死因子(TNF-α)。结果显示,番茄叶水提物单次灌胃给药的LD50为46.44g/kg,95%可信限为39.52~54.60 g/kg。3.19 g/kg番茄叶水提物可明显降低LPS诱导的急性炎症小鼠血清IL-1、IL-6和TNF-α水平。番茄叶水提物抑制LPS诱导急性炎症的作用机制可能与其降低血清炎症因子水平有关。  相似文献

目的观察油橄榄叶提取物(olive leaf extract,OLE)联合美沙酮对海洛因依赖大鼠肾功能及活性物质的影响。方法将40只健康大鼠随机分成正常组、模型组、OLE+美沙酮处理组、美沙酮处理组。按递增法采用皮下注射海洛因建立海洛因依赖大鼠模型,造模后药物处理30 d,自动生化分析仪分析血浆尿酸(UA)、尿素氮(UN)、肌酐(Cr)含量,比色法检测肾脏组织一氧化氮(NO)含量、一氧化氮合酶(NOS)活性,免疫组织化学法检测肾脏组织中环氧化酶-2(COX-2)的表达情况。结果与正常组相比,模型组海洛因依赖大鼠血浆UA、UN、Cr含量明显增高,肾脏组织中NO含量和NOS活性明显降低,COX-2的表达则显著升高;与模型组相比,OLE+美沙酮处理组和美沙酮处理组大鼠血浆UA、UN、Cr含量明显降低,肾脏组织中NO含量和NOS活性升高,COX-2表达明显减少。OLE联合美沙酮对海洛因依赖大鼠肾脏的治疗效果比美沙酮处理组更明显。结论 OLE联合美沙酮对海洛因依赖大鼠肾功能损害有明显的治疗作用,机制可能与其降低血尿酸、减少尿酸盐在肾小管沉积、升高肾组织NO含量和NOS活性、抑制COX-2的表达有关。  相似文献

Several red wines were studied to find a correlation between the physicochemical parameters characterizing the antioxidant status of wine and the total content of phenols in samples. The content of dissolved oxygen (its value varied from 0.75 to 3.28 mg/ml), pH (3.10–3.63), redox potential (–186 to –106 mV), mass concentration of free and total sulfur dioxide (10–30 and 36–200 mg/dm3, respectively), absorption spectra, and total phenol content were determined. The wines fell into two main groups—with a relatively low (1850–2050 mg/dm3) and high (2300–2900 mg/dm3) content of polyphenols. It was demonstrated that the physicochemical parameters (except for the content of sulfur dioxide) correlate with the total phenol content in the wines studied.  相似文献

Cancer protection associated with the consumption of olive products is well established, but not for leukemia. The protective effects of olive (Olea europaea L.) leaves were investigated by incubating human promyelocytic leukemia HL-60 cells with olive leaf extracts (OLEs) from seven principal Tunisian olive varieties, namely, Chemchali, Chemlali, Chétoui, Gerboui, Sayali, Zalmati and Zarrazi. The results showed significant growth inhibition of HL-60 cells incubated for 48 h with a 100-fold dilution of each OLE which had been obtained by incubating 10 g of dried leaves in 100 ml of 70% ethanol for one week with subsequent ultrafiltration. DNA fragmentation was observed in the cells incubated for 19 h with a 100-fold dilution of the Chemchali, Chemlali and Zalmati extracts. The results of a nitroblue tetrazolium (NBT) assay revealed NBT reduction, a differentiation marker, by the OLE-treated cells after an overnight incubation. The Gerboui extract showed the highest NBT reduction ability at more than 90%. An HPLC analysis revealed the presence of apigenin 7-glucoside in the extract, which was found in subsequent experiments to be responsible for the Gerboui extract-mediated cell differentiation.  相似文献

目的:以青橄榄为原料研究提取分离黄酮类化合物的方法与条件,建立最佳提取工艺。方法:采用醇提法,通过单因素试验考察浸提溶剂浓度、浸提温度、浸提时间等三个因素对青橄榄中黄酮类化合物提取效率的影响,同时采用不同显色方法及紫外、红外测定其性质。结果:在单因素的基础上,通过正交试验L9(34),得三因素中乙醇浓度的影响最大,并初步判断了提取物的性质。结论:青橄榄中黄酮类化合物最佳提取工艺为:乙醇浓度60%,浸提温度70℃,浸提时间120min,提取率达19.87mg/g。另外初步判断提取物中可能含有查尔酮类化合物。  相似文献

Sodium caseinate is a commonly used emulsifier in foods, as it adsorbs on the surface of oil droplets and stabilizes them via electrostatic and steric stabilization, forming a polyelectrolyte layer at the interface. Since the protein interface is affected by varying environmental conditions such as pH, ionic strength, concentration of unadsorbed polymers, these emulsions are prone to a variety of destabilization mechanisms. The objective of the present work was to observe the destabilization of sodium caseinate stabilized oil in water emulsions using electroacoustic spectroscopy. This technique can be utilized for the characterization of concentrated colloidal systems in situ, without dilution. The electroacoustic and ultrasonic properties of soy oil in water emulsions were determined for sodium caseinate stabilized emulsions under conditions known to cause destabilization. Ultrasonic attenuation and electrophoretic mobility (ζ-potential) could clearly follow the changes occurring in the emulsion droplets, under minimal sample disruption. This is critical for these systems in a very fragile, metastable state. The emulsions were stable to the addition of high methoxyl pectin (HMP) up to 0.1% HMP. Addition of free sodium caseinate induced depletion flocculation, causing a decrease in the attenuation and electrophoretic mobility measured. The presence of HMP limited depletion interactions. Acidification of the emulsion droplets resulted in a clear sol–gel transition, as shown by a steep increase in the particle size and a decrease in attenuation. Again, destabilization was limited by HMP addition. It was concluded that ultrasonics and electroacoustics are suitable techniques to understand the details of the destabilization processes occurring to food emulsions, measured in situ.  相似文献

To encapsulate piperine (Pip), as a poor water-soluble bioactive compound, zein-sodium caseinate-xanthan gum (Z-SG-XG) nanocomplex was prepared as a colloidal delivery system. The effect of different parameters involved in complexation process, including concentration of proteins, polysaccharide, and Pip on the encapsulation efficiency of Pip, particle size and stability of the nanocomplexes was investigated. Powders obtained by freeze-drying of the colloidal solution had relatively uniform particles compared to those obtained from conventional drying system and showed well redispersibility in water. At the optimal condition, a stable and homogeneous nanocomplex with a mean particle size of 145.9 ± 2.7 nm, PDI of 0.27 ± 0.01, and ζ-potential of −39.7 ± 1.3 mV was obtained. The antioxidant activity of Pip was significantly improved by encapsulation into the Z-SC-XG nanocomplex. Also, the in vitro release of Pip from the synthesized nanocomplexes in phosphate-buffer saline (PBS) solution and simulated gastrointestinal fluids (SGIF) was investigated and the release kinetic was studied as well. The Pip/Z-SG-XG nanocomplex showed a slower release in SGIF compared to the free Pip and nanoparticles without XG and SC, while its antioxidant activity was remarkable. Results suggested a possible utilization of Z-SC-XG nanocomplex for improving the water solubility, bioavailability and storage stability of Pip.

The ethnomedicinal uses of the aqueous leaf extract of Andrographis paniculata Nees (AP) include treatment of pain and inflammation, malaria, asthma and common cold. We designed this study to characterize some effects of AP and those of its andrographolide constituent. Guinea pig tracheal rings suspended in organ baths containing PSS were precontracted with histamine or carbachol and then exposed to cumulative concentrations of AP, andographolide or theophylline. The effect of AP was tested in Ca2+-depleted tracheal rings stimulated with the EC50 of histamine in Ca2+-free PSS. IC50 and Emax values were calculated for each relaxant. Results showed that both AP and andrographolide possessed relaxant effects on the tracheal smooth muscle. While AP was more effective on histamine-induced contraction, andrographolide and theophylline were more effective on carbachol-induced contraction. The IC50 values of andrographolide were significantly higher than those of theophylline in the two contractile agents. The presence of AP significantly attenuated the contractile force produced by 6.4 x 10-3 M Ca2+ in Ca2+-depleted rings. It is concluded that andographolide contributes at least in part to the relaxant action of AP on tracheal smooth muscles. The mechanism of action is related to inhibition of Ca2+ influx into tracheal smooth muscle cells.  相似文献

The rheological attributes of polymers as wheat dough are strongly related to its microstructure. To quantify dough protein microstructure confocal laser scanning microscopy combined with image analysis was used. The effect of three experimental factors pH (addition of lactic acid and sodium hydroxide), water addition, and sodium chloride (NaCl) addition on empirical and fundamental rheological properties as well as microstructural protein properties were studied and modeled by applying a response surface methodology. The obtained models revealed high correlations between the experimental factors and the complex shear modulus (R 2?=?0.97), dough resistance (Rmax k; R 2?=?0.91) and stickiness (R 2?=?0.93). Furthermore it was possible to determine microstructural attributes as the area fraction (R 2?=?0.88) and Feret??s diameter (R 2?=?0.86) as a function of pH, water and NaCl addition. Especially measures of Rk max revealed highly significant correlations with the protein microstructure as the branching index (r?=?0.79).  相似文献

Snakebites are a serious public health problem due their high morbi-mortality. The main available specific treatment is the antivenom serum therapy, which has some disadvantages, such as poor neutralization of local effects, risk of immunological reactions, high cost and difficult access in some regions. In this context, the search for alternative therapies is relevant. Therefore, the aim of this study was to evaluate the antiophidic properties of Jatropha gossypiifolia, a medicinal plant used in folk medicine to treat snakebites. The aqueous leaf extract of the plant was prepared by decoction and phytochemical analysis revealed the presence of sugars, alkaloids, flavonoids, tannins, terpenes and/or steroids and proteins. The extract was able to inhibit enzymatic and biologic activities induced by Bothrops jararaca snake venom in vitro and in vivo. The blood incoagulability was efficiently inhibited by the extract by oral route. The hemorrhagic and edematogenic local effects were also inhibited, the former by up to 56% and the latter by 100%, in animals treated with extract by oral and intraperitoneal routes, respectively. The inhibition of myotoxic action of B. jararaca reached almost 100%. According to enzymatic tests performed, it is possible to suggest that the antiophidic activity may be due an inhibitory action upon snake venom metalloproteinases (SVMPs) and/or serine proteinases (SVSPs), including fibrinogenolytic enzymes, clotting factors activators and thrombin like enzymes (SVTLEs), as well upon catalytically inactive phospholipases A2 (Lys49 PLA2). Anti-inflammatory activity, at least partially, could also be related to the inhibition of local effects. Additionally, protein precipitating and antioxidant activities may also be important features contributing to the activity presented. In conclusion, the results demonstrate the potential antiophidic activity of J. gossypiifolia extract, including its significant action upon local effects, suggesting that it may be used as a new source of bioactive molecules against bothropic venom.  相似文献
