In experiments in three years, seed tubers were inoculated before planting with either Fusarium solani var. coeruleum or F. sulphureum to initiate a rot, or were contaminated by dipping in soil slurries containing spore suspensions of one or other of the pathogens. Transmission to progeny tubers was tested by uniformly wounding and incubating tubers and by dilution plating of soil samples. In two years, transmission of F. sulphureum was greater from highly contaminated than from rotting seed and was greater on cv. Pentland Crown than on cvs Desiree and Maris Piper. F. solani var. coeruleum appeared to be transmitted most readily from rotting seed and Maris Piper was the cultivar most extensively contaminated. In experiments with different harvest dates, transmission of both fungi from highly contaminated seed could be detected by late June or mid-July. More progeny tuber wounds rotted in F. sulphureum than in F. solani var. coeruleum plots and in one year, F. sulphureum caused more rots on cv. Record than on cv. Maris Piper. These differences between the pathogens may be related to their differing abilities to sporulate underground on the surface of seed tubers and on stem bases.  相似文献

Potato tubers artificially inoculated with Fusarium solani var. coeruleum or F. sulphureum 3 months after harvest were uniformly wounded and held at 5, 10 or 15°C for up to 21 days before immersion in fungicide suspensions. Holding tubers for 14 days at 15°C (curing conditions) or at 5°C did not alter the incidence of dry rot subsequently developing on tubers stored at 10°C, and holding tubers for up to 21 days at 15°C slightly increased disease caused by both pathogens. Thiabendazole, imazalil and prochloraz applied to tubers immediately after wounding almost completely prevented dry rot. Treatment after 3 days was less effective and the amount of disease increased with further delay; fungicides were more effective on tubers held at 5°C than at 10 or 15°C before treatment and storage, and efficacy of the fungicide was decreased by increasing the amount of inoculum on tubers. Wounds became less susceptible to infection by F. solani var. coeruleum and F. sulphureum when tubers were held at 15°C before inoculation, and the incidence of rots was decreased by 70–80% by delaying inoculation for 7 days. Treating tubers with dichlorophen immediately after wounding slightly increased the disease. The effects of fungicide treatment, curing conditions and wound healing are discussed.  相似文献

Dryopteris crassirhizoma dryocrassin ABBA treatment was tested for its effectiveness in controlling Fusarium solani var. coeruleum growth in vitro and for prevention of postharvest dry rot of potato tubers and slices. Dryocrassin ABBA strongly inhibited mycelial growth, resulting in reductions in both mycelium dry weight and per cent spore germination of F. solani var. coeruleum at concentrations of 2.0, 0.5 and 0.1 mg/ml. Scanning electron microscopy (SEM) observations showed that treatment induced abnormal, tightly twisted morphological changes in hyphae. Moreover, in vivo experiments demonstrated that dryocrassin ABBA treatment at 2 mg/ml effectively controlled dry rot of potato tubers inoculated with mycelial discs of F. solani var. coeruleum. After dryocrassin ABBA treatment, the content of soluble proteins, peroxidase (POD) and superoxide dismutase (SOD) activity increased, and malondialdehyde (MDA) content remained the stable situation. In addition, the expression level of plant lipid‐transfer proteins (LTPs) genes – StLTPa1, StLTPa7, StLTPb1 and StLTPb3 – was upregulated. These results collectively demonstrate that dryocrassin ABBA can inhibit growth of Fusarium pathogens to induce disease resistance. On the other side, dryocrassin ABBA maybe induce potato defence responses.  相似文献

Potato glycoalkaloids were determined in tubers of a wide range of genotypes in order to study their influence on resistance to storage rots caused by Fusarium solani var. coeruleum and Phoma exigua var. foveata. No relationships were found. This indicates that breeders selection for low glycoalkaloid potato clones is not in conflict with the demands for high levels of resistance to these pathogens. It was also observed that genotypes susceptible to skin-breaking types of, damage were more easily infected than genotypes susceptible to other types of damage.  相似文献

Potato seed tubers of seven cultivars derived from stem cuttings in 1965 (healthier seed) were grown in 1969–72 at two sites, one clay with flints soil (Rothamsted) and the other sandy loam soil (Woburn). Inoculating sprouted tubers at planting with Polyscytalum pustulans did not affect the number of stems/plant or total yield but increased stem base and tuber infection. The yields of large tubers (57–83 cm) were increased and small tubers (>57 cm) decreased, indicating a decrease in tuber numbers. Rhizoctonia solani inoculated at planting decreased numbers of stems/plant and yield by up to 14% at Woburn but not at Rothamsted. At both sites, yields of large tubers were increased with cvs Majestic and Record and decreased with Pentland Crown. Stem canker and tuber infection were increased but infection was also prevalent on tubers from non-inoculated seed at Woburn. When both pathogens were inoculated together yields of large tubers were increased in cvs King Edward, Majestic and Record and decreased in Pentland Crown. Infection of stem bases and tubers was sometimes less than when either pathogen was inoculated singly.  相似文献

Interactions between Fusarium solani and Phytophthora parasitica or F. solani and P. citrophthora influenced the development of root rot of citrus but depended on the temporal order of inoculation with F. solani or the two Phytophthora spp. Inoculation of citrus with either Fusarium solani and Phytophthora parasitica or Phytophthora citrophthora increased root rot compared to inoculation with P. parasitica or P. citrophthora alone when plants were inoculated with Phytophthora by dipping their roots in zoospore suspensions and subsequently transplanted into soil infested with F. solani. However, root rot was not increased by simultaneous co-inoculation of P. parasitica and F. solani or when plants were inoculated with F. solani first. Root rot was not increased when heat-stressed or non-stressed plants were inoculated with P. parasitica 30 days after transplanting into soil infested with F. solani. In most but not all experiments, F. solani alone reduced growth of tops or roots a small but significant amount.Co-inoculation of citrus by root-dipping into zoospore suspensions of P. parasitica and transplanting into soil infested with F. solani reduced feeder root length by 62% and root weight by 61% but did not significantly reduce the percentage of living roots when compared to inoculation with P. parasitica alone. When citrus roots were immersed in zoospore suspensions of P. citrophthora and transplanted into soil infested with F. solani, feeder root length was reduced by 68%, but feeder root weight and the percentage of living roots were not significantly reduced when compared to plants inoculated with P. citrophthora alone.Propagule densities of both P. parasitica and P. citrophthora in the rhizosphere of plants inoculated by root-immersion and then transplanting into soil infested with F. solani were not significantly different than propagule densities from plants transplanted into non-infested soil. Propagule densities of P. parasitica were suppressed an average of 41% when citrus was inoculated with P. parasitica 30 days after transplanting into soil infested with F. solani and by 41% when citrus was co-inoculated by transplanting into soil infested with both F. solani and P. parasitica.  相似文献

Stolons attached to developing potato tubers were inoculated with the soft rot bacterium Erwinia carotovora var. atroseptica. Almost all the stolons rotted, but soft rots developed in less than 10% of new tubers; the bacterium was isolated later from these tubers. No rots developed in the other tubers but the bacterium was later isolated from about half of them. It could not be isolated from tubers attached to inoculated stolons where the rot on them did not extend to the tuber or from tubers attached to stolons that were not inoculated though many of these rotted. The bacterium was reisolated from almost all arrested lesions in tubers inoculated 8 month earlier with E. carotovora var. atroseptica. Blackleg did not develop from plants grown fom these tubers under various soil conditions. It did develop in a large proportion of plants from tubers inoculated shortly before planting and grown in cool, wet soil. Less than 1% blackleg developed in plants grown from tubers from plants with blackleg or from plants immediately adjacent. The presence of pectolytic bacteria and E. caratovora var. atroseptica in seed and new tubers was investigated during June, July and August. Although E. caratovora var. atroseptica was obtained from c. 40% tubers, only c. 0·3% of c. 8400 plants developed blackleg. The bacterium was isolated from only three of 160 new tubers sampled during the summer.  相似文献

Rhizoctonia solani, the causal agent of stem canker and black scurf on potato, survives as sclerotia on tubers, in soil and in plant residues. The objective of the present study was to evaluate the importance of inoculum source on disease development. Disease‐free minitubers and seed tubers contaminated with low levels of R. solani were planted in fumigated or artificially inoculated growth mixture in greenhouse experiments. Black scurf incidence and severity were significantly higher when the inoculum was present in both seed tubers and soil, compared with either of them separately. The severity of disease symptoms on the subterranean parts of the plant also were significantly higher in plots where both seed tubers and soil were contaminated, compared with plots where the inoculum source was either the seed tubers or the soil. Thus, both major sources of inoculum, seed tubers and soil, are important in disease development. However, when both sources are present, black scurf incidence and severity are increased, leading to economical damage to tuber yield and quality. Additional results from field trials support these findings. Disease incidence and severity on daughter tubers were correlated with levels of contamination in seed tubers and soil. When seed tubers and soil were heavily infested, the levels of black scurf incidence and severity on daughter tubers were very high; when seed tuber and soil infestation were very low, black scurf incidence and severity on progeny were also lower. Disease levels were reduced by in‐furrow fungicide treatment, but were less effective when the initial levels of the fungus on the seed tubers and in the soil were high.  相似文献

Over 3 yr, the development of dry rot, caused by Fusarium solani var. coeruleum, and the efficacy of treating potato tubers with imazalil was examined in relation to the date of haulm destruction and harvest, and the interval between haulm destruction using diquat dibromide and harvest. The effect of these factors on skin set was also assessed. Planting inoculated seed tubers bearing small rots produced more dry rot on the daughter tubers than planting naturally contaminated seed tubers in 1992. The incidence of dry rot was higher on daughter tubers wounded by a standardised method than on those passed over a reciprocating riddle (riddling). The relationship between the two assessment methods was significantly (P<0.05) correlated in 2 out of 3 yr. Overall, the interval between haulm destruction and harvest had less effect on the incidence of dry rot on daughter tubers after riddling than the date of harvest. In 2 out of 3 yr, the incidence of dry rot on riddled tubers was least on those harvested in August and was much greater on September‐harvested tubers. On tubers harvested in October, the pattern was variable, with the incidence declining in 1 yr but increasing in the other. In the third year, the development of dry rot was similar on August‐ and September‐harvested tubers and was least on those harvested in October. Dipping tubers in imazalil gave significant reductions in dry rot although the amount of the reduction was variable and not affected by time of treatment or the amount of fungicide deposited within the range 6.7 to 19.4 mg imazalil kg?1. Skin set at harvest, as measured by skin strength or the amount of scuff damage, increased the later the tubers were harvested but did not appear to be affected by the interval between haulm destruction and harvest.  相似文献

Tecnazene (up to 33 ppm) and dichloran (up to 500 ppm) had little effect on germination of spores or growth of Fusarium solani isolated from and causing a rot of potato tubers; they also did not decrease rotting when applied to wounds later inoculated with the pathogen. Benomyl and thiabendazole (up to 500 ppm) also had little effect on spore germination but did greatly decrease growth at 5 ppm. A pronounced pink coloration developed in cultures growing slowly in the presence of benomyl; a similar though less striking effect appeared in agar cultures containing thiabendazole. Benomyl suspended in water or diluted with Fuller's earth gave good control of rotting when applied to wounds inoculated later with F. solani. Still better control was obtained with thiabendazole; dusts containing 1% a.i. substantially decreased rots and those containing 10 % a.i. gave almost complete control when applied to wounds shortly before inoculation. Thiabendazole was also very effective when used 24 h after inoculation and a fair measure of control was obtained when it was applied 24 h later. Benomyl and thiabendazole placed on apparently intact surfaces of tubers caused tissue 5 mm deep to become toxic to F. solani 10 days later, and, unexpectedly, this tissue prevented spore germination. Fuller's earth alone substantially decreased rotting. The results obtained suggest that dusts containing thiabendazole have some promise for the control of Fusarium rots of potato tubers, especially of early crops.  相似文献

Fusarium javanicum Kds. has been isolated from wilted vegetable marrows and melons and from the soil of a citrus grove in Israel. The morphology of the species is described, and measurements and drawings of macroconidia are presented.In vitro tests showed thatF. javanicum failed to develop at either 12 or 40° C, and developed better at 24° C than 18 or 30° C. Mycelial growth on potato dextrose agar was always better at pH 7 than at pH 4.2. Darkness favored such growth to a marked extent only at 18° C and pH 4.2. In glasshouse inoculation tests one isolate ofF. javanicum from vegetable marrow proved clearly pathogenic to cucumber, melon, watermelon and marrow. Measurements of macroconidia have been made, for comparative purposes, on isolates ofF. javanicum var.ensiforme and ofF. solani f.cucurbitae furnished from abroad, and on isolates ofF. solani from cucurbits in Israel. Comparing the data for 3-septate macroconidia with those given in literature forF. solani and forF. javanicum and its var.radicicola and var.ensiforme, it is concluded that two groups can be distinguished: One group comprises, with one exception, all isolates ofF. javanicum and its varieties as well asF. solani f.cucurbitae, and is characterized by slender macroconidia. The other group is represented byF. solani with its typically sausage shaped conidia. It is concluded thatF. javanicum should be maintained as a species distinct fromF. solani. It is further suggested that what has been calledF. solani f.cucurbitae may have closer affinity toF. javanicum than toF. solani.
Zusammenfassung Fusarium javanicum Koorders wurde in Israel von verwelkten Cucurbitaceen(Cucurbita pepo Kussa andCucumis melo) sowie von dem Boden einer Citruspflanzung isoliert. Die Morphologie des Pilzes wird hier beschreiben. Inin vitro Versuchen entwickelte sichF. javanicum auf Kartoffel-Dextrose Agar weder bei 12 noch bei 40° C, und wuchs besser bei 24 als bei 18 oder 30° C. Das Wachstum des Myzeliums war stets besser bei pH 7 als bei pH 4.2. Dunkelheit förderte dieses Wachstum erheblich nur bei 18° C und pH 4.2. In Impfungsversuchen im Glasshaus wirkte ein Isolat desF. javanicum vonC. pepo stark pathogenisch auf Gurken, Melonen, Wassermelonen undC. pepo. Zwecks taxonomischer Vergleiche wurden Makrokonidien aus dem Ausland erhaltener Isolate vonF. javanicum var.ensiforme undF. solani f.cucurbitae gemessen, sowie solche desF. solani von Cucurbitaceen in Israel. Die Resultate dieser Messungen wurden mit den in der Literatur angegebenen Werten fürF. solani, F. javanicum und dessen var.radicicola und var.ensiforme verglichen. Dieser Vergleich zeigte 2 klar getrennte Gruppen: Die erste enthält, mit einer Ausnahme, alle Isolate desF. javanicum und desF. solani f.cucurbitae, und besitzt typisch schlanke Makrokonidien. Die zweite Gruppe besteht ausF. solani mit seinen typisch dickeren, Würstchenartigen Sporen. Dies führt zu dem Schluß daßF. javanicum weiterhin als eine vonF. solani getrennte Spezies zu betrachten ist.

Three tetraploid somatic hybrid lines produced by protoplast fusion between a dihaploid potato, Solanum tuberosum, cultivar BF15 and the wild potato species Solanum berthaultii were evaluated here for their response to different soil‐borne pathogens, that is Fusarium solani, Pythium aphanidermatum and Rhizoctonia solani as well as to infection by potato virus Y (PVY). Both hybrid and BF15 plants grown in vitro were inoculated with the tested pathogen strains, that is R. solani, P. aphanidermatum, or F. solani. The growth level and disease severity index of these plants were compared to the susceptible commercial cultivar Spunta. A better growth of inoculated hybrid plants and restricted disease symptoms were observed in comparison with the commercial plants. Under glasshouse conditions and after inoculation with R. solani and P. aphanidermatum, improved resistance of the hybrid plants to these pathogens was confirmed. Indeed, these plants showed no significant damage following inoculation and a better development in R. solani‐infected plants. The susceptibility of the hybrid tubers to R. solani, P. aphanidermatum, and to F. solani infection was also determined. A significant reduction of tissue colonisation was observed in all the hybrid lines compared to the cultivated cultivars. The STBc and STBd hybrids also showed improved resistance to the PVY ordinary strain (PVYo) under glasshouse conditions.  相似文献

Potato seed tubers of six cultivars from commercial stocks and from stocks derived from stem cuttings (healthier seed) were fumigated with 2-aminobutane 2 wk after lifting or treated with benomyl or thiabendazole in January. 2-aminobutane prevented skin spot and gangrene developing on treated tubers. Experiments were planted at Rothamsted (clay with flints soil) and at Woburn (sandy loam soil) in 1973–75. Healthier seed produced more stems/plant than commercial stocks and yielded on average 8% more at Rothamsted in 1973 and 1974 and respectively 5 and 10% more at Woburn in 1973 and 1975. Seed treatments did not consistently affect stem numbers or increase yield although all treatments tended to decrease tuber size. Infection of stem bases and tubers by Polyscytalum pustulans and Rhizoctonia solani was usually less from healthier than from commercial seed and was decreased by benomyl and thiabendazole in 1973 and 1974. Infection by Helminthosporium solani of the skin around tuber eyes was greater from healthier than from commercial seed but was decreased by benomyl and thiabendazole. 2-aminobutane sometimes decreased infection of tubers by P. pustulans and R. solani but neither of stem bases nor of tubers by H. solani. Gangrene on tubers uniformly wounded at lifting was not consistently affected by seed source or seed treatment. Treating seed with benomyl or thiabendazole in 1975 decreased skin spot and black scurf in tubers stored until March 1976. These treatments also decreased silver scurf on the produce of commercial seed at Rothamsted but gangrene was not consistently affected by seed treatments.  相似文献

To exploit natural products for plant disease control, the essential oil of Syzygium aromaticum (L.) Merr. & Perr. (clove) was investigated for its antifungal activity against Rhizopus stolonifer and Fusarium solani, the postharvested yam tuber rot pathogens. The essential oil was obtained by hydrodistillation using a Clevenger‐type apparatus. The chemical composition of the oil was determined by gas chromatography and gas chromatography coupled with mass spectrometry. Antifungal activities of the oil were tested in vitro against mycelia growth and spores germination. In situ tests were conducted on healthy yam tubers, and necrosis symptoms were assessed. Results showed that eugenol (79.4%), eugenylacetate (9.2%) and isocaryophyllene (7.0%) were the major components. The oil exerted antifungal activities with total inhibition (TI) of the mycelia growth of R. stolonifer and F. solani was recorded at 200 and 300 ppm, respectively, while TI of spores germination was recorded at 31.25 and 250 ppm, respectively. For the standard fungicide (Ridomil®), TI value of mycelia growth was 1600 ppm for the both pathogens, while TI of spores germination were 200 ppm and 1600 ppm, respectively, for Rhizopus and Fusarium. In situ tests showed complete inhibition of yam tuber rot when the essential oil was applied at 2000 ppm for preventive tests. This oil also reduced significantly (P ≤ 0.05) necrosis development on yam tuber for curative test at the same concentration. Total inhibition of the necrosis by Ridomil (3000 ppm) was observed only for Rhizopus on preventive test. There were positive correlations between the oil concentration and the reduction of necrosis cause by R. stolonifer and F. solani. These findings showed that clove essential oil may serve as environmental friendly bio‐fungicide for the management of postharvest yam tuber rot.  相似文献

Ko & Hora's (1971) selective medium (FM) for counting Rhizoctonia solani propagules from soil has been modified. FM medium minus gallic acid and fenaminosulf i.e. mineral antibiotic (MA) medium, was amended with inhibitors and fungicides at different concentrations. The modified medium consisting of MA + gallic acid (400 μg ml-1) + fosetyl-Al (250μgml-1) was found most efficient in selective isolation of R. solani from soil even in the presence of Macrophomina phaseolina (Rhizoctonia bataticold) and other soil mycoflora. This amended medium was effective in isolating R. solani from soil even when the number of propagules of M. phaseolina present was 10 times greater. The sensitivity of this medium was five times better than Ko & Hora's medium.  相似文献

The development of dry rot caused by Fusarium solani f. sp. eumartii was evaluated in susceptible (Huinkul) and resistant (Spunta) potato cultivars. Fungal proteolytic and polygalacturanase activities were measured at different days postinoculation either with the pathogenic F. solani f. sp. eumartii, isolate 3122 or with the non‐pathogenic F. solani, isolate 1042. After inoculation with the pathogenic fungus, proteolytic and polygalaturonase activities were higher in the susceptible than in the resistant cultivar. In addition, we found a correlation between the levels of proteolytic activity detected in the intercellular washing fluids with the size of the lesion area caused by F. solani f. sp. eumartii in Huinkul tubers. The action of the proteolytic activity over cell wall proteins of both potato cultivars was assayed. An extracellular potato protein with homology to proteinase inhibitors of the Kunitz family was identified as a substrate of the proteolytic activity in the susceptible cultivar. A microscopic study revealed differences between the potato genotypes in the rate of response to infection by F. solani f. sp. eumartii. In addition, the cell wall alteration caused by F. solani f. sp. eumartii in cortical cells of susceptible tubers was evaluated. The data with respect to the correlation between the course of cyto‐ and biochemical events of the two host–pathogen interactions were discussed.  相似文献

Isolates of Fusarium were obtained from samples of barley grain at harvest in 1971, 1973 and 1974. The ability of these isolates to produce the myco-toxin zearalenone was examined, the grain also being tested for its presence in 1974. The most common isolate was F. culmorum, 60% of which in 1974 produced zearalenone. Other species isolated which produced this myco-toxin were F. monilijorme, F. graminearum, F. avenaceum, F. nivale and F. sambucinum var coeruleum, the last three species have not previously been recorded as producing zearalenone.  相似文献

The soil-borne fungus, Fusarium solani f. sp. phaseoli, attacks roots and hypocotyls of bean (Phaseolus vulgaris) plants causing a devastating disease called root and foot rot. In a study of the host-pathogen relationship it was found that young bean roots, with the radicle just emerging, were highly tolerant to the pathogen, whereas older bean seedlings, with a fully developed root system, were completely susceptible. Investigations by low-temperature scanning electron microscopy demonstrated that significantly fewer spores and hyphae were present on the root surface of young bean seedlings as compared to older ones. A similar pattern of attachment was found when bean roots were inoculated with spores of F. solani f. sp. pisi, a related pathogen causing disease on peas but not on beans. Light microscopic studies showed that F. solani f. sp. pisi did not penetrate the root but rapidly formed thick-walled resting spores on the root surface. F. solani f. sp. phaseoli on the other hand quickly penetrated the root and formed an extensive network of fungal hyphae. These results demonstrate that the ability of fungal propagules to adhere to and to penetrate host tissues are two distinct processes. Furthermore, the data indicate that young bean roots lack a surface component necessary for attachment of fungal spores which may help explain their tolerance to Fusarium root rot.  相似文献

Rhizoctonia solani AG-3 forms sclerotia on potato tubers, reducing quality and marketability. Potato discs with sclerotia were exposed to saturated soil, and following toothpick baiting, hyphae of R. solani parasitised with mycelia that either coiled around or penetrated cell walls were observed. Similar parasitic behaviour was observed in dual cultures. Clonostachys rosea was identified as the mycoparasite by morphology and ITS sequencing. When co-inoculated onto stems grown from disease-free seed potatoes, tubers of the C. rosea?+?R. solani inoculated plants had significantly lower black scurf (P?0.0001) and higher yield (P?=?0.0002) than R. solani inoculated controls.  相似文献

Criconemella xenoplax, Fusarium solani, and F. oxysporum caused necrosis of Nemaguard peach feeder roots in greenhouse tests. Root necrosis was more extensive in the presence of either fungus than wtih C. xenoplax alone. Shoot growth and plant height were less for plants inoculated with F. oxysporum or F. solani than for plants inoculated with the fungi plus C. xenoplax. Neither synergistic nor additive effects on root necrosis or plant growth occurred between C. xenoplax and the fungal pathogens.  相似文献
