A chemometric-assisted kinetic spectrophotometric method has been developed for simultaneous determination of ascorbic acid (AA), uric acid (UA), and dopamine (DA). This method relies on the difference in the kinetic rates of the reactions of analytes with a common oxidizing agent, tris(1,10-phenanthroline) and iron(III) complex (ferritin, [Fe(phen)3]3+) at pH 4.4. The changes in absorbance were monitored spectrophotometrically. The data obtained from the experiments were processed by chemometric methods of artificial neural network (ANN) and partial least squares (PLS). Acceptable techniques of prediction set, randomization t test, cross-validation, and Y randomization were applied for the selection of the best chemometric method. The results showed that feedforward artificial neural network (FFANN) is more efficient than the other chemometric methods. The parameters affecting the experimental conditions were optimized, and it was found that under optimal conditions Beer’s law is followed in the concentration ranges of 4.3–74.1, 4.3–78.3, and 2.0–33.0 μM for AA, UA, and DA, respectively. The proposed method was successfully applied to the determination of analytes in serum and urine samples.  相似文献

3-(N-Methylamino)glutaric acid has been identified as a new free amino acid in extracts from Prochloron didemnii (Lewin), a unique prokaryotic algal symbiont associated with certain didemnid ascidians. The aim of the present study was to determine the effects of the potential methane reducer fumaric acid on the fattening performance and acid–base balance of growing bulls fed two different silage types as roughage (maize and grass silage). A total of 62 fattening bulls (German Holstein breed, initial body weight: 266 ± 42 kg), randomly assigned to eight feeding groups, received four levels of fumaric acid (0, 100, 200 or 300 g/d) at each silage type. The daily feed and water intake and the live weight were measured over the whole testing period of 280 days. In blood samples, blood cells and blood gases as a parameter of acid–base status were analysed. Feed and faeces were collected to determine the apparent nutrient digestibility. Five animals from each group fed maize or grass silage, supplemented with 0 g or 300 g fumaric acid, were slaughtered at 580 kg body weight. After slaughter, rumen fluid pH was measured and dressing percentage was calculated. Neither the total feed intake (8.81 ± 0.07 kg/d) nor the daily weight gain (1277±24 g/d) was influenced significantly by treatments. Fumaric acid supplementation did not influence the erythrocyte count or the blood gas concentration. The silage type significantly influenced the apparent digestibility of the whole diet. The dressing percentage was slightly higher (p < 0.1) after fumaric acid supplementation. No signs of an incompatibility to fumaric acid on the animals were observed over the whole experimental period. However, it seems to be necessary to conduct more long-term studies with different silage types and addition of organic acids combined with direct measurements of methane.  相似文献

An experiment was performed to ascertain whether changing the dietary intake of two substances, cystine and margaric acid (heptadecanoic acid), that affect the flux through pathways involving the two vitamin B12-depednent enzymes, methionine synthase and methylmalonyl-CoA mutase, would affect the interaction between nickel and vitamin B12. Rats were assigned to treatment groups of six in a fully crossed, four-factorial arrangement. The independent variables, or factors, were: per kg of fresh diet, nickel analyzed at 25 and 850 μg; vitamin B12 supplements of 0 and 50 μg; margaric acid supplements of 0 and 5 g; andl-cystine supplements of 0 and 12 g. The diet without cystine was marginally deficient in sulfur amino acids. Nickel affected growth, liver wt/body wt ratio (LB/BW), and a number of variables associated with iron, calcium, zinc, copper, and magnesium metabolism. Most of the effects of nickel were modified by the vitamin B12 status of the rat. In numerous cases, the interaction between nickel and vitamin B12 was dependent on, or altered by, the cystine or margaric acid content of the diet. Thus, the findings showed that the extent and the direction of changes in numerous variables in response to nickel deprivation varied greatly with changes in diet composition. These variables include those previously reported to be affected by nickel deprivation, including growth and the distribution or functioning of iron, calcium, zinc, copper, and magnesium. The findings also support the hypothesis that nickel has a biological function in a metabolic pathway in which vitamin B12 is important.  相似文献

Littorina saxatilis and Littorina arcana collected from a boulder field low in the intertidal zone had a uric acid concentration significantly higher than snails collected from nearby cliff crevices that were at the upper limit of their vertical range on the shore. The absolute concentrations varied with different collections, suggesting a possible seasonal fluctuation in uric acid. L. arcana had a greater concentration of uric acid than did L. saxatilis when both were from the boulder field, but the two did not differ significantly when taken from crevices. Samples from the two sites were marked and transplanted; by four weeks the transplanted animals showed a tendency to develop a uric acid concentration similar to that of natives of the site, suggesting a physiological rather than a genetic adaptation. Xanthine dehydrogenase activity differed between animals from the two sites in the same manner as the uric acid concentration.  相似文献

Summary A method has been developed to determine the age of fossil bone samples based on amino acid racemization (AAR). Approximately one hundred fossil bone samples of known age from Hungary were collected and analysed for D- and L-amino acids. As the racemization of amino acids is affected by temperature, pH, metal content of the soil, and time passed since death, these factors were eliminated by comparing the estimated age to age determined by the radiocarbon method. Determining the D- and L-amino acid contents in samples of known age, determining the half life of racemization and plotting the D/L ratio as a function of time, calibration curves were obtained. These curves can be used for the age estimation of samples after determining their D- and L-amino acid content. The D/L ratio for 2 to 3 amino acids was determined for each sample and the mean value of estimated ages based on calibration curves was considered to estimate age of the fossil samples.  相似文献

In 1987, there was an episode of shellfish poisoning in Canada with human fatalities caused by the diatom Pseudo-nitzschia multiseries, which produced the toxin domoic acid. In order to examine whether domoic acid in this diatom serves as a grazing deterrent for copepods, we compared feeding rates, egg production rates, egg hatching success and mortality of the calanoid copepods Acartia tonsa and Temora longicornis feeding on unialgal diets of the toxic diatom P. multiseries and the similarly-sized non-toxic diatom Pseudo-nitzschia pungens. Copepods were collected in summers of 1994, 1995 and 1996 from Shediac Bay, New Brunswick, Canada, near Prince Edward Island, the site of the 1987 episode of domoic acid shellfish poisoning. Rates of ingestion of the toxic versus the non-toxic diatom by A. tonsa and T. longicornis were similar, with only one significantly different pair of values obtained in 1994, for which A. tonsa had a higher mean rate of ingestion of the toxic than the non-toxic diatom. Thus, domoic acid did not appear to retard grazing. Analyses of copepods with high performance liquid chromatography (HPLC) revealed that copepods accumulated domoic acid when feeding on P. multiseries. Egg production rates of copepods when feeding on P. multiseries and P. pungens were very low, ranging from 0 to 2.79 eggs female–1 d–1. There did not appear to be differential egg production or egg hatching success on diets of the toxic and non-toxic diatoms. Mortality of females on the toxic diet was low, ranging from 0 to 20%, with a mean of 13%, and there was no apparent difference between mortality of copepods feeding on toxic versus non-toxic diatoms. Egg hatching success on both diets, although based on few eggs, ranged between 22% and 76%, with a mean percentage hatching of 45%. Diets of the non-toxic diatom plus natural seawater assemblages supplemented with dissolved domoic acid, revealed similar rates and percentages when compared to previous experiments. In summary, none of the variables measured indicated adverse effects on copepods feeding on the toxic compared to the non-toxic diatom.  相似文献

We determined the fatty acid signatures in milk, serum, and blubber samples collected from young free‐ranging Steller sea lions (Eumetopias jubatus) and investigated the partitioning of fatty acids among these matrices. We assessed the relationship of fatty acids in each matrix with region, age, season, sex, and body condition to gain information needed to determine the most appropriate type of samples to collect and analyze to address future research questions. The variability of fatty acid composition was almost entirely explained by sample type, highlighting the importance of selecting a matrix consistent with the study objectives. Regional differences in fatty acid composition were found in all sample types and these differences varied among matrices. The proportion of fatty acids in milk and serum were influenced by season of capture whereas blubber samples were more affected by age of the pup. The influence of season on the fatty acid signatures in milk and serum highlights the use of these samples over blubber in studies investigating seasonal changes in diet. Further, our findings suggest that the use of milk samples collected from the stomachs of pups may be a viable alternative to directly collecting milk from adult females.  相似文献

Lecanoric acid (1), orsellinic acid methyl ester (2), orcinol (3), and usnic acid (4) were isolated from the lichen Parmelia subrudecta, collected on Palma of the Canary Islands, Spain. Compounds 1, 2, 3, and 4 were purified by solvent extraction, silica gel column chromatography, and preparative high-performance liquid chromatography (HPLC) consecutively. The structures of the four compounds were elucidated by one- and two-dimensional nuclear magnetic resonance (NMR) experiments and mass spectrometric investigations. These compounds showed activity against important gram-positive and gram-negative pathogens like mycobacteria and multiresistant staphylococci. This activity is combined with antiproliferative activity and cytotoxicity.  相似文献

本文对几种海南野生石斛(红头金石斛、重唇石斛、流苏金石斛、华石斛、密花石斛、刀叶石斛)及人工栽培的金钗石斛,进行了人体及植物必需微量元素和人体必需氨基酸的含量分析.结果表明:每种石斛中均含有植物和人体所需的7种微量元素和7种人体必需氨基酸,其中流苏金石斛的微量元素和氨基酸含量均高于其他种.人工栽培的金钗石斛其微量元素和氨基酸含量与海南野生石斛比较,差异不大,仅在Fe、Mn两个元素均低于野生种.本文还进行了植物必需微量元素与必需氨基酸相关性的讨论.  相似文献

Three species of microalgae able to produce eicosapentaenoic acid(EPA) were collected from brackish and sea water around Japan. The species were identified as Navicula saprophila, Rhodomonassalina and Nitzschia sp. EPA as a proportion of total fatty acids increased in the presence of acetic acid for Rhodomonas salina and Nitzschia sp. However, Navicula saprophila displayed the greatest productivity of EPA and the EPA content of its biomass was enhanced under mixotrophic conditions in the presence of acetic acid. AIMS: To investigate how carbon dioxide affects the acid resistance of Escherichia coli. METHODS AND RESULTS: Escherichia coli W3110 was grown in minimal EG medium at pH 7.5, and cells were adapted at pH 5.5 at 37 degrees C with and without supply of carbon dioxide and nitrogen gases. The number of colonies grown on LB medium was measured after cells were challenged in minimal EG medium of pH 2.5 at 37 degrees C under various conditions. When carbon dioxide was supplied at both the acid adaptation and challenge stages, 94% of cells survived after the acid challenge for 1 h, while the survival rates were 50 and 67% when nitrogen gas and glutamate were supplied respectively. After the acid challenge for 3 h, the survival rate observed with the carbon dioxide gas supply was again 2.5-fold higher than those with the nitrogen gas supply. CONCLUSION: Carbon dioxide was shown to participate in the maintenance of high viability under acidic conditions. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides useful information for research into bacterial pathogenesis, fermentation and food preservation.  相似文献

选择国内外7种不同产地蜂胶,采用氨基酸自动分析仪分别测定其氨基酸含量。结果表明,所测蜂胶均含有17种以上氨基酸,其构成模式比较接近,但氨基酸含量差异较大,北京蜂胶总氨基酸含量最低,为26.96mg·100g-1,杭州蜂胶总氨基酸含量最高,为165.74mg·100g-1,谷氨酸为各产地蜂胶中含量最高的氨基酸,其中杭州蜂胶谷氨酸含量居首,为30.74mg·100g-1,大部分蜂胶中约30%的氨基酸为必需氨基酸。不同产地蜂胶氨基酸含量差异,可能是影响其生物活性的一个重要因素。  相似文献

The effects of gibberellic acid and kinetin with ethylene plus carbon dioxide on the thermodormancy of lettuce seeds (Lactuca sativa L. cv. Mesa 659) at 35 C in the dark were studied. The combination of gibberellic acid plus kinetin with ethylene plus carbon dioxide was most effective in overcoming thermodormancy in these Great Lakes type seeds, alleviating any induced light requirement. Gibberellic acid action required at least a minimal level of ethylene plus carbon dioxide. Kinetin action was independent of ethylene plus carbon dioxide but interacted with the gases when the gases were added. A schematic representation of the interaction is presented.  相似文献

Phloem sap collected from Yucca and coconut inflorescence stalks was shown to contain abscisic acid (ABA) and trace amounts of 2-trans ABA. Aloe greatheadii var. davyana is the most important indigenous South African bee plant. Fresh, bee-collected and stored pollen of this aloe was collected and analysed for its nutritional content, including amino acid and fatty acid composition. Highly significant differences were found between the three types of pollen. Collection and storage by the bees resulted in increased water (13-21% wet weight) and carbohydrate content (35-61% dry weight), with a resultant decrease in crude protein (51-28% dry weight) and lipid content (10-8% dry weight). Essential amino acids were present in equal or higher amounts than the required minimum levels for honeybee development, with the exception of tryptophan. Fatty acids comprised a higher proportion of total lipid in fresh pollen than in bee-collected and stored pollen. This study is the first to compare the changes that occur in pollen of a single species after collection by honeybees.  相似文献

Phytogenous Phenolic and benzene compounds have been described as being responsible for many biological activities including antifungal effects. The effect of caffeine and cinnamic and caffeic acids on a model fungus, Aspergillus nidulans, was investigated at its initial stage of germination. Conidia did not germinate in the presence of cinnamic acid (1 mM). Caffeine and caffeic acid exerted a negative effect on germination, on the nuclear duplication cycle and on first septum formation. The effects of caffeine were dose-dependent; effects of caffeic acid (1 mM) were more intense than those of caffeine (10 mM).  相似文献

In the selection of acid-tolerantRhizobium meliloti, procedures for the collection and isolation of rhizobia, and the assessment of acid tolerance, have not been critically evaluated. Such procedures form the basis of this study. Root nodules were collected fromMedicago spp. found growing on acid soil in Sardinia. Their encumbent bacteria were isolated directly onto media adjusted over a range of pH values, and then assessed for acid tolerance in both the laboratory and field. Strains ofRhizobium meliloti isolated onto low pH media were, in general, more acid-tolerant than sister isolates from high pH media, when tested in both the laboratory and field. Dilution (10 or 100 fold) of the inocula used in the laboratory assessment did not greatly influence the rating derived, although there was some effect of bacterial colony type on growth rating. The link between polysaccharide production and acid tolerance was not strong. There was a poor correlation between the growth ratings derived from the laboratory screening and acid tolerance as expressed in the field.  相似文献
