The versatility of palladium(II) as a template for Mannich-type macrocyclization is illustrated. Reaction of (bis(3-aminopropyl)piperazine)palladium(II) with formaldehyde and nitroethane in basic aqueous solution yields the ‘reinforced’ macrocycle 7-methyl-7-nitro-1,5,9,13-tetraazabicyclo[11.2.2]heptadecane as its palladium(II) complex. The crystal structure shows the palladium ion lies in a slightly tetrahedrally distorted square plane of four nitrogen donors, with distances to the two tertiary donors [av. 2.059(3) Å] slightly shorter than those to the secondary amines [av. 2.066(3) Å]. The 3-methyl-3-nitro-1,5,9,13-tetraazacyclohexadecane as its palladium(II) complex was prepared by an analogous route. In a separate reaction based on the [Pd(en)(chxn)]2+ (en = ethane-1,2-diamine; chxn = cyclohexane-1,2-diamine) intermediate, an unsymmetrical macrocycle with a fused cyclohexane ring, 4,11-dimethyl-4,11-dinitro-2,6,9,13-tetraazabicyclo[12.4.0]octadecane was isolated as its palladium(II) complex. Accessibility to an isolable mixed-ligand precursor is a key to this reaction, provided by using palladium(II) as the templating metal. Reaction of (4,8-diazaundecane-1,11-diamine)palladium(II) with formaldehyde and diethyl malonate in basic aqueous solution yields, with ester hydrolysis and decarboxylation, the carboxylate-pendant macrocycle 1,5,9,13-tetraazacyclohexadecane-3-carboxylic acid as its palladium(II) complex. The crystal structure is comprised of hydrogen-bonded dimers {[Pd(L)][Pd(L-H)]}3+ where the pair of inversion related square-planar complexes share a single proton between their pendant carboxylates. Bis(3-aminopropyl)(piperazine)palladium(II) yields the macrocyclic complex ion (1,5,9,13-tetraazabicyclo[11.2.2]heptadecane-7-carboxylic acid)palladium(II), in a similar reaction.  相似文献

18,20-Epoxy-11 beta,17 alpha,20 beta,21-tetrahydroxypregn-4-en-3-one was synthesized by the application of hypoiodite reaction to the cortisol acetonide. The intermediary 18-iodo derivative was converted to the 11-oxo steroid by chromic acid prior to silver ion-assisted solvolysis. Removal of the protective group with hydrochloric acid was finally carried out to give the desired 11 beta,17 alpha,18,21-tetrahydroxypregn-4-ene-3,20-dione as the hemiacetal form. 18,20-Epoxy-11 beta-17 alpha,20 beta,21- tetrahydroxypregna-1,4-dien-3-one was also prepared from prednisolone through a similar reaction sequence.  相似文献

The macrocyclisation reaction of 3,3′-(3,6-dioxaoctane-1,8-diyldioxy)-bis(2-hydroxybenzaldehyde) (1) with S-methylisothiosemicarbazide hydroiodide (H2NNC(SCH3)NH2·HI) in the presence of potassium triflate, followed by addition of M(CH3COO)2·nH2O, where M=Ni, Cu, Zn, afforded [NiLKI3] (2), [NiLK(CF3SO3)] (3), [CuLK(CF3SO3)(CH3OH)] (4) and [(ZnILK)2CH3OH] (5), respectively. Compounds 2-5 have been characterised by X-ray crystallography. IR, electronic, mass, 1H, 13C{1H} and 19F{1H} NMR spectra are reported. Magnetic susceptibility measurements and ESR spectra of 4 indicate weak intermolecular spin-spin interactions, which are mostly dipolar in origin.  相似文献

We studied the behaviour of 8-azido-2'-deoxyadenosine and 8-bromo-2'-deoxyadenosine in aqueous solutions of ammonia and primary and secondary amines. Unexpectedly, 8-Azido-2'-deoxyadenosine is converted to 8-amino-2'-deoxyadenosine in excellent yields. The use of this reaction for the preparation of 8-aminoadenine derivatives needed for the preparation of oligonucleotides carrying 8-aminoadenine is discussed.  相似文献

The unique physicochemical properties of a thioamide bond, which is an ideal isostere of an amide bond, have not been fully exploited because of the tedious synthesis of thionated amino acid building blocks. Here, we report a purification‐free and highly efficient synthesis of thiobenzotriazolides of Fmoc‐protected and orthogonally protected 20 naturally occurring amino acids including asparagine, glutamine, and histidine. The near‐quantitative conversion to the respective thioamidated peptides on solid support demonstrates the robustness of the synthetic route. Furthermore, the unaltered incorporation efficiency of thiobenzotriazolides from their stock solution till 48 h suggests their compatibility toward automated peptide synthesis. Finally, utilizing an optimized cocktail of 2% DBU + 5% piperazine for fast Fmoc‐deprotection, we report the synthesis of a thioamidated Pin1 WW domain and thioamidated GB1 directly on solid support.  相似文献

Macrocycles are key structural elements in numerous bioactive small molecules and are attractive targets in the diversity-oriented synthesis of natural product-based libraries. However, efficient and systematic access to diverse collections of macrocycles has proven difficult using classical macrocyclization reactions. To address this problem, we have developed a concise, modular approach to the diversity-oriented synthesis of macrolactones and macrolactams involving oxidative cleavage of a bridging double bond in polycyclic enol ethers and enamines. These substrates are assembled in only four or five synthetic steps and undergo ring expansion to afford highly functionalized macrocycles bearing handles for further diversification. In contrast to macrocyclization reactions of corresponding seco acids, the ring expansion reactions are efficient and insensitive to ring size and stereochemistry, overcoming key limitations of conventional approaches to systematic macrocycle synthesis. Cheminformatic analysis indicates that these macrocycles access regions of chemical space that overlap with natural products, distinct from currently targeted synthetic drugs.  相似文献

Convenient high yielding methods for the preparation of 4,6-di-O-benzyl-myo-inositol, myo-inositol 1,3,5-orthoformate and myo-inositol 1,3,5-orthoacetate, without involving chromatography are described. Myo-inositol was converted to racemic 2,4-di-O-benzoyl-myo-inositol 1,3,5-orthoformate by successive treatment with triethyl orthoformate and benzoyl chloride. The dibenzoate obtained on benzylation with benzyl bromide and silver(I) oxide gave 2-O-benzoyl-4,6-di-O-benzyl-myo-inositol 1,3,5-orthoformate. Deprotection of the benzoate and the orthoformate with isobutylamine and aqueous trifluoroacetic acid, respectively gave 4,6-di-O-benzyl-myo-inositol in an overall yield of 67%. Myo-inositol orthoformate and orthoacetate were prepared and isolated as their tribenzoates. The free orthoesters were regenerated by deprotection of the benzoates by aminolysis with isobutylamine.  相似文献

A facile synthesis of oligodeoxynucleotides (ODN) containing 2'-deoxy-6-thioinosine (dI6S) based on the convertible nucleoside O6-phenyl-2'-deoxyinosine is presented. After standard solid-phase DNA synthesis and removal of the cyanoethyl protecting groups with DBU treatment with aqueous sodium hydrogen sulfide introduces the sulfur functionality, deprotects the other nucleobases and cleaves the ODN from the solid support in a one-pot reaction. In addition, the extinction coefficient of 2'-deoxy-6-thioinosine is determined by enzymatic fragmentation of the resulting ODN in the presence of adenosine deaminase.  相似文献

A lysyl-lysine bifunctional derivative of 9-aminoacridine has been synthesized and its DNA-binding capacity established by electron-paramagnetic-resonance study. For this purpose the binding parameters of a spin-labelled aminoacridine probe were estimated and the affinities of the lysylacridinyl-lysyldiamino-octane dimer and of 9-amino-acridine could be evaluated by competitive assays. The competition study provided quantitative results concerning the dissociation constant (KD) of the dimer. The obtained value was closely similar to the KD of 9-aminoacridine determined by the same method and to the KD previously reported for the anti-tumour and antibiotic bifunctional intercalator quinomycins.  相似文献

The human calcitonin peptide chain was assembled using Fmoc solid-phase peptide synthesis chemistry. The combinations of cleavage Reagent H with trans-[Pt(en)(2)Cl(2)](2+) and Reagents B, K, and R with trans-[Pt(CN)(4)Cl(2)](2-) provide convenient methods for the synthesis of human calcitonin and its methionine sulfoxide derivative; the formation of intramolecular disulfide bonds by the above Pt(IV) oxidants is essentially quantitative.  相似文献

A convenient synthesis of 2'-deoxy-2-fluoroadenosine from commercially available 2-fluoroadenine is described. The coupling reaction of silylated 2-fluoroadenine with phenyl 3,5-bis[O-(t-butyldimethylsilyl)]-2-deoxy-1-thio-D-erythro-pentofuranoside gave the corresponding 2-fluoro-2'-deoxyadenosine derivative (alpha/beta = 1:1) in good yield. The alpha- and beta-anomers were separated by chromatography, and then desilylated to give compounds 1a and 1b.  相似文献

Two synthetic strategies have been developed for the synthesis of spaced sugars with lipophilic 1,4-phenylene core. A building block combining the usefulness of Weinreb amide functionality and modified Julia olefination reaction has been explored towards this objective. This building block offers complete flexibility in attaching any desired sugar derivative across phenylene spacer via C-C bond formation. The other strategy uses carbohydrate based building blocks for the synthesis of symmetrical as well as unsymmetrical 1,4-phenylene spaced sugars. This is the first report for the synthesis of 1,4-phenylene spaced sugars via C-C bond formation.  相似文献
