Phosphoenolpyruvate carboxylase (PEPC) [EC 4.1.1.31 [EC] ] of plantsundergoes regulatory phosphorylation in response to light ornutritional conditions. However, the nature of protein kinase(s)for this phosphorylation has not yet been fully elucidated.We separated a Ca2+-requiring protein kinase from Ca2+-independentone, both of which can phosphorylate maize leaf PEPC and characterizedthe former kinase after partial purification. Several linesof evidence indicated that the kinase is one of the characteristicCa2+-dependent but calmodulin-independent protein kinase (CDPK).Although the Mr, of native CDPK was estimated to be about 100kDa by gel permeation chromatography, in situ phosphorylationassay of CDPK in a SDS-polyacrylamide gel revealed that thesubunit has an Mr of about 50 kDa suggesting dimer formationor association with other protein(s). Several kinetic parameterswere also obtained using PEPC as a substrate. Although the CDPKshowed an ability of regulatory phosphorylation (Ser-15 in maizePEPC), no significant desensitization to feedback inhibitor,malate, could be observed presumably due to low extent of phosphorylation.The kinase was not specific to PEPC but phosphorylated a varietyof synthetic peptides. A procedure is described for the purification of phosphoenolpyruvatecarboxylase (EC 4.1.1.31 [EC] ) and NADP-dependent malic enzyme (EC1.1.1.40 [EC] ) from sugar cane leaves. Each enzyme was purified tohomogeneity as judged by sodium dodecyl sulfate-polyacrylamidegel electro-phoresis, with about 30% yield. Phosphoenolpyruvatecarboxylase was purified 54-fold. A molecular weight of 400,000and a homotetrameric structure were determined for the nativeenzyme. The purified carboxylase had a specific activity of20.0 {diaeresis}mol (mg protein)–1 min–1, and wasactivated by glucose-6-phosphate and inhibited by L-malate.Km values at pH 8.0 for phosphoenolpyruvate and bicarbonatewere 0.25 and O.l0 mM, respectively. NADP-malic enzyme, 356-foldpurified, exhibited a specific activity of 71.2 {diaeresis}mol(mg protein)–1 min–1 and was characterized as ahomotetramer with native molecular weight of 250,000. Purifiedmalic enzyme showed an absolute specificity for NADP+ and requireda divalent metal ion for activity. Km values of 0.33 and 0.008mM for L-malate and NADP+, respectively, were determined. Thisenzyme was inhibited by several organic acids, including ketoand amino acids; while succinate and citrate increased the enzymeactivity when assayed with 10{diaeresis}M L-malate. Phosphoenolpyruvate carboxylase (PEPC) (EC 4.1.1.31 [EC] ) assayedin extracts of Panicum maximum Jacq. loses up to 50% of itsactivity after incubation for 60 minutes at 0C while the enzymefrom P. miliaceum L. is completely stable under these conditions.Following dilution at room temperature the enzyme from P. maximumis labile, while that from P. miliaceum is stable. The P. maximumenzyme can be largely stabilized against dilution and againstcold-inactivation by D2O which stabilizes hydrophobic bondsand the compatible solutes proline, betaine and trimethylamine-N-oxide.Mineral ions, previously demonstrated to be protective againstcold inactivation of pyruvate, Pi dikinase from maize, provideno protection of P. maximum PEPC against either cold or dilution.The chaotropic ion SCN- causes partial inactivation of the enzymefrom P. miliaceum in the cold. The possible interrelationshipbetween inactivation by dilution and inactivation by cold isdiscussed. At constant external [CO2], rates of dark-CO2 fixation of theunicellular green alga Eremosphaera viridis were drasticallyincreased (up to 40-fold) by addition of ammonium (NH3+ NH4+)at external pH values (pH0) between 6.0 and 8.0. The cytosolicpH was monitored under identical conditions by micro-pH-electrodemeasurements, and cytosolic and vacuolar pH by the 31P-NMR technique.Addition of ammonium (5.0 mol m pH0 7.0) caused a rapidand transient acidification of the cytosol during the first4 min. Thereafter, the cytosolic pH remained constant at itsoriginal value. A rather constant cytosolic pH was also confirmedby 31P-NMR measurements, which, in addition, indicated a slowalkalization of the vacuole (about 0.5 units within 30 min afteraddition of ammonium). Since the dramatic stimulation of dark-CO2 fixation by ammoniumis not mediated by an alkalization of the cytosol, nor by directammonium effects on phosphoenolpyruvate carboxylase (PEPC, E.C.4.1.1.31 [EC] ), the role of vacuolar alkalization as a possible triggerfor the stimulation of PEP-carboxylase is discussed. The pattern for primary products of CO2-fixation and the chloroplaststructure of Amaranthus retrqflexus L., a species which incorporatescarbon dioxide into C4 dicarboxylic acids as the primary productof photosynthesis, were compared in various chlorophyll containingtissues,i.e., foliage leaves, stems, cotyledons and pale-greencallus induced from stem pith. Despite some morphological differencesin these assimilatory tissues, malate and aspartate were identifiedas the major compounds labelled during a 10 sec fixation of14CO2 in all tissues. Whereas, aspartate was the major componentin C4-dicarboxylic acids formed in foliage leaves, malate predominatedas the primary product in stems, cotyledons and the pale-greencallus. Glutamate dehydrogenase (GDH) (EC 1.4.1.3 [EC] .) purified from greentobacco callus mitochondria was activated markedly by Ca2$ inthe amination reaction. This activation was detectable evenat concentrations below 5 µM Ca2$. Saturation curves for the three substrates of the aminationreaction showed normal Michaelis-Menten kinetics in the presenceof 1 mM of Ca2$, but pronounced substrate inhibition occurredwithout Ca2$. The effect of Ca2$ was chiefly on the maximalvelocity. The saturation curve for NH4Cl in the presence of Ca2$ was modulatedby a change in pH. The apparent Km value for NH4Cl markedlydecreased whereas that for -ketoglutarate increased slightlywhen the pH was raised from 7.3 to 9.0. In contrast, the Kmfor NADH was little affected by raising the pH. Eisenia bicyclis, Arame, was allowed to photosynthesize in seawatercontaining H14CO3, and 14C-mannitol was isolated fromits fronds. The ratio of 14C-total/14C1 + 14C6 in the 14C-mannitolwas found to be about 8.0 at 1 min-illumination, but graduallydecreased with time to 3.0, showing uniform radioactivity distribution.Mannitol therefore seems to be formed in brown algae throughthree carbon compounds. Enzymes which may be involved in the possible biosynthetic pathwayof mannitol, i.e. aldolase, hexose diphosphatase, mannitol-1-phosphataseand glucosephosphate isomerase were present in extracts fromseveral brown algae. Mesophyll chloroplasts were isolated from leaves of a Na+-requiringNAD-malic enzyme type, dicotyledonous C4 plant, Amaranthus tricolorL. Mitochondria isolated from leaves of Mesembryanthemum crystallinumoxidized malate by both NAD malic enzyme and NAD malate dehydrogenase.Rates of malate oxidation were higher in mitochondria from plantsgrown at 400 mil NaCl in the rooting medium and performing Crassulaceanacid metabolism (CAM) than in mitochondria from plants grownat 20 mM NaCl and exhibiting C3-photosynthetic CO2 fixation.The mitochondria isolated from plants both in the CAM and C3modes were tightly coupled and gave high respiratory control.At optimum pH for malate oxidation (pH 7.0), pyruvate was themajor product in mitochondria from CAM-M. crystallinum, whereasmitochondria from C3-M. crystallinum produced predominantlyoxaloacetate. Both the extracted NAD malic enzyme in the presenceof CoA and the oxidation of malate to pyruvate by the mitochondriafrom plants in the CAM mode had a pH optimum around 7.0 withactivity declining markedly above this pH. The activity of NAD-malicenzyme, expressed on a cytochrome c oxidase activity basis,was much higher in mitochondria from the CAM mode than the C3mode. The mechanism of light-dependent active transport of pyruvatein C4 mesophyll chloroplasts has not been clarified, particularlyin Na+-type C4 species, in which the pyruvate uptake into mesophyllchloroplasts is enhanced by illumination or by making a Na+gradient (Na+-jump) across the envelope in the dark. We re-investigatedhere the effect of Na+ on the active transport of pyruvate inmesophyll chloroplasts of Panicum miliaceum, a Na+-type C4 species,by comparing the rate of pyruvate uptake at various externalpHs under four conditions; in the light and dark together with/withoutNa+-jump: (1) At neutral pH, the rate of pyruvate uptake inthe dark was enhanced by Na+-jump but scarcely by illumination.(2) While the enhancement effect by Na+-jump was independentof external pH, that by illumination increased greatly at pHover 7.4, and the effects of light and Na+ at the alkaline pHwere synergistic. (3) The light-enhanced pyruvate uptake wasrelated to stromal alkalization induced by illumination. Infact, pyruvate uptake was induced by H+-jump in the medium frompH 8.0 to 6.7. (4) Stromal pH was lowered by the addition ofK+-pyruvate and more by Na+-pyruvate into the medium at pH 7.8in the light. (5) However, the pH and ATP levels in the stromawere not affected by Na+-jump. Carbonic anhydrase (CA, EC. 4.2.1.1 [EC] ) activity in air-grown Characorallina was detected mainly in the intracellular fraction,most of which composed of chloroplasts and cytoplasmic gel,and not on the cell surface. Only minor levels of CA activity,on the basis of equivalent volumes, were detected in the cellsap and the cytoplasmic sol. The maximum rate of photosynthetic O2 evolution by air-grownChara corallina at pH 6.0 was twice that at pH 7.6, while theapparent Km for external inorganic carbon (Ci) at pH 7.6 wasabout three times that at pH 6.0. However, the apparent Km(CO2)was about three times larger at pH 6.0 than at pH 7.6. The Km(Ci)-valueat pH 7.6 increased severalfold in the presence of acetazolamide(AZA), an inhibitor of CA, but no inhibition was observed atpH 6.0. The pH-dependence may be due to differences in the permeabilityof AZA at the given pH values. Fixation of 14CO2 at 20 µMand of H14CO3 at 200 µM over the course of 5 swas very similar at pH 7.4. Addition of CA significantly suppressedthe photosynthetic 14CO2-fixation but it stimulated the H14CO3-fixation.This result indicates that free CO2 is an active species ofCi that is incorporated into the cell during photosynthesis. Three isoforms (Types 1, 2 and 3) of phosphoenolpyruvate (PEP)carboxylase in young leaves of the Crassulacean acid metabolism(CAM) plant Kalanchoe daigremontiana were separated by DEAE-cellulosecolumn chromatography and preparative polyacrylamide-agarosegel electrophoresis, and their enzymatic properties were characterized. All three isoforms had similar molecular weights of about 234,000.At pH 8.0 Type 1 showed a high affinity to PEP, (Km=0.08 mM),whereas Type 3 showed a low affinity (Km=1.0mM). Km values forMgCl2 were 0.26 HIM in Types 1 and 3 and 0.5 nut in Type 2.All three types exhibited the same pH optimum at 8.0, but Type1 showed relatively low activity below pH 6.0, whereas Type3 showed high activity. Type 3 was more acid stable than theother forms. In the presence of glucose-6-phosphate, the Kmvalues of Types 1, 2 and 3 for PEP lowered to 0.027, 0.037 and0.044 mu at pH 8.0, respectively. Inhibition of activity byorganic acids such as malate and pyruvate was pronounced inType 3. Type 2 exhibited properties intermediate to Types 1and 3 with regard to pH curve, affinity to PEP and its effectof various metabolites. NADP-malic enzyme (EC 1.1.1.40 [EC] ), which is involved in Crassulaceanacid metabolism (CAM), was purified to electrophoretic homogeneityfrom the leaves of the inducible CAM plant Mesembryanthemumcrystallinum. The NADP-malic enzyme, which was purified 1,146-fold,has a specific activity of 68.8 µmol (mg protein)–1min–1. The molecular weight of the subunits of the enzymewas 64 kDa. The native molecular weight of the enzyme was determinedby gel-filtration to be 390 kDa, indicating that the purifiedNADP-malic enzyme is a hexamer of identical subunits. The optimalpH for activity of the enzyme was around 7.2. Double-reciprocalplots of the enzymatic activity as a function of the concentrationof L-malate yielded straight lines both at pH 7.2 and at pH7.8 and did not reveal any evidence for cooperativity of bindingof L-malate. The Km value for L-malate was 0.35 mM. Hill plotsof the activity as a function of the concentration of NADP+indicated positive cooperativity in the binding of NADP+ tothe enzyme with a Hill coefficient (nH) of 2.0. An S0.5 value(the concentration giving half-maximal activity) of 9.9 µMfor NADP+ was obtained. Oxaloacetate inhibited the activityof the NADP-malic enzyme. Both malate and aspartate were decarboxylated at the 4-carbonposition by isolated bundle sheath strands of C4 plants butto different extents depending upon the species. In Digitariasanguinalis, an NADP-malic enzyme (NADP-ME) species, 100 µMoxalic acid blocked malate decarboxylation through NADP-ME withoutaffecting aspartate decarboxylation which apparently occursthrough NAD-ME. In several phosphoenolpyruvate carboxykinase(PEP-CK) type C4 species, 200 µM 3-mercaptopicolinic acid(3-MPA), an inhibitor of PEP-CK, specifically inhibited themalate decarboxylation and partially inhibited aspartate decarboxylation.The aspartate decarboxylation insensitive to 3-MPA may occurthrough NAD-ME. Neither inhibitor prevented C4 acid decarboxylationin bundle sheath cells of NAD-ME species. Phosphoenolpyruvate carboxylase (PEPC) from several C3 plants was compared to maize PEPC by immunoblotting using an antibody against maize PEPC and by peptide mapping. In C3 gramineous plants, PEPCs of slightly different monomeric sizes were detected as two bands for wheat and barley leaves, as three bands for etiolated maize leaves and as four bands for rice leaves by SDS-polyacrylamide gel electrophoresis and immunoblotting, whereas only one PEPC band was detected for maize leaves, a C4 plant, or tobacco leaves, a dicotyledonous C3 plant. The peptide fragment patterns of the lower molecular weight PEPC (major band in immunoblotting) in wheat leaves was similar to that of maize PEPC in peptide mapping by protein staining or by immunological detection, but the upper one (minor band) had a different pattern from the lower one in peptide mapping by immunological detection and few peptide fragments from this were recognized by the anti-(maize) PEPC antibody. These results suggest that there are multiple forms of PEPC subunits in the gramineous plants tested, and the major PEPC has a primary structure similar to that of maize PEPC. To obtain information about the expression of PEPCs in C3 plants, changes in the amount of PEPC protein were investigated during the greening of rice and wheat seedlings. Judging from the regulation by light, there were two types of PEPCs in greening rice seedlings, one induced by light and the other reduced by it. Greening wheat seedlings also show a PEPC band induced by light. These findings indicate that some PEPCs in C3 gramineous plants not only have structures similar to that of maize PEPC, but also are regulated by light in a similar manner.  相似文献

The effects of Na application on growth and nitrate reductaseactivity of seven C4 plant species, Zea mays, Echinochloa crus-galli,Panicum miliaceum, Panicum coloratum, Panicum dichotomiflorum,Panicum maximum and Chloris gayana were studied. Growth of Pseudomonas stutzeri(VAN NIEL strain) in the presenceof a limiting amount of nitrate under anaerobic conditions ischaracterized by 2 logarithmic phases separated distinctly byan intermediate phase where the growth rate is very low. Inthe first logarithmic phase nitrate is reduced stoichiometricallyto nitrite stage, and in the second phase nitrite is reducedto nitrogen gas. The nitrite reducing activity of cells in the second growthphase is 3–4 times higher than that of cells in the firstphase. The effect of carbonic anhydrase (CA) on time courses of photosynthetic14C incorporation in the presence of 14CO2 or NaH14CO3 was studiedwith cells of Chlamydomonas reinhardtii which had been grownunder ordinary air (low-CO2 cells) or air enriched with 4% CO2(high-CO2 cells). Experimental data obtained at 20°C andpH 8.0 suggested that the major form of inorganic carbon utilizedby high-CO2 cells was CO2, while that utilized by low-CO2 cellswas HCO3. The cell suspension showed CA activity which was comparableto that observed in the sonicate of cells. Both activities werehigher in low-CO2 cells than in high-CO2 cells.
