An unknown substance(s) produced in the cotyledons of seedlings of the Japanese morning glory (Pharbitis nil) during a defined period of darkness triggers the subsequent initiation of floral buds at apical and axillary meristems. Recent studies have concentrated on characterizing molecular changes as a possible mechanism associated with its synthesis, but these have failed to eliminate interference due to lack of development unity in the sampled population and to consider different kinetic alternatives of those potential changes. The current study demonstrates that numerous age-related changes occur in polypeptides from cotyledons during growth under noninductive conditions, but that these are minimal in older seedlings selected for improved synchrony of the floral response. Polypeptides from older seedlings sampled at various times during and after a dark inductive period were examined by two-dimensional polyacrylamide gel electrophoresis (2-D PAGE). No differences associated with floral induction could be detected. The results indicate that changes in gene expression correlated with floral-induction do not occur in the abundant polypeptide fraction within the limitations of the techniques employed.  相似文献

Fossil seeds and seedlings of a Metasequoia-like taxodiaceous conifer occur in Paleocene deposits at the Munce's Hill and Gao Mine localities of central Alberta, Canada. Compression/impression specimens are preserved in upright growth positions among seedlings of the cercidiphyllaceous dicot Joffrea speirsii Crane & Stockey. There are a large number of seeds, a few of which were buried while germinating and show a radicle or short primary root. More than 500 Metasequoia-like seedlings have been identified that have two linear cotyledons with parallel margins and rounded tips. Three specimens have been found that display three cotyledons. Slightly older seedlings show decussate pairs of leaves attached to the stem distal to the cotyledons. Still older seedlings have axillary branches that show varying sizes and numbers of opposite leaves arranged in a single plane distal to the opposite pairs. These specimens reveal that both Joffrea and this extinct taxodiaceous conifer were early colonizers of North American floodplain communities at the beginning of the Tertiary.  相似文献

The effect of age of radish seedlings on changes in chlorophyll concentration caused by ethylene was examined. Ethylene was produced at 2–4 nl g?1 h?1 following excision of cotyledons from 5-to 20-day-old seedlings. The youngest cotyledons maintained this rate, whereas ethylene synthesis declined by as much as 80% during a 24-h period in older cotyledons. The youngest cotyledons continued to accumulate chlorophyll in the dark, but after 7 days cotyledons lost chlorophyll and the proportion of chlorophyll lost increased with age. Ethylene promoted, and norbornadiene inhibited, this loss of chlorophyll; in combined treatments the effects of ethylene and norbornadiene were competitive. The maximal rate of chlorophyll loss occurred in 1μl L?1 ethylene; extrapolation of the response to concentration indicated that half-maximum loss would occur at 0.005–0.01 μl L?1 ethylene. In cotyledons from 20-day-old seedlings, chlorophyll degradation occurred mainly after 24 h from excision and transfer to the dark. Chlorophyll degradation during 48 h in the dark was affected by norbornadiene or ethylene applied from 0–24 h or from 24–48 h.  相似文献

Gibberellin A3 (GA3) stimulated flowering when it was appliedto the shoot apex of seedlings of Pharbitis nil, dwarf strainKidachi; but, not when it was applied to the cotyledons. GA3applied to the plumule before or shortly after the start ofan inductive dark period promoted both flowering and shoot elongation;but, the later the time of application during the dark periodless the promotion of flowering, although marked promotion ofshoot elongation always took place. The variation with time in the response of flowering to GA3indicates that early floral processes at the apex are stimulatedby GA3, but that subsequent processes are insensitive to it.The early processes of floral stimulus produced by a 16 hr inductivedark period probably are completed within 20 hr at 28°Cafter the end of the dark period. At low temperatures, suchas 15 and 20°C, early floral processes continued for morethan 40 hr. When cotyledons were removed at various times, the export ofthe floral stimulus to the shoot apex was apparent within hoursof the generation of the floral stimulus in the cotyledons,which started with the passage of the critical 9-hr dark period. Seedlings of Pharbitis nil, Strains Violet, Tendan and Kidachi,initiated floral buds under Continuous light when exposed totemperatures lower than 15, 15 and 21?C, respectively, throughoutthe experimental period, or to 13–14?C for a minimum durationof 10, 8 and 4 days, respectively. Cotyledons were necessaryfor floral initiation when the seedlings at the start of coldtreatment were 8 days old (10 days old for Kidachi) or younger,although neither cotyledons nor foliage leaves were necessarywhen the plants were older. When the cotyledons in young seedlingswere removed immediately after exposure to cold temperature(13–14?C) for 14 (Violet), 12 (Tendan) or 8 (Kidachi)days (cold treatment begun when the cotyledons had just unfolded),only a few plants initiated floral buds under continuous light.However, when the cotyledons were left attached for 2 more daysat 23?C, the plants produced as many flower buds as those withintact cotyledons, suggesting that cotyledons exposed to coldtemperature produce a floral stimulus which can be translocatedto buds even after the end of the cold treatment. Pseudomonas solanacearum was transmitted from contaminated seed to the cotyledons of capsicum (Capsicum frutescens) at 92, 73 and 60% r. h. and to the cotyledons of tomato (Lycopersicon esculentum) seedlings at 92% r. h. Subsequent epiphytic colonisation of the true leaves of capsicum occurred at 92 and 73% r. h. An increase in the population on capsicum cotyledons was detected at 92 and 73% r. h. but only at the higher r. h. was an increased population of P. solanacearum associated with the colonisation of the true leaves. Lesions developed on the true leaves of capsicum at 92% r. h. Transmission of P. solanacearum from capsicum seeds was affected by inoculum concentration, occurring at an infestation level of ∽ 103 propagules seed-1 but not at 50 propagules seed-1. Pseudomonas solanacearum was detected on the cotyledons of capsicum held at 98% r. h. after germination of seed in soil infested with 2 × 108 propagules g-1 soil; lesions were detected on cotyledons 11 days after planting and invasion occurred in 10 stems and one root of the 20 seedlings sampled. The movement of the pathogen from the soil to the seedlings was affected by the level of soil infestation. Pseudomonas solanacearum was detected in four pairs of cotyledons, two stems and one root of the 36 seedlings sampled from soil infested with 107 propagules g-1 soil but it was not isolated from seedlings sampled from soil infested with 105 or 4 × 103 propagules g-1 soil. Leaf and stem prints demonstrated the epiphytic nature of this organism on the cotyledons and stems of seedlings.  相似文献

Summary Cytological and histochemical characterization of the structures from which an obscure substance is secreted via open stomata to the abaxial surface of Japanese morning glory (Pharbitis nil Choisy cv. Violet) cotyledons has been carried out. Observation of intact cotyledons using the light microscope revealed randomly distributed semi-transparent structures. These structures, which were shown to be the same as those previously described as giant oil cells are referred to here as cotyledonary bodies. These bodies can be easily isolated and purified after enzymatic digestion of the cotyledons. Using different staining procedures we have confirmed that each cotyledonary body originates from an individual mesophyJl cell during embryo development. Purified bodies consist of (i) a thick shell-like envelope; (ii) a transparent, hydrophilic zone; (iii) a hydrophobic core. Hydrophobic contents of the bodies were readily extracted with methanol and shown to contain fatty acids and phenolic compounds using the gas chromatography/mass spectrometry (GC/MS) technique. Methanolic extracts of cotyledonary bodies showed high fluorescence with two excitation and emission maxima. Using a fluorescence microscope we have shown that the bodies isolated from seedlings grown in continuous light, conditions non-inductive for flowering, and those grown under conditions inductive for flowering (a single 16 h, long dark period) have different fluorescence emission spectra. Different levels of free Ca2+ inside cotyledonary bodies isolated from light-grown and single dark-period treatedP. nil seedlings were found using the fluorescent calcium indicator dye Fluo-3 under a confocal scanning laser microscope. On the basis of these observations we speculate that cotyledonary bodies could be involved in floral induction.  相似文献

Cotyledons of gourd (Cucurbita maxima Duchesne) and bean (Phaseolus vulgaris L.) were used to study the changes in the activities of catalase, peroxidase, acid inorganic pyrophosphatase and alkaline inorganic pyrophosphatase during ageing and the diversion in such changes that occur when cotyledon senescence was retarded by detopping the seedlings above the cotyledons. Catalase, acid inorganic pyrophosphatase and alkaline inorganic pyrophosphatase activities declined during the senescence of the cotyledons. When cotyledon senescence was retarded by detopping as marked by the increase in the levels of chlorophyll and protein, there was also an increase in the activities of these enzymes. Peroxidase activity, on the other hand, increased during the senescence of the cotyledons and detopping the seedlings resulted in a further increase in the peroxidase activity. It can be suggested that some root factor(s) probably cytokinin(s) is (are) mobilised into the cotyledons of the detopped seedlings which otherwise would have been mobilised into the shoot apices, and help retard or even reverse the senescence of the cotyledons.  相似文献

Summary Isolated zygotic embryos of Pinus radiata D. Don germinated and their cotyledons, hypocotyl and root grew and developed further on the optimized culture medium, named LPSH2 based on the accompanying paper. The resulting plantlets appeared normal, but were one-third the size of the natural seedlings grown on water-agar medium only. When the isolated embryos were cultured on water-agar medium, they grew little and more importantly root development did not occur. Studies on biochemical changes during germination and early seedling growth showed that the patterns of changes in soluble sugar and starch content were generally different between isolated embryos and seedlings. Early on during culture the cotyledons and hypocotyls of natural seedlings had higher levels of soluble sugars and starch than the counterparts of the isolated embryos grown on the LPSH2-medium. Conversely, the root of the isolated embryos contained more soluble sugars and starch than that of the seedlings throughout the 3 wk of culture. However, little difference was found between the isolated embryos and seedlings as far as total protein concentrations and their sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) protein profiles were concerned.  相似文献

Floral induction in seedlings of Pharbitis nil Choisy cv. Violet, with one cotyledon removed, was manipulated by applying various photoperiodic treatments to the remaining cotyledon. Populations of polyadenylated RNA from treated cotyledons were examined to identify messages specifically involved in floral induction. The RNA was translated in vitro using a wheat-germ system, and the resulting translation products were analysed by two-dimensional polyacrylamide gel electrophoresis. Substantial qualitative and quantitative differences were found between mRNA from cotyledons of seedlings kept in continuous light (non-induced) and of seedlings given a 16-h dark period (induced). This investigation was conducted to observe changes in the compositions of fatty acids, glycolipids (GL) and phospholipids (PL) in cotyledons of soybean seeds which were germinated either in the dark or the light at 28°C for 8 days. The patterns of changes in lipid composition depended on the germinating conditions tested. In general, non-polar lipids were metabolized at a faster rate than polar lipids. Changes in lipid contents in cotyledons were also observed more clearly with the polar lipids than with the non-polar ones, especially in the light-grown seedlings. The major component of lipid, GL in chloroplasts, appeared rapidly at an earlier stage in the cotyledons of light-grown seedlings. During germination of soybean seeds, acyl sterylglucoside in cotyledons decreased rapidly, but monogalactosyl diglyceride and digalactosyl diglyceride (DGD) increased in the light-grown seedlings, whereas sterylglucoside and DGD increased in the dark-grown seedlings.

The major PL present immediately after immersion were phosphatidyl ethanolamine (PE), phosphatidyl choline (PC) and phosphatidyl inositol (PI). During germination under both conditions, light and dark, PE in cotyledons decreased with PC or PI, while phosphatidic acid increased rapidly, and phosphatidyl glycerol and diphosphatidyl glycerol also increased slightly. These changes in glycolipid and phospholipid compositions during germination seem to occur from the formation of photosynthetic tissues and the metabolic interconversion of phospholipids.  相似文献

Severe chlorosis and ultrastructural modifications of chloroplasts occur in sunflower in response to infection by Pseudomonas syringae pv. tagetis. Chlorosis became apparent within 2 days after the cotyledons of 10-day-old sunflower seedlings were inoculated with the bacteria. The first symptoms generally appeared in the center of leaves at the second node above the cotyledons. Leaves above the second node lost essentially all of their pigmentation but remained turgid and continued to expand. Grana thylakoids became dilated and separated from the granal stacks. These thylakoid membranes did not chemically breakdown as in the case in chromoplast formation or normal chloroplast senescence. Both grana and stroma thylakoid membranes coalesced to form a large membrane sheet within the plastid. The ultrastructural changes are unlike those reported to be caused by other chlorosis-inducing bacteria or chlorosis associated with normal senescence.  相似文献

'Alaska’ peas (Pisum sativum L.) grown under a 16-hr photoperiod at 20 ± 1 C and an 8-hr dark period at 16 ± 1 C in their ontogeny exhibit two periods of sensitivity to applied gibberellin (GA), namely, prior to and subsequent to but not during the linear phase of stem elongation. This paper describes experiments conducted primarily with seedlings. Growth-saturating doses of GA, applied to dry seeds before planting (10−3 m) and to the shoot tips of 3-day-old seedlings (10 μg), evoked growth rates equal to the growth rate of etiolated seedlings. Sensitivity of seedlings to applied GA decreased with age through the first 2 to 3 weeks of development; by the time seedlings were about 14 days of age and had four elongating internodes they no longer responded to applied GA. As endogenous growth rate diminished late in ontogeny, the plants again became sensitive to applied GA. Growth response was used as a criterion for determining apparent translocation of applied GA. ‘Alaska’ pea seedlings appeared to transport GA, both acropetally from the cotyledons and basipetally from the shoot tip, to all internodes with remaining extension potential. Excision of both cotyledons at any time during the first 9 days of development caused a significant reduction of growth rate, and applied GA did not restore normal growth rate. No evidence was found that the cotyledons supply endogenous GA to the shoot axis in normal seedling development. It is suggested that the normal growth rate of light-grown ‘Alaska’ peas is correlated with the rate of synthesis of GA and that GA is rate-limiting for stem elongation during early seedling development and during the period of decreasing growth rate and onset of apex senescence.  相似文献

Rooting is shown to occur in excised cotyledons of Sinapis alba when grown in petri dishes on moist filter paper. Cotyledons were excised at intervals from 6 hours after the start of imbibition, when they were yellow, unexpanded and enclosed within the testa, to 27 days after sowing when the cotyledons were green and fully expanded and on plants possessing up to 3 foliage leaves. Rooting generally began 5 or 6 days after excision and was completed dining the following 5 days. The age of cotyledons at t ho time of excision had three effects on rooting: the lag period be-fore rooting began and the period during which rooting took place both increased with age: but the most marked effect was on the total number of cotyledons which were able to form roots, which increased until cotyledon expansion was almost complete, then decreased as the mature cotyledons became older. Optimal rooting was shown by cotyledons detached 8 days alter sowing, when they were half expanded. At this age S5 % of them formed roots between 6 days and 8 days after excision.  相似文献

To obtain information on the importance of a functional mevalonate synthesis for plant growth and development, we investigated the effect of mevinolin, a highly specific inhibitor of 3-hydroxy-3-methylglutaryl (HMG) coenzyme A reductase (the mevalonate-producing enzyme) on growth, sterol accumulation and pigment formation of radish seedlings (Raphanus sativus L. cv. Saxa Treib) and in part also wheat seedlings (Triticum aestivum L. cv. Kolibri). Mevinolin applied during germination inhibits root elongation and development of lateral roots in etiolated and light-grown radish seedlings. This effect cannot be overcome by exogenous GA3, but by addition of mevalonic acid, the product of the internally inhibited reaction. This emphazises the specifity of the mevinolin effect and indicates that the biosynthesis of mevalonic acid is a mandatory requirement for root growth. In light-grown radish seedlings mevinolin also affects hypocotyl length-growth and inhibits sterol accumulation, but has little effect on the chlorophyll and carotenoid accumulation in the chloroplasts of the cotyledons. This indicates the possible presence of an independent mevalonate synthesizing pathway within the plastids and suggests a low transport rate of mevinolin from the radish roots to the cotyledons. When mevinolin is directly applied to the leaves at higher concentrations, it also reduces the light-induced chlorophyll and carotenoid accumulation as has been shown with etiolated primary leaves of wheat. This inhibition is age-dependent and proceeds to a higher extent in older than in younger etiolated leaf tissue. From our results we conclude that plastids possess an independent HMG-CoA reductase. In the cotyledons of radish, mevinolin seems to induce a senescence retardation and sun-type growth response, as has been evaluated by measuring the fast and slow chlorophyll fluorescence induction kinetics (Kautsky effect). These responses may be due to inhibitor-induced changes in the intracellular phytohormone balance.  相似文献

Excision of both cotyledons of Lupinus albus L. (L. termis Forssk.) seedlings at different morphological stages during the first 8 days after sowing, retarded significantly the growth of 16-day-old seedlings and decreased the content of IAA in the shoots. The effect was more obvious at the early excision (4 days after sowing). A pattern similar to that of control plants with intact cotyledons was attained when cotyledons were excised later than 8 days after sowing. It is believed that the cotyledons control the IAA content of the shoot during the early stages of development. At day 36 flower primordia were observed in controls but not in cotyledon-less plants. At this time the IAA content of the shoot apices and the growth of cotyledon-less plants were the same as in controls except for plants that lost cotyledons 4 days after sowing, which showed similar values at week 11, but did not produce flower primordia. The other cotyledon-less plants produced flower primordia 4–11 days after controls. It is postulated that the cotyledons convey certain factor(s) to the developing axis at an early stage of the development which promote flowering.  相似文献

When a brassinosteroid biosynthesis inhibitor, brassinazole (Brz), was applied at concentrations ranging from 0.1 to 2 μM, Arabidopsis thaliana (L.) Heynh seedlings grown in the dark exhibited morphological features of light-grown plants, i.e. short hypocotyls, expanded cotyledons, and true leaves, in a dose-dependent manner. Control (non Brz-treated) seedlings grown in the dark for 40 d did not develop leaf primordia. However, treatment with the lowest concentration of Brz induced the development of leaf buds, although it hardly induced any short hypocotyls, and treatment with the highest concentration of Brz induced both short hypocotyls and leaves. Labeling experiments with the thymidine analogue 5-bromo-2′-deoxyuridine revealed that amplification of cell nuclei and organellar nucleoids is activated in the shoot apical meristems of dark-grown Brz-treated seedlings. These results suggest that Brz-treatment induces development of true leaves. Furthermore, condensation and scattering of plastid nucleoids, which is known to occur during the differentiation of etioplasts into chloroplasts, was observed in the plastids of dark-grown Brz-treated cotyledons. In addition, high levels of ribulose-1,5-bisphosphate carboxylase-oxygenase proteins accumulated in the plastids of the cotyledons. Electron microscopy showed that the plastids were etioplasts with a prolamellar body and few thylakoid membranes. These results suggest that Brz treatment in the dark induces the initial steps of plastid differentiation, which occur prior to the development of thylakoid membranes. This is a novel presumed function of brassinosteroids. These cytological changes seen in Brz-treated Arabidopsis were exactly the same as those seen in a brassinosteroid-biosynthesis-deficient mutant, det2, supporting the hypothesis that Brz has no side-effects except inhibiting brassinosteroid biosynthesis, and should prove a useful tool in clarifying the role of brassinosteroids. Earlier experiments demonstrated that DNA from young plants of 5-azacytidine-induced flax (Linum usitatissimum) lines that flower earlier-than-normal is hypomethylated relative to DNA from their control lines and detected differences in methylation level between plants sampled at different ages, which suggested that the methylation level in flax changes during development. To investigate this possibility, and its potential impact on the difference in methylation level between early-flowering and control lines, developmental profiles were established for the cytosine methylation levels in DNA from post-germination seedlings and from the shoot tips of main stems and the cotyledons sampled throughout vegetative phase. The methylation profiles for two early-flowering lines and their control lines were compared. The methylation profiles were then compared to profiles for DNA content, tissue weight and chlorophyll content (green tissues); these additional parameters provided information on tissue status in terms of cell division, tissue expansion and/or photosynthetic maturity. With one exception, methylation levels were either static or increased with plant age and/or tissue maturity; the highest methylation levels were seen in senescent cotyledons. Although DNA from immature plants or tissues of the early-flowering lines was usually hypomethylated, the hypomethylation was not always apparent in tissues from older plants.  相似文献

Xanthomonas compestris pv.cucurbitae strain 62 was isolated from diseased cucumber seedlings grown in an open field in Egypt and identified in this study. Inoculation of cucumber seedlings with this strain resulted in reduction of relative water content (RWC) and increase in free proline content of infected cotyledons. RWC was found to be decreased by the increase of disease severity. The reverse was true for free proline content which was found to be increased by the increase of disease severity. These results indicate that water stress increased in direct proportion to the disease severity. Highly significant increase in free proline content was found to occur in organs of cucumber seedlings inoculated with strain 62 and showing disease symptoms on cotyledons only. This implied that water stress was not limited to symptomatic cotyledons but extended to the whole seedling.  相似文献
