为了探讨解偶联蛋白(UCP)基因-3826多态性与UCPmRNA表达水平之间可能存在的联系,应用RT-PCR方法测定了UCP基因-3826多态性野生型(AA),杂合子(AG)和突变纯合子(GG)3组人群脂肪组织中UCPmRNA的表达水平.定量结果指出:3种基因型(AA、AG和GG)携带者腹膜内脂肪的UCPmRNA表达水平存在极显著差异(P<0.01),并显示突变等位基因(G)的数量与UCPmRNA表达水平呈负相关.此结果表明UCP基因A→G(-3826)变异与UCPmRNA表达水平降低密切相关.但该变异导致UCPmRNA表达水平降低的机制还有待进一步研究  相似文献

本文报道对中国新疆东部一起戊型肝炎流行高峰期间,由HE患者烘便中分离到的型肝炎病毒中国XT-179株进行全基因cDNA克隆、核苷酸和氨基酸序列测定及分析结果。所阐明的HEV-XT-179株基因组全序列由7194个核苷酸和3'端的多聚腺嘌呤核苷酸尾组成。四种核苷酸的含量腺嘌呤(A)为17.0%,胞嘧啶(C)为31.9%,鸟嘌呤(G)为26.0,尿嘧啶(U)为25.1%。G+C含量为57.9%。全基因  相似文献

通过PCR定点突变的技术,将蛇毒蛋白Echistatin基因的C端进行了突变(Ala48→Arg48→,Thr49→Val49),模拟纤维蛋白N端的四肽(Gly-Pro-Arg-Val),以期增加Ecs(Echistatin)的活性。突变的基因重组到表达质粒pJC264上,经IPTG诱导,以CheY-Ecs融合蛋白方式进行了表达,表达量占菌体总蛋白的15~20%。SephadexG-75初步纯化该融合蛋白,然后用CNBr裂解,透析,冻干,反相HPLC纯化C端突变体Ecs蛇毒蛋白,N端十个氨基酸分析与天然的相符,在PRP(platelet-richplasma)测活体系中,10μmol/L的ADP诱导,C端突变体Ecs抑制血小板凝聚的活性约为野生型4倍。得到了Ecs的C端突变后使Ecs抑制血小板凝聚的活性提高的结果。  相似文献

将尿激酶原(pro-UK)cDNA和组织型纤溶酶原激活剂(t-PA)A链cDNA克隆到M13mp18中,经过二次寡核甘酸诱导的大片段定点删除和一次寡核苷酸诱导的多位点突变,得到u-PA(Leu144-Gly408)/t-PA(Ser1-Thr263)(ut-PA)融合基因.将ut-PA融合基因克隆到表达载体pCM-βneo中,与pCM-dhfr共转染CHO/DHFR-细胞,筛选稳定表达株.收集无血清表达上清,经苯甲脒柱纯化得到ut-PA纯品,SDS-PAGE和纤维蛋白自显影显示ut-PA有两种分子量形式,分子量分别为68kD和61kD.纤维蛋白亲和性试验表明,LUK(低分子量尿激酶)对纤维蛋白没有亲和性,而含有LUK的ut-PA则对纤维蛋白表现出很强的亲和性,但ut-PA的亲和性略低于亲本t-PA.  相似文献

人胃癌细胞系中p53抑癌基因变异的检测及其序列分析   总被引:3,自引:0,他引:3  
p53抑癌基因由于点突变,缺失或易位等方式而丧失活性是多种肿瘤发生发展的重要机理之一。本文应用聚合酶链式反应─—单链构象多态性分析(PCR-SSCP)方法,对四种人胃癌细胞系MGC803,BGC823,GC7901和PAMC-82中p53基因的第5、6、7、8四个外显子进行检测,结果发现,PAMC-82的第5和第8外显子,GC7901的第6外显子存在突变。PCR-直接测序证明,它们分别在174位、280位、204位密码子发生缺失G,GC→CG,AT→CG转变,因而可使其编码的p53蛋白分别发生移码突变,Arg→Thr,Glu→Ala,而丧失肿瘤抑制功能。实验结果表明,p53基因在胃癌发生发展过程中,尤其是较晚阶段具有重要作用。  相似文献

RRM RNA 结合蛋白在基因的转录后调控中起着重要作用.人GRY-RBP 是我们实验室最近克隆的一个新的全长cDNA,编码一个RRM RNA 结合蛋白.GRY-RBP 的全编码区用PCR扩增后,克隆到pET30a+ 表达载体中,在E.coli中获得了高效表达,表达的GRY-RBP重组蛋白占细菌总蛋白的21% .利用融合在GRY-RBPN 端的His.Tag,采取亲和层析的方法纯化了表达的重组蛋白.为了检测GRY-RBP的RNA 结合活性及其所结合的RNA 性质,将表达的蛋白与poly(A)-Sepharose 4B结合,发现GRY-RBP能与polyA (A)结合,结合活性能被可溶性的poly(A)、poly(C)减弱.改变NaCl浓度和加入不同浓度的酵母tRNA竞争物后,poly(A)结合活性不变.  相似文献

用四氮唑蓝光化学还原法对所合成的KCu(IDA)(Ser)·2H2O、KCu(IDA)(Ala)·H2O、Cu(IDA)(en)、KCu(IDA)(Gly)·H2O和Cu(IDA)·2H2O(IDA=N(羧基甲基)-甘氨酸,Ser=丝氨酸,Ala=丙氨酸,en=乙二胺,Gly=甘氨酸)等5种氨基酸─铜(Ⅱ)配合物进行了活性测定,发现它们均具有天然超氧化物歧化酶活性,其活性依次为0.34、0.45、0.50、0.54、0.72Cuμmol·L-1。  相似文献

Pseudomonas aeruginosa strain K (PAK) bears polar pili that promote infection by at least six bacteriophages. Moreover, a recently isolated mutant of strain K (PAK/2PfS) is many times more piliated than the wild-type strain and facilitates the preparation of large amounts of pure pili for biochemical studies. The present investigation was carried out to establish the structural relatedness of PAK and PAK/2PfS pili and to determine their biochemical composition. A purfication procedure is described for PAK and PAK/2PfS pili that yields about 8 mg of pure pili per 100 g (wet weight) of PAK/2PfS cells and 0.8 mg of pure pili per 100 g (wet weight) of PAK cells. PAK and PAK/2PfS pili were found to be free from phosphate, carbohydrate, and lipid and to contain a single polypeptide subunit of 17,800 daltons. Isopycnic centrifugation studies revealed that PAK and PAK(2PfS pili have the same buoyant density in sucrose (1.221) and CsC1 (1.295). Both types of pili banded at pH 3.9 when subjected to isoelectric focusing. Amino acid analyses showed that PAK and PAK/2PfS pili have identical amino acid compositions, whereas microimmunodiffusion studies revealed that the two types of pili are immunologically indistinguishable. It was concluded that PAK and PAK/2PfS pili are identical and that the mutation responsible for producing the multipiliated state in PAK/2PfS is probably located outside the structural gene for PAK pili.  相似文献

The characteristics of exotoxin inhibition of deoxyribonucleic acid (DNA) dependent ribonucleic acid (RNA) polymerase isolated from Escherichia coli and Bacillus thuringiensis were investigated. RNA polymerase isolated from a variety of growth stages was partially purified and assayed using several different native and synthetic DNA templates, and exotoxin inhibition patterns were recorded for each. Although 8 to 20-h RNA polymerase extracts of E. coli retained normal sensitivity to exotoxin (50% inhibition at a concentration of 7.5 X 10(-6) M exotoxin), RNA polymerase isolated from late exponential and ensuing stationary-phase cultures of B. thuringiensis were nearly 50% less sensitive than exponential RNA polymerase activity. Inhibition patterns relating culture age at the time of RNA polymerase extraction to exotoxin inhibition suggested a direct correlation between diminishing exotoxin sensitivity and sporulation. Escherichia coli RNA polymerase could be made to mimic the B. thuringiensis exotoxin inhibition pattern by removal of sigma from the holoenzyme. After passage through phosphocellulose, exotoxin inhibition of the core polymerase was 30% less than the corresponding inhibition of E. coli holoenzyme. Heterologous enzyme reconstruction and assay were not possible due to loss of activity from the B. thuringiensis preparation during phosphocellulose chromatography, apparently from the removal of magnesium. In enzyme velocity studies, inhibition with exotoxin was noncompetitive with respect to the DNA template in the RNA polymerase reaction.  相似文献

The entire amino acid sequence for Pseudomonas aeruginosa PAO pilin was determined through peptide sequencing and from the complete nucleotide sequence encoding the pilin gene. The precursor PAO pilin is 149 amino acids in length which includes a 6-amino-acid positively charged leader sequence. Comparison of the amino acid sequences of pilin produced by P. aeruginosa PAO and PAK reveals a region of high homology corresponding to the leader peptide and residues 1 to 54 of the mature pilin. The amino acid sequence of the peptide encompassing the major antigenic determinant of PAK differs greatly from that of the equivalent region in PAO. The C-terminal regions of these proteins are semiconserved. Few major differences were found when the predicted secondary structures for PAO and PAK pilins were compared. Major nucleotide sequence variation between the equivalent restriction fragments from PAO and PAK occurred within the areas coding for the peptides containing the immunodominant site for PAK pilin and the C termini.  相似文献

Limited proteolysis of Pseudomonas aeruginosa exotoxin A by four proteases (chymotrypsin, Staphylococcal serine proteinase, pepsin A and subtilisin) resulted in the formation of polypeptides having a molecular mass of approximately 25 kDa. They possessed both enzymatic activity and residual antigenicity. Their N-terminal sequence analysis showed that the different proteases cleaved exotoxin A in a very restricted area within domain Ib (amino acids 365-404). As a result, the polypeptides contained a large portion (13-34 amino acids) of domain Ib linked to the adjacent C-terminal domain III (amino acids 405-613). The major fragment derived from subtilisin cleavage, at a final yield of 35% (S-fragment; residues 392-613; 24201 Da; pI 4.7) possessed the same level of ADP-ribosyltransferase activity as uncleaved exotoxin A (by mass), and a 37-fold higher NAD-glycohydrolase activity. Polyclonal antibodies from rabbits against exotoxin A completely inhibited the ADP-ribosyltransferase activity of both exotoxin A and the S-fragment, but not the NAD-glycohydrolase activity of the S-fragment. Antibodies against the S-fragment neutralized the ADP-ribosyltransferase activity of exotoxin A. These data determine the primary proteolytic cleavage site of exotoxin A, suggest that some residues in the amino acid sequence 392-404 of exotoxin A seem to have a role in binding or positioning elongation factor 2 (EF-2) and show that antibodies recognize the EF-2-binding site but not the NAD(+)-binding site.  相似文献

Bad is a critical regulatory component of the intrinsic cell death machinery that exerts its death-promoting effect upon heterodimerization with the antiapoptotic proteins Bcl-2 and Bcl-x(L). Growth factors promote cell survival through phosphorylation of Bad, resulting in its dissociation from Bcl-2 and Bcl-x(L) and its association with 14-3-3tau. Survival of interleukin 3 (IL-3)-dependent FL5.12 lymphoid progenitor cells is attenuated upon treatment with the Rho GTPase-inactivating toxin B from Clostridium difficile. p21-activated kinase 1 (PAK1) is activated by IL-3 in FL5.12 cells, and this activation is reduced by the phosphatidylinositol 3-kinase inhibitor LY294002. Overexpression of a constitutively active PAK mutant (PAK1-T423E) promoted cell survival of FL5.12 and NIH 3T3 cells, while overexpression of the autoinhibitory domain of PAK (amino acids 83 to 149) enhanced apoptosis. PAK phosphorylates Bad in vitro and in vivo on Ser112 and Ser136, resulting in a markedly reduced interaction between Bad and Bcl-2 or Bcl-x(L) and the increased association of Bad with 14-3-3tau. Our findings indicate that PAK inhibits the proapoptotic effects of Bad by direct phosphorylation and that PAK may play an important role in cell survival pathways.  相似文献

The sensitivities of 21 mammalian cell lines to the exotoxins of Pseudomonas aeruginosa and Corynebacterium diphtheriae were measured. Each line exhibited 1-4 log differences in sensitivities to the two toxins. No species-specific sensitivities were noted for Pseudomonas exotoxin while diphtheria exotoxin was most potent in cells of monkey origin, followed by human and hamster cells. Rat- and mouse-derived cell lines were very insensitive to diphtheria exotoxin. The rates of cellular intoxication by both toxins exhibited apparent first-order kinetics and were indistinguishable from one another when equipotent doses were used. Our preparation of diphtheria exotoxin appeared to have a slightly higher ADP-ribosylating efficiency than did Pseudomonas toxin. However, neither toxin exhibited cell line-specific differences in ribosylating efficiencies which could have explained the wide range in potencies for intact cells. Our results suggest that there are significant differences in the mechanisms of cellular intoxication by Pseudomonas and diphtheria exotoxins and that these differences probably exist in the attachment or internalization stages of toxin action.  相似文献

Six human PAK members are classified into groups I (PAKs 1–3) and II (PAK4–6). Previously, only group I PAKs were thought to be auto-inhibited but very recently PAK4, the prototype of group II PAKs, has also been shown to be auto-inhibited by its N-terminal regulatory domain. However, the complete auto-inhibitory domain (AID) sequence remains undefined and the mechanism underlying its auto-inhibition is largely elusive. Here, the N-terminal regulatory domain of PAK4 sufficient for auto-inhibiting and binding Cdc42/Rac was characterized to be intrinsically unstructured, but nevertheless we identified the entire AID sequence by NMR. Strikingly, an AID peptide was derived by deleting the binding-unnecessary residues, which has a Kd of 320 nM to the PAK4 catalytic domain. Consequently, the PAK4 crystal structure complexed with the entire AID has been determined, which reveals that the complete kinase cleft is occupied by 20 AID residuescomposed of an N-terminal α-helix and a previously-identified pseudosubstrate motif, thus achieving auto-inhibition. Our study reveals that PAK4 is auto-inhibited by a novel mechanism which is completely different from that for PAK1, thus bearing critical implications for design of inhibitors specific for group II PAKs.  相似文献

Transforming growth factor beta (TGF-beta) causes growth arrest in epithelial cells and proliferation and morphological transformation in fibroblasts. Despite the ability of TGF-beta to induce various cellular phenotypes, few discernible differences in TGF-beta signaling between cell types have been reported, with the only well-characterized pathway (the Smad cascade) seemingly under identical control. We determined that TGF-beta receptor signaling activates the STE20 homolog PAK2 in mammalian cells. PAK2 activation occurs in fibroblast but not epithelial cell cultures and is independent of Smad2 and/or Smad3. Furthermore, we show that TGF-beta-stimulated PAK2 activity is regulated by Rac1 and Cdc42 and dominant negative PAK2 or morpholino antisense oligonucleotides to PAK2 prevent the morphological alteration observed following TGF-beta addition. Thus, PAK2 represents a novel Smad-independent pathway that differentiates TGF-beta signaling in fibroblast (growth-stimulated) and epithelial cell (growth-inhibited) cultures.  相似文献

Lysophosphatidic acid (LPA) has been shown to be a potent mitogen for vascular smooth muscle cells. Src-dependent transactivation of receptor tyrosine kinases has been previously demonstrated to mediate LPA-induced activation of MAP kinase ERK1/2. Furthermore, generation of reactive oxygen species (ROS) by LPA is also known to contribute to MAP kinase activation. Rho family small G-proteins Rac and Cdc42, and their immediate downstream effector p21-activated kinase (PAK), have been demonstrated to mediate important effects on the cytoskeleton that are relevant for cell migration and proliferation. In the present report we evaluated stimulation of PAK by LPA in rat aortic vascular smooth muscle cells (VSMC) by PAK immunocomplex MBP in-gel kinase assay. LPA increased PAK activity 3-fold, peaking at 5 min and showing sustained activation up to 45 min. Inhibition of tyrosine kinases by pretreatment of VSMC with genistein or specific inhibition of Src by PP1 greatly diminished LPA-induced PAK activation, whereas specific inhibition of PDFG- and EGF receptor kinase by tyrphostin AG1296 and AG1478 had no effect. Furthermore, inhibition of Galpha(i) by pertussis toxin and inhibition of NADH/NADPH oxidase by diphenylene iodonium also diminished LPA-induced stimulation of PAK. This is the first study to demonstrate that LPA activates PAK. In VSMC, PAK activation by LPA is mediated by Galpha(i) and is dependent on Src, whereas EGF- or PDGF receptor transactivation are not involved. Furthermore, generation of ROS is required for LPA-induced activation of PAK.  相似文献

An insecticidal exotoxin from Bacillus thuringiensis var. thuringiensis (Berliner) has been purified. The efficiency of each stage of the purification has been ascertained and the yield of toxic material estimated by means of a quantitative bioassay. It is shown that the exotoxin is an adenine derivative substituted at position 9 and having a molecular weight of approximately 825. It can be dephosphorylated enzymically or chemically under conditions that define the exotoxin as a phosphomonoester. This results in loss of toxicity, both to insects and to mice. Spectroscopic and kinetic data are presented which suggest that a beta-ribofuranosyl moiety may be attached to the adenine. Glucose and allomucic acid have been positively identified as hydrolysis fragments from the exotoxin. These results are discussed and compared with the results of others on similar (or possibly identical) compounds.  相似文献
