The stearoyl-CoA desaturase (delta-9-desaturase; SCD) gene is a candidate gene for fatty acid composition. It is located on pig SSC14 in a region where quantitative trait loci (QTL) for fatty acid composition were previously detected in a Duroc purebred population. The objective of the present study was to fine map the QTL, to identify polymorphisms of the pig SCD gene and to examine the effects of SCD polymorphisms on fatty acid composition and melting point of fat in the population. The pigs were examined for fatty acid composition and melting point of inner and outer subcutaneous fat and inter- and intramuscular fat; the number of pigs examined was 479-521. Two SNPs (g.-353C>T and g.-233T>C) were identified in the promoter region of the SCD gene and were completely linked in the pigs from the base generation. In all pigs, 19 microsatellite markers and SCD haplotypes were then genotyped. Different statistical models were applied to evaluate the effects of QTL and the possible causality of the SCD gene variants with respect to the QTL. The results show that all significant QTL for C14:0, C18:0, C18:1 and melting point of fat were detected in the same region, located near the SCD gene. The results also show a significant association between SCD haplotypes and fatty acid composition and fat melting point in this population. These results indicate that the haplotype of the SCD gene has a strong effect on fatty acid composition and melting point of fat.  相似文献

Omega-3-PUFAs, eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), are associated with prevention of various aspects of metabolic syndrome. In the present studies, the effects of oil rich in EPA on gene expression and activation of nuclear receptors was examined and compared with other ω3-PUFAs. The EPA-rich oil (EO) altered the expression of FA metabolism genes in THP-1 cells, including stearoyl CoA desaturase (SCD) and FA desaturase-1 and -2 (FASDS1 and -2). Other ω3-PUFAs resulted in a similar gene expression response for a subset of genes involved in lipid metabolism and inflammation. In reporter assays, EO activated human peroxisome proliferator-activated receptor α (PPARα) and PPARβ/γ with minimal effects on PPARγ, liver X receptor, retinoid X receptor, farnesoid X receptor, and retinoid acid receptor γ (RARγ); these effects were similar to that observed for purified EPA. When serum from a 6 week clinical intervention with dietary supplements containing olive oil (control), DHA, or two levels of EPA were applied to THP-1 cells, the expression of SCD and FADS2 decreased in the cells treated with serum from the ω3-PUFA-supplemented individuals. Taken together, these studies indicate regulation of gene expression by EO that is consistent with treating aspects of dyslipidemia and inflammation.  相似文献

茄科雷尔氏菌(Ralstonia solanacearum)是一种危害严重的土传植物致病菌,其宿主范围广泛,在世界各地严重影响重要经济作物的生产.研究茄科雷尔氏菌的生理特性,探索其致病机理,有利于研发防治青枯病的技术与方法.脂肪酸是细菌细胞重要的组成物质,但是茄科雷尔氏菌脂肪酸合成的机制尚不清晰.本文以茄科雷尔氏菌GMI1000为材料,鉴定了该菌的脂酰Co A脱饱和酶和环丙烷脂肪酸合成酶,并分析了这两种酶在不饱和脂肪酸和环丙烷脂肪酸合成中的作用.结果显示,茄科雷尔氏菌RSc2450编码脂酰Co A脱饱和酶,参与其不饱和脂肪酸合成,但是该菌还存在其他不饱和脂肪酸合成途径.同时发现在茄科雷尔氏菌编码两个可能的环丙烷脂肪酸合成酶蛋白质中,仅有Cfa1(RSc0776)参与了该菌环丙烷脂肪酸的合成,并在低p H和高渗透压的耐受中起作用.该研究结果为深入研究茄科雷尔氏菌脂肪酸合成代谢特点及致病机理奠定了基础.  相似文献

Herbaceous peony has been widely cultivated in China due to its substantial ornamental and medicinal value. In the present study, the phenotypic characteristics, total fatty acid (FA) content, and nine FA compositions of herbaceous peony seeds from 14 populations belonging to six species and one subspecies were determined by normal test and gas chromatography/mass spectrometry (GC/MS). The results showed that the phenotypic characteristics of seeds varied dramatically among species. The concentrations of five major FAs in seed oils were as follows: linoleic acid (173.95–236.51 μg/mg), linolenic acid (227.82–302.71 μg/mg), oleic acid (135.32–208.81 μg/mg), stearic acid (6.52–11.7 μg/mg), and palmitic acid (30.67–47.64 μg/mg). Correlation analysis demonstrated that oleic acid had the highest partial correlation coefficient with total FAs and might be applied to develop a model of phenotypic characteristics. FAs were significantly influenced by the following environmental factors: latitude, elevation, and annual average temperature. Based on the FA levels in the seed oils, clustering analysis divided 14 populations into two clusters. It was found that the average contents of oleic acid, linoleic acid, and total FAs in cluster I (147.16 μg/mg, 200.31 μg/mg, and 671.24 μg/mg, respectively) were significantly lower than those in cluster II (196.65 μg/mg, 220.16 μg/mg, and 741.78 μg/mg, respectively). Cluster I was perfectly consistent with subsect. Foliolatae, while cluster II was in good agreement with subsect. Dissectifoliae. Therefore, the FA composition of wild herbaceous peony seed oil might be used as a chemotaxonomic marker.  相似文献

为了研究蕨麻的化学成分以更好地利用蕨蔴资源,采用索氏提取法提取了青海和甘肃两个地区产的蕨麻中的脂溶性成分,利用气相色谱-质谱联用(GC-MS)对蕨麻中的脂肪酸成分进行分析。结果表明,来自甘肃产地的蕨麻含有11种脂肪酸,青海地区产的蕨麻含有7种脂肪酸,二者的共同成分有7种;甘肃产蕨麻中硬脂酸含量最高,辛烷含量最低;青海产蕨麻中邻苯二酚含量最高,棕榈酸含量最低。表明不同产地的蕨麻所含脂肪酸种类和脂肪酸含量均存在差异。  相似文献

寄生于人体的肠道菌群是一个高度动态化和个体化的复杂生态系统,受遗传、环境、饮食、年龄和运动等因素的影响,并通过其产生的代谢物与机体众多组织器官产生广泛的应答效应。短链脂肪酸(short chain fatty acid, SCFA)主要是由位于盲肠和结肠内的菌群以膳食纤维为底物发酵产生,其被吸收进入肠系膜上下静脉,随后汇入门静脉至肝。部分短链脂肪酸被肝作为糖异生和脂质合成的底物,剩余的短链脂肪酸以游离脂肪酸的形式经肝静脉进入外周循环。研究发现,运动可使产生SCFA的肠道菌群组分的丰度提高和参与调控SCFA生成的相关基因表达增加,使肠道中短链脂肪酸含量增加。由短链脂肪酸刺激结肠内分泌细胞合成分泌的胰高血糖素样肽1(glucagon like peptide-1, GLP-1)可促使胰岛B细胞合成分泌胰岛素,进而调节骨骼肌的葡萄糖摄取与糖原合成。此外,短链脂肪酸通过提高骨骼肌胰岛素受体底物1(insulin receptor substrate 1,IRS1)基因转录起始位点附近的组蛋白乙酰化水平,增强骨骼肌的胰岛素敏感性。同时,短链脂肪酸通过激活腺苷酸活化蛋白质激酶(AMP-activated protein kinase, AMPK)促进骨骼肌的脂肪酸摄取、脂肪分解和线粒体生物发生,抑制脂肪合成。本文就肠道菌群代谢物——短链脂肪酸概述、运动对产生短链脂肪酸的肠道菌群的影响和运动介导肠道菌群代谢物——短链脂肪酸对骨骼肌代谢调控机制的最新研究进展进行综述,为骨骼肌运动适应的新机制研究提供理论依据。  相似文献

In this study, we examined the effect of fatty acids on choline acetyltransferase (ChAT) activity. ChAT is unstable in a solution of low protein concentration, so serum albumin (BSA) is usually added to stabilize the enzyme. However, we found that ChAT from bovine caudate nucleus rapidly lost its activity when diluted with a buffer containing commercial preparations of BSA. This effect was caused by octanoic acid, which was found in the gas chromatography/mass spectrometry system of lipid extract in commercial BSAs. The inhibition of ChAT activity by octanoic acid depended on the concentrations of the octanoic acid and of the albumin. We also found that ChAT activity was decreased by some long-chain fatty acids, arachidonic acid having exhibited the strongest effect. The extent to which arachidonic acid inhibited ChAT activity depended on the molar ratio of arachidonic acid and albumin, rather than upon the concentration of arachidonic acid. The effect of octanoic acid and arachidonic acid on ChAT activity appeared to increase in the presence of albumin.  相似文献

通过气相色谱-质谱法分析了水酶法提取的石榴籽油的脂肪酸组成。结果表明:石榴籽油含有丰富的不饱和脂肪酸,总不饱和脂肪酸含量达94%以上。采用Schall烘箱法,以过氧化值(POV)为参考指标,研究了光线、温度及抗氧化剂对石榴籽油氧化稳定性能的影响。结果表明,温度、光线对石榴籽油的氧化过程都有影响,温度的影响更为显著;叔丁基对苯二酚(TBHQ)对石榴籽油具有良好的抗氧化效果,并与抗坏血酸(VC)具有较好的协同增效作用;生育酚(VE)和没食子酸丙酯(PG)对石榴籽油无显著抗氧化作用。  相似文献

Cell function is influenced by surface structure and molecules. Molecules that enhance cellular differentiation can be applied to tissue scaffold surfaces to stimulate endogenous tissue regeneration. The application of this approach to bone implants yields surfaces coated with factors (proteins, peptides, etc...) that promote the differentiation of osteoblasts, the cells that make bone. Increased bone formation leads to increased healing and union of the implant with endogenous bone. To obtain better control over surface coating we developed PLLA copolymers with allyl (PLLA-co-DAG) and 3-hydroxypropyl (PLLA-co-HP) side chains to which we can attach functional groups. Given the potential of fatty acids being able to incorporate into lipid bilayers and/or influence gene expression, we grafted different fatty acid side chains to PLLA-co-HP by esterifying the corresponding fatty acids with the PLLA-co-HP 3-hydroxypropyl side chains. The effects of the polymer modifications on osteoblasts were then evaluated. While cellular morphology differed between surface coatings, they did not reflect changes in cellular phenotype. Changes in gene expression were most evident with arachidonate and 3-hydroxypropyl side-chains which exhibited osteoblast differentiating capabilities. Linoleate, myristate, oleate, and stearate ester side-chains did not have a significant influence on osteoblast phenotype. Growth characteristics of osteoblasts did not differ between the fatty acid copolymer films, although cells grown on PLLA-co-HP exhibited a trend toward increased growth. Taken together our findings demonstrate that surface fatty acid composition can impact osteoblast phenotype.  相似文献

The effects of arachidonic acid and other polyunsaturated fatty acids (PUFAs) on both oxidative and metabolic perturbation were studied in primary cultures of rat cerebral cortical astrocytes. In the presence of 0.1 mM arachidonic acid, the rate of the reduction of nitroblue tetrazolium (NBT) to nitroblue formazan (NBF) was stimulated from 0.65 +/- 0.10 to 1.43 +/- 0.15 and from 0.092 +/- 0.006 to 0.162 +/- 0.009 nmol/min/mg protein in intact and broken cell preparations, respectively. The rate of superoxide radical formation, as measured by the superoxide dismutase (SOD)-inhibitable NBT reduction was 0.042 nmol/mg protein in broken cells and was negligible in intact cells. The latter is due to the impermeability of SOD into the intact cell preparation. NBF formation in intact astrocytes stimulated by arachidonic acid was both time- and dose-dependent. Other PUFAs, including linoleic acid, linolenic acid, and docosahexaenoic acid, were also effective in stimulating NBF formation in astrocytes, whereas saturated palmitic acid and monounsaturated oleic acid were ineffective. Similar effects of these PUFAs were observed in malondialdehyde formation in cells and lactic acid accumulation in incubation medium. These data indicate that both membrane integrity and cellular metabolism were perturbed by arachidonic acid and by other PUFAs. The sites of superoxide radical formation appeared to be intracellular and may be associated with membrane phospholipid domains, because liposome-entrapped SOD, which was taken up by intact astrocytes, reduced the level of superoxide radicals and lactic acid content, whereas free SOD was not effective.  相似文献

用电子自旋共振(ESR)技术对液晶态多烯脂肪酸多相脂质体与癌细胞膜相互作用进行了研究. 并探讨了其在抑制和杀伤癌细胞过程中可能具有的生物学意义.实验发现:油酸多相脂质体的影响使自旋标记物在Ec腹水肝癌细胞膜上的强固定化作用减弱, 弱固定化作用增强,使自旋标记物运动自由度增加.亚油酸多相脂质体的影响使自旋标记物在乳腺癌细胞膜上的强固定化作用增强, 弱固定化作用减弱,使自旋标记物运动自由度受到限制.蓖麻酸多相脂质体的影响使自旋标记物在S180实体瘤细胞膜上的强固定化作用增强, 弱固定化作用减弱,使自旋标记物运动自由度受到限制.结果表明,多烯脂肪酸多相脂质体作用于膜蛋白引起了膜蛋白构象的变化.  相似文献

Abstract: The present study was undertaken to determine whether polyunsaturated fatty acid metabolism is affected by high glucose levels in cerebral and retinal microvascular endothelial cells. The metabolism of [3-14C]22:5n-3 and [1-14C]18:2n-6 was studied in cells previously cultured for 5 days in normal (5 m M ) or high (30 m M ) glucose medium. After incubation of retinal endothelial cells with [3-14C]22:5n-3 in the high glucose condition, the formation of labeled 24:6n-3 and 22:6n-3 was increased, and that of labeled 24:5n-3 was decreased, compared with the normal glucose condition. The changes were found for fatty acids esterified in cellular lipids and those released into the medium. After incubation with [1-14C]18:2n-6, levels of all elongation/desaturation products were increased at the expense of the precursor in retinal endothelial cells cultured in high glucose medium. The changes were primarily found for esterified fatty acids, with the release of n-6 fatty acids being minor in both glucose concentrations. By contrast, high glucose levels did not affect the metabolism of [3-14C]22:5n-3 and [1-14C]18:2n-6 in cerebral endothelial cells. The changes in metabolic activity of retinal endothelial cells were not reflected in the fatty acid composition. The present data suggest that high glucose can increase the desaturation process in retinal but not cerebral endothelial cells. This may produce some lipid abnormalities in retinal microvasculature and contribute to altered vascular function observed in diabetic retinopathy.  相似文献

The kinetics of lipid-hydrolysis by Candida rugosa lipase was investigated in a membrane reactor and in an emulsion system. Two models were chosen to describe the kinetics of the enzyme:

(1) The hydrolysis of triglycerides to fatty acids was considered to be a chain reaction with the intermediary products di- and mono-glyceride; each step was assumed to be a reversible second-order reaction. The reaction rate constants were determined from batch experiments. The experimental results could be described with this model.

(2) For process optimization and control, a model based on the power law was developed. For this model, the rate of hydrolysis was measured as a function of fatty acid and glycerol concentrations. Relations for the initial rate and equilibrium ester fraction as a function of the glycerol concentration were determined. Further, the reaction rate could be described with the power-law model with a power of 1.75 in the hydrolyzable ester fraction for a wide range of glycerol concentrations. The model with power 1.75 gave much better results when compared to a similar first order model. Although simpler, the first order model can not be used. The power law model was applied in the simulation of a reactor composed of three modules. The fatty acid production rate was calculated for this reactor system as a function of the outgoing glycerol concentration at different conditions.  相似文献

The fatty acid composition of ER, Golgi and peribacteroid membrane (PBM) from root nodules formed on Glycine max after infection with different strains of Bradyrhizobium japonicum has been analysed by gas chromatography. In each plant-microsymbiont combination the fatty acid composition (FAC) of the PBM is distinct from ER and Golgi. The similarity between ER and PBM fatty acid composition is significantly stronger than between Golgi and PBM. In addition the fatty acid composition of all membrane systems in nodules is affected by the microsymbiont strain. A comparison of four strains of Bradyrhizobium japonicum grown in agar surface culture and isolated as the symbiotic bacteroids reveals a decrease in oleic acid during bacteroid differentiation.  相似文献
