Photosynthetic parameters were determined in disks from leaves of C. arabica cv. Red Catuaí and C. canephora cv. Kouillou grown in the field. Kouillou showed a relatively higher irradiance requirement for saturating photosynthesis, lower chlorophyll (Chl) content, and higher Chl a/b ratio than Catuaí. Photoinhibition of photosynthesis under bright irradiance was manifested by decreases in maximum photochemical efficiency (evaluated by the variable to maximum fluorescence ratio, Fv/Fm), as a consequence of an increased initial and a quenched maximum fluorescence. Restoration of Fv/Fm following photoinhibition in low irradiance was faster in Kouillou than in Catuaí. Chloramphenicol both accelerated photoinhibition (mainly in Kouillou) and blocked its recovery for at least 190 min in either cultivar. Photosynthetic oxygen evolution under photoinhibitory conditions was decreased by chloramphenicol; in control leaf disks this decrease was only observed in C. arabica, but with a rapid recovery within 90 min of low irradiance exposure. In both coffee cultivars, the depressed photochemical efficiency of photosystem 2 was not accompanied by a concomitant lowering in oxygen evolution during reversal from photoinhibition.  相似文献

Coffea arabica cv. Red Catuaí and C. canephora cv. Kouillou were grown in pots beneath a plastic shelter. When they were 14 months old, irrigation was withheld until the leaf pre-dawn water potential was about -1.5 and -2.7 MPa (designated mild and severe water stress, respectively). Under mild stress, net photosynthetic rate (PN) decreased mainly as a consequence of stomatal limitations in Kouillou, whereas such decreases were dominated by non-stomatal limitations in Catuaí. Under severe drought, further decreases in PN and apparent quantum yield were not associated to any changes in stomatal conductance in either cultivar. Decreases were much more pronounced in Catuaí than in Kouillou, the latter maintained carbon gain at the expense of water conservation. In both cultivars the initial chlorophyll (Chl) fluorescence slightly increased with no changes in the quantum efficiency of photosystem 2. In response to rapidly imposed drought, the Chl content did not change while saccharide content increased and starch content decreased. Photoinhibition and recovery of photosynthesis, as evaluated by the ratio of variable to maximum fluorescence and by the photosynthetic O2 evolution, were unaffected by mild drought stress. Photoinhibition was enhanced under severe water deficit, especially in Catuaí. In this cultivar the O2 evolution did not resume upon reversal from photoinhibition, in contrast to the complete recovery in Kouillou.  相似文献

In the bark of Populus tremula L. photochemical efficiency of photosystem 2 (PS2) determined as Fv/Fm decreased during winter. The strongest reduction was found after cold periods. The degree of reduction depended on irradiance since the lowest levels of Fv/Fm were found on the sun-exposed side of the stem and below thin phellem. Therefore, photoinhibition was partly responsible for the reduction in Fv/Fm. The photochemical efficiency of PS2 recovered in late April about a month before the trees got leaves. In the laboratory, Fv/Fm recovered within about a week under low irradiance at 20 °C. Rapid recovery of photochemical efficiency of PS2 in the bark may be important to reduce respiratory loss of CO2 from the stem before the trees get leaves.  相似文献

In maize (Zea mays L., cv Contessa), nitrogen (NO3) limitation resulted in a reduction in shoot growth and photosynthetic capacity and in an increase in the leaf zeaxanthin contents. Nitrogen deficiency had only a small effect on the quantum yield of CO2 assimilation but a large effect on the light-saturated rate of photosynthesis. Linear relationships persisted between the quantum yield of CO2 assimilation and that of photosystem II photochemistry in all circumstances. At high irradiances, large differences in photochemical quenching and nonphotochemical quenching of Chl a fluorescence as well as the ratio of variable to maximal fluorescence (Fv/Fm) were apparent between nitrogen-deficient plants and nitrogen-replete controls, whereas at low irradiances these parameters were comparable in all plants. Light intensity-dependent increases in nonphotochemical quenching were greatest in nitrogen-deficient plants as were the decreases in Fv/Fm ratio. In nitrogen-deficient plants, photochemical quenching decreased with increasing irradiance but remained higher than in controls at high irradiances. Thermal dissipative processes were enhanced as a result of nitrogen deficiency (nonphotochemical quenching was elevated and Fv/Fm was lowered) allowing PSII to remain relatively oxidised even when carbon metabolism was limited via nitrogen limitation.  相似文献

Cold acclimation and photoinhibition of photosynthesis in Scots pine   总被引:13,自引:0,他引:13  
Cold acclimation of Scots pine did not affect the susceptibility of photosynthesis to photoinhibition. Cold acclimation did however cause a suppression of the rate of CO2 uptake, and at given light and temperature conditions a larger fraction of the photosystem II reaction centres were closed in cold-acclimated than in nonacclimated pine. Therefore, when assayed at the level of photosystem II reaction centres, i.e. in relation to the degree of photosystem closure, cold acclimation caused a significant increase in resistance to photoinhibition; at given levels of photosystem II closure the resistance to photoinhibition was higher after cold acclimation. This was particularly evident in measurements at 20° C. The amounts and activities of the majority of analyzed active oxygen scavengers were higher after cold acclimation. We suggest that this increase in protective enzymes and compounds, particularly Superoxide dismutase, ascorbate peroxidase, glutathione reductase and ascorbate of the chloroplasts, enables Scots pine to avoid excessive photoinhibition of photosynthesis despite partial suppression of photosynthesis upon cold acclimation. In different marine red algae (Chondrus crispus, Delesseria sanguinea, Membranoptera alata, Phycodrys rubens, Phyllophora truncata, Polyneura hilliae) photoinhibition of photosynthesis has been investigated by means of both fluorescence and oxygen measurements. Measurements of absolute oxygen production show that photoinhibition causes a decline in the initial slope and in the rate of bending of the fluence rate-response curve (i.e. the photosynthetic efficiency at non-saturating fluence rates), as well as a decline in the photosynthetic capacity (Pm) at saturating fluence rates. Fluorescence data (Fv/Fm) were consistent with the results of oxygen measurements. Under excessive light photoinhibition protects photosynthesis against photo-damage in red algae. However, an increase in the initial fluorescence (Fo) after photoinhibitory treatment indicates that it could not prevent photodamage entirely. Action spectra of photoinhibition demonstrate that the main photoinhibition site in Polyneura hiliae is PS II, because far red light absorbed by PS I was ineffective. The strong increase of Fo in the blue wavelength range and the slight and partial recovery in weak blue light indicate that blue light especially causes photodamage. Recovery of photosynthesis requires dim white light conditions. Experiments with monochromatic light also show a wavelength dependence of recovery. Moreover, the recovery of photosynthesis after a photoinhibitory treatment is strongly temperature dependent, indicating participation of enzymatic processes. The comparison of fluorescence and oxygen measurement of the recovery shows different results in some species. The rate of oxygen production in red control light increased immediately after photoinhibited algae were exposed to weak light conditions. Surprisingly, the ratio of variable to maximum fluorescence (Fv/Fm) of Phyllophora truncata and the maximum fluorescence (Fm) of Polyneura hilliae show first a delay of the recovery under weak light conditions. Thus, in recovery experiments fluorescence and oxygen data are not quite consistent.  相似文献

Photoinhibition of photosynthesis in the brown alga, Dictyota dichotoma, was studied with a PAM fluorometer (Walz, Effeltrich, Germany) and a homemade oxygen measuring device. As a measure of fluorescence, Fv/Fm, and for the photosynthetic yield, ΔF/Fm', were used. Oxygen measurements show clearly that the observed degree, as well as the time course, of photoinhibition depends on the fluence rate of the light used to measure changes of the production rate. After photoinhibition of photosynthesis the depression of oxygen production caused by non-saturating fluence rates was generally much more pronounced than that caused by saturating or nearly saturating fluence rates. At minimal photoinhibition the initial slope and the convexity of the fluence rate-response curve of oxygen evolution decrease, whereas the level of light saturation decreases only after strong photoinhibition. Nevertheless, at different degrees of photoinhibition, changes in the degree of the upper bending of the fluence rate-response curve of oxygen production are also linearly correlated to changes in the fluorescence ratios (Fv/Fm and ΔF/Fm'). The action spectrum of photoinhibition, calculated on the basis of changes of Fv/Fm, indicates that the reaction center of PS I is not involved in photoinhibition. The lower effectiveness of blue light in comparison to effects of green and red light may be due to chloroplast displacement, as in the so-called strong light position, the light absorbed by the thalli in vivo is decreased.  相似文献

A study was conducted, using rapid time course of chlorophyll (Chl) fluorescence parameters, and light-response curves of Chl fluorescence parameters, to determine the induction requirements and response of photosystem II (PSII) photochemistry and non-photochemical reactions after changes in irradiance in greenhouse mulberry plants. The induction of PSII photochemistry rapidly approached to steady state after leaves were treated from darkness to low irradiance (LI). When irradiance of leaves changed from darkness to high irradiance (HI), a biphasic induction was observed. A slight photoinhibition occurred in the leaves exposed to sunlight coming to the greenhouse, whereas a chronic photoinhibition occurred in the leaves fully exposed to sunlight outside the greenhouse. The chronic photoinhibition was demonstrated by sustained reduction of maximal quantum yield of PSII photochemistry (Fv/Fm). Moreover, the leaves of mulberry plants in greenhouse were sensitive to abrupt changes in irradiance and the sensitivity of leaves suffered in a short-term (1h) high light treatment was reduced, based on the changes in photosynthetic quantum conversion. These results demonstrated an inducible response of photosynthetic quantum conversion to changes in irradiance in mulberry.  相似文献

Effect of photoinhibition of sorghum leaves and isolated chloroplasts on chlorophyll fluorescence, peroxidation of thylakoid lipids and activity of antioxidant enzymes were studied. Photoinhibition of intact leaves and isolated chloroplasts decreased Fv/Fm ratio and qP, while qN increased. Photoinhibitory damage was more at 5 degrees C than at 30 degrees or 50 degrees C. Peroxidation of thylakoid lipids was 5 times greater when photoinhibited at 50 degrees C compared to control. Photoinhibition of chloroplasts under low oxygen condition or when supplemented with anti-oxidants (beta-carotene, ascorbate and GSH) resulted in significantly less damage to photosynthesis (Fv/Fm ratio) and peroxidation level. Photoinhibition also resulted in many fold increase in the activity of superoxide dismutase (SOD) and ascorbate peroxidase (APX) and decrease in catalase. Data presented here suggest that photoinhibition resulted in production of oxygen radicals and photoinhibition of chloroplasts in the presence of low oxygen level or when supplemented with antioxidants decreased the damage to Fv/Fm ratio and peroxidation level to a great extent since former prevented the formation of oxygen radicals and later could scavenge the oxygen radicals thus the protection. Increase activity of SOD and APX may also be to metabolise the oxygen radicals produced during photoinhibition treatment, thereby, protecting the seedlings against photooxidative damage.  相似文献

Diurnal heliotropic leaf movements, photosynthetic gas exchange, and the ratio of variable fluorescence to maximum fluorescence (Fv/Fm) of unrestrained and horizontally restrained leaves from soybean (Glycine max cv. Cumberland) plants grown in two different water and two different nitrogen treatments were measured. Leaves of plants grown in low water or low nitrogen availability treatments displayed more pronounced diaheliotropism (solar tracking) in the afternoon and a longer period of paraheliotropism (light avoiding) at midday relative to those of well-watered, high-nitrogen-grown plants. Photosaturated photosynthetic rates and the photon flux required to saturate photosynthesis were reduced by water stress and nitrogen deficiency. Compared to horizontal leaves, irradiance on orienting leaves was nearer to the breakpoint of the photosynthetic light response curve, where photosynthesis is co-limited by ribulose biphosphate regeneration and carboxylation. This would increase the carbon return on investments of nitrogen into photosynthesis. A positive linear relationship between Fv/Fm and quantum yield of photosynthesis was measured. Leaves of low-nitrogen-grown plants had earlier and more prolonged reductions in Fv/Fm at midday compared to leaves of high nitrogen grown plants of the same water treatment. Within the same water and nitrogen treatment, horizontally restrained leaves had lower midday Fv/Fm in relation to orienting leaves. Nitrogen deficiency and water stress enhanced this difference such that horizontally restrained leaves of low water and low nitrogen grown plants had earlier and longer midday depressions in Fv/Fm.  相似文献

Photosynthetic light curve, chlorophyll (Chl) content, Chl fluorescence parameters, malondialdehyde (MDA) content, phosphoenolpyruvate carboxylase (PEPC) activity and reactive oxygen metabolism were studied under drought stress in two autotetraploid rice lines and corresponding diploid rice lines. Net photosynthetic rate decreased dramatically, especially under severe drought stress and under high photosynthetic active radiation in diploid rice, while it declined less under the same conditions in autotetraploid lines. Compared with the corresponding diploid lines, the Chl content, maximum photochemical efficiency of photosystem (PS) II, and actual photochemical efficiency of PSII were reduced less in autotetraploid lines. PEPC activities were higher in autotetraploid rice lines. PEPC could alleviate inhibition of photosynthesis caused by drought stress. The chromosome-doubling enhanced rice photoinhibition tolerance under drought stress. The lower MDA content and superoxide anion production rate was found in the autotetraploid rice indicating low peroxidation level of cell membranes. At the same time, the superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities were higher in autotetraploid rice lines. SOD, POD, and CAT could effectively diminish the reactive oxygen species and reduced the membrane lipid peroxidation.  相似文献

To determine whether the net loss of D1 protein is the main cause of photoinhibition of photosynthesis in wheat leaves under field conditions in the absence of any environmental stress other than strong sunlight, the D1 protein content, photosynthetic evolution of oxygen and chlorophyll a fluorescence parameters were measured in field grown wheat leaves. After exposure to midday strong light for about 3 h, apparent photosynthetic quantum efficiency (Φ), Fv/Fm and Fo in wheat leaves declined, and these parameters recovered almost completely 1 h after transfer to the weak light of 30~40 ttmol photons · m-2 · s-1. No evident change in the D1 protein content was observed in the leaves after exposure to midday strong light for 3 h. After 3 hours exposure to strong light, the slow-relaxed fluorescence quenching in the leaves treated with streptomycin (SM) increased much more than that in the control leaves, but there was no effect SM on the recovery of Fv/Fm and F0; dithiothretol (DTT) treatment enhanced photoinhibition of photosynthesis and reduced the D1 protein content in the leaves after exposure to midday strong light. These results indicated that under field conditions with no environmental stress other than strong sunlight, photoinhibition of photosynthesis in wheat leaves was not due to the net loss of D1 protein, and it could be attributed mainly by the increased nonradiative energy dissipation.  相似文献

Under 30-min high irradiance (1500μmol m^-2 s^-1), the roles of the xanthophyll cycle and D1 protein turnover were investigated through chlorophyll fluorescence parameters in morning glory (Ipomoea setosa) leaves, which were dipped into water, dithiothreitol (DTT) and lincomycin (LM), respectively. During the stress, both the xanthophyll cycle and D1 protein turnover could protect PSI from photoinhibition. In DTT leaves, non-photochemical quenching (NPQ) was inhibited greatly and the oxidation level of P700 (P700^+) was the lowest one. However, the maximal photochemical efficiency of PSII (Fv/Fm) in DTT leaves was higher than that of LM leaves and was lower than that of control leaves. These results suggested that PSI was more sensitive to the loss of the xanthophyll cycle than PSII under high irradiance. In LM leaves, NPQ was partly inhibited, Fv/Fm was the lowest one among three treatments under high irradiance and P700^+ was at a similar level as that of control leaves. These results implied that inactivation of PSII reaction centers could protect PSI from further photoinhibition. Additionally, the lowest of the number of active reaction centers to one inactive reaction center for a PSII cross-section (RC/CSo), maximal trapping rate in a PSll cross-section (TRo/CSo), electron transport in a PSll cross-section (ETo/CSo) and the highest of 1-qP in LM leaves further indicated that severe photoinhibition of PSII in LM leaves was mainly induced by inactivation of PSII reaction centers, which limited electrons transporting to PSh However, relative to the LM leaves the higher level of RC/CSo, TRo/CSo, Fv/Fm and the lower level of 1-qP in DTT leaves indicated that PSI photoinhibition was mainly induced by the electron accumulation at the PSI acceptor side, which induced the decrease of P700^+ under high irradiance.  相似文献

对两种不同生长光强下(自然光的8%和50%)西双版纳热带雨林木本植物藤黄(Garcinia han-buryi)幼苗经夜间低温(4℃)处理后荧光特性和活性氧代谢的研究结果表明,低温使藤黄叶片光合机构PSⅡ原初光能转化效率(Fv/Fm)、PSⅡ非环式电子传递的量子效率(ФPSⅡ)、非光化学猝灭系数(NPQ)下降,原初荧光(F0)上升。低温胁迫消除后,生长在50%光强下藤黄叶片的Fv/Fm和F0在3d后仍不能完全恢复,而生长在8%光强下藤黄叶片的Fv/Fm和F0基本恢复,说明低温使生长在8%光强下藤黄的光合机构PSⅡ反应中心受到可逆失活,而生长在50%光强下藤黄的光合机构受到氧化伤害。随着低温胁迫时间的延长,两种生长光强藤黄叶片活性氧保护酶(SOD,CAT,APX)的活性虽升高,但O2^-的生成速率、H2O2和MDA含量积累增加。而在恢复阶段,生长在8%光强比生长在50%光强下藤黄叶片的活性氧含量下降得快,进一步说明生长在高光强的植物比生长在低光强的植物受低温伤害大。  相似文献

Net rates of photosynthesis (PN) saturated by irradiance of >500 mol m-2 s-1 (PAR) significantly decreased in water-stressed potato (Solanum tuberosum L. cv. Kufri Sindhuri) plants. The quantum yield of photochemical energy conversion (Fv/Fm), relative electron transport rate (ETR), and photochemical quenching (Qp) exhibited a parallel decline at high irradiance. A slight decrease in relative water content (RWC) was accompanied by a drastic decline in leaf water potential (w) from -0.2 to -1.0 MPa. Dehydrated leaves showed an increase in the amount of total soluble sugars per unit leaf area which inhibited the photosynthesis in a feedback manner. After rewatering, PN and Fv/Fm were restored to the values of control plants within 24 h, and the restoration was accompanied by a proportionate lowering of content of total soluble sugars in the leaves.  相似文献

以湖南省永州市冷水滩采穗圃中的美国山核桃为试材,研究了叶绿素荧光参数的日变化规律。结果表明:初始荧光(Fo)、最大荧光(Fm)、PSII原初光能转化效率(Fv/Fm)、光合量子产额(Yield)、光化学猝灭系数(qP)、非光化学猝灭系数(qN)和表观电子传递速率(ETR)均存在着明显的日变化。其中Fv/Fm、Fm、Yield、qP均呈先下降后上升的趋势,在中午强光下降低到最低值;qN则呈先上升后下降的趋势,在中午时分达到峰值;Fo呈下降趋势,部分品种傍晚稍有回升,但仍比早晨低;ETR日变化呈双峰曲线。不同品种间Fv/Fm、Yield、ETR、qP、qN对光强和温度的响应也存在着明显差异,可作为鉴定品种耐光抑制能力大小的指标。  相似文献

The responses to photoinhibition of photosynthesis at low temperature and subsequent recovery were examined in Arabidopsis thaliana (ecotype Columbia) developed at 4°C cold-acclimating conditions, 23°C non-acclimating conditions and for non-acclimated plants shifted to 4°C (cold-shifted). These responses were determined in planta using Chl fluorescence imaging. We show that cold acclimation results in an increased tolerance to photoinhibition in comparison with non-acclimated plants and that growth and development at low temperature is essential for this to occur. Cold-shifted plants were not as tolerant as the cold-acclimated plants. In addition, we demonstrate this tolerance is as a result of growth under high PSII excitation pressure, that can be modulated by growth temperature or growth irradiance. Cold-acclimated and cold-shifted plants fully recover from photoinhibition in the dark, whereas non-acclimated plants show reduced levels of recovery and demonstrate a requirement for light. The role of the PSII repair cycle, PSII quenching centres, and the use of Chl fluorescence imaging to monitor photoinhibitory responses in planta are discussed.  相似文献

以建兰栽培品种‘八宝奇珍’Cymbidium ensifolium ‘Ba Bao Qi Zhen’为材料,对其正常绿色叶片及黄化变异叶片的光合色素含量、叶绿素荧光参数进行比较。结果表明,‘八宝奇珍’黄化变异叶片的叶绿素总量、叶绿素a 和叶绿素b 含量均显著低于正常绿色叶片,且随着黄化面积的增大呈现递减趋势;黄化变异叶片的初始荧光量(Fo)、最大荧光产量(Fm)、Kautsky 诱导效应最大荧光(Fp)、稳态光适应光化学淬灭系数(qP)以及光适应稳态荧光产量(Ft_Lss)均显著低于正常绿色叶片;PSⅡ原初光能转化效率(Fv/Fm)与正常绿色叶片无明显差异;稳态非光化学荧光淬灭系数(NPQ)高于正常绿色叶片。  相似文献
