目的:探讨CD90在肝细胞肝癌(HCC)患者肿瘤组织中的表达及其与临床病理参数的关系,为HCC的临床治疗提供参考。方法:选择2014年3月-2015年3月在我院接受治疗的HCC患者80例,收集患者肿瘤组织及癌旁组织,另选择同期就诊的80例正常肝脏组织作为对照组。观察并比较CD90在肝癌组织、癌旁组织及正常肝脏组织中的表达情况。结果:CD90在肝癌组织、癌旁组织及正常肝脏组织中呈不同程度阳性表达(P0.05)。CD90在肝癌组织和癌旁组织中的表达显著高于正常肝脏组织,差异具有统计学意义(P0.05)。CD90在肝癌组织中的表达显著高于癌旁组织,差异具有统计学意义(P0.05)。CD90在HCC组织中的表达与肿瘤大小、肝内转移、TMN分期和病理分级有关(P0.05)。结论:CD90在肝癌组织中呈高表达,其表达水平与肝癌临床病理参数有关,可以作为HCC诊断、治疗及预后评估的参考指标。  相似文献

This study aimed to investigate the expression of Twist in gastric cancer tissues and its correlation between Twist and the epithelial-mesenchymal transition (EMT). By means of RT-PCR and Western blot, the mRNA and protein expressions of Twist, E-cadherin, and Vimentin in 61 gastric cancer tissues and adjacent normal tissues were detected. The positive rates of Twist, E-cadherin, and Vimentin mRNA expression in gastric cancer tissues were 73.9. 40.6, and 60.9 %, respectively; compared to the expression of these genes in adjacent normal tissues (2.9, 75.4, and 27.5 %), the differences were significant (p < 0.05). The E-cadherin protein expression level in gastric cancer tissues was significantly lower than that in the adjacent normal tissues (p < 0.05). After the transfection of Twist siRNA into the MKN45 cells, the protein expression of Twist was significantly reduced (p < 0.05), the protein expression of E-cadherin was significantly increased, and the number of cells that passed through the Transwell chamber was significantly lower than that in the non-transfected control group as well as the transfected control group (p < 0.05). Twist may be associated with the epithelial-mesenchymal transition in gastric cancer and the tumorigenesis, invasion, and metastasis of gastric cancer.  相似文献

目的:探讨乳腺癌中NF2基因启动子甲基化状态及其mRNA水平与乳腺癌发病的关系.方法:应用甲基化特异性聚合酶链反应(MSP)和逆转录-聚合酶链反应(RT-PCR)技术,检测47例乳腺癌组织及相应的癌旁组织和15例乳腺良性病变组织,分析NF2基因的甲基化与某些临床参数及mRNA表达的关系.结果:NF2基因启动子区在乳腺癌、癌旁和乳腺良性病变组织中的甲基化频率分别为57.4%(27/47)、23.4%(23/47)和0%(0/15).且乳腺癌组明显高于其余两组(P<0.05).NF2基因发生甲基化与发病年龄、组织分型、转移和组织分级无相关性.乳腺癌组NF2基因mRNA的相对表达量(0.16±0.11)明显低于相应的癌旁组(0.27±0.14)及乳腺良性病变组(0.64±0.17)(P<0.05).NF2基因启动子区甲基化频率与其mRNA表达呈负相关(Spearman's r=-0.314,P<0.05).结论:NF2基因发生甲基化与乳腺癌的发生密切相关,NF2mRNA表达与NF2基因启动子高甲基化呈负相关.  相似文献

It has been demonstrated recently that upper forelimbs of axolotls comprised of symmetrically arranged soft tissues do not regenerate (P. W. Tank, 1978,J. Exp. Zool.204, 325–336). These double-half forelimb stumps contained skin, muscle, and loose connective tissues in symmetrical arrangement. The present study explores the roles of muscle, skin, and epidermis in the regeneration of double-half forelimbs by grafting them separately to create forelimb stumps bearing symmetrical arrangements of these individual tissues. Forelimb stumps bearing symmetrically arranged flexor and extensor muscles and normally arranged skin underwent complete regeneration (96%). Forelimbs comprised of double-half skin overlying normally arranged muscles and deep tissues formed hypomorphic structures and nonregenerates (56%) with some single and multiple regenerates. Limbs with double-half deep tissues and complete epidermis either regenerated distally incomplete patterns (47%), single patterns (33%), or multiple patterns (20%). Those forelimbs comprised of double-half skin and no muscle regenerated incomplete patterns in the majority of cases (56%) but single and multiple limbs also were formed. Based on these results it can be concluded that no single type of tissue is solely responsible for the regenerative failure experienced by double-half forelimbs in the earlier study. The complete failure of forelimb regeneration occurs only when all types of soft tissues tested (skin, muscle, and deep connective tissues) are present in symmetrical arrangement.  相似文献

A diverse array of bacterial species, including several potential human pathogens, was isolated from edible crabs collected in cold waters. Crabs collected near Kodiak Island, Alaska, contained higher levels of bacteria than crabs collected away from regions of human habitation. The bacteria associated with the crabs collected near Kodiak included Yersinia enterocolitica, Klebsiella pneumoniae, and coagulase-negative Staphylococcus species; the pathogenicity of these isolates was demonstrated in mice. Although coliforms were not found, the bacterial species associated with the tissues of crabs collected near Kodiak indicate possible fecal contamination that may have occurred through contact with sewage. Compared with surrounding waters and sediments, the crab tissues contained much higher proportions of gram-positive cocci. As revealed by indirect plate counts and direct scanning electron microscopic observations, muscle and hemolymph tissues contained much lower levels of bacteria than shell and gill tissues. After the death of a crab, however, the numbers of bacteria associated with hemolymph and muscle tissues increased significantly. Microcosm studies showed that certain bacterial populations, e.g., Vibrio cholerae, can be bioaccumulated in crab gill tissues. The results of this study indicate the need for careful review of waste disposal practices where edible crabs may be contaminated with microorganisms that are potential human pathogens and the need for surveillance of shellfish for pathogenic microorganisms that naturally occur in marine ecosystems.  相似文献

Disturbances of nitrogen metabolism under acute ammonium toxicosis have been studied in tissues of rabbit. A sharp increase of the ammonium content in the blood and tissues of the liver and kidneys is accompanied by an increase in the glutamine and glutamate level in all tissues. The level of urea nitrogen in the blood of rabbits increases. The activity of phosphate-independent and phosphate-activated glutaminase also increases in tissues of the liver and kidneys, while arginase activity decreases as compared with the control, which is connected with fall of the ATP level under hyperammonemia. 目的:检测网织钙结合蛋白2(RCN2)和伪足富集的非典型激酶1(PEAK1)蛋白在结直肠癌组织中的表达情况,分析RCN2和PEAK1表达与患者临床病理特征和预后的关系。方法:免疫组织化学法检测90例结直肠癌组织及其癌旁正常组织中RCN2和PEAK1蛋白表达情况,分析结直肠癌组织RCN2和PEAK1表达与患者临床病理特征的关系,Kaplan-Meier生存曲线分析RCN2和PEAK1表达对患者预后的影响,Spearman等级相关检验结直肠癌组织RCN2和PEAK1表达的相关性。结果:RCN2和PEAK1蛋白在结直肠癌组织中的阳性表达率均明显高于癌旁正常组织(P0.05)。结直肠癌组织RCN2表达与肿瘤直径、浸润深度和TNM分期均有关(P0.05),PEAK1表达与肿瘤浸润深度、淋巴结转移和TNM分期均有关(P0.05)。Log Rank检验结果显示,RCN2阳性表达组和PEAK1阳性表达组患者的术后5年总生存率均分别低于RCN2阴性表达组和PEAK1阴性表达组患者(P0.05)。结直肠癌组织RCN2和PEAK1表达呈正相关性(r=0.586,P=0.000)。结论:RCN2和PEAK1蛋白在结直肠癌组织中呈高表达,且均与肿瘤恶性进展和不良预后关系密切。RCN2和PEAK1可作为结直肠癌治疗靶标的候选分子。  相似文献

Summary The fine structure of the plastids in the leaf's basal meristem and in the leaf tissues 2 cm immediately above has been studied in maize leaves of different ages. In the young leaves the proplastids of the basal meristem differentiate, in the tissues within 2 cm above the meristem, into chloroplasts containing two or more prolamellar bodies, indipendently whether the tissues have been fixed 11 hours after a period of illumination or of darkness. In the oldest leaves, in the tissues immediately above the basal meristem, no prolamellar body is present in the plastids, and the proplastids differentiate directly into chloroplasts, without passing through an etio-chloroplast stage.Supported by a grant of C.N.R.  相似文献

Summary Transformation of plant tissues into crown gall tumors has been associated with the transfer of a portion of a tumor-inducing plasmid (Ti-plasmid) into plant DNA. Various laboratories have regenerated normal-appearing plants from a number of crown gall tumors. This study investigates the fate of the foreign DNA in a series of tissues derived from various parts of a plant regenerated from the tumor BT-37 by Braun and his coworkers. It was found that all the foreign DNA sequences were lost from tissues that had lost all their tumorous traits; whereas the plasmid DNA sequences were still present in tissues that appeared normal but still exhibited tumorous traits when returned to tissue culture media. From these studies it would appear that the presence of the Ti-plasmid sequences in the plant DNA is required for the maintenance of the transformed state. Osteoarthritis (OA) affects all articular tissues and finally leads to joint failure. Although articular tissues have long been considered unresponsive to estrogens or their deficiency, there is now increasing evidence that estrogens influence the activity of joint tissues through complex molecular pathways that act at multiple levels. Indeed, we are only just beginning to understand the effects of estrogen deficiency on articular tissues during OA development and progression, as well as on the association between OA and osteoporosis. Estrogen replacement therapy and current selective estrogen receptor modulators have mixed effectiveness in preserving and/or restoring joint tissue in OA. Thus, a better understanding of how estrogen acts on joints and other tissues in OA will aid the development of specific and safe estrogen ligands as novel therapeutic agents targeting the OA joint as a whole organ.  相似文献

Protein disulphide-isomerase activity was determined in homogenates of chick-embryo tissues. Activities were highest in tissues active in procollagen synthesis and were maximal at the developmental stage of maximal procollagen synthesis. These variations in protein disulphide-isomerase activity correlate closely with those previously observed for protocollagen hydroxylase activities.  相似文献

目的:分析肝细胞癌组织中FHIT基因启动子甲基化状态及其与FHIT基因表达和肝细胞癌临床病理特征的关系。方法:运用甲基化特异性PCR(MSP)方法分析肝细胞癌组织和癌旁正常肝脏组织中FHIT基因启动子甲基化状况;应用RT-PCT和Western免疫印迹方法检测FHIT基因mRNA和蛋白的表达情况;统计学分析FHIT基因启动子甲基化与肝细胞癌临床病理特征的关系。结果:MSP分析结果表明肝细胞癌组织中FHIT基因甲基化率(60.8%)显著高于癌旁正常组织中FHIT基因甲基化(16.2%;x2=31.071,P=0.000)。同时我们还发现:发生完全或者部分甲基化的肝细胞癌组织或者癌旁正常肝组织中FHIT基因mRNA和蛋白表达水平显著降低。FHIT基因启动子甲基化和肝细胞癌患者的临床分期和肝外转移情况密切相关(P=0.006和0.049),而与其他临床病理特征无相关性(P>0.05)。结论:FHIT基因甲基化是导致FHIT基因在肝细胞癌中失活的一个重要因素,与肝细胞癌的发生密切相关,有望成为肝细胞癌早期诊断的分子检测标志物和分子治疗新靶点。  相似文献

Psammoma bodies and small calcifications are frequently seen in a wide variety of tissues. These deposits of calcium salts render tissues difficult to section. Conventional decalcification alters the tissues; consequently it is not advisable to decalcify tissues containing only small calcium deposits. A simple and rapid method to remove small amounts of calcium salts using citric acid alone is described. This method does not alter the antigenic properties of the substances studied.  相似文献

Total lipid and lipid class composition of selected tissues and serum were determined for juvenile steelhead trout Salmo gairdneri , during the smolting period between 25 January and 19 April 1982. The total lipid content of the serum, liver, light muscle and dark muscle was significantly depleted during smolt transformation. Mesenteric fat total lipid concentration fluctuated little over the sampling period. Phospholipids of muscle and mesenteric fat exhibited little seasonality. Cholesterol was significantly depleted from dark muscle and liver. Large stores of triacylglycerol (TGL) in the parrs' mesenteric fat, dark muscle and liver implicate these tissues as lipid depot organs. In those tissues depleted of lipid, TGL concentrations were reduced more than any other lipid class. This supports the hypothesis that increased catabolism is the cause of loss of body fat during salmonid smolt transformation.  相似文献

The content of adrenaline and noradrenaline in the tissues of the heart, adrenal glands, spleen and brain of rats was studied in experimental myocardial infarction. A significant decrease in the catecholamine levels was revealed in the tissues. Malaben promoted normalization of the catecholamine tissue content in myocardial infarction. It is suggested that the said effect of malaben is due to its antihistaminic properties.  相似文献

In this study, concentrations of some major and minor elements were determined in the larynx tissues with and without cancer, and results obtained were statistically compared. No meaningful differences were found between sodium, potassium, calcium and copper concentrations in cancer tissues, corresponding cancer-free adjacent tissues and in control larynx tissues. Phosphate concentrations of the cancer tissues were higher compared with cancer-free adjacent tissues and control tissues. Iron, zinc and magnesium concentrations were found increased in both cancer and corresponding cancer-free adjacent tissues relative to control values. Intra- and inter-element correlations established within and between groups indicated that relations between elements were also disordered in the cancer tissues. We suggest that the changed element status of cancerous larynx tissues may arise from increased requirements of cancer tissues for some elements such as iron, zinc, magnesium and phosphate.  相似文献

HPV-16、EB病毒对细胞增殖的影响及与子宫颈癌发病的关系   总被引:1,自引:0,他引:1  
目的 了解宫颈癌患者人乳头瘤病毒16(HPV 16)、EB病毒(EBV)感染情况,探讨HPV-16、EBV对细胞核增殖性抗原(PCNA)表达的影响及在子宫颈癌发病中的意义。方法 免疫组织化学SP法检测59例宫颈癌和20例非癌性子宫颈上皮细胞中HPV-16、EBV蛋白表达和PCNA表达的情况,并分析它们的表达与病理参数的关系。结果 子宫颈癌和非癌性子宫颈上皮细胞中HPV-16、EBV及PCNA的阳性表达率依次分别为69.49%、57.63%及77.97%和30%、25%及15%;子宫颈癌和非癌性子宫颈上皮细胞中HPV-16、EBV的共同阳性表达率分别为35.59%和0%,子宫颈癌中HPV-16、EBV及PCNA的阳性表达率均高于非癌性子宫颈上皮(P〈0.05)。病理Ⅰ级、Ⅱ级和Ⅲ级子宫颈癌中HPV-16、EBV、PCNA的阳性表达率分别为65.00%、55.00%、60.00%,69.57%、60.87%、82.61%和75.OO%、56.25%、93.75%。各级子宫颈癌间HPV-16、EBV的阳性表达率差异无显著性,但PCNA的表达率随病理分级的增加显著上升(P〈0.05)。不同期别子宫颈癌问HPV-16、EBV及PCNA的阳性表达率差异无显著性。HPV-16阳性与阴性组子宫颈上皮PCNA的阳性表达率分别为82.98%(39/47)与43.75%(14/32),两者差异有显著性(P〈0.05)。EBV阳性及阴性组子宫颈上皮PCNA的阳性表达率分别为71.79%(28/39)及42.50%(17/40),两者差异有显著性(P〈0.05)。HPV-16和EBV共同阳性表达的21例子宫颈癌PCNA阳性表达率均为100%。结论 HPV-16、EBV通过增加PCNA表达的促细胞增生作用可能是子宫颈癌的发生机制之一。  相似文献
