小尾寒羊高繁殖力候选基因ESR的研究   总被引:15,自引:0,他引:15

利用PCR—SSCP技术对高繁殖力绵羊品种(小尾寒羊、湖羊、德国肉用美利奴羊)和低繁殖力绵羊品种(多赛特羊、萨福克羊)的雌激素受体(estrogen receptor,ESR)基因第一外显子部分序列进行单核苷酸多态性研究。结果表明:小尾寒羊、湖羊和德国肉用美利奴羊中存在3种基因型(AA、BB、AB),而在多赛特羊和萨福克羊中只存在两种基因型(AA、AB)。统计结果表明:湖羊、德国肉用美利奴羊、小尾寒羊、萨福克羊和多赛特羊A等位基因频率分别为0.672、0.786、0.846、0.857和0.867,B等位基因频率分别为0.328、0.214、0.154、0.143和0.133。测序结果表明:BB型和AA型相比在外显子1第363位发生1处碱基突变(C→G)。独立性检验表明:小尾寒羊和湖羊之间基因型分布差异极显著(P〈0.01),湖羊和多赛特羊之间基因型分布差异显著(P〈0.05),其他各个绵羊品种之间基因型分布差异均不显著。A8基因型和BB基因型小尾寒羊产羔数比AA基因型分别多0.51只(P〈0.05)和0.7只(P〈0.05)。研究结果表明:ESR基因可能是控制小尾寒羊多胎性能的一个主效基因或与之存在紧密的遗传连锁。  相似文献

以绵羊BMPR-IB基因为候选基因,应用PCR-RFLP方法通过分析湖羊、夏洛来、陶赛特、萨福克、罗米丽、中国美利奴羊、中国美利奴肉用多胎品系以及陶赛特×中国美利奴羊和萨福克×中国美利奴羊杂交后代共615只个体的FecB基因多态性,以及BMPR-IB基因多态性对产羔数、体尺和体重的影响.结果表明,BMPR-IB基因在不同品种(系)绵羊中共有3种基因型(BB、B+和++),但基因型频率分布在各品种(系)间差异极显著(P<0.01).在湖羊中仅有BB基因型;在中国美利奴肉用多胎品系中BB、B+和++基因型频率分别为51%、30%和19%;而其他品种(系)羊中则仅有++基因型.对中国美利奴羊肉用多胎品系研究,发现BB和B+基因型群体平均产羔数分别为2.8和2.3,显著高于++基因型群体(1.2,P<0.01).在90日龄时,BB和B+基因型群体的体重分别为18.6±3.70 kg和18.0±3.31 kg,显著高于++基因型群体(15.6±2.22kg,P<0.05);此外,90日龄时,BB和B+基因型群体比++基因型群体胸围、胸宽较大(P<0.05);但这些差异在120日龄时消失.另外,我们还发现不同地区群体的第一胎产羔数存在明显差别.这些结果表明,BMPR-IB基因为影响绵羊产羔数的主效基因,并首次证明该基因对后代羔羊出生后生长发育具有加性效应.  相似文献

以绵羊GDF9基因FecG^H突变和BMP15基因FecX^B和FecX^G突变为候选基因,采用PCR—RFLP方法研究其在湖羊、夏洛来、陶赛特、萨福克、中国美利奴肉用多胎品系、中国美利奴羊和罗米丽羊7个品种中的多态性.结果发现,湖羊群体中存在GDF和BMP15基因的FecG^H和FecX^B突变,但发生率极低,分别为0.645%(2/310)和0.968%(3/310):而其它品种中则没有发现GDF9和BMP15基因的相应突变.这一发现对于建立绵羊基因标记辅助选择方法具有重要意义.  相似文献

为了探讨LEPR在绵羊季节性发情中的表达特点,以季节性发情的中国美利奴和常年发情的多浪羊为研究对象,利用实时荧光定量PCR技术检测繁殖季节不同发情阶段LEPRb m RNA在下丘脑、卵巢和子宫组织中的表达变化。结果显示,在发情间期、前期和发情期中国美利奴卵巢和子宫组织LEPRb的相对表达水平都低于下丘脑,但在发情后期卵巢和子宫LEPRb的相对表达水平显著高于下丘脑。不同发情时期,两品种绵羊同一组织的LEPRb的表达变化幅度有所差异,但趋势相似。且中国美利奴各组织LEPRb的表达水平都高于常年发情的多浪羊。LEPRb的表达变化与绵羊发情存在相关关系。  相似文献

以控制BooroolaMerino羊高繁殖力的BMPR-IB基因为候选基因,以小尾寒羊及其杂交羊、东北半细毛羊、澳洲美利奴羊、德国肉用美利奴羊、萨福克羊、特克塞尔羊、夏洛莱羊为试验对象,采用PCR-限制性片段长度多态性(PCR-RFLP)方法进行基因单核苷酸多态性(SNP)检测和基因型分析,同时研究基因对高繁殖力的影响.研究结果表明:小尾寒羊及其杂交羊、东北半细毛羊和夏洛莱羊群体中发现了与BooroolaMerino羊相同的A746G碱基突变,而小尾寒羊及其杂交羊群体的B等位基因频率明显高于其他2个品种.另外4个品种中未发现此突变.携带B等位基因的群体较非携带B等位基因群体排出更多的卵子,排卵后黄体直径较小.移植入冷冻胚胎后, 、B 和BB3种基因型群体的妊娠率分别为38.78%、45.71%和66.67%.由此推断,BMPR-IB基因突变很有可能从增加卵巢排卵数和提高胚胎着床及妊娠建立效率两个方面同时影响绵羊高繁殖力性状.所得BB型群体冻胚移植妊娠率明显高于 和B 型群体,已接近鲜胚移植水平,通过PCR-RFLP方法进行基因型分析,选用合适基因型群体作为胚胎移植受体,有可能为提高绵羊胚胎移植受胎率提供新的方向.  相似文献

中国7个地方绵羊品遗传多样性的微卫星分析   总被引:14,自引:1,他引:14  
利用26个微卫星标记分析了中国7个地方绵羊(Ovis aries)品种的遗传多样性。通过计算基因频率,平均杂合度(H)、多态信息含量(PIC)及有效等位基因数(Ne),并根据Nei氏标准遗传距离,利用UPGMA法进行了聚类分析,评估其种内遗传变异和品种间遗传关系。结果表明:26个微卫星位点共检测到278个等位基因,Ne在2.1288-13.3924之间;26个位点均为高度多态位点,PIC在0.6628-0.8712之间,聚类分析表明哈萨克羊、阿勒泰羊和巴音布鲁克羊遗传关系最近;然后与白藏羊,黑藏羊聚为一类,湖羊和晋中羊聚为一类,各绵羊各种的聚类关系与其来源,育成史及地理分布基本一致。  相似文献

以控制Belclare和Cambridge绵羊高繁殖力的骨形态发生蛋白 15 (bonemorphogeneticprotein 15 ,BMP15 )基因和生长分化因子 9(growthdifferentiationfactor 9,GDF9)基因为候选基因 ,采用PCR RFLP技术检测BMP15基因和GDF9基因在高繁殖力绵羊品种 (小尾寒羊、湖羊 )以及低繁殖力绵羊品种 (多赛特羊、特克塞尔羊、德国肉用美利奴羊 )中的单核苷酸多态性 ,同时研究这两个基因对小尾寒羊高繁殖力的影响。结果表明 :在 5个绵羊品种中都没有检测到GDF9基因的G8突变 (C→T) ,也没有检测到BMP15基因的B4突变 (G→T)。高繁殖力的小尾寒羊在BMP15基因编码序列第 718位碱基处发生了与Belclare绵羊和Cambridge绵羊相同的B2突变 (C→T) ,而其余 4个绵羊品种则没有发生这种突变。对于BMP15基因的B2突变 ,在小尾寒羊中检测到AA、AB两种基因型 ,A等位基因频率为 0 734,B等位基因频率为 0 2 6 6。小尾寒羊与其余 4个绵羊品种间B2突变基因型分布差异极显著 (P <0 0 0 1)。突变杂合基因型 (AB)小尾寒羊平均产羔数比野生纯合基因型 (AA)多 0 6 2只 (P <0 0 1)。研究结果表明 ,BMP15B2突变对小尾寒羊高繁殖力影响作用十分明显 ,同时排除了GDF9G8突变和BMP15B4突变影响小尾寒羊高繁殖力的可能性  相似文献

目的:对新疆4个绵羊品种内皮型一氧化氮合酶(eNOS)基因第8外显子的多态性进行鉴定。方法:利用PCR-SSCP和测序的方法对76只中国美利奴羊、51只无角陶赛特羊、57只萨福克羊、37只哈萨克羊共4个绵羊品种进行单核苷酸多态性(SNP)检测,并用生物信息学方法对检测出的SNP进行统计分析。结果:在中国美利奴羊、无角陶赛特羊、萨福克羊、哈萨克羊中共检测到AA、AB、BB等3种基因型,AA基因型的频率分别为0.0526、0.0980、0.1754和0.2973,BB基因型的频率分别为0.6316、0.1961、0.5614和0.1892,AB基因型的频率分别为0.3158、0.7059、0.2632和0.5135。通过测序,在eNOS基因第8外显子上发现了一个新的SNP位点(ss974768653),位于绵羊eNOS基因第8外显子142 bp处(A142G)。结论:中国美利奴羊和哈萨克羊的多态性位点(P0.05)处于Hardy-Weinberg平衡状态,萨福克和无角陶赛特羊的多态性位点(P0.05)不处于Hardy-Weinberg平衡状态。  相似文献

绵羊微卫星BMS2508和FecB基因的多态及连锁分析   总被引:2,自引:1,他引:2  
文章分析与绵羊高繁殖力主效基因FecB紧密连锁的微卫星座位BMS2508在高繁殖力绵羊品种(小尾寒羊)和低繁殖力绵羊品种(特克塞尔、多赛特和中国美利奴)中的遗传多态性, 同时探讨该微卫星座位与小尾寒羊FecB基因的连锁不平衡关系。高繁殖力品种小尾寒羊在骨形态发生蛋白受体IB(Bone morphogenetic protein receptor IB, BMPR-IB)基因编码序列第746位碱基处发生了与Booroola Merino绵羊相同的FecB突变(A746G), 而在低繁殖力的特克塞尔、多赛特和中国美利奴绵羊中没有检测到该突变; 小尾寒羊BB、B+、++的基因型频率分别为0.485、0.398和0.117。微卫星座位BMS2508在4个绵羊品种的438个个体中共检测到8个等位基因和15种基因型, 最小等位基因为94 bp, 最大等位基因为116 bp; 小尾寒羊(n = 307)、特克塞尔(n = 45)、多赛特(n = 46)、中国美利奴(n = 40)和BB型(n = 149)、B+型(n = 122)、++型(n = 36)小尾寒羊群体中优势等位基因分别是100 bp、94 bp、94 bp、112 bp、100 bp、100 bp、112 bp, 其频率分别为0.453、0.544、0.802、0.475、0.483、0.439、0.389。连锁不平衡分析显示小尾寒羊FecB基因B等位基因与BMS2508微卫星座位100 bp等位基因之间存在一定的连锁不平衡(D′=0.408), 而+等位基因与BMS2508微卫星座位110 bp和114b p等位基因均存在一定的连锁不平衡(D′=0.513)。  相似文献

研究选择了与产肉性能相关的5个微卫星标记,分析了这5个位点在杜泊、德国美利奴、特克塞尔、道塞特和右玉本地绵羊5个品种中的遗传多态性.通过遗传分析预测了4个国外引进肉用绵羊品种与右玉本地绵羊的杂种优势,并与实际测定结果进行了比较分析.结果表明,利用微卫星DNA多态性进行品种间杂种优势预测是可行的,杜泊羊与右玉本地绵羊杂交的杂种优势最大,与实际测定结果一致.  相似文献

The PrP genotype of sheep of the improved Valachian breed   总被引:4,自引:0,他引:4  
In a worldwide majority of sheep breeds an excessive susceptibility to scrapie associated with the PrP gene alleles coding for valine (V; at the 136 codon) and glutamine (Q; at the 171 codon) (e.g., VRQ/VRQ, VRQ/ARQ, or ARQ/ARQ) was demonstrated. Particularly the PrPVRQ allele is closely associated with the high-risk development of the disease; the PrPARQ allele can also fulfill this function but under certain limited conditions. Polymorphism in the PrP gene sequences (conclusively related to the increased susceptibility of sheep to scrapie) of improved Valachian sheep from two Slovak regions, Orava and Spis, was determined. Examination of 735 sheep showed that ARR/ARQ was the most frequent genotype (45.2%). High-risk genotypes were determined in 32.4% of sheep (ARQ/ARQ 19.3, ARR/VRQ 9.0, ARR/VRQ 3.5, VRQ/VRQ 0.3, ARR/VRR 0.3). Low-risk genotypes were found in 67.7% of sheep (ARR/ARQ 45.2, ARR/ARR 10.9, ARR/AHQ 5.7, ARQ/ARQ 4.9, AHQ/AHQ 0.7, ARR/AHR 0.3). Despite the geographically distant flocks of improved Valachian sheep investigated no difference in the occurrence of individual PrP genotypes was observed.  相似文献

To determine the polymorphisms of the prion protein gene in sheep from the state of Paraná, Brazil, 323 animals of meat breeds (Suffolk, Hampshire Down, Texel, Ile de France, Dorper, Dorset, Santa Inês and crossbreds) were genotyped by restriction fragment length polymorphism (RFLP) analysis. The most frequent allele was ARQ, with a frequency of 0.61, followed by ARR (0.30). VRQ and AHQ alleles were present at very low frequencies (0.13 and 0.05 respectively), and the ARH allele was not found. Seven genotypes were identified (ARR/ARR, ARR/ARQ, ARQ/ARQ, ARR/VRQ, ARR/AHQ, ARQ/VRQ and ARQ/AHQ), of which ARQ/ARQ was the most frequent (0.41). The Santa Inês breed and crossbred animals showed the highest genotypic variability.  相似文献

The status of scrapie in Africa is largely unknown. The susceptibility to scrapie and its pathology is determined by amino acid polymorphisms at positions 136 (A/V), 154 (R/H) and 171 (Q/R/H) of the ovine PrP gene (PrP genotype) of the animals. Despite the widely studied PrP gene polymorphisms worldwide, limited data is available on PrP genotypes of sheep from the African continent. Previously, we have reported six PrP genotypes derived from four different alleles in Red Maasai and Black Herd Persian (BHP), the ARQ/ARQ genotype being more frequent in the Red Maasai than in the BHP sheep. The highly susceptible VRQ allele was not found in any of the sheep breeds; the ARR allele was absent in the Red Maasai or occurred at a low frequency in the BHP. The lack of the highly susceptible VRQ alleles among Tanzanian sheep examined, necessitated further examinations on genetic susceptibility in sheep of the same breeds, but originating from an entirely different part of Tanzania. Consistently, ARQ/ARQ genotype was observed in 88% of Red Maasai and 64% of BHP sheep, ARQ/AHQ genotype was present in 12% in Red Maasai and 36% in BHP. Neither the highly resistant (ARR/ARR), nor the highly susceptible (VRQ/VRQ) genotypes were found. The ARQ and AHQ were the only alleles observed. The ARQ allele constituted 94%, 82% and 67% in the Red Maasai, BHP and cross-bred sheep, respectively. The AHQ constituted 6%, 18% and 33%, respectively. Data reported here, provide additional information on genetic susceptibility of the Red Maasai, BHP and their crosses; they may be helpful in policy formulation for future prevention of scrapie.  相似文献

Scrapie, an invariably fatal disease of sheep and goats, is a transmissible spongiform encephalopathy (TSE). The putative infectious agent is the host-encoded prion protein, PrP. The development of scrapie is closely linked to polymorphisms in the host PrP gene. The pathogenesis of most TSEs involves conversion of normal, cellular PrP into a protease-resistant, pathogenic isoform called PrPSc. The conversion to PrPSc involves change in secondary structure; it is impacts on these structural changes that may link polymorphisms to disease. Within the structured C-terminal part of PrP polymorphisms have been reported at 15 and 10 codons of the sheep and goat PrP genes respectively. Three polymorphisms in sheep are acutely linked to the occurrence of scrapie: A136V, R154H and Q171R/H. These generate five commonly observed alleles: ARQ, ARR, AHQ, ARH and VRQ. ARR and AHQ are associated with resistance; ARQ, ARH and VRQ are associated with susceptibility. There are subtle effects of specific allele pairings (genotypes). Generally, more susceptible genotypes have younger ages at death from scrapie. Different strains of scrapie occur which may attack genotypes differently. Different sheep breeds vary in the assortment of the five alleles that they predominantly encode. The reason for this variation is not known. Furthermore, certain genotypes may be susceptible to scrapie in some breeds and resistant in others. The explanation is not known, but may relate to different scrapie strains circulating in different breeds, or there may be effects of other genes which modulate the effect of PrP.  相似文献

Of 34 breeds kept in the Czech Republic 45,604 sheep were genotyped for codons 136, 154 and 171 in the prion protein gene (PRNP) during the years 2006–2014. In this cohort, haplotypes ARR, ARQ, ARH, AHQ, VRQ, AHR and ARK were detected. The haplotype AF141RQ associated with susceptibility to atypical scrapie was observed in nine out of 30 breeds analysed for this purpose. In addition, six rare nonsynonymous substitutions producing haplotypes AT137RQ, AN138RQ, AG151RQ, AH151RQ, ARL168Q and ARQE175 were identified in various breeds. Due to their low frequencies, these polymorphisms are of no potential importance for the breeding programme. With regard to their genetic particularity, Sumavka, Valachian and Cameroon breeds were screened for additional polymorphisms. Further haplotypes, AR143RQ and AS146RQ, were found in Sumavka and Cameroon, and in Valachian sheep, respectively. Frequencies of the ARR (resistance-associated), VRQ (susceptibility-associated) haplotypes, and of the most resistant ARR/ARR genotype calculated for sheep born in the years 2001–2003 and 2011–2013 documented effects of the 10 year-lasting national breeding programme. The total frequency of ARR doubled from 36.8 to 75.8 %, while the frequency of VRQ decreased from 4 to 0.7 %. The total frequency of the ARR/ARR genotype increased from 17.7 to 59 %. These data show that the national scrapie resistance breeding programme has had an important desirable effect on haplotype and genotype frequencies of PRNP in Czech sheep.  相似文献

The UK national scrapie plan (NSP) for sheep is based on selection for the resistant ARR/ARR genotype and elimination of susceptible types of the ovine prion protein (PrP) gene. The aim of this study was to estimate the possible association of the PrP genotype and performance traits by using data from the CAMDA Welsh Mountain flock. Four alleles (ARH, ARQ, ARR and VRQ) and 10 genotypes covering all five NSP risk groups were present in the CAMDA flock. Overall, the most common allele was ARR (35.2%), and VRQ was the least common (5.4%). The commonest genotypes were ARR/ARQ (23.7%) and ARR/AHQ (23.1%). The most resistant genotype, ARR/ARR, and the most susceptible genotype, VRQ/VRQ, were found in 10.2% and 0.3%, respectively, of the population tested. The associations of PrP genotypes with weight and ultrasonically scanned traits were investigated in three analyses, the first using genotypes, the second using risk categories and the third using number of alleles. These associations were evaluated by univariate analysis of each trait using an animal model with maternal effects where appropriate, and PrP was included as a fixed effect. Selection for scrapie resistance will not adversely affect progress in the traits considered and is consistent with improvements in muscle depth.  相似文献

Sheep scrapie is a transmissible spongiform encephalopathy that can be transmitted horizontally. The prion protein gene (PRNP) profoundly influences the susceptibility of sheep to the scrapie agent and the tissue levels and distribution of PrPSc in affected sheep. The purpose of this study was to compare the survival time and PrPSc tissue distribution in sheep with highly resistant and highly susceptible PRNP genotypes after intracranial inoculation of the agent of scrapie. Five sheep each of genotype VRQ/VRQ, VRQ/ARR or ARQ/ARR were inoculated. Sheep were euthanized when clinical signs of scrapie became severe. Clinical signs, microscopic lesions, and western blot profiles were uniform across genotypes and consistent with manifestations of classical scrapie. Mean survival time differences were associated with the 171 polymorphic site with VRQ/VRQ sheep surviving 18 months, whereas VRQ/ARR and ARQ/ARR sheep survived 60 and 56 months, respectively. Labeling of PrPSc by immunohistochemistry revealed similar accumulations in central nervous system tissues regardless of host genotype. Immunoreactivity for PrPSc in lymphoid tissue was consistently abundant in VRQ/VRQ, present but confined to tonsil or retropharyngeal lymph node in 4/5 VRQ/ARR, and totally absent in ARQ/ARR sheep. The results of this study demonstrate the susceptibility of sheep with the ARQ/ARR genotype to scrapie by the intracranial inoculation route with PrPSc accumulation in CNS tissues, but prolonged incubation times and lack of PrPSc in lymphoid tissue.  相似文献

Screening for polymorphism at the prion protein (PrP) locus (PRNP) was carried out in 33 flocks of Local Awassi (LA) sheep in Israel, the Palestinian Authority (PA) and Jordan. In addition, PRNP genotyping was carried out in two flocks of Improved Awassi (IA) in Israel and the PA and in 11 flocks of Assaf sheep in Israel. Most of the rams present in the flocks at the time of the survey 328, 44 and 298 rams from the LA, IA and Assaf breeds, respectively, were genotyped. The ARQ allele was found to be predominant in all three breeds with average frequencies of 0.74, 0.78 and 0.86 for the LA, IA and Assaf breeds, respectively. Frequencies of the desirable ARR allele in these breeds were 0.10, 0.05 and 0.06, respectively. ARH, AHQ and ARK alleles were identified at low/absent frequencies. Only one Assaf ewe carried the undesirable V allele at codon 136. A few scrapie outbreaks have been documented in the past in the region. To increase ARR allele frequencies in the Awassi and Assaf populations, distribution of homozygous ARR/ARR rams is recommended.  相似文献

Genetic susceptibility to scrapie, a fatal disease of sheep and goats, is modulated by polymorphisms in the prion protein (PrP). Neither the frequency of the PrP genotypes nor their association with animal performance has been investigated in a large multibreed Irish sheep population. Scrapie genotypes were available on 16 416 animals; the breeds represented included purebred Belclare (733), Charollais (333), Suffolk (739), Texel (1 857), Vendeen (191), and crossbreds (12 563). Performance data on lambing, lamb and ewe performance as well as health traits were available. The association between alternative approaches of describing the PrP genotype (i.e. 15 individually called PrP genotypes, five genotype classes representing susceptibility to scrapie, or number of ARR haplotypes) and animal performance were quantified using animal linear mixed models. All 15 of the possible scrapie genotypes were detected, although the frequency differed by breed. The frequency of the five PrP haplotypes in the entire population were 0.70 (ARR), 0.15 (ARQ), 0.11 (ARH), 0.02 (AHQ) and 0.01 (VRQ); the most susceptible haplotype (VRQ) was only detected in purebred Texels and crossbreds. No association was detected between the PrP genotype of either the animal or dam and any of the lambing traits (i.e. lambing difficulty score, perinatal mortality and birth weight). With the exception of ultrasound muscle depth, no association between the PrP genotype and any of the lamb performance traits (i.e. lamb BW and carcass) was observed. Lambs carrying the category four PrP genotype (i.e. ARR/VRQ) had 1.20 (SE = 0.45) mm, 1.38 (SE = 0.12) mm, 1.47 (S = 0.25) mm shallower ultrasound muscle depth relative to lambs of the less susceptible scrapie categories of 1, 2, 3, respectively (P < 0.05). Nonetheless, no association between PrP genotype and lamb carcass conformation, the ultimate end goal of producers, was detected. Ewe litter size, body condition score or lameness did not differ by PrP genotype of the ewe (P > 0.05). For ewe mature BW, ARH/VRQ ewes differed from most other ewe PrP genotypes and were, on average, 3.79 (SE = 1.66) kg heavier than ARR/ARR genotype ewes. Lamb dag score differed by dam PrP genotype (P < 0.05), although the differences were small. Results from this study show that scrapie is segregating within the Irish sheep population, but the PrP genotype was not associated with most traits investigated and, where associations were detected, the biological significance was minimal. Preclinical sheep with the highly scrapie-susceptible VRQ/VRQ PRNP genotype secrete prions from the oral cavity. In order to further understand the significance of orally available prions, buccal swabs were taken from sheep with a range of PRNP genotypes and analyzed by serial protein misfolding cyclic amplification (sPMCA). Prions were detected in buccal swabs from scrapie-exposed sheep of genotypes linked to high (VRQ/VRQ and ARQ/VRQ) and low (ARR/VRQ and AHQ/VRQ) lymphoreticular system involvement in scrapie pathogenesis. For both groups, the level of prion detection was significantly higher than that for scrapie-resistant ARR/ARR sheep which were kept in the same farm environment and acted as sentinel controls for prions derived from the environment which might contaminate the oral cavity. In addition, sheep with no exposure to the scrapie agent did not contain any measurable prions within the oral cavity. Furthermore, prions were detected in sheep over a wide age range representing various stages of preclinical disease. These data demonstrate that orally available scrapie prions may be a common feature in sheep incubating scrapie, regardless of the PRNP genotype and any associated high-level accumulation of PrP(Sc) within lymphoreticular tissues. PrP(Sc) was present in buccal swabs from a large proportion of sheep with PRNP genotypes associated with relatively low disease penetrance, indicating that subclinical scrapie infection is likely to be a common occurrence. The significance of positive sPMCA reactions was confirmed by the transmission of infectivity in buccal swab extracts to Tg338 mice, illustrating the likely importance of orally available prions in the horizontal transmission of scrapie.  相似文献
