Synthesis of germ tube wall is a major quantitative event during germination penetration of fungi on host plants, but little is known of germ tube composition or metabolic regulation. Sonic oscillation was used to separate germ tubes from germinating uredospores of Uromyces phaseoli var. typica. Uniformly 14C-labeled wall fractions from both structures were prepared by repeated low speed centrifugation and extraction with polar and nonpolar solvents. Based on amino acid analysis, approximately 6 and 16% of the carbon from uredospore and germ tube walls, respectively, was present in amino acids readily accessible to protease. Covalent linkages between amino acid and carbohydrate of walls was indicated by analysis of fragments prepared by mild hydrolytic procedures and separated by column chromatography and paper electrophoresis. The existence of protein in wall structures may resolve some previous uncertainty about the occurrence of protein biosynthesis during germination of rust fungi. Glucose, mannose, and glucosamine were the only carbohydrate components identified in both germ tubes and uredospore walls but different percentages were observed (germ tubes 28: 16: 16; uredospore 6: 36: 6). In germ tubes, most of the glucosamine was present in linkages hydrolyzed only by strong acid treatment, suggesting chitin-like polymers. In uredospore walls, glucosamine appears to be associated with red uredospore pigment which has properties similar to those of a melanin. Approximately 20% of the carbon in walls could not be identified with known compounds, partially because of degradation during the analytical procedures.  相似文献

The germ tubes ofColletotrichum lagenarium showed negative phototropism to UV-blue (300–520 nm) and far-red (>700 nm) regions with maximum in the near ultraviolet (NUV) region, while monochromatic radiations of 575–700 nm (yellow-red region) induced positive phototropism with maximum in the red region. Green light (520–575 nm) was ineffective. Negative phototropism-inducing wavelength regions inhibited germ tube growth and positive phototropism-inducing wavelength regions promoted it significantly.Bipolaris oryzae did not show any phototropic response and light did not affect the germ tube growth. These results indicate that the lens effect, in combination with the light growth reaction and light growth inhibition, is the mechanism of the phototropism of germ tubes ofC. lagenarium. NUV radiation, which induced negative phototropism ofC. lagenarium, promoted appressorium formation, while red light, which induced positive phototropism, suppressed it significantly. In the case ofB. oryzae, light did not affect the infection structure formation. These results indicate that negative phototropism of germ tubes ofC. lagenarium favors the infection process by facilitating the contact of the tips of germ tubes with the host surface, while positive phototropism has the opposite effect.  相似文献

This study compared different temperatures and dormancy‐reversion procedures for preservation of Phakopsora pachyrhizi uredospores. The storage temperatures tested were room temperature, 5°C, ?20°C and ?80°C. Dehydrated and non‐dehydrated uredospores were used, and evaluations for germination (%) and infectivity (no. of lesions/cm2) were made with fresh harvested spores and after 15, 29, 76, 154 and 231 days of storage. The dormancy‐reversion procedures evaluated were thermal shock (40°C/5 min) followed or not by hydration (moist chamber/24 h). Uredospores stored at room temperature were viable only up to a month of storage, regardless of their hydration condition. Survival of uredospores increased with storage at lower temperatures. Dehydration of uredospores prior to storage increased their viability, mainly for uredospores stored at 5°C, ?20°C and ?80°C. At 5°C and ?20°C, dehydrated uredospores showed increases in viability of at least 47 and 127 days, respectively, compared to non‐dehydrated spores. Uredospore germination and infectivity after storage for 231 days (7.7 months), could only be observed at ?80°C, for both hydration conditions. At this storage temperature, dehydrated and non‐dehydrated uredospores exhibited 56 and 28% of germination at the end of the experiment, respectively. Storage at ?80°C also maintained uredospore infectivity, based upon levels of infection frequency, for both hydration conditions. Among the dormancy‐reversion treatments applied to spores stored at ?80°C, those involving hydration allowed recoveries of 85 to 92% of the initial germination.  相似文献

Summary A comparison was made of fixed and non-fixed uredospores of the wheat stem rust fungusPuccinia graminis f. sp.tritici, using thin sectioning and freeze-etching. The latter technique yielded new information on the ultrastructure of dormant, germinating, and photo-inhibited uredospores, although the distribution of intramembranous particles (IMPs) was similar in each case. A stereological analysis of thin sectioned specimens provided quantitative data on organelle volumetric densities complementing the qualitative information. Evidence suggests that light acts by preventing the dissolution of wall material in the germ pore regions. Methylcis-ferulate, the uredospore self-inhibitor, is also believed to act at this stage in the germination process, and possible links between photo-inhibition and self-inhibition are discussed.  相似文献

The development of infection structures by the directly infecting soybean rust fungus of different artificial membranes was followed by light and scanning electron microscopy. On water agar uredospores developed germ tubes without appressoria. On dialysis membranes more than 80% of the uredospores formed appressoria. With low frequencies (1–7%) also primary hyphae and/or penetration hyphae were present. When cellulose nitrate membrane filters with pore diameters ≤ 0.2 μm were used, uredospores germinated but showed a strongly reduced appressoria formation. Membranes with pores ≥ 0.1 μm allowed a development of infection structures similar to that on dialysis membranes. In experiments with paraffin oil incorporated into collodion membranes more than 90% of the uredospores formed appressoria, about 50% of the appressoria developed hyphae. Ungerminated spores and germ tubes always contained 2 nuclei. In fully developed appressoria 4 nuclei were present. Compared with stomata entering rust fungi appressoria formation by Phakopsora pachyrhizi occurred more frequently and seemed to be less dependent on specific stimuli. Moreover, in most cases only few of the appressoria formed penetration or primary hyphae. The induction of these structures seemed to be dependent on further unknown stimuli.  相似文献

Temperature markedly affected germination and germ tube length of A. niger. More than 90% of the spores were germinated in the range 30°–34 °C and formed maximum length of germ tubes. At temperatures from 38° to 43 °C, the proportion of the spores that germinated as well as the germ tube length were both gradually decreased. However, at 47°C germ tube formation was completely inhibited up to 15 hrs. after inoculation.High relative humidity was found necessary for the spore germination of A. niger. Germination failed to occur at 76% relative humidity. At 78 and 81% relative humidity germination was detected 15 hrs. after inoculation while at the higher humidities germination was started after 6 hrs. only.Conidiospores of A. niger were very sensitive to changes in the hydrogen ion concentration, pH. Complete inhibition of germination was found at pH less than 3.5. The germination and the length of the formed germ tubes increased with pH to reach their maximum rates at pH 4.5.  相似文献

Polyphosphate (polyP) is the form in which phosphorus (P) is transferred from extraradical hyphae into arbuscles in the symbiotic stage of arbuscular mycorrhizal fungi. However, polyP dynamics in the presymbiotic stage are less understood. In this study, we aimed to investigate polyP accumulation in Gigaspora margarita as influenced by nitrogen (N) and/or P supply during germination. Spores of G. margarita were incubated on medium with or without P or N addition. PolyP content in the fungal tissue was monitored using a polyP kinase/luciferase system, and polyP synthetic activity was determined with 32P labeling. The results showed that both N and P were necessary for polyP accumulation in germ tubes. Nitrate increased the polyP content in germ tubes, but ammonium did not. Along with germination, polyP content decreased in spores, but increased in germ tubes. 32P labeling indicated that polyP synthetic activity increased in germ tubes along with germination, but was negligible in spores. Our results suggest that, in the presymbiotic stage of G. margarita, uptake of environmental N and P increases polyP content in germ tubes, and that polyP synthesis occurs mainly therein, leading to polyP accumulation. The possible mechanism of transfer of polyP from spores to hyphae remains to be elucidated.  相似文献

Digital video microscopy in conjunction with the DNA-binding fluorescent probe 4′,6-diamidino-2-phenylindole was used to determine the relative DNA content of single nuclei in germ tubes of uredospores of the bean rust fungus (Uromyces phaseoli), a parasite of bean plants (Phaseolus vulgaris). The uredospores develop a series of infection structures in response to contact stimuli, and the first structure to appear, the appressorium, occupies the stomatal opening. This study was made to determine the time after the start of germination on an inductive surface that DNA replication and mitosis occur. It was found that the start of DNA replication detected by increased nuclear fluorescence power of some individual nuclei in the population was nearly coincident with mitosis between 2.0 and 2.5 h after the start of germination and that the appressorium was completed about 30 min later. The fluoresence power of nuclei was about the same before DNA replication began as at the end of mitosis; hence the germ tube nuclei were in the G1 phase of the cell cycle before mitosis.  相似文献

Summary The dry mass of two-celled Diplodia maydis spores was measured both before and after germination by quantitative interference microscopy. The dry mass of spores declined approximately 50% during germination. However, the dry mass of germinating spores plus the dry mass of their germ tubes was greater than the dry mass of spores before germination. We conclude that the germinating spores absorbed nutrients released from non-germinating spores.The dry mass of fungal spores can be estimated by weighing large numbers of spores and determining the mean from sample spore counts. Mumford and Pappelis(4) determined the total dry mass of individual spores of Fusarium roseum and the contained lipid bodies before and after spores germinated using quantitative interference microscopy. The mean spore dry mass before germination was 57 pg. Lipid bodies accounted for about 61% of that mass and decreased as spores germinated. The total dry mass of the spore and germ tube 24 hr later greatly exceeded that of the spore before germination. Quantitative interference microscopy has been used to measure the dry mass of various types of cells. Kulfinski and Pappelis (3) recently reviewed how this technique has been applied to plant cells. Technical aspects of interference microscopy have been described by Ross (6).The purpose of this study was to examine the dry mass changes in Diplodia maydis (Berk.) Sacc. with and without germ tubes through the use of interference microscopy.  相似文献

Newly regenerated thalli were used to study the phototropism of Bryopsis hypnoides Lamouroux under different qualities of light. Positive phototropism in the thalli and negative phototropiam In the rhizoida of B. hypnoides were investigated and analyzed in terms of bending. Both thaiii and rhlzoids developed from thallus segments exhibited typical tip growth, and their photoreceptive sites for phototroplam were also restricted to the apical hemisphere. The bending curvature of rhizoids and thalli were determined with unilateral lights at various wavelengths and different fluence rates after a fixed duration of Illumination. The trends of bending from the rhizoid and thallus were coincident, which showed that the action spectrum had a large range, from ultraviolet radiation (366.5 nm) to green light (524 nm). Based on the bending curvatures, blue light had the highest efficiency, while the efficiency of longer wavelengths (〉500 nm) was significantly lower. External Ca^2+ had no effect on the bending curvature of thalli and rhlzolda. Blue light (440 nm) induced thallus branching from rhizoids, while red light (650 nm) had no such effect. The amounts of two growth inhibitors in diffusates from illuminatedhalves of phototropically stimulated oat (Avena sativa L.)coleoptile tips were larger than those from shaded halves. The less polarinhibitor was isolated from diffusates from oat coleoptile tips illuminatedwithblue light, and identified as uridine from 1H NMR spectrum. Thedistribution of endogenous uridine in diffusates from the illuminated andshadedsides of coleoptile tips unilaterally exposed to blue light for 3, causing a first positive phototropic curvature, and fromdark-control tips, was determined using a physicochemical assay. The uridineconcentration was significantly higher in the diffusates from the illuminatedside than in those from the shaded side and the dark-control. Uridine inhibitedthe growth of etiolated oat coleoptile tips at concentrations of 30 and above. These results suggest that uridine plays a role inthe phototropism of oat coleoptiles.  相似文献

Summary Germ tube apices ofGilbertella persicaria contain cytoplasmic vesicles, similar to the secretory vesicles found at the tips of vegetative hyphae. The vesicles are present at all stages of development, from the time of germ tube initiation to the establishment of branched hyphae. In contrast to the abundant vesicles at tips of established hyphae, the germ tubes have only a few apical vesicles in a layer next to the plasma membrane. When germinated spores are treated by washing and centrifuging prior to fixation, the cytoplasm is often disrupted near the apex, and the clusters of apical vesicles disappear. The findings indicate the delicate nature of hyphal tips and the necessity of avoiding prefixation stresses in order to preserve the apical apparatus of growing hyphae.  相似文献

Blue light induces the phosphorylation of a 116 kDa oat protein found in plasma membrane preparations from coleoptile tips. We developed a very sensitive in vitro method that allowed us to determine the tissue distribution of protein phosphorylation after applying unilateral and bilateral blue light pulses in vivo. We found that following unilateral in vivo irradiation the degree in phosphorylation of the 116 kDa protein is significantly higher at the irradiated than at the shaded side of the coleoptile tip. This asymmetry can be considered as previously missing criterion that protein phosphorylation represents an early event within the transduction chain for phototropism.  相似文献

Botrytis cinerea is one of the most serious post-harvest pathogens of fruits and vegetables. Volatiles generated by Bacillus subtilis JA significantly inhibited both spore germination and elongation of germ tubes in Botrytis cinerea using a two-compartment agar-plate assay. On artificial polyethylene membranes providing a thigmotropic signal, uredospores of the broad bean rust fungus Uromyces viciae-fabae differentiated a series of infection structures which in nature are necessary to invade the host tissue through the stomata. Within 24 h germ tubes, appressoria, substomatal vesicles, infection hyphae and haustorial mother cells were developed successively. Alterations in protein metabolism during infection structure differentiation of this obligate plant pathogen were analyzed in the absence of the host plant by high resolution two-dimensional polyacrylamide gel electrophoresis (2-DE) and silver staining. The norm pattern representing the 2-DE protein patterns of the whole developmental sequence of infection structures of U. viciae-fabae showed 733 spots. During infection structure differentiation 55 proteins were newly formed, altered in quantity, or disappeared. Major alterations in the protein pattern occurred during uredospore germination and when infection hyphae were formed. A simple method for the isolation of single ascospores of the fission yeast Schizosaccharomyces pombe was examined. Single spores in the 7-day-old sporulating culture of a homothallic strain were separated from remaining vegetative cells by isopycnic centrifugation in the linear gradient from 10 to 60% of Urografin solution at 700 X g for 20 min. Protein content of isolated spores was very low as compared with that of vegetative cells. The isolated spores germinated through the following steps when cultured in a liquid medium at 25--35 degrees C; loss of refractility (darkening) under a phase-contrast microscope, spherical growth (swelling), emergence of germ tubes, elongation of germ tubes, cell plate formation, and cell separation. The absorbance at 650 nm of the spore suspension initially decreased, accompanied by darkening of spores, and then increased with spherical growth. The germination rate of isolated spores reached almost 100%.  相似文献

Young seedlings of Lavatera cretica L. exhibit positive phototropism. The hypocotyl perceives unilateral illumination with blue light and curves towards the light source by unequal growth. In addition, the cotyledonary laminas perceive the vectorial component of unilateral illumination with blue light and reorient normal to the beam by creating a turgor differential in their pulvini. Excision of one cotyledon resulted in negative organotropic curvature of the hypocotyl, away from the remaining cotyledon. Illumination of the cotyledonary lamina did not participate in the phototropic curvature of the hypocotyl, so long as the lamina was free to reorient to face the beam. When the lamina was continuously exposed to vectorial photoexcitation, elongation of the hypocotyl on the side carrying the cotyledon could be enhanced, or inhibited, depending on the direction of the beam striking its lamina.  相似文献

Aeciospores of the long-cycle heteroecious rust fungus, Cronartium fusiforme, were found to have an extremely thick cell wall with striking spicules protruding from it. The wall was readily degraded by commercial chitinase, but spicules were unaffected. Quiescent spores contained two nuclei with distinct nuclear membranes possessing many pores. Numerous membrane-bounded lipid bodies were found both in wild-type orange and in white mutant aeciospores. An abundance of irregularly ovoid mitochondria was present in quiescent spores. After glutaraldehydeosmium fixation, the surface of the mitochondria appeared to be covered with ribosomes or microtubules in a paracrystalline array, whereas after permanganate fixation only smooth outer mitochondrial membranes were noted. The latter fixative revealed abundant vesicular endoplasmic reticulum in the spore. Spores incubated at 20 C on agar produced one to five distinct germ tubes within 65 to 180 min. These thin-walled tubes exhibited varying degrees of branching, and reached a total hyphal length of 300 to 500 mu prior to rupturing. Emergence of germ tubes took place through a pore in the spore wall and appeared to be mainly a physical flowing of cytoplasm from the spore into the germ tube without division of nuclei or other cell organelles. On completion of germination, the protoplasm of the germ tube contained both nuclei and nearly all of the other spore contents. Mitochondria had smooth outer membranes, were greatly elongated, and possessed distinct longitudinal cristae. A limited amount of rough endoplasmic reticulum was arranged parallel to the germ tube wall. Other organelles seen in germ tubes were lipid bodies, concentric membrane figures, and numerous ribosomes. Lipid bodies appeared smaller and fewer in number than in quiescent spores.  相似文献

Plant seed lectins are well-characterized proteins and glycoproteins whose natural function remains unknown. We found that eleven purified seed lectins (representing five groups of lectin sugar specificities) bound to the germ tubes of asexual spores of Neurospora crassa, Aspergillus amstelodami , and Botryodiplodia theobromae. The lectins caused several types of quantifiable growth disruption during germination of these seed- or soil-borne fungal spores, including sensitivity to osmotic lysis, adventitious branching of the spore germ tubes, and inhibition of germ tube elongation. These anti-fungal effects of purified lectins, which were reversible with the sugar hapten specific for each lectin, were partially duplicated by lectin-like factors in the homologous crude seed extracts. The seed lectins may disrupt fungal growth by interfering with normal cell wall deposition and assembly.  相似文献

We have studied the effects of two polyamine biosynthetic inhibitors,-difluoromethylor-nithine (DFMO) and -difluoromethylarginine(DFMA), and of polyamines (PAs), alone and in combination onuredospore germination and germ tube growth in Uromyces phaseoliL, race O. Both the inhibitors at concentrations 0.01, 0.1 and1.0 mM produce successively inhibition of uredospore germinationin vitro. The inhibitors also delay the timing of spore germinationfor 15–30 min and restrict germ tube elongation. Stimulationof spore germination and germ tube growth was noticed in culturescontaining PAs (putrescine or spermidine) alone, while culturesfortified with inhibitor plus PA resulted in a partial reversionof the inhibitory effect, suggesting that PAs may be requiredfor normal germination and outgrowth of fungal spores. Sporegermination was completely inhibited on the surface of unifoliolatebean leaves treated with 0.5 mM or higher DFMO 1 d before inoculationwith pathogen, while DFMO treated 1 d after inoculation showedgreater damage of uredosporelings. In contrast, DFMA confersno effect even at 5 mM. Spores collected from bean plants givena pre- and post-inoculatory treatments with DFMO and DFMA showno significant differences in germination and pathogenicity,however, the higher doses caused significant decrease.
