Plasminogen activator inhibitor 1 (PAI-1), a major modulator of the fibrinolytic system, is an important factor in cardiovascular disease (CVD) susceptibility and severity. PAI-1 is highly heritable, but the few genes associated with it explain only a small portion of its variation. Studies of PAI-1 typically employ linear regression to estimate the effects of genetic variants on PAI-1 levels, but PAI-1 is not normally distributed, even after transformation. Therefore, alternative statistical methods may provide greater power to identify important genetic variants. Additionally, most genetic studies of PAI-1 have been performed on populations of European descent, limiting the generalizability of their results. We analyzed >30,000 variants for association with PAI-1 in a Ghanaian population, using median regression, a non-parametric alternative to linear regression. Three variants associated with median PAI-1, the most significant of which was in the gene arylsulfatase B (ARSB) (p = 1.09 x 10−7). We also analyzed the upper quartile of PAI-1, the most clinically relevant part of the distribution, and found 19 SNPs significantly associated in this quartile. Of note an association was found in period circadian clock 3 (PER3). Our results reveal novel associations with median and elevated PAI-1 in an understudied population. The lack of overlap between the two analyses indicates that the genetic effects on PAI-1 are not uniform across its distribution. They also provide evidence of the generalizability of the circadian pathway’s effect on PAI-1, as a recent meta-analysis performed in Caucasian populations identified another circadian clock gene (ARNTL). 相似文献

Background
Chronic inflammation is involved in the pathogenesis of chronic age-associated, degenerative diseases. Pro-inflammatory host responses that are deleterious later in life may originate from evolutionary selection for genetic variation mediating resistance to infectious diseases under adverse environmental conditions.Methodology/Principal Findings
In the Upper-East region of Ghana where infection has remained the leading cause of death, we studied the effect on survival of genetic variations at the IL10 gene locus that have been associated with chronic diseases. Here we show that an IL10 haplotype that associated with a pro-inflammatory innate immune response, characterised by low IL-10 (p = 0.028) and high TNF-α levels (p = 1.39×10−3), was enriched among Ghanaian elders (p = 2.46×10−6). Furthermore, in an environment where the source of drinking water (wells/rivers vs. boreholes) influences mortality risks (HR 1.28, 95% CI [1.09–1.50]), we observed that carriers of the pro-inflammatory haplotype have a survival advantage when drinking from wells/rivers but a disadvantage when drinking from boreholes (pinteraction = 0.013). Resequencing the IL10 gene region did not uncover any additional common variants in the pro-inflammatory haplotype to those SNPs that were initially genotyped.Conclusions/Significance
Altogether, these data lend strong arguments for the selection of pro-inflammatory host responses to overcome fatal infection and promote survival in adverse environments. 相似文献3.

This article focuses on Ghanaian popular cinema,' which emerged in the late 1980s and has become a major form of entertainment for urban audiences. It shows that the filmmakers, who produce films in the video format which are very close to everyday life experiences and dreams, offer visions which they and their audiences regard as being similar to those provided by religious prophets and preachers. Watching these films allows for complex negotiations between the desires for transgression of the moral order, the longing to feel morally superior, and the striving for knowledge to understand one's world. On the basis of a detailed analysis of the film Women in Love, which conjures up a fantasy space located at the bottom of the sea where money and commodities are generated in exchange for sex and blood, this article argues that popular cinema is part and parcel of a new culture of vision, one characterized by the visualization of otherwise invisible fantasy spaces as well as by its easy accessibility through such modern mass media as video and TV. The article argues that these fantasy spaces should not be regarded as misguided distortions, but as sources for gaining insight into what Marx called “the religion of everyday life,” and thus as revelations of the fantasy dimension of global capitalism. 相似文献

Molecular Population Genetics of Sex Determination Genes: The Transformer Gene of Drosophila Melanogaster 下载免费PDF全文
The transformer locus (tra) produces an RNA processing protein that alternatively splices the doublesex pre-mRNA in the sex determination hierarchy of Drosophila melanogaster. Comparisons of the tra coding region among Drosophila species have revealed an unusually high degree of divergence in synonymous and nonsynonymous sites. In this study, we tested the hypothesis that the tra gene will be polymorphic in synonymous and nonsynonymous sites within species by investigating nucleotide sequence variation in eleven tra alleles within D. melanogaster. Of the 1063 nucleotides examined, two synonymous sites were polymorphic and no amino acid variation was detected. Three statistical tests were used to detect departures from an equilibrium neutral model. Two tests failed to reject a neutral model of molecular evolution because of low statisitical power associated with low levels of genetic variation (Tajima/Fu and Li). The Hudson, Kreitman, and Aguade test rejected a neutral model when the tra region was compared to the 5'-flanking region of alcohol dehydrogenase (Adh). The lack of variability in the tra gene is consistent with a recent selective sweep of a beneficial allele in or near the tra locus. 相似文献

Incubation temperature determines sex in the mugger crocodile,Crocodylus palustris. Exclusively females are produced at constanttemperatures of 28.0°C through 31°C. At 32.5°C,only males are produced. Both sexes are produced in varyingproportions at 31.5, 32.0, and 33.0°C. Embryo survival isnot affected within this range, but developmental rate and totalincubation time are strongly temperature dependent. In naturalnests laid in breeding enclosures, cool incubation temperaturesproduced only females whereas males were produced only in warmnests. Clutch sex ratios were female or male biased. Yearlysex ratios (=percent male) varied from 0.05 to 0.58; overallsex ratio during six nesting seasons was 0.24 (1 male: 3 females).Sex ratio and incubation time vary with nest location and temperaturein a manner consistent with the constant temperature results.Incubation time decreases with increasing incubation temperature,and is an accurate predictor of sex ratio in the field and laboratory. To date, temperature-dependent sex determination (TSD) has beenreported in five species of Crocodylus and in three speciesof Alligatorinae; but the TSD patterns in these groups differ.The TSD pattern of C. palustris is similar to that of C. porosus.Nesting in C. palustris is synchronized with the seasonal availabilityof thermal regimes suitable for incubation. Resultant sex ratiosare a consequence of when and where eggs are laid. Early nestsare located in warm, sunny sites; in contrast, late season nestsare located in the shade. An egg transplant experiment demonstratedthat sex ratios could be altered by simple manipulations ofnest temperatures in the field. Abstract: Sex chromosomes have been reported in several dioecious plants. The most general system of sex determination with sex chromosomes is the XY system, in which males are the heterogametic sex and females are homogametic. Genetic systems in sex determination are divided into two classes including an X chromosome counting system and an active Y chromosome system. Dioecious plants have unisexual flowers, which have stamens or pistils. The development of unisexual flowers is caused by the suppression of opposite sex primordia. The expression of floral organ identity genes is different between male and female flower primordia. However, these floral organ identity genes show no evidence of sex chromosome linkage. The Y chromosome of Rumex acetosa contains Y chromosome-specific repetitive sequences, whereas the Y chromosome of Silene latifolia has not accumulated chromosome-specific repetitive sequences. The different degree of Y chromosome degeneration may reflect on evolutionary time since the origination of dioecy. The Y chromosome of S. latifolia functions in suppression of female development and initiation and completion of anther development. Analyses of mutants suggested that female suppressor and stamen promoter genes are localized on the Y chromosome. Recently, some sex chromosome-linked genes were isolated from flower buds of S. latifolia. 相似文献

近几十年来,发现很多爬行动物具有温度依赖型性别决定机制(TSD),即性别分化取决于胚胎发育过程中温度敏感期(TSP)的环境温度高低.龟鳖类存在两种TSD模式,即低温产雄性、高温产雌性的TSD Ⅰ a,低温、高温均产雌性而中间温度产雄性的TSDⅡ.TSD机制在生理水平的作用机制主要受到性腺类固醇激素的控制,温度通过影响芳香化酶和5α-还原酶的活性控制雌、雄激素转化,进而决定了个体的性别分化.在分子水平的研究发现:Sf1、Mis、Sox9、Dax1、Wt1和Dmrt1等基因的表达受到温度的影响,参与了龟鳖类性别分化.介绍了关于TSD的进化意义提出的假说,有待进一步验证. 相似文献

In Caenorhabditis elegans, the decision to develop as a hermaphrodite or male is controlled by a cascade of regulatory genes. These genes and other tissue-specific regulatory genes also control sexual fate in the hermaphrodite germline, which makes sperm first and then oocytes. In six pairings (one female × three males and vice versa) of honmoroko Gnathopogon caerulescens , although in one pairing the sex ratio of the offspring did not deviate significantly from 1:1, in four pairings the proportion of females decreased significantly with an increase in temperature. Heavy mortality due to disease was observed in the remaining pair. There were highly significant differences in sex ratios among the broods produced by different mothers with the same father and vice versa, and the response of sex ratio to temperature treatments differed considerably within pairings. The progeny of five out of 20 males produced at 34°C were almost all females, two were male-biased, and the remaining had balanced (1:1) sex ratios. These results suggest that the sex determination system in honmoroko is close to female homogamety but is influenced by temperature, genetic factors and genotype-temperature interactions. 相似文献
