In the editorial inaugurating this journal, Levine (1989) pointed to a new reductionism in biology, which--unlike the old reductionism that led to specialization and isolation of areas concerned with different aspects of a complex biological problem--is providing a renewed sense of unity. This development is the result of widespread use of common experimental methodology and the emergence of signal transmission and differential gene expression as themes that are central to many areas of modern biology. I describe here a set of complementary developments in molecular biology that focus attention on the problems of complexity and organization. Simple examples are given that illustrate the difficulty of relating systemic behavior to the properties of the underlying molecular determinants, and the outlines of a general approach to this problem are presented. These developments, together with those highlighted by Levine, are leading us to a new, more integrative intellectual paradigm whose fruits will be the elucidation of fundamental issues concerning network function, design, and evolution that cannot be addressed by the current paradigm.  相似文献

It is clear that the selection of the best possible algorithm for a computer program is essential for the creation of a useful tool. After this first step is taken, however, the usefulness of such a program may be greatly enhanced or impeded by the way it is implemented. We illustrate this point by describing our implementation of the well known FASTP/FASTIN algorithm in an interactive software environment.  相似文献

The tremendous complexity of even the simplest living unit makes a correct theoretical guess as to its mechanism very difficult. It is therefore suggested that, following the example of the physical sciences, a number of purely abstract cases in molecular biology be studied mathematically at first. Subsequent comparison of the different conclusions of such an abstract theory with available data would enable us to decide which of the conceivable situations are likely to occur in reality. As a first step toward such a study the problem of the minimal size of a living unit is studied. Usually the minimal size is considered to be determined by information-theoretical requirements. It is shown that the minimal size may have a very different origin. It may be determined by the possibility that too small a system, even though performing all necessary biological functions, may not be viable unless it is a member of a large group of other similar systems. This approach is developed both from a metric and from a relational point of view. Some relational characteristics of systems of reactions which possess the elementary metabolic properties of organisms are studied.  相似文献

The observation that phosphorothioate analogues of the nucleoside triphosphates are substrates for DNA- and RNA-polymerases has proven a boon for the molecular biologist. As these phosphorothioate-containing polymers are stable to degradation by nucleases and the sulfur atom confers many favourable chemical properties, several applications in molecular biology have been developed, including new methods for site-directed mutagenesis and DNA sequencing.  相似文献
