利用水提醇沉法对桦褶孔菌Lenzites betulina粗多糖HZKJc进行提取,随后经Sevage法和酶法联合脱蛋白,再经DEAE‐Sephadex A‐25,G‐100柱层析纯化后得到多糖纯品HZKJv。通过高效液相色谱(HPLC)测定上述纯化多糖的纯度和相对分子量分布;经纸层析(PC)、气相色谱(GC)、红外光谱(IR)进行单糖组成分析;并研究其清除自由基活性。分离纯化后得到的HZKJv为纯多糖,相对分子质量约为11 687。当HZKJv溶液浓度为3.0mg/m L时,其DPPH自由基清除率可达82%;浓度为2.0mg/m L时,对?OH自由基的清除率可达85%。表明HZKJv对?OH与DPPH自由基存在很好的清除作用。  相似文献

One polysaccharide PTP was isolated and purified from the roots of Polygala tenuifolia. It consisted of galactose, glucose and galactose in the ratio of 3.1:3.7:2.5, and a small amount of rhamnose, mannose and xylose. 17 general amino acids were identified to be components of the protein-bound polysaccharide analyzed by automatic amino acid analyzer. In order to test the anti-cancer activity of PTP, we investigated its effect against the growth of human ovarian cancer cells SKOV3 in vitro and in ovarian cancer rats. The intracellular reactive oxygen species (ROS) and glutathione (GSH) in SKOV3 cells following PTP treatment were also quantified to explore the possible mechanism underlying the antitumor activity of the polysaccharide. The result showed that PTP is effective on inhibiting the proliferation of SKOV3 cells in a concentration-dependent manner. Furthermore, treatment with PTP caused a rapid depletion of intracellular GSH content and accumulation of intracellular ROS, thus resulting in the apoptosis, which may prove to be a pivotal mechanism for its cancer protection action. In addition, a significant tumor growth inhibition effect was observed in nude mice after PTP administration for 7 weeks. All above indicated PTP could be beneficial towards ovarian cancer therapy.  相似文献

正交实验确定提取工艺后,用热水提取法得到苦瓜多糖(MCP).对MCP进行DEAE-32离子交换层析分离,得到3个多糖组分MCP1、MCP2和MCP3. 进一步采用Sephacryl S-400凝胶层析进行分离,经凝胶层析和高效液相色谱检测表明,MCP1、MCP2为均一性多糖组分.通过高效液相凝胶色谱法测定了两者的相对分子质量分别为1.16×106和7.45×105.用PMP衍生化法测定其单糖,结果表明: MCP1系由Man、Rham、GlcUA、GalUA、Glu、Gal、Xyl、Ara等单糖组成的杂多糖,摩尔比为1.03:2.93:1.00:14.95:2.16:30.70:2.85:4.50.MCP2系由Rham、GalUA、Gal、Xyl、Ara等单糖组成的杂多糖,对应的摩尔比为1.63:21.88:4.66:1.00:1.29.紫外光谱表明该多糖不含蛋白质和核酸.  相似文献

Pleurotus sajor-caju was produced on a basal medium containing spent shiitake substrate plus 10% wheat bran and 10% millet. An analysis of the fibrous composition of the spent shiitake substrate revealed that 85% of the original hemicellulose, 44% of the original cellulose and 77% of the original lignin was not consumed during production of a full crop (78% biological efficiency) of shiitake (63-day harvest period). To produce P. sajor-caju, the spent shiitake substrate was ground, air dried, supplemented, pasteurized with live steam and spawned. Highest yields (79% biological efficiency) of P. sajor-caju were obtained by supplementing the spent shiitake basal medium with 12% soybean and 1% CaCO3. Increases in percentage biological efficiency and mushroom size were positively correlated with increasing levels of CaCO3 added to the basal medium.  相似文献

A study was conducted to determine the effect of broiler poultry litter (BL) supplementation to spent mushroom substrate (SMS) on its storage and feed-nutritional value improvement. In Exp. 1, the sawdust-based SMS from a king oyster mushroom (Pleurotus eryngii) farm was mixed with BL at 0%, 25%, 50%, 75% and 100% levels on a wet basis and deepstack stored for short-term (1-wk) and long-term (3-wk). At 1-wk of short-term deepstack storage, SMS with more than 50% BL levels showed favorable conservation. At 3-wk of long-term storage, all treatments except for BL 100% had a serious fungal problem. Based on chemical parameters, BL-blending to SMS practically improved the feed-nutritional value of the mixtures. Since the deepstacking method was not effective for long term storage, in Exp. 2 SMS ensiled with or without BL was attempted to improve long-term (3-wk) storage. All the ensiled treatments (SMS 100%, SMS 75%+BL 25% and SMS 50%+BL 50%) had desirable fermentation. As in deepstacking, BL-blending to SMS improved the nutritive value of the ensiled mixtures. The populations of total bacteria, lactic acid bacteria and yeast were highest (P<0.05) when 75% SMS was blended with 25% BL. In conclusion, blending 50% or more BL with SMS was effective for the short-term (1-wk) deepstack storage. For long-term (3-wk) storage of SMS, an ensiling method was effective with or without the addition of BL.  相似文献

六妹羊肚菌Morchella sextelata是一种珍贵的食药用真菌,具有重要的经济价值。本研究在单因素试验基础上,采用响应面分析法优化其多糖提取工艺,利用Sevage法及透析法对粗多糖进行初步纯化,获得六妹羊肚菌多糖(Morchella sextelata polysaccharide,MSP)组分。采用凝胶色谱-示差-多角度激光光散射(gel permeation chromatography- refractive index-multi angle laser light scattering,GPC-RI-MALLS)、高效阴离子交换色谱(high performance anion exchange chromatography,HPAEC)、傅里叶变换红外光谱仪(fourier transform infrared spectrometer,FTIR)和气相色谱质谱联用仪(gas chromatography-mass spectrometry,GC-MS)等对其进行结构分析,并检测了该多糖清除自由基活性的能力。结果表明,六妹羊肚菌多糖最优提取工艺为:提取温度89.94℃、液料比31.07mL/g、提取时间162.86min。在此工艺条件下,提取率为23.98%。该多糖主要由分子量为4.655×10 6Da和6.571×10 4Da的两个组分组成,其单糖组成为葡萄糖、甘露糖、半乳糖,摩尔百分比分别为71.60%、23.70%和4.70%。该多糖的糖苷键主要有T-Glc、1,2-Man、1,4-Glc、1,6-Man、1,3,4-Man、1,4,6-Gal。此外,该多糖具有较强的清除2,2‐联氮基双‐3‐乙基苯并噻唑啉‐6‐磺酸[2,2‐azino bis (3‐ethylbenzothiazoline‐6‐sulfonic acid),ABTS]、1,1‐二苯基‐2‐苦基肼(1,1‐diphenyl‐2‐picrylhydrazyl,DPPH)和羟自由基活性的能力。本研究结果为六妹羊肚菌多糖功能食品的开发和利用提供研究基础。  相似文献

以槲蕨块茎为材料,采用水提醇沉法提取槲蕨多糖.在单因素考察的基础上,以多糖的得率为响应值,料液比、提取温度、提取时间为自变量,建立数学模型,利用响应面法获得槲蕨多糖的最佳提取工艺.采用纸片扩散法比较了不同浓度多糖的抑菌效果.结果表明:槲蕨多糖最佳提取工艺条件为:料液比1:25,提取时间2.0h,提取温度为85℃,在此优化条件下多糖得率为7.57%,与响应面分析预测值7.61%基本相符.槲蕨多糖对杆菌有明显的抑制作用,对供试球菌无明显抑制效果,多糖最低抑菌浓度为10 mg/mL.  相似文献

A small-silo study was conducted to develop an effective ensiling storage method for the use of cotton waste-based spent mushroom substrate (SMS) as an animal feed. The SMS was ensiled with 5% molasses (DM basis), 0.5% (v/w) lactic acid bacteria (LAB, Lactobacillus plantarum) inoculant or 0.5% (v/w) yeast (Saccharomyces cerevisiae) inoculant. The treatments included 100% SMS (control), 95% SMS+5% molasses (T1), 95% SMS+5% molasses+0.5% LAB (T2) and 95% SMS+5% molasses+5% LAB+0.5% yeast (T3). The treatments were ensiled for 10. Change in chemical compositions was little (P>0.05) according to the ensiling process and treatments. Compared with those before ensiling, 100% SMS (control) after ensiling showed unstable fermentative properties with high pH (5.2) and little lactic acid production. Compared with the ensiled control, treatments (T1, T2 and T3) resulted in decreased pH, 18-20 times higher concentrations of lactic acid, and greater populations of total bacteria (P<0.07), LAB and yeast (P<0.07). The addition of 5% molasses, 0.5% LAB and 0.5% yeast (T3) to the SMS resulted in the lowest pH (4.25) and the greatest microbial populations. Treatment T3 was selected for a large scale silo study which was ensiled for 10, 20 and 30 d. As in the small-silo study, the T3 treatment showed favorable fermentative and microbial parameters, compared with the control, by decreasing pH and increasing lactic acid concentrations, LAB and yeast populations. The minimum ensiling period was 20 d, when pH was reasonably low and LAB and yeast populations were greatest. In conclusion, molasses and microbial inoculation improved silage quality of SMS.  相似文献

采用水提醇沉法提取芦荟多糖,经DEAE-C32柱层析分离,Sephades G-100进一步纯化,得AⅠ、AⅡ和AⅢ三种芦荟多糖。Sephadex G-100凝胶色谱表明,AⅠ组分为均一组分,其分子量约为3.8×10~4。借助气相色谱技术,研究了芦荟粗多糖和AⅠ组分的单糖组成。另外,红外光谱鉴定芦荟多糖主要为吡喃多糖。  相似文献

Insect cecropins are small basic polypeptides synthesized in fat body and hemocytes in response to bacterial infections or hypodermic injuries. To explore a new approach for high expression of soluble cecropin in Escherichia coli cells, we fused the sequence encoding Musca domestica mature cecropin (named Mdmcec) in-frame to thioredoxin (TRX) gene to construct an expression vector pTRX-6His-Mdmcec. An enterokinase cleavage site was introduced between the 6xHis-tag and Mdmcec to facilitate final release of the recombinant Mdmcec. The fusion protein TRX-6His-Mdmcec was purified successfully by HisTrap HP affinity column and a high yield of 48.0mg purified fusion protein was obtained from 1L culture. Recombinant Mdmcec was readily obtained by enterokinase cleavage of the fusion protein followed by HPLC chromatography, and 11.2mg pure active recombinant Mdmcec was obtained from 1L E. coli culture. The molecular mass of recombinant Mdmcec determined by electrospray ionization-mass spectrometry (ESI-MS) is identical to that of native cecropin. Analysis of recombinant Mdmcec by circular dichroism (CD) indicated that recombinant Mdmcec contained predominantly alpha-helix with some random coil. Antimicrobial activity assays demonstrated that recombinant Mdmcec had a broad spectrum of activity against fungi, Gram-positive and negative bacteria. The procedure described in this study will provide a reliable and simple method for production of different cationic peptides for biological studies.  相似文献

为进一步开发和利用金缕半枫荷,该研究以超声时间、液料比、乙醇体积分数和超声温度为考察因素,用金缕半枫荷总多酚提取率作为评价指标,采用单因素和正交试验优化提取工艺确定最佳工艺,并以最佳工艺制备金缕半枫荷多酚;进一步采用DPPH·和羟自由基清除法评价金缕半枫荷多酚的抗氧化能力,以抑菌圈和最小抑菌浓度(MIC)为指标,采用牛津杯和肉汤稀释法考察其抗菌活性。结果表明:最佳提取工艺为乙醇浓度70%,料液比为1∶16,提取时间为60 min,提取温度为60℃。金缕半枫荷多酚清除DPPH·自由基的IC_(50)为5.22μg·mL~(-1),与阳性对照维生素C的IC_(50)值4.31μg·mL~(-1)无显著差异,羟自由基清除率IC_(50)为105μg·mL~(-1),显著低于维生素C的IC_(50)值(180μg·mL~(-1))。对金黄色葡萄球菌和大肠杆菌具有抑制作用,抑菌圈直径分别为(13.7±1.2) mm和(10.0±1.3) mm,MIC分别为0.393和0.785 mg·mL~(-1)。这表明金缕半枫荷多酚具有抗菌和显著抗氧化能力,为开发利用金缕半枫荷资源提供了理论依据。  相似文献

Summary Various spent agro-residues obtained after cultivation of the edible mushroom Pleurotus sajor-caju were used in anaerobic digestors for production of biogas. The changes that take place in the residues during bioconversion were quantified in terms of composition of cellulose, hemicellulose, lignin, carbon and nitrogen. These mycostraws resulted in increased biogas production over the untreated ones, which varied from 21.5% in the case of spent bagasse to 38.8% in the case of spent paddy straw. The increased biogas generation by the spent residues seems to be due to the increased susceptibility to digestion and more favourable C/N ratio of the residues. Offprint requests to: V. S. Polysaccharides, extracted from the herbs of Chinese natural Solanum lyratum Thunb (SLT), which is a traditional Chinese medicine with hot water and 0.04 M sodium hydroxide successively, were fractionated and purified by ion-exchange and gel-filtration chromatography. According to methylation, periodate oxidation, NMR spectroscopy, partial and graded acid and enzymic hydrolysis analysis, the results indicated the D-glucan to be linear and to contain both (1-->3)- and (1-->4)-linkages. The anomeric NMR and IR spectra measurements confirmed that the sugar residues were beta-glycosidically linked.  相似文献

Mycelial growth and extracellular polysaccharide production of Phellinus linteus were optimal at pH 5 and 25 °C. Maximum biomass production (14.2 g l–1) was after 15 d of cultivation, whereas, extracellular polysaccharide was maximal (3.5 g l–1) after 21 d. The hypoglycemic effect of the polysaccharide, investigated in streptozotocin-induced diabetic rats, decreased plasma glucose, total cholesterol and triacylglycerol concentrations by 49%, 32%, and 28%, respectively, and aspartate aminotransferase activity by 20%. The results indicate the potential of this polysaccharide to prevent hyperglycemia in diabetic patients.  相似文献

本研究对水溶醇沉法提取的鸡腿菇粗多糖脱蛋白方法进行了比较,最终确定Sevage法为最优的脱蛋白方法。经DEAE-纤维素52离子交换及Sephadex G-200分子层析柱分级、纯化,最终得到2个主要的多糖组分Ccp-I-A和Ccp-I-B。理化性质测定结果表明,二者均为白色絮状固体,能溶于水,不溶于无水乙醇、丙酮等有机溶剂;与斐林试剂,CTAB、硫酸-咔唑、碘-碘化钾及三氯化铁反应均为阴性。GC测定结果可知Ccp-I-A主要由甘露糖、葡萄糖和半乳糖组成,摩尔比为2.03∶9.52∶1;Ccp-I-B主要由岩藻糖和半乳糖组成,其摩尔比为1∶5.21。另外,Ccp-I-A和Ccp-I-B对DPPH和·OH显示出良好的清除能力,而且,相比于Ccp-I-B,Ccp-I-A的清除能力更高,当浓度为300μg/mL,其对DPPH和·OH的清除能力可分别达到72.1%和55.3%。  相似文献

Spent mushroom substrate (SMS) was treated with dilute sulfuric acid followed by cellulase and xylanase treatment to produce hydrolysates that could be used as the basis for media for the production of value added products. A L9 (34) orthogonal experiment was performed to optimize the acid treatment process. Pretreatment with 6% (w/w) dilute sulfuric acid at 120 °C for 120 min provided the highest reducing sugar yield of 267.57 g/kg SMS. No furfural was detected in the hydrolysates. Exposure to 20 PFU of cellulase and 200 XU of xylanase per gram of pretreated SMS at 40 °C resulted in the release of 79.85 g/kg or reducing sugars per kg acid pretreated SMS. The dilute sulfuric acid could be recycled to process fresh SMS four times. SMS hydrolysates neutralized with ammonium hydroxide, sodium hydroxide, or calcium hydroxide could be used as the carbon source for cultivation of Lactococcus lactis subsp. lactis W28 and a cell density of 2.9 × 1011 CFU/mL could be obtained. The results provide a foundation for the development of value-added products based on SMS.  相似文献

An aqueous ethanol extract of Ziziphus jujuba leaves was fractionatedby solvent extraction. The active chloroform-ethanol fractionwas separated into two components - ZjE-A, which had surfaceactive properties, and ZjE-B, which did not. Bioassay by psychophysicaltests on humans revealed sweetness-modifying activity in ZjE-A,but not in ZjE-B. The combined chemistry and bioassay resultsindicated that the active component is either closely associatedwith, or is itself, an amphipathic molecule and supported thenotion of a role for surface active properties in the tastemodifyingaction of Z. jujuba. Thin layer chromatography (TLC) and otherdata indicated and/or suggested that the active component inZ. jujuba consists of a group of compounds (ziziphins), whichprobably are triterpene saponin glycosides, as had been indicatedpreviously for the taste-modifying component in Gymnema sylvestre(gymnemic acids). However, the ziziphins differed from the gymnemicacids in TLC mobility and in coloration by a Liebermann-Burchardreagent. Therefore, the ziziphins are not the same compoundsas the gymnemic acids.
