Magadi soda (MS) is a commercially available form of sodium sesquicarbonate in Kenya. Soaking high-tannin sorghum (HTS) grain (2.5% catechin equivalents) in a 6 g l−1 MS solution was shown to eliminate measurable tannin content. A feeding trial was conducted to investigate the performance of broiler chicks fed soyabean-based diets containing HTS or MS-treated HTS as the sole cereal source from 1-3 weeks of age. A third diet containing sorghum with no detectable tannin content served as the control. Diets containing 220 g kg−1 crude protein were fed to 6 pens of 5 birds per pen for each treatment from 1-2 weeks of age and to 4 pens of 4 birds per pen per treatment in weeks 2-3. Feed intake, weight gain and feed efficiency (FE) were determined weekly. Apparent dry matter digestibility (DMD), metabolizable energy (AMEn) and nitrogen retention (NR) were measured over the last 3 days of each period. Gain and feed intake were not affected by treatment (P > 0.05), but FE and DMD were lower (P < 0.05) for the HTS diet than for the MS-treated HTS and control diets, which themselves were similar. Absolute NR was higher (P < 0.05) for the control diet than either of the HTS diets in week 2 but was the same for all diets in week 3. As % of intake, NR was higher (P < 0.05) for the control in both weeks 2 and 3 than for the HTS diets, although this was improved (P < 0.05) by MS treatment in week 2. AMEn values showed a similar pattern to that for NR in week 2 but in week 3 was the same for the MS-treated HTS and control diets, which were higher than the HTS diet (P < 0.05). Though total tannin content did not influence growth of birds in this study, MS treatment of HTS did reduce deleterious effects on FE, AMEn and NR as a % of intake.  相似文献

Multi-year spatial overlay patterns of plants, insects and soil water may yield insights for management for reducing biotic and abiotic stresses in dryland crops. A study of non-irrigated grain sorghum (Sorghum bicolor (L.) Moench) was conducted in a Pullman clay loam on the semi-arid High Plain of Texas during 2002–2005. The objectives of the 4-year study were to understand the mechanisms of plant spatial and temporal responses to stress from drought, infestations of greenbug, corn leaf aphid (CLA) and maize dwarf mosaic virus (MDMV) disease and soil water content (SWC) heterogeneity, and to reduce plant biotic and abiotic stress using their underlying relationships in space and time. Infrared IRt/c sensed-canopy temperature was measured at 18 or 54 sites along transects in a 6 m × 6 m grid across the years. Greenbugs, CLA, MDMV, SWC and hyperstectral reflectance were determined at each IRt/c site. Natural infestations of greenbugs and CLA on sorghum occurred in early July and insect populations peaked in late July or early August. Insect attacks resulted in plant water stress and sorghum yield loss except a late replanting in early July in 2004. Sorghum grain yield was negatively correlated with canopy temperature, greenbug and CLA (−0.38 < r < −0.75, P < 0.05), and positively correlated with SWC and plant near infrared reflectance (0.25 < r < 0.67, P < 0.05). The IRt/c temperature decreased with SWC but increased with greenbugs and CLA (0.26 < R2 < 0.64). Crosscorrelation analysis showed that these insect, crop, and soil variables were correlated in space within 48–54 m. Late planting in July or spray control in late July or early August would be options to reduce dryland sorghum water stress and yield loss from drought and insect attacks.  相似文献

In a 4 × 4 Latin-square experiment with a 2 × 2 factorial arrangement of treatments, 4 cattle fitted with a rumen and duodenal cannula were given four grass-containing diets [480 g kg−1 of the total dry matter (DM) intake] and barley (BU), barley + molasses (2:1) (BM), sugar-beet pulp (SU) or sugar-beet pulp + molasses (SM). Duodenal flow was estimated using Cr-mordanted straw and CoEDTA as markers, and microbial nitrogen entering the small intestine using purine bases of nucleic acids.

Molasses-containing diets had a higher (P < 0.01) organic matter (OM) digestibility. The proportion of digestible OM apparently disappearing in the rumen averaged 0.72 and was not significantly affected by the diet. When cattle received molasses, the quantity of microbial N entering the small intestine was higher (P < 0.05) and there was a trend towards a higher efficiency of microbial N synthesis (28.8 vs. 25.6 g N kg−1 OM apparently digested in the rumen). When S diets were consumed, total non-ammonia N flow at the duodenum exceeded N intake by 7.0 g day−1 and when B diets were consumed, it was 0.7 g day−1 less than N intake. Feed N degradability in the rumen and apparent N digestibility of S diets were lower (P < 0.05; P < 0.001) than those of B diets.

Rumen (P < 0.05) and total (P < 0.001) digestibility of neutral detergent fibre (NDF) and acid detergent fibre (ADF) was higher when S diets were given. The proportion of digestible fibre disappearing in the rumen was not affected by the diet. The rate and extent of silage and hay DM degradation were not significantly affected by the diet. However, dietary inclusion of molasses decreased (P < 0.05) the lag time of both hay and silage DM degradation.

The rumen dilution rate of liquid averaged 0.097 and that of particles, 0.049; neither was significantly different for either B and S diets or U and M diets. Duodenal liquid flow was higher (P < 0.05) for M diets.

Average rumen pH was not affected by the diet, but the molasses diets increased (P < 0.05) the range in rumen pH. The BM diet was associated with higher (P < 0.01) rumen ammonia concentration than the other diets. Low rumen ammonia concentrations (< 2 mM) were observed for long periods between feeds. The molar proportion of butyrate was higher on B diets and there was a trend towards a higher proportion of acetate and propionate on S diets. Molasses tended to increase the molar proportion of propionate and butyrate.  相似文献

The aim of the experiment was to study the oestrous behaviour and the peripheral blood plasma profiles of luteinizing hormone (LH), progesterone and the prostaglandin metabolite, 15-keto-13,14-dihydro-PGF2, during oestrus and metoestrus in repeat breeder (RBH) and virgin heifers (VH). Ten animals of each category were utilized. The RBH had a history of at least three inseminations without conception, and the VH were sexually mature animals not previously inseminated or mated. Oestrous symptoms were recorded and blood collected from the onset of prooestrus to 7 days after oestrus. The animals were inseminated during oestrus and their embryos were collected by a nonsurgical technique 7 days after insemination. The morphology of the embryos was evaluated.

The duration of oestrus was longer (P < 0.05) in the RBH (31.5 ± 3.6 h) than in the VH (23.8 ± 2.0 h). No differences in duration of prooestrus or in the interval from the end of oestrus to postoestrous bleeding were found between the heifer categories. The interval from the onset of oestrus to the preovulatory LH peak was longer (P < 0.05) in the RBH (12.2 ± 2.8 h) than in the VH (4.8 ± 1.5 h). There was a lower LH release in the RBH than in the VH, measured as the magnitude of the preovulatory LH peak (P < 0.05; 28.0 ± 4.0 vs. 40.7 ± 3.6 μg/l) or as the area under the curve of the LH peak (P < 0.01; 1141 ± 164 vs. 1765 ± 144 mm2). The progesterone levels were higher (P < 0.05) in the RBH than in the VH during the interval 5–48 h and lower (P < 0.05) during the interval 121–168 h after the LH peak. Peaks of the prostaglandin metabolite were seen during oestrus in both heifer groups. There were more prostaglandin metabolite peaks in the RBH than in the VH during the interval 13–24 h after the LH peak. Fewer normal embryos (P < 0.05) and more degenerated embryos (P < 0.01) were found in the RBH than in the VH group 7 days after insemination. No apparent relation was found between the morphology of the embryos and the hormonal changes.

The result of the study indicates a hormonal imbalance in the RBH. The hormonal asynchronism starts before or early in oestrus, which presumably leads to a sequence of improper hormonal changes responsible for the elevated embryonic loss in repeat breeder animals.  相似文献

Recent EU legislation discourages the practice of resecting piglets’ needle teeth. However, the effect of leaving piglets’ teeth intact on the welfare of sows in farrowing crates is poorly understood. Therefore, the aim of the study was to compare the effects of clipping and grinding piglets’ needle teeth, compared to leaving them intact, on the welfare of sows in farrowing crates.

Six days pre-partum 60 multiparous sows were assigned to one of three treatments. Litters had their teeth clipped (C), ground (G) or left intact (I) at birth. Sows’ teats were inspected for lesions pre-partum (day −3) and on days 1, 4, 11, 18 and 27 post-partum. Instantaneous scan samples (5 min intervals) of sow behaviour were carried out during three 2 h periods on days 1, 4, 8, 14, 21 and 26. On days 1, 4 and 11 all piglets were removed from the crate for 60 min. On re-introduction of the piglets, sow maternal behaviour was recorded continuously for 20 min.

The number of sows with teat lesions tended to differ between treatments on days 11 (P = 0.06) and 18 (P = 0.10). There was an interactive effect between treatment and day on sow dog-sitting behaviour throughout lactation (P < 0.001) and a tendency for an interactive effect on posture-changing behaviour (P = 0.08). On day 21, I sows were dog-sitting in more observations than C sows (P < 0.05) and on day 26 in more observations than C and G sows (P < 0.001). There was an interaction between treatment and day in the latency of sows to suckle their piglets following 60 min separation (P < 0.05). On day 4, I sows had a shorter latency to suckle than C and G sows (P < 0.05). There was an effect of treatment on the number of sows that terminated bouts of post-suckling udder massage (P < 0.05). On day 4, more I than C sows terminated post-suckling udder massage (P = 0.01). Finally, there was an effect of treatment on the time spent lying in the ventral posture in the observations of maternal behaviour (P < 0.05). C sows spent less time lying in the ventral posture than G and I sows (P < 0.05).

There were indications that leaving the teeth intact and to a lesser extent grinding caused injury and disturbance to sows. In farrowing crates, leaving piglets’ teeth intact cannot be recommended.  相似文献

Brittle rachis is an important trait to elucidate the domestication process in barley. Brittle rachis in wild barley (Hordeum vulgare ssp. spontaneum (C. Koch.) Thell) is controlled by two dominant complementary genes, Btr1 and Btr2. Cultivated barley (H. vulgare L. ssp. vulgare) lost the brittle rachis character during domestication as a result of mutation at the Btr1 or Btr2 locus. In this study, a high-resolution map of the btr1 locus was constructed using an F2 population of cultivar (cv. ‘Kanto Nakate Gold’) × wild barley (line OUH602). We cloned and sequenced 26 AFLP markers linked with the btr1 and btr2 loci. Ten converted STS markers were located on the short arm of chromosome 3H only, and at least 9 of the 10 STS markers were allelic with their original AFLPs. Efficient conversion of co-dominant STS markers using BAC clones was successful. No suppression of recombination was observed in the btr1 region even though wild barley was used as one of the parents. Initial results of BAC screening confirmed the resolution power of the developed high resolution map.  相似文献

Iso-nitrogenous and iso-energetic diets based on wheat, rye and 3 triticale cultivars were given with or without a supplement with a technical enzyme preparation containing pentosanase activity. The 10 diets obtained were given to a total of 600 broiler chickens from the age of 1 to 34 days. Considerable variation in chemical composition and productive value of the triticale cvs. was found. The triticale cv. Lasko had a chemical composition which closely resembled that of the wheat (cv. Holme), particularly regarding pentosan content. The rye (cv. Kungs II) and the triticale Sv 8008 had higher pentosan contents, while the triticale WW 31433 was intermediate between the wheat and rye.

At 21 days of age, live weights, feed intakes and feed conversion ratios were influenced by cereal type (P < 0.001), enzyme supplementation (P < 0.001) and the interaction of cereal type and enzyme (P < 0.001). In general, Lasko and WW 31433 (triticales) and wheat supported similar poultry production and were only marginally improved by enzyme supplementation. The rye gave an inferior production which, despite substantial improvement with enzyme supplementation, was still lower than that of wheat. Triticale Sv 8008 gave an intermediate production level which increased to that of the wheat following enzyme supplementation. An adaptation to the diets, particularly those rich in pentosans, was apparent as the chickens grew older and this reduced the beneficial effects of enzyme supplementation; however, statistically significant effects were still observed at 34 days of age.

The results indicate that pentosan content may be a negative indicator for evaluating the productive potential of triticale. It was also evident that supplementation with enzyme preparations high in pentosanase activity can improve the nutritive value of diets based on cereal grains with pentosan-rich endosperm cell walls.  相似文献

This study has been undertaken to determine the effect of a diet enriched with olive oil (OO) and high-oleic sunflower oil (HOSO) on fatty acid composition of erythrocyte membrane phospholipids and blood pressure in healthy women. OO and HOSO were used as natural sources of monounsaturated fatty acids (MUFAs) in a random-order sequence over two 4-week periods with a 4-week washout period between both MUFA diets. HOSO diet resulted in significant increases in oleic [(18:1n-9) 8.6%, P < 0.001], eicosenoic [(20:1n-9) 33.3%, P < 0.05], arachidonic [(20:4n-6) 6.2%, P < 0.05], and docosapentaenoic [(22:5n-6) 56.0%, P < 0.001] acids, whereas OO diet besides increased the content of stearic acid [(18:0) 13.6%, P < 0.01] and long-chain polyunsaturated fatty acids (PUFAs) of the n-3 family (22:5n-3 and 22:6n-3), when compared with the baseline [a diet high in saturated fatty acids (SFAs) and low in MUFAs]. In contrast, there was a significant decrease in linoleic acid [(18:2n-6) 21.8%, P < 0.001] for both MUFA diets. Consistent with these data, dietary intake of OO significantly raised total PUFAs (7.2%, P < 0.05), the n-3 fatty acids (22.2%, P < 0.01) and the PUFAs/SFAs ratio (9.3%, P < 0.01), as well as decreased the ratio of cholesterol to phospholipids (26,1%, P < 0.001) versus HOSO-based diet. Interestingly, dietary OO, but not HOSO, was able to significantly reduce the systolic (3%, P < 0.05) and diastolic (4%, P < 0.05) blood pressures. Although both vegetable oils provided a similar content of MUFAs (mainly oleic acid), our findings rather indicate that OO has important benefits to modulate the fatty acid composition of membranes and the mechanisms involved in the regulation of blood pressure in human.  相似文献

An experiment examined the intake, growth responses and rumen digestion of young sheep fed ad libitum oat or barley straws alone or supplemented with approximately isonitrogenous amounts of barley grain and urea (Bar/N), safflower seed meal (SAF) or linseed meal (LIN) supplements provided at 3 day intervals. The supplements comprised 15–22% of total dry matter (DM) intake. Sheep offered either of the straws alone consumed 35.0–37.2 g DM/kg liveweight (LW0.75) per day of straw and an estimated 2.03–2.07 MJ metabolizable energy (ME) per day, and lost 85–97 g LW per day. Supplements increased (P<0.05 or <0.001) voluntary intake of straw and of total DM, and the organic matter (OM) digestibility of the entire diet. Each of the supplements increased (P<0.001) the estimated ME intake to a similar extent and changed the rapid LW loss of sheep fed straw alone to approximate LW maintenance. Rumen ammonia concentrations in sheep fed barley and oat straws alone (12 and 24 mg NH3/l, respectively) were expected to be deficient for microbial activity, but were increased (P<0.001) by provision of the supplements. Digestion of straw in synthetic fibre bags incubated in the rumen was markedly increased (P<0.01 or <0.001) when supplements were provided. Rumen pH was depressed briefly to pH <6.0 by the Bar/N, but not by the LIN or SAF, supplements. In young sheep fed cereal straws and losing LW rapidly the oilseed meal supplements increased wool growth more than the barley grain–urea supplements, but both types of supplement increased ME intake similarly and were equally effective to reduce the extent of LW loss.  相似文献

Twenty-four growing Barki rams, 5–6 months old with an average body weight of 22 kg, were divided into asbestos shaded and unshaded groups during summer. Ad libitum feeding on roughage with 0.5 kg of concentrates per head per day was practiced. Drinking water was available twice daily in the early morning and afternoon. Biweekly observations included rectal temperature (RT; °C), and respiration rate (RR; breaths per minute) at 06:00 h and 14:00 h were recorded. Meteorological data, plasma glucose, calcium, inorganic phosphorus and packed cell volume were also measured. Results indicated that experimental animals developed hyperthermia during June to September, as evidenced by higher (P < 0.01) RT and RR than normal (40 ± 0.05 and 103.9 ± 3.87 vs. 38.9 ± 0.10 and 40 ± 6.56, respectively). Diurnal variations in these physiological phenomena were closely associated (P < 0.05) with ambient temperature changes. As compared with the unshaded group, providing an asbestos shed reduced (P < 0.05) RT and RR in the hyperthermic animals during the day (39.9 ± 0.07 and 94.7 ± 3.75 vs. 40.1 ± 0.08 and 113.1 ± 4.74, respectively), but it increased (P < 0.05) these parameters during summer nights (39.5 ± 0.05 and 82.4 ± 0.95 vs. 39.2 ± 0.07 and 71.6 ± 2.41, respectively). It also increased (P < 0.05) packed cell volume 1 h after morning drinking (35 ± 0.97 vs. 33.2 ± 0.60); reduced (P < 0.05) plasma glucose (43.3 ± 5.88 vs. 53.2 ± 6.31); caused hypocalcemia (10.9 ± 0.35 vs. 11.5 +- 0.43; P < 0.05) and hypophosphatemia (3.9 ±0.35 vs. 4.6 ± 0.34; P < 0.05) as a result of hyperthermia. Monthly variations in all parameters studied were higher (P < 0.01) during early summer. It is concluded that providing an asbestos shed for growing Barki rams under Egyptian summer conditions will not protect the animals from hyperthermia by day and night, as it interferes with extra body heat dissipation to the surroundings during summer nights. Although the unshaded animals were more hyperthermic during summer days, they tended to be normal during the night.  相似文献

A 2×3 factorial arrangement was used to investigate the interaction between lactose level (170 g/kg versus 275 g/kg), enzyme supplementation (with or without endo-1,3 (4)-β-glucanase) and toasting (raw barley versus toasted barley) in barley-based diets (250 g/kg) on piglet performance and diet digestibility postweaning. One hundred and forty-four weaned piglets (24 days old, 6.5 kg live weight) were blocked on the basis of live weight and were assigned to one of six dietary treatments (n = 6) for 21 days as follows: (1) 170 g lactose/kg and barley (250 g/kg), (2) 275 g lactose/kg and barley (250 g/kg), (3) 170 g lactose/kg and toasted-flaked barley (250 g/kg), (4) 275 g lactose/kg and toasted-flaked barley (250 g/kg), (5) 170 g lactose/kg and barley (250 g/kg) plus β-glucanase, (6) 275 g lactose/kg and barley (250 g/kg) plus β-glucanase. There was an interaction (P<0.05) between lactose level and β-glucanase supplementation on average daily gain (ADG), food conversion ratio (FCR) and coefficient of total tract apparent digestibilities (CTTAD) of dry matter (DMD), organic matter (OMD) and nitrogen (N). Pigs offered 170 g lactose/kg and β-glucanase supplementation had an improved FCR and ADG compared to the treatment based on 170 g lactose/kg and unsupplemented barley. However, β-glucanase supplementation at 275 g lactose/kg had no significant effect on FCR and ADG compared to the 275 g lactose/kg and unsupplemented barley. Pigs offered diets containing 170 g lactose/kg plus β-glucanase had a significantly higher CTTAD for DMD, OMD and nitrogen compared to the 275 g lactose/kg plus β-glucanase diet. However, there was no significant effect of lactose level in the unsupplemented diets. Pigs offered high lactose diets showed significantly increased total VFA concentration, molar proportions of butyric acid and reduced acetic acid compared to those offered low lactose diets. Pigs offered β-glucanase supplemented diets showed reduced (P<0.05) total VFA concentration compared to unsupplemented diets. In conclusion, β-glucanase supplementation improved diet digestibility and pig performance compared to the unsupplemented diet at the low lactose level only. There was no response of β-glucanase supplementation at the high level of lactose.  相似文献

A trial was performed to examine the effects of levels of barley substitution and supplementation with β-glucanase in a corn-soybean diet on growth performance and intestinal characteristics of broiler chickens. The experiment involved five levels of barley substituted for corn (0, 125, 250, 500, and 1000 g/kg) and two levels of β-glucanase supplement (with 0.5 g/kg and without) in a factorial arrangement with two replicates. Four hundred day-old commercial strain Harber broiler chickens were randomly allocated into twenty groups of ten dietary treatments for a six-week feeding trial, growing (0-3 week) and finishing period (4-6 week). Unless supplemented with β-glucanase, broilers receiving the diet with more than 250 g/kg of the barley substitute gained slower during the growth period. Conversely, supplementing β-glucanase did not improve total weight gain (0-6 weeks) with a diet of 500 g/kg barley substitution. As the level of barley substitution increased, feed intake in the growing period decreased significantly and viscosity of the intestinal contents increased. However, such an increase did not significantly influence feed conversion (P > 0.05). Supplementation with β-glucanase on diet up to 250 g/kg of barley substitute not only enhanced body weight gains of growing broilers, but also improved the live-weight of six-week-old broilers (P < 0.05).  相似文献

Teeth resection is a method of controlling the injurious effects of the aggression displayed when newborn piglets fight to establish a teat order. Recent European legislation discourages the practice. The objective of this study was to assess the effect of clipping and grinding piglets’ needle teeth, compared to leaving them intact, on the welfare of piglets in farrowing crates.

Six days pre-partum, 60 sows were assigned to one of three treatments. Litters had their teeth clipped (C), ground (G) or left intact (I) at birth. The time taken to carry out each procedure was recorded. Piglet weights and facial lesions, which were scored according to severity, were recorded on days 1, 4, 11, 18 and 27. Piglet weights were also recorded at birth. Mouth lesions were recorded on days 1, 4 and 27. Instantaneous scan samples of piglet behaviour were carried out for 30 min post-teeth resection procedure (1 min intervals), and for 6 h on days 1, 4, 8, 14, 21 and 26 (5 min intervals). One male and one female piglet per litter were chosen as focal animals and observed for 5 min each post-procedure and for 10 min each twice per day on days 1, 5, 12, 20 and 26. Mortalities were recorded throughout lactation.

Grinding took significantly longer than clipping the teeth or leaving them intact (F = 638.87, P < 0.001). I piglets had higher facial lesion scores than C and G piglets (F = 10.58, P < 0.001). A smaller proportion of piglets in I litters than C and G litters and a smaller proportion of piglets in G litters than C litters had at least one mouth lesion (F = 4.74, P < 0.001). During 30 min post-procedure, I piglets were active on the heatpad in more observations than C and G piglets (F = 3.49, P < 0.05). During 5 min post-procedure C piglets spent longer chomping than I piglets (F = 5.92, P = 0.05). On day 21, I piglets were active in more observations than G piglets (F = 2.11, P < 0.05). On day 26, G piglets were inactive in more observations than C and I piglets (F = 5.02, P < 0.05). On days 14 and 26, C piglets were sleeping in more observations than G piglets (F = 2.87, P = 0.05). There was a tendency for a larger proportion of I than C piglets to die due to overlying (F = 2.68, P = 0.08).

In conclusion, although all three options were associated with welfare problems, grinding can be recommended in preference to clipping or leaving the teeth intact.  相似文献

Lymphocytes from sows maintained in a constant hot environment (32°C) showed reduced proliferative responses to mitogens PHA (P < 0.02) and PWM (P < 0.01) in comparison to sown maintained in a constant cool environment (21°C). In the piglets the hot constant temperature slightly reduced (P < 0.05) proliferative responses of lymphocytes to PHA.
2. 2. No significant effects of a cycling hot environment (27–32°C) were found for any proliferative responses of lymphocytes from sows and litters.

Plasma levels of copper and lipid peroxidation were evaluated in 14 smokers as compared to 14 nonsmokers. Plasma copper concentrations were higher in smokers than in nonsmokers (122.5 ± 19.15 vs. 101.5 ± 16.2 μg/dl, P < .01). Plasma lipoperoxidation, evaluated as fluorescent damage products of lipid peroxidation (FDPL), also was higher in smokers than in nonsmokers (20.35 ± 2.6 vs. 17.1 ± 2.95 units of relative fluorescence/ml, P < .01). A significant and positive correlation between the number of cigarettes smoked, expressed as pack years, and the levels of either FDPL (r = .61, P < .025) or copper (r = .55, P < .05) was found. Moreover, a significant and positive relationship between copper and FDPL values was observed in smokers (r = .64; P < .025), but not in nonsmokers. We have investigated the role of steroid hormones as coronary risk factors in Helsinki Heart Study population of dyslipidemic middle-aged men. We compare here the effects of gemfibrozil and placebo on the serum levels of dehydroepiandrosterone (DHEA), its sulfate (DHEAS), their metabolite androstanediol glucuronide (3-AdiolG), androstenedione, cortisol, testosterone, and sex-hormone binding globulin (SHBG) in non-smokers. We also examined the associations between steroid and lipoprotein levels in both treatment groups. Compared with placebo gemfibrozil treatment was associated with significant elevations of the mean levels of DHEA 10.2 vs 8.0 nmol/1; P<0.005, of DHEAS 8.0 vs 5.8 μmol/1; P<0.001, of 3AdiolG 18.3 vs 8.4 nmol/1; P<0.001, of androstenedione 5.7 vs 5.1 nmol/1; P<0.02, and of cortisol 426 vs 358 nmol/1; P<0.001. The mean SHBG levels decreased from 46.4 to 41.7 nmol/1; P=0.03 with gemfibrozil treatment. No difference was found in testosterone levels 17.7 vs 18.8 nmol/1; P=0.11, or the ratio of testosterone/SHBG 0.45 vs 0.43; P=0.23. Positive correlations were found between high density lipoprotein-cholesterol and DHEAS (r=0.267; P<0.01) and DHEA (r=0.282; P<0.01) levels and negative correlations between low density lipoprotein-cholesterol and 3-AdiolG (r=−0.400; P<0.001) and cortisol (r=−0.281; P<0.01) levels in the gemfibozil group. Our results indicate that gemfibrozil treatment increases the production and turnover of adrenal androgens and cortisol, and suggest that activation of the adrenocorticol function and increased metabolism of androgens are related to the improved lipoprotein pattern during gemfibrozil treatment.  相似文献

The rate-limiting step in luteal biosynthesis of progesterone consists of cleavage of the side chain of cholesterol by mitochondrial cytochrome P450 side-chain cleavage enzyme (P450scc) to form pregnenolone. Luteal mRNA encoding P450scc, quantitated on selected days of the 16-day ovine estrous cycle, was similar on days 3 and 6, increased by 2-fold on day 9 (P < 0.05) and remained elevated on day 15. Levels of P450scc mRNA on day 15 of pregnancy were not different from those found on any day of the cycle (P < 0.05). To determine whether levels of mRNA encoding P450scc are hormonally regulated, ewes on day 10 of the estrous cycle were injected with hCG or prostaglandin F2 (PGF2). P450scc mRNA was not increased for up to 36 h after injection of hCG, nor decreased within 8 h after injection of PGF2 (P < 0.05). An assay for P450scc activity was developed which utilized ovine small and large luteal cells in the presence of 22R-hydroxycholesterol and ovine high density lipoprotein. Enzyme activity was quantitated by measurement of progesterone production. In small luteal cells activation of the protein kinase A (PKA) second-messenger system by treatment with LH resulted in 910% increase in progesterone production without altering activity of P450scc. Activation of the protein kinase C (PKC) second-messenger system with phorbol 12-myristate 13-acetate caused a 51% reduction in progesterone secretion from large luteal cells but did not alter activity of P450scc. These findings suggest that in mature luteal tissue steady state levels of mRNA encoding P450scc, and enzyme activity are independent of acute regulation by activation of PKA or PKC second-messenger systems.  相似文献

Hydrous fuel ethanol (95%) and distiller's wet grain (DWG) were produced in a farm-scale plant (< 4 million liters ethanol year−1) from corn, wheat, and grain sorghum particles of various sizes, from corn combined with thin stillage-whey, and from various other cereal grains. These variations were made in a search to find the best set of conditions for maximizing the energy balance (energy output divided by energy input) and minimizing the cost of ethanol production. We found that the optimum hammermill screen size for corn, wheat, and grain sorghum was 1·59–2·38 mm. In tests with thin stillage and whey a higher energy balance (2·91) occurred when one part whey was mixed with three parts stillage, rather than the reverse (2·69). However, the reverse (three parts whey and one part stillage) gave a lower ethanol cost ($0.45 liter−1) than the original ($0.47 liter−1). Tests with various cereal grains (corn, oats, wheat, barley, rye, and grain sorghum) gave identical energy balance values (2·26) when 10% (v/v) ethanol beers were produced. However, rye ($0.50 liter−1), grain sorghum ($0.46 liter−1), and corn ($0.51 liter−1) yielded ethanol at the lowest net cost. Recommendations for farm-scale plants are also provided.  相似文献

Starch from AC Hill oat grains (Avena nuda) was isolated and some of the characteristics determined. The yield of starch was 23·4% on a whole grain basis. The shape of the granule was polyhedral to irregular, with granules 6–10 μm in diameter. Lipids were extracted by acid hydrolysis and by selective solvent extraction with chloroform-methanol 2:1 v/v (CM) at ambient temperature, followed by n-propanol-water 3:1 v/v (PW) at 90–100°C. The acid hydrolyzed extracts which represented the total starch lipids (TSL) was 1·13%. The free lipids in the CM extract (1% TSL) was 6·2%, whereas the free and bound lipids in the PW extracts was 93.0%. Neutral lipids formed the major lipid class in the CM and PW extracts. The monoacyl lipid content in both CM and PW extracts was 61·0%. The total amylose content was 19·4%, of which 13·9% was complexed by native lipids. X-ray diffraction was of the ‘A’ type. Oat starch differed from wheat starch in showing a higher swelling factor, decreased amylose leaching, coleaching of a branched starch component and amylose during the pasting process, higher peak viscosity and set-back, low gel rigidity, greater susceptibility towards acid hydrolysis, greater resistance to -amylase action and a higher freeze-thaw stability. Furthermore, in comparison to wheat starch, the amylose chains of oat starch appear to be more loosely arranged in the amorphous regions, whereas in crystalline regions, oat starch chains are more compactly packed. Lipid removal from oat and wheat starches decreased their swelling factor, peak viscosity, set-back, gelatinization temperatures, freeze-thaw stability and paste clarity (at pH > 4·0), and increased their thermal stability, amylose leaching, enthalpy of gelatinization, susceptibility towards -amylase and paste clarity (at pH < 4·0). The results also showed that the properties of AC Hill oat starch is not representative of oat starch in general.  相似文献
