OBSERVATIONS ON FEEDING METHODS IN SOME FLORIDA ASCOGLOSSANS   总被引:2,自引:0,他引:2  
Feeding methods are described for 12 Florida ascoglossans. Relationshipsbetween size and shape of radular teethand dimensions of foodalgae are analysed. Feeding in a diatom-eating species (Eylsiaevelinae) is described for the first time. The suctorial feedingof Ascobulla ulla and the structure of its radula and alimentarysystem clearly reveal that it is an ascoglossan, not a cephalaspid.The very complicated method of feeding in Oxynoe spp. is consideredan evolutionary "dead end", and the branching of the digestivegland and position of pharyngeal appendices seem to indicatethat the Elysiidae did not evolve from the Oxynoidae. New anddetailed observations on feeding in Elysia ornata, Placida kingstoni,and Ercolania funerea show that after cell sap has been suckedinto the pharynx, it is returned to the algal filament. Thisprocess may be repeated once or twice before the cell sap iseventually pumped into the stomach. Cell sap is probably mixedwith saliva in the pharynx to reduce its viscosity, and thepharyngeal regurgitation facilitates withdrawal of the remainingsap. *Present address: Ellebjergvej 22, 1 t.h., DK-2450 CopenhagenSV, Denmark. Cylindrical 52.5-nm-long phycobilisomes were observed in Anabaenavariabilis, differing from the generally accepted hemidiscoidalmorphology. The central part of such a phycobilisome has a network-likefine structure of slightly greater diameter (16 nm) than theconnected end parts of stacked-disc structure (12 nm in diameter).On the basis of this morphology, the molecular mass of thisphycobilisome was calculated to be 3.27?106, about 60% of whichis accounted for by phycocyanin with the rest being due to allophycocyanin.Separately prepared 23 S allophycocyanin particles with a molecularmass of 1.13?106 have the dimensions (16?23 nm) and network-likefine structure similar to the central part of phycobilisomes,while an aggregate form of phycocyanin (18 S) has a fine structureof stacked discs similar to the connecting end part of phycobilisomes,suggesting that the central part constitutes the core at whichthese phycobilisomes attach to the thylakoid membranes. Salps were gently captured in a perspex chamber by a SCUBA diver,who then injected a suspension of 14C-labeled phytoplanktonand 3H-labeled bacteria into the chamber. After 0.5 h incubationin situ, filtering rates were estimated from incorporation ofthe two isotopes, and expressed as a function of each salp'ssize. Both bacteria and phytoplankton were grazed by Thalia,the latter at higher rates. Weight-specific grazing rates increasedwith increasing size of salp. Estimated hourly rations rangedfrom 1% of bodily C for small Thalia feeding only on phytoplanktonto 8% of bodily C for large Thalia feeding on all particulateorganic C. The effects of shortening distance at Vu,the unloaded shortening speed, and filament overlap on the amount ofextra Ca2+ released during relaxation in muscle, asindicated by the bump area, were studied. Single, intactfrog skeletal muscle fibers at 3°C were used. The myoplasmic freeCa2+ concentration ([Ca2+]i) wasestimated by using fura 2 salt injected into the myoplasm. Ramps wereapplied, either at full overlap with different sizes or at varyingoverlaps with a fixed size, in the linear phase of relaxation. At fulloverlap, a plot of bump area vs. ramp size was fit by using a sigmoidalcurve with one-half of the bump area equal to 25.9 nm. With a fixedramp size of 100 nm/half-sarcomere, the plot of bump area vs. meansarcomere length (SLm) was fit by a straight lineintersecting the SLm axis at ~3.5 µm, close to just nooverlap. The results suggest that the transition in the distribution ofattached cross bridges from the isometric case to one appropriate forunloaded shortening at Vu is completed within 50 nm/half-sarcomere and support the view that attached crossbridges in the overlap zone influence the affinity of Ca2+for troponin C in the thin filament.

Partially purified homoserine dehydrogenase was prepared frompea seedlings. The optimum pH for this enzyme is approximately 5.4. The Kmvaluesfor ASA and TPNH are 4.6xl0–4Af and 7.7xl0–5M, respectively.This enzyme can also utilize DPNH but less effectively thanTPNH. In contrast with yeast homoserine dehydrogenase whichis insensitive to — SH reagents, the pea enzyme is inhibitedalmost completely by 10–4MPCMB and 10–5MHgCl2, theinhibition being removed by 10–2M thioglycolate. Homoserinedehydrogenase was found not only in decotylized seedlings, butalso in cotyledons. The significance of this enzyme in homoserine biosynthesis ingerminating pea seeds has been discussed. Cladoceran in situ feeding rates on natural bacteria labelledwith [methyl-3H] were studied in parallel with feeding ratedeterminations on 14C-labelled Chlorella in a hypertrophic subtropicalreservoir (Lake Hartbeespoort) through spring and summer (1986/87).Community filtration rates (CFR5) on bacteria and algae weresimilar, but selection for Chlorella (relative to natural bacteria)increased from midsummer in association with declining bacterialdensity and increasing dominance of ‘inedible’ componentsof the natural phytoplankton. Species-specific filtration rates(SSFRs) were determined for Daphnia pulexllongispina, Ceriodaphniareticulata, Diaphanosoma excisum, Bosmina longirostris and Moinamicrura during their respective seasonal occurrence in the studyperiod. SSFRs on algae and bacteria increased with body length(L, mm) in all species apart from Bosmina. Species-specificdifferences in absolute feeding rate (FR, ml animal–1day–1), the slope of the FR-L relationship and bacteriaselectivity were evident. The objective of this study was to quantify the functional responsein feeding rate in the various developmental stages of Calanusfinmarchicus to different concentrations of the diatoms Thalassiosiranordenskioeldii and Porosira glacialis, and the haptophyseanPhaeocystis pouchetii. Grazing of copepodite stage I–VC.finmarchicus was measured using two different approaches.Feeding rates were obtained from either incubation experiments,estimating the rate of removal of particles from suspension,or by quantifying the turnover rate of the plant pigments inthe gut. Clearance as a function of algal concentration (1–30µg plant pigment 1–1) was described in juvenilestages of C.finmarchicus fed the diatoms T.nordenskioeldii [20µm equivalent spherical diameter (ESD)], P.glacialis (40µm ESD), and two size categories (30–100 µmand >100 µm ESD) of the gelatinous alga P.pouchetii.When the copepodite stages were fed T.nordenskioeldii, the gutcontent of plant pigments was in general higher than when fedP.glacialis. Rates obtained were variable when the same copepoditestages were offered the two size categories of P.pouchetii,but within the same order of magnitude as those obtained forthe larger diatom. At unialgal diets, diatoms were more readilyconsumed than the larger size fraction among colonies of P.pouchetiiby copepodite stage I–III C.finmarchicus. But given anappropriate prey size, C.finmarchicus grazed both diatoms andcolonies of gelatinous algae at equal rates. A linear relationshipbetween gut content and food concentrations <10 µgchlorophyll 1–1 was found. Post-embryonic development time, egg production rate and adultbody size of calanoid copepod Eurytemora affinis from Lake Ohnuma,Japan, were determined under six temperature-food conditions(103,5 x 103, 104 and 5 x 104 cells ml–1 at 15°C,5 x 104 cells ml–1 at 10and20°C) in the laboratory.The measured parameters varied with both temperature and foodconcentration. Development time from hatching to adult femalewas 9.2, 11.4 and 22.8 days at 20, 15 and 10°C respectively,at the highest food concentration. The males developed to adultat one to two days earlier than the females. An effect of foodshortage on development time occurred at the lowest food concentration.This development time was 24.8 days even at 15°C, beingtwice as long as that at the highest food concentration. Prosomelength of these food-limited females was approximately 75% ofwell-fed ones, which reduced by only 10% with increasing temperaturefrom 10 to 20°C. Clutch size (C, eggs clutch–1) ofwell-fed individuals depended on prosome length of the adultfemale (L, mm), and was expressed as an equation: C = 65.2 L383. Clutch size of individuals reared at less than 104 cellsml–1, however, mostly laid below the estimated curve,especially at the lowest food concentration being only 10% ofthat at the highest food concentration. These results suggestthat food availability is a more important factor affectingpopulation growth of E.affinis in Lake Ohnuma than variationof temperature.  相似文献

The growth of the anther filaments in the Lilium bud is essentiallyexponential after a length of 5 mm is reached, when the celldivision rate declines and growth is by cell extension, particularlyof cells in the central part of the filament. During the periodof extension, fresh vascular tissue is formed and xylem continuityis conserved up to the time of anther dehiscence, in contrastto the Graminae where the filament extends much more rapidly.The expansion of the lily filament depends in part on the continuoustransfer of potassium ions from the receptacle throughout theperiod of growth. Although starch to sugar conversion may bean important source of osmoticum as the cells expand, regulationof K+ ion movement would be likely to provide a more sensitiveand faster method of regulation of cell osmoticum. The redox kinetics of P700 induced by far-red light and a pulseof strong white light in wild type cells were compared withthose in NAD(P)H dehydrogenase (NDH)-defective mutants of thecyanobacterium Synechocystis PCC 6803. The wild type cells showedthe electron donation from the respiratory donor and the photoreductantgenerated in PS I to P700+ through the plastoquinone, whichis mediated by a Hg2+-sensitive enzyme. The NDH-defective mutantcells, however, did not show the electron transfer to P700+through the plastoquinone from both the photoreductant in PSI and cytosolic electron donors using pyndine nucleotides asan intermediate. Thus, NDH appears to be the site of main entryof electrons into the plastoquinone pool in the NAD(P)H-mediatedcyclic electron flow and the respiratory electron flow in Synechocystis. We investigated the seasonal occurrence, wet : dry : carbon: nitrogen weight ratios, population biomass, gastric pouchcontents, and rates of feeding, growth and respiration of thescyphomedusa Aurelia aurita in the central part of the InlandSea of Japan. Aurelia aurita medusae began to appear in January/Februaryas ephyrae, reached annual maximum body size in July/August,and disappeared, presumably due to death, by November. Initialslow growth in early spring was followed by a period of exponentialgrowth (mean growth rate: 0.069 d–1) between April andJuly. In the Ondo Strait, which is characterized by strong tidalmixing, the A. aurita population (mean carbon biomass: 66.0mg C m–3) overwhelmingly dominated the zooplankton-communitybiomass (mean biomass of micro- and mesozooplankton: 23.7 mgC m–3) between May and early August The gastric contentanalysis revealed that A. aurita ate almost all micro- and mesozooplankters,of which small copepods were most important. On the basis ofdigestion time for small copepods (60 min) and their abundancein the gastric pouch of field-collected A. aurita, we determinedthe weight specific feeding rates and clearance rates. The formerincreases linearly with increasing copepod abundance, but thelatter was relatively constant irrespective of the food supply.We also measured the respiration rates of A. aurita and expressedthem as functions of body weight and temperature. These physio-ecologicalparameters enabled us to construct the carbon budget of theA. aurita population typical of early summer in the Ondo Strait.Predicted population-feeding rate (6.07 mg C m–3 d–1)was higher than the population-food requirement for both metabolismand growth (4.55 mg C m–3 d–1), indicating thatfood supply was sufficient to sustain the observed growth rate.This feeding rate was equivalent to 26% of micro- and mesozooplanktonbiomass, a significant impact on zooplankton.  相似文献

Using X-ray photography and flow cytometry, the internal morphologyand DNA replication activity of wild type (wt), GA- (gib-1 )and ABA-deficient (sitw ) tomato (Lycopersicon esculentum Mill.cv. Moneymaker) mutant seeds were studied. During seed formation,from 30 to 45 d after pollination (DAP) the endosperm becomessolid and the seed starts to gain desiccation tolerance. Atthis time significant changes occur in the amounts of DNA inradicle tip cells. At 30 DAP, radicle tip cells of the threegenotypes manifest about 60% of 2C, 30% of 4C and 10% of 8Camounts of DNA. Upon maturation (45 DAP onwards), most cellsin the seeds of the three genotypes arrest in the G1phase ofthe cell-cycle with 2C amounts of DNA. However, a relativelyhigh proportion of cells with 4C amounts of DNA was detectedin the radicle tip cells ofsitw compared with wild type andgib-1. At the well-matured stage (60 DAP), there were about 2% ofseeds with free space in wild type andgib-1 , and about 13%insitw . At the over-matured stage (75 DAP), even more seedswith free space were found insitw , whereas no increase in theproportion of the seeds with free space was detected in theother two genotypes. In -1.0 MPa PEG-6000 with or without 10µM GA4+7, no germination occurred in well-matured wildtype andgib-1 seeds, whether or not they were dried after harvest.However,sitw seeds were able to germinate both in over-maturefruit and in -1.0 MPa PEG-6000. Priming of dried seeds in -1.0MPa PEG induced a large amount of free space in almost all seedsof the three genotypes, and nuclear DNA synthesis in the radicletip cells of wild type andsitw seeds. However, PEG priming offresh (non-dried) seeds had no effect on the amount of freespace and 2C/4C DNA ratios in wild type orgib-1 seeds, but didinduce free space in about 20–25% ofsitw seeds and provoked4C signals insitw seeds. Removal of the endosperm and testaopposite the radicle tip of seeds resulted in root protrusion,the induction of free space and an increase of 4C DNA signalsin the three genotypes. It is concluded that ABA is crucialfor the efficient arrest of tomato embryo radicle tip cellsin G1phase upon maturation, whereas GAs play an important rolein re-initiating 4C DNA levels upon germination. 2C DNA content values for 70 orchid species from 26 genera,including 37Dendrobiumspecies from eight taxonomic sections,were analysed using flow cytometry. The resulting nuclear DNAcontent values for species other thanDendrobiumranged from 1.91pg 2C-1to 15.19 pg 2C-1nuclei forCadetia tayloriandVanilla phaeantha,respectively.Dendrobiumnuclear DNA content values ranged from1.53 pg 2C-1to 4.23 pg 2C-1nuclei forD. cruentumandD. spectabile,respectively. Fracture toughness was estimated for a 'least tough' path inthe leaves of woody species from three sclerophyllous plantcommunities. Most of the species from Mediterranean, tropicalheath forest and lowland tropical rain forest habitats had verytough leaves, with toughness generally 600-1300 J m-2, whichis two to four times higher than soft-leaved tropical pioneertrees. The toughest leaf (2032 J m-2), Parishia insignis, camefrom the canopy of the lowland rain forest. The goal of this research is to enhance our knowledge of thecontributions of doliolids to the planktonic community as consumersand secondary producers. The objectives are to quantify feedingand growth rates of Dolioletta gegenbauri gonozooids at fourfood concentrations and four temperatures in order to determinetheir impact as grazers throughout the water column. Althoughdoliolids are abundant in numerous regions of the coastal ocean,and are considered to be major planktonic grazers, data on ratesof feeding and growth are scarce. Laboratory experiments wereconducted at 16.5, 20, 23.5 and 26.5°C to quantify removalof a 50:50 volumetric concentration of Thalassiosira weissflogiiand Rhodomonas sp. at four different food concentrations of20, 60, 160 and 390 µg C l–1. Results from theseexperiments suggest that clearance rates are similar at concentrationsfrom 20 to 60 µg C l –1, and decrease as the foodconcentrations increase to 160 and 390 µg C l –1.The ingestion rates increase over a range of phytoplankton concentrationsfrom 20 to 160 µg C l –1, then decrease when abnormallyhigh concentrations of 390 µg C l –1 are offered.Clearance and ingestion rates increase as temperature increasesfrom 16.5 to 26.5°C. The exponential growth rates rangefrom k = 0.2–0.7, with the lowest rates occurring at thehighest food concentration. Growth rates increase with increasingtemperature from K = 0.1–0.3 at 16.5°C to 0.45–0.7at 26.5°C. In each case, the small- and medium-sized zooidshad higher growth rates than the larger gonozooids. These resultssuggest that doliolid feeding and growth rates are a functionof environmental food concentrations and temperatures, and implythat they can be important consumers in a changing neritic environment.  相似文献

By manipulation of various growth regulators and physical conditions,plants have been regenerated from excised roots, stem segments,cotyledons, leaves, and callus cultures of red cabbage (Brassicaoleracea var. capitata) grown under in vitro conditions. Shootbuds were induced on isolated root segments (1 cm long) culturedon Murashige and Skoog's medium and the frequency of bud formationwas greatly enhanced by the addition of kinetin (0.5 part 10–6).Callus obtained from the seeds, cotyledons, and hypocotyl segmentscultured on a medium fortified with 2,4-D (1 part 10–6),kinetin (0.1 part 10–6), and coconut milk (10%, v/v) hasbeen repeatedly subcultured. The callus is slow growing, andon transference to a kinetin (2 parts 10–6) and IAA (2parts 10–6) medium underwent morphogenesis to give riseto plants. Metabolism of Inorganic Carbon Taken Up by Roots in Salix Plants   总被引:1,自引:0,他引:1  
The metabolic products of inorganic carbon taken up throughthe roots from nutrient solution were studied in willow plants.Willow cuttings (Salix cv. Aquatica gigantea) were suppliedwith unlabelled or 14C-labelled NaHC03 for 1, 5, 10, and 24h in light or in darkness. After feeding, the plants were dividedinto six samples (upper and lower leaves and corresponding stems,cuttings and roots), which were frozen in liquid N2. Freeze-driedground samples were extracted into water-soluble, chloroform-solubleand insoluble fractions. The water-soluble fraction was furtherseparated into basic, acidic, and neutral fractions by ion-exchangechromatography. In the light experiment pronase treatment wasused to separate the insoluble fraction into proteins and insolublecarbohydrates. After I h feeding time, most of the 14C was fixed into organicacids and amino acids both in light and in darkness in all partsof the plants. In the roots a large part of the l4C-carbon wasincorporated into the protein and insoluble fractions alreadyduring short feeding times, and the amounts incorporated increasedwith time. In the leaves, after 1 and 5 h the main labelledcompounds were the organic acids and amino acids, but after10 h about half of the total 14C was in protein and in the insolublefraction. A further analysis of amino acids and organic acidswith HPLC showed that C-4 acids were labelled initially andthat over time the proportion of different acids changed. These results indicate that the metabolism of carbon in rootsmight take place via ß-carboxylation of PEP. Partof the fixed 14C is transported from the roots, probably asamino acids and organic acids, to the shoot. Soluble proteins extracted from leaves of Chenopodium albumcatalyzed the conversion of pheophorbide a to a precursor ofpyropheophorbide a, putatively identified as C-132-carboxyl-pyropheophorbidea. The precursor was then decarboxylated non-enzymatically toyield pyropheophorbide a. Soluble proteins and pheophorbidea, as the substrate, were required for the formation of theprecursor, and boiled proteins were enzymatically inactive.The maximum rate of conversion of pheophorbide a to the precursoroccurred at pH 7.5. The Km for pheophorbide a was 12.5 µMat pH 7.0. Both pheophorbide b and bacteriopheophorbide a couldserve as substrates, but protopheophorbide a could not. Formationof methanol was detected during the enzymatic reaction, an indicationthat the enzyme is an esterase. Among seven alcohol analogstested, only methanol inhibited the enzymatic activity uncompetitively,with a K1 of 71.6 mM. Mass-spectrometric (MS) analysis of theprecursor yield a peak at m/z 579 that indicated the releaseof a methyl group from pheophorbide a. It appears thereforethat the enzyme catalyzes the demethylation of the carbomethoxygroup at C-132 of pheophorbide a by hydrolysis to yield methanoland the precursor, C-132-carboxyl-pyropheophorbide a, whichis converted to pyropheophorbide a by spontaneous decarboxylation.We have tentatively designated the enzyme "pheophorbidase".The presence of the enzyme was dependent on plant species andit was expressed constitutively. 1Present address: Faculty of Science, Shizuoka University, Ohya,Shizuoka, 422 Japan  相似文献

Current work at Old Peak (Ravenscar) on the northeast coastof England indicates that boulder-dwelling Lutonna saxatilis(Olvi) comprise at least two distinct forms, inhabiting high-and mid-shore boulders Using multivariate morphometrics we showthat these forms are significantly different in shape. Rareintermediates between these two distinct forms were occasionallyfound, usually with the low shore animals The two forms showdifferent reproductive strategies. High shore females (L. saxatilisH) mature at a smaller size and produce a smaller number oflarge eggs giving rise to large crawlaways The mid-shore form(L. saxatilis M) matures at a larger size and produces a largenumber of small eggs which give rise to small crawlaways. Anovel observation is that intermediate females (L. saxatilisI) contain eggs of both sizes, giving rise to a bimodal distributionof egg sizes within the brood pouch. The usual incidence ofabortion (observed in H and M forms) is Very low (<1%) However,in the intermediates the large eggs showed a high incidenceof abortion (about 60%) and the incidence of abortion in thesmall eggs was also increased (about 30%) Thus there appearsto be a partial reproductive barrier between the two forms,presumably resulting in reduced fitness due in the intermediatesto reproductive dysfunction. These findings extend to othershores in the region, over total distance of about 66 km. The carbon and nitrogen content of Noctiluca scintillans cellsfrom the Seto Inland Sea, Japan was investigated in order toestimate its biomass in natural samples. The carbon contentof N.scintillans ranged from 123 to 627 ng C cell–1 witha mean value of 353 ng C cell–1, or 1.12 to 2.67 fg Cµm–3 with a mean value of 1.98 fg C µm–3.The nitrogen content ranged from 36.0 to 232 ng N cell–1with a mean value of 131 ng N cell–1, or 0.499 to 0.910fg N µm–3 with a mean value of 0.694 fg N µm–3.Total cell carbon and nitrogen increased but the carbon andnitrogen per cell volume decreased with increasing cell volume.The C/N ratio of the cells ranged from 2.3 to 4.4, which wasrelatively low compared with the Redfield ratio. The carbonand nitrogen content was extremely low (91.2 ng C cell–1,41.8 ng N cell–1) for starved cells, whereas it was extremelyhigh (528 ng C cell–1, 205 ng N cell–1) for cellswhich had ingested the large diatom, Coscinodiscus wailesii.Our results suggest that the carbon and nitrogen content ofN.scintillans varies depending on its physiological conditionand the type of food that it has recently consumed.  相似文献
