Most of the biochemical, physiological and behavioural events in living organisms show diurnal fluctuations, normally synchronized with 24-h environmental rhythms, such as the light-dark cycle. The suprachiasmatic nucleus (SCN) of the hypothalamus is considered to be a pacemaker of the circadian rhythms in several mammals. The light-dark cycle is the primary synchronizing agent for many of the circadian rhythms which are regulated by the SCN. The photic information reaches the SCN also through a neuropeptide Y(NPY)-like immunoreactive pathway from the ventro-lateral geniculate nucleus. We found that in 12-h-dark and 12-h-light housed rats the NPY-like immunoreactive innervation of the ventro-lateral part of the SCN shows a 24 h rhythm with values rising gradually during the light phase and falling during the dark phase. Besides this rhythm, we found two peaks corresponding to the switching on and switching off of the light. The average level of NPY-like immunoreactivity, as assessed by means of semiquantitative immunocytochemistry and expressed in 'arbitrary units', is reduced in rats housed in total darkness for 2 weeks. These results confirm the physiological role of NPY in the timing of the circadian activity of the SCN.  相似文献

Two experiments were conducted to determine whether unilateral implantation of dihydrotestosterone propionate (DHTP) into different brain regions of castrated rats, bearing silastic capsules containing estradiol, could augment sexual behavior without appreciable leakage of androgen into the peripheral circulation. In Experiment 1 implanation of pulverized crystalline DHTP (via 25-gauge, 1-mm-long pellets) facilitated mating significantly without stimulating penile spine growth, provided the pellets were positioned in the lateral septum or medial amygdala. Insertion of DHTP pellets into the preoptic area-anterior hypothalamus, caudate-putamen, or the border of the substantia nigra and ventral tegmental nucleus or of cholesterol pellets into lateral septum or medial amygdala had no behavioral effects. Implanation of DHTP into the lateral septum also failed to activate penile erections in rats restrained in a supine position. In Experiment 2 implantation of different bone wax dilutions of DHTP (via 25-gauge, 1-mm-long pellets) into the preoptic area-anterior hypothalamus augmented males' sexual performance only in that group in which penile spine growth was also significantly stimulated. The results sugggst that 5α-reduced androgen is capable of activating mating in the male rat by acting locally in the lateral septum and/ or medial amygdala.  相似文献

Tyrosine transaminase activity in liver, kidney, intestine, stomach, skin, adipose tissue, striated muscle and brain in fed and 24-hour fasted rats, has been studied. Maximal activity has been found in liver, with only fractional activity in the other tissues. 24 hour fasting induced significant decrease in liver and adipose tissue activity, while no changes have been detected in the other tissues. The distribution pattern of neuropeptide Y-like immunoreactivity (NPY-Li) in cat hypothalamus was studied using avidin-biotin modification of immunocytochemical method. This study showed cell bodies containing NPY-Li in the periventricular and the infundibular nuclei and also a moderate number of neurons with NPY-Li in the ventromedial nucleus, an observation not reported in earlier studies. Fibers with NPY-Li were noted throughout the hypothalamus, but most prominently within the periventricular regions. The location of NPY cells within the hypothalamus suggests the possibility of an interaction with dopaminergic and other proopiomelanocortinergic neurons.  相似文献

Summary The pattern of distribution and reactivity of the neuropeptides vasopressin (AVP), vasoactive intestinal peptide (VIP), neuropeptide Y (NPY), substance P (SP), and thyrotropin-releasing hormone (TRH) were studied in the suprachiasmatic nucleus (NSC) of 20 Richardson's ground squirrels (and 7 European hedgehogs) of both sexes during hibernation and euthermia. The total area of immunostained structures revealed by application of the individual immunocytochemical techniques was measured by means of computer-aided image analysis. In both species, elements of all peptide systems examined were related to particular subdivisions of the NSC. The pattern of immunoreactivity was strongly correlated with the physiological stage of hibernation or euthermia both in ground squirrels and hedgehogs. The immunoreactivities to AVP and SP increased in area during hibernation (AVP: 25%; SP: 25%), whereas the respective area immunoreactive to NPY and VIP decreased (NPY: 45%; VIP: 100%) in comparison to nonhibernating controls. The sleep-wake cycle is virtually the most prominent circadian rhythm in mammals. In the timing system of sleep and wakefulness, the intrinsic neuropeptide systems of the suprachiasmatic nuclei (SCN) may play an important role. To elucidate this possible influence in the golden hamster, the immunoreactivity patterns of the suprachiasmatic gastrin-releasing peptide (GRP), vasoactive intestinal polypeptide (VIP), and arginine-vasopressin (AVP) systems were investigated in relation to the day-night and sleep-wake cycle by use of immunocytochemistry combined with semiquantitative planimetric analysis. For the GRP system, the highest level of immunoreactivity (expressed as area density) was observed in sleeping hamsters. Intermediate levels were found in awake, motorically active evening animals, whereas the lowest levels of immunoreactivity were detected in awake, motorically inactive hamsters studied in the morning. The immunoreactivity of the VIP system showed a completely opposite pattern, indicating highest area density in awake morning, intermediate area density in awake evening and lowest area density in sleeping golden hamsters. The immunoreactivity pattern of the AVP system, displaying highest levels in sleeping individuals, was virtually identical to that of the GRP system. Together with the related signs of neuronal activity, the present results favor an important role of these neuropeptide systems for the integration of central nervous information related to the sleep-wake cycle with photic information of the retinal input.  相似文献

A substance presenting a beta-casein-like immunoreactivity (beta-cas-LI) was found in male serum and tissues. This beta-cas-LI was measured by a specific radioimmunoassay using a purified beta-casein and an anti beta-casein kindly supplied by L. M. Houdebine. Serial dilutions of serum or tissues samples showed a good parallelism to the standard curve. In male rat, beta-cas-Li was found in all sera and in some tissues = mammary gland, prostate and testis which contained respectively 6,200, 408 and 38 ng per organ. beta-cas-LI value in the serum was regulatable. Hypophysectomy and long-term castration strongly reduced basal levels. Insertion of testosterone silastic capsules (25 mg/capsule) was able to restore control values, while estradiol (E2) (4 mg/kg E2 valerianate) to castrated or intact rats increased serum beta-cas-LI above basal values. In conclusion, these results showed, for the first time, the presence in male rat serum and tissues of a beta-casein immunoreactive-like substance. Moreover, this beta-cas-LI seemed to be under pituitary and gonad regulation.  相似文献

The distribution of neuropeptide Y (NPY) like immunoreactivity was investigated in the forebrain of the rat with immunohistochemical methods. Specificity of the antisera was established by the absence of all immunoreactive staining in tissue incubated in antisera which had been preabsorbed with the pure NPY antigen. NPY containing cells were distributed widely in the forebrain. These included neocortex, basal forebrain, septum, bed nucleus of the stria terminalis, neostratium, hypothalamic arcuate nucleus, and intergeniculate leaflet. This study also demonstrated an extensive network of NPY fibers in various areas of the forebrain such as the prepotic area, the hypothalamus and the paraventricular nucleus of the thalamus. The distribution of avian pancreatic polypeptide and NPY was compared, and the possible importance of NPY is briefly discussed.  相似文献

Melanin concentrating hormone (MCH) precursor-derived neuropeptide EI (NEI) has not yet been extensively studied. The aim of this study was to determine the effect of neuropeptide EI on serum levels of LH in normal male rats and chronically ovariectomized (CHR-OVX) female rats treated with estrogen benzoate (EB) and with a low dose of progesterone. The peptide, administered intracerebroventricularly in male and chronically ovariectomized female rats, increased LH serum levels compared to the controls injected with artificial cerebrospinal fluid. It is important to note that there is some relation between neuropeptide EI-melanin concentrating hormone and alpha-melanocyte stimulating hormone (alpha-MSH) indicating that all three peptides are associated in a complex inter-relationship. Therefore, the question that arises is if neuropeptide EI could also be related with the receptors for melanin concentrating hormone or alpha-melanocyte stimulating hormone.  相似文献

Summary Recent studies have accomplished the establishment of a collagenous fiber-fringe matrix upon dental root surfaces in vitro. The present study was undertaken to follow the development of such a matrix in vitro and to test the possible effects of root surface treatments upon this matrix. Periodontal ligament cells, 0.1 to 0.2-mm thick dental root discs, and alveolar bone cells were derived after extraction from four partially erupted third molars and the accompanying interradicular bony septa of 1 male patient. Autologous serum was obtained by venipuncture. Cultures were initiated by delivering a 1-ml suspension of 50000 tritiated thymidine-labeled periodontal ligament cells and 50000 alveolar bone cells onto each of 42 culture sets. The following day, demineralized or non-demineralized root discs treated with autologous serum, fibronectin or complete medium were placed in pairs, separated by a 0.1–1.0 mm gap, upon the initial cell layer. Representative cultures were terminated after 2, 3, 4, 5 and 6 weeks, and processed for light- and electron microscopy, morphometric analysis and autoradiography. An outstanding feature of the early cultures (2, 3 and 4 weeks) was a patchwise, random distribution of matrix making a precise developmental study impossible, although collagen fibrils were produced within the first 2 weeks. Some 3-week cultures already demonstrated a mature fiber-fringe characterized electron-microscopically as oriented, densely packed collagen fibrils closely abutting the cementum-lined root discs. The treatments (including autologous serum) used in this study had no appreciable morphologic or morphometric effect upon the fiber-fringe formed. Because none of the cultures in the present or past studies have demonstrated a true cementoid matrix, this model may not be suitable for the in-vitro study of cementum formation.  相似文献

The neonatal leptin surge, occurring from postnatal day (PND) 5 to 13 and peaking at PND9 in rodents, is important for the development of neuroendocrine circuits involved in metabolic control and reproductive function. We previously demonstrated that treatment with a leptin antagonist from PND 5 to 9, coincident with peak leptin levels in the neonatal surge, modified trophic factors and markers of cell turnover and neuronal maturation in the hypothalamus of peri-pubertal rats. The kisspeptin system and metabolic neuropeptide and hormone levels were also modified. Here our aim was to investigate if the timing of pubertal onset is altered by neonatal leptin antagonism and if the previously observed peripubertal modifications in hormones and neuropeptides persist into adulthood and affect male sexual behavior. To this end, male Wistar rats were treated with a pegylated super leptin antagonist (5 mg/kg, s.c.) from PND 5 to 9 and killed at PND102–103. The appearance of external signs of pubertal onset was delayed. Hypothalamic kiss1 mRNA levels were decreased in adult animals, but sexual behavior was not significantly modified. Although there was no effect on body weight or food intake, circulating leptin, insulin and triglyceride levels were increased, while hypothalamic leptin receptor, POMC and AgRP mRNA levels were decreased. In conclusion, alteration of the neonatal leptin surge can modify the timing of pubertal onset and have long-term effects on hypothalamic expression of reproductive and metabolic neuropeptides.  相似文献

These experiments were undertaken to investigate the effects of systemically administered neuropeptide Y (NPY) on gonadotropin secretion in the intact male rat and to determine whether the effects observed might be mediated by a direct action of NPY alone on the anterior pituitary gland (APG). Subcutaneous administration of 10 micrograms of NPY caused a greater than 2-fold increase in serum luteinizing hormone (LH) concentration at 15 min after injection but was without effect on serum follicle-stimulating hormone (FSH) or thyrotropin-stimulating hormone (TSH) levels. The addition of NPY (final concentrations of 10(-8) to 10(-11) M) or the structurally similar neuropeptide, rat pancreatic polypeptide, to culture medium containing hemi-APG did not alter the release of LH, FSH, or TSH. The results indicate that systemically administered NPY can elevate serum LH concentration in intact male rats. This effect does not appear to be due to NPY acting alone at the level of the APG.  相似文献

Using a porcine neuropeptide Y directed radioimmunoassay it was shown that acid extracts of human phaeochromocytoma tumour tissue contained a neuropeptide Y-like peptide. Further fractionation and purification of this immunoreactivity showed that this human neuropeptide Y-like immunoreactivity was closely similar in molecular size and separation characteristics to porcine neuropeptide Y. The possible contribution of neuropeptide Y to the hypertension characterizing human phaeochromocytoma is discussed.  相似文献

The occurrence of neuropeptide Y-like substances has been verified in the nervous system and alimentary tract of the ascidian Styela plicata. Neuropeptide Y-like immunoreactivity is present in a few small neurons and in a network of beaded nerve fibres of the cerebral ganglion. Neuropeptide Y-like immunoreactive material can be also localized in the endostyle and in a few cell bodies of the branchial walls. Moreover, immunofluorescent endocrine-like cells of the "open" type occur in the gastric folds. Finally, some possible functions of the ascidian neuropeptide Y are discussed.  相似文献

Summary In colchicine-pretreated cells of sympathetic ganglia, intensely NPY-immunoreactive material was localized within vacuoles and vesicles of the disorganized, widely dispersed Golgi apparatus. Intensely positive large granular vesicles, which are known to be one of major storage sites of various peptides in the autonomic nerve endings, were essentially unobserved in the perikaryal cytoplasm. The present finding provides evidence that one pool of NPY-like immunoreactivity is localized in the Golgi apparatus of colchicine-pretreated as well as normal sympathetic ganglion cells. It is also clear that visualization of NPY-immunoreactive somata by colchicine-pretreatment in the sympathetic ganglia is due to the accumulation of the neuropeptide in the disorganized Golgi stacks instead of increased amount of the large granular vesicles containing NPY.  相似文献

The invertebrate neuropeptide Y (NPY) homolog, neuropeptide F (NPF), has been characterized for a wide range of invertebrate phyla, including platyhelminthes, molluscs, and arthropods. Current hypotheses suggest that NPF may be capable of regulating responses to diverse external cues related to nutritional status and feeding. The qualitative and quantitative distribution of an NPF-like peptide in fifth instar Rhodnius prolixus was undertaken using an antiserum raised against Drosophila NPF. Immunohistochemistry reveals NPF-like immunoreactive neurons and processes in the central nervous system, stomatogastric nervous system and peripheral nervous system. The distribution of NPF-like immunoreactivity within the medial neurosecretory cells of the brain and neurohemal areas of the corpus cardiacum and dorsal vessel, suggests NPF may act as a neurohormone. Immunoreactive processes are present over the surface of the hindgut and the immunoreactivity in these processes is greatly reduced in intensity 24h post-feeding. The quantification of partially purified NPF-like material in the CNS of R. prolixus was conducted by HPLC fractionation and radioimmunoassay. The results suggest that NPF-like material is present in fifth instar R. prolixus and likely released into the hemolymph following a blood meal.  相似文献

Summary The septal region represents an important telencephalic center integrating neuronal activity of cortical areas with autonomous processes. To support the functional analysis of this brain area in the guinea pig, the afferent connections to the lateral septal nucleus were investigated by the use of iontophoretically applied horseradish peroxidase (HRP). Retrogradely labeled perikarya were located in telencephalic, diencephalic, mesencephalic and metencephalic sites. The subnuclei of the lateral septum (pars dorsalis, intermedia, ventralis, posterior) receive afferents from the (i) medial septal nucleus, diagonal band of Broca (pars horizontalis and pars ventralis), and the principal nucleus of the stria terminalis, the hippocampus, and amygdala (nucleus medialis); (ii) the medial habenular nucleus, and the para- (peri-) ventricular, parataenial and reuniens nuclei of the thalamus; the anterior, lateral and posterior hypothalamic areas in particular, the medial and lateral preoptic, suprachiasmatic, periventricular, paraventricular, arcuate, premammillary, and supramammillary nuclei; (iii) the periaquaeductal grey, ventral tegmental area, nucleus interfascicularis, nucleus reticularis linearis, central linear nucleus, interpeduncular nucleus; (iv) dorsal and medial raphe complex, and locus coeruleus. Each subnucleus of the lateral septum displays an individual, differing pattern of afferents from the above-described regions.
