Shoot and root growth are differentially sensitive to water stress. Interest in the involvement of hormones in regulating these responses has focused on abscisic acid (ABA) because it accumulates in shoot and root tissues under water-limited conditions, and because it usually inhibits growth when applied to well-watered plants. However, the effects of ABA can differ in stressed and non-stressed plants, and it is therefore advantageous to manipulate endogenous ABA levels under water-stressed conditions. Studies utilizing ABA-deficient mutants and inhibitors of ABA synthesis to decrease endogenous ABA levels, and experimental strategies to circumvent variation in plant water status with ABA deficiency, are changing the view of the role of ABA from the traditional idea that the hormone is generally involved in growth inhibition. In particular, studies of several species indicate that an important role of endogenous ABA is to limit ethylene production, and that as a result of this interaction ABA may often function to maintain rather than inhibit shoot and root growth. Despite early speculation that interaction between these hormones may influence many of the effects of water deficit, this topic has received little attention until recently.  相似文献

The phytohormone abscisic acid (ABA) plays a major role in regulating root growth. Most work to date has investigated the influence of root‐sourced ABA on root growth during water stress. Here, we tested whether foliage‐derived ABA could be transported to the roots, and whether this foliage‐derived ABA had an influence on root growth under well‐watered conditions. Using both application studies of deuterium‐labelled ABA and reciprocal grafting between wild‐type and ABA‐biosynthetic mutant plants, we show that both ABA levels in the roots and root growth in representative angiosperms are controlled by ABA synthesized in the leaves rather than sourced from the roots. Foliage‐derived ABA was found to promote root growth relative to shoot growth but to inhibit the development of lateral roots. Increased root auxin (IAA) levels in plants with ABA‐deficient scions suggest that foliage‐derived ABA inhibits root growth through the root growth‐inhibitor IAA. These results highlight the physiological and morphological importance, beyond the control of stomata, of foliage‐derived ABA. The use of foliar ABA as a signal for root growth has important implications for regulating root to shoot growth under normal conditions and suggests that leaf rather than root hydration is the main signal for regulating plant responses to moisture.  相似文献

在田间持水量分别保持于35%、55%和75%±5%的土壤水分条件下,利用盆栽实验研究了土壤干旱和氮素营养对玉米内源激素和气孔导度的影响.结果表明,土壤干旱下氮素营养明显降低了玉米根系木质部汁液ABA浓度,而正常供水下施氮处理间则无显著差异(施氮处理仍较低),同时测定的叶片ABA浓度则呈相反的变化趋势,表现为干旱下施氮处理要高于不施氮处理;施氮处理木质部汁液中ZRs浓度应低于相应的不施氮处理,在调控气孔行为方面并未表现拮抗ABA作用;3种土壤水分条件下,施氮玉米叶片的气孔导度均高于不施氮处理,与木质部汁液ABA浓度呈负相关,说明施氮处理较低的根源ABA浓度是导致其气孔导度较大的主要原因.  相似文献

Roots of maize (Zea mays L.) seedlings continue to grow at low water potentials that cause complete inhibition of shoot growth. In this study, we have investigated the role of abscisic acid (ABA) in this differential growth sensitivity by manipulating endogenous ABA levels as an alternative to external applications of the hormone. An inhibitor of carotenoid biosynthesis (fluridone) and a mutant deficient in carotenoid biosynthesis (vp 5) were used to reduce the endogenous ABA content in the growing zones of the primary root and shoot at low water potentials. Experiments were performed on 30 to 60 hour old seedlings that were transplanted into vermiculite which had been preadjusted to water potentials of approximately −1.6 megapascals (roots) or −0.3 megapascals (shoots). Growth occurred in the dark at near-saturation humidity. Results of experiments using the inhibitor and mutant approaches were very similar. Reduced ABA content by either method was associated with inhibition of root elongation and promotion of shoot elongation at low water potentials, compared to untreated and wild-type seedlings at the same water potential. Elongation rates and ABA contents at high water potential were little affected. The inhibition of shoot elongation at low water potential was completely prevented in fluridone-treated seedlings during the first five hours after transplanting. The results indicate that ABA accumulation plays direct roles in both the maintenance of primary root elongation and the inhibition of shoot elongation at low water potentials.  相似文献

A glasshouse experiment was conducted with plants of Phaseolus grown in liquid culture. Root growth parameters (biomass, diameter, length, growth rate, zone of cell division), root rheological components (wall extensibility, water potential yield threshold, water potential), shoot growth, carbon allocation, and abscisic acid (ABA) concentration were measured in Phaseolus acutifolius A. Gray at ambient (550 μmol mol-1) and elevated (700 μmol mol-1) atmospheric CO2 concentrations. For contrast, measurements of above- and belowground growth were conducted on Phaseolus vulgaris L. in the same treatments. Under nonlimiting conditions of water and nutrients, elevated CO2 increased root and shoot growth of P. acutifolius but not P. vulgaris. While root mass was increased by nearly 60% in P. acutifolius, there was no effect of atmospheric CO2 on any of the rheological components measured. In contrast, starch and ABA accumulated in roots of P. acutifolius. The concentration of starch in roots of P. acutifolius increased by 10-fold, while root concentrations of ABA doubled. From the data it is concluded that CO2 enrichment is favorable for root growth in some species in that more carbon is allocated to belowground growth. In addition, ABA may play a role in growth responses and/or allocation of photosynthates at elevated CO2 in P. acutifolius.  相似文献

在土壤干旱情况下,根源信号一方面向植物地上部分的长距离传输,为地上部分提供了土壤水分获取能力的测度,另一方面调控气孔开度,抑制蒸腾作用并提高植物的水分利用效率.文中综述了根源信号参与调控植物水分利用的生理机制和理论模型,指出该模型与根系吸水模型、气孔导度模型耦合,能够更好地反映植物叶片对土壤干旱以及大气干旱的响应、评述了在根源信号参与调控植物水分关系的基础上发展的调亏灌溉(RDI)、部分根系干旱(PRD)和控制性交替灌溉(CAI)等有效灌溉手段,有助于合理配置根系层供水量,通过根土相互作用和信号物质的传输,降低蒸腾和提高水分利用效率、另外,根源信号在调控根系生长发育、延缓地上部分生长以调节根冠比例,优化资源分配以利于生殖生长等方面均有所为,为全面提高农田水分利用效率提供节水生理基础。  相似文献

Wheat germ agglutinin is found in wheat embryos and a similar lectin is present in the roots of older plants. We report here that 10 micromolar abscisic acid (ABA) produces an average two to three-fold enhancement in the amount of lectin in the shoot base and the terminal portion of the root system of hydroponically grown wheat seedlings. Although ABA stunts seedling growth, a similar growth inhibition produced by ancymidol is not accompanied by elevated lectin levels. To further clarify the role of ABA, wheat callus cultures were employed. Callus derived from immature embryos was grown on growth medium containing various combinations of ABA and 2,4-dichlorophenoxyacetic acid. Those grown in the presence of 10 micromolar ABA exhibit the largest increases in lectin compared to material grown on other regimes. The involvement of ABA in lectin accumulation was further probed with fluridone, an inhibitor of carotenoid synthesis which has also been linked to depressed levels of endogenous ABA. Wheat seedlings grown in the presence of 1 or 10 milligrams per liter fluridone have few or no carotenoids, and wheat germ agglutinin levels in the shoot base and roots are lower compared to controls. The greatest effect (a 39% reduction in the shoot base) is produced at an herbicide concentration of 10 milligrams per liter. Exogenous 10 micromolar ABA greatly stimulates lectin accumulation in the presence of fluridone, but the levels are not as high as those produced by ABA alone. These results indicate that lectin synthesis is under ABA control in both wheat embryos and adult plants.  相似文献

Previous studies have reported that a purified sedimentary humic acid (PHA) was able to increase the concentration of nitric oxide (NO), indole-acetic acid (IAA) and ethylene in cucumber roots. Here, we investigated if these effects are functionally related to the ability of PHA to improve shoot growth. The effect of specific inhibitors of NO, IAA and ethylene functionality and signaling on PHA-induced shoot growth was studied. Likewise, the effect of these inhibitors on the synthesis and activity of the phytoregulators concerned by PHA action in cucumber roots was also explored. The results show that shoot growth promoted by PHA is due to an increase of IAA concentration in the root through both a NO-dependent and a NO-independent pathway. In addition, the increased ethylene production in the root is regulated by an IAA-dependent pathway. Finally, results also showed that the increase of ABA concentration in the root is regulated through both IAA- and ethylene-dependent pathways. In summary, the shoot growth promoting action of PHA involves a complex hormonal network. On one hand, the PHA action is functionally linked to increments in NO and IAA concentration in roots. And on the other hand, PHA action also increases ethylene and ABA root concentration mediated by NO-IAA dependent pathways.  相似文献

We studied the possible involvement of ABA in the control of water relations under conditions of increased evaporative demand. Warming the air by 3°C increased stomatal conductance and raised transpiration rates of hydroponically grown Triticum durum plants while bringing about a temporary loss of relative water content (RWC) and immediate cessation of leaf extension. However, both RWC and extension growth recovered within 30 min although transpiration remained high. The restoration of leaf hydration and growth were enabled by increased root hydraulic conductivity after increasing the air temperature. The use of mercuric chloride (an inhibitor of water channels) to interfere with the rise on root hydraulic conductivity hindered the restoration of extension growth. Air warming increased ABA content in roots and decreased it in shoots. We propose this redistribution of ABA in favour of the roots which increased the root hydraulic conductivity sufficiently to permit rapid recovery of shoot hydration and leaf elongation rates without the involvement of stomatal closure. This proposal is based on known ability of ABA to increase hydraulic conductivity confirmed in these experiments by measuring the effect of exogenous ABA on osmotically driven flow of xylem sap from the roots. Accumulation of root ABA was mainly the outcome of increased export from the shoots. When phloem transport in air-warmed plants was inhibited by cooling the shoot base this prevented ABA enrichment of the roots and favoured an accumulation of ABA in the shoot. The comparative study of shoot and root growth was carried out, and the level of ABA therein determined in the mutant af and tl and wild-type isogenic lines of pea. The recessive af mutation transformed the leaflets into tendrils, and the tl mutation transformed the tendrils into leaflets. These mutations did not affect the length and number of internodes. In all plants, the level of ABA in the leaves was 3–10 times greater than in the roots, and in the course of vegetative growth it rose in both organs. An increase in the shoot area of tl mutant did not change the dry weight of underground and above-ground parts; therefore, the ratio shoot/root in the mutant was identical to that in the wild-type plants. The maintenance of shoot dry weight in the tl mutant at the level of wild-type plant while its area considerably increased was accounted for by a decrease in the thickness of the leaflet and stipule blades. The level of ABA in the stipules of mutant plants was greater than in the wild-type plants. A decrease in the shoot area in the af mutant brought about a decline in its dry weight; however, the ratio root/shoot was maintained at the wild-type level due to a reduced accumulation of dry weight by the root. The level of ABA in the roots of the af mutant was twice greater than in the leafy forms. ABA was assumed to participate in the control over the root growth exerted by the shoot. The absence of leaflets in the af plants was partially compensated for by expanding stipules. The level of ABA therein was three times higher than in the plants of wild type and comparable with the level in the leaflets of the tl mutant and in the wild-type plants. The role of ABA in the growth and final size of leaf blades is discussed.  相似文献

Abstract. It is now clear that drying of the soil does not always result in an early change in shoot water status. This may be because stomata close and leaf growth slows to reduce water loss. When this is the case, it is necessary to ask how the change in soil water status has been 'sensed'by the shoot. The current view is that soil drying results in some type of chemical signalling between roots and shoots. The sensitivity of the response and experiments involving the manipulation of small parts of root systems suggest that the signalling involves more than a simple change in root activity in response to soil drying. In this paper, we consider the evidence for chemical signalling between roots and shoots and discuss the possible candidates for such signals. In some plants, root-sourced ABA can apparently influence shoot physiology and growth in the absence of any perturbation of shoot water relations. The ABA produced is quantitatively sufficient to account for the responses observed. Applied ABA can mimic many of the effects of soil drying on plants, including effects at the plasma membrane and on gene expression. Perhaps uniquely, ABA seems to be involved in signalling between different plant organs, and in signalling at the transmembrane and genome levels. We review the effects of ABA on leaf cells with a view to gaining some understanding of how soil drying may influence plant development.  相似文献

Underground roots normally reside in darkness. However, they are often exposed to ambient light that penetrates through cracks in the soil layers which can occur due to wind, heavy rain or temperature extremes. In response to light exposure, roots produce reactive oxygen species (ROS) which promote root growth. It is known that ROS‐induced growth promotion facilitates rapid escape of the roots from non‐natural light. Meanwhile, long‐term exposure of the roots to light elicits a ROS burst, which causes oxidative damage to cellular components, necessitating that cellular levels of ROS should be tightly regulated in the roots. Here we demonstrate that the red/far‐red light photoreceptor phytochrome B (phyB) stimulates the biosynthesis of abscisic acid (ABA) in the shoots, and notably the shoot‐derived ABA signals induce a peroxidase‐mediated ROS detoxification reaction in the roots. Accordingly, while ROS accumulate in the roots of the phyb mutant that exhibits reduced primary root growth in the light, such an accumulation of ROS did not occur in the dark‐grown phyb roots that exhibited normal growth. These observations indicate that mobile shoot‐to‐root ABA signaling links shoot phyB‐mediated light perception with root ROS homeostasis to help roots adapt to unfavorable light exposure. We propose that ABA‐mediated shoot‐to‐root phyB signaling contributes to the synchronization of shoot and root growth for optimal propagation and performance in plants.  相似文献

The phytohormone abscisic acid (ABA) has been proposed to act as a mediator in plant responses to a range of stresses, including salt stress. Most studies of ABA response apply ABA as a single dose. This may not resemble the prolonged increasing endogenous ABA levels that can occur in association with slowly increasing salinity stresses in nature or field situations. Salt stress response based on method of ABA application was examined in four potato genotypes of varying salt stress resistance: the sensitive ABA-deficient mutant and its normal sibling, a resistant genotype line 9506, and commercial cultivar ‘Norland’ of moderate resistance. ABA was applied by root drench at 0, 50, 75, or 100 μM concentrations through a single dose, or by slowly increasing multiple ABA doses in a sand-based growing system under greenhouse conditions. Salt tolerance was then evaluated after 2 weeks of exposure to 150–180 mM NaCl stress. The method of ABA application had a marked effect on the responses to salt stress. Plant responses to the method of ABA application were differentiated according to (1) growth rate, (2) root water content, and (3) apparent shoot growth response. Under a single dose, growth rate increased in all genotypes under salt stress, whereas slowly increasing multiple ABA applications generally maintained stable growth rates except in the ABA-deficient mutant where there was an upward growth trend. Percent root water content was elevated only under slowly increasing multiple ABA doses in two genotypes, whereas none of the single-dose treatments induced any change. The single ABA dose enhanced vertical growth, whereas the slowly increasing multiple ABA dose applications enhanced lateral shoot growth. Because exogenous application is still an artificial system, endogenous ABA was supplied through grafting of ABA-deficient mutant scions onto rootstocks with known elevated ABA levels. Multiple exogenous ABA applications as low as 50 μM elicited similar shoot water content responses as grafting treatments without ABA application in the mutant genotype but had no effect on the ABA normal sibling. Shoot dry weight was significantly increased through grafting over all exogenous ABA treatments. Our study further indicates that the method of ABA application regime in itself can alter plant responses under salt stress and that certain application regimes may reflect responses to elevated endogenous levels of ABA.  相似文献

The research aim was to assess the effects of the plant hormone abscisic acid (ABA) and the growth regulator paclobutrazol (PBZ) on root system development during the in vitro culture of different birch and aspen genotypes. The studied genotypes involved two aspen (Populus tremula and Populus tremuloides × P. tremula) and two silver birch (Betula pendula) trees, with one of the birches characterized by its inability to root in vitro. For experiments, apical shoot segments were cultured on nutrient medium enriched with either ABA or PBZ. Additionally, the analysis of the endogenous hormones in shoots developed on hormone‐free medium was conducted by high‐performance liquid chromatography. The endogenous concentration of auxin indole‐3‐acetic acid was much higher in the aspens than that in the birches, while the highest concentration of ABA was found in the root‐forming birch. The culturing of this birch genotype on medium enriched with ABA resulted in an increased root length and a higher number of lateral roots without any negative effect on either shoot growth or adventitious root (AR) formation, although these two processes were largely inhibited by ABA in the aspens. Meanwhile, PBZ promoted AR formation in both aspen and birch cultures but impaired secondary root formation and shoot growth in birches. These results suggest the use of ABA for the in vitro rooting of birches and PBZ for the rooting of aspens.  相似文献

To examine whether the reduced shoot growth of abscisic acid (ABA)-deficient mutants of tomato is independent of effects on plant water balance, flacca and notabilis were grown under controlled-humidity conditions so that their leaf water potentials were equal to or higher than those of well-watered wild-type plants throughout development. Most parameters of shoot growth remained markedly impaired and root growth was also greatly reduced. Additional experiments with flacca showed that shoot growth substantially recovered when wild-type levels of ABA were restored by treatment with exogenous ABA, even though improvement in leaf water potential was prevented. The ability of applied ABA to increase growth was greatest for leaf expansion, which was restored by 75%. The ethylene evolution rate of growing leaves was doubled in flacca compared to the wild type and treatment with silver thiosulphate to inhibit ethylene action partially restored shoot growth. The results demonstrate that normal levels of endogenous ABA are required to maintain shoot development, particularly leaf expansion, in well-watered tomato plants, independently of effects on plant water balance. The impairment of shoot growth caused by ABA deficiency is at least partly attributable to ethylene.  相似文献

Plants undergo cell division throughout their life in order to maintain their growth. It is well known that root and shoot tip of plants possess meristems, which contain quiescent cells. Fluridone (1-methyl-3-phenyl-5-(3-trifluoromethyl (phenyl))-4-(1H)-pyridinone) is an established inhibitor of both ABA and carotenoid biosynthesis. However, the other functions of fluridone remain undiscovered. In this report, we provide experimental evidence that fluridone plays a role in the division of the quiescent centre of the Arabidopsis root meristem. This study examined the effects of exogenous fluridone and ABA on the development of the stem cell niche in Arabidopsis root. We show that fluridone promoted the division of stem cells in the quiescent centre, whereas exogenous ABA suppressed quiescent centre division. Furthermore, we established a novel regulatory function for fluridone by demonstrating that it plays an important role in postembryonic development.  相似文献

Roots often continue to elongate while shoot growth is inhibited in plants subjected to low-water potentials. The cause of this differential response to water deficit was investigated. We examined hypocotyl and root growth, polysome status and mRNA populations, and abscisic acid (ABA) content in etiolated soybean (Glycine max [L.] Merr. cv Williams) seedlings whose growth was inhibited by transfer to low-water potential vermiculite or exogenous ABA. Both treatments affected growth and dry weight in a similar fashion. Maximum inhibition of hypocotyl growth occurred when internal ABA levels (modulated by ABA application) reached the endogenous level found in the elongating zone of seedlings grown in water-deficient vermiculite. Conversely, root growth was affected to only a slight extent in low-water potential seedlings and by most ABA treatments (in some, growth was promoted). In every seedling section examined, transfer of seedlings into low-water potential vermiculite caused ABA levels to increase approximately 5- to 10-fold over that found in well-watered seedlings. Changes in soluble sugar content, polysome status, and polysome mRNA translation products seen in low-water potential seedlings did not occur with ABA treatments sufficient to cause significant inhibition of hypocotyl elongation. These data suggest that both variation in endogenous ABA levels, and differing sensitivity to ABA in hypocotyls and roots can modulate root/shoot growth ratios. However, exogenous ABA did not induce changes in sugar accumulation, polysome status, and mRNA populations seen after transfer into low-water potential vermiculite.  相似文献

The growth-inhibiting effects of indole-3-acetic acid (IAA) at high concentration and the synthetic auxins 7-chloro-3-methyl-8-quinolinecarboxylic acid (quinmerac), 2-methoxy-3,6-dichlorobenzoic acid (dicamba), 4-amino-3,6, 6-trichloropicolinic acid (picloram), and naphthalene acetic acid, were investigated in cleavers (Galium aparine). When plants were root treated with 0.5 mM IAA, shoot epinasty and inhibition of root and shoot growth developed during 24 h. Concomitantly, 1-aminocyclopropane-1-carboxylic acid (ACC) synthase activity, and ACC and ethylene production were transiently stimulated in the shoot tissue within 2 h, followed by increases in immunoreactive (+)-abscisic acid (ABA) and its precursor xanthoxal (xanthoxin) after 5 h. After 24 h of treatment, levels of xanthoxal and ABA were elevated up to 2- and 24-fold, relative to control, respectively. In plants treated with IAA, 7-chloro-3-methyl-8-quinolinecarboxylic acid, naphthalene acetic acid, 2-methoxy-3,6-dichlorobenzoic acid, and 4-amino-3,6,6-trichloropicolinic acid, levels of ethylene, ACC, and ABA increased in close correlation with inhibition of shoot growth. Aminoethoxyvinyl-glycine and cobalt ions, which inhibit ethylene synthesis, decreased ABA accumulation and growth inhibition, whereas the ethylene-releasing ethephon promoted ABA levels and growth inhibition. In accordance, tomato mutants defective in ethylene perception (never ripe) did not produce the xanthoxal and ABA increases and growth inhibition induced by auxins in wild-type plants. This suggests that auxin-stimulated ethylene triggers ABA accumulation and the consequent growth inhibition. Reduced catabolism most probably did not contribute to ABA increase, as indicated by immunoanalyses of ABA degradation and conjugation products in shoot tissue and by pulse experiments with [(3)H]-ABA in cell suspensions of G. aparine. In contrast, studies using inhibitors of ABA biosynthesis (fluridone, naproxen, and tungstate), ABA-deficient tomato mutants (notabilis, flacca, and sitiens), and quantification of xanthophylls indicate that ABA biosynthesis is influenced, probably through stimulated cleavage of xanthophylls to xanthoxal in shoot tissue.  相似文献

We describe the involvement of abscisic acid (ABA) in the control of differential growth of roots and shoots of nutrient limited durum wheat plants. A ten-fold dilution of the optimal concentration of nutrient solution inhibited shoot growth, while root growth remained unchanged, resulting in a decreased shoot/root ratio. Addition of fluridone (inhibitor of ABA synthesis) prevented growth allocation in favour of the roots. This suggests the involvement of ABA in the redirecting of growth in favour of roots under limited nutrient supply. The ABA content was greater in shoots and growing apical root parts of starved plants than in nutrient sufficient plants. Accumulation of ABA in shoots of nutrient deficient plants was linked to a decrease in leaf turgor. Increased flow of ABA in the phloem apparently contributed to the accumulation of ABA in the apical part of the roots. Thus, partitioning of growth between roots and shoots of wheat plants limited in mineral nutrients appears to be modulated by accumulation of ABA in roots. This ABA may originate in the shoots, where its synthesis is stimulated by the loss of leaf turgor.  相似文献

The effects of heating at 38°C of whole cucumber (Cucumis sativus L.) seedlings or local heating of their shoots and roots on ABA content and heat tolerance of leaves and roots were investigated. During the initial period of the high-temperature treatment of whole seedlings, the ABA concentration in leaves and roots increased considerably. Local heating of the shoot or root resulted in an increase in the ABA concentration not only in the heated organ, but also in unheated seedling parts. A high-temperature treatment of the whole seedlings and the local treatment of shoots or roots caused an increase in the heat tolerance of leaf cells. The heat tolerance of root cells virtually did not change after heating of the whole seedlings or shoots, but decreased after heating of roots. The possible role of ABA in changing the heat tolerance of leaf and root cells by local heating of the seedling is discussed.  相似文献
