在不同的高温胁迫条件下 ,对湿地匍灯藓 (Plagiomnium acutum)和大羽藓 (Thuidium cymbifolium)过氧化物酶 (POD)活性及其与处理时间和处理温度的关系进行了初步研究。结果表明 ,在一定的温度范围内 ,随着温度的升高 ,POD活性增加 ,二者成线形关系。在一定温度条件下 ,一般随着处理时间的延长 ,POD活性增加。但是当超过一定的温度 (4 5~ 5 0°C)以及一定的处理时间 (4~ 6h) ,POD活性有所下降。结果还表明 ,湿地匍灯藓的POD活性显著高于大羽藓。而且在高温胁迫下 ,湿地匍灯藓 POD活性变化比大羽藓活跃 ,其变化幅度也比大羽藓大。  相似文献

The activity and spectrum of proteinases in the intestines of host fishes change upon infestation with cestodes. Serine proteinases are found to make a greater contribution to the total proteolytic activity. The reduction of proteolytic activity is associated with adsorption of the enzymes of the host on the surface of cestodes, and the increase in the activity is caused by the injury of the intestinal mucosa by the attachment apparatuses of cestodes. The inhibition of proteainase activity indicates the possible participation of microbiota enzymes in protein hydrolyses.  相似文献

研究了不同肥力水平的宫川蜜柑根际土壤酶的活性及其与土壤农化特性的关系。结果表明 :高产园的土壤酶活性显著高于低产园的土壤酶活性。经统计分析 ,土壤酶活性与养分含量均呈极显著相关。而且酶的活性在土壤中的分布有一定的规律性。其水平分布是在树冠内半径的 4 /5处至树冠滴水线范围内 ,酶的活性最高 ,由此处向内向外酶的活性逐渐降低 ;其垂直分布是 0~ 2 0 cm土层酶的活性最高 ,随土层的加深而逐渐降低  相似文献

A histochemical study has been carried out on the eye muscles of the carp. On the base of the ATP-ase and SDH activity and with regard to the localization and diameter of muscle fibres six types of muscle fibres situated in the defined zones can be distinguished. The type 1 and 2 fibres display a moderately high ATP-ase activity at pH 9.4 and rather low activity after alkaline and acid preincubation. Type 1 shows a high SDH activity in contrast to type 2 with a low SDH activity. The other four types of fibres have the high ATP-ase activity at pH 9.4. Type 3 contains fibres with a moderately high ATP-ase activity after alkaline preincubation with a rather low activity after acid preincubation and with a low SDH activity. The fibres of type 4 characterized by the high ATP-ase activity after alkaline and acid preincubation and by the high SDH activity. The fibres of type 5 display high ATP-ase activity after alkaline and acid preincubation and the low SDH activity. They are situated in the white and intermediate fibre zones. The fibres of type 6 are comparable to the fibres of type 5, however they differ diameter and localization, i.e. they are situated in small diameter fibre zone. Using the electron microscope four types of fibres (A, B, C, and D) are found. They vary in the localization of T system, the organization of Z-line and in M-line appearance. In the type B of muscle fibres two different localizations of T system have been discerned.  相似文献

A genetic polymorphism of the purine nucleoside phosphorylase is described in red cells of cattle. Electrophoresis distinguishes two types, one associated with a high activity (H), the other with a low activity (L). Breeding tests suggest the segregation of two alleles: a dominant allele for high activity ( H ) and a recessive allele for low activity (L). On the basis of quantitative estimation of enzyme activity, the individuals of the H phenotype can be separated in two classes, the enzyme activity in one class being twice as high as the activity in the other class. It is suggested that the class with the highest enzymatic activity corresponds to the homozygote for the H allele, the other class to the heterozygote. In fact, an A.I. bull (H phenotype and with the highest enzymatic activity) mated with cows of the L phenotype, gave offspring of the H phenotype, only. The decline of activity, with age, is stressed. Four breeds have been typed. The Charolais breed is characterized by a high frequency of the H allele.  相似文献

In the previous work we have found two peaks of catalase activity at acid and neutral pH in partially destroyed bacteria E. coli K12 KS400. The present study indicates that catalase activity with two pH-optimums is sensitive to pH of cultivation medium. The relative catalase activity of frozen-thawed bacteria preparations measured at pH 3.5 increased two-fold and activity measured at pH 7.0 didn't change by shift of medium pH from value 5.5 to 7.0. In analogical preparations of bacteria grown in slightly alkaline media activity with acid maximum was not observed, but activity with neutral maximum rose to 130% in comparison with the intact cells was revealed. Two peaks of activity differed in their sensitivity to bacteria destruction, heating, inhibition by NaN3 and AMT, oxidative stress. The analysis of recent literature information and experimental data leads us to conclude that the activity with neutral pH-optimum consists of two known catalase forms HPI and HPII in E. coli. The ratio of HPI and HPII is 70 and 30%, respectively what was concluded from inhibition of catalase activity with neutral pH-optimum by AMT. Properties of catalase activity with acid pH-optimum didn't corresponding to any known enzyme forms. It is suggested the activity measured at pH 3.5 is results of some unstable activator which acts in acid pH range. It is possible that the described activity with acid pH-optimum is specific for the used E. coli strain. Investigation of another strain of E. coli K12 AB1157 confirmed this idea where the activity peak with acid pH-optimum was not detected.  相似文献

The activity of S-adenosylmethionine decarboxylase in rat liver homogenates is localized chiefly in the crude nuclear fraction, probably associated with membrane fragments, with the remainder in the supernatant fraction. This distribution is not paralleled by the activity of the cytoplasmic enzyme, lactate dehydrogenase. The spermidine-synthesizing activity of whole homogenate is recovered entirely in the supermidine-synthesizing activity of whole homogenate is recovered entirely in the supernatant fraction. Measurement of various kinetic parameters in crude fractions provided not positive evidence for isozymes of S-adenosylmethionine decarboxylase. Some species do not possess a sedimentable fraction of S-adenosylmethionine decarboxylase activity in liver. In those species all activity present in the whole homogenate of liver is released into the supernatant fraction.  相似文献

α-Chymotrypsin (CT) was lyophilized from an aqueous solution in the presence of hydroxypropyl-β-cyclodextrin (HP-β-CyD). The enzyme preparation was used as a catalyst for transesterification between N-acetyl-l-tyrosine ethyl ester and methanol in a mixed solvent of acetonitrile/water (97/3 (v/v)). The enzyme preparation had much higher catalytic activity than free CT. The activity increased with an increase of HP-β-CyD/CT ratio and reached a maximum activity at the weight ratio of 4. Also, the activity of HP-β-CyD/CT increased with an increase in water content in the reaction media, and the maximum activity was obtained at 5–10% water. The fluorescence spectroscopic analysis suggested that the co-lyophilization with HP-β-CyD increased the structural stability of CT in acetonitrile/water. Upon co-lyophilization with HP-β-CyD, the activity of CT increased in any of the solvents used, but the activity depended strongly on the nature of the organic solvents. The catalytic activity of subtilisin Carlsberg (STC) also increased by co-lyophilization with α-, β-, γ-CyD or tri-O-methyl-β-CyD. α-CyD gave the best result, while HP-β-CyD diminished the activity of STC.  相似文献

The DNA-dependent RNA polymerase activity of isolated nuclei from human peripheral blood has been shown to increase following stimulation with phytohaemagglutinin (PHA). Using the toxin α-amanitin it has been possible to demonstrate that within 4 h of the addition of PHA there is a two-fold increase in the amanitin-resistant polymerase activity (polymerase A) with little increase in the sensitive polymerase activity (polymerase B). 24 h following PHA stimulation the amanitin-resistant activity is stimulated 4–5 fold and the amanitin-sensitive activity less than two-fold. The susceptibility of this increased amanitin-resistant activity to low doses of actinomycin D both in vivo and in vitro indicates that the amanitin-resistant enzyme is mainly engaged in ribosomal RNA precursor synthesis. These changes in DNA-dependent RNA polymerase activity closely correspond to the observed changes in ribosomal and non-ribosomal RNA synthesis following lymphocyte stimulation.The increased polymerase A activity is diminished by a 1 h incubation of the cells with cycloheximide added 24 h after PHA whereas polymerase B activity remains unaffected. This indicates that the polymerase A activity observed after transformation is dependent on continuing protein synthesis.In our incubation conditions the polymerase activity observed in isolated nuclei appeared to be almost wholly attributable to elongation of nascent RNA molecules attached to the endogenous DNA template.  相似文献

The intra-acinar distribution pattern of phosphoenolpyruvate carboxykinase activity (PEPCK) was investigated in microdissected samples of livers from normal, castrated, castrated and estradiol- or testosterone-treated, and uncastrated and testosterone- or estradiol-treated male and female rats. The total PEPCK activity showed a marked sex dependency, with 1.8 times higher activity in males. The intra-acinar distribution profiles were also sex-dependent. The periportal-to-perivenous gradient was steeper in males. Castration resulted in an approximation of PEPCK activity and its acinar distribution pattern between the sexes due to a reduction in males and an increase in females. Estrogen treatment of castrated males had no further effect on PEPCK activity and its acinar gradient, whereas in ovariectomized animals the activity was reduced to levels near normal. Testosterone treatment of castrated male or female animals led to a marked increase in enzyme activity with a concomitant steepening of the acinar gradient. Administration of estradiol to normal male rats also led to a reduction in activity, together with a change in the acinar activity gradient. Testosterone treatment of normal females resulted in an induction of PEPCK activity which was most prominent in the periportal zone. The most drastic changes were observed in the perivenous zones. In all experiments a periportal-to-perivenous activity gradient persisted thus marking the periportal zone as the area with highest gluconeogenic capacity.  相似文献

Diacylglycerol kinase activity is found in both adult and fetal lung. Approximately 27 and 52% of the total activity is found in microsomes and cytosol, respectively. The activity is maximal at pH 7.4. The apparent Km for ATP is 0.11 mM and 0.21 mM for cytosol and microsomes, respectively. The apparent Km for dioleoylglycerol is 0.05 mM for cytosol and 0.14 for microsomes. Maximal activity in cytosol and microsomes is obtained with 2.0 mM dexoycholate. Other detergents cannot substitute for deoxycholate. Phosphatidylglycerol stimulates activity in the absence and in the presence of deoxycholate. Phosphatidylserine also stimulates activity, whereas phosphatidylethanolamine was inactive and phosphatidylcholine inhibited the reaction. Linoleic acid produced inhibition. The general properties of the enzyme were similar for fetal and adult lung. Diacylglycerol kinase from microsomes and cytosol fraction from both fetal and adult lung was most active with dioleoylglycerol and diacylglycerol from egg phosphatidylcholine. Significantly lower activity was obtained with dipalmitoylglycerol. Phosphatidylglycerol did not alter the relative substrate preferences. The activity in microsomes increased with development from 19 days gestation to a maximal activity at 21 days gestation. Maximal activity was about 2-fold higher than the adult. The activity dropped rapidly reaching adult values prior to birth (22 days gestation). The activity in cytosol fractions increased gradually from 19 days gestation, reaching adult values by 22 days gestation.  相似文献

AIMS: To investigate properties of the proteolytic activity of a yeast cell wall lytic soil bacterium identified as an Arthrobacter species. METHODS AND RESULTS: The organism was grown at pH 7.5 and 30 degrees C in shake flasks on media with different complex subtrates. Highest proteolytic activity assayed with azocaseine was detected in media with wheat gluten. In addition, l-leucine, l-alanine exopeptidase activity and esterase activity were found. The proteolytic activity showed stability up to pH 12, with a maximum at pH 11. The temperature optimum was at 55 degrees C, but there was a loss in enzyme activity of 50% within 2 h. The proteolytic activity was inhibited by 3,4-dichloroisocumarin, whereas there was little or no effect with EDTA, pepstatin A or E64. CONCLUSIONS: The proteolytic activity is highly alkaline stable. The formation of the enzyme can be induced by media with high protein content.  相似文献

The relationship between the early-age activity of Mediterranean fruit flies (medflies) or other fruit flies and their lifespan has not been much studied, in contrast to the connections between lifespan and diet, sexual signaling, and reproduction. The objective of this study is to assess intra-day and day-to-day activity profiles of female Mediterranean fruit flies and their role as biomarker of longevity as well as to explore the relationships between these activity profiles, diet, and age-at-death throughout the lifespan. We use advanced statistical methods from functional data analysis (FDA). Three distinct patterns of activity variations in early-age activity profiles can be distinguished. A low-caloric diet is associated with a delayed activity peak, while a high-caloric diet is linked with an earlier activity peak. We find that age-at-death of individual medflies is connected to their activity profiles in early life. An increased risk of mortality is associated with increased activity in early age, as well as with a higher contrast between daytime and nighttime activity. Conversely, medflies are more likely to have a longer lifespan when they are fed a medium-caloric diet and when their daily activity is more evenly distributed across the early-age span and between daytime and nighttime. The before-death activity profile of medflies displays two characteristic before-death patterns, where one pattern is characterized by slowly declining daily activity and the other by a sudden decline in activity that is followed by death.  相似文献

The antioxidant activity of wild Pueraria mirifica collected from 28 of the 76 provinces of Thailand and Pueraria lobata collected from China were assessed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. P. mirifica tuberous extracts showed weak antioxidant activity in comparison with alpha-tocopherol. Six plant samples exhibited stronger antioxidant activity than the mean value of the P. mirifica population. In addition, the mean value of the P. mirifica population indicated significantly lower antioxidant activity than P. lobata. The analysis of the antioxidant activity of isoflavonoids revealed that puerarin and daidzein exhibited the same level of antioxidant activity as alpha-tocopherol. The results showed convincingly that puerarin and daidzein in the plant tubers may play an important role in antioxidant activity. The correlation analysis between antioxidant activity and major isoflavonoid contents of plant tubers indicated a significant correlation only with puerarin and a significant lack of correlation with daidzin, daidzein and genistein.  相似文献

The activity of glutamate dehydrogenase (l-glutamate: NAD oxidoreductase, EC 1.4.1.2.; GDH) of rice plants changes in response to the nitrogen source supplied to the culture solution. The activity of NADH-GDH(aminating) in roots is rapidly increased by the addition of ammonia, whereas the activity in shoots is much less affected by nitrogen supply. The activity increased with increasing concentration of ammonia at least up to 14.3 mM. In roots GDH activity was found in both the mitochondrial and soluble fractions. The increase of NADH-GDH activity caused by the ammonia treatment occurs mainly in the latter fraction. The new band with GDH activity was detected on the zymogram of polyacrylamide gel electrophoresis and this inducible enzyme is active with both NAD and NADP. On the other hand, the constitutive enzyme activity active with NAD is also increased by the ammonia treatment. The increase of enzyme activity is prevented by the addition of cycloheximide or chloramphenicol to culture medium. The incorporation of 14C-leucine(U) into GDH proteins was also studied using polyacrylamide gel electrophoresis. Higher radioactivity was found in induced samples than in non-induced ones. These results show that the increase of GDH activity in roots by ammonia treatment seems to depend on de novo protein synthesis.  相似文献

Cyclic AMP phosphodiesterase (PDE) activity was assayed in the plasma membrane, mitochondrial and microsomal fractions of rat brain. The specific activity of the enzyme was highest in the plasma membrane fraction followed by mitochondrial and then the microsomal fraction. Phosphodiesterase activity of all three fractions was reduced after pretreatment with lecithinase C (PCase) from Clostridium perfringens but less markedly affected by the pretreatment with sphingomyelinase (SMase) from human placenta. The PDE activity of the plasma membrane fraction was more sensitive to PCase treatment compared with the other two particulate fractions, which showed only a slight loss of activity. Temperature seemed to affect PDE activity of the plasma membrane. The enzyme was quite stable at 30 degrees C but its activity dropped by approximately 46% at 37 degrees C after 90 min of incubation. Pretreatment of the plasma membrane at 30 degrees C with PCase at a concentration of more than 5 U caused a marked loss of PDE activity and the decrease in activity reached a plateau at concentrations above 10 U.  相似文献

The flower-inducing and -inhibiting activities of phloem exudate (PE) prepared from cotyledons of Pharbitis seedlings were examined, using apex cultures in vitro from Pharbitis as a bioassay system.The PE was prepared from photoperiodically-induced cotyledons (SD-PE). The SD-PE was subjected to the following fractionations: When the SD-PE was extracted with CHCl3 and then ethyl acetate, the inducing activity was located in the final aqueous fraction. The activity was localized in the diffusate when the aqueous fraction was dialyzed (molecular weight cut off was 10,000). The diffusate was fractionated by ion exchange chromatography, and flower-inducing activity was found in the fraction adsorbed onto anion exchange resin. When the fraction was applied to a Sep-Pak C18 cartridge, the activity eluted with 25% MeOH. As a result of the above fractionation, activity was increased about 30-fold.The nature of the flower-inhibiting activity of the PE taken from cotyledons exposed to continuous-light conditions was examined (CL-PE). The inhibiting activity was decreased as the cotyledons were exposed to longer dark periods; it appeared to be heat-stable. The CL-PE also inhibited flowering in Lemna. The CL-PE was subjected to the following fractionations: When the CL-PE was extracted with CHCl3 and ethyl acetate, activity was located in the final aqueous fraction. Activity was localized in the diffusate when the aqueous fraction was dialyzed (molecular weight cut off was 10,000). When the diffusate was fractionated by ion exchange chromatography, the activity was found in the flow-through fraction. When the fraction was applied to a hydroxyapatite cartridge, the activity eluted with 25 mM sodium phosphate buffer. When the fraction was re-dialyzed (molecular weight cut off was 1,000), the diffusate contained the activity. As a result of the above fractionation, activity was increased about 10-fold.  相似文献

The diurnal activity rhythm of Clethrionomys rufocanus was studied under three different food conditions in a laboratory. The activity occurred both in the daytime and at night, but mainly at night (19:00--4:00). The mice fed with Mouse food exhibited six small peaks in their daily activity, and those fed with grass or hay exhib-ited seven, but the peak of the out-nest night activity was 2 ~ 3 hours earlier. The activity amount of out-nest was 30 518.0 ± 3 694.9 s for those fed with grass, the next was 21 811.7 ± 2 288.0 s for those fed with hay,and the least was 15 038.0 ± 666.0 s for those fed with mouse food. The activity amount of feeding was 10867.3± 1 612.1 s for those fed with grass, much more than those fed with mouse food and hay. The activity amount for drinking was 988.5 ± 79.1 s (fed with hay), 568.9 ± 60.9 s (fed with mouse food) and 139.3 ±47.2 s (fed with grass).  相似文献

The ATPase preparations from the hog thyroid was preincubated with various amounts of trypsin. The activity of Mg-ATPase was consistently elevated. On the contrary, the Na, K-ATPase activity decreased with increasing amounts of trypsin. The effects were similar to those which were observed in the enzyme preparations treated with basis polyamino acids as previously reported. This phenomenon seemed to be specific in the preparations from the thyroid. The Mg-dependent activity was increased after pretreatment with trypsin or poly-L-lysine (PLL) when CTP, ITP and UTP were used as substrate. Thus the substrate specificity of Mg-ATPase was low. The enzyme-kinetics using ATP as substrate showed that the increase in activity was due to an increase in Vmax and not to a change in Km. The activity of Mg-ATPase was increased even after 30 min of preincubation with trypsin, while the Na, K-ATPase activity was almost diminished. These results suggest that the activity of Mg-ATPase in the preparation from the thyroid is specifically changed by the modification of the molecular environment of the enzyme with trypsin or basic polyamino acids.  相似文献
