Newborn babies are able, like the young of many animal species, to recognize their mothers' body odors and to orient towards them. The question arises whether infants can also use artificial odors applied to their mothers' bodies in the same way as animals can. In experiments, breast-feeding mothers perfumed their breasts for two weeks and their infants were tested in a choice test situation. At the age of one week the infants oriented their heads significantly more often to that side where they smelled their mothers' perfumes than to the side where they smelled a control odor. At the age of four weeks (mothers had stopped perfuming at two weeks) the infants did not prefer their mothers' perfumes any more.All the infants showed the “tonic neck reflex,” i.e., they significantly preferred one side, mostly the right, for their head turning. Consequently, these two preferences sometimes caused a conflict of response for the infants in our experiments. The interplay of the preference for turning to the side of mother's perfume and turning to a special side, as well as the biological meaning of both, is discussed and compared with the findings of other authors.  相似文献

As the population ages, there is an ever-increasing need for therapeutic agents that can be used safely and efficaciously to manage symptoms related to postmenopausal estrogen deficiency. Endogenous estrogens, e.g., 17beta-estradiol, of exogenous mammalian origin, e.g., horses, have long been used to manage such symptoms. There are more than 20 different classes of phytochemicals that have demonstrated affinity for human estrogen receptors in vitro. Some studies on exogenous estrogenic substances of botanical origin (phytoestrogens), such as standardized formulations of plant extracts with in vitro and in vivo estrogenic activity from soy (Glycine max Merill.) and red clover (Trifolium pratense L.), suggest clinical efficacy. Few clinical data for phytoestrogens other than isoflavonoids are available. In an exhaustive review of the literature through 2003, only two clinical trials were identified that were designed to evaluate the effect of hops (Humulus lupulus L.) on symptoms related to menopause. Folkloric, chemical, and biological literature relating primarily to the use of hops for their estrogenic activity, and two human clinical trials, are reviewed.  相似文献

INTRODUCTION     
Foliage-feeding forest insects have served as model systems in the study of animal populations for more than 50 years. Early studies emphasized identification of "key" mortality agents or density-dependent sources of mortality. However, these efforts became burdened by rhetorical ambiguity, and population ecologists are increasingly focusing on characterizing population behavior and identifying the processes that generate that behavior. Two types of behavior seem to be common in forest insect populations: periodic oscillations ("population cycles") and spatial synchrony (synchronous fluctuations over large geographic areas). Several population processes (e.g., host–pathogen interactions) have been demonstrated to be capable of producing periodic oscillations, but the precise identity of these processes remains uncertain for most forest insects and presents a challenge to future research. As part of these efforts, a greater emphasis is needed on the use of statistical methods for detecting periodic behavior and for identifying other types of population behavior (e.g., equilibrium dynamics, limit cycles, transient dynamics). Spatial synchrony appears to be even more ubiquitous in forest insect populations. Dispersal and regional stochasticity ("Moran effect") have been shown to be capable of producing synchrony, but again more research is needed to determine the relative contribution of these processes to synchrony observed in natural populations. In addition, there is a need to search for other types of time–space patterns (e.g., traveling waves, spiral waves) in forest insect populations and to determine their causes. The effects on self management of asthma of a specially prepared book and audiocassette tape with similar contents were observed in a controlled study of 177 patients with asthma in general practice. After a run in period of six months patients were randomly given the book, the tape, both the book and tape, or neither. Patients'' knowledge of the use of drugs, perceptions of their disability, skill in using an inhaler, consumption of drugs, consultations with their general practitioners, morbidity (from patients'' entries on diary cards), and use of the educational material were measured. Knowledge about the use of drugs was significantly increased in the groups who received the material after three months and persisted after 12 months. Patients who had been given the tape or the book and tape increased their scores of knowledge of drugs more than patients given the book alone. Patients in all groups given the material considered that their disability was reduced. There were no other significant changes. Patients given both the book and the tape preferred the book. Patients with asthma can obtain useful information from such material. The paradoxical result whereby patients learnt more from the tape but preferred the book suggests that a distinction can be made between information that patients need, which may be acquired better from an audiocassette, and information that they want, which may be acquired better from a book.  相似文献

Animal models of osteoarthritis are used to study the pathogenesis of cartilage degeneration and to evaluate potential antiarthritic drugs for clinical use. Animal models of naturally occurring osteoarthritis (OA) occur in knee joints of guinea pigs, mice and other laboratory animal species. Transgenic models have been developed in mice. Commonly utilized surgical instability models include medial meniscal tear in guinea pigs and rats, medial or lateral partial meniscectomy in rabbits, medial partial or total meniscectomy or anterior cruciate transection in dogs. Additional models of cartilage degeneration can be induced by intra-articular iodoacetate injection or by administration of oral or parenteral quinolone antibiotics. None of these models have a proven track record of predicting efficacy in human disease since there are no agents that have been proven to provide anything other than symptomatic relief in human OA. However, agents that are active in these models are currently in clinical trials. Methodologies, gross and histopathologic features and comparisons to human disease will be discussed for the various models.  相似文献

Only cefprozil and cefpodoxime have been shown to be more effective than comparison agents, and they appear to better tolerated than amoxicillin-containing regimens. The potent Gram-negative activity of cefpodoxime is attractive for selective clinical situations (e.g., in place of a quinolone). However, it has not been tested for the treatment of serious Gram-negative infections. Either of these agents would appear to be preferable to cefaclor in terms of efficacy, safety, and cost. Usage patterns similar to that currently applicable to cefaclor should also be applied to the use of either of these drugs, as they should be reserved for patients who have not responded to first-line therapy or in penicillin-allergic patients who cannot tolerate erythromycin or trimethoprimsulfamethoxazole.  相似文献

The platinum-based drugs, cisplatin and carboplatin, represent major agents in the chemotherapeutic treatment of a variety of types of cancer. Novel, "third-generation" agents aimed at broadening the clinical activity of this class of drug are currently undergoing clinical evaluation. These include oxaliplatin, ZD0473 and BBR3464. Clinical trials and preclinical studies are also being conducted with liposomal (SPI-077 and L-NDDP) and polymeric platinum complexes (linked to HPMA or albumin). Combination studies of cisplatin/carboplatin with other anticancer drugs such as gemcitabine and UCN-01 (7-hydroxystaurosporine) and agents designed to reduce platinum drug toxicities (e.g., BNP-7787, DIMESNA) are ongoing. Preclinically, there is interest in trans platinum complexes, terpyridine platinum(II) complexes and other metal-containing agents (ruthenium and gold).  相似文献

The multifactorial ulcer-producing actions of non-steroidal anti-inflammatory drugs (NSAIDs) are briefly reviewed and the main actions highlighted as the focus for potential strategies for reducing the ulcerogenic effects of these drugs. While some clinical benefits are evident from long-term clinical studies from application of PG analogues (misoprostol) and H+/K(+)-ATP-ase inhibitors (omeprazole) these are, ultimately, expensive approaches. Chemical structural properties of the NSAIDs underlying differences in their ulcerogenicity are analyzed with the objective of establishing the reasons for the low ulcerogenicity of some of these drugs (e.g. azapropazone). These studies serve as a basis for developing less gastrotoxic drugs in the future. In the studies we have undertaken analysis of the benefits of micronutrients and of modulating eicosanoid metabolism have been considered. The results of some clinical trials with micronutrients have proven encouraging. These and other approaches and pitfalls reported give further encouragement to explore the mechanisms of the protective effects of these latter agents and serve as a basis for future developments.  相似文献

Pharmacodynamic modeling has been increasingly used as a decision support tool to guide dosing regimen selection, both in the drug development and clinical settings. Killing by antimicrobial agents has been traditionally classified categorically as concentration-dependent (which would favor less fractionating regimens) or time-dependent (for which more frequent dosing is preferred). While intuitive and useful to explain empiric data, a more informative approach is necessary to provide a robust assessment of pharmacodynamic profiles in situations other than the extremes of the spectrum (e.g., agents which exhibit partial concentration-dependent killing). A quantitative approach to describe the interaction of an antimicrobial agent and a pathogen is proposed to fill this unmet need. A hypothetic antimicrobial agent with linear pharmacokinetics is used for illustrative purposes. A non-linear functional form (sigmoid Emax) of killing consisted of 3 parameters is used. Using different parameter values in conjunction with the relative growth rate of the pathogen and antimicrobial agent concentration ranges, various conventional pharmacodynamic surrogate indices (e.g., AUC/MIC, Cmax/MIC, %T>MIC) could be satisfactorily linked to outcomes. In addition, the dosing intensity represented by the average kill rate of a dosing regimen can be derived, which could be used for quantitative comparison. The relevance of our approach is further supported by experimental data from our previous investigations using a variety of gram-negative bacteria and antimicrobial agents (moxifloxacin, levofloxacin, gentamicin, amikacin and meropenem). The pharmacodynamic profiles of a wide range of antimicrobial agents can be assessed by a more flexible computational tool to support dosing selection.  相似文献

Previous studies of the motor cortex in behaving animals were focused on the relations between the activity of single cells, usually pyramidal tract neurons, and parameters of isometric contraction (e.g., intensity of force) or parameters of movement along one axis (e.g., flexion-extension) of a single joint (e.g., elbow or wrist). However, the commonly meaningful behavioral parameter is the trajectory of the hand in extrapersonal space, which is realized by simultaneous motions about two or three joints (e.g., elbow, shoulder, wrist) and concurrent engagement of several muscles. The spatial parameters of a straight trajectory are its direction and extent. We hypothesized that a major function of the motor cortex, among other possible roles, is the specification and control of the direction of the movement trajectory in space. This reference of motor cortical function to the control of spatial aspects of the trajectory differentiated our approach from the other approaches outlined above. We investigated the directional selectivity cells in the arm area of the motor cortex by recording their activity while monkeys moved their hands in various directions in space towards visual targets. There were two salient findings of these studies. First, the intensity of the discharge of single cells varies in an orderly fashion with the direction of movement in space, so that the discharge rate is highest with movements in a preferred direction, and decreases progressively with movements made in directions more and more away from the preferred one. Cancer is becoming an increasingly significant disease worldwide. Currently, more than 7 million people die each year from cancer. With the existing knowledge, at least one-third of worldwide cancer cases could be prevented. Searching for naturally occurring agents in routinely consumed foods that may inhibit cancer development, although challenging, constitutes a valuable and plausible approach to the control and prevention of cancer. To date, the use of the micronutrient selenium (Se) in human clinical trials is limited, but the outcome indicates that Se is among the most promising agents. Although it is convenient to describe the effects of Se in terms of the element, it must always be kept in mind that the chemical form of Se and the dose are determinants of its biological activities. Hyphenated techniques based on coupling chromatographic separation with inductively coupled plasma mass spectrometric (ICP-MS) detection are now established as the most realistic and potent analytical tools available for real-life speciation analysis. These speciation investigations provide evidence that the Se compounds, which can generate monomethylated Se (e.g., Se-methylselenocysteine and methylseleninic acid), are more efficacious than other Se compounds because of their chemoprevention activity.  相似文献

Mansonella perstans filariasis is widely present in Africa and equatorial America and its pathogenicity has been recently reconsidered. Although M. perstans infection has been considered a minor filariasis, remaining asymptomatic in most of infected subjects, more recent studies have shown that M. perstans is capable of inducing a variety of clinical features, including angioedemas, swellings like the "Calabar swellings" of loiasis, pruritus, fever, headache, pain in bursae and/or joint synovia, or in serous cavities. It is likely that some of the pathological changes observed are induced by the immune response to the infection. Eosinophilia is present in many cases of infection. Moreover M. perstans filariasis is difficult to be treated. Effective treatment is lacking and there is no consensus on optimal therapeutic approach. The most commonly used drug is diethylcarbamazine (DEC) that is however often ineffective. Although other drugs have been tried (e.g. praziquantel, ivermectin), none has proven to be reliably and rapidly effective. Mebendazole seemed more active than DEC in eliminating the infection, with a comparable rate of overall responses. Thiabendazole evidenced a small, but significant activity against the infection. Combination treatments (DEC plus mebendazole) resulted in a significantly higher activity compared with the single drugs.  相似文献

Microparticles have been effectively used for many years as delivery systems for drugs and therapeutic proteins. Their application to the delivery of vaccines is not as extensive, but is growing. Utility has been demonstrated for the delivery of various types of vaccines (e.g., recombinant proteins, plasmid DNA, and peptides) and other vaccine components (e.g., immune potentiators). With respect to delivery of immune potentiators, synergistic effects are often observed whereby much more potent immune responses are induced with a combination than with either component alone. Hence, the prospects for broad application of microparticle-based delivery systems for vaccines are excellent.  相似文献

The studies reviewed herein support the precept that "systemic dose-intensity" (i.e., systemic exposure) may be more informative than "administered dose-intensity" for certain anticancer drugs. This does not mean that the administered dose-intensity should be ignored; in fact these data indicate the importance of documenting and assessing administered dose-intensity as an initial step toward identifying those situations where systemic dose-intensity may be most important. The studies described in this review were selected as representative examples of successful clinical pharmacodynamic studies; other published examples include vincristine AUC versus severity of neurotoxicity, etoposide systemic exposure versus leukopenia, red cell concentration of mercaptopurine metabolites versus neutropenia in children with ALL, and ARA-CTP retention in leukemic blasts versus clinical response in acute non-lymphocytic leukemia. As is the case with other types of clinical trials in cancer patients, there are also examples of negative pharmacodynamic studies (i.e., no relationship found between concentration and effects). There are several possible reasons for such negative findings, including the lack of such a relationship for some drugs, measuring the inappropriate drug moiety (e.g., failure to measure all active metabolites), measuring drug concentrations in the wrong biological fluid, evaluating systemic exposure over too narrow a range (i.e., all patients have either sub- or supra-therapeutic systemic exposure), selecting inappropriate sampling times or pharmacokinetic parameters, inadequately assessing drug toxicity or response, or simply studying an inadequate number of patients or patients with drug-resistant cancers. Therefore, negative findings in some pharmacodynamic studies should not deter the investigation of other drugs and/or other malignant diseases, just as negative therapeutic trials do not preclude subsequent clinical trials in oncology. Also, finding a relation between systemic exposure and drug toxicity, in the absence of a clear relation to antitumor effects, is potentially of great clinical utility. Such data should allow more objective escalation of drug dosages in individual patients, to ensure maximum dose-intensity while avoiding host toxicity. Obviously, if such dose escalation could be guided by more easily measured patient characteristics (e.g., age, weight, CrCl, shoe size, etc.), then using drug concentrations in individual patients might be obviated.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

The ability to choose the partners we interact with is thought to have been an important driver in the evolution of human social behavior, and in particular, our propensity to cooperate. Studies showing that humans prefer to interact with cooperative others is often cited as support for partner choice driving the evolution of cooperation. However, these studies are largely drawn from Western samples, where conditions for partner choice to operate may be especially favorable. Here, we investigate qualities associated with being a preferred partner (i.e., campmate) in Hadza hunter-gatherers of Tanzania in 2016 and 2019. A total of 156 Hadza participants from 17 camps ranked their campmates on generosity, foraging ability, and their preference for them as future campmates. In 2016, Hadza preferred more generous people and better hunters as campmates, with evidence suggesting a stronger preference for better hunters; however, the relationship between generosity and being a preferred campmate was greater in 2019 than in 2016, such that the preference for generous people was stronger than the preference for better foragers, suggesting that campmate preferences are changing. These new findings contrast with reports on data from nearly a decade ago, suggesting that the Hadza do not prefer more cooperative campmates. Further, in 2019, there was anecdotal evidence that Hadza with greater exposure to outside cultural institutions (e.g., schooling, having a job, or living in a village) had a stronger preference for generous campmates than those with less exposure. Taken together, the results suggest that preferences for social partners may, in part, be culturally shaped.  相似文献

Attitudes toward animals are influenced by both animal traits (e.g., similarity to humans, aesthetic quality, size) and individual human attributes (e.g., gender, age, educational level, cultural factors). Although the examination of children's interest in animals, and their preference for different species, may evidence specific trends and help explain the development of attitudes, the vast majority of research has not considered children younger than 6 years. The present study was aimed at assessing preferences for a variety of animal species in a sample of 3–6 year-old Italian children, using a forced-choice task and visual aids (images of the animals). Pictures of 48 animal species, ranging from mammals to invertebrates, were presented to the children. Two photographic stimuli were simultaneously displayed and participants were asked to indicate their preference. Results show that the children preferred higher-order species, and domestic over wild animals. Apart from a few exceptions, invertebrates were the most disliked group of species among the children. Girls showed more negative and fear-related attitudes than the boys. Results are discussed taking into account different factors that may affect children's preferences for various animal species, that is, similarity to humans and aesthetical appeal. Greater knowledge on early attitudes toward animals has implications for promoting interest in animals and for building educational interventions for kindergarten children. This is particularly important in light of the growing use of different animals in educational and therapeutic contexts, as well as from an animal welfare perspective.  相似文献

Optimal foraging, specialization, and a solution to Liem's paradox   总被引:5,自引:0,他引:5  
Species that appear highly specialized on the basis of their phenotype (e.g., morphology, behavior, and physiology) also sometimes act as ecological generalists. This apparent paradox has been used to argue against the importance of competition as a diversifying evolutionary force. We provide an alternative explanation based on optimal foraging theory. Some resources are intrinsically easy to use and are widely preferred, while others require specialized phenotypic traits on the part of the consumer. This asymmetry allows optimally foraging consumers to evolve phenotypic specializations on nonpreferred resources without greatly compromising their ability to use preferred resources. The evolution of phenotypic specialization on nonpreferred resources can be driven by competition, but the specialists act as ecological generalists whenever their preferred resources are available. Our model identifies at least three different concepts of specialization that need to be distinguished, based on diet, prey utilization efficiencies, and phenotypic adaptations. The relationships among these concepts are complex and often counterintuitive. Specialists should often reject the very resources that they have evolved traits to use. The most extreme phenotypic specializations should occur in the absence of a trade-off between using preferred and nonpreferred resources. Our model may explain why extreme phenotypic-specializations evolve more often in fish communities than in terrestrial vertebrate communities and provides a mechanism whereby species can coexist in stable communities despite common preferences for some resources.  相似文献
