Carbon-13 nuclear magnetic resonance (NMR) spectroscopy has been applied to the direct observation of acetate and pyruvate metabolism in suspension cultures of Zea mays (var Black Mexican Sweet). Growth of the corn cells in the presence of 2 millimolar [2-13C]acetate resulted in a rapid uptake of the substrate from the medium and initial labeling (0-4 hours) of primarily the intracellular glutamate and malate pools. Further metabolism of these intermediates resulted in labeling of glutamine, aspartate, and alanine. With [1-13C]acetate as the substrate very little incorporation into intermediary metabolites was observed in the 13C NMR spectra due to loss of the label as 13CO2. Uptake of [3-13C]pyruvate by the cells was considerably slower than with [2-13C]acetate; however, the labelling patterns were similar with the exception of increased [3-13C] alanine generation with pyruvate as the substrate. Growth of the cells for up to 96 hours with 2 millimolar [3-13C]pyruvate ultimately resulted in labeling of valine, leucine, isoleucine, threonine, and the polyamine putrescine.  相似文献

Malate synthesis by CO2 fixation in wheat (Triticum aestivum L.) and lupin (Lupinus luteus) roots was investigated by labeling with NaH13CO3 as well as with NaH14CO3. The distribution of 14C label in the malate was examined, using enzymic degradation methods (malic enzyme, pyruvate decarboxylase) and, in the case of 13C, gas chromatography-mass spectrometry. In long-term experiments (2 to 12 hours), both methods showed that the [1-C] and [4-C] positions of malic acid are approximately equally labeled, in agreement with former findings. Short-term experiments (15, 30 seconds) showed that 14C is confined initially to the [4-C] position of malate but then is distributed quickly to the [1-C] atom. Neither labeling pattern nor rate of randomization was influenced by salt treatment. Analysis of malate from roots by gas chromatography-mass spectrometry, a procedure which was tested against in vitro-prepared [1-13C]-, [4-13C]-, and [1,4-13C] malate, gave strong evidence for the existence of only singly labeled malate molecules. These data suggest that only one carboxylation step, catalyzed by phosphoenolpyruvate carboxylase and/or phosphoenolpyruvate carboxykinase, is responsible for malic acid synthesis in roots and that malate label is randomized by a fumarase-like reaction, presumably in mitochondria.  相似文献

14C-Labeled 2-carboxyarabinitol-1,5-bisphosphate was bound to both nonactivated and CO2and Mg2+ activated forms of ribulose bisphosphate carboxylase/oxygenase. The complex could be precipitated with 20% polyethylene glycol and 20 mm MgCl2 for quantitation of the moles of the affinity label bound per mole of enzyme. The [14C]carboxyarabinitol-P2 bound to the nonactivated enzyme could be exchanged with a 100-fold excess of the unlabeled compound. With the activated enzyme the binding of [14C]carboxyarabinitol-P2 was so tight that it did not exchange with the unlabeled compound and a binding stoichiometry of one molecule per active site was assumed. This tight binding was dependent upon pretreatment of the enzyme with both CO2 and MgCl2 in the same manner that enzyme activation depended on CO2 and Mg2+ concentrations. Various enzyme preparations from spinach leaves tightly bound [14C]carboxyarabinitol-P2 in proportion to their specific activities. By extrapolating to a maximum binding of 8 mol of [14C]carboxyarabinitol-P2 per mole of this A8B8 enzyme a theoretical specific activity of 2.8 μmol · min?1 · mg protein?1 was indicated. Enzyme preparations purified from spinach leaves generally have a specific activity in the range of 1.0 to 2.3.  相似文献

The metabolism of [13C]malate was studied in the Crassulacean plant Kalanchoë tubiflora following exposure to 13CO2 for 2 hour intervals during a 16 hour dark cycle. Nuclear magnetic resonance spectroscopy of [13C]malate extracted from labeled tissue revealed that the transient flux of malate to the mitochondria, estimated by the randomization of [4-13C]malate to [1- 13C]malate by fumarase, varied substantially during the dark period. At both 15 and 25°C, the extent of malate label randomization in the mitochondria was greatest during the early and late parts of the dark period and was least during the middle of the night, when the rate of 13CO2 uptake was highest. Randomization of labeled malate continued for many hours after malate synthesis had initially occurred. Internally respired 12CO2 also served as a source of carbon for malate formation. At 15°C, 15% of the total malate was formed from respired 12CO2, while at 25°C, 49% of the accumulated malate was derived from respired 12CO2. Some of the malate synthesized from external 13CO2 was also respired during the night. The proportion of the total [13C]malate respired during the dark period was similar at 15 and 25°C, and respiration of newly formed [13C]malate increased as the night period progressed. These data are discussed with regard to the relative fluxes of malate to the mitochondria and the vacuole during dark CO2 fixation.  相似文献

13C-nuciear magnetic resonance (NMR) spectroscopy was used to investigate the products of glycerol and acetate metabolism released by Leishmania braziliensis panamensis promastigotes and also to examine the interaction of each of these substrates with glucose or alanine. The NMR data were supplemented by measurements of the rates of oxygen consumption and substrate utilization, and of 14CO2 production from 14C-labeIed substrate. Cells incubated with [2-13C]glycerol released acetate, succinate and D-lactate in addition to CO2. Cells incubated with acetate released only CO2. More succinate C-2/C-3 than C-l/C-4 was released from both [2-13C]glycerol and [2-13C]glucose, indicating that succinate was formed predominantly by CO2 fixation followed by reverse flux through part of the Krebs cycle. Some redistribution of the position of labeling was also seen in alanine and pyruvate, suggesting cycling through pyruvate/oxaloacetate/phosphoenolpyruvate. Cells incubated with combinations of 2 substrates consumed oxygen at the same rate as cells incubated with 1 or no substrate, even though the total substrate utilization had increased. When promastigotes were incubated with both glycerol and glucose, the rate of glucose consumption was unchanged but glycerol consumption decreased about 50%, and the rate of 14CO2 production from [l,(3)-14C]glycerol decreased about 60%. Alanine did not affect the rates of consumption of glucose or glycerol, but decreased 14CO2 production from these substrates by increasing flow of label into alanine. Although glucose decreased alanine consumption by 70%, it increased the rate of 14CO2 production from [U-14C]- and [l-14C]alanine by about 20%. This is consistent with rapid equilibration of alanine with pyruvate derived from glucose and yet little decrease in the specific activity of the large alanine pool.  相似文献

Andrographolide, a diterpene lactone, is isolated from Andrographis paniculata which is well known for its medicinal properties. The biosynthetic route to andrographolide was studied using [1-13C]acetate, [2-13C]acetate and [1,6-13C2]glucose. The peak enrichment of eight carbon atoms in the 13C NMR spectra of andrographolide suggested that deoxyxylulose pathway (DXP) is the major biosynthetic pathway to this diterpene.The contribution of the mevalonic acid pathway (MVA) is indicated by the observed 13C-labeling pattern, and because the labeling patterns indicate a simultaneous contribution of both methyl erythritol phosphate (MEP) and MVA pathways it can be deduced that cross-talk occurs between plastids and cytoplasm.  相似文献

The labeling patterns in malic acid from dark 13CO2 fixation in seven species of succulent plants with Crassulacean acid metabolism were analysed by gas chromatography-mass spectrometry and 13C-nuclear magnetic resonance spectrometry. Only singly labeled malic-acid molecules were detected and on the average, after 12–14 h dark 13CO2 fixation the ratio of [4-13C] to [1-13C] label was 2:1. However the 4-C carboxyl contained from 72 to 50% of the label depending on species and temperature. The 13C enrichment of malate and fumarate was similar. These data confirm those of W. Cockburn and A. McAuley (1975, Plant Physiol. 55, 87–89) and indicate fumarase randomization is responsible for movement of label to 1-C malic acid following carboxylation of phosphoenolpyruvate. The extent of randomization may depend on time and on the balance of malic-acid fluxes between mitochondria and vacuoles. The ratio of labeling in 4-C to 1-C of malic acid which accumulated following 13CO2 fixation in the dark did not change during deacidification in the light and no doubly-labeled molecules of malic acid were detected. These results indicate that further fumarase randomization does not occur in the light, and futile cycling of decarboxylation products of [13C] malic acid (13CO2 or [1-13C]pyruvate) through phosphoenolpyruvate carboxylase does not occur, presumably because malic acid inhibits this enzyme in the light in vivo. Short-term exposure to 13CO2 in the light after deacidification leads to the synthesis of singly and multiply labeled malic acid in these species, as observed by E.W. Ritz et al. (1986, Planta 167, 284–291). In the shortest times, only singly-labeled [4-13C]malate was detected but this may be a consequence of the higher intensity and better detection statistics of this ion cluster during mass spectrometry. 13C NMR spectroscopy was applied to studying lysine biosynthesis in Corynebacterium glutamicum ATCC 21543, a lysine producing mutant. It was cultured in a medium containing [1-13C]glucose or [6–13C]glucose as the sole carbon source and the 13C NMR spectrum of the culture filtrate was measured. C labeling patterns of l-lysine produced were well explained by the putative metabolic pathways of the bacterium. Fixation of 13CO2 liberated from the labeled substrates and the operation of the tricarboxylate cycle in the fermentation were obviously observed. The dual operations of the classical diaminopimelate pathway and the diaminopimelate dehydrogenase bypath were supported. Calculation of the contribution ratios of the metabolic pathways was attempted.  相似文献

When ribulose-1,5-bisphosphate carboxylase is assayed under N2 using [3H]ribulose 1,5-bisphosphate and 14CO2, [3H]3-phosphoglycerate and [14C]3-phosphoglycerate are produced in nonstoichiometric amounts in a ratio which approaches 7 at low concentrations of CO2 (2 micromolar) assuming a 1:1 ratio at Vmax (280 micromolar). The log of the molar ratio varies as a linear function of log[CO2]. Nonstoichiometry could be explained by CO2 contaminatio of the reactants or tritium contamination of the products. However, the magnitude of CO2 contamination required (18 ± 4 micromolar) is far in excess of controlled CO2 (<0.1 micromolar), and the required tritium contaminant would have to vary from 30 to 85% of the purified 3-phosphoglycerate at the 58 and 2 micromolar CO2 assay levels, respectively. This contrasts with detectable tritium contamination which is only 1 to 4% and correctable. Nonstoichiometry is evident using either 1 or 5 labeled [3H]ribulose 1,5-bisphosphate. When 3-phosphoglycerate is reisolated as glycerate the 3H/14C ratio remains unchanged.  相似文献

Background

Pathogen-specific metabolic pathways may be detected by breath tests based on introduction of stable isotopically-labeled substrates and detection of labeled products in exhaled breath using portable infrared spectrometers.

Methodology/Principal Findings

We tested whether mycobacterial urease activity could be utilized in such a breath test format as the basis of a novel biomarker and diagnostic for pulmonary TB. Sensitized New-Zealand White Rabbits underwent bronchoscopic infection with either Mycobacterium bovis or Mycobacterium tuberculosis. Rabbits were treated with 25 mg/kg of isoniazid (INH) approximately 2 months after infection when significant cavitary lung pathology was present. [13C] urea was instilled directly into the lungs of intubated rabbits at selected time points, exhaled air samples analyzed, and the kinetics of δ13CO2 formation were determined. Samples obtained prior to inoculation served as control samples for background 13CO2 conversion in the rabbit model. 13CO2, from metabolic conversion of [13C]-urea by mycobacterial urease activity, was readily detectable in the exhaled breath of infected rabbits within 15 minutes of administration. Analyses showed a rapid increase in the rate of 13CO2 formation both early in disease and prior to treatment with INH. Following INH treatment, all evaluable rabbits showed a decrease in the rate of 13CO2 formation.

Conclusions/Significance

Urea breath testing may provide a useful diagnostic and biomarker assay for tuberculosis and for treatment response. Future work will test specificity for M. tuberculosis using lung-targeted dry powder inhalation formulations, combined with co-administering oral urease inhibitors together with a saturating oral dose of unlabeled urea, which would prevent the δ13CO2 signal from urease-positive gastrointestinal organisms.  相似文献

13C spin diluted protein samples can be produced using [1-13C] and [2-13C]-glucose (Glc) carbon sources in the bacterial growth medium. The 13C spin dilution results in favorable 13C spectral resolution and polarization transfer behavior. We recently reported the combined use of [1-13C]- and [2-13C]-Glc labeling to facilitate the structural analysis of insoluble and non-crystalline biological systems by solid-state NMR (ssNMR), including sequential assignment, detection of long-range contacts and structure determination of macromolecular assemblies. In solution NMR the beneficial properties of sparsely labeled samples using [2-13C]-glycerol (13C labeled Cα sites on a 12C diluted background) have recently been exploited to provide a bi-directional assignment method (Takeuchi et al. in J Biomol NMR 49(1):17–26, 2011 ). Inspired by this approach and our own recent results using [2-13C]-Glc as carbon sources for the simplification of ssNMR spectra, we present a strategy for a bi-directional sequential assignment of solid-state NMR resonances and additionally the detection of long-range contacts using the combination of 13C spin dilution and 3D NMR spectroscopy. We illustrate our results with the sequential assignment and the collection of distance restraints on an insoluble and non-crystalline supramolecular assembly, the Salmonella typhimurium type III secretion system needle.  相似文献

Extracts of Pseudomonas C grown on methanol as sole carbon and energy source contain a methanol dehydrogenase activity which can be coupled to phenazine methosulfate. This enzyme catalyzes two reactions namely the conversion of methanol to formaldehyde (phenazine methosulfate coupled) and the oxidation of formaldehyde to formate (2,6-dichloroindophenol-coupled). Activities of glutathione-dependent formaldehyde dehydrogenase (NAD+) and formate dehydrogenase (NAD+) were also detected in the extracts.The addition of d-ribulose 5-phosphate to the reaction mixtures caused a marked increase in the formaldehyde-dependent reduction of NAD+ or NADP+. In addition, the oxidation of [14C]formaldehyde to CO2, by extracts of Pseudomonas C, increased when d-ribulose 5-phosphate was present in the assay mixtures.The amount of radioactivity found in CO2, was 6.8-times higher when extracts of methanol-grown Pseudomona C were incubated for a short period of time with [1-14C]glucose 6-phosphate than with [U-14C]glucose 6-phosphate.These data, and the presence of high specific activities of hexulose phosphate synthase, phosphoglucoisomerase, glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase indicate that in methanol-grown Pseudomonas C, formaldehyde carbon is oxidized to CO2 both via a cyclic pathway which includes the enzymes mentioned and via formate as an oxidation intermediate, with the former predominant.  相似文献

Complexes of the type [Pt(amine)4]I2 were synthesized and characterized mainly by multinuclear (195Pt, 1H and 13C) magnetic resonance spectroscopy. The compounds were prepared with different primary amines, but not with bulky amines, due to steric hindrance. In 195Pt NMR, the signals were observed between −2715 and −2769 ppm in D2O. The coupling constant 3J(195Pt-1H) for the MeNH2 complex is 42 Hz. In 13C NMR, the average values of the coupling constants 2J(195Pt-13C) and 3J(195Pt-13C) are 18 and 30 Hz, respectively. The crystal structure of [Pt(EtNH2)4]I2 was determined by X-ray diffraction methods. The Pt atom is located on an inversion center. The structure is stabilized by H-bonding between the amines and the iodide ions. The compound with n-BuNH2 was found by crystallographic methods to be [Pt(n-BuNH2)4]2I3(n-BuNHCOO). The crystal contains two independent [Pt(CH3NH2)4]2+ cations, three iodide ions and a carbamate ion formed from the reaction of butylamine with CO2 from the air. When the compound [Pt(CH3NH2)4]I2 was dissolved in acetone, crystals identified as trans-[Pt(CH3NH2)2(H3CNC(CH3)2)2]I2 were isolated and characterized by crystallographic methods. Two trans bonded MeNH2 ligands had reacted with acetone to produce the two N-bonded Schiff base Pt(II) compound.  相似文献

The precursors and directionality of synthesis of the methyl branched cuticular hydrocarbons and the female contact sex pheromone, 3,11-dimethyl-2-nonacosanone, of the German cockroach, Blattella germanica, were investigated by radiotracer and carbon-13 NMR techniques. The amino acids [G-3H]valine, [4,5-3H]isoleucine and [3,4-14C2]methionine labeled the hydrocarbon fraction in a manner indicating that the carbon skeletons of all three amino acids serve as the methyl branch group donor. The incorporation of [1,4-14C2]- and [2,3-14C2]succinates into the hydrocarbon and acylglycerol/polar lipid fractions indicated that succinate also served as a precursor to methylmalonyl-CoA. Carbon-13 NMR analyses showed that [1-13C]propionate labeled the carbon adjacent to the tertiary carbon, and, for the 3,x-dimethylalkanes, that carbon-4 and not carbon-2 was enriched. [1-13C]Acetate labeled carbon-2 of these hydrocarbons. This indicates that the methyl branching groups of the 3,x-dimethylalkanes were inserted early in the chain elongation process. [3,4,5-13C3]Valine labeled the methyl, tertiary and carbon adjacent to the tertiary carbon of the methyl branched alkanes. Thus, the methyl branched hydrocarbon was formed by the insertion of methylmalonyl units derived from propionate, isoleucine, valine, methionine and succinate early in chain elongation.  相似文献

Methyl 13CHD2 isotopomers of all methyl-containing amino-acids can be observed in residually protonated samples of large proteins obtained from [U-13C,1H]-glucose/D2O-based bacterial media, with sensitivity sufficient for a number of NMR applications. Selective detection of some subsets of methyl groups (Alaβ, Thrγ2) is possible using simple ‘out-and-back’ NMR methodology. Such selective methyl-detected ‘out-and-back’ NMR experiments allow complete assignments of threonine γ2 methyls in residually protonated, [U-13C,1H]-glucose/D2O-derived samples of an 82-kDa enzyme Malate Synthase G. [U-13C,1H]-glucose/D2O-derived protein samples are relatively inexpensive and are usually available at very early stages of any NMR study of high-molecular-weight systems.  相似文献

We grew a non-bicarbonate using red seaweed, Lomentaria articulata (Huds.) Lyngb., in media aerated with four O2 concentrations between 10 and 200% of current ambient [O2] and four CO2 concentrations between 67 and 500% of current ambient [CO2], in a factorial design, to determine the effects of gas composition on growth and physiology. The relative growth rate of L. articulata increased with increasing [CO2] up to 200% of current ambient [CO2] but was unaffected by [O2]. The relative growth enhancement, on a carbon basis, was 52% with a doubling of [CO2] but fell to 23% under 5× ambient [CO2]. Plants collected in winter responded more extremely to [CO2] than did plants collected in the summer, although the overall pattern was the same. Discrimination between stable carbon isotopes (Δ13C) increased with increasing [CO2] as would be expected for diffusive CO2 acquisition. Tissue C and N were inversely related to [CO2]. Growth in terms of biomass appeared to be limited by conversion of photosynthate to new biomass rather than simply by diffusion of CO2, suggesting that non-bicarbonate-using macroalgae, such as L. articulata, may not be directly analogous to C3 higher plants in terms of their responses to changing gas composition.  相似文献

dl-Ornithine-[2,3-13C2] was synthesized from acetate-[1-13C] and ethyl acetamidocyanoacetate-[2-13C]. This labelled material was mixed with dl-ornithine-[5-14C] and fed to Nicotiana glutinosa plants by the wick method. After 10 days the plants were harvested affording radioactive nicotine and nornicotine (0.14% and 0.051% specific incorporations, respectively). Even at these low specific incorporations an examination of their 13C NMR spectra established the incorporation of ornithine symmetrically into the pyrrolidine rings of these alkaloids. Satellites were observable at the signals due to C-2′, 3′, 4′ and 5′ positions, arising by the presence of contiguous carbons at C-2′, 3′ and C-4′, 5′.  相似文献
