Summary Adult Cichlasoma biocellatum were kept in deionized water, 25% sea-water and tap-water. The mucous cells in the epithelium of the gills and the buccal floor were not affected by the osmotic experiments. In animals kept in deionized water the mucous cells in the skin remained unaffected, but the ionocytes (chloride cells) in the gills were strongly stimulated. Keeping the animals in salt water led to a strong regression of the epidermis including the mucous cells, and of the ionocytes. The regression could be counteracted by prolactin injections. The changes in the skin and the ionocytes could be correlated with the activity of the prolactin producing cells in the adenohypophysis. It is suggested that the epidermis and its mucous cells as well as the ionocytes are under prolactin control.The authors gratefully acknowledge the interest and helpful suggestions of Prof. Dr. P. G. Summary The development and activity of the basophils in the meso-adenohypophysis of Anopthichthys jordani were studied in relation to the maturation of the gonads. In addition, pituitaries and ovaries of post-spawning females were studied during a period 0–11 days subsequent to spawning. The large and mainly PAS-positive basophilic cells in the meso-adenohypophysis were the only cell type that showed a correlation with the development and maturation of the ovaries and testes. These cells were subjected to a partial or complete degranulation in females following oviposition, when yolk formation in the ovary was at a maximum. It is concluded that the large PAS-positive basophils in the mesoadenohypophysis have a gonadotropic function and that only one gonadotropic cell type is present in the adenohypophysis of Anoptichthys jordani.This study was partly supported by a grant from the Netherlands Organization for the Advancement of Pure Research (Z.W.O.).I wish to express my gratitude to Prof. Dr. P. Summary In order to identify the TSH cells in the pituitary gland of Anoptichthys jordani, the development of the adenohypophysial cell types was studied in relation with the differentiation of the thyroids. In addition experiments were performed in young and adult animals with propylthiouracil (PTU) and thyroxine. At the time of the first thyroid development two meso-adenohypophysial cell types in the pituitary gland of Anoptichthys jordani are distinctly differentiated: orangeophilic cells and Alcian Blue (AB)-positive basophils. Because thyroxine and PTU could evoke changes in activity of the latter but had no effect on the orangeophils, it is concluded that the angular AB-positive basophils in the meso-adenohypophysis of Anoptichthys are responsible for the TSH production. Summary Staining sagittal sections of the pituitary of the Blind Mexican Cave Fish, Anoptichthys jordani, with a number of methods including Herlant's tetrachrome, Herlant's Alcian Blue (AB)-periodic acid Schiff (PAS)-orange G, Gabe's aldehyde fuchsin and lead hematoxylin according to Mac Conaill, made it possible to distinguish seven tinctorial cell types in the adenohypophysis: in the pro-adenohypophysis erythrosin-positive acidophils, and lead hematoxylin-positive amphiphils, lining branches of the neural lobe; in the meso-adenohypophysis orange G-positive acidophils, large, weakly AB- and AF-positive, strongly PAS- positive basophils, and smaller strongly AB- and AF-positive basophils; in the metaadenohypophysis lead hematoxylin-positive and -negative cells. Summary The ultrastructure of the secretory cells of the adenohypophysis of juvenile sockeye salmon was investigated. Pituitary glands were collected from immature fish transferred experimentally to sea water and subsequently returned to fresh water. The rostral pars distalis contained three cell types: ACTH cells, prolactin cells, and non-secretory cells. The prolactin and non-secretory cells were joined together in the form of follicles by desmosomes and they both had cilia and microvilli projecting into the follicle lumen. Various follicular structures such as lumen, multivesicular structures, and peripheral basement membrane are discussed as possible sites of prolactin cell granule release. The columnar ACTH cells were found at the junction of the rostral pars distalis and the neurohypophysis. The cytoplasmic granules in these cells were characteristically separated from their limiting membrane by a clear space. Multivesicular structures were also found in association with this cell type. The caudal pars distalis also contained three cell types: one acidophil (putative somatotrop) and two basophils (putative thyrotrops and gonadotrops), all of which were similar to those described in adult fish. The pars intermedia contained only one cell type. They appeared to be active cells and were characterized by containing membrane-bounded granules similar to those found in the ACTH cells. Changes in ambient salinity had no apparent effect on any cell type described.The work was supported by a grant in aid of research from the National Research Council of Canada. Prolactin receptors were monitored by measuring 125I-labeled prolactin binding to collagenase-dissociated mammary epithelial cells of lactating BALB/c mice. Specific receptors for iodine-labeled prolactin with an apparent dissociation constant (Kd) of 0.99 . 10(-9) M were present on the dissociated mammary cells. The binding was inhibited by ovine prolactin, human growth hormone and human placental lactogen but not by follicle stimulating hormone, luteinizing hormone, thyroid stimulating hormone, bovine growth hormone or insulin. Adrenal ablation of nursing mothers caused a reduction of the number of prolactin receptors and this effect was preventable by hydrocortisone therapy. Hydrocortisone injections to mothers 3 days after adrenalectomy also induced a replenishment of the prolactin receptors on the mammary cells. Injections of progesterone failed to sustain the high level of mammary cell prolactin receptors in adrenalectomized animals. Stimultaneous injections of hydrocortisone and progesterone to animals 3 days after adrenalectomy caused a partial suppression of the stimulatory action of hydrocortisone alone. The results suggest that hydrocortisone can exert a modulatory influence on mammary cell prolactin receptors in non-hypophysectomized post-partum mice without altering the dissociation constant (Kd) of the receptors.  相似文献

Normal fed and 2 days fasted Warren chickens were injected intravenously with 100 micrograms of ovine growth hormone (GH) and ovine prolactin and plasma concentrations of thyroid hormones were assayed prior and up to 2 h after injection. Fasting alone decreases T3, but increases T4. An injection of GH resulted in increases of plasma T3 concentrations in two fasting experiments by 40% (after 3/4 h) and 104% (after 1 h). In normal fed animals no increase is observed in the first experiment, whereas a 35% increase occurs in the second one. An injection of 100 micrograms prolactin does not influence T3 in normal fed or fasting animals. Both GH and prolactin, however, may decrease plasma concentrations of T4. In a separate experiment 50 micrograms and 200 micrograms of GH raised the decreased T3 levels after fasting by 39% and 60% respectively 1 h after injection and by 24 and 61% respectively in normal fed chicken, whereas prolactin was ineffective in this regard. Using Hisex chickens, the influence of an injection of 100 micrograms GH on plasma concentrations of thyroid hormones could be confirmed. At the same time GH increases the liver 5'-monodeiodinase activity by 330% after 1 h and by 147% after 2 h. The peroxidase activity is not influenced in normal fed chickens, but GH decreases this activity in food deprived animals after 1 h and 2 h. It is concluded that ovine GH, but not prolactin, stimulates the peripheral conversion of T4 into T3 in both normal fed and food deprived chicken and that this effect is dose dependent.  相似文献

Summary Normal and propylthiouracil (PTU) treated Xenopus laevis tadpoles were fixed during all stages of metamorphosis and sagittal sections were stained with aldehyde fuchsin (AF) or pseudoisocyanine (PIC). Whereas AF positive neurosecretory material could only be demonstrated in the preoptic nucleus from late prometamorphosis, increasing amounts of PIC positive material were found in cells of the dorsal part of the preoptic region from early premetamorphosis. The development of these cells correlated with that of the thyrotropic cells and the thyroids. Likewise, signs of hyperactivity in thyroids and thyrotropic cells of PTU treated larvae were accompanied by a depletion of the dorsal PIC positive cells. In the ventral preoptic region PIC positive cells developed from late prometamorphosis in control larvae, but failed to do so in PTU treated animals. It is argued that the differentiation of the PIC positive cells is largely dependent on thyroid hormones; that the dorsal PIC positive cells may produce a thyrotropin releasing factor; and that the function of these dorsal cells is inhibited by thyroid hormones.The authors thank Dr. P. Summary In the male black molly, Poecilia latipinna, morphological and functional aspects of the gonadotropic (GTH-)cells have been studied at the ultrastructural level. The cells exclusively occupy the ventral and lateral areas of the meso-adenohypophysis. In the black molly there is evidence of the presence of only one type of gonadotropic cell. In the GTH-cells of most specimens, the rough endoplasmic reticulum is weakly developed. The secretory vesicles are characterized by cores with varying diameters; this variation was not observed in the secretory vesicles of the other types of pituitary cells, except in the TSH-cells. After applying a histochemical method for the demonstration of polysaccharides, small black deposits appear in the core of the secretory vesicles of the GTH and TSH-cells only; this indicates the glycoproteinaceous nature of the hormones produced in these cells.Male black mollies treated with methyl-testosterone have significantly smaller GTH-cells and a lesser number of secretory vesicles and mitochondria in these cells. GTH-cell activity in Poeciliinae may be thus influenced by androgens by means of a negative feed-back mechanism. The GTH-cells are innervated by both type A and type B neurosecretory fibres. Summary The ultrastructure of the prolactin cells in nine-spined sticklebacks (Pungitius pungitius L.) with and without large pituitary cysts was compared. Prolactin cells from animals with cysts were less active as regards hormone synthesis and release than those from animals without cysts. Rough endoplasmic reticulum, mitochondria, Golgi apparatus and exocytotic figures were all more prominent in animals without cysts. Lysosomes and mature secretory granules were more plentiful in animals with cysts. The correlation between inactive prolactin cells and pituitary cysts argues against the hypothesis that cysts store prolactin released by holocrine secretion. It is more likely that the origin of cysts from degenerating cells signals extreme inactivity in the rostral pars distalis.  相似文献

Summary Tadpoles of Xenopus laevis were treated with propylthiouracil from the second half of prometamorphosis. Sagittal sections of the head region were stained a.o. with pseudoisocyanine. The goitrogen caused a degranulation of neurosecretory cells in the dorsal part of the preoptic region of the hypothalamus, suppressed the development of ventral neurosecretory cells and of the outer zone of the median eminence, stimulated the thyrotropic cells in the adenohypophysis, caused a hypertrophy of the thyroids, and impaired metamorphosis. Returning the animals to tap water had reciprocal effects and restored the normal activity of the hypothalamus, adenohypophysis and thyroid glands. It is concluded that thyroid hormones exert a morphogenetic influence upon hypothalamic centres and the outer zone of the median eminence and that a negative feed back relation exists between the thyroids on the one hand and the dorsal neurosecretory cells and the thyrotropic cells on the other.The author thanks Prof. Dr. P. The present communication deals with a histological study of the pituitary gland of the teleost fish Mugil cephalus , found in the estuarine waters of Cochin area. Six different cell types were identified in the pituitary gland on the basis of their grouping, distribution and staining properties. The prolactin and the TSH cells (thyroid stimulating hormone producing cells or thyrotrops) were identified in the rostral pars distalis and the ACTH cells (adrenocorticotropic hormone producing cells or corticotrops) in the interphase between the neurohypophysis and the rostral pars distalis. The STH cells (somatotropic hormone producing cells or somatotrops) and the gonadotropic cells were distinguished in the proximal pars distalis and the MSH cells (melanin stimulating hormone producing cells or melanotrops) in the pars intermedia.  相似文献

Summary The frontal brain was removed from premetamorphic larvae ofXenopus laevis. Part of the animals was kept in a 0.01% propylthiouracil (PTU) solution for 21, 30 or 90 days, together with unoperated controls. Others were reared in tap water for the same length of time. The hypothalamus, the pituitary and the thyroids were studied in sagittal sections stained with aldehyde fuchsin or pseudoisocyanine (PIC).Operated tadpoles lacking the telencephalon only, could not be distinguished from unoperated controls. Extirpation of the preoptic region of the hypothalamus lowered the activity of the thyrotropic (TSH) cells and of the thyroids, and blocked metamorphosis. In animals with intact PIC-positive cells in the dorsal preoptic region, PTU induced a degranulation of these cells, stimulated the TSH cells and caused a strong hypertrophy of the thyroids. It is concluded that these neurosecretory cells in the dorsal preoptic region produce a thyrotropin-releasing factor (TRF), indispensable for the endocrine regulation of metamorphosis, and that the thyroid hormones have a negative feed-back influence upon the TRF cells.In part of the animals lacking the rostral hypothalamus, new PIC-positive cells were observed to develop immediately behind the preoptic region. In these animals PTU was somewhat less effective than in the animals with an intact dorsal preoptic region. This means either that the newly differentiated PIC-positive cells have a TRF function or that the thyroids can exert a direct negative feed-back upon the TSH cells. The latter possibility is supported by the fact that even in the absence of any neurosecretory cell PTU stimulated the thyroids.Blocking of thyroid hormone production by PTU inhibited the differentiation of the hypothalamus, of the median eminence and of the portal vessels. Summary Cells binding anti-bovine TSH serum were found exclusively in the rostral lobe of the adenohypophysis of the drake using the peroxidase-antiperoxidase complex unlabelled antibody method. The specificity of the binding of the anti-serum to TSH cells was established by relating the morphology and relative abundance of immunochemically stained cells to the TSH content of the adenohypophysis after experimentally altering the activity of the pituitary-thyroid axis. The TSH activity of the adenohypophysis was assessed indirectly, by the weight of the thyroid glands, and directly, by bioassay. As determined by bioassay, the TSH content of the rostral lobe of the adenohypohysis was much greater than that of the caudal lobe. Compared with control drakes, immunochemically stained cells in birds fed a goitrogen, methimazole, seemed to be enlarged and were closer together, while the stained cells in drakes injected with thyroxine were shrunken and less intensely stained. The TSH content of the adenohypophysis was increased in drakes fed methimazole. Castration did not alter the TSH content of the adenohypophysis or change the morphology of immunochemically stained cells. These observations suggest that in the drake: 1) anti-bovine TSH serum binds specifically to TSH cells; 2) the TSH cells occur in the rostral and not in the caudal lobe of the adenohypophysis; and 3) the activity of TSH cells is not inhibited by the feedback effects of gonadal steroids.We thank Dr. L.E. The mucous gland of the red-spotted newt, Notophthalamus viridescens viridescens, Rafinesque was examined by histochemical and ultrastructural techniques and its cytological responses to various hormonal conditions were studied. Its secretory epithelial cells produce and release in merocrine fashion a neutral, unsulphated mucosubstance. The secretory epithelium is bounded peripherally by a thin, but apparent non-functional, myo-epithelium. The duct of this mucous gland consists of a single keratinized tubular cell that extends from the neck region of the gland to the surface of the epidermis. Mucous secretion is absent or greatly reduced on the skins of newts maintained under laboratory conditions for a few weeks but reappears after injection of ovine prolactin. Mucous glands in laboratory conditioned animals show a 4-fold increase in volume brought about by the engorgement of their epithelial cells with secretory granules. Ovine prolactin reduces the volume of the glands to unconditioned levels with a corresponding reduction in granular content, suggesting that prolactin functions in the release of the granules. This view is reinforced by the findings that autotransplantation of the pituitary gland prevents the conditioning effect and that glandular volume increases in auto-transplanted animals given ergocornine. Granular accumulation begins also in hypophysectomized newts but ceases after a week, indicating the need for some hypophyseal factor in the synthesis as well as the release of the granules. Ovine prolactin restores mucous glands of hypophysectomized newts to the unconditioned state. Contrary to earlier findings, ovine prolactin induces a reduction in the volume of the mucous gland in thyroidectomized newts.  相似文献

Summary In the eel, ovine prolactin (oPrl) treatment (0.018 IU/day·g body weight), for 8 to 13 days modifies neither iodide absorption from the water nor excretion, extrathyroidal metabolism and plasma level of iodide.Thyroid activity, evaluated by epithelial cell height, radioiodine uptake and absolute iodide uptake is approximately twice that of controls. However, the amounts of total iodine, thyroxine (T4) and triiodothyronine (T3) in thyroid are unaltered by oPrl. Therefore, the decrease of plasma T4 and the increase of plasma T3, previously observed in oPrl-treated eels, do not result from a preferential thyroidal secretion of T3, but only from a stimulation of peripheral conversion of T4 to T3. Summary In the alewife the orohypophyseal duct, a remnant of Rathke's pouch, persists in adults as a tube passing from the rostral pars distalis to the pharyngeal region. Its lumen is not open to the buccal cavity. The prolactin cells are situated around the bifurcations of this duct in the rostral pars distalis. Contents from prolactin cells, such as granules, nuclei, mitochondria and Golgi structures were found in these bifurcations. These contents were indistinguishable from those of intact prolactin cells. Evidence of actual release into the duct was often noted. At the presumptive point of release, the cells lining the lumen separate and the contents, probably of an entire prolactin cell, are extruded. The cilia usually found at the point of extrusion arise from prolactin cells. The prolactin cells of freshwater fish were more heavily granulated than those from a marine environment. Prolactin cells of fish entering freshwater streams were not heavily granulated but showed evidence of increased activity. Granule size was not affected by salinity. The ACTH cells are arranged in bands along branches of the neurohypophysis in the rostral pars distalis. Summary The relative volume of the RPD region of the pituitary gland of Gasterosteus aculeatus form leiurus was greater in animals collected in the spring than in winter. A morphometric analysis of the prolactin cells from spring animals showed only slight changes in cell ultrastructure compared with winter animals. However, in spring the prolactin cells apparently formed and released more secretory granules. Three distinct sites for the release of secretory granules are described. A preliminary study of the response of prolactin cells from spring animals to 70% sea water did not reveal any large scale changes in cell ultrastructure. The significance of this finding is discussed. It is concluded that although the freshwater stickleback cannot be regarded as physiologically hypophysectomised: with regard to prolactin secretion in the winter, it still shows a seasonal change in prolactin cell ultrastructure.This work formed part of a thesis submitted for the degree of Doctor of Philosophy in 1973 and for which the author was in receipt of an S.R.C. studentship. Summary Pituitaries of the African catfish (Clarias lazera) were studied with immunocytochemical methods, at the light-microscopic and ultrastructural levels, for the characterization and localization of gonadotropic and thyrotropic cells. Two immunostaining procedures with the use of different markers were carried out: (i) with peroxidase-antiperoxidase, (ii) with protein A-gold. In routinely stained sections for light microscopy two types of basophils were identified in the proximal pars distalis: (1) large, round, purple cells, and (2) small, angular, light-blue cells. Both types were immunolabeled with antibodies against Clarias ,-gonadotropin (GTH) and salmon G100-GTH. Only the large basophils were immunolabeled with anti-carp -GTH, whereas the small basophils were the only cells immunolabeled with anti-human thyrotropin beta subunit (anti-h TSH-). It was concluded that the large basophils represent the gonadotrops and the small basophils the thyrotrops. At the ultrastructural level the immunostaining of the GTH-cells was confined to three types of inclusions: (i) secretory vesicles, (ii) globules, and (iii) electron-dense, membrane-bound irregular masses. Especially the protein A- gold method, in combination with the use of a highly diluted homologous antiserum, resulted in a distinct localization of GTH. The presence of two types of nerve fibres, synaptically contacting the gonadotrops, is discussed with regard to the presence of a peptidergic (stimulatory) and an aminergic (inhibitory) control of GTH-secretion.Part of the results were presented as a poster at the IXth conference of the European Society of Comparative Endocrinology, Jerusalem, August 1981, and published as an abstract (Peute et al. 1982)  相似文献
