Stability properties of the barren state of a kelp forest-sea urchin system were studied in northern Norway. The ability of the sea urchin Strongylocentrotus droebachiensis to maintain high population densities and recover from perturbations, and the succession of kelp forest revegetation, were studied experimentally by reducing the sea urchin density on a barren skerry. Additional information was obtained from community changes following a natural, but patchy, sea urchin mortality that varied between sites. On the barren grounds, high sea urchin densities (30 50 per m2) is maintained by annual recruitment. Severe reductions of sea urchin densities initiated luxuriant kelp growth, while more moderate reductions allowed establishment of opportunistic algae (during spring and early summer), but no kelps. Succession of algal growth, after the severe decline in sea urchin densities, followed a predictable pattern. At first the substrate was colonized by filamentous algae, but within few weeks they were outcompeted by the fast growing kelp Laminaria saccharina. After 3–4 years of the removal experiment, the slower-growing, long-lived kelp L. hyperborea became increasingly dominant. Increased food availability after reduction in sea urchin density led to increased individual growth of the remaining sea urchins. However, the population density did not increase, neither from recruitment nor immigration from adjacent areas with high sea urchin densities. Possibly, early establishment of a dense kelp stand, may represent a breakpoint in the ability of sea urchins to reestablish a barren state. The ability of L. saccharina quickly to invade and monopolize an area may have both positive and negative effects on the succession towards the climax L. hyperborea kelp forest. Competitive interactions may slow the process, but development of a dense stand of L. saccharina will also reduce grazing risk on scattered recruits of the more slowly growing L. hyperborea.  相似文献

Summary Fibronectin, with a subunit molecular weight of 220,000 daltons, was isolated from the ovary of the sea urchin,Pseudocentrotus depressus, using affinity chromatography on heat-denatured mammalian collagen coupled to Sepharose 4B. The distribution of fibronectin in the sea urchin ovary was examined by indirect immunofluorescence using antifibronectin serum. The basement membrane and the connective tissues exhibited strong fluorescence. The fibronectin was localized closely together with collagen bundles in the sea urchin ovary. Biochemical and immunological examinations indicate that sea urchin fibronectin has similar properties as those of mammalian fibronectin.  相似文献

Trophic cascade hypotheses for biological communities, linking predation by upper trophic levels to major features of ecological structure and dynamics at lower trophic levels, are widely subscribed and may influence conservation policy. Few such hypotheses have been evaluated for temporal or spatial generality. Previous studies of sea otter (Enhydra lutris) predation along the outer coast of North America suggest a pattern, often elevated to the status of paradigm, in which sea otter presence leads to reduced sea urchin (Strongylocentrotus spp.) biomass and rapid increases in abundance and diversity of annual algal species, followed by a decline in diversity as one or a few perennial algal species become dominant. Both sea otter predation and commercial sea urchin harvest are ecologically and economically important sources of urchin mortality in nearshore benthic systems in northern Washington marine waters. We recorded changes in density of macroalgae in San Juan Channel, a marine reserve in the physically protected inland waters of northern Washington, resulting from three levels of experimental urchin harvest: (1) simulated sea otter predation (monthly complete harvest of sea urchins), (2) simulated commercial urchin harvest (annual size-selective harvest of sea urchins), and (3) no harvest (control). The two experimental urchin removal treatments did not significantly increase the density of perennial (Agarum and Laminaria) or annual (Desmarestia, Costaria, Alaria and Nereocystis) species of macroalgae after 2 years, despite significant and persistent decreases in urchin densities. Our results suggest that other factors such as grazing by other invertebrates, the presence of dense Agarum stands, and recruitment frequency of macroalgae and macroinvertebrates may play a large role in influencing community structure in San Juan Channel and other physically protected marine waters within the range of sea otters. This study evaluates and projects the effect of experimental removal of two species of wrasses, Thalassoma bifasciatum and Halichoeres bivittatus, on the demographic structure of the Caribbean sea urchin Diadema antillarum. For census periods ‘before’ and ‘after’ fish removal at treatment and un-manipulated control sites, size-based matrix population projections revealed the most important change in the sea urchin demography was increased survival of the medium size-class following removal of wrasses. The asymptotic growth rate (λ) exhibited no differences between periods for the control; however, the treatment displayed a significant increase in λ from 0.94 to 1.0. During the before period, the treatment population displayed lower λ than the control population, indicating site differences in urchin recruitment from the outset of the experiment, however after one year of maintenance of the predator removal treatment, the treatment population exhibited and increased growth rate to become similar to the control population; indicating predatory-release for the treatment population. Physiological status of sea urchins, as determined by righting times, exhibited no difference between treatment and control populations, or through time. Nil correlation was evident between righting activity and urchin size, indicating that urchin physiological status was not influenced by predation. Long-term demographic simulation indicated that the sea urchin population growth at the treatment site was negative and thus unviable predatory wrasse. However, only one year after fish removal, sea urchin population growth rate became positive. Therefore local population recovery for D. antillarum appears enhanced when abundance of wrasses is kept low.  相似文献

Peroxisomes are essential subcellular organelles that appear to be derived from pre-existing organelles. To test the presence of peroxisomes in sea urchin (Tetrapigus niger) sperm and eggs, we performed biochemical and morphological experiments to evaluate the subcellular distribution of catalase as the typical peroxisomal marker. In sea urchin sperm, we found that catalase is localized in the cell cytosol. In contrast, sea urchin eggs contain sedimentable catalase, presumably contained in peroxisome-like structures detected by immunomicroscopy and by cytochemistry. Our results show, for the first time, evidence for the presence of peroxisome-like structures in sea urchin eggs and provide evidence for the peroxisome biogenesis hypothesis by division of pre-existing organelles.  相似文献

Participatory management approaches are increasingly recognized as an effective strategy for enabling the sustainable use of natural resources. The southeast coast of St. Lucia is one of the sites where a particular form of participatory management, a co-management regime, was recently developed to control the sea urchin fishery. The Caribbean Natural Resources Institute (CANARI), a NGO based in Vieux-Fort, St. Lucia, played a key role in the development of this co-management arrangement. This case study of the sea urchin fishery in Vieux-Fort examines the extent of the devolution of authority to locally-based sea urchin harvesters, explores the potential contribution of local knowledge to the understanding of sea urchin behavior, and points to elements of a strategy aimed at strengthening the organizational capacity of the core group of sea urchin harvesters. The study addresses both present practice and future possibilities in response to concrete questions raised by participants in the study.  相似文献

Reporter genes have been used as a powerful tool to analyze cis-regulatory elements responsible for temporal and spatial expression in the early development of sea urchin. However, here we show that the transgenes introduced into the sea urchin embryos undergo suppression in larval stage. The transgene silencing could be one of major obstacle in the analysis of regulatory regions in the late stages of development. We previously demonstrated that a DNA fragment (ArsI) located in the upstream region of sea urchin (Hemicentrotus pulcherrimus) arylsulfatase gene has the property of an insulator. We show that tandem ArsI prevents silencing of a transgene in sea urchin larvae when the ArsI is fused to the 5′ end of the reporter gene. Furthermore, we demonstrate that DNA of the reporter gene introduced into the sea urchin eggs is methylated during development and that the ArsI protects the transgene from the DNA methylation.  相似文献

Sea urchin gametes and embryos serve as a model system to evaluate toxicity in the marine environment. In this study, the toxicity of complex chemical mixtures in leachate samples to sea urchin development was examined with a focus on ammonia, which was the main contaminant of concern in most samples. Two rapid tests, the submitochondrial particle function and bacterial luminescence tests, were also used. Ammonia is highly toxic to sea urchin embryos with an EC50 of 1.3 mg l−1 for the embryos of the Australian sea urchin Heliocidaris tuberculata. Leachate ammonia levels were well above these EC50 concentrations. To assess the contribution of ammonia to leachate toxicity in sea urchin development, we compared the predicted toxic units (PTU) and observed toxic units (OTU) for ammonia for each sample. The PTU/OTU comparison revealed that the sensitivity of the sea urchin embryos to ammonia were altered (enhanced or decreased) by other chemicals in the leachates. This result emphasises the need for parallel chemical analyses and a suite bioassays for evaluating the toxicity of complex and variable chemical mixtures.  相似文献

Ongoing changes along the northeastern Atlantic coastline provide an opportunity to explore the influence of climate change and multitrophic interactions on the recovery of kelp. Here, vast areas of sea urchin‐dominated barren grounds have shifted back to kelp forests, in parallel with changes in sea temperature and predator abundances. We have compiled data from studies covering more than 1,500‐km coastline in northern Norway. The dataset has been used to identify regional patterns in kelp recovery and sea urchin recruitment, and to relate these to abiotic and biotic factors, including structurally complex substrates functioning as refuge for sea urchins. The study area covers a latitudinal gradient of temperature and different levels of predator pressure from the edible crab (Cancer pagurus) and the red king crab (Paralithodes camtschaticus). The population development of these two sea urchin predators and a possible predator on crabs, the coastal cod (Gadus morhua), were analyzed. In the southernmost and warmest region, kelp forests recovery and sea urchin recruitment are mainly low, although sea urchins might also be locally abundant. Further north, sea urchin barrens still dominate, and juvenile sea urchin densities are high. In the northernmost and cold region, kelp forests are recovering, despite high recruitment and densities of sea urchins. Here, sea urchins were found only in refuge habitats, whereas kelp recovery occurred mainly on open bedrock. The ocean warming, the increase in the abundance of edible crab in the south, and the increase in invasive red king crab in the north may explain the observed changes in kelp recovery and sea urchin distribution. The expansion of both crab species coincided with a population decline in the top‐predator coastal cod. The role of key species (sea urchins, kelp, cod, and crabs) and processes involved in structuring the community are hypothesized in a conceptual model, and the knowledge behind the suggested links and interactions is explored.  相似文献

Recurrent sea urchin mass mortality has recently affected eastern Atlantic populations of the barren‐forming sea urchin Diadema africanum. This new episode of die‐off affords the opportunity to determine common meteorological and oceanographic conditions that may promote disease outbreaks. The population dynamics of this sea urchin species are well known—urchin barrens have persisted for many decades along most of the coastlines off the archipelagos of Madeira, Selvages, and the Canary Islands, where they limit macroalgae biomass growth. However, this new and explosive mortality event decimated the sea urchin population by 93% on Tenerife and La Palma Islands. Two severe episodes of southwestern rough sea that led to winter storms, in February 2010 (Xynthia) and February 2018 (Emma), preceded both mass mortality events. The autumn and winter months of those years were anomalous and characterized by swells with an average wave height above 2 m that hit the south and southwest sides of the islands. The amoeba Paramoeba brachiphila was the only pathogen isolated this time from the moribund and dead sea urchins, suggesting that the amoeba was the primary cause of the mortality. This new sea urchin die‐off event supports the “killer‐storm” hypothesis that has been already described for western Atlantic coasts. These anomalous southwest storms during winters generate pronounced underwater sediment movement and large‐scale vertical mixing, detected in local tide gauge, which may promote paramoebiasis. This study presents valuable insights about climate‐mediated changes in disease frequency and its impacts on the future of coastal marine ecosystems in the Atlantic.  相似文献

The actin gene family of the sea urchin Lytechinus pictus includes a single muscle actin gene, LpM, and four cytoskeletal actin genes: LpC1, LpC2, LpC3, and LpC4. The origin and relationship of these actin genes to members of the actin gene family of the sea urchin Strongylocentrotus purpuratus were considered. Comparison of deduced amino acid sequences suggested a close relationship between LpC1 and the CyI–CyII subfamily of S. purpuratus actin genes, and between LpC2 and the CyIII subfamily of S. purpuratus actin genes; the muscle actin genes were orthologous. It is proposed that two divergent cytoskeletal actin genes of the common ancestral sea urchin gave rise by duplication to the extant cytoskeletal actin genes of these species, some of which have changed 3 noncoding sequences while others have maintained a terminus highly conserved among sea urchin actin genes.Correspondence to: B.P. The hemocytes/coelomocytes DNA content in five selected marine invertebrates (sea mouse Aphrodita aculeata, spiny crab Maja crispata, sea star Echinaster sepositus, sea urchin Paracentrotus lividus, and tunicate Phallusia mammillata) was investigated by flow cytometry. The cell cycle analyses identified sea mouse coelomocytes as proliferating cells and revealed that spiny crab hemocytes and sea urchin coelomocytes complete their division in the hemolymph and coelom, respectively. The genome sizes of sea mouse and spiny crab are reported for the first time. The diploid DNA content (2C) in sea mouse A. aculeate was 1.24 pg, spiny crab M. crispata 7.76 pg, red starfish E. sepositus 1.52 pg and sea urchin P. lividus 1.08 pg. The mean diploid DNA content in tunicate P. mammillata was 0.11 pg with a high interindividual variability (45%). The presented results provide a useful database for future studies in the field of invertebrate physiology, ecotoxicology, biodiversity, species conservation and phylogeny.  相似文献

Background  

Vaults are intriguing ribonucleoprotein assemblies with an unknown function that are conserved among higher eukaryotes. The Pacific coast sea urchin, Strongylocentrotus purpuratus, is an invertebrate model organism that is evolutionarily closer to humans than Drosophila and C. elegans, neither of which possesses vaults. Here we compare the structures of sea urchin and mammalian vaults and analyze the subcellular distribution of vaults during sea urchin embryogenesis.  相似文献

Summary New methods have been applied to the determination of single copy DNA sequence differences between the sea urchin speciesStrongylocentrotus purpuratus, S. franciscanus, S. drobachiensis, andLytechinus pictus. The thermal stability of interspecies DNA duplexes was measured in a solvent (2.4 M tetraethylammonium chloride) that suppresses the effect of base composition on melting temperature. The lengths of duplexes were measured after digestion with S1 nuclease and correction made for the effect of length on thermal stability. The degree of base substitution that has occurred in the single copy DNA during sea urchin evolution is significantly larger than indicated by earlier measurements. We estimate that 19% of the nucleotides of the single copy DNA are different in the genomes of the two sea urchin congeners,S. purpuratus, andS. franciscanus, which apparently diverged only 15 to 20 million years ago.  相似文献

Epithelial‐mesenchymal transition (EMT) is an evolutionarily conserved cellular program, which is a prerequisite for the metastatic cascade in carcinoma progression. Here, we evaluate the EMT process using the sea urchin Paracentrotus lividus embryo. In sea urchin embryos, the earliest EMT event is related to the acquisition of a mesenchymal phenotype by the spiculogenetic primary mesenchyme cells (PMCs) and their migration into the blastocoel. We investigated the effect of inhibiting the epidermal growth factor (EGF) signaling pathway on this process, and we observed that mesenchyme cell differentiation was blocked. In order to extend and validate our studies, we investigated the migratory capability and the level of potential epidermal growth factor receptor (EGFr) targets in a breast cancer cell line after EGF modulation. Altogether, our data highlight the sensitivity of the sea urchin embryo to anti‐EMT drugs and pinpoint the sea urchin embryo as a valuable in vivo model system for studying EMT and the screening of anti‐EMT candidates.  相似文献

The sea urchin provides a relatively simple and tractable system for analyzing the early stages of embryo development. Here, we use the sea urchin species, Paracentrotus lividus, to investigate the role of Alix in key stages of embryogenesis, namely the egg fertilization and the first cleavage division. Alix is a multifunctional protein involved in different cellular processes including endocytic membrane trafficking, filamentous (F)‐actin remodeling, and cytokinesis. Alix homologues have been identified in different metazoans; in these organisms, Alix is involved in oogenesis and in determination/differentiation events during embryo development. Herein, we describe the identification of the sea urchin homologue of Alix, PlAlix. The deduced amino acid sequence shows that Alix is highly conserved in sea urchins. Accordingly, we detect the PlAlix protein cross‐reacting with monoclonal Alix antibodies in extracts from P. lividus, at different developmental stages. Focusing on the role of PlAlix during early embryogenesis we found that PlAlix is a maternal protein that is expressed at increasingly higher levels from fertilization to the 2‐cell stage embryo. In sea urchin eggs, PlAlix localizes throughout the cytoplasm with a punctuated pattern and, soon after fertilization, accumulates in larger puncta in the cytosol, and in microvilli‐like protrusions. Together our data show that PlAlix is structurally conserved from sea urchin to mammals and may open new lines of inquiry into the role of Alix during the early stages of embryo development.  相似文献

The sea urchin,Tripneustes gratilla, which feeds mainly on living leaves of the seagrass,Thalassia hemprichii, was studied in its habitat on the southern coast of Papua New Guinea, and its roles in decomposition and nutrient cycling in a seagrass bed were assessed through the excretion of ammonium and metabolism of feces produced by the sea urchin. Carbon content of the fresh feces (21% of dry weight) was similar to that of intact dead leaves of the same species (22–23%). Carbon/nitrogen and carbon/phosphorus ratios of the feces (21.7 and 466, respectively), however, were significantly lower than those of the dead leaves (25.9–27.7 and 656–804, respectively), indicating that the feces retain more nitrogen and phosphorus in comparison with carbon. Net consumption of ammonium and orthophosphate typically concurred with oxygen consumption during dark incubation of both the dead leaves and the sea urchin feces. Compared with the same oxygen consumption rate, however, the dead leaves consumed more orthophosphate than the feces. Under sunlight, dead leaves showed a net accumulation of carbon by epiphytic algae, while the feces showed a carbon loss. Ammonium excretion by this sea urchin (1.7–5.4 mg nitrogen/individual/day) would thus appear to make a significant contribution to nitrogen recycling since biological communities associated with dead leaves and sea urchin feces tend to demand an external supply of nitrogen, such as ammonium.  相似文献

This study revealed a new lectin (MBL-SN) in the coelomic fluid of the sea urchin Strongylocentrotus nudus. Based on the peculiarities of molecular structure and carbohydrate specificity, MBL-SN can be assigned to the mannan-binding lectin family. Using polyclonal monospecific rabbit antibodies against MBL-SN, the presence of MBL-SN in the sea urchin was detected in two forms: a soluble form dissolved in the coelomic fluid and an extracellular matrix-bound form. The biosynthesis site of this lectin may be one of the subpopulations of morula cells-coelomic fluid cells that perform heterosynthesis. Our results demonstrate the similarity of the sea urchin lectin MBL-SN to the previously investigated MBLs of the holothurians Cucumaria japonica and Apostichopus japonicus, and suggest a similarity to MBLs of vertebrates, which also have soluble and bound forms.  相似文献
