分离并测定黄柏中盐酸药根碱、盐酸巴马汀、盐酸小檗碱的含量.方法:Shim-Pack型C18柱,0.5% w/v SDS-Britton-Robinson缓冲液 pH2.5 -乙腈-三乙胺 0.5∶56∶34∶0.5 为流动相,检测波长:345nm;柱温:40℃;流速1mL·min-1.盐酸药根碱回归方程为Y=38315164.0X-1897.7,r=0.9991 n=6 ,线性范围0.05~0.30μg,平均回收率为95.90%,RSD=3.10%;盐酸巴马汀回归方程为Y=1982872.0X-2471.1,r=0.9998 n=6 ,线性范围0.09~0.54μg,平均回收率100.2%,RSD=2.01%;盐酸小檗碱回归方程为Y=9450164.2X-3456.9,r=0.9999 n=6 ,线性范围0.50~3.00μg,平均回收率103.1%,RSD=1.12%;结果表明:在以上条件下,盐酸药根碱、盐酸巴马汀、盐酸小檗碱成分可以完全分离,并测定了黄柏药材含量.本法可用于黄柏药材、饮片、含黄柏中成药的质量控制.  相似文献

建立同时测定桑叶中绿原酸、芦丁和槲皮素含量的分析方法。色谱柱为NUCLEODUR C18 RP(250 mm×4.6 mm,5μm),光电二极管检测器,流动相为甲醇-质量分数0.5%磷酸水溶液,梯度洗脱程序为0 min(30:70)-15min(30:70)-25min(50:50)-35min(85:15)-40min(30:70);流速0.8 ml.min-1,检测波长为350 nm。绿原酸、芦丁和槲皮素的线性范围分别为0.1144~1.0296μg(r=0.9996)、0.0436~0.3924μg(r=0.9999)和0.0452~0.4068μg(r=0.9997),平均回收率分别为97.7%(RSD=1.7%)、98.4%(RSD=2.2%)和100.6%(RSD=1.5%)。方法快速简便,专属性强,重复性好,可作为桑叶中绿原酸、芦丁和槲皮素的定量分析方法。  相似文献

采用HPLC-ELSD法,Prevail C18柱,体积分数0.06%HFBA溶液-乙腈(88∶12)为流动相,柱温30℃,流速为0.8 ml.min-1,ELSD漂移管温度110℃,气体流量2.8 L.min-1,建立了测定L-鸟氨酸的方法。鸟氨酸在0.1-0.6 g.L-1范围内,线性回归比非常显著(r2=0.9998),平均回收率98.94%。该法简便、快速、准确性高、重复性好,适合于L-鸟氨酸的定量分析。  相似文献

目的:建立以高效液相色谱法测定经舒胶囊中丹皮酚含量的方法。方法:采用Platisil ODS色谱柱(4.6 mm×250 mm 5μm);以甲醇-水(45:55)为流动相;柱温:40℃;流速:1.0ml.min-1;检测波长:274nm。结果:在建立的色谱条件下,丹皮酚进样量在0.08487~0.50922μg范围内与峰面积呈良好的线性关系(r=0.9999),平均回收率(n=6)为98.75%(RSD=1.74%)。结论:方法简便可靠,分离度好,可用于经舒胶囊的质量控制。  相似文献

目的:建立测定人血浆中关利曲辛浓度的高效液相串联质谱法(HPLC-MS/MS),用于氟哌噻吨美利曲辛片的生物等效性研究.方法:以Agilent ZORBAX Eclipse Plus C18(4.6mm× 150mm,5μm)为色谱柱,流动相为乙腈(含1%甲酸):0.02mo·L-1甲酸铵水溶液(80∶20,V∶),流速:0.8mL·min-1;柱温:40℃,以醋酸乙酯:二氯甲烷(4∶1,V∶V)为提取剂.样品经电喷雾离子源正离子化后,通过三重四级杆串联质谱仪,采用选择反应监测(SRM)对美利曲辛(m/z 292.2→232.2)和阿米替林(m/z 278.1-91.0)进行测定.结果:美利曲辛的高(50μg·L-1)、中(20μg·L-1)、低(0.5μg·L-1)3个浓度的平均回收率分别为97.53%、104.03%和106.87%,日内(n=5)、日间(n=3)RSD均小于15%;分析方法的最低定量限为0.2μg·L-1.线性范围为:0.2~60μg·L-1,回归方程为:F=1.8691ρ+0.0555,r=0.9986(n=9),权重为1/ρ2.结论:该方法灵敏、准确、简单、快速,可用于临床血浓监测和药动学研究.  相似文献

本文建立了黄芩黄酮总苷元提取物中总黄酮和3种主要成分的含量测定方法。采用HPLC法测定黄芩素、汉黄芩素和千层纸素A的含量。使用Agilent 1200型高效液相色谱仪,Eclipse XDB-C18(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-0.2%磷酸水(50∶50),流速1 m L/min,柱温35℃,检测波长275 nm的方法。黄芩素、汉黄芩素和千层纸素A线性范围分别为3.501~70.016μg/m L(r=0.9999),0.832~16.640μg/m L(r=0.9998),0.418~8.352μg/m L(r=0.9999);平均加样回收率(n=6)分别为99.55%、98.86%、9.997%,RSD分别为1.74%、1.74%、1.70%。采用UV法测定总黄酮含量,黄芩素线性范围为1.71~8.54μg/m L(r=0.9991);平均加样回收率(n=6)为100.93%,RSD为2.63%。黄芩素、汉黄芩素、千层纸素A和总黄酮的含量分别为48.22%~60.36%、12.09%~14.46%、5.05%~8.81%、78.11%~90.12%;研究结果表明该方法准确、可靠,适用于黄芩黄酮总苷元提取物的含量测定。  相似文献

当年8月份至次年3月份采集紫花含笑(♀)×钙土含笑(♂)杂种F1代优良单株的顶芽和侧芽,置于H+0.25mg·L-16-BA+0.125mg·L-1NAA+0.10mg·L-1KT+0.05mg·L-1IAA上培养,不定芽诱导率可达115%;诱导的不定芽在H+0.3mg·L-16-BA+0.3mg·L-1KT培养基中增殖及生长较好。经过壮苗后的芽条在H+2.8mg·L-1NAA上的生根率可达100%,且单株根数多,芽条生长健壮。间苯三酚(phloroglucinol)明显促进根的发生,但对根的后期生长有一定的抑制效应。组培苗移栽至红土:腐殖土:珍珠岩=1:1:1的混合基质中,60d时成活率可达90%左右。  相似文献

目的:建立RP-HPLC法测定盐酸托莫西汀的含量和有关物质.方法:采用Kromasil C18(4.6mm×250 mm,5μm)色谱柱,流动相:乙腈-水-三乙胺溶液(40:60:0.5,冰醋酸调pH至6.0),流速:1.0 mL·min-1,检测波长:270 nm,柱温:室温.结果:有关物质与盐酸托莫西汀分离良好,盐酸托莫西汀在2.0~50.0mg·L-1 范围内线性关系良好(r=:0.9998),平均回收率为99.8%(RSD=1.7%),检测限为80 μg·L-1(S/N=3).盐酸托莫西汀与有关物质分离良好.结论:该方法操作简便、重复性好、专属性强,可用于测定盐酸托莫西汀的含量和有关物质.  相似文献

目的:建立同时测定龙胆泻肝制剂中黄芩苷、汉黄芩苷、黄芩素和汉黄芩素4种有效成分含量的HPLC分析方法.方法:色谱柱:Dikma Diamonsil C18柱(250 mm×4.6 mm,5μm);流动相:乙腈- 0.05%磷酸溶液,梯度洗脱;波长:278 nm.结果:黄芩苷、汉黄芩苷、黄芩素和汉黄芩素的线性范围分别为0.418~4.18μg(r=0.9999),0.2~2μg(r=0.9999),0.233~2.33 μg(r=0.9999),0.215~2.15(r=0.9998).2种制剂4种成分的平均回收率分别为98.10% (RSD=0.59%),98.05%( RSD=1.37%),98.88%( RSD=1.44%),98.10%( RSD =0.59%)和98.40% (RSD=0.46%),99.33% (RSD=1.74%),99.09%( RSD=1.22%),98.82%(RSD=1.42%).结论:该方法快速、简便、准确、具有良好的重复性和回收率,可作为龙胆泻肝制剂中这4种成分的含量测定方法.  相似文献

在暗培养条件下,黄芩愈伤组织生长和黄酮类物质积累的最佳培养条件为:基本培养基MS中的氮源浓度为60mmol·L-1(NH4+:NO3-为1:1),KH2PO4为1.5mmol·L-1,附加80g·L-1蔗糖、0.3mg·L-1IAA、2mg·L-16-BA和200mg·L-1蛋白胨,培养温度为(25±1)℃。培养40d后,愈伤组织生物量为28.7g·L-1,总黄酮产量达10.2g·L-1。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
