Development of diurnal rhythm in some metabolic parameters in foals   总被引:1,自引:0,他引:1  
1. The development of diurnal rhythm activity of FDPA, AspAT and A1AT and in levels of cortisol, T3 and T4 was observed in the blood serum of six foals. 2. The studies began when a foal was 7 days old and were repeated every month until foals reached 1 year of age. Blood samples were taken every 4 hr for one day each month. 3. As a control group four barren mares were used, kept and examined in the same conditions. 4. In mature mares, diurnal rhythms in activity of A1AT (acrophase at 2200 hr), AspAt (2400 hr) and cortisol (0630 hr) but in T3 only in summer months (acrophase at 0100 hr) were observed. 5. During the first 6 months of foal life, significantly higher mean levels of FDPA, A1AT, T3 and T4 than in control mares were found. 6. The cortisol level in foals was half as much as that of mature mares throughout the year. 7. In foals the diurnal rhythm in A1AT activity occurred in the 5th month and in AspAt--in the 12th month (acrophase at 2400 hr), but in cortisol levels it was developed already in the second month of foal life (acrophase at 0830 hr).  相似文献

1. The diurnal changes in the level of total protein, cortisol, T3 and T4 were studied in four barren and four pregnant standard-bred mares, kept and examined under the same conditions. 2. Blood samples were taken every 4 hr. for one day each month, throughout one year. 3. In barren mares, a diurnal rhythm in cortisol level (acrophase at 0530 hr in summer and at 0830 hr in winter) and in T3 level (acrophase at 1330 hr in summer and at 1800 hr in winter) was found. 4. In pregnant mares, a diurnal rhythm in cortisol level only till 5th month of pregnancy was observed. 5. A diurnal rhythm in T3 level was found throughout the pregnancy, with acrophase always at 1400 hr. 6. No diurnal rhythm in the total protein content and in the T4 level was observed. 7. In both groups of mares the seasonal cyclicity in T3 and T4 levels were found. A seasonal cyclicity in cortisol level was found only in pregnant mares. 8. Pregnancy abolished seasonal cyclicity in total protein and showed it in cortisol level. 9. Pregnancy in mares modifies diurnal rhythms as well as seasonal cycles in secretion and metabolism of the hormones studied.  相似文献

In 34 pure breed Arabian horses divided into four groups (Gr. I--10 pregnant mares, Gr. II--7 barren mares, Gr. III--10 foals born in 1981, Gr. IV--7 foals born in 1982) seasonal changes in total blood serum protein, its electrophoretic fractions and the activity of AspAT and AlAT were studied. Seasonal cyclicity was found in all groups in the amount of total serum proteins, and alpha 2- and beta 1-globulin fractions. Cyclicity was found in the level of albumin and activity of AspAT in three groups, not Gr. II, and in gamma-globulin, not Gr. IV. beta 2-globulin and AlAT cyclicity was found in two groups and alpha 1-globulin cyclicity was found only in Gr. II. Out of nine indices studied, cyclicity was found in eight of them in pregnant mares, Gr. I; in seven in older foals, Gr. III; in six in the young foals, Gr. IV; and in five in barren mares, Gr. 1. In six standard-bred mares and their foals diurnal changes in the cortisol and glucose levels and in FDPA activity were studies for 13 weeks of foal life. 2. In the cortisol level diurnal rhythm was found in the 3rd, 7th and 11th week of foal life and in the 11th week of lactation in mares. 3. In mares the mean diurnal cortisol level changed from 32 ng/ml in the first week to 57 in the 11th week and in foals from 24 in the first week to 16 ng/ml in the 11th week. 4. In the glucose level no diurnal rhythm was observed. 5. In mares the mean diurnal glucose level after parturition was about 58 mg/100 ml and increased to 83-85 mg/100 ml. In foals it changed from 105 to 128 mg/100 ml. 6. In the activity of FDPA no diurnal rhythm was observed. 7. The diurnal changes in the levels of lactic (LA) and pyruvic (PA) acids and in values of pH, pO2 and pCO2 were studied in the blood of barren and later on in pregnant and lactating mares, throughout three subsequent years. 2. Blood samples were taken every 4 hr, for one day, each month, throughout 3 years. 3. The mares were kept and fed in the same conditions, lighting was natural. 4. In barren mares, diurnal rhythm in LA, PA, pO2 and pCO2 was found. 5. The pregnancy as well as lactation masked diurnal rhythms in parameters studied, except the LA level during lactation but then the acrophase was shifted by 3-4 hr. 6. Seasonal cyclicity was found in the values of LA, PA and pCO2 in barren mares. The pregnancy abolished cyclicity in LA level and modified the behaviour of PA and pCO2 values causing a shift of acrophases and lowering the amplitudes of the indices. 7. In the pO2 tensions no seasonal cycles were observed. 8. In the values of pH neither diurnal rhythms nor seasonal cycles throughout study years were observed.  相似文献

In 34 pure-bred Arabian horses, divided into four groups (Gr. I, 10 pregnant mares; Gr. II, seven barren mares; Gr. III, 10 foals born in 1981; Gr. IV, seven foals born in 1982), seasonal changes in haemoglobin level, haematocrit value, sedimentation rate, red blood cell number and diameter, percentage of erythroblasts and reticulocytes, and index F were studied. Seasonal cyclicity was found in all groups in the haemoglobin level, haematocrit value and RBC diameter. It was also found in the sedimentation rate (PCV) and in index F, but not for the youngest foals (Gr. IV). For the RBC number the cyclicity is given in both groups of foals, and the erythroblast and reticulocyte percentage only in the older foals (Gr. III). Out of eight indices studied cyclicity was found in all foals in Gr. III, in five of the mares in Groups I and II, and in four of the youngest foals in Gr. IV. There is no difference in the cyclicity of indices studied between pregnant and barren mares.  相似文献

Seasonal periodicity was found in all groups for the number of lymphocytes, segmented neutrophils and eosinophils and cortisol level. Leukocyte periodicity was found in three groups, but not in the barren mares. In lyzozyme activity there was periodicity in three groups but not in the youngest foals. In the stab neutrophils, basophils and monocytes no cycle was observed. The behaviour of the indices studied showed the influence of age of the horses (mature vs young) and the physiological state of the mares (pregnancy or barrenness).  相似文献

The objective of the present study was to evaluate the beneficial effect of hCG injected immediately after mating in Arabian barren mares treated with uterine lavage and oxytocin. Arabian barren mares (n = 36) with PMIE were subjected to detailed clinical examinations including palpation per rectum, vaginoscopy, and cytological examination. After mating the 36 mares were randomly divided into four groups. The mares in group 1 (n = 10) were immediately after breeding injected with hCG 3000 IU IM. Uterine lavage with 1 L of N-saline containing 4 million IU of crystalline penicillin and 4 g of streptomycin sulphate was performed 4 h after breeding. Then mares received two injections of oxytocin 40 IU IM 2 h apart after 6 h of mating. Mares in group 2 (n = 10) treated with uterine lavage and oxytocin as group 1. While mares in group 3 (n = 10) received uterine lavage only. A control group (n = 6) as group 4 did not received any treatment. The results of clinical examination indicated that 69.4% of PMIE mares were harboring severe endometritis and 30.6% with a moderate form of endometritis. Significant (P < 0.01) increase in lymphocytes were founded in barren mares included in this study. Higher pregnancy rate (P < 0.01) was founded in Arabian barren mares 80% injected with hCG immediately after breeding and uterine lavage and oxytocin. No significant difference was found in mares received uterine lavage and oxytocin and uterine lavage only. In a conclusion, administration of hCG immediately after mating and intrauterine lavage containing antibiotics performed 4 h and two injections of oxytocin 40 IU IM 2 h apart after 6 h of mating had improved fertility of Arabian barren mares.  相似文献

The aim of this study was to evaluate the effect of active immunization against GnRH on ovarian activity, plasma progesterone and estradiol concentrations and on estrous behavior in adult mares. Eighteen cyclic mares were randomly divided into a treatment and control group. Nine mares were immunized twice with 2 mL (400 microg GnRH-protein conjugate) of a GnRH-vaccine (Improvac, CSL Limited, Australia) administered intramuscularly, 4 weeks apart. Control mares received the same amount of saline solution. Ovaries and uterus of all mares were examined weekly by ultrasonography from 3 weeks before to 60 weeks after first immunization. Thereafter, vaccinated mares were evaluated monthly until 100 weeks after first vaccination. In addition, mares were teased with a stallion for assessment of estrous behavior and blood was collected for progesterone, estradiol-17beta and GnRH antibody titer determination. Results demonstrate that vaccination against GnRH significantly (P<0.05) influenced all parameters, except estradiol-17beta concentration. All vaccinated mares ceased reproductive cyclicity (plasma progesterone <1 ng/mL, follicles <3 cm) within 8 weeks after the first injection and ovarian activity remained suppressed for a minimum of 23 weeks. Five mares resumed cyclicity (follicles >3 cm, progesterone >1 ng/mL) while three mares showed only follicular activity (follicles >3 cm) and one mare remained completely suppressed for the entire duration of the study. In spite of ovarian suppression, four mares expressed sporadic and one mare continuous estrous behavior. In conclusion, reproductive cyclicity in adult mares can be successfully suppressed by immunization against GnRH but the timing of resumption of cyclicity is highly variable and estrous behavior may occur in spite of ovarian suppression.  相似文献

In Experiment I, 32 mares were equally allotted to the following treatments: mares fed to A) high body condition from 90 days prepartum to foaling and maintained in high body condition to 90 days postpartum, B) high body condition from 90 days prepartum to foaling and allowed to lose body condition to 90 days postpartum, C) lose body condition from 90 days prepartum to foaling and maintained in low body condition to 90 days postpartum, and D) lose body condition from 90 days prepartum to foaling and allowed to gain weight after foaling to attain a high level of body condition by 90 days postpartum. After three cycles, pregnancy rate at 30 days postovulation was lower (P<0.05) in C mares (50%) than in those in the other three groups (100%). Maintenance of pregnancy to 90 days was also reduced (P<0.05) in C mares (25%) when compared with A, D (both 100%) and B mares (88%). Foal growth to 90 days of age was similar in all treatments. In Experiment II, 927 mares were evaluated for body condition and monitored for reproductive performance. Pregnancy rate was lower (P<0.05) and number of cycles/conception was higher (P<0.05) for barren and maiden mares entering the breeding season in thin condition and for pregnant mares foaling in thin condition (condition score less than 5.0) when compared with mares with a higher level of condition. Also, onset of estrus and ovulation appeared to be delayed in barren and maiden mares entering the breeding season in thin condition. Breeding efficiency was enhanced in mares entering the breeding season or foaling at a condition of 5.0 or above. Initial excess stores of body fat enhanced fertility. There were no detrimental effects of excess body fat stored in late gestation.  相似文献

The effects upon equine endometrial histological features produced by 10 to 30% concentrations of dimethyl sulfoxide (DMSO) in sterile saline were compared with the effects of sterile saline (0.9% NaCl) alone as an intrauterine infusion therapy in 16 barren mares. No harmful histological changes were noted (P > 0.05) as a result of the therapy. Thirty percent intrauterine DMSO therapy produced a significant (P < 0.01) improvement (i.e., reduction of chronic inflammatory cell infiltrates and reduction of periglandular fibrosis) in endometrial biopsy classification in 18 of the 27 barren mares evaluated; whereas only 2 of 18 barren mares improved following intrauterine saline treatment in the control group. In subsequent breeding trials, the pregnancy rates following intrauterine therapy were no different between DMSO-treated mares and saline-treated control mares (P > 0.05); however, there was a trend toward a higher pregnancy rate following DMSO therapy. The inability to control for sire variability in the breeding trials may have contributed toward the result that no significant difference could be demonstrated in the pregnancy rates between treated and control mares.  相似文献

Endometrial cytology and culture specimens (n=2123) were collected concurrently with a guarded uterine culture instrument from 970 mares (738 barren, 1230 foaling and 155 maiden mares) during three breeding seasons (2001-2004). Results were compared to the 28-d pregnancy rate for the cycle from which the samples were taken. Cytological smears were evaluated for inflammation at x100 and graded as: not inflammatory (0-2 neutrophils/field), moderate inflammation (2-5 neutrophils/field), severe inflammation (>5 neutrophils/field), or hypocellular (scant epithelial cells and no neutrophils). Uterine culture swabs were plated within 6h, incubated for 72 h and results determined at 24, 48, and 72 h. Approximately, 20% (n=423) cytology samples were positive for inflammation (>2 neutrophils), whereas approximately 11% (n=231) of cultures had microorganisms recovered. A majority (64%) of the positive cultures (147/231) had inflammation on cytology smears. Streptococcus equi subsp. zooepidemicus was associated with more positive cytology results than coliforms (P<0.01). Mares with positive cytology or culture had lower pregnancy rates than mares with normal findings (P<0.01). Lowest pregnancy rates were recorded for mares with severe endometrial inflammation (21%, versus moderate inflammation 48%). Isolation of a microorganism from mares with endometrial inflammation was not associated with a further reduction in pregnancy rates. In barren, foaling and maiden mares, cytology was positive in 28, 17, and 5%, respectively, and culture was positive in 12.2, 11.1, and 3.2%. Foaling and maiden mares had higher pregnancy rates than barren mares (62, 69, and 44%, respectively, P<0.001). In conclusion, a positive cytology was twice as common as a positive culture, and isolation of microorganisms was associated with reduced pregnancy rates, even in the apparent absence of inflammation.  相似文献

Mean corpuscular haemoglobin changed distinctly from one study day to another. 5. Background

Mares have an annual reproductive rhythm, with a phase of inactivity in midwinter. The aim of this study was to determine the impact of food restriction on physiological and metabolic hallmarks of this rhythm.

Methods

Over three successive years, 3 groups of 10 mares were kept under natural photoperiod. A 'well-fed' group was fed to maintain the mares in good body condition; a 'restricted' group received a diet calculated to keep the mares thin and a 'variable' group was fed during some periods like the 'restricted' group and during some other periods like the 'well-fed' group, with the aim of mimicking the natural seasonal variation of pasture availability, but a few months in advance of this natural rhythm.

Results

Winter ovarian inactivity always occurred and was long in the restricted group. In contrast, in the 'well-fed' group, 40% of mares showed this inactivity, which was shorter than in the other groups. Re-feeding the 'variable' group in autumn and winter did not advance the first ovulation in spring, compared with the 'restricted' group. Measurements of glucose and insulin concentrations in mares from the 'restricted' group during two 24 h periods of blood sampling, revealed no post-prandial peaks. For GH (Growth hormone), IGF-1 and leptin levels, large differences were found between the 'well-fed' group and the other groups. The glucose, insulin, GH and leptin levels but not melatonin level are highly correlated with the duration of ovulatory activity.

Conclusions

The annual rhythm driven by melatonin secretion is only responsible for the timing of the breeding season. The occurrence and length of winter ovarian inactivity is defined by metabolic hormones.  相似文献

Conceptuses were obtained from pony mares on each day of pregnancy between Days 12 and 28, and on Days 39, 45, 65 and 100. Endometrium was obtained from mares at Days 12, 14, 16, 18, 39, 45, 65 and 100 of pregnancy, and from non-pregnant mares during anoestrus, during transition into the breeding season, at oestrus, or during dioestrus. Tissues were incubated in vitro for 24 h with L-[3H]leucine. Proteins synthesized and released into the culture medium were analysed by two-dimensional polyacrylamide gel electrophoresis (2-D PAGE) and fluorography. Conceptuses obtained before Day 14 after ovulation released a characteristic pattern of labelled proteins. These included two groups of apparent isoelectric variants of relative molecular weights (Mr) 30,000-40,000 (pI values 4.5-5.5 and 6-7), one group of Mr approximately 22,000 (pI 6.5-7), and large protein(s) that did not enter the 10% polyacrylamide gel. After Day 14 the array of labelled proteins had changed and resembled that produced by isolated yolk sac at the later stages of pregnancy studied. Included amongst these were several acidic polypeptides with Mr 20,000 (pI 5-6). The endometrial samples released an array of non-dialysable polypeptides into the culture medium. Fluorograms could be assigned to one of three general groups, with endometrium from mares within each group producing similar patterns of labelled proteins. The first group consisted of anoestrous, transitional and ovariectomized mares, and mares at oestrus or Day 1 or Day 18 after ovulation. The epiphyseal neurohormone melatonin (MEL) exhibits circadian cyclicity, as noted extensively in diurnal vertebrates although very little information is available regarding nocturnal species. We have studied the MEL rhythmicity with 24-hour periodicity in a tropical nocturnal bird, Indian spotted owlet Athene brama, which possesses a well-developed pineal organ. We performed our study during two crucial reproductive phases (active and inactive), when the pineal gland activity in owlet exists in inverse states, i.e., inactive and active respectively. Independent of sex, the circadian rhythm of plasma MEL in owlets showed a two-peak cyclicity with a smaller peak at around 1400 h and the higher one at about 0200 h, while the lowest value was found at 1000 h. The night (0200 h) peak activity of plasma MEL in owlet has a resemblance with the earlier findings in diurnal birds and strongly suggests that independent of species habit the peak activity of MEL is invariably dark dependent. However, the daytime peak of MEL may be due to the daytime hiding nature of this nocturnal bird. Interestingly, it was also noted that the hours of peak activity of MEL (1400 and 0200 h) were the same during both of the reproductive phases, though the environmental day length was longer and ambient temperature was higher during the reproductively inactive phase. During daytime these birds hide in a dark burrow where, in general, the intensities of light and temperature are less, and the amplitude of variations of these factors is not prominent. Hence, the seasonal variations in these oscillatory components may not have affected the entrainment of the owlet pineal oscillator, which regulates the daily MEL rhythm in a similar pattern during both the studied phases. On the other hand, a single circadian peak (around 1000 h) circadian cyclicity of gonadal steroids (i.e., testosterone in the male and estradiol and progesterone in the female) showed an inverse relationship with plasma MEL. The aim of this study was tracing of changes in the activity of glutathione peroxidase (GSHPx), glutathione transferase (GSH S-Tr), aspartate aminotransferase (AspAT) and alanine aminotransferase (A1AT) in the brain as a result of diet enrichment with antioxidants: selenium (Se), vitamin E and vitamin B15 (pangamic acid). The experiment was carried out on Wistar rats with initial body weight 150 g. Following prolonged enrichment of diet with Se (0.1 ppm of sodium selenite), vitamin E (6 mg/100 g of diet) and vitamin B15 (2.5 mg/100 g of diet) the following results were obtained. The activity of GSHPx in brain microsomes was not changed after one year of vitamin E administration when it was measured against hydrogen hydroxide and against cumene hydrochloride; vitamin E administration increased the activity of GSH S-Tr in the cytoplasmic fraction of brain cells. Diet enrichment with selenium increased after 12 and 18 months the activity of GSHPx measured against both substrates, and GSH S-Tr activity increased considerably. Presence of vitamin B15 in diet reduced GSHPx activity after one-year or longer administration, after 18 months the activity of GSH S-Tr was reduced also. No changes were noted in the activity of AspAT and A1AT.  相似文献

In stud management, broodmares are commonly fed concentrates in late pregnancy. This practice, however, was shown to correlate with an increased incidence of osteochondrosis in foals, which may be related to insulin sensitivity. We hypothesized that supplementation of the mare with barley in the last trimester of pregnancy alters the pre-weaning foal growth, glucose metabolism and osteoarticular status. Here, pregnant multiparous saddlebred mares were fed forage only (group F, n=13) or both forage and cracked barley (group B, n=12) from the 7th month of pregnancy until term, as calculated to cover nutritional needs of broodmares. Diets were given in two daily meals. All mares and foals returned to pasture after parturition. Post-natal growth, glucose metabolism and osteoarticular status were investigated in pre-weaning foals. B mares maintained an optimal body condition score (>3.5), whereas that of F mares decreased and remained low (<2.5) up to 3 months of lactation, with a significantly lower bodyweight (-7%) than B mares throughout the last 2 months of pregnancy. B mares had increased plasma glucose and insulin after the first meal and after the second meal to a lesser extent, which was not observed in F mares. B mares also had increased insulin secretion during an intravenous glucose tolerance test (IVGTT). Plasma NEFA and leptin were only temporarily affected by diet in mares during pregnancy or in early lactation. Neonatal B foals had increased serum osteocalcin and slightly increased glucose increments and clearance after glucose injection, but these effects had vanished at weaning. Body measurements, plasma IGF-1, T4, T3, NEFA and leptin concentrations, insulin secretion during IVGTT, as well as glucose metabolism rate during euglycemic hyperinsulinemic clamps after weaning, did not differ between groups. Radiographic examination of joints indicated increased osteochondrosis relative risk in B foals, but this was not significant.
