Abstract Three different methods, (i) PEG, (ii) electroporation and (iii) biolistic, were employed to transform Metarhizium anisopliae using benomyl resistance as a selectable marker. Transformation frequencies and mitotic stability varied for each method, from 0.8 to 6.9 transformants μg−1 of DNA and 46%, respectively, by the PEG method; 1.3 to 1.8 transformants μg−1 of DNA and 67% by electroporation; and 32 to 201 transformants μg−1 of DNA and 90% by biolistic. We demonstrate by PCR that 60% of the transformants were generated by gene conversion.  相似文献

Abstract The pre-shock incubation of cells plus DNA and the methylation state of plasmid DNA were found to play a role in the electroporation-based transformation of Clostridium perfringens 3626B. Following pre-shock incubation, the highest number of C. perfringens 3626B transformants was obtained when plasmid pGK201 was both dam+ dcm+ modified, while no transformants were obtained when pGK201 was not methylated or only dcm methylated. This is consistent with the observation that plasmid pGK201 was protected against digestion by C. perfringens 3626B cell-associated nucleases for up to 3 min when methylated by both methylases. C. perfringens 3626B was successfully transformed only within a narrow cell recovery rate window. The erm AM gene associated with pGK201 and pAK102 was found to integrate into the chromosome of C. perfringens strains 13A and 3626B.  相似文献

通过向枯草芽孢杆菌Ki-2-132染色体和/或细胞质导入来自枯草杆菌168菌株的degU32(Hy)和degR基因,以及来自芽孢杆菌解淀粉菌株(Bacillus amyloliquefaciens)的degQa基因,对上述基因对枯草芽孢杆菌Ki-2-132细胞的生长、孢子发生、蛋白酶发酵的影响进行了研究。尽管上述多效调控基因来自不同的芽孢杆菌种和菌株,它们在枯草芽孢杆菌Ki-2-132中依然表现多效性。枯草杆菌Ki-2-132degU32(Hy)表现出增高了的蛋白酶产量;当和质粒或染色体上的degQa基因协作,可以进一步依赖葡萄糖的水平和degQa的基因剂量影响细胞生长,增加蛋白酶产量,以及影响孢子的形成。与此不同,degR在degU32(Hy)突变体中并不显著影响其蛋白酶的产量,这一发现支持DegR蛋白通常稳定磷酸化的DegU,而其在degU32(Hy)菌株中不再进一步放大该突变体内已被磷酸化的DegU的调控作用。  相似文献

Abstract An arg7, cw15, mt+ strain of Chlamydomonas reinhardtii (CC1618) was transformed with pARG7.8, a plasmid containing the wild-type ARG7 gene. Over 2300 arg+ transformants were selected on TAP media. Upon subsequent analysis on TAP plus cadmium plates, five of the transformants failed to grow at a level of 400 μM cadmium and were designated as cadmium sensitive (Cds) mutants. Hybridization data indicated that vector (pBR329) sequences were present in these five mutants, but not in the untransformed parental strain. Two of the mutants have been back crossed to an arg7, cw15, Cd+, mt strain (CC425) and found to have progeny which always cosegregate the arg+ and Cds phenotypesin these two mutants results from the insertion of the plasmid pARG7.8 into a gene involving cadmium detoxification, and it provides a method by which to clone the interrupted gene(s).  相似文献

Two classes of mutations were identified in the degS and degU regulatory genes of Bacillus subtilis, leading either to deficiency of degradative enzyme synthesis (degS or degU mutations) or to a pleiotropic phenotype which includes overproduction of degradative enzymes and the loss of genetic competence (degS(Hy) or degU(Hy) mutations). We have shown previously that the DegS protein kinase and the DegU response regulator form a signal transduction system in B. subtilis. We now demonstrate that the DegS protein kinase also acts as a DegU phosphatase. We present evidence that the DegU response regulator has two active conformations: a phosphorylated form which is necessary for degradative enzyme synthesis and a nonphosphorylated form required for expression of genetic competence. The degU146-encoded response regulator, allowing expression of genetic competence, has been purified and seems to be modified within the putative phosphorylation site (D56----N) since it is no longer phosphorylated by DegS. Both the degU146 mutation as well as the degS220 mutation, which essentially abolishes DegS protein kinase activity, lead to deficiency of degradative enzyme synthesis, indicating the requirement of phosphorylated DegU for the expression of this phenotype. We also purified the degU32(Hy)-encoded protein and showed that this response regulator is phosphorylated by the DegS protein kinase in vitro. In addition, the phosphorylated form of the degU32(Hy)-encoded protein presented a strongly increased stability as compared with the wild type DegU protein, thus leading to hyperproduction of degradative enzymes in vivo.  相似文献

SUMMARY: Deoxyribonucleic acid (final concentration 100 μg/ml) extracted from wild-type cells of Methylococcus capsulatus was used to transform a p -amino benzoic acid (PABA) requiring strain (paba-). Maximum levels of transformants were obtained towards the end of the exponential phase of growth in shake flask culture and the level of the population of transformants was not significantly altered by post-treatment incubation in enriched medium. The maximum transformation frequency was 0.17% of the initial population of PABA requiring cells and the back mutation frequency was 1 : 3.12 × 108.  相似文献

Abstract An arg 7 mutant of the green alga Chlamydomonas reinhardtii was transformed with pARG7.8, a plasmid bearing the wild-type ARG 7 gene. Out of 4100 arg+ transformants selected on an arginine-free medium supplemented with acetate, nine failed to grow on acetate-free medium (ac mutants). The results of the genetic and molecular analysis of several ac mutants are in agreement with the hypothesis that they originated from insertion of the incoming plasmid into the nuclear genome. These mutants should constitute valuable tools for isolating the corresponding wild-type genes after plasmid rescue into Escherichia coli .  相似文献

Abstract An efficient protoplast transformation system and a procedure of plasmid transfer by means of protoplast fusion is described for Streptococcus lactis . Protoplasts of S. lactis IL1403 and S. lactis MG1363 were transformed by pGK12 [2.9 MDa erythromycin resistance (Emr)] with an efficiency of 3 × 105 transformants per μg plasmid DNA. This high efficiency was obtained by the inclusion in the transformation mixture of liposomes composed of cardiolipin and phosphatidyl choline in a molar ratio of 1 to 6 in the presence of 22.5% polyethylene glycol (PEG). This paper also reports an efficient plasmid transfer method between lactic and streptococci and Bacillus subtilis by means of protoplast fusion. When S. lactis and B. lactis protoplasts undergo fusion mediated by exposure to 37.5% polyethylene glycol, plasmid pGKV21 (3.2 MDa; Emr) was transfered from one host to the other with a frequency of 10−3−10−5 transformants per regenerating recipient protoplast.  相似文献

Abstract Plasmid transformation of the nitrogen-fixing bacterium Azospirillum brasilense is described. A modification of the method of Hanahan [1] was used to transform this bacterium with the 20-kb plasmid pRK290. The efficiency of transformation ranged from 200–1000 transformants per μg of plasmid DNA according to DNA concentration. Ca2+, Mn2+ and K+ were essential for competence, while Rb+ and hexamine cobalt(III) chloride did not appear necessary. The length and the temperature of heat-pulse during transformation affected the efficiency of transformation. The response to different numbers of plasmid molecules was linear, in the range of 0.05–1.0 μg of DNA. No transformants were obtained with pRK290 plasmid DNA linearized with Eco RI. The transformability of different strains of Azospirillum has been compared.  相似文献

Aims:  The ability to transform Vibrio spp. is limited by the extracellular nuclease that their cells secrete. The reported transformation efficiency of this organism is 102–105 transformants per microgram DNA. We tried different buffers and conditions, aiming to elevate its transformation efficiency.
Methods and Results:  MgCl2 and sucrose are often included in the washing and/or electroporation buffers to stabilize the cell membrane. However, Mg2+ is required for production and activity of the extracellular nuclease. A simple electroporation buffer lacking Mg2+ was found to increase transformation efficiency dramatically, to levels 50-fold more than the buffers containing Mg2+. To maintain the stability of the cell membranes, Mg2+ was replaced with high concentrations of sucrose, from 272 to 408 mmol l−1. With the new buffers, the transformation efficiency of Vibrio parahaemolyticus was increased to 2·2 × 106 transformants per microgram DNA.
Conclusions:  Mg2+ in the buffer adversely affected transformation of V. parahaemolyticus by electroporation. The cell membranes of vibrio can be stabilized by high concentration of sucrose when Mg2+ is absent.
Significance and Impact of the Study:  A greater transformation efficiency can facilitate the genetic analysis of an organism and its pathogenicity. Buffers lacking Mg2+ can be used for other nuclease-producing organisms.  相似文献

Abstract An optimized polyethylene glycol (PEG) method of transformation was developed for Methanococcus maripaludis using the pKAS102 integration vector. The frequency of transformation with 0.8 μg of plasmid and 3×109 cells was 4.8×10−5 transformants cfu−1, or 1.8×105 transformants μg−1, which was four orders of magnitude greater than with the natural transformation method. A Pst I restriction activity in M. maripaludis was also identified. Methylation of the plasmid with Pst I methylase increased the methanococcal transformation frequency at least four-fold. Also, chromosomal DNA from M. maripaludis was resistant to digestion by the Pst I endonuclease.  相似文献

A vacuolar Na^ /H^ antiporter cDNA gene was successfully isolated fromHordeum brevisubulatum (Trin.) Link using the rapid amplification ofcDNA ends (RACE) method. The gene was named HbNHXI and was found to consist of 1 916 bp encoding a predicted polypeptide of 540 amino acids with a conserved amiloride-binding domain. Phylogenetic tree analysis of the Na^ /H^ antiporters showed that the HbNHXI gene shares 55.3%-74.8% similarity with the vacuolar-type Na^ /H^ antiporters. Transgenic tobaccos that contain the HbNHXI gene, integrated by forward insertion into the tobacco genome, were obtained via Agrobacterium tumerfaciens and characterized for the determination of the concentration of Na^ and K^ ions, as well as proline, in the presence of 300 mmol/L NaCl. The T1 transgenic plants showed more tolerance to salt and drought than did wild-type plants. Our data suggest that overexpression of the HbNHXI gene could improve the tolerance of transgenic tobaccos to salt and drought through the function of the vacuolar Na^ /H^ antiporter.  相似文献

Abstract A genetic transformation system for the aflatoxin-producing fungus Aspergillus parasiticus using two autonomously replicating plasmids from A. nidulans (ARp1 and pDHG25) is reported. Transformation frequencies using the plasmid pDHG25 were from 5 × 102 to 2.5 × 104 transformants per 106 viable protoplasts and μg DNA. The stability of the plasmids in the transformants was also studied. This transformation system offers a new opportunity to clone genes related to aflatoxin production using appropriate aflatoxin-defective mutants.  相似文献

A marked increase in the Na+, K+-ATPase activity of sea urchin embryos occurred following an elevation of its mRNA level, revealed by Northern blotting analysis, in developmental period between the swimming blastula and the late gastrula stage. cDNA clone of Na+, K+-ATPase α-subunit, obtained from γgt10 cDNA library of sea urchin gastrulae, was digested with EcoRl ad Hindlll. The obtained 268 bp cDNA fragment, hybridized to a 4.6 Kb RNA, was used as probe for Northern blotting analysis. The level of Na+, K+-ATPase mRNA was higher in embryo-wall cell fraction isolated from late gastrulae (ectoderm cells) than the level in the bag fraction, containing mesenchyme cells (mesoderm cells) and archenteron (endoderm cells). The activity of Na+, K+-ATPase and the level of its mRNA were higher in animalized embryos obtained by pulse treatment with A23187 for 3 hr, starting at the 8–16 cell stage and were considerably lower in vegetalized embryos induced by 3 hr treatment with Li+ than that in normal embryos at the post gastrula corresponidng stage. Augmentation of Na+, K+-ATPase gene expression can be regarded as a marker for ectoderm cell differentiation at the post gastrula stage, which results from determination of cell fate in prehatching period.  相似文献

Abstract: A rapid and effective method is described for electroporation of Brevibacterium sp. R312, a coryneform strain producing nitrile hydratase and amidase. The transformation efficiency of the method is 108 transformants per μg of plasmid under optimal conditions. Parameters optimised included field strength (11.8 kV cm−1), pulse length (2.4 ms), plasmid DNA concentration (0.25 μg ml−1 and cell density (1010 cells ml−1). Surprisingly, the transformation efficiency did not vary with the growth stage, in contrast to results in the literature. A shuttle vector was constructed containing several unique cloning sites down-stream of the SP6 RNA polymerase promoter.  相似文献

Polymorphism for two autosomal alleles of equine plasminogen, PLG 1 and PLG 2, was demonstrated in plasma by isoelectric focusing and immunofixation, with a goat anti-human plasminogen antibody. The frequency of PLC 2 was 0.16 in 150 Standardbreds, 0.20 in 96 Thoroughbreds, and 0.39 in 32 Shetland ponies. No evidence for linkage of PLG with any of 13 marker loci was found.  相似文献

Abstract: Myelin membrane prepared from mouse sciatic nerve possesses both kinase and substrates to incorporate [32P]PO43− from [γ-32P]ATP into protein constituents. Among these, P0 glycoprotein is the major phosphorylated species. To identify the phosphorylated sites, P0 protein was in vitro phosphorylated, purified, and cleaved by CNBr. Two 32P-phosphopeptides were isolated by HPLC. The exact localization of the sequences around the phosphorylated sites was determined. The comparison with rat P0 sequence revealed, besides a Lys172 to Arg substitution, that in the first peptide, two serine residues (Ser176 and Ser181) were phosphorylated, Ser176 appearing to be modified subsequently to Ser181. In the second peptide, Ser197, Ser199, and Ser204 were phosphorylated. All these serines are clustered in the C-terminal region of P0 protein. This in vitro study served as the basis for the identification of the in vivo phosphorylation sites of the C terminal region of P0. We found that, in vivo, Ser181 and Ser176 are not phosphorylated, whereas Ser197, Ser199, Ser204, Ser208, and Ser214 are modified to various extents. Our results strongly suggest that the phosphorylation of these serine residues alters the secondary structure of this domain. Such a structural perturbation could play an important role in myelin compaction at the dense line level.  相似文献

Protoplasts were isolated from leaves of tomato seedlings ( Lycopersicon esculentum Mill., cv. Marmande) at the 2nd to 4th true leaf stage and were loaded with the calcium binding tetra[acetoxymethyl+] ester of the fluorescent stilbene chromophore, Fura 2. Although the loading efficiency of the dye in these protoplasts was low, many protoplasts loaded only in the cytosol were always obtained. Changes in the cytosolic calcium concentration ([Ca2+]cyt) were determined in single protoplasts in a temperature-controlled perfusion chamber by use of fluorescence photometry microscopy after excitation at 340 and 380 nm. When the protoplasts were subjected to chilling temperatures (10–15°C) by a circulating solution, the [Ca2+]cyt increased in 64% of the analysed protoplasts. Depending on the initial resting level of [Ca2+]cyt, three main types of kinetics were obtained in these protoplasts: (1) In 21% of the protoplasts, [Ca2+]cyt increased to a maximum within 10–20 s from the start of temperature decrease, followed by a fast decrease; (2) in 11% of the protoplasts, the [Ca2+]cyt both increased and decreased somewhat slower; and (3) in 32% a constant increase of [Ca2+]cyt was obtained 1 min after the start of temperature decrease.  相似文献
