Abstract The Bacillus subtilis spoIIA and spoIIAB genes were fused to the Escherichia coli lacZ gene on a novel integrational plasmid vector. The constructs were integrated into the B. subtilis chromosome, and used to show that the spoIIA locus was expressed at two times during sporulation.  相似文献

Abstract We isolated pepT from Bacillus subtilis , a gene with homology to various tripeptidases from different bacterial sources, pepT is preceded by genes encoding a two component regulatory system. Its expression is activated during stationary phase. In minimal medium this activation is boosted in the presence of phosphate. The response regulator is preceded by putative promoter consensus sequences recognized by the stationary phase specific sigma factors σ H, σ F, and σ G. This is in accordance with the initiation of expression at the beginning of stationary phase. Inactivation of pepT causes no obvious phenotype.  相似文献

A pleiotropic mutation (cpm) which is localized in the vicinity of the spoOA gene is described in Bacillus subtilis. The mutation inhibits spore formation, rendering bacteria auxotrophic for adenine and tyrosine, enhances sensitivity to antibiotics, decreases cell motility, inhibits the ability to grow on pentoses and to maintain bacteriophage multiplication, induces severalfold the activities of alkaline proteinase and alpha-amylase. At the same time, the cpm mutant starts to excrete inosine into the growth medium. This excretion most probably is explained by a 50-fold increase in the activity of inosine monophosphate: 5'-nucleotidase and a 10-fold decrease in the activity of purine nucleoside phosphorylase. The inosine production and Ade- phenotype of the cpm mutant is not accompanied by the change in the activity of succinyl adenosine monophosphate synthetase. The nature of the mutation is discussed.  相似文献

Abstract During sporulation of Bacillus subtilis strain 168 initiated by step-down conditions, resistance to chlorhexidine diacetate (CHA) developed at about t 3.5, before heat but after toluene resistance. Mutants blocked at stage IV of sporulation were sensitive to all three treatments. Stage V mutants were toluene resistant but moderately sensitive to heat and CHA. A stage VI mutant was resistant to all three treatments. Thus, chlorhexidine resistance is likely to be a result of spore coat, rather than of cortex, development.  相似文献

克隆Bacillus subtilis NX-2中的聚谷氨酸合成酶基因pgsBCA并进行测序。应用生物信息学分析方法和工具对PgsB、PgsC、PgsA蛋白质的理化性质、跨膜区域、信号肽、细胞定位等进行分析和预测,并探讨它们的作用方式。结果表明:PgsB蛋白不含有跨膜区,它与ATP结合并催化ATP的水解,为PGA合成提供能量;PgsC蛋白保守性最高,其含有4个跨膜区域,是疏水性膜结合蛋白;PgsA为亲水性稳定蛋白,在N端存在1个跨膜区域。  相似文献

Two Bacillus subtilis strains L10 and G1, previously isolated from fermented pickles, were tested for antibacterial activity against Vibrio harveyi using a well-diffusion agar assay. Antibiotic biosynthesis genes were then detected in both bacterial strains and included the following genes: bacA, bacB and bacD genes for bacilysin production, ppsE/fenB gene for plipastatin/fengycin production, albF and albA genes for subtilosin production, and srfAB and sfP genes for surfactin production. Based on these results, two B. subtilis strains could be considered as potential biological control agents in aquaculture.  相似文献

Abstract A Bacillus subtilis response regulator, DegU9, carrying an amino acid alteration caused by the degU9 (Hy) mutation was partially purified, and phosphorylation and dephosphorylation of the protein was studied. The extent of phosphorylation was not as high as the level attained with wild-type DegU, but the DegU9-phosphate once formed was more stable than the wild-type DegU-phosphate. An in vivo study with a degU9 mutant showed that degS was necessary for the overproduction of exoproteases. These results suggest that phosphorylation is necessary for the mutant DegU9 to exert its effect and that the higher stability of phosphorylated DegU9 is responsible for the overproulation phenotype.  相似文献

Dephosphorylation of SpoIIAA-P by SpoIIE is strictly dependent on the presence of the bivalent metal ions Mn2+ or Mg2+. Replacement by Ala of one of the four Asp residues, invariant in all representatives of protein phosphatase 2C, completely abolished the SpoIIE phosphatase activity in vitro, whilst replacement of the Asp residues by another acidic amino acid, Glu, had varying effects on the activities of the resulting mutated proteins. D610E and D795E exhibited some residual activity while D628E and D745E were without enzymatic activity. The results suggest that the functional model in which metal-associated water molecules are involved in the dephosphorylation reaction catalyzed by human protein phosphatase 2C alpha can also be applied to the bacterial protein phosphatase 2C-like protein.  相似文献

Abstract The transformation system currently used for Bacillus subtilis protoplasts has been improved. Special emphasis was made on three parameters of practical importance:
(a) conditions for direct selection of transformants, (b) optimization of the transformation system for Rec mutants, and (c) conservation of protoplast suspensions for further use.
Selective regeneration was efficiently achieved for kanamycin or neomycin. Chloramphenicol, tetracycline and erythromycin were only expressed when low concentrations of the antibiotics were used to select transformants during regeneration.  相似文献

微生态制剂是饲用抗生素的绿色有效替代品。枯草芽孢杆菌在逆境中可形成抗逆性强的芽孢,在生产和应用过程中保持高活性,是一种高效的微生态制剂菌种。提高枯草芽孢杆菌活菌数及芽孢率是保证微生态制剂产品质量的关键。本文综述了枯草芽孢杆菌芽孢形成的分子生物学机制及影响芽孢形成的重要因素,进一步比较枯草芽孢杆菌微生态制剂不同发酵方式的特点,重点阐述了提高枯草芽孢杆菌有效生物量的工艺优化,最后介绍了枯草芽孢杆菌微生态制剂的应用,并对将来研究思路进行了讨论。  相似文献

通过向枯草芽孢杆菌Ki-2-132染色体和/或细胞质导入来自枯草杆菌168菌株的degU32(Hy)和degR基因,以及来自芽孢杆菌解淀粉菌株(Bacillus amyloliquefaciens)的degQa基因,对上述基因对枯草芽孢杆菌Ki-2-132细胞的生长、孢子发生、蛋白酶发酵的影响进行了研究。尽管上述多效调控基因来自不同的芽孢杆菌种和菌株,它们在枯草芽孢杆菌Ki-2-132中依然表现多效性。枯草杆菌Ki-2-132degU32(Hy)表现出增高了的蛋白酶产量;当和质粒或染色体上的degQa基因协作,可以进一步依赖葡萄糖的水平和degQa的基因剂量影响细胞生长,增加蛋白酶产量,以及影响孢子的形成。与此不同,degR在degU32(Hy)突变体中并不显著影响其蛋白酶的产量,这一发现支持DegR蛋白通常稳定磷酸化的DegU,而其在degU32(Hy)菌株中不再进一步放大该突变体内已被磷酸化的DegU的调控作用。  相似文献

A transformation system with plasmids was developed for Bacillus subtilis NB22, an antibiotic iturin producing strain. Treatment of B. subtilis NB22 with 4 M KCl was effective for the induction of competence, followed by uptake of plasmid DNA in the presence of polyethylene glycol. The efficiency of transformation of this bacterium with pC194 and pUB110 was 4.1 X 10(3) and 1.5 X 10(3) transformants per micrograms DNA, respectively and the transformation frequency was 3.3 X 10(-3) and 7.2 X 10(-4), transformants per viable cell, respectively. This method was much faster and three orders of magnitude more efficient in transformation efficiency than protoplast transformation methods.  相似文献

Abstract Using promoter-probe plasmids, more than 200 promoter-containing fragments from Bacillus stearothermophilus and Bacillus subtilis were cloned in B. subtilis . Among these, 15 promoter fragments were highly temperature-dependent in activity compared to the promoter sequence (TTGAAA for the −35 region, TATAAT for the −10 region) of the amylase gene, amyT , from B. stearothermophilus . Some fragments exhibited higher promoter activities at elevated temperature (48°C), others showed higher activities at lower temperature (30°C). Active promoter fragments at higher and lower temperatures were obtained mainly from the thermophile ( B. stearothermophilus ) and the mesophile ( B. subtilis ), respectively. A promoter fragment active at high temperature was sequenced, and the feature of the putative promoter region was discussed.  相似文献

枯草芽孢杆菌是芽孢杆菌中比较具应用潜力的菌种之一。近年来国内外对于芽孢杆菌各方面应用的研究日益增多,枯草芽孢杆菌作为一种生防细菌越来越引起人们的关注。主要综述了枯草芽孢杆菌在抑制植物病原菌生物防治领域的研究进展,阐述了枯草芽孢杆菌的控病作用机制,包括竞争作用、拮抗作用、溶茵作用、诱导植物产生抗性及促进植物生长5个方面。简要介绍了枯草芽孢杆菌及其制剂在国内外的应用情况及在植物病害防治应用中存在的问题、解决措施及发展前景。  相似文献

We have established the co-linear regions of Bacillus licheniformis, an industrially important bacterium, and Bacillus subtilis, a model bacterium. In the co-linear regions, revealed by PCR, gene content and order are presumed to be conserved. These regions constitute approximately 60% of the compared chromosomes. Sequencing of the competence genes of B. licheniformis allowed us to validate the approach, and to demonstrate how it can be used for the comparative analysis of complex genetic systems. A new insertion sequence, designated IS3Bli1, was discovered in the competence region of the analyzed B. licheniformis strain.  相似文献
