The elongation growth of the Avena first internode segments was studied in the presence of one or several of the following growth substances: indoleacetic acid (IAA), 6-fur-furylamino purine (FAP, kinetin), 6-benzylamino purine (BAP), gibberellin A3 (GA3) and A4+7 (GA4+7), and abscisic acid (ABA). The cytokinins at concentrations of 10?7 to 10?6M stimulated growth with 4 to 6 per cent but this effect was not statistically significant. Concentrations higher than 5 × 10?6M inhibited growth. FAP and BAP (from 10?8M to 10?6M) had no significant interaction with any other growth substance used. The two-factor interactions of IAA × ABA, IAA × GA3, and GA3× ABA, as well as the three-factor interaction IAA × ABA × GA3 were significant. However, the IAA × ABA interaction was significant only when high concentration (10?6M) of ABA was used. The growth inhibition produced by 10?7 and 10?6M ABA was overcome by about equimolar concentrations of IAA. The stimulation of growth by GA3 and GA4+7 (10?9 to 10?7M) was prevented by simultaneous application of ABA, and it was reduced significantly by application of IAA (10?7 to 10?8M). GA3 at 10?8M combined with different concentrations of IAA gave slightly higher elongation than IAA alone but the observed values were significantly lower than expected assuming independent additive action.  相似文献

The amount of γ-amino butyric acid as measured by thin-layer chromatography, and the specific activity of glutamic decarboxylase as measured both by Warburgapparatus (CO2) and by thin-layer chromatography (γ-ABA) were determined in leaves of different ages. Datura suaveolens, Medicago sativa and Salvinia natans were used as experimental plants. The amount of γ-ABA increases in the leaves of Salvinia natans, Medicago sativa and Datura suaveolens in proportion to the age of the leaves. In the case of Salvinia natans, the amount of γ-ABA in old leaves is about twice as much as in young ones: in the case of Medicago sativa and Datura suaveolens the increase is slightly less. In Salvinia natans the amount of γ-ABA reaches its greatest value in the third to fifth leaf, in Medicago sativa and Datura suaveolens later. This increase in the amount of γ-ABA with respect to the age variation of the leaves, is reflected in the growth of leaves. In Salvinia natans the third or fourth leaf is full-grown, in Medicago sativa and Datura suaveolens this stage is reached in the seventh or eighth leaf. The activity of glutamic decarboxylase parallels the γ-ABA accumulation in that it increases when the leaves grow. The increase of glutnmic decarboxylase activity takes place more rapidly in the aging leaves of Medicago sativa than in those of Datura suaveolens. The activities observed are somewhat higher when the amounts of γ-ABA are analysed than when the CO2 evolved is measured. KM-values for glutamic decarboxylase were calculated using various substrate concentrations to test the enzyme activity. The KM-values obtained were 7.0 · 10?3M in the case of Medicago sativa, and 7.9 · 10?3M in the case of Datura suaveolens.  相似文献

γ-Aminobutyrate synthesis, the first step in this pathway, is catalysed by L-glutamate-1-carboxy-lyase (E.G. 4.1.1.15). The purification procedure and some of in vitro properties of this enzyme isolated from fruit-bodies of Agaricus bisporus 19 Lge were investigated. Glutamate decarboxylase has been partially purified from a homogenate by a combination of ammonium sulfate fractionation and hydroxylapatite column chromatography. All kinetic studies were carried out manometrically in a nitrogen atmosphere at 35°C by conventional Warburg technique. The decarboxylase is a pyridoxal-phosphate requiring enzyme. The pH optimum was found to be between 5.5 and 5.6 and the Km value for glutamate was calculated to be 4 × 10-2M from a Lineweaver-Burk plot. Of the amino acids tested the enzyme is specific for glutamate. γ-Aminobutyrate is not carboxylated to form glutamate. Inhibition by malate, malonate, α-ketoglutarate and NAD were found to be non-competitive with respect to glutamate, and those by succinate to be uncompetitive. Fractionation of the subcellular components shows that the enzyme is localized in the hyaloplasm. The results are discussed in relation to the γ-aminobutyrate bypath, a probable shunt to the Krebs cycle.  相似文献

The growth of wheat seedlings (Triticum sativum) is inhibited by abscisic acid (ABA). The inhibition increases with the concentration of ABA (from 10-6M to 5 × 10-5M) and is stronger in the case of coleoptiles and first leaves than in roots. In contrast, naphthaleneacetic acid (ANA), at 10-5M, exerts its greatest inhibitory effect on the roots. The inhibitory effect of ABA on coleoptiles can be partially overcome by kinetin and to a much smaller degree by gibberellic acid. Neither of these two compounds, at 10-5M, had any effect on the ABA-induced inhibition of root growth. The RNA and DNA contents per plant organ are considerably reduced after treatment of the seedlings with ABA, particularly in the coleoptiles and the first leaves. The incorporation of uracil-2-14C and uridine T (G) into RNA of treated seedlings is reduced in the case of coleoptiles and first leaves, but considerably enhanced in roots. The mechanism of the action of ABA is discussed in the light of these results.  相似文献

Lens roots form chlorophylls in well-developed chloroplasts when they are cultivated in continuous light. Chlorophyll accumulation was increased when kinetin (10?4M) was added for a short time (24 h) of treatment. For a long time (72 h), the increase of chlorophyll content was reduced when roots were cultivated in light and with kinetin. When the roots were transferred from light to dark, chlorophyll content was decreased. This reduction was inhibited when roots were treated with kintin. From these present result it was supposed that kinetin acts on the catabolism and the anabolism of chlorophyll. The necessity of kinetin for chloroplast from amyloplast maturation and differentiation was discussed.  相似文献

Metabolism of γ-Aminobutyrate in Agaricus bisporus. III. The Succinate-Semialdehyde: NAD (P)+ Oxidoreductase. The succinate-semialdehyde:NAD(P)+ oxidoreductase (E.C. 1.2.1.16) is responsible for the second step in the catabolism of γ-aminobutyrate: the irreversible enzymatic conversion of succinic semialdehyde (SSA) to succinate. Succinate semialdehyde dehydrogenase was extracted from mitochondrial fraction of fruit-bodies of Agaricus bisporus Lge. The mitochondrial pellet was sonicated and centrifuged at 110,000 g; the supernatant obtained was designated the “crude extract”. The enzyme was extremely unstable on storage, unless 1 mM EDTA and 20% glycerol were added. Kinetic studies were carried out at 30°C, and the formation of NADH or NADPH was followed by measuring increase of absorbance at 340 nm with a spectrophotometer. The dehydrogenase was completely inactive when the reaction was run in the absence of thiol and was more active with NAD+ than with NADP+. In the “crude extract” the activity with NADP+ had a pH optimum between 8.6 and 9.1 and the Km values for SSA and NADP+ were 2.0 × 10?4M and 1.4 × 10?4M respectively. The pH optimum with NAD+ was found between 8.6 and 8.8 and the Km value for SSA is 4.8 × 10?4M and for NAD+ 2.0 × 10?3M. With NAD+, the kinetic values (pH, Km) of the “crude extract” chromatographed on hydroxylapatite were unchanged. Inhibition by thiamine pyrophosphate (TPP) was uncompetitive with respect to NAD+, those by malate, ATP, ADP and NADPH non-competitive and that by NADH competitive. These results and the fact that activity with NAD+ was lost more slowly than with NADP+ indicate the possibility of at least two mitochondrial succinate-semialdehyde dehydrogenases, even though the activities of this enzyme assayed with NAD+ and NADP+ respectively were not able to be separated from each other by hydroxylapatite column chromatography. Some speculations on the metabolic regulation of this dehydrogenase and considerations on the significance of these results in the physiology of respiration in Agaricus bisporus Lge are given.  相似文献

Abstract: A γ-aminobutyric acidA (GABAA) receptor (GABAAR) γ2 subunit (short form) was cloned from an adult human cerebral cortex cDNA library in bacteriophage λgt11. The 261-bp intracellular loop (IL) located between M3 and M4 was amplified using the polymerase chain reaction and inserted into the expression vectors λgt11 and pGEX-3X. Both γ-galactosidase (LacZ) and glutathione-S-transferase (GST) fusion proteins containing the γ2IL were purified, and a rabbit antibody to the LacZ–γ2IL was made. The antibody reacted with the γ2IL of both LacZ and GST fusion proteins and immunoprecipitated the GABAAR/ benzodiazepine receptor (GABAAR/BZDR) from bovine and rat brain. The antibody reacted in affinity-purified GABAAR/BZDR immunoblots with a wide peptide band of 44,000–49,000 Mr. Immunoprecipitation studies with the anti-γ2IL antibody suggest that in the cerebral cortex, 87% of the GABAARs with high affinity for benzodiazepines and 70% of the GABAARs with high affinity for muscimol contain at least a γ subunit, probably a γ2. These results indicate that there are [3H]muscimol binding GABAARs that do not bind [3H]flunitrazepam with high affinity. Immunoprecipitations with this and other anti-GABAAR/BZDR antibodies indicate that the most abundant combination of GABAAR subunits in the cerebral cortex involves α1, γ2 (or other γ), and β2 and/or β3 subunits. These subunits coexist in >60% of the GABAAR/BZDRs in the cerebral cortex. The results also show that a considerable proportion (20–25%) of the cerebellar GABAAR/BZDRs is clonazepam insensitive. At least 74% of these cerebellar receptors, which likely contain α6, also contain γ2 (or other γ) subunit(s). The α1 and β2 or β3 subunits are also frequently associated with γ2 (or other γ) and α6 in these cerebellar receptors.  相似文献

Abstract: To see the effect of a γ-aminobutyric acid GABA uptake inhibitor on the efflux and content of endogenous and labeled GABA, rat cortical slices were first labeled with [3H]GABA and then superfused in the absence or presence of 1 mM nipecotic acid. Endogenous GABA released or remaining in the slices was measured with high performance liquid chromatography, which was also used to separate [3H]GABA from its metabolites. In the presence of 3 mM K+, nipecotic acid released both endogenous and [3H]GABA, with a specific activity four to five times as high as that present in the slices. The release of labeled metabolite(s) of [3H]GABA was also increased by nipecotic acid. The release of endogenous GABA evoked by 50 mM K+ was enhanced fourfold by nipecotic acid but that of [3H]GABA was only doubled when expressed as fractional release. In a medium containing no Ca2+ and 10 mM Mg2+, the release evoked by 50 mMK+ was nearly suppressed in either the absence or the presence of nipecotic acid. In the absence of nipecotic acid electrical stimulation (bursts of 64 Hz) was ineffective in evoking release of either endogenous or [3H]GABA, but in the presence of nipecotic acid it increased the efflux of endogenous GABA threefold, while having much less effect on that of [3H]GABA. Tetrodotoxin (TTX) abolished the effect of electrical stimulation. Both high K+ and electrical stimulation increased the amount of endogenous GABA remaining in the slices, and this increase was reduced by omission of Ca2+ or by TTX. The results suggest that uptake of GABA released through depolarization is of major importance in removing GABA from extracellular spaces, but the enhancement of spontaneous release by nipecotic acid may involve intracellular heteroexchange. Depolarization in the presence of Ca2+ leads to an increased synthesis of GABA, in excess of its release, but the role of this excess GABA remains to be established.  相似文献

Abstract: In this study, we have investigated the effect of mivazerol, [3-(1H-imidazol-4-yl)methyl-1]-2-hydroxy-benzamide hydrochloride, a new α2-agonist lacking hypotensive properties and a potential anti-ischemic drug, on the evoked release of norepinephrine, aspartate, and glutamate in tissue preparations from hippocampus, spinal cord T1–T5 section, rostrolateral ventricular medulla, and nucleus tractus solitarii of the brainstem of rat. A simple and efficient in vitro procedure to study pharmacologically the release of norepinephrine and glutamate is described. Tissues were chopped into (0.3 × 0.2 × 0.2 mm3) sections and the resulting minces were used for this study. Exposure to KCl (10–75 mM) for 5 min served as a stimulus for the release response. One, S (for aspartate and for glutamate release), or two such stimuli, S1 and S2 (for norepinephrine release) were conducted. The release of norepinephrine (+150% above baseline) was inhibited in a dose-dependent manner by mivazerol in hippocampus (IC50 = 1.5 × 10?8M), spinal cord (IC50 = 5 × 10?8M), rostrolateral ventricular medulla (IC50 = 10?7M), and nucleus tractus solitarii (IC50 = 7.5 × 10?8M), and by clonidine in hippocampus (IC50 = 5 × 10?8M), spinal cord (IC50 = 4.5 × 10?8M), rostrolateral ventricular medulla (IC50 = 2.5 × 10?7M), and nucleus tractus solitarii (IC50 = 10?7M). This effect was counteracted by the selective α2-antagonists yohimbine and rauwolscine. A significant glutamate and aspartate release response was also induced by KCl (35 mmol/L) in hippocampus (+250 and +135%, respectively) and spinal cord (+120 and +55%, respectively), in vitro. However, neither mivazerol nor clonidine, at doses up to 10 µM, had any significant effect on KCl-induced glutamate release in spinal cord, whereas mivazerol blocked completely the release of both amino acids in hippocampus and only the release of aspartate in spinal cord. On the other hand, clonidine (1 µM) was only effective in reducing by 40% the release of aspartate in hippocampus. These data indicate that (1) inhibition of KCl-induced norepinephrine release by mivazerol is mediated by its action on α2-adrenergic receptors; (2) at concentrations selective for α2-adrenergic receptors, only mivazerol was effective in blocking the KCl-induced glutamate release in hippocampal tissue; and (3) at the same concentrations, both mivazerol and clonidine were unable to inhibit glutamate release in the spinal cord. These data suggest that prevention of hyperadrenergic activity by mivazerol in perioperative patients may be mediated through its effect on the release of norepinephrine and/or the release of glutamate and aspartate in regions of the CNS that are involved in the control of cardiovascular homeostasis.  相似文献

Abstract: Morphine-induced release of adenosine from the spinal cord is believed to contribute to spinal antinociception. Although this release is Ca2+ dependent, little is known of the nature of this dependence. In this study, the effects of the dihydropyridine L-type Ca2+ channel agonist Bay K 8644 and the antagonist nifedipine, the N-type Ca2+ channel antagonist ω-conotoxin, and ruthenium red, a blocker of Ca2+ influx induced by capsaicin, on release of adenosine evoked by morphine were determined. The effect of partial depolarization with a minimally effective concentration of K+ on morphine-evoked release of adenosine also was examined. Morphine 10?5-10?4M produced a dose-dependent enhancement of adenosine release from dorsal spinal cord synaptosomes. Following the addition of 6 mM K+ (total K+ concentration of 10.7 mM), 10?6M morphine also enhanced release, and an additional component of action at 10?8M was revealed. Release was Ca2+-dependent as it was not observed in the absence of Ca2+ and presence of EGTA. Bay K 8644 (10 nM) and nifedipine (100 nM) had no effect on the release of adenosine evoked by morphine, but ω-conotoxin (100 nM) markedly reduced such release in both the absence and the presence of the additional 6 mM K+. Morphine-evoked adenosine release was not altered in the presence of a partially effective dose of capsaicin, nor by ruthenium red. These results indicate that morphine can stimulate two distinct phases of adenosine release from the spinal cord (nanomolar and micromolar), and that both phases of release are due to Ca2+ entry via ω-conotoxin-sensitive N-type Ca2+ channels.  相似文献

Abstract: Mesencephalic cell cultures were used as a model to investigate the effects of interleukin-2 (IL-2) on evoked release of [3H]dopamine ([3H]DA) and γ-[3H]-aminobutyric acid ([3H]GABA). At low concentrations (10?13-10?12M), IL-2 potentiated [3H]DA release evoked by the excitatory amino acids N-methyl-D-aspartate (NMDA) and kainate, whereas higher IL-2 concentrations (10?9-10?8M) had no effect. IL-2 (10?14-10?8M) modulated K+-evoked [3H]DA release in a biphasic manner, with low concentrations (10?12-10?11M) of IL-2 potentiating and higher concentrations (10?9-10?8M) inhibiting K+-induced [3H]DA release. IL-2 (10?14-10?8M) by itself failed to alter spontaneous [3H]DA release. The inhibition by IL-2 of K+-evoked [3H]DA release was reversible and not due to neurotoxicity, as preexposure to IL-2 (10?8M) had no significant effect on the subsequent ability of dopaminergic cells to take up and to release [3H]DA. Under our experimental conditions, IL-2 (10?8 M) did not alter Ca2+-independent [3H]GABA release evoked by either K+ or NMDA. The results of this study indicate that IL-2 is able to potentiate [3H]DA release evoked by a number of different stimuli, including K+ depolarization and activation of both NMDA and non-NMDA receptor subtypes in mesencephalic cell cultures. IL-2 is active at very low concentrations, a finding that indicates a potent effect of IL-2 on dopaminergic neurons and implicates a physiological role for this cytokine in the modulation of DA release.  相似文献

The kinetics of t-[3H]butylbicycloorthobenzoate (TBOB) binding to the convulsant sites of the γ-aminobutyric acidA (GABAA) receptor-ionophore complex were examined in synaptosomal membrane preparations of rat brain. On and off rates of TBOB binding were accelerated by 1 μM GABA and decelerated by 1 μM bicuculline methochloride, a GABAA antagonist. The presence of GABA and bicuculline methochloride created rapid and slow phases of dissociation, respectively. The three groups of rate constants distinguished for the dissociation of 4 nM and 30 nM [3H]TBOB represent multiaffinity states of the convulsant sites depending on the presence of GABA or bicuculline methochloride. Apparent association rate constants do not obey the equation kapp=koff±kon [TBOB] without assuming interconvertibility of the kinetic states during binding. Avermectin B1a (AVM B1a), a chloride channel opening agent, accelerated the association and dissociation of TBOB and resulted in a biphasic effect on TBOB binding, i.e., enhancement at low concentrations (EC50, 7.8 nM) followed by displacement at high concentrations (IC50 6.3 μM) of AVM B1a. AVM B1a resulted in similar biphasic effects on t- [35S]butylbicyclophosphorothionate binding. DIDS, an isothiocyanatostilbene derivative with irreversible anion channel blocking effect, selectively inhibited basal [3H]TBOB binding (IC50 125 μM DIDS) leaving the enhancement by AVM B1a unaffected.  相似文献

The insecticide DDT (1,1,1 trichloro-2,2-bis-(p-chlorophenyl) ethane) was found to affect the amount of 32P from [γ-32P] ATP incorporated into proteins derived from lobster peripheral nerves in a variety of ways depending upon the relative concentrations of ATP, Na+, K+, Mg2+, and Ca2+. When a high concentration of ATP (2.5 ± 10?5m ) was used DDT (10?5m ) inhibited the incorporation and reversed the increased incorporation caused by ouabain. At low concentration of ATP (8.6 ± 10 ?8m ), DDT inhibited the incorporation when the buffer contained Mg2+ and Na+ or K+ alone, but when the buffer contained Mg2+ and both Na+ and K+ together, DDT consistently caused an increased amount of 32P to be incorporated. The ability of DDT to cause increases in incorporation of 32P into the lobster nerve proteins was found to parallel many of the properties that this compound shows when allowed to poison crustacean nerves. For example: the effect was negatively temperature dependent, Ca2+ lowered the incorporation in the presence of DDT, and DDT consistently caused greater amounts of 32P to be incorporated than its less potent analog DDE (1,1-dichloro-2,2-bis-(p-chlorophenyl)ethylene). The proteins that were affected by DDT were microsomal in nature and could be centrifuged from supernatant by recentrifuging at 149,000 g. The possibility that this system may be the actual target through which DDT causes its characteristic interferences of the ionic conductance changes associated with the action potential are discussed.  相似文献

Abstract: The effects of γ-aminobutyric acid (GABA) on the spontaneous release of endogenous glutamic acid (Glu) or aspartic acid (Asp) and the effects of Glu on the release of endogenous GABA or [3H]GABA were studied in superfused rat cerebral cortex synaptosomes. GABA increased the outflow of Glu (EC5017.2 μM) and Asp (EC50 18.4 μM). GABA was not antagonized by bicuculline or picrotoxin. Neither muscimol nor (-)-baclofen mimicked GABA. The effects of GABA were prevented by GABA uptake inhibitors and were Na+ dependent. Glu enhanced the release of [3H]GABA (EC50 11.5 μM) from cortical synaptosomes. Glu was not mimicked by the glutamate receptor agonists N-methyl-d -aspartic, kainic, or quisqualic acid. The Glu effect was decreased by the Glu uptake inhibitor D-threo-hydroxyaspartic acid (THA) and it was Na+ sensitive. Similarly to Glu, D-Asp increased [3H]GABA release (EC50 9.9 μM), an effect blocked by THA. Glu also increased the release of endogenous GABA from cortex synaptosomes. In this case the effect was in part blocked by the (RS)-α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor antagonist 6-cyano-7-nitroquinoxaiine-2, 3-dione, whereas the 6-cyano-7-nitroquinoxaline- 2, 3-dione-insensitive portion of the effect was prevented by THA. GABA increased the [3H]D-Asp outflow (EC50 13.7 μM) from hippocampal synaptosomes in a muscimol-, (-)- baclofen-, bicuculline-, and picrotoxin-insensitive manner. The GABA effect was abolished by blocking GABA uptake and was Na+ dependent. Glu increased the release of [3H]- GABA from hippocampal synaptosomes (EC50 7.1 μM) in an N-methyl-d -aspartic acid-, kainic acid-, or quisqualic acid-insensitive way. The effect of Glu was prevented by THA and was Na+ dependent. As in the cortex, the effect of Glu was mimicked by D-Asp in a THA-sensitive manner. It is proposed that high-affinity GABA or Glu heterocarriers are sited respectively on glutamatergic or GA- BAergic nerve terminals in rat cerebral cortex and hippocampus. The uptake of GABA may modulate Glu and Asp release, whereas the uptake of Glu may modulate the release of GABA. The existence of these heterocarriers is in keeping with the reported colocalization of GABA and Glu in some cortical and hippocampal neurons. Preliminary data suggest that these mechanisms may also be present in rat cerebellum and spinal cord.  相似文献

Abstract: Three novel antisera to the γ2 subunit of the γ-aminobutyric acidA (GABAA) receptor/benzodiazepine receptor (GABAAR/BZDR) complex have been made. Anti-γ2S and anti-γ2L are specific antibodies to synthetic peptides that recognize the γ2S (short) and γ2L (long) forms, respectively, of the γ2 subunit. An antibody (anti-γ2IL2) to staphylococcal protein A fusion protein of the large intracellular loop (γ2IL) located between the putative transmembrane segments M3 and M4 of γ2S recognizes both γ2S and γ2L subunits. The antibodies immunoprecipitated both the solubilized and affinity-purified GABAAR/BZDR from rat and bovine brain. Immunoblots with membranes from rat brain cerebral cortex as well as with affinity-purified receptor from bovine cortex show that anti-γ2S and anti-γ2L recognize peptides of 45,000 and 47,000 Mr, respectively. Immunoprecipitation experiments indicate that γ2S is more prevalent in hippocampus, whereas γ2L is more abundant in cerebellum. Intermediate values for each form are found in the cerebral cortex. The results suggest that in the rat brain there is a considerable amount of colocalization of γ2S and γ2L in the same receptor complex. In the cerebral cortex, 15% of the BZDRs contain both γ2S and γ2L subunits and 41–48% of the γ2L subunit coexists with γ2S in the same receptor complex. In cerebellum, in 27% of the clonazepam-sensitive and 39% of the clonazepam-insensitive BZDRs the γ2S and γ2L coexist in the same receptor complex. The latter are presumably localized in granule cells and also contain α6. In addition, almost all (93%) the clonazepam-insensitive BZDRs that contain γ2L also contain a γ2S subunit in the same receptor complex. The most likely interpretation of the results is that there is an important population of granule cell receptors that contain α6, γ2S, and γ2L coexisting in the same receptor complex. Nevertheless, 31% of the cerebellar receptors that contain α6 subunit(s) have neither γ2S nor γ2L subunits. There are also species differences with respect to the relative abundance of γ2S and γ2L. These results might be relevant for understanding the molecular mechanisms underlying some of the GABAAR/BZDR-mediated effects of ethanol intoxication involving cerebellar granule cells.  相似文献

—The inhibition of γ-glutamylcysteine synthetase and its influence on the concentration of intermediates associated with the metabolism of glutathione was studied in mice receiving methionine sulfoximine, a convulsant agent. The activity of the enzyme decreased significantly in the liver and kidney 1-4 h after administration of methionine sulfoximine; the activity of the enzyme in the brain was unchanged after 1 and 2 h but decreased significantly after 4 h. There was a rapid and sharp decrease in the concentration of glutathione in the kidney and a slower decrease in the liver. Brain glutathione concentrations were unaffected. Methionine sulfoximine in vivo, inhibited the synthesis of l -γ-glutamyl-l -α-aminobutyrate after administration of l -α-aminobutyrate, a reaction catalyzed by γ-glutamylcysteine synthetase. The inhibitor also lowered the concentration of pyrrolidone carboxylate in mouse tissues and prevented the accumulation of this intermediate after administration of l -α-aminobutyrate. The results show that methionine sulfoximine in vivo affects the metabolism of glutathione and that this action may contribute to its convulsive properties.  相似文献

Action of some phytohormones on the respiration and on the absorption of phosphate by aging potato tuber discs. Discs of potato tuber incubated in aerated medium show an increase of the rates of respiration and phosphate absorption with aging time; the rates increase by two and nine respectively during the time period between 5 and 24 h of aging. Adenine or some N-6 substituted adenines [benzylaminopurine (BAP), furfurylaminopurine (FAP), methylaminopurine (MAP)], which present variable degrees of cytokinin activity, partially inhibit the increase of the rate of phosphate absorption and, to a lesser extent, the increase of the rate of respiration. Also abscisic acid (ABA), indole 3-acetic acid (IAA), and gibberellic acid (GA3) produce inhibition of the increase of the rate of phosphate absorption with varied effects on the respiration. With regard to phosphate uptake, the effects of ABA, 1AA and GA3 were additive to those of BAP. The effects on respiration were different from the effects on phosphate uptake, so that there is no direct relationship between inhibition of respiration and inhibition of phosphate uptake.  相似文献

Abstract: G protein activation mediated by serotonin 5-HT1A and 5-HT1B/D receptors in guinea pig brain was investigated by using quantitative autoradiography of agonist-stimulated [35S]GTPγS binding to brain sections. [35S]GTPγS binding was stimulated by the mixed 5-HT1A/5-HT1B/D agonist L694247 in brain structures enriched in 5-HT1A binding sites, i.e., hippocampus (+140 ± 14%), dorsal raphe (+70 ± 8%), lateral septum (+52 ± 12%), cingulate (+36 ± 8%), and entorhinal cortex (+34 ± 5%). L694247 caused little or no stimulation of [35S]GTPγS binding in brain regions with high densities of 5-HT1B/D binding sites (e.g., substantia nigra, striatum, central gray, and dorsal subiculum). The [35S]GTPγS binding response was antagonized by WAY100635 (10 µM) and methiothepin (10 µM). In contrast, the 5-HT1B inverse agonist SB224289 (10 µM) did not affect the L694247-mediated [35S]GTPγS binding response, and the mixed 5-HT1B/D antagonist GR127935 (10 µM) yielded a partial blockade. The distribution pattern of the [35S]GTPγS binding response and the antagonist profile suggest the L694247-mediated response in guinea pig brain to be mediated by 5-HT1A receptors. In addition to L694247, 8-hydroxy-2-(di-n-propylamino)tetralin, and flesinoxan also stimulated [35S]GTPγS binding; their maximal responses varied between 46 and 52% compared with L694247, irrespective of the brain structure being considered. Sumatriptan, rizatriptan, and zolmitriptan (10 µM) stimulated [35S]GTPγS binding in the hippocampus by 20–50%. Naratriptan, CP122638, and dihydroergotamine stimulated [35S]GTPγS binding to a similar level as L694247 in hippocampus, lateral septum, and dorsal raphe. It appears that under the present experimental conditions, G protein activation through 5-HT1A but not 5-HT1B/D receptors can be measured in guinea pig brain sections.  相似文献

The hypothesis of Mann et al. that carbamate herbicides act as anti-derepressor substances was tested in two systems: cell enlargement and proliferation in cultured tobacco pith, and auxin-induced Avena first internode extension. Isopropyl-N-phenylcarbamate (IPC) and isopropyl-N-(3-chIorophenyl carbamate (CIPC) both severely inhibited kinetin-induced cell proliferation in tobacco pith at 10?6M. Auxin-induced tobacco cell enlargement was inhibited to the same degree only at 10?3M. Avena internode extension was insensitive to these compounds in the range 10?3 to 10?7M of IPC and 10?4 to 10?8M CIPC. The results support but do not prove Mann's hypothesis. Results of various studies with carbamates are interpreted in terms of this mechanism.  相似文献
