In vitro anticryptosporidial activity of dinitroaniline herbicides   总被引:4,自引:0,他引:4  
Abstract Despite the evaluation of over 100 antimicrobial drugs, the diarrheal disease cryptosporidiosis has remained refractory to treatment. We report the evaluation of five dinitroaniline herbicides including trifluralin, profluralin, nitralin, pendimethalin, and fluchloralin for anticryptosporidial activity in an in vitro cultivation model of Cryptosporidium parvum . All five compounds exhibited significant anticryptosporidial activities with no corresponding evidence of toxicity. The most active compound was pendimethalin with an IC50 of 0.19 μM while nitralin was the least active with an IC50 of 4.5 μM. These compounds should be evaluated further in an animal model of cryptosporidiosis.  相似文献

In field experiments on a sandy loam at Wellesbourne, England in 1972 and on a silt loam at Agassiz, British Columbia in 1973, combinations of herbicides and insecticides were applied at sowing to determine their effects on weeds and invertebrate populations and on the growth and yield of cauliflowers grown at high density There was good agreement between the results from the two locations. The two combinations of herbicides, 0.6 kg trifluralin/ha incorporated pre-drilling plus 2.2 kg propachlor/ha pre-emergence and 2.2 kg nitrofen/ha plus 2.2 kg propachlor/ha both applied pre-emergence, gave good weed control, their relative effectiveness depending on the species composition of the weed population. The insecticides isophenphos, carbofuran, chlorfenvinphos and fensulfothion were applied as bow-wave treatments. None of them, whether in combination with herbicides or not, adversely affected crop stand or yield. Yield was reduced when either weeds or root-fly maggots (Hylemya brassicae (Bouché)) were not controlled. Only in one experiment was there any evidence of any herbicide-insecticide interactions. Where trifluralin and carbofuran were applied together at Agassiz, the control of both weeds and maggots was less than that with the other combinations. None of the treatments affected the populations of predatory beetles, but the numbers of earthworms were greatly reduced by carbofuran and to a lesser extent by chlorfenvinphos. Except for carbofuran in one experiment, the treatments had no effects on the numbers of aphids, lepidopterous larvae or leaf miners present at harvest.  相似文献

Nitrogen fixing bacteria, rhizobia and phosphate solubilizing bacteria (PSB) are the commonly applied microbial inoculants in grain legumes (Pulses). It is important to apply herbicides to control weeds in order to augment yield of the crop. The herbicides may however, be incompatible with the microbial inoculants. This study compared the effect of the recommended pre-plant incorporated herbicide, fluchloralin (20.25 × 104 ppm) and pre-emergence herbicide, pendimethalin in two doses (9 × 104 and 15 × 104 ppm) on the growth and survival of mungbean Rhizobium and PSB, under laboratory conditions. These herbicides were also used under field conditions in conjunction with biofertilizers (R, PSB) to improve grain yield of mungbean. It was found that fluchloralin (20.25 × 104 ppm) and the lower dose of pendimethalin (9 × 104 ppm) had no adverse effect on growth of Rhizobium and PSB. The higher dose of pendimethalin (15 × 104 ppm) was safe on PSB but it imposed a retarding effect on the growth of Rhizobium.  相似文献

Summary Alachlor was more inhibitory toRhizoctonia solani growth than fluchloralin in potato dextrose broth (PDB). Infective capacity of the pathogen was not altered by growing it in a medium containing either of the herbicides. Cowpea seedlings grown in alachlor-treated soil were more susceptible toR. solani than those treated with fluchloralin and the untreated seedlings. Pre-sowing application of alachlor in soil (5 l a.i./kg) aggravated damping-off whereas fluchloralin decreased the disease incidence to nearly half of that in untreated soil in greenhouse pot tests (av. temperature 31±5°C). Both herbicides reduced damping-off in pots kept at constant temperature of 30°C and increased the disease incidence at 20°C. Fungus growth in culture was stimulated at 20° but was strongly inhibited at 30°C by both herbicides. Growth inhibition by herbicides was maximum in PDB of pH 8 and decreased steadily up to pH 5.Impact of fluchloralin and alachlor onR. solani damping-off of cowpea appears to be due to the predisposing effect by the herbicides on the susceptibility of the host and is influenced by atmospheric temperature.  相似文献

Field, greenhouse and laboratory investigations were conducted to determine the effect of four dinitroaniline herbicides on rhizobia, nodulation and nitrogen fixation of four groundnut cultivars. Benefin, dinitramine and nitralin used at recommended levels decreased nodule dry weight, nitrogenase activity and total nitrogen of groundnut tops and pod yield in three cultivars Kadiri 71-1, Kadiri-2, ICGS-11 and not for a fourth cultivar, Kadiri-3 of groundnut (Arachis hypogaea L.), but fluchloralin used at the recommended level increased the nodulation rate, nitrogenase activity and total nitrogen of groundnut tops and pod yield compared to untreated plants. Studies were conducted in vitro to determine the relative toxicity of the herbicides on four Rhizobium strains isolated from the nodules of four cultivars of groundnut. It was found that various strains of rhizobia differ in their sensitivity to different rates of the herbicides tested. Carbon dioxide exchange rate (CER) of all the cultivars which received herbicide treatment was measured at different time intervals to determine the relationship between photosynthesis and inhibition of nodulation. The lack of adverse effect on the CER of four cultivars when treated at recommended concentrations indicated that nitrogen fixation was affected in cultivars Kadiri 71-1, Kadiri-2 and ICGS-11 due to inhibition of nodulation.  相似文献

Experiments were performed to reveal the extent to which individual heterotrophic substrates of a mixture contribute to the overall carbon and energy metabolism. For this reason Hansenula polymorpha MH 20 was chemostatically (C-limited) cultivated at different growth rates on mixtures of methanol and glucose fed at proportions of 3:1 and 1:3 (in weight units), respectively. The distributions of 14C-carbon from methanol in biomass as well as carbon dioxide (and supernatant) fractions were determined. From these results it followed, firstly, that energy derived from methanol dissimilation was used in part for the incorporation of glucose carbon, resulting in carbon conversion efficiencies for this substrate equivalent to yield coefficients of 0.61–0.69 g/g. Secondly, the growth yield data revealed that the efficiency of methanol conversion had to be increased in order to account for the experimentally determined yield figures. This was further confirmed by theoretical treatment of the growth yield data which showed that these could only be obtained if P/O-quotients for methanol conversion similar to those for glucose, i.e. 2.0–2.5, were considered. Primary productivity in marine waters is widely estimated by the measurements of 14C incorporation, the underwater light climate, and the absorption spectra of phytoplankton. In bio-optical models the quantum efficiency of carbon fixation derived from 14C incorporation rates, the photosynthetically absorbed radiation derived from the underwater light climate, and the phytoplankton absorption spectra are used to calculate time- and depth-integrated primary productivity. Due to the increased sensitivity of commercially available fluorometers, chlorophyll a in vivo fluorescence became a new tool to assess the photosynthetic activity of phytoplankton. Since fluorescence data yield only relative photosynthetic electron transport rates, a direct conversion into absolute carbon fixation rates is not possible. Here, we report a procedure how this problem can be adressed in freshwater phytoplankton. We adapted a marine bio-optical model to the freshwater situation and tested if this model yields realistic results when applied to a hypertrophic freshwater reservoir. Comparison of primary productivity derived from 14C incorporation to primary productivity derived from Chl a fluorescence showed that the conversion of fluorescence data into carbon fixation rates is still an unsolved problem. Absolute electron transport rates calculated from fluorescence data tend to overestimate primary production. We propose that the observed differences are caused mainly by neglecting the package effect of pigments in phytoplankton cells and by non-carbon related electron flow (e.g., nitrogen fixation). On the other hand, the 14C incorporation rates can be artificially influenced by "bottle effects", especially near the water surface, where photoinhibition, photorespiration, and Mehler reaction can play a major role.  相似文献

Abstract

Aromatic amine herbicides, including propanil, fluometuron, alachlor, trifluralin, and pendimethalin, were examined for their ability to form haemoglobin adducts in rats as potential biomarkers of exposure. Many aromatic amines are known to form haemoglobin adducts via conversion to the nitroso metabolite and binding of this metabolite to cysteinyl groups on haemoglobin. Since red blood cells are long lived, adducts formed with these cells may be reliable biomarkers of exposure with the potential for showing progressive accumulation. Gas chromatographic-mass spectrometric analyses of haemoglobin revealed that adducts were formed in rats treated with the rice herbicide propanil and the cotton herbicide fluometuron. Adducts were not detected with the herbicides alachlor, trifluralin, or pendamethalin.  相似文献

Cosubstrates fermentation is such an effective strategy for increasing subject metabolic products that it could be available and studied in propionic acid production, using glycerol and glucose as carbon resources. The effects of glycerol, glucose, and their mixtures on the propionic acid production by Propionibacterium acidipropionici CGMCC1.2225 (ATCC4965) were studied, with the aim of improving the efficiency of propionic acid production. The propionic acid yield from substrate was improved from 0.475 and 0.303 g g−1 with glycerol and glucose alone, respectively, to 0.572 g g−1 with co-fermentation of a glycerol/glucose mixture of 4/1 (mol/mol). The maximal propionic acid and substrate conversion rate were 21.9 g l−1 and 57.2% (w/w), respectively, both significantly higher than for a sole carbon source. Under optimized conditions of fed-batch fermentation, the maximal propionic acid yield and substrate conversion efficiency were 29.2 g l−1 and 54.4% (w/w), respectively. These results showed that glycerol/glucose co-fermentation could serve as an excellent alternative to conventional propionic acid fermentation.  相似文献

The effects of four herbicides and hand weeding and no weeding on soybean disease development and seed quality were studied in the field. Chloramben, dinoseb, fluchloralin, and trifluralin were preplant incorporated at recommended rates. In addition, the effect of an in-furrow oat (Avena sativa L.) amendment used as an inoculum carrier was studied in all treatments. Compared with the hand-weeded control, yield, incidence of Septoria leafspot (Septoria glycines), and the recovery of Phomopsis spp. from pod and stem tissues were reduced in the week-infested control and dinosebtreated plots. Recovery of Alternaria spp. was greater from crown tissues of plants grown in chloramben- and fluchloralin-treated plots compared with, either the weed-infested or hand-weeded control. Septoria leafspot ratings and recovery of Phomopsis spp. from various plant tissues were higher in plants from hand-weeded plots compared with weed-infested plots, whereas the opposite was truefor, the recovery of Fusarium spp. Chloramben-treated plots were high-yielding and had the best weed control of all herbicide treatments. Seed quality was not affected by herbicide treatment. Plants from furrows with autoclaved oats had a lower incidence of Septoria glycines, a greater recovery of Fusarium spp. from pod and stem pieces, and produced seed with lower quality than plants from rows with no oat amendment.  相似文献

The potential impact of three widely used herbicides, pendimethalin, prometryn, and trifluralin, on populations of five plant disease suppressing bacterial isolates (three isolates of Pseudomonas fluorescens and two isolates of Burkholderia cepacia) in the rhizosphere of cotton seedlings was investigated. All isolates are efficient cotton root colonizers and each is capable of suppressing a plant disease. In microcosm experiments, application of each of the test herbicides at the rates of 1, 2, and 4 µg active ingredient (a.i.) g-1 soil caused significant (p<0.05) reductions in populations of most of the isolates in the rhizosphere, 14 days after the release of bacteria into the soil by seed coating. The responses of the isolates to the herbicides varied depending on the isolate and the type and concentration of the herbicides. In microcosm experiments the impact of pendimethalin, prometryn, and trifluralin at the respective concentrations of 2.4, 3.6, and 1.8 g a.i. g-1 soil on the population of isolate D1 in the cotton rhizosphere declined with time during a four week period of monitoring following the release of the isolate into the soil by seed coating. The impact of soil applied test herbicides on the population sizes of D1 in cotton rhizosphere was also studied in two field experiments (Safford and Tucson, Arizona) where the bacteria were added as a soil drench. In the Safford experiment pendimethalin and prometryn, but not trifluralin, caused significant (p <0.05) reductions in the population of the bacterium 15 days after sowing. In the Tucson experiment a significant (p < 0.05) reduction in the population of the bacterium was observed 15 and 25 days after sowing in soils treated with pendimethalin and prometryn and 25 days after sowing in soils treated with trifluralin.  相似文献

Glucose but not aniline increased the yield of products formed during propachlor cometabolism in sewage. No microorganism able to use propachlor as a sole source of carbon and energy was isolated, but bacteria isolated from sewage and lake water metabolized this chemical. During the metabolism of this herbicide by two of the bacteria, none of the carbon was assimilated. Our data indicate that cometabolism of these pesticides takes place at concentrations of synthetic compounds that commonly occur in natural waters.  相似文献

The colR4 and colR15 beta 2-tubulin missense mutations for lysine-350 in Chlamydomonas reinhardtii (Lee and Huang, 1990) were originally isolated by selection for resistance to the growth inhibitory effects of colchicine. The colR4 and colR15 mutants have been found to be cross resistant to vinblastine and several classes of antimitotic herbicides, including the dinitroanilines (oryzalin, trifluralin, profluralin, and ethafluralin); the phosphoric amide amiprophos methyl; and the dimethyl propynl benzamide pronamide. Like colchicine and vinblastine, the antimitotic effects of these plant-specific herbicides have been associated with the depolymerization of microtubules. In contrast to their resistance to microtubule-depolymerizing drugs, the mutants have an increased sensitivity to taxol, a drug which enhances the polymerization and stability of microtubules. This pattern of altered sensitivity to different microtubule inhibitors was found to cosegregate and corevert with the beta-tubulin mutations providing the first genetic evidence that the in vivo herbicidal effects of the dinitroanilines, amiprophos methyl, and pronamide are related to microtubule function. Although wild-type like in their growth characteristics, the colR4 and colR15 mutants were found to have an altered pattern of microtubules containing acetylated alpha-tubulin, a posttranslational modification that has been associated with stable subsets of microtubules found in a variety of cells. Microtubules in the interphase cytoplasm and those of the intranuclear spindle of mitotic cells, which in wild-type Chlamydomonas cells do not contain acetylated alpha-tubulin, were found to be acetylated in the mutants. These data taken together suggest that the colR4 and colR15 missense mutations increase the stability of the microtubules into which the mutant beta-tubulins are incorporated and that the altered drug sensitivities of the mutants are a consequence of this enhanced microtubule stability.  相似文献

Groundnut (Arachis hypogaea L.) productivity in India is low, because of many problems beset in its cultivation. One of the serious problems are weeds. Groundnut yield losses due to weeds have been estimated as high as 24 to 70 percent. This has created a scope for using herbicides in groundnut crop. A field investigation was carried out during kharif (rainy) season of 2001-2002 on a sandy loam soil at College Agronomy Farm, B.A. College of Agriculture, Gujarat Agricultural University, Anand, India to study the effect of weed management practices and Rhizobium inoculation on growth and yield of groundnut (Arachis hypogaea L.). Ten weed control treatments, comprising four treatments of sole application of fluchloralin, pendimethalin, butachlor and metolachlor, respectively each applied at 1.0 kg ha(-1); four treatments comprising of an application of the same herbicides at the same levels coupled with one hand weeding at 30 DAS; one weed-free treatment (hand weedings at 15, 30, 45 DAS); and one unweeded control. All 10 treatmets were combined with and without Rhizobium inoculation (i.e. a total of 20 treatment combinations) under a factorial randomized complete block design (FRBD) with four replications. Minimum weed dry matter accumulation (70 kg/ha) with higher weed control efficiency (90.70%) was recorded under an integrated method i.e. pendimethalin at 1.0 kg ha(-1) + hand weeding at 30 DAS, which also resulted in maximum pod yield (1773.50 kg ha(-1)). This treatment was comparable to fluchloralin applied at 1.0 kg ha(-1) combined with hand- weeding at 30 DAS. Weedy conditions in the unweeded control treatment reduced pod yield by 29.90-35.95% as compared to integrated method. Significantly higher pod yield was obtained with Rhizobium inoculation than the mean value of all treatments without inoculation. For most agronomical parameters examined, Rhizobium inoculation and weed control treatments were independent in their effect.  相似文献

Possibility to chemically control weeds in spring rapeseed has been tested in two locations ( Novi Sad and Kragujevac) and following herbicides (a.i.) : trifluralin, clomazone, quizalofop-p-ethyl and clopyralid. We tested the effect of the herbicides on yield and hectoliter weight of seed and oil and protein contents in seed. In the trial in Kragujevac, a large number of weed species were present, with somewhat increased density and uneven distribution of weed plants. This was particularly evident with grassy weeds and with the species Rubus caesius in several plots. Rapeseed yield and quality were determined by measuring and analyzing the following parameters: grain yield (kg/plot (30 m2), hectoliter weight, oil content (%) and protein content (%) in seed.Basic statistical calculations of rapessed yield and quality were done by the t-test. The tested herbicides showed no adverse effect on the yield and hectoliter weight of seed in either location, with the exception of quizalofop-p-ethyl in Kragujevac, which affected the control variants. Oil content was negatively affected by the combination, trifluralin + clopyralid in the location of Novi Sad and by quizalofop-p-ethyl in the other location. Trifluralin and quizalofop-p-ethyl exhibited a negative effect on protein content in the location of Novi Sad, while there were no statistically significant negative effects in the other location.  相似文献

Mixtures of (14)C-labelled glucose plus pyruvate were incubated either with rat diaphragm or slices of rat liver. Incorporation of glucose carbon into glycogen was compared with its incorporation into glucose 6-phosphate relative to the incorporation of pyruvate carbon into these metabolic products. There was no preferential incorporation of glucose carbon relative to pyruvate carbon into glycogen compared with glucose 6-phosphate in the liver slices, but there was in diaphragm. On the assumption that glucose 6-phosphate is a necessary intermediate in the conversion of pyruvate carbon into glycogen, this is evidence for the existence in muscle, but not in liver, of more than one pool of glucose 6-phosphate or of a pathway from glucose to glycogen without glucose 6-phosphate as an intermediate. Galactose carbon, relative to pyruvate carbon, was preferentially incorporated into liver glycogen, so that a substrate converted in liver into glycogen without glucose 6-phosphate as an intermediate could be detected by this approach.  相似文献

Using this new dopant for carbon nanotube–silicon solar cells, advanced substrate design, and an optimized antireflective texture fast etch with organic base, breakthrough performance is obtained from research grade devices with active areas of 1 and 5 cm2, which yield power conversion efficiencies of 17.2 and 15.5%, respectively.  相似文献

1. Microbial parameters were determined at five sampling sites in the River Danube up-and downstream of Vienna, Austria, twice monthly over an annual cycle. Bacterial production (BP) was estimated from thymidine and leucine incorporations; additionally, the effect of turbulence on BP and the conversion factors for converting incorporation rates into bacterial cell production were determined using the cumulative approach. 2. BP under turbulent conditions was not significantly different from that under stagnant conditions. For thymidine, a mean annual conversion factor of 3.2 ± 1018 cells mol?1 thymidine incorporated was calculated. For leucine, the corresponding factor was 0.07 ± 1018 cells mol?1 leucine. Average annual BP calculated by thymidine incorporation was significantly higher than BP calculated from leucine incorporation and ranged from 47.2 to 77.5 μg C 1-?1 day?1 depending on the tracer and the conversion factor used. 3. Bacterial growth rates ranged from 0.1 day?1 during winter to 1.7 day?1 in the summer. A strong correlation was found between temperature as well as chlorophyll a and bacterial growth when temperature was greater than 5 °C; a major spring phytoplankton bloom at a temperature below 5 °C did not increase BP. 4. Dissolved organic carbon (DOC) concentrations varied between 2 and 7.2 mg C 1-?1 and comprised between 50 and 92% of the total organic carbon pool in the River Danube, Based on the DOC concentration and an assumed bacterial growth yield of 20% we calculated mean DOC turnover times of around 60 days in the winter and less than 8 days during the summer.  相似文献

5-Hydroxymethylcytosine has recently been characterized as the ‘sixth base’ in human DNA. To enable research on this DNA modification, we report an improved method for the synthesis of 5-hydroxymethyl-2′-deoxycytidine (5-HOMedC) phosphoramidite for site-specific incorporation into oligonucleotides. To minimize manipulations we employed a temporary protecting group-free 2′-deoxyuridine to 2′-deoxycytidine conversion procedure that utilizes phase transfer catalysis. The desired 5-HOMedC phosphoramidite is obtained in six steps and 24% overall yield from 2′-deoxyuridine.  相似文献
