研究了不同浓度的水杨酸时水华鱼腥藻生长的影响以及对鲤鱼的毒性作用.结果表明,一定浓度的水杨酸能有效抑制水华鱼腥藻的生长;抑藻作用的最佳浓度为0.6 mmol/L,用该浓度的水杨酸溶液处理鲤鱼1周后,鲤鱼血清中的天门冬氨酸氡基转移酶(AST)、丙氨酸氨基转移酶(ALT)、γ-谷氨酰基转移酶(GGT)、碱性磷酸酶(ALP)的活性没有明显影响,对肝脏、鳃和肌肉中的超氧化物歧化酶(SOD)及膜脂过氧化产物(MDA)的活性也没有明显影响,提示水杨酸时鲤鱼无毒性作用,时水华治理有一定的应用前景.  相似文献

为研究3'-大豆苷元磺酸钠(DSS)的体外抗氧化活性,测定了DSS对超氧自由基(O2·)、羟基自由基(·OH)和1,1-二苯基-2.苦苯肼自由基(DPPH·)的清除能力,及其对Fe2 诱导的脂质过氧化反应和β-胡萝卜素/亚油酸自氧化体系的抑制作用.结果表明,DSS对DPPH·有一定的清除能力,对超氧自由基的清除能力较强,能很好地清除·OH自由基,对脂质过氧化有一定的抑制作用,对β-胡萝卜素/亚油酸自氧化体系有明显的抑制作用.说明DSS的药理作用可能与其较强的抗氧化能力有关.  相似文献

研究了水杨酸对盐胁迫下黄瓜幼苗生长抑制的缓解效应及其机制。结果证明,盐胁迫条件下水杨酸能提高幼苗相对含水量,降低Na^ 、K^ 向上运输的选择性,通过促进子叶内超氧化物歧化酶、过氧化物酶活性降低膜脂过氧化产物丙二醛含量和质膜透性,缓解了盐胁迫对幼苗生长的抑制。  相似文献

为了探讨外源水杨酸(SA)提高植物抗旱性的相关机制,研究了干旱胁迫下(基质含水量为饱和持水量的60%和50%),根际施用外源SA对黄瓜幼苗生长、膜脂过氧化、脯氨酸积累、水分利用效率、净光合速率(Pn)和叶绿素荧光参数的影响.结果表明:SA处理能够缓解干旱胁迫对黄瓜幼苗生长、Pn和水分利用效率的抑制,减小膜脂过氧化程度,促进脯氨酸的积累;添加外源SA显著减小了干旱胁迫下黄瓜幼苗的PSⅡ最大光化学效率、PSⅡ实际光化学效率、PSⅡ潜在活性、PSⅡ有效光化学效率和光化学猝灭系数的下降幅度,抑制了非光化学猝灭系数的升高.添加外源SA可以缓解干旱胁迫造成的膜脂过氧化对膜系统的氧化损伤,并通过增强PSⅡ反应中心活性提高了Pn,有助于水分的利用,同时增大渗透调节能力,减少水分的散失,提高水分利用效率,从而增强植株对干旱的适应能力.  相似文献

本工作研究了氧自由基在大鼠冷冻束缚应激性胃溃疡中的发病学意义。实验结果如下:(1)以超氧自由基清除剂超氧化物歧化酶(SOD)或羟自由基清除剂二甲亚砜和甘露醇预先处理大鼠,均可显著地减轻胃粘膜损伤;(2)应激时,胃粘膜内的脂质过氧化分解产物丙二醛的含量显著升高;(3)组织化学的研究显示,胃粘膜层含有丰富的黄嘌呤氧化酶,其活性在应激时明显升高,预先用别嘌呤醇处理大鼠以抑制黄嘌呤氧化酶的活性,可使胃粘膜损伤显著减轻。上述结果提示,氧自由基是应激性胃溃疡的重要致病因子,而黄嘌呤氧化酶活性的升高似可能为应激时氧自由基生成增加的重要原因。  相似文献

研究了原子氧自由基阴离子(O-)对大肠杆菌的失活作用和形貌变化的影响.实验所用的O-自由基由新研制的O-发生器制备,其中[Ca24Al28O64]4· 4O-(缩写为C12A7-O-)材料是O-发生器中发射O-的部分.实验结果表明,大肠杆菌的失活率随O-强度的变化而变化,在O-强度为1.5μA/cm2时,细胞的死亡率大大加强,作用120min后,细胞死亡率超过3个对数量级.通过场发射扫描电子显微镜观察发现O-对细胞结构具有破坏作用.通过丙二醛(MDA)的形成证实了O-诱导大肠杆菌发生脂质过氧化反应过程的存在,这可能是大肠杆菌死亡的潜在原因.当1.5μA/cm2的O-流通入到大肠杆菌悬浊液后,丙二醛浓度开始升高,15min后达到最高值1.2μmol/g,然后缓慢下降.结果显示,原子氧自由基阴离子能失活大肠杆菌,诱导脂质过氧化反应,这对发展一种新的净化微生物污染方法和研究微生物与原子氧自由基阴离子相互作用具有潜在的意义.  相似文献

不同温光条件(4d)下,以粳稻(Dryza sativaL.sp.japonica)“9516”和籼稻(O.sativaL.sp.indica)“汕优63”为材料,测定了与PSⅡ光能转化效率(Fv/Fm)和膜脂过氧化作用有关的生理指标。结果表明:适温、中等光强下两品种Fv/Fm和膜脂过氧化作用的产物(MDA)无变化,未见光抑制和光氧化表现;适温、强化下籼稻“汕优63”的Fv/Fm明显下降,MDA未见变化,有光抑制无光氧化表现;低温、强光下两品种有光抑制和光氧化表现。低温、强光下结合抑制剂实验证明,与粳稻相比,籼稻的D1蛋白量和SOD活性下降较多,叶黄素循环和非光化学猝灭(qN)受抑制程度较大,膜脂过氧化产物MDA含量较高,因而光抑制和光氧化现象较明显。实验提出:光能转化效率和膜脂过氧化表现是预测光氧化的关键指标。  相似文献

氢气是一种高效,无毒的氧自由基清除剂,研究表明氢气能通过清除氧自由基减轻缺血、中毒引起的心肝肾以及胃肠和神经等多种组织损伤,促进溃疡的修复。本研究通过观察注射氢盐水对运动性贫血大鼠红细胞及铁代谢指标的影响,探讨防治运动性贫血的新思路和方法。结果表明,在建立大鼠运动性贫血模型后,注射氢盐水大鼠的血红蛋白(Hb)、红细胞计数(RBC)和红细胞压积(Hct)等指标明显高于实验对照组。注射氢盐水可减少红细胞氧化损伤造成的溶血,提高红细胞相关指标,改善大鼠的运动性贫血状况。  相似文献

以小麦品种‘新麦18’为材料,采用室内水培实验研究了不同浓度水杨酸(SA)处理对300 mg.L-1锌胁迫下小麦种子萌发和幼苗生长的影响。结果表明:在Zn2+胁迫下,小麦种子的发芽势和发芽率、幼苗根长、芽长以及幼苗叶片的可溶性蛋白含量、根系活力显著降低,而脯氨酸和丙二醛(MDA)含量显著增加(P<0.05);外施SA显著提高了Zn2+胁迫下小麦种子的发芽势和发芽率,同时也使Zn2+胁迫7 d后的小麦幼苗的根长、芽长,幼苗叶片的脯氨酸和可溶性蛋白含量以及根系活力显著升高,膜脂过氧化产物MDA含量却显著降低(P<0.05)。由此可见,外施SA可通过提高小麦幼苗根长和芽长来增加幼苗根系活力,通过提高小麦幼苗可溶性蛋白含量、脯氨酸含量来维持细胞膜的稳定性,降低膜脂过氧化伤害程度,从而缓解了Zn2+胁迫对幼苗生长的抑制,并以14 mg.L-1外源水杨酸缓解效果最好。  相似文献

The passive permeation rates of DMPO and DEPMPO spin traps and their hydroxyl radical adducts through liposomal membranes were measured using ESR spectroscopy. For the spin traps, we measured the time-dependent change in the signal intensity of the OH-adduct, which is formed by a reaction between the penetrated spin trap and hydroxyl radicals produced by the UV-radiolysis of H(2)O(2) inside the liposomes. The hydroxyl radicals produced outside the liposomes were quenched with polyethylene glycol. For the OH-adduct, pre-formed adduct was mixed with liposomes and the time-dependent change of the ESR signal was measured in the presence of a line-broadening reagent outside the liposomes to make the signal outside the liposomes invisible. Both the spin traps and their OH-adducts diffused across the lipid membranes rapidly and reached equilibrium within tens of seconds. These findings suggest that if used for the detection of free radicals inside cells, these spin traps should be well distributed in cells and even in organelles.  相似文献

The effect of caffeic acid, a kind of catechol, on the Fenton reaction was examined by using the ESR spin trapping technique. Caffeic acid enhanced the formation of hydroxyl radicals in the reaction mixture, which contained caffeic acid, hydrogen peroxide, ferric chloride, EDTA, and potassium phosphate buffer. Chlorogenic acid, which is an ester of caffeic acid with quinic acid, also stimulated the formation of the hydroxyl radicals. Quinic acid did not stimulate the reaction, suggesting that the catechol moiety in chlorogenic acid is essential to the enhancement of the hydroxyl-radical formation. Indeed, other catechols and related compounds such as pyrocatechol, gallic acid, dopamine, and noradrenaline effectively stimulated the formation of the hydroxyl radicals. The above results confirm the idea that the catechol moiety is essential to the enhancement. Ferulic acid, 4-hydroxy-3-methoxybenzoic acid, and salicylic acid had no effect on the formation of the hydroxyl radicals. The results indicate that the enhancement by the catechols of the formation of hydroxyl radicals is diminished if a methyl ester is formed at the position of the hydroxyl group of the catechol. In the absence of iron chelators such as EDTA, DETAPAC, desferrioxamine, citrate, and ADP, formation of hydroxyl radicals was not detected, suggesting that chelators are essential to the reaction. The enhancement of the formation of hydroxyl radicals is presumably due to the reduction of ferric ions by the catechols. Thus, the catechols may exert deleterious effects on biological systems if chelators such as EDTA, DETAPAC, desferrioxamine, citrate, and ADP are present.  相似文献

This paper describes the use of complex liposomes as real membrane models to evaluate the potential benefits of several antioxidants in relation to lipid peroxidation. The xanthine oxidase/Fe(3+)-ADP-EDTA and the Fe(2+)/H2O2 systems have been used to generate hydroxyl radicals and the water soluble azo-compound 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH) to generate carbon centered radicals (A*) by thermal decomposition. The antioxidant behavior of the rosemary and citrus plant extracts and vitamin-E and vitamin-E acetate alpha-tocopherols have been analyzed. The order of effectiveness in avoiding radical chain reactions has been established by using the colorimetric thiobarbituric acid reaction and the fluorescent probe DPH-PA. ESR spectroscopy has been used to carry out the pursuit of the oxidation processes on the basis of the identification of the radical species resulting from the oxidant system and the ability of the antioxidants to act as scavengers for hydroxyl and AAPH-derived radicals. The modification of the main transition temperature for the lipid mixture and the splitting of the calorimetric peak in the presence of the antioxidants were demonstrated by differential scanning calorimetry. The results obtained showed that the phenols-containing plant extracts and alpha-tocopherols perturb the phase behavior of the BBE lipid bilayer and have a fluidifying effect that could favor the known antioxidant capability and scavenging characteristics of these compounds. 31P-NMR results could be interpreted as, after the incorporation of these antioxidants, those lipid molecules interacting with antioxidants give rise to lamellar phase spectral components with resonance position at lower fields or to isotropic signals in accordance with a higher motion of their phosphate groups.  相似文献

The antioxidant activities of trans-resveratrol (trans-3,5,4′-trihydroxystilbene) and trans-piceid (trans-5,4′-dihydroxystilbene-3-O-β-d-glucopyranoside), its more widespread glycosilate derivative, have been compared measuring their inhibitory action on peroxidation of linoleic acid (LA) and the radical scavenging ability towards different free radicals (such as DPPH) and radical initiators. It has been found that the two stilbenes have similar antioxidant capacity, while the comparison with BHT (2,6-di-tert-butyl-4-methylphenol) and -tocopherol (vitamin E, vit. E), taken as reference, points out a slower but prolonged protective action against lipid peroxidation. Furthermore, piceid appears more efficacious than resveratrol as a consequence of the reaction of the latter with its radical form.

The DSC profiles of phosphatidylcholine liposomes of various chain lengths, and EPR measurements of spin labelled liposomes demonstrated that the susceptible hydroxyl group of these compounds are located in the lipid region of the bilayer close to the double bonds of polyunsatured fatty acids, making these stilbenes particularly suitable for the prevention and control of the lipid peroxidation of the membranes.  相似文献

A novel conjugate of melatonin 2 and alpha-lipoic acid 4 has been prepared using DCC mediated coupling. The conjugate named melatoninolipoamide has been assigned its structure 1 on the basis of spectral analysis (UV, IR, NMR, and EI-MS). Pulse radiolysis studies of the conjugate were carried out in aqueous solutions with both oxidizing and reducing radicals. The results indicate that the melatonin moiety of the conjugate reacts preferably with oxidizing radicals and the lipoic acid moiety exhibits preferential reaction with reducing radicals. The in vitro radioprotection ability of 1 was examined by gamma-radiation induced lipid peroxidation in liposomes and hemolysis of erythrocytes, and compared the results with those of melatonin and alpha-lipoic acid. The studies suggest that the conjugate can be explored as a probable radioprotector.  相似文献

SUMMARY

The ability of hydroxylated metabolites of salicylic acid to scavenge reactive oxygen species and to inhibit arachidonic acid metabolism was investigated. The tested trihydroxybenzoic acids (THBAs) were potent scavengers of hydroxyl and superoxide anion radicals produced by Fenton reaction and xanthine/xanthine oxidase system or activated macrophages respectively. In the same tests, salicylic acid possessed moderate O2? and low OH'scavenging activities.

Our results demonstrate that adding two hydroxyl groups to salicylic acid strongly increases the reactive oxygen species (ROS) scavenging activities. Adding two hydroxyl groups at position 4 and 5 (2,4,5-THBA) affords the most active ROS scavenging activity probably due to the ortho unsubstituted catechol moiety. In fact, we can consider that the ROS scavenging properties of salicylic acid are essentially due to its metabolic products such as 2,3- and 2,5-DHBAs, catechol and also to THBAs.  相似文献

Although the presence of free radicals has been indicated in ischemic-reperfused heart, the exact nature and source of these free radicals are not known. The present study utilized a chemical trap, salicylic acid, to trap hydroxyl radical which could be detected as hydroxylated benzoic acid using high pressure liquid chromatography. Since the hydroxylated product is extremely stable, heart was subjected to subcellular fractionation after ischemia and reperfusion, and each fraction was separately examined for the presence of hydroxyl radical. The results indicated for the first time the presence of hydroxyl radical in the mitochondrial fraction during early reperfusion, which decreased in intensity as the reperfusion progressed.  相似文献

This work stresses the need to combine antioxidant assays and drug-membrane interaction studies to describe more accurately the antioxidant profile of non-steroidal anti-inflammatory drugs (NSAIDs). Different experiments performed in liposomes and aqueous solution were compared and used to evaluate the protective effect of etodolac in lipid peroxidation. Lipid peroxidation was induced by the peroxyl radical (ROO*) derived from 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH) and hydroxyl radical (HO*) generated by the Fenton reaction and was assessed by the fluorescence intensity decay of three fluorescence probes with distinct lipophilic properties--fluorescein; hexadecanoyl aminofluorescein (HDAF) and diphenylhexatriene propionic acid (DPHPA). Membrane fluidity changes due to lipid peroxidation were also evaluated by steady-state anisotropy measurements. Interactions of etodolac with lipid bilayers were evaluated by membrane zeta-potential measurements. Results indicate a drug location near the membrane surface and show that etodolac can scavenge the radicals studied but to a variable extent, depending on the assayed media and reactive species. The use of different probes and liposomes as membrane mimetic systems allowed us to conclude that membrane lipoperoxidation is not only related to the scavenging characteristics of the antioxidants, but also to their ability to interact with lipid bilayers.  相似文献

Here we show that lipid peroxidation of liposomal membranes was suppressed in the presence of Caroverine, a spasmolytic drug used in some countries. In order to understand the mechanism of this antioxidant action of Caroverine we studied the interaction of Caroverine with superoxide radicals, hydroxyl radicals and peroxynitrite. The results of the study show that the reaction of Caroverine with O2-* radicals is of marginal significance. However, it is efficient in removing peroxynitrite and a particular high reaction constant was found for reaction with hydroxyl radicals. The strong antioxidant activity of Caroverine is therefore based both on the partial prevention of the formation and the highly active scavenging of hydroxyl radicals.  相似文献

本文研究了过氧化氢与亚铁离子体系产生的羟基自由基对兔脑微粒体脂质过氧化作用及对膜上(Na~++K~+)-ATP酶活性的影响.结果表明,羟基自由基导致兔脑微粒体脂质过氧化,增加丙二醛的含量.羟基自由基还使微粒体膜巯基数下降,(Na~++K~+)-ATP酶活力受到抑制.阿魏酸钠对抑制微粒体脂质过氧化及对膜巯基和(Na~++K~+)-ATP酶均有保护作用.自旋捕集实验结果进一步证明药物对羟基自由基的猝灭作用.  相似文献
