Regulation of nodule permeability in response to short-term changes in environmental and physiological conditions is thought to occur by occlusion of intercellular spaces in the nodule inner cortex. To test this hypothesis, the permeability of legume nodules was altered by adapting them to either 20 or 80% O2 over a 2.5-h period. The nodules were then rapidly frozen, cryo-planed and examined under cryo-scanning electron microscopy for differences in the number, area or shape factor of intercellular spaces. Comparisons were made between whole nodules and specific nodule zones (outer cortex, middle cortex, inner cortex and central zone) in each treatment. Gas analysis measurements indicated that nodules equilibrated at 20% O2 had a 6.6-fold higher permeability than those equilibrated at 80% O2 However, no significant differences were observed between pO2 treatments in the number of open intercellular spaces, the cross-sectional area of those spaces, or the proportion of the tissue area present as open space in whole nodules or any nodule zone. Also, although nodules in both treatments possessed a boundary layer of tightly packed cells in the inner cortex, the total area of intercellular spaces between cells bordering this layer did not differ between treatments. Together these observations do not support the currently favored hypothesis that nodule permeability is regulated by opening or occlusion of intercellular spaces in the nodule inner cortex. Highly significant differences (P= 0.0006) were observed between O2 treatments in the shape factor of the open intercellular spaces in all nodule zones. Nodules equilibrated at 80% O2 had significantly more isodiametric spaces while those equilibrated at 20% O2 had more long, narrow spaces. This observation suggests that the critical step in the regulation of nodule permeability to O2 may be localized in the central, infected zone and involve changes in the ratio of the surface area of the intercellular space to the volume of the infected cell.  相似文献

A structural analysis was conducted to determine whether glycoprotein‐containing intercellular space occlusions are involved in medium‐term regulation of O2 diffusion in soybean (Glycine max) nodules. Alterations in O2 diffusion were induced by a 3 h detopping treatment, and glycoprotein was immunolocalized with the monoclonal antibodies MAC236 and MAC265. Western blots of unstressed nodules revealed that these antibodies recognize antigens with two different molecular weights in soybean nodules. Tissue printing of halved nodules showed that both antigens were present in fresh nodules from control and 3 h detopped plants. The main localization appeared to be the inner cortex, but some immunolabelling also occurred in the infected region. ELISAs demonstrated a significant increase in total nodule concentration of intercellular glycoprotein following detopping, and cryosections of fresh nodules from this treatment also showed localization of antigens within the intercellular spaces of the infected region. The production of intercellular space occlusions in both the mid‐cortex and infected regions after 3 h detopping was confirmed by light microscopy and silver‐enhanced immunolabelling; cortical changes were quantified by image analysis techniques. Electron microscopy revealed that the occlusions within the infected region were less dense and less heavily labelled than those in the cortex. Polyclonal antibodies raised against NAD-dependent glyceraldehyde-3-phosphate dehydrogenase (D-glyceraldehyde-3-phosphate:NAD+ oxidoreductase [phosphorylating], EC 1.2.1.12) from the plant cytosolic fraction of soybean [ Glycine max (L.) Merr. cv. Williams] nodules were used to study the subcellular location of the enzyme and its relative distribution between infected, interstitial and cortical cells of soybean (cv. Lincoln) nodules. Post-embedding immunogold labelling was carried out on nodules harvested 5, 12, 19 and 25 days after the first sign of nodulation. Labelling for glyceraldehyde-3-phosphate dehydrogenase was observed over the cytoplasm and nuclei of infected and uninfected cells, as well as over the nucleoid regions of bacteroids. In 5-day-old nodules, label also bound adjacent to the peribacteroid membranes. Statistical analysis of the number of gold particles per cell area indicated that in 5-day-old nodules, glyceraldehyde-3-phosphate dehydrogenase was distributed equally between infected, interstitial and cortical cells, but in older nodules the enzyme was more prominent in the interstitial and cortical cells than in infected cells.  相似文献

Ammonia, the primary product of nitrogen fixation is rapidly incorporated into a number of amino acids such as glutamate and aspartate. A novel enzyme system glutamine: 2-oxoglutarate aminotransferase oxidoreductase, which probably has an important role in ammonia assimilation has been detected, in the present studies, in the rhizobial fraction of soybean root nodules and in Rhizobium japonicum grown in culture. The role of this latter enzyme and other enzymes such as glutamate dehydrogenase, aspartate aminotransferase and alanine aminotransferase in ammonia assimilation by soybean nodules is discussed.  相似文献

Measurements of the short-term response of nodulated roots of soybean ( Glycine max L. Merr, cv. Harosoy: Bradyrhizobium japonicum USDA 16) to rapid changes in surrounding pO2 indicate that their ability to reversibly adjust gaseous diffusive resistance is retained whether plants are cultured in rhizospheres of very low (2.8%) or very high (61.2%) pO2. Thus the capacity for reversible short-term diffusion adjustment is additional to structural changes in the fixed diffusional barriers of nodules which allow their continued fixation of N2 in unfavourably high or low external pO2. Anatomical evidence, involving quantitative measurement of intercellular spaces in the cortical tissues using electron microscopy of thin sections, indicates that the major fixed diffusional barrier is a boundary layer of cells in the inner cortex which may be as small as one cell thick in nodules from 2.8% O2 to 5 or 6 cells thick, and almost completely devoid of intercellular spaces, in those from 61.2% O2. The data are interpreted to indicate that the variable diffusion harrier is distinct from the boundary layer and is most likely to be a property of cells and/or intercellular spaces inside the boundary layer of the nodule cortex.  相似文献

The impact of exclusion of solar UV-B (280–320 nm) and UV-A+B (280–400 nm) radiation on the root nodules was studied in soybean(Glycine max var. MACS 330). Soybean plants were grown in the tropical region of Indore (Latitude-22.4°N), India under field conditions in metal cages covered with polyester exclusion filters that specifically cut off UV-B (<320 nm) and UV-A+B (<400 nm) radiation; control plants were grown under ambient solar radiation. Leghemoglobin content was analyzed in the root nodules on the 50th day after emergence of seedlings. Exclusion of UV radiations significantly enhanced the leghemoglobin content in the nodules on fresh weight basis; 25% and 45% higher amount of leghemoglobin were present in the nodules after the exclusion of UV-B and UV-A+B radiation respectively. Analysis by native and SDS-PAGE showed high intense bands of leghemoglobin after the exclusion of UV-A+B as compared to control. Exclusion of UV radiation also enhanced the growth of roots as well as aerial parts of the plants. UV Exclusion increased nodulation by increase in the number and size of nodules. The results are discussed in the light of advantage of exclusion for enhancing protein/nitrogen content in the plants.  相似文献

Oxygen and the regulation of nitrogen fixation in legume nodules   总被引:3,自引:0,他引:3  
In N2-fixing legume nodules, O2 is required in large amounts for aerobic respiration, yet nitrogenase, the bacterial enzyme that fixes N2, is O2 labile. A high rate of O2 consumptition and a cortical barrier to gas diffusion work together to maintain a low, non-inhibitory O2 concentration in the central, infected zone of the nodule. At this low O2 concentration, cytosolic leghemoglobin is required to facilitate the diffusion of O2 through the infected cell to the bacteria. The resistance of the cortical diffusion barrier is variable and is used by legume nodules to regulate the O2 concentration in the infected cells such that it limits aerobic respiration and N2 fixation at all times. The resistance of the diffusion barrier and therefore the degree of O2 limitation seems to be regulated in response to changes in the O2 concentration of the central infected zone, the supply of phloem sap to the nodule, and the rate of N assimilation into the end products of fixation.  相似文献

The linked structural genes lux A and lux B, encoding bacterial luciferase of a marine bacterium Vibrio harveyi, were fused with the nitrogenase nifD promoter from Bradyrhizobium japonicum and with the P1 promoter of pBR322. Both fusions were integrated into the B. japonicum chromosome by site-specific recombination. Soybean roots infected with the two types of rhizobium transconjugants formed nitrogen-fixing nodules that produced bright blue-green light. Cells containing the P1 promoter/lux AB fusion resulted in continuously expressed bioluminescence in both free-living rhizobium and in nodule bacteriods. However, when under control of the nifD promoter, luciferase activity was found only in introgen-fixing nodules. Light emission from bacteroids allowed us to visualize and to photograph nodules expressing this marker gene fusion in vivo at various levels of resolution, including within single, living plant cells. Localization of host cells containing nitrogen-fixing bacteroids within nodule tissue was accomplished using low-light video microscopy aided by realtime image processing techniques developed specifically to enhance extreme low-level luminescent images.  相似文献

Analysis of apoplastic solutes in the cortex of soybean nodules   总被引:3,自引:0,他引:3  
Various techniques were used to extract solutes from the free space of intact soybean [ Glycine max (L.) Merr.] nodules. A variety of solutes (carbohydrates, amino acids, organic acids, ions) was found, but the major solute obtained with all methods was allantoic acid. Most work was done with a technique involving vacuum infiltration of intact detached nodules with water. This approach provided rapid sampling of the apoplastic solutes, and the results indicated that solutes were not derived from the xylem and phloem of ruptured vascular bundles. Infiltration of intact nodules with Fast Green showed dye penetration only to the barrier in the inner cortex, indicating that infected tissues did not contribute to solute composition. Although allantoic acid was the only ureide which could be detected in solute samples, no evidence was obtained for the presence of allantoinase in the cortical apoplast. The results suggest the transport of allantoic acid by an apoplastic route in nodules or the release of allantoic acid to the cortical apoplast in response to treatments which disrupt ureide export. Calculated values for solute concentrations in the cortical apoplast were in the hundred millimolar range, suggesting that apoplastic solutes may represent a significant osmotic component in the nodule cortex.  相似文献

High-performance liquid chromatography analysis is used to examine the distribution of isoflavones in different parts of soybean sprouts. Between the seed leaf and hypocotyl, the ratio of isoflavones differs depending on the aglycone type. Glycitein exists predominantly in the hypocotyl. Two phosphofructokinases (EC 2.7.1.11; PFK) have been isolated from the host fraction of nitrogen–fixing chickpea ( Cicer arietinum L.) root nodules that were formed with Rhizobium sp. (Cicer) CC1192. The predominant enzyme, which is suggested to be cytosolic, was isolated as a large aggregate of molecular mass near 2 000 kDa and was purified to a high degree. The less abundant enzyme, suggested to be of plastid origin, had a native molecular mass of 186 kDa. Both forms of PFK displayed typical hyperbolic kinetics with MgATP and fructose-6-phosphate. The major form of PFK was strongly inhibited by phosphoenolpyruvate, 2-phosphoglycerate and 3-phos-phoglycerate, and to a lesser extent by ADP, pyrophosphate, glucose-6-phosphate and 6-phosphogluconate. KCl and phosphate were activators and relieved the effect of inhibitors. The major PFK was disaggregated in the presence of ATP into a species which had a molecular mass of approximately 550 kDa and a lower affinity for fructose-6-phosphate.  相似文献

Legume root nodules use the ascorbate-glutathione pathway to remove harmful H2O2. In the present study. effective and ineffective nodules from soybean and alfalfa were compared with regard to this pathway. Effective nodules had higher activity of all 4 enzymes (ascorbate peroxidase, EC 1. 11. 1. 11: monodehydroascorbate reductase, EC 1. 6. 5. 4: dehydroascorbate reductase, EC 1. 8. 5. 1: and glutathione reductase, EC 1. 6. 4. 2). The concentration of thiol tripeptides (primarily homoglutathione) was about 1 m M in effective nodules – a level 3–4-fold higher than in ineffective nodules. Effective nodules contained higher levels of NAD+. NADP+ and NADPH. but not of NADH or ascorbate. The increased capacity for peroxide scavenging in effective nodules as compared to ineffective nodules emphasizes the important protective role that this pathway may play in processes related to nitrogen fixation.  相似文献

The aim of this study was to examine how the pools of non-structural carbohydrates in soybean nodules are affected under water stress conditions depending on the nature of the symbiont strains with particular emphasis on the plant-borne carbohydrates sucrose and pinitol, and on trehalose, a compatible solute synthesized by the bacteroids. Soybean ( Glycine max [L.] Merr. cv. Maple Arrow) plants were inoculated with the nitrogen-fixing strains Bradyrhizobium japonicum 61-A-101 or USDA 110 spc4 and cultivated axenically under conditions in which nodules formed in an upper soil compartment while roots for water supply grew into a compartment filled with nutrient solution. When the nodules were well established (1 month post inoculation), 10% (w/v) PEG 6000 was added to the nutrient solution. This led to a slowly progressing, moderate water stress, as determined by measuring the decrease of transpiration, and to a decrease in nitrogen fixation. The pool sizes of the major non-structural nodule carbohydrates changed during progression of water stress. Sucrose, the major soluble carbohydrate in nodules of unstressed plants (2 and 4%, respectively of nodule dry weight depending on symbiont strain), strongly increased in nodules of stressed plants, reaching nearly 10% of dry weight. The activities of two major sucrose-consuming enzymes, sucrose synthase and alkaline invertase, decreased markedly in nodules of stressed plants. Starch decreased only transiently upon water stress. Pinitol, a cyclitol serving as compatible solute in many plants, increased more than 4 times, reaching about 1% of nodule dry weight during the stress. Trehalose, the major soluble carbohydrate synthesized by the bacteroids, increased in nodules colonized by USDA 110 spc4 from about 0.2 to 0.8% of nodule dry weight, while in nodules colonized by 61-A-101 it amounted to more than 1.5% of dry weight both with and without stress.  相似文献

A field experiment was conducted to determine the effects of light intensity and nitrate nutrition on soybean (Glycine max (L) Merr.) root growth and development. Relative growth rates, total, root and nodule dry weights, and the rates of increase in the number of roots indicated that nitrogen fixation limited growth relative to that achieved with nitrate and that the response to nitrate increases with light intensity and varies with plant age. Nitrate increased with rate of taproot extension but light intensity had no effect.  相似文献

Abstract. A glycoprotein which occludes intercellular spaces in the inner cortex of legume nodules may be involved in controlling oxygen diffusion into rhizobial-infected cells. Here we investigated this possibility by localizing the glycoprotein using monoclonal antibodies and immunogold labelling in nodulated roots of soybean cv. Clarke inoculated with Bradyrhizobium japonicum strain RCR3442 exposed to atmospheres with either 10, 21 (control) or 40% oxygen for 28d. Infected cells showed evidence of premature senescence when grown in above or below ambient pO2 particularly at 10% oxygen, although cortical cells appeared to be little altered by oxygen treatment. In the inner cortical cells, more glycoprotein was seen to be occluding intercellular spaces of those nodules subjected to 40% oxygen and less in those nodules exposed to 10% oxygen, when compared to controls. This observation, made at the light microscope level (using silver enhancement) was confirmed under the TEM using immunogold labelling. Therefore, it is suggested that intercellular space glycoprotein is one of the structural components of the diffusion resistance in the cortex of legume nodules.  相似文献
