Irradiation of puparia in Mediterranean fruit fly, Ceratitis capitata (Wiedemann), sterile insect release programs can negatively affect adult fly performance. Emergence, survivorship, lure attraction, and mating competition tests were performed on irradiated and unirradiated Mediterranean fruit flies in Hawaii. Unirradiated flies of the Vienna-7 (tsl) strain had higher emergence, flight ability, and survivorship compared with irradiated flies. In general, unirradiated flies were more responsive to trimedlure, but this effect was not consistent for all strains at every age. Laboratory strains, of both unirradiated and irradiated flies, responded to trimedlure at a younger age than wild flies, which may be a result of inadvertent selection for decreased development time in laboratory-reared flies. Mating competition tests with irradiated and unirradiated flies showed no significant differences. Costs associated with the irradiation process and the development of alternative control techniques are discussed.  相似文献

In greenhouse behavioural assessments conducted at Seibersdorf, Austria that allowed flies the choice of different types of potted host trees and plants, it was found that mature laboratory mass-reared Mediterranean fruit fly males, Ceratitis capitata (Wied), (Dipt. Tephritidae), strain Vienna-42 (temperature-sensitive lethal genetic sexing strain) showed the same preference for orange foliage, over other plants types and parts, as wild males. They aggregated in leks, released pheromone, and courted approaching females almost exclusively on orange foliage. Even though a number of other trees and plants were available to Vienna-42 males, less than 1% of these sexual activities occurred on the foliage or other plant parts of non-Citrus trees and not a single mating occurred there. In choice tests carried out in Chios, Greece in large field cages housing naturally planted orange trees, it was determined that mature and sterile Mediterranean fruit fly males of the same sexing strain responded to the odour emanating from the flavedo (due to superficial cuts made in the fruit peel containing the oil glands) of ripening sweet, sour oranges and lemons, in a similar intensity, order of fruit preference and time of day as did mature males of the local wild population. Males of both strains were frequently observed contacting with their mouth parts the oils exuding from the cuts made in the flavedo of the orange peel, or exhibiting homosexual activity on, or near the cuts. The results show that mass-reared, sterile males of the Vienna-42, genetic sexing strain behave in a comparable way to wild males in an important component of habitat location (odour of host foliage and host fruit) and hence are competitive with them in these respects. None of the standard international quality control tests for sterile Mediterranean fruit flies evaluates this important habitat location behaviour, which is a prerequisite for the successful encounter of the sexes in a sterile release programme.  相似文献

The high-temperature treatment of eggs of mass-reared tsl genetic sexing strains in Mediterranean fruit fly, Ceratitis capitata (Wiedemann), during late embryogenesis (the low-high protocol) conserves more male flies than treatment during early embryogenesis. A tsl strain, AUSTRIA 6-97, was constructed to follow the fate of aneuploid individuals during male-only production. Aneuploid individuals are produced following segregation in the translocation heterozygous males, and they can survive to the pupal stage where they compromise quality because they do not eclose as adults. Hatching, emergence, and male fly production were quantified and the heat-treatment protocol was characterized. The low-high egg treatment conserves the number of euploid-balanced males, and there is a very low survival of aneuploid males. After heat treatment of eggs, at least 95% of the male pupae were euploid compared with only 71% from untreated eggs. The quality of euploid male pupae was diminished with successive daily collections, an effect previously attributed to aneuploid survivors. Reduced yield of euploid males from early heat treatments was the result of an emergence effect, in addition to a maternal effect. A third detrimental effect of heat was found, occurring after hatching and before pupation, that reduces the survivorship of euploid males. The low-high treatment protocol yielded more males, with a higher accuracy than other heat treatments. However, although it avoids both the maternal and emergence effects, the production of euploid males was 30% less than the potential production, implying that the low-high heat protocol for killing female embryos in tsl genetic sexing strains can be fine-tuned.  相似文献

The effect of releases of bisexual (males and female) and unisexual (male only) sterilized medflies was compared in three large field evaluations over a 3-yr period (1995-1997) in southwestern Guatemala. The two strains tested were a genetic sexing strain, Vienna-4/Tol-94, carrying the temperature sensitive tsl gene to eliminate females in the egg stage, and the standard bisexual Petapa strain. Flies were mass-reared, sterilized by irradiation as pupae, shipped to a field center, and released by air as young adults over 2 km by 2 km core areas in the centers of separate 6 km by 6 km test plots. Strain performance was monitored weekly by trapping sterile and wild male adults in core and buffer areas and by collecting eggs from coffee berries to determine induced sterility. Results indicated a several-fold advantage for the males-only strain as measured by the level of induced sterility, especially at the very high release ratios of 100:1 recorded in 1997. During that final test year, sterile-fly release rates were increased to provide high sterile:wild (S:W) fly ratios in the field, and egg sterility reached levels in excess of 70% in plots were the male-only strain was used. However, in the plots where the bisexual strain was released, induced sterility only reached 12% despite S:W ratios above 1,000:1.  相似文献

The sterile insect technique (SIT) is used to control wild Mediterranean fruit fly introductions in California and Florida in the U.S. In the past, bait sprays containing malathion proved invaluable in treating new outbreaks or large populations before the use of SIT. Recently, a spinosad protein bait spray, GF-120, has been developed as a possible alternative to malathion, the standard insecticide in protein bait sprays. In this study, protein-deficient and protein-fed Vienna-7 (sterile, mass-reared, "male-only" strain) flies and wild males and females were evaluated to determine the effectiveness of the GF-120 protein bait containing spinosad with respect to bait attraction, feeding, and toxicology. There were no effects of diet or fly type on feeding duration in small laboratory cages. Wild flies, however, registered more feeding events than Vienna-7 males. Flies that fed longer on fresh bait died faster. Protein-deficient flies were more active and found the bait more often than protein-fed flies. Data suggest that adding protein to the diet of SIT flies may decrease their response to baits, therefore, reduce mortality, and thus, allow the concurrent use of SIT and bait sprays in a management or eradication program.  相似文献

To restore male mating competitiveness of Mexican fruit flies, Anastrepha ludens (Loew) (Diptera: Tephritidae), reared for sterile insect releases by the Mexican Fruit fly Eradication Campaign, two strain replacement techniques were evaluated. Field cage male competitiveness tests revealed that laboratory males of the Metapa strain mated 3 times less often with wild females than field-collected wild males. A strain developed from the cross of wild males and laboratory females (hybrid strain) was similar to a strain developed from the cross of laboratory males and females (laboratory strain) in that its females produced similar amounts of eggs and the eggs displayed similar levels of hatch and egg-to-pupa transformation in artificial diet. By contrast, a strain developed from the cross of wild males and females (wild strain), forced into artificial rearing, experienced a series of bottlenecks involving reduced egg laying and extremely poor development in diet. The male F1 progeny of the hybrid strain and field-collected wild males outcompeted Fl laboratory males in field cage tests for matings with field-collected wild females. In conclusion, we found that strains developed from the cross of wild males and laboratory females allowed us to restore male mating competitiveness of F1 Mexican fruit flies without compromising mass-rearing production.  相似文献

The recent development of medfly genetic sexing strains (GSS) enables only male adults to be produced and released for area-wide control using the sterile insect technique (SIT). Before these strains can be incorporated into large operational field programmes, information is required on all aspects of field performance.
In a pilot project in Tunisia, medfly pupae from GSS SEIB-40 [based on a pupal colour mutation ( wp ) and VIENNA-43/44 [based on a temperature sensitive lethal mutation ( tsl ) were shipped from the FAO/IAEA Agriculture and Biotechnology laboratory (Seibersdorf, Austria) and the emerged sterile males released weekly from the ground in oases of the Tozeur Governorate from mid-February until October 1994. During that period, flies emerging from an average of 5000 pupae per hactare were released weekly throughout 105 hectares. Transportation problems were encountered which resulted in damage to the pupae. Nevertheless, the monitoring of the wild population through a trapping network using Jackson and McPhail traps showed that both strains performed equally well inducing a three- to five-fold decrease of the wild population compared with the control oasis (10 wild flies per Jackson trap per day in October in Ain-El-Karma, five in Tamerza and 30 in Bir Kastilia control oasis). Considering the economical and technical benefits inherent in the use of these new strains in the field and as a result of their good field performance, similar strains have been introduced into mass-rearing facilities in Guatemala (1994), Argentina (1995) and Madeira (1996).  相似文献

The sterile insect technique (SIT), when used for the control of the Mediterranean fruit fly (medfly), Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), generally relies on the release of sterile flies of only the male sex. Male selection is achieved through the use of a genetic sexing strain (GSS) in which females are killed by heat treatment in the generation prior to release. Transgenic sexing strains (TSS) have been developed that perform the same function of female-lethality, this time by withholding tetracycline (or related compounds) from the larval diet. The use of TSS may allow for certain problems associated with conventional GSS, such as strain instability and reduced productivity in mass-rearing, to be avoided. The performance, and principally the sexual competitiveness, of released male flies is important for the success of an SIT control programme. This study describes field cage experiments in which the competitiveness of males from a TSS (OX3376B) was compared with that of a conventional GSS (VIENNA-8) and two wild-type strains (TOLIMAN and ARG). When competing for female mates with wild-type males, OX3376B male performance was acceptable. When OX3376B males competed directly for mates with VIENNA-8 males, VIENNA-8 slightly outperformed the TSS males. Parallel tests, in which wild-type males competed with either OX3376B or VIENNA-8 males, showed that males from both sexing strains were highly competitive with wild-type males. These results suggest that OX3376B in particular, and TSS in general, show sufficiently good mating competitiveness to merit further research into their suitability for eventual use in SIT programmes.  相似文献

Dopamine (DA) content, tyrosine decarboxylase (TDC) activity and survival were studied under normal and environmental stress conditions in the ste and e strains carrying ebony mutation increasing DA level and the octopamineless strain Tbetah(nM18) of Drosophila melanogaster. Wild-type strains Canton S and Oregon R, and strain p845 from which Tbetah(nM18) strain was derived were used as controls. Sexual dimorphism of TDC activity, DA content, and survival in flies of all D. melanogaster strains under study was found. Tbetah(nM18) mutation sharply reduced TDC activity in females, while ebony had no such effect. DA content and survival under heat stress in Tbetah(nM18) flies did not differ from those in the wild type. ste and e flies had drastically increased DA content under normal conditions, dramatically decreased survival under heat stress, but increased survival under starvation. DA content and survival under heat stress were also studied in the reciprocal hybrids (males) F(1) of the cross D. virilis strains 101 (wild type) and 147 with X-linked mutation, which significantly increases DA content. 147x101 males had a considerably higher DA content and lower survival than 101x147 ones. Individuals of all D. melanogaster strains under study developed the stress reaction, as judged by changes in TDC activity and DA levels. To examine possible effects of space radiation on living organism, we have analyzedtwo types of mutations, sex-linked recessive lethal mutations and somatic mutations, in fruit fly of the species Drosophila melanogaster. Drosophila strains used were wild type strains and a radiation-sensitive strain mei-41. Two different developmental stages of samples were sent into space; young adult males to analyze sex-linked recessive lethal mutations and about 30hr-old larvae to detect somatic mutations in wing epidermal cells. For wild type and mei-41 strains each, about 200 adult male flies and about 6,000 larvae were loaded on space shuttle Endeavour. The male flies returned from space were mated to virgin female flies of a tester strain, and the presence of the lethal mutations was analyzed at F2 generation. The frequencies of sex-linked recessive lethal mutations in flight groups were 2 and 3 times higher for wild type Canton-S and mei-4 1, respectively, than those in ground control groups. Most larvae sent to space emerged as adult flies within about 10 days after the landing. The presence of wing-hair somatic mutations, which give morphological change in hairs growing on the surface of wing epidermal cells, was analyzed under microscope. In wild type strain Muller-5, the frequency of wing hair mutant spots in flight group was about 1.5-fold higher than that in ground control, and in Canton-S-derived wild type strain the frequencies were similar between the two groups. By contrast, for mei-41 strain the mutation frequency was lower in flight group than in control group. The observed higher frequency of lethal mutations in the flight group might be due to a possibility that radiation effects on reproductive cells could be greatly enhanced under micro gravity. However, if this would be the case, we do not have appropriate explanation for the apparent absence of such synergistic effects on somatic wing-hair mutation system.  相似文献

The presence of males was shown to affect the rate of female ovarian development in the Caribbean fruit fly, Anastrepha suspensa (Loew). Virgin females were maintained either in the absence or presence of males. Those sharing a common space with males had either visual contact with the opposite sex or no visual contact. Three strains of Caribbean fruit fly were tested: mass reared strain (flies in colony for more than 20 years); semi-wild strain, flies recently adapted to the laboratory conditions (ca. 12 months); and a wild strain collected in the field. We found that: (1) Mass reared, semi-wild, and wild strains had different female maturation rates, as measured by the presence of mature oocytes, regardless of male presence. (2) Male presence accelerated maturation in wild females and to a lesser extent in semi-wild flies, but had no effect on the long-domesticated mass reared strain. (3) A barrier between female and male cages removed any possibility of visual communication but had no effect on the males’ effect on female maturation. We discuss the adaptive significance of facultative ovarian maturation and the use of male-produced cues to regulate sexual development, and comment on the rapid rate of selection on female maturation under mass-rearing conditions.  相似文献

Mating success was examined in groups of Drosophila melanogaster carrying mutation flamMS (SS, MSn1-2, and MSn1-3) and in wild-type flies. The proportion of normally copulating males was significantly lower in the mutant strains. The reduction in mating efficiency was caused by changes in male behavior rather than in female attractiveness. Individual analyses showed that male mating behavior in strains carrying flamMS was qualitatively and quantitatively different from that in the wild-type strain Canton S. The proportion of males that performed consecutive courtship stages was significantly lower in the mutant strains. The sequence and duration of some courtship stages (in particular, orientation and wing vibration) in mutant flies was shown to be altered. The significance of the flamenco locus in regulation of processes occurring at the organismal level are discussed.  相似文献

The Mediterranean fruit fly (medfly), Ceratitis capitata (Wiedemann), is a key pest of citrus in Spain because of significant yield losses and to quarantine restrictions. Biologically based control methods, such as the Sterile Insect Technique (SIT), which relies on the sterilization by irradiation of large numbers of insects, is gaining an increasing role in the control of medfly in Mediterranean areas. However, gamma-irradiation might damage the midgut epithelium cells, causing a lowering of nutritive assimilation that can negatively affect adult performance. Irradiation effects on digestive physiology are well established for a number of insect pests, but there is no information on medfly. Our aim was to determine the effects of gamma-irradiation on C. capitata digestive protease activity. Both larvae and adults were found to use a similar proteolytic system based on aspartyl-, trypsin-, chymotrypsin-, amino peptidase-, and carboxypeptidase A- and B-like activities. Pupae of the Vienna-7 (tsl) strain were irradiated at 70 or 140 Gy, two days before emergence, and the adults fed during 5 days on sugar-protein (4:1) diets. Protease activity was measured in midgut extracts and compared with males non-irradiated reared in the same conditions. The results showed that the irradiation doses tested had no effect on the digestive proteolytic activities of medfly adults. Moreover, the longevity of irradiated medflies at the highest dose (140 Gy) was similar to that of controls.  相似文献

Recent laboratory studies of mass‐reared flies in small cages have found that periods of just 24‐ or 48‐h access to yeast hydrolysate can substantially enhance mating performance of mass‐reared male Queensland fruit flies, Bactrocera tryoni (Froggatt) (Diptera: Tephritidae) (‘Q‐flies’). Using field cage tests that provide a better approximation of nature, we here investigated whether access to yeast hydrolysate for 48 h after adult emergence improves the ability of male and female mass‐reared, sterile Q‐flies to compete sexually with wild‐type flies that had been provided continuous access to yeast hydrolysate. Mating probability of sterile males was significantly increased by 48‐h access to yeast hydrolysate; sterile males provided 48‐h access to yeast hydrolysate had mating probability similar to that of wild males provided continuous access to yeast hydrolysate, whereas sterile males deprived of access to yeast hydrolysate had much lower mating probability. Unlike males, access to yeast hydrolysate for 48 h did not increase mating probability of sterile female Q‐flies. We instead found that wild females provided continuous access to yeast hydrolysate had higher mating probability than sterile females that did or did not have 48‐h access to yeast hydrolysate. This result raises the possibility that a bisexual Q‐fly strain might operate essentially as a male‐only release when the flies are given access to yeast hydrolysate during a 48‐h pre‐release holding period. Sterile males given access to yeast hydrolysate for 48 h mated significantly earlier in the evening than wild males and, as in other recent studies, this tendency was associated with an increased tendency to mate on the trees rather than the cage walls. There was no evidence of sexual isolation in this study, as wild and sterile mass‐reared flies showed no evidence of preferential mating with their own kind. Further studies are now needed to assess the potential for pre‐release access to yeast hydrolysate to improve sexual performance and longevity of sterile, mass‐reared, Q‐flies in the field.  相似文献

To study the effect of mutagenic/carcinogenic agents on P-element transposition, the P strains used should be defined, especially with respect to the number of intact and functional P elements present. In this investigation, the relation between the number of complete P elements present in dysgenic males and P-insertion mutagenesis was studied in several MR (P) strains. The main conclusions from this investigation are: (1) Complete P elements can be present in the genome without genetic activity (even in a 'dysgenic' cross). As a consequence, the number of complete P elements present in particular dysgenic flies, is not necessarily an indication of their dysgenic genetic activity. (2) The MR-h12/Cy strain carries two complete P elements, one on the X chromosome without and one on the MR chromosome with genetic activity (making this strain most suitable for studies on P-transposition mechanisms).  相似文献

Mutants defective in chemolithoautotrophic growth (Aut-) have been isolated from Alcaligenes eutrophus strains H16, N9A, G27, and TF93. Spontaneous Aut- mutants were obtained only with strain TF93. Mutants of the other strains were selected after conventional mutagenesis or treatment with mitomycin. Most of the mutants, including the spontaneous Aut- strains, lacked hydrogenase activity (Hox-) but possessed the ability to fix carbon dioxide (Cfx+). Agar mating of A. eutrophus H16 with Hox- mutants of the various strains resulted in transconjugants which had recovered the ability to grow autotrophically and to express activity of hydrogenase as examined by enzymatic and immunochemical analysis. Transfer of hydrogen-oxidizing ability occurred in the absence of a mobilizing plasmid such as Rp4. The transfer frequency was particularly high (ca. 10(-2) per donor) when the spontaneous Hox- mutants of strain TF93 were used as recipients. These strains proved to be plasmid free, whereas donors, transconjugants, and the mutagen-treated Hox- mutants contained a large plasmid (molecular weight, 270 +/- 10 X 10(6) revealed by agarose gel electrophoresis. The results allow the conclusion that A. eutrophus H16 harbors a self-transmissible plasmid designated pHG1, which carries information for hydrogen-oxidizing ability.  相似文献

Summary Host cell reactivation and UV reactivation and mutagenesis of UV-irradiated phage were measured in tsl recA + and tsl recA host mutants. Host cell reactivation was slightly more efficient in the tsl recA strain compared to the tsl + recA strain. Phage was UV-reactivated in the tsl recA strain with about one-half the efficiency of that in the wild type strain, but there was no corresponding mutagenesis of phage. UV-reactivation was also slightly lower and mutagenesis several-fold lower than normal in the tsl recA + strain. To account for these observations, we propose that there is an inducible, error-free pathway of DNA repair in E. coli that competes with error-prone repair for repair of phage lesions.  相似文献

The house fly, Musca domestica L., is an important ectoparasite with the ability to develop resistance to insecticides used for their control. Thiamethoxam, a neonicotinoid, is a relatively new insecticide and effectively used against house flies with a few reports of resistance around the globe. To understand the status of resistance to thiamethoxam, eight adult house fly strains were evaluated under laboratory conditions. In addition, to assess the risks of resistance development, cross-resistance potential and possible biochemical mechanisms, a field strain of house flies was selected with thiamethoxam in the laboratory. The results revealed that the field strains showed varying level of resistance to thiamethoxam with resistance ratios (RR) at LC50 ranged from 7.66-20.13 folds. Continuous selection of the field strain (Thia-SEL) for five generations increased the RR from initial 7.66 fold to 33.59 fold. However, resistance declined significantly when the Thia-SEL strain reared for the next five generations without exposure to thiamethoxam. Compared to the laboratory susceptible reference strain (Lab-susceptible), the Thia-SEL strain showed cross-resistance to imidacloprid. Synergism tests revealed that S,S,S-tributylphosphorotrithioate (DEF) and piperonyl butoxide (PBO) produced synergism of thiamethoxam effects in the Thia-SEL strain (2.94 and 5.00 fold, respectively). In addition, biochemical analyses revealed that the activities of carboxylesterase (CarE) and mixed function oxidase (MFO) in the Thia-SEL strain were significantly higher than the Lab-susceptible strain. It seems that metabolic detoxification by CarE and MFO was a major mechanism for thiamethoxam resistance in the Thia-SEL strain of house flies. The results could be helpful in the future to develop an improved control strategy against house flies.  相似文献

Wild strain, mated, female Mexican fruit flies, Anastrepha ludens (Loew), with no prior experience with fruit (naive), were not attracted to grapefruit, a preferred cultivated host, in wind tunnel experiments. Naive, mated laboratory strain females were attracted. Prior experience with grapefruit increased attraction of both laboratory and wild strains. More females were attracted to fruit with peel damage than to undamaged fruit, indicating that fruit odor mediated attraction. More naive than experienced females attempted to oviposit on the sides of the wind tunnel. Experienced laboratory males, but not wild males or naive males, were attracted to grapefruit. Attraction and oviposition behavior of laboratory flies was greater than that of wild flies.  相似文献

In Escherichia coli, the cell division block observed during the SOS response requires the product of the sfiA gene, whose expression is regulated negatively by the LexA repressor and positively by the RecA protease. We have studied the effect on sfiA expression of sfiA, sfiB, infA, and infB mutations, which are known to affect SOS-associated filamentation. To measure sfiA expression in the different strains, we first constructed a lambda transducing phage carrying an sfiA::lac operon fusion. Mutations at the sfiA locus (dominant and recessive) and the sfiB locus (recessive) had no effect on sfiA expression. The mutations tif (at the recA locus) and tsl (at the lexA locus) are known to induce filamentation and a high level of sfiA expression at 42 degrees C. The infB1 mutation, which suppresses filamentation in a tif tsl strain at 42 degrees C, reduced sfiA expression at 42 degrees C in tif tsl infB1 and tsl infB1 strains but not in a tif infB1 strain. The infA3 mutation, which suppresses tif-mediated filamentation, reduced induction of sfiA expression in a tif infA3 strain at 42 degrees C or after UV irradiation. The isolation and characterization of sfiA constitutive strains revealed only lexA-linked mutations in a sfiA-background, suggesting that LexA is the only readily eliminated repressor of the sfiA gene. Nevertheless, the infA and infB mutations could define elements involved in the regulation of sfiA expression.  相似文献
