The growth of the freshwater microalga Scenedesmus obliquus was studied at 30°C in a mineral culture medium with phosphorus concentrations of between 0 and 372 μ . The values for the specific growth rates, between and , fitted a semistructured substrate-limitation model with μm1 = 0·0466 h−1, μm2 = 0·0256 h−1 and . The specific uptake rate of phosphorus reached a maximum value of qSm1 = 658·01 × 10−4 μmol P mg−1 biomass h−1.  相似文献

Recently, we demonstrated that angiotensin-(1–7) (Ang-(1–7)) stimulates the Na+-ATPase activity through a losartan-sensitive angiotensin receptor, whereas bradykinin inhibits the enzyme activity through the B2 receptor [Regul. Pept. 91 (2000) 45; Pharmacol. Rev. 32 (1980) 1]. In the present paper, the effect of bradykinin (BK) on Ang-(1–7)-stimulated Na+-ATPase activity was evaluated. Preincubation of Na+-ATPase with 10−9 M Ang-(1–7) increases enzyme activity from 7.9±0.9 to 14.1±1.5 nmol Pi mg−1 min−1, corresponding to an increase of 79% (p<0.05). This effect is reverted by bradykinin in a dose-dependent manner (10−14–10−8 M), reaching maximal inhibitory effect at 10−9 M. Des-Arg9 bradykinin (DABK), an agonist of B1 receptor, at the concentrations of 10−9–10−7 M, does not mimic the BK inhibitory effect, and des-Arg9-[Leu8]-BK (DALBK), a B1 receptor antagonist, at the concentrations of 10−10–10−7 M, does not prevent the inhibitory effect of BK on Ang-(1–7)-stimulated enzyme. On the other hand, HOE 140, an antagonist of B2 receptor, abolishes the inhibitory effect of BK on the Ang-(1–7)-stimulated enzyme in a dose-dependent manner, reaching maximal effect at 10−7 M. Taken together, these data indicate that stimulation of B2 receptors by BK can counteract the stimulatory effect of Ang-(1–7) on the proximal tubule Na+-ATPase activity.  相似文献

It is well recognized that estradiol (E2) is one of the most important hormones supporting the growth and evolution of breast cancer. Consequently, to block this hormone before it enters the cancer cell or in the cell itself, has been one of the main targets in recent years. In the present study we explored the effect of the progestin, nomegestrol acetate, on the estrone sulfatase and 17β-hydroxy-steroid dehydrogenase (17β-HSD) activities of MCF-7 and T-47D human breast cancer cells. Using physiological doses of estrone sulfate (E1S: 5 × 10−9 M), nomegestrol acetate blocked very significantly the conversion of E1S to E2. In the MCF-7 cells, using concentrations of 5 × 10−6 M and 5 × 10−5 M of nomegestrol acetate, the decrease of E1S to E2 was, respectively, −43% and −77%. The values were, respectively, −60% and −71% for the T-47D cells. Using E1S at 2 × 10−6 M and nomegestrol acetate at 10−5 M, a direct inhibitory effect on the enzyme of −36% and −18% was obtained with the cell homogenate of the MCF-7 and T-47D cells, respectively. In another series of studies, it was observed that after 24 h incubation of a physiological concentration of estrone (E1: 5 × 10−9 M) this estrogen is converted in a great proportion to E2. Nomegestrol acetate inhibits this transformation by −35% and −85% at 5 × 10−7 M and 5 × 10−5 M, respectively in T-47D cells; whereas in the MCF-7 cells the inhibitory effect is only significant, −48%, at 5 × 10−5 M concentration of nomegestrol acetate. It is concluded that nomegestrol acetate in the hormone-dependent MCF-7 and T-47D breast cancer cells significantly inhibits the estrone sulfatase and 17β-HSD activities which converts E1S to the biologically active estrogen estradiol. This inhibition provoked by this progestin on the enzymes involved in the biosynthesis of E2 can open new clinical possibilities in breast cancer therapy.  相似文献

Conscious rats with chronic gastric fistula were trained for drinking a 14-ml milk meal. The activity of an intestinal hormone, oxyntomodulin (OXM), was studied in this model and compared to that observed when histamine was the stimulus. Under histamine (0.25 mg·kg−1·h−1) stimulation, OXM at doses (60–120 pmol·kg−1·h−1) that induced physiological circulating levels inhibited gastric acid secretion up to 50%. Under meal stimulation, OXM reduced up to 29% acid secretion at doses (1–1.5 nmol·kg−1·h−1) inducing supraphysiological levels. We conclude that at physiological concentrations OXM cannot counteract the complex processes triggered by a meal. OXM would be a component of enterogastrone, a combination of several intestinal hormones acting in synergy. The OXM action is related to pathways recognizing the C-terminal 19–37 moiety of the molecule.  相似文献

The rat pineal gland is known to release melatonin in response to noradrenergic stimulation. Since vasopressin (VP)- and oxytocin (OT)-containing fibers innervate the pineal gland, the effects of VP and OT on melatonin release from perifused rat pineal glands were investigated. VP (10−7 M) and OT (10−6 M) decreased the basal melatonin secretion. No dose-dependent effect was observed. At high concentrations (10−5) these peptides potentiated the isoproterenol-induced increase of melatonin secretion. Below 10−5 M no potentiation was observed. Fragments of VP {[pGlu4,Cys6]VP(4–9)} and OT {[pGlu4,Cys6]OT(4–9)} did not display any effect on the isoproterenol-induced melatonin secretion.  相似文献

The phosphinoalkenes Ph2P(CH2)nCH=CH2 (n= 1, 2, 3) and phosphinoalkynes Ph2P(CH2)n C≡CR (R = H, N = 2, 3; R = CH3, N = 1) have been prepared and reacted with the dirhodium complex (η−C5H5)2Rh2(μ−CO) (μ−η2−CF3C2CF3). Six new complexes of the type (ν−C5H5)2(Rh2(CO) (μ−η11−CF3C2CF3)L, where L is a P-coordinated phosphinoalkene, or phosphinoalkyne have been isolated and fully characterized; the carbonyl and phosphine ligands are predominantly trans on the Rh---Rh bond, but there is spectroscopic evidence that a small amount of the cis-isomer is formed also. Treatment of the dirhodium-phosphinoalkene complexes with (η−CH3C5H4)Mn(CO)2thf resulted in coordination of the manganese to the alkene function. The Rh2---Mn complex [(η−C5H5)2Rh2(CO) (μ−η11−CF3C2CF3) {Ph2P(CH2)3CH=CH2} (η−CH3C5H4)Mn(CO)2] was fully characterized. Simi treatment of the dirhodium-phosphinoalkyne complexes with Co2(CO)8 resulted in the coordination of Co2(CO)6 to the alkyne function. The Rh2---Co2 complex [(η−C5H5)2Rh2(CO) (μ−η11−CF3C2CF3) {Ph2PCH2C≡CCH3}Co2(CO)2], C37H25Co2F6O7PRh2, was fully characteriz spectroscopically, and the molecular structure of this complex was determined by a single crystal X-ray diffraction study. It is triclinic, space group (Ci1, No. 2) with a = 18.454(6), B = 11.418(3), C = 10.124(3) Å, = 112.16(2), β = 102.34(3), γ = 91.62(3)°, Z = 2. Conventional R on |F| was 0.052 fo observed (I > 3σ(I)) reflections. The Rh2 and Co2 parts of the molecule are distinct, the carbonyl and phosphine are mutually trans on the Rh---Rh bond, and the orientations of the alkynes are parallel for Rh2 and perpendicular for Co2. Attempts to induce Rh2Co2 cluster formation were unsuccessful.  相似文献

The rate of electron transfer between reduced cytochrome ƒ and plastocyanin (both purified from parsley) has been measured as k = 3.6 · 107 M−1 · s−1, at 298 °K and pH 7.0, with activation parameters ΔH = 44 kJ · mole−1 and ΔS = +46 J · mole−1 · °K−1. Replacement of cytochrome ƒ with red algal cytochrome c-553, Pseudomonas cytochrome c-551 and mammalian cytochrome c gave rates at least 30 times slower: k = 5 · 105, 7.5 · 105 and 1.0 · 106 M−1 · s−1, respectively.

Similar measurements made with azurin instead of plastocyanin gave k = 6 · 106 and approx. 2 · 107 M−1 · s−1 for reaction of reduced azurin with cytochrome ƒ and algal cytochrome respectively.

Rate constants of 115 and 80 M−1 · s−1 were found for reduction of plastocyanin by ascorbate and hydroquinone at 298 °K and pH 7.0. The rate constants for the oxidation of plastocyanin, cytochrome ƒ, Pseudomonas cytochrome c-551 and red algal cytochrome c-553 by ferricyanide were found to be between 3 · 104 and 8 · 104 M−1 · s−1.

The results are discussed in relation to photosynthetic electron transport.  相似文献

Bacterivory by the rotifer Brachionus plicatilis Müller, nauplii and copepodites of the copepods Centropages Krøyer sp. and Acartia tonsa Dana, and the tintinnid Favella panamensis Kofoid & Campbell was examined using fluorescently labelled bacteria (FLB) and epifluorescence microscopy. FLB were < 1 μm in diameter, and were offered at environmental concentrations (1.47−9.08 × 106 cells·ml−1). FLB were visible within rotifers, nauplii, copepodites, and tintinnids, confirming ingestion. Rotifer clearance rates (32–418 μl·animal−1·h−1) exhibited no relation with FLB concentration. In some cases rates of clearance of FLB by rotifers were different with alternative phytoplankton food (Nanochloris Naumann sp.) than in replicates with FLB alone, whereas in other cases presence of alternative food exhibited no clear effects on rates of ingestion of FLB. Clearance rates for all six naupliar stages of A. tonsa nauplii (0–320 μl·animal−1·h−1) were stage-related, with higher rates by NIII-VI nauplii than NI-II nauplii. Nauplii had higher rates of clearance of FLB in the absence of alternative phytoplankton food (Isochrysis Parke sp.). Clearance rates of FLB by a single stage of Centropages sp. nauplii, A. tonsa CI copepodites and F. panamensis (each obtained at only a single food concentration of either 1.5 or 5.0 × 106 cells·ml−1) were within the range of 85–142 μl·animal−1·h−1. These ranges were similar to those of rotifers and A. tonsa nauplii. This is the first report of FLB ingestion by metazoan marine microzooplankton. Although rotifers and ciliates might be expected to ingest small particles such as FLB using ciliary induced feeding currents, the means by which nauplii and copepodites eat FLB is less clear. We propose that they may “eat” bacteria as they “drink” to osmoregulate.  相似文献

Oxygenation of [CuII(fla)(idpa)]ClO4 (fla=flavonolate; IDPA=3,3′-iminobis(N,N-dimethylpropylamine)) in dimethylformamide gives [CuII(idpa)(O-bs)]ClO4 (O-bs=O-benzoylsalicylate) and CO. The oxygenolysis of [CuII(fla)(idpa)]ClO4 in DMF was followed by electronic spectroscopy and the rate law −d[{CuII(fla)(idpa)}ClO4]/dt=kobs[{CuII(fla)(idpa)}ClO4][O2] was obtained. The rate constant, activation enthalpy and entropy at 373 K are kobs=6.13±0.16×10−3 M−1 s−1, ΔH=64±5 kJ mol−1, ΔS=−120±13 J mol−1 K−1, respectively. The reaction fits a Hammett linear free energy relationship and a higher electron density on copper gives faster oxygenation rates. The complex [CuII(fla)(idpa)]ClO4 has also been found to be a selective catalyst for the oxygenation of flavonol to the corresponding O-benzoylsalicylic acid and CO. The kinetics of the oxygenolysis in DMF was followed by electronic spectroscopy and the following rate law was obtained: −d[flaH]/dt=kobs[{CuII(fla)(idpa)}ClO4][O2]. The rate constant, activation enthalpy and entropy at 403 K are kobs=4.22±0.15×10−2 M−1 s−1, ΔH=71±6 kJ mol−1, ΔS=−97±15 J mol−1 K−1, respectively.  相似文献

The kinetics of substitution reactions of [η-CpFe(CO)3]PF6 with PPh3 in the presence of R-PyOs have been studied. For all the R-PyOs (R = 4-OMe, 4-Me, 3,4-(CH)4, 4-Ph, 3-Me, 2,3-(CH)4, 2,6-Me2, 2-Me), the reactions yeild the same product [η5-CpFe(CO)2PPh3]PF6, according to a second-order rate law that is first order in concentrations of [η5-CpFe(CO)3]PF6 and of R-PyO but zero order in PPh3 concentration. These results, along with the dependence of the reaction rate on the nature of R-PyO, are consistent with an associative mechanism. Activation parameters further support the bimmolecular nature of the reactions: ΔH = 13.4 ± 0.4 kcal mol−1, ΔS = −19.1 ± 1.3 cal k−1 mol−1 for 4-PhPyO; ΔH = 12.3 ± 0.3 kcal mol−1, ΔS = 24.7 ±1.0 cal K−1 mol−1 for 2-MePyO. For the various substituted pyridine N-oxides studied in this paper, the rates of reaction increase with the increasing electron-donating abilities of the substituents on the pyridine ring or N-oxide basicities, but decrease with increasing 17O chemical shifts of the N-oxides. Electronic and steric factors contributing to the reactivity of pyridine N-oxides have been quantitatively assessed.  相似文献

Palythoa psammophilia Walsh & Bowers has a well coordinated, stereotyped feeding response, the culminating step of which is ingestion; this may be elicited by the synergistic effect of the tripeptide glutathione and the -imino acid, proline. Either activator acting separately causes responses only at high concentrations (above 10−5 M for glutathione; above 10−4 M for proline) in a reduced number of animals and at a low rate (5.00 ± 1.73 min in 5 × 10−3 M solutions of glutathione; 11.10±3.74 min in 5 × 10−3 M solutions of proline). Highest percentages of response were obtained in combinations where glutathione was at a concentration of 5 × 10−3 M and proline at 5 × 10−4 M or in combinations of glutathione at concentrations 5 × 10−6 M and proline at 5 × 10−5 M. The speed of ingestion is considerably enhanced when these activators are combined (1.17±1.18 min).  相似文献

In the present paper, the modulation of the basolateral membrane (BLM) Na+-ATPase activity of inner cortex from pig kidney by angiotensin II (Ang II) and angiotensin-(1–7) (Ang-(1–7)) was evaluated. Ang II and Ang-(1–7) inhibit the Na+-ATPase activity in a dose-dependent manner (from 10−11 to 10−5 M), with maximal effect obtained at 10−7 M for both peptides. Pharmacological evidences demonstrate that the inhibitory effects of Ang II and Ang-(1–7) are mediated by AT2 receptor: The effect of both polypeptides is completely reversed by 10−8 M PD 123319, a selective AT2 receptor antagonist, but is not affected by either (10−12–10−5 M) losartan or (10−10–10−7 M) A779, selective antagonists for AT1 and AT(1–7) receptors, respectively. The following results suggest that a PTX-insensitive, cholera toxin (CTX)-sensitive G protein/adenosine 3′,5′-cyclic monophosphate (cAMP)/PKA pathway is involved in this process: (1) the inhibitory effect of both peptides is completely reversed by 10−9 M guanosine 5′-O-(2-thiodiphosphate) (GDPβS; an inhibitor of the G protein activity), and mimicked by 10−10 M guanosine 5′-O-(3-thiotriphosphate) (GTPγS; an activator of the G protein activity); (2) the effects of both peptides are mimicked by CTX but are not affected by PTX; (3) Western blot analysis reveals the presence of the Gs protein in the isolated basolateral membrane fraction; (4) (10−10–10−6 M) cAMP has a similar and non-additive effect to Ang II and Ang-(1–7); (5) PKA inhibitory peptide abolishes the effects of Ang II and Ang-(1–7); and (6) both angiotensins stimulate PKA activity.  相似文献

1. Rate constants for reduction of paraquat ion (1,1′-dimethyl-4,4′-bipyridy-lium, PQ2+) to paraquat radical (PQ+·) by eaq and CO2· have been measured by pulse radiolysis. Reduction by eaq is diffusion controlled (k = 8.4·1010 M−1·s−1) and reduction by CO2· is also very fast k = 1.5·1010 M−1·s−1).

2. The reaction of paraquat radical with oxygen has been analysed to give rate constants of 7.7·108 M−1·s−1 and 6.5·108 M−1·s−1 for the reactions of paraquat radical with O2 and O2·, respectively. The similarity in these rate constants is in marked contrast to the difference in redox potentials of O2 and O2· (− 0.59 V and + 1.12 V, respectively).

3. These rate constants, together with that for the self-reaction of O2·, have been used to calculate the steady-state concentration of O2· under conditions thought to apply at the site of reduction of paraquat in the plant cell. On the basis of these calculations the decay of O2· appears to be governed almost entirely by its self-reaction, and the concentration 5 μm away from the thylakoid is still 90% of that at the thylakoid itself. Thus, O2· persists long enough to diffuse as far as the chloroplast envelope and tonoplast, which are the first structures to be damaged by paraquat treatment. O2· is therefore sufficiently long-lived to be a candidate for the phytotoxic product formed by paraquat in plants.  相似文献

Abstraction of chloride from the Pd complex {[η3-2,6-(tBu2PCH2)2C6H3)]PdCl with AgBF4 in THF gives {[η3-2,6-(tBu2PCH2)2C6H3)]Pd(THF)}+BF4 −. Attemped crystallization of this THF complex produced the aqua complex {[η3-2,6-(tBu2PCH2)2C6H3)]Pd(OH2)}+BF4 −. Crystal structures of two crystalline forms of this compound are reported. In {[η3-2,6-(tBu2PCH2)2C6H3)]Pd(OH2)}+BF4 −·THF, one hydrogen of the water is hydrogen bonded to the oxygen of the THF, and the other hydrogen is hydrogen bonded to an F of the BF4 − anion. Another crystalline form has no THF, but has both of the hydrogens of water hydrogen bonded to different BF4 − anions, such that two different BF4 − anions bridge two {[η3-2,6-(tBu2PCH2)2C6H3)]Pd(OH2)}+ cations. A crystal structure is also reported for the palladium chloride complex [η3-2,6-(tBu2PCH2)2C6H3)]PdCl.  相似文献

Genistein is an isoflavone and phytoestrogen that is a potent inhibitor of cell proliferation and angiogenesis. This study was designed to investigate the binding of genistein to human serum albumin (HSA) under physiological conditions with drug concentrations in the range of 6.7 × 10−6 to 2.0 × 10−5 mol L−1 and HSA concentration at 1.5 × 10−6 mol L−1. Fluorescence quenching methods in combination with Fourier transform infrared (FT-IR) spectroscopy and circular dichroism (CD) spectroscopy was used to determine the binding mode, the binding constant and the protein structure changes in the presence of genistein in aqueous solution. Changes in the CD spectra and FT-IR spectra were observed upon ligand binding, and the degree of tryptophan fluorescence quenching change did significantly in the complexes. These data have proved the change in protein secondary structure accompanying ligand binding. The change in tryptophan fluorescence intensity was used to determine the binding constants. The thermodynamic parameters, the enthalpy change (ΔH) and the entropy change (ΔS) were calculated to be −22.24 kJ mol−1and 19.60 J mol−1 K−1 according to the van’t Hoff equation, which indicated that hydrophobic and electrostatic interactions play the main role in the binding of genistein to HSA.  相似文献

The reaction of peroxynitrous acid with monohydroascorbate, over the concentration range of 250 μM to 50 mM of monohydroascorbate at pH 5.8 and at 25°C, was reinvestigated and the rate constant of the reaction found to be much higher than reported earlier (Bartlett, D.; Church, D. F.; Bounds, P. L.; Koppenol, W. H. The kinetics of oxidation of L-ascorbic acid by peroxynitrite. Free Radic. Biol. Med. 18:85–92; 1995; Squadrito, G. L.; Jin, X.; Pryor, W. A. Stopped-flow kinetics of the reaction of ascorbic acid with peroxynitrite. Arch. Biochem. Biophys. 322:53–59; 1995). The new rate constants at pH 5.8 are k1 = 1 × 106 M−1 s−1 and k−1 = 500 s−1 for 25°C and k1 = 1.5 × 106 M−1 s−1 and k−1 = 1 × 103 s−1 for 37°C. These values indicate that even at low monohydroascorbate concentrations most of peroxynitrous acid forms an adduct with this antioxidant. The mechanism of the reaction involves formation of an intermediate, which decays to a second intermediate with an absorption maximum at 345 nm. At low monohydroascorbate concentrations, the second intermediate decays to nitrate and monohydroascorbate, while at monohydroascorbate concentrations greater than 4 mM, this second intermediate reacts with a second monohydroascorbate to form nitrite, dehydroascorbate, and monohydroascorbate. EPR experiments indicate that the yield of the ascorbyl radical is 0.24% relative to the initial peroxynitrous acid concentration, and that this small amount of ascorbyl radicals is formed concomitantly with the decrease of the absorption at 345 nm. Thus, the ascorbyl radical is not a primary reaction product. Under the conditions of these experiments, no homolysis of peroxynitrous acid to nitrogen dioxide and hydroxyl radical was observed. Aside from monohydroascorbate's ability to “repair” oxidatively modified biomolecules, it may play a role as scavenger of peroxynitrous acid.  相似文献

The aim of our study was to determine whether a meal modifies the antisecretory response induced by PYY and the structural requirements to elicit antisecretory effects of analogue PYY(22–36) for potential antidiarrhea therapy. The variations in short-circuit current (Isc) due to the modification of ionic transport across the rat intestine were assessed in vitro, using Ussing chambers. In fasted rats, PYY induced a dose- and time-dependent reduction in Isc, with a sensitivity threshold at 5 × 10−11 M (ΔIsc −2 ± 0.5 μA/cm2). The reduction was maximal at 10−7 M (Isc −23 ± 2 μA/cm2), and the concentration producing half-maximal inhibition was 10−9 M. At 10−7 M, reduction of Isc by PYY reached 90% of response to 5 × 10−5 M bumetanide. The PYY effect was partly reversed by 10−5 M forskolin (Isc +13.43 ± 2.91 μA/h·cm2, p < 0.05) or 10−3 M dibutyryl adenosine 3′,5′ cyclic monophosphate (Isc +12 ± 1.69 μA/cm2, p < 0.05). Naloxone and tetrodotoxin did not alter the effect of PYY. In addition, PYY and its analogue P915 reduced net chloride ion secretion to 2.85 and 2.29 μEq/cm2 (p < 0.05), respectively. The antisecretory effect of PYY was accompanied by dose- and time-dependent desensitization when jejunum was prestimulated by a lower dose of peptide. The antisecretory potencies exhibited by PYY analogues required both a C-terminal fragment (22–36) and an aromatic amino acid residue (Trp or Phe) at position 27. At 10−7 M the biological activity of PYY was lower in fed than fasted rats (p < 0.001). Our results confirm the antisecretory effect of PYY, but show that the fed period is accompanied by desensitization, similar to the transient desensitization observed in the fasted period with cumulative doses. This suggests that PYY may act as a physiological mediator that reduces intestinal secretion.  相似文献

Estimation of the ammonia production of the shrimp C. crangon in two littoral ecosystems (oligotrophic sand and eutrophic mud) was determined in winter and summer conditions from laboratory observations in experimental microcosms. The ammonia excretion rate of C. crangon was not influenced by either the sediment type or the ammonia concentration of the overlying water; on the other hand, the mean excretion rate and the response to initial handling stress increased markedly as shrimp were deprived of soft substratum.

The daily ammonia production of C. crangon was 16 μmol NH3 · g −1 wet wt · day −1 in winter and 40 μmol in summer. A gross production of 12 μmol NH3 · m−2 · day −1 and 300–700 μmol μ m−2 · day−1, respectively, could be expected in the two ecosystems studied. This would account for 5% (winter) and 2–4% (summer) of the total NH+4 flux at the sediment-water interface. The contribution of the excretion of all macrofauna to the NH+4 flux from the sediment is discussed.  相似文献

Amperometric choline biosensors were fabricated by the covalent immobilization of an enzyme of choline oxidase (ChO) and a bi-enzyme of ChO/horseradish peroxidase (ChO/HRP) onto poly-5,2′:5′,2″-terthiophene-3′-carboxylic acid (poly-TTCA) modified electrodes (CPMEs). A sensor modified with ChO utilized the oxidation process of enzymatically generated H2O2 in a choline solution at +0.6 V. The other one modified with ChO/HRP utilized the reduction process of H2O2 in a choline solution at −0.2 V. Experimental parameters affecting the sensitivity of sensors, such as pH, applied potential, and temperature were optimized. A performance comparison of two sensors showed that one based on ChO/HRP/CPME had a linear range from 1.0×10−6 to 8.0×10−5 M and the other based on ChO/CPME from 1.0×10−6 to 5.0×10−5 M. The detection limits for choline employing ChO/HRP/CPME and ChO/CPME were determined to be about 1.0×10−7 and 4.0×10−7 M, respectively. The response time of sensors was less than 5 s. Sensors showed good selectivity to interfering species. The long-term storage stability of the sensor based on ChO/HRP/CPME was longer than that based on ChO/CPME.  相似文献

Nitrogen dioxide (NO2) is a key biological oxidant. It can be derived from peroxynitrite via the interaction of nitric oxide with superoxide, from nitrite with peroxidases, or from autoxidation of nitric oxide. In this study, submicromolar concentrations of NO2 were generated in < 1 μs using pulse radiolysis, and the kinetics of scavenging NO2 by glutathione, cysteine, or uric acid were monitored by spectrophotometry. The formation of the urate radical was observed directly, while the production of the oxidizing radical obtained on reaction of NO2 with the thiols (the thiyl radical) was monitored via oxidation of 2,2′-azino-bis-(3-ethylthiazoline-6-sulfonic acid). At pH 7.4, rate constants for reaction of NO2 with glutathione, cysteine, and urate were estimated as 2 × 107, 5 × 107, and 2 × 107 M−1 s−1, respectively. The variation of these rate constants with pH indicated that thiolate reacted much faster than undissociated thiol. The dissociation of urate also accelerated reaction with NO2 at pH > 8. The thiyl radical from GSH reacted with urate with a rate constant of 3 × 107 M−1 s−1. The implications of these values are: (i) the lifetime of NO2 in cytosol is < 10 μs; (ii) thiols are the dominant ‘sink’ for NO2 in cells/tissue, whereas urate is also a major scavenger in plasma; (iii) the diffusion distance of NO2 is 0.2 μm in the cytoplasm and < 0.8 μm in plasma; (iv) urate protects GSH against depletion on oxidative challenge from NO2; and (v) reactions between NO2 and thiols/urate severely limit the likelihood of reaction of NO2 with NO• to form N2O3 in the cytoplasm.  相似文献
